SUMMARY: The present study aimed to investigate the in vitro effect of human adipose derived stem cells (hADSCs) and their secretions on viability of retinal cells and the axonal growth. After isolation, the rat retina was kept in containers covered with poly-D-lysine/laminin, after preparing the supernatant of hADSCs, the retina along with cells and hADSCs conditioned medium (CM) were co-cultured. The viability was assessed by 3-[4,5- dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide (MTT) assay at days 1, 7 and 14, and axonal growth of retinal cells was analyzed by Image J software using Neurite J plug-in. The expression of PI3K/Akt pathway genes and its proteins were evaluated through real-time polymerase chain reaction (PCR) and western blot, respectively. Anti-oxidant effects of hADSCs and their CM were measured by measuring the retina cells level of thiobarbituric acid reactive substance (TBARS), ferric reducing ability of plasma (FRAP) and glutathione (GSH). Brain-derived neurotrophic factor (BDNF), neurotrophin-3 (NT-3) and NT-4 in the CM were measured by enzyme-linked immunosorbent assay (ELISA). The CM contained high levels of BDNF, NT-3 and NT-4 compared to the control group. The expression of genes and proteins of the PI3K/Akt pathway along with levels of TBARS, FRAP and GSH was also increased in the group of hADSCs and their CM compared to the control group. The results of this study showed that hADSCs can increase the axonal growth and survival of rat retinal cells through anti-inflammatory, anti-apoptotic, and paracrine stimulation, as well as through the PI3K/Akt pathway.

RESUMEN: El presente estudio tuvo como objetivo investigar el efecto in vitro de las células madre derivadas de tejido adiposo humano (hADSC) y sus secreciones sobre la viabilidad de las células de la retina y el crecimiento axonal. Después del aislamiento, la retina de la rata se mantuvo en recipientes cubiertos con poli-D-lisina/laminina, después de preparar el sobrenadante de las hADSC, se co-cultivaron la retina junto con las células y el medio acondicionado (CM) de las hADSC. El CM contenía altos niveles de BDNF, NT-3 y NT-4 en comparación con el grupo control. La expresión de genes y proteínas de la vía PI3K/Akt junto con los niveles de TBARS, FRAP y GSH también aumentaron en el grupo de hADSC y sus CM en comparación con el grupo control. ABSTRACT Current corn sowing equipment shows relatively low levels of automation and intelligent response. Issues such as wheel slipping and chain jumping occur in seeders driven by wheels and chains, leading to a decrease in the seeding qualification rate. To solve the problems of wheel slipping and low control strategy accuracy, an electric control sowing system has been designed. This system uses radar to collect the locomotive speed and automatically adjusts the motor speed. To improve the control accuracy of the electric control system, a dual closed-loop control method is employed, and the proportional-integral-derivative (PID) controller parameters are optimized by combining the particle swarm optimization (PSO) algorithm and the fuzzy algorithm. Control accuracy tests and field tests are conducted for both the traditional PID control system and the fuzzy PID control system based on the PSO algorithm. The average error of this system is 0.622%, which is 1.6% lower than that for the traditional method. Field tests show that in operation, the sowing system achieves the average seeding qualification index of 93.99%, which is 3.19% higher than that for the traditional PID control method. The fuzzy PID control system shows improved sowing effectiveness compared to the traditional PID control method.

Abstract A novel kiwifruit wine (KW) fermented by Aspergillus niger and Aspergillus oryzae was developed with elegant flavor and poor alcoholic strength. The effects of natural aging, microwave and ultrasonic treatment on the quality of KW were investigated. A total of 61 aroma components were detected in the KW fermented by Aspergillus niger and Aspergillus oryzae. The results showed that microwave and ultrasonic treatments could promote the maturation of KW and increase the aroma components of KW. The variety of flavor components increased with a longer storage time. Combined with the taste evaluation analysis. The esters, alcohols, acids, and aldehydes in ultrasonic-aged wines were in dynamic balance and harmony with each other. The wine was soft and mellow, making the kiwi wine more typical.

ABSTRACT Purpose: To investigate the role and mechanism of β1,3-N-acetylglucosaminyltransferase-3 gene (B3GNT3) in esophageal cancer (ESCA). Methods: The starBase database was used to evaluate the expression of B3GNT3. B3GNT3 function was measured using KYSE-30 and KYSE-410 cells of esophageal squamous cell carcinoma (ESCC) cell lines. The mRNA levels were detected by quantitative real-time polymerase chain reaction (qRT-PCR). Cell counting kit-8, clone formation assay and transwell assay were used to detect the changes of proliferation, invasion and migration. Results: B3GNT3 expression was higher in ESCA tissues than in normal tissues. The overall survival rate of ESCA patients with high B3GNT3 expression was lower than that of ESCA patients with low B3GNT3 expression. In vitro functional experiments showed that the proliferation ability, migration and invasion ability of KYSE-30 and KYSE-410 cells with B3GNT3 interference were lower than those of the control, and the overexpression of B3GNT3 had the opposite effect. After silencing B3GNT3 expression in ESCC cell lines, the growth of both cell lines was inhibited and the invasiveness was decreased. Knockdown of B3GNT3 reduced the growth rate and Ki-67 expression level. Conclusions: B3GNT3, as an oncogene, may promote the growth, invasion and migration of ESCC cell. Abstract This study was intended to elucidate the effects of HACE1 in Citrobacter rodentium-Induced Murine Colitis. Male C57BL/6 mice orally administered with 100 µLof Citrobacter rodentium DBS-100 (109 CFU mL−1) for 14 days. The expression of HACE1 in Citrobacter rodentium-Induced Murine Colitis was increased. HACE1 promoted inflammation and ROS levels in macrophage through the inactivation of Nrf2/NLRP3 signaling pathway. Meanwhile, si-HACE1 reduced inflammation and ROS levels in macrophage via the inhibition of Nrf2/NLRP3 signaling pathway. Sh-HACE1 reduced inflammatory responses in Citrobacter rodentium-Induced Murine Colitis through the promotion of Nrf2/NLRP3 signaling pathway. IP showed that HACE1 protein catenated with Nrf2 protein to suppress ROS-induced NLRP3 in macrophage through the promotion of Nrf2 Ubiquitination. Nickel-based alloys containing high amount of Cr are desirable materials for the production of critical components that operate under high temperature and severe corrosion conditions. The description of flow behaviors for these alloys is key to designing the hot formation process for achieving the excellent mechanical properties of components. This work aims to study the flow behaviors of Ni-38Cr-3.8Al alloy during thermal deformation. A series of isothermal compression experiments were carried out at temperatures of 1148 K, 1223 K, 1298 K, 1373 K, 1448 K and 1523 K, and strain rates of 0.01 s-1, 0.1 s-1, 1 s-1 and 10 s-1. Results show that the obtained true stress-strain curves exhibit the softening characteristics of dynamic recrystallization (DRX) and dynamic recovery (DRV). The flow stress decreases with temperature increasing and strain rate decreasing. To further quantitatively describe the flow behaviors, an improved segmented Arrhenius constitutive equation was developed according to the softening characteristics under different temperatures. In these equations, the variable coefficients, including activation energy Q and material constants (structure factor A, stress exponent n, stress parameters α), were fitted as the polynomial functions of true strain. Furthermore, a typical Arrhenius constitutive equation was also solved. The comparisons between experimental stress and predicted results obtained from the typical and improved constitutive equations were conducted, and the correlation coefficient (R) and average absolute relative error (AARE) were calculated as 0.932, 15.83%, and 0.9963, 5.61%, respectively. It suggests that the improved constitutive equation can adeptly describe the flow behaviors of Ni-38Cr-3.8Al alloy.

Portanto, fortalecer a resistência física dos jogadores de futebol através de métodos específicos de treinamento é um importante fator para a otimização do desempenho atual. Objetivo Estudar a aplicação do treinamento funcional no condicionamento físico dos jogadores de futebol. Métodos 20 atletas estudantes de educação física de futebol júnior em faculdades e universidades foram selecionados como objeto de pesquisa. A formação funcional global foi dividida em três etapas: estágio de prática, fase de adaptação e etapa de promoção. Os dados foram comparados, integrados e analisados antes e após a intervenção. Resultados A realização de treinamentos funcionais direcionados para jogadores de futebol pode efetivamente aumentar a resistência física dos atletas, reduzindo a ocorrência de lesões esportivas e melhorando a pontuação geral do condicionamento físico a nível técnico e de estabilidade. Conclusão A partir da pesquisa deste artigo, pode-se ver que há falta de aptidão física e força técnica no futebol chinês atual. A realização de treinamento funcional direcionado é relevante e merece ser aplicado ao treinamento de futebol. ABSTRACT Introduction Although Chinese soccer has experienced many updates in its methods, there is still a large gap in players’ physical endurance compared to the world powers. Therefore, strengthening soccer players’ physical endurance through specific training methods is important in optimizing current performance. Objective Study the application of functional training in soccer players’ physical conditioning. Methods 20 junior soccer physical education student-athletes in colleges and universities were selected as the research object. The global functional training was divided into three stages: practice, adaptation, and promotion. Data were compared, integrated, and analyzed before and after the intervention. Results Conducting targeted functional training for soccer players can effectively increase athletes’ physical endurance, reducing sports injuries and improving overall fitness scores at the technical and stability level. Conclusion From the research of this article, it can be seen that there is a lack of physical fitness and technical strength in Chinese soccer today. El entrenamiento funcional global se dividió en tres etapas: etapa de práctica, etapa de adaptación y etapa de promoción. Los datos se compararon, integraron y analizaron antes y después de la intervención. Resultados La realización de un entrenamiento funcional específico para los futbolistas puede aumentar eficazmente la resistencia física de los deportistas, reduciendo la aparición de lesiones deportivas y mejorando la puntuación global de la aptitud a nivel técnico y de estabilidad. Conclusión De la investigación de este artículo se desprende que en el fútbol chino actual hay una falta de aptitud física y de fuerza técnica. El rendimiento del entrenamiento funcional dirigido es relevante y merece ser aplicado al entrenamiento del fútbol. ABSTRACT Introduction The improvement of soccer sports skills depends on many training efforts and is closely related to intrinsic scientific methods. Attention to the integral quality of the lower limb muscles and the performance of specific exercises of technical movements is essential to reach the optimal state of physical performance in players. Objective Analyze the kinematic effect of lower limb movement techniques in soccer training. Methods 10 athletes were marked with reflective spheres and submitted to the kinematic training method of the lower limbs designed by the coaches in 60 minutes, three times a week, for six weeks. Before and after the experiment, data captured by reflective spheres were captured, compared, classified, and analyzed. Results Kinematic training can effectively optimize movement time, swing amplitude, swing angle, and other aspects of lower limb mechanical structure, thus improving energy expenditure and making the movement concise, conveying strength and precision. ABSTRACT Due to its high nutritional value, broccoli (Brassica oleracea var. italica Plenck) is one of the most popular vegetables worldwide. This study assessed 36 phenotypic characteristics of 111 broccoli varieties to understand the phenotypic diversity of new broccoli varieties and improve their breeding speed with advantages and characteristics in China, including 108 new varieties and three varieties of common knowledge. The genetic diversity, the principal component, and the cluster of phenotypic characteristics of broccoli varieties were further investigated. The results showed that the coefficients of variation of 36 characteristics ranged between 11.18 % and 94.99 %, with their diversity index between 0.26 and 1.82. The 111 broccoli varieties were further classified into eight groups, primarily attributed to the differences in phenotypic characteristics, including curd weight, main stem thickness, plant development degree, plant height, and anthocyanin coloration. The cumulative contribution rate of the first five principal components reached 81.186 %, corresponding to 12 representative phenotypic traits. The analysis indicated that the phenotypic characteristics of broccoli were rich in diversity, especially for several characteristics appreciated by the market, such as weight, curd firmness, and anthocyanin coloration. This study revealed the basic information on the genetic diversity of new broccoli varieties in China from 2017 to 2019 and provided potential breeding strategies for broccoli to meet diverse market demands.

Abstract Background Duchenne muscular dystrophy (DMD) is an X-linked lethal genetic disorder for which there is no effective treatment. Previous studies have shown that stem cell transplantation into mdx mice can promote muscle regeneration and improve muscle function, however, the specific molecular mechanisms remain unclear. DMD suffers varying degrees of hypoxic damage during disease progression. This study aimed to investigate whether induced pluripotent stem cells (iPSCs) have protective effects against hypoxia-induced skeletal muscle injury. Results In this study, we co-cultured iPSCs with C2C12 myoblasts using a Transwell nested system and placed them in a DG250 anaerobic workstation for oxygen deprivation for 24 h. We found that iPSCs reduced the levels of lactate dehydrogenase and reactive oxygen species and downregulated the mRNA and protein levels of BAX/BCL2 and LC3II/ LC3I in hypoxia-induced C2C12 myoblasts. Meanwhile, iPSCs decreased the mRNA and protein levels of atrogin-1 and MuRF-1 and increased myotube width. Furthermore, iPSCs downregulated the phosphorylation of AMPKA and ULK1 in C2C12 myotubes exposed to hypoxic damage. Conclusions Our study showed that iPSCs enhanced the resistance of C2C12 myoblasts to hypoxia and inhibited apoptosis and autophagy in the presence of oxidative stress. Further, iPSCs improved hypoxia-induced autophagy and atrophy of C2C12 myotubes through the AMPK/ULK1 pathway. This study may provide a new theoretical basis for the treatment of muscular dystrophy in stem cells.

RESUMO Introdução: A atividade física acontece por meio coordenação de vários sistemas de órgãos do corpo humano, e os exercícios físicos podem ter impacto positivo nas atividades de vários desses sistemas. Uma vez que o impacto de esportes de alta intensidade nos órgãos humanos é diferente de acordo com a temperatura do ambiente e com a intensidade do exercício, é necessário realizar observações e discussões mais detalhadas. Objetivo: Analisar as mudanças de forma e função do corpo, e das funções dos órgãos de alunos universitários antes e depois de atividade física de alta intensidade, através do estudo do movimento humano. Métodos: Pesquisa experimental sobre as características da troca gasosa em exercícios de alta intensidade com aumento gradual de carga em alunos universitários. Resultados: Houve diferenças estatísticas significativas na incidência de reações fisiológicas dos estudantes que passaram pela atividade de alta intensidade com aumento gradual de carga, incluindo dores abdominais, dispneia, taquicardia, náusea, tonturas, e dores musculares (p<0,05). Na presença de hipotermia havia uma queda na frequência cardíaca durante os exercícios. Conclusão: Exercícios de sobrecarga podem mediar funções fisiológicas eficientemente. Para melhorar a performance esportiva em ambientes de baixa temperatura é importante realizar exercícios de aquecimento para aumentar a temperatura do corpo. ABSTRACT Introduction: Physical activity is accomplished by the coordination of various organ systems of the human body, and physical exercise can positively impact the activities of many of these systems. Because the impact of high-intensity sports on human organs is different according to the environmental temperature and exercise intensity, we must make more detailed observations and discussions. Objective: To analyze the changes in the body shape, function, and organ function of middle school students before and after high-intensity physical exercise through the study of human movement. Methods: Through the experimental research on the characteristics of metabolic gas exchanges in high-intensity incremental load exercise of college students. Results: There were statistically significant differences in the incidence of physiological reactions shown by the students who underwent high-intensity incremental load exercise, such as abdominal pain, dyspnea, tachycardia, nausea, dizziness, and muscle aches (P<0.05). Hypothermia was accompanied by a decrease in heart rate during exercise. Conclusion: Carrying out overload training can effectively mediate physiological functions. It is an important, in improving sports performance, to carry out warm-up activities in a low-temperature environment to increase body temperature. RESUMEN Introducción: La actividad física ocurre por medio de la coordinación de varios sistemas de órganos del cuerpo humano y los ejercicios físicos pueden tener impacto positivo en las actividades de varios de estos sistemas. Dado que el impacto de deportes de alta intensidad en los órganos humanos es diferente de acuerdo con la temperatura del ambiente y con la intensidad del ejercicio, es necesario realizar observaciones y discusiones más detalladas. Objetivo: Analizar los cambios de forma y función del cuerpo, y de las funciones de los órganos de alumnos universitarios antes y después de la actividad física de alta intensidad, a través del estudio del movimiento humano. Métodos: Investigación experimental sobre las características del intercambio de gases en ejercicios de alta intensidad con aumento gradual de carga en alumnos universitarios. Resultados: Hubo diferencias estadísticas significativas en la incidencia de reacciones fisiológicas de los estudiantes que pasaron por la actividad de alta intensidad con aumento gradual de carga, incluyendo dolores abdominales, disnea, taquicardia, náuseas, mareos y dolor muscular (p<0,05). En la presencia de hipotermia se produjo un descenso cardíaco durante los ejercicios. Conclusión: Los ejercicios de sobrecarga pueden mediar las funciones fisiológicas eficientemente. Para mejorar el rendimiento deportivo en ambientes de baja temperatura es importante realizar ejercicios de calentamiento para aumentar la temperatura corporal. Abstract The processes of racial and socioeconomic school segregation, the inequalities that generates in learning opportunities and its relationship with low compulsory school completion rates, constitute a serious obstacle to social and educational equity. In this article we report a design-based research currently under development and lasting more than four years, which is part of a set of actions for educational change in an institute-school from Barcelona (Catalonia), Our aim is to carry out a historical analysis of the transformations and tensions in this system of activity and among its participants: educational team and school students, undergraduate students and researchers. We observe that the legitimization and negotiation of their reasons and goals have led to processes of appropriation and agency of the project, and we identify tensions and contradictions that are analyzed under the power relations that happens in schools.

Resumen Los procesos de segregación educativa por motivos raciales y socioeconómicos, las desigualdades que genera en las oportunidades de aprendizaje y su relación con las bajas tasas de finalización de estudios obligatorios constituyen un serio obstáculo para la equidad social y educativa. En este artículo damos cuenta de una investigación basada en el diseño, actualmente en desarrollo y con más de cuatro años de duración, que forma parte de un conjunto de medidas para el cambio educativo en un instituto-escuela en la ciudad de Barcelona. Nuestro objetivo es realizar un análisis histórico atendiendo a las transformaciones y tensiones en este sistema de actividad y entre sus participantes: equipo educativo y alumnado del centro, estudiantes universitarias e investigadoras. Observamos que la legitimación y negociación de sus motivos y metas han llevado a procesos de apropiación y agencia del proyecto e identificamos tensiones y contradicciones que son analizadas a la luz de las relaciones de poder en las escuelas.

Abstract High fat diet may cause obesity, diabetes, atherosclerosis, hypertension, hyperlipidemia and other diseases, and even mortality. The recombinant rice based on mixing lotus seed starch and broken rice flour was used to feed high-fat diet rats, and its effects on body weight and organ index were determined. The changes of intestinal flora in rats were studied by macrogenomic technology, and the effects of lotus seed starch-broken rice flour recombinant rice (LSBR) on intestinal flora in rats were discussed. The results showed that adding enough LSBR could effectively inhibit the weight gain of rats, reduce the liver index from 0.3498 to 0.2836, and the kidney index from 0.0079 to 0.0072. The reduction of intestinal flora abundance and diversity caused by high-fat diet were improved, the relative abundance ratio of Firmicutes/Bacteroidete was reduced from 11.04 to 2.17, while the relative abundance of beneficial bacteria was increased.

Abstract Mesenchymal stem cells (MSCs) derived exosomes (Exos) are one of the most promising candidate for the treatment of this condition. However, the underlying molecular mechanism remains uncertain. Here we investigated the therapeutic effect of exosomal miR-181c-5p (ExomiR-181c-5p) on a rat model of neuropathic pain induced by sciatic nerve chronic constriction injury (CCI). In this study NP model was established using the CCI method. NP levels were assessed using PWT and PWL. Microarray analysis and RT-PCR were used to determine the relative expression of miR-181c-5p. MSC-derived exosomes were extracted using the total exosome isolation reagent characterized by WB and NTA. MiR-181c-5p was loading into Exos using electroporation. The inflammation response in microglia cells and CCI rats were assessed by ELISA assay respectively. Our study demonstrates that miR-181c-5p expression was obviously decreased in a time-dependent manner in CCI rats. MiR-181c-5p was effectively electroporated and highly detected in MSC-derived Exos. ExomiR-181c-5p internalized by microglia cells and inhibit the secretion of inflammation factors. ExomiR-181c-5p intrathecal administration alleviated neuropathic pain and neuroinflammation response in CCI rats. Taken together, ExomiR-181c-5p alleviated CCI-induced NP by inhibiting neuropathic inflammation. ExomiR-181c-5p may be a valid alternative for the treatment of neuropathic pain and has vast potential for future development.
