Abstract Lyophyllum decastes is a common macrofungi for both medicinal and economic value, and its polysaccharide has excellent development potential as a new immune regulator. In the present study, the bioactive component polysaccharide of L decastes was extracted, and its chain structure was characterized in detail by Fourier transform infrared (FT-IR) spectroscopy, high-performance gel-permeation chromatography (HP-GPC), and gas chromatography–mass spectrometry (GC–MS). LDP-W had a molecular weight of 2.12 × 104 Da, highly branched β type pyranose. At the same time, it exhibited potential immunomodulatory activity, which can modulate the numbers of CD3+, CD4+, CD8+ and CD19+ cells in the spleen; optimize the CD4+/CD8+ ratio; promoted T and B lymphocyte proliferation and it can elevate the levels of IL-2, IL-6, IFN-γ and TNF-α in immunosuppressed mice by influencing the phosphorylation of MAPK signaling-related proteins. These studies fill the gap in the immune regulation mechanism of L decastes polysaccharide, providing a theoretical basis for the potential application of LDP-W as immunomodulatory drugs or functional foods.

ABSTRACT The objective of this study was to assess the effect of Suanzaoren decoction on stress response of horses in transportation. A total of six male Ili horses were equally divided into Suanzaoren decoction treatment group (n = 3; basal diet supplemented with Suanzaoren decoction, three times/day) and control group (n = 3; basal diet, three times/day). After feeding for five days, all horses were transported for 8 h to simulate stress. Blood and serum samples were obtained before transport (BT), during transport (T), and after transport (AT). Results showed that there was significant interaction between road transport and dietary Suanzaoren decoction supplementation for the white blood cells (WBC), intermediate cell ratio (MID%), granulocyte ratio (GRAN%), alanine aminotransferase (ALT), glucose (GLU), aspartate transaminase (AST), creatine (CK), cortisol (COR), human growth hormone (HGH), adrenocorticotropic hormone (ACTH) and arginine vasopressin content, insulin (INS), and thyronine 4 (T4) in blood and serum. The highest WBC was found in T group. The highest GRAN%, ALT, GLU, AST, CK, T4, ACTH, and INS contents were observed in serum from AT groups, which was decreased by treatment. In conclusion, dietary Suanzaoren decoction supplementation did relieve horse transportation stress. This study provides a useful clinical therapy for relieving transportation stress of horses.

Abstract Basic leucine zipper (bZIP) is a conserved transcription factor (TF) widely present in eukaryotes, and it plays an important role in regulating plant growth and stress responses. To better understand the white pear bZIP gene family, comprehensive bioinformatics analysis of the pear genome was performed. A total of 84 PbbZIP genes were identified, which were divided into 13 subfamilies by phylogenetic analysis. The 84 PbbZIP genes were all located in the nucleus, and 77 of those genes were unevenly distributed across the 17 chromosomes of white pear. The other 7 PbbZIP genes were located on the scaffold. Subsequent expression profile analysis showed that PbbZIP genes in exocarp were significantly upregulated or downregulated in ‘Huangguan’ pear with brown spot (BS) compared with healthy pear and in response to hormonal treatment with gibberellin A3 (GA3). These results provide helpful insights into the characteristics of PbbZIP genes and their responses to BS in ‘Huangguan’ pear.

Abstract To observe the eradication rate, safety and compliance of bismuth quadruple therapies based on minocycline in the newly diagnosed patients with Helicobacter pylori (Hp) infection. We selected 150 penicillin allergic patients for the initial treatment of Hp infection treated in Beijing Jishuitan hospital from January 2018 to February 2019. Subsequently, the patients were randomly selected into the minocycline and metronidazole-containing group (RMMB) and minocycline and levofloxacin-containing group (RMLB). The total course of treatment is 14 days. Follow-up clinical manifestations 3 to 5 days after treatment. The 13C urea breath test was performed 4 to 12 weeks to confirm the eradication of Hp. There were no significant differences in baseline data between the two groups (p>0.05). According to ITT analysis, the eradication rates of RMMB group and RMLB group were 80.3% (61/76) and 89.2% (66/74), respectively (χ2=2.301, p=0.129). According to PP analysis, the eradication rates of the RMMB group and the RMLB group were 83.6% (61/73) and 90.4% (66/73), respectively (χ2=1.513, p=0.21). The overall incidences of adverse events were 47.4% and 38.8% in the RMMB and RMLB groups, respectively (p=0.924). In patients with penicillin allergy, the eradication rate of minocycline-based bismuth quadruple therapies is ideal and the side effects are few. It is recommended for the eradication treatment of Hp-infected patients with penicillin allergy.

Abstract In recent decades, with the development of the pathogenesis of Acute severe pancreatitis (SAP) and the improvement of clinical treatment techniques, the mortality rate of SAP is still over 10% . Recent studies have shown that immune dysregulation plays an important role in the pathogenesis of AP, especially in SAP. A meta-study points to widespread concern for the discovery of immunosuppression by networked cytokines in the course of AP. In the early stages of AP, high cytokine levels play a leading role in the development of SIRS pathological conditions, which result in a cascade of inflammatory cytokines that induce the release of immunocompetent cells, leading to an excessive immune response. In the later stages of Compensatory anti-inflammatory response syndrome (CARS), the body over-releases anti-inflammatory agents and has an immunosuppressive process, but contributes to the development of the risk of secondary infection and increases the likelihood of Multiple organ disfunction syndrome (MODS). The study was aimed to measure T-lymphocyte immune function changes of patients with acute pancreatitis and to explore BISAP scoring system in clinical applications.

Abstract In order to reduce post-harvest losses, the classification of fresh apples is crucial. Taking the hierarchical transmission control system as the object, the research was carried out on the verification of the bus network can flexibly expand the motor equipment, the stable and reliable operation of the motor, and the accuracy of Apple's classification. Combine Labview virtual instrument technology to realize the design of Apple's hierarchical transmission control system based on Controller Area Network technology. Fuzzy PID and traditional PID algorithms are used to simulate and realize the operation of brushless DC motor, and compare the advantages of brushless DC motor control based on fuzzy PID to ensure the safe and stable operation of the system. Using the machine learning algorithms model for color detection, the Support Vector Machine algorithm model finally achieved the classification of the three types of apple samples with a recognition rate of 96.7%.

ABSTRACT Purpose: To explore the potential immunomodulatory effects of total extract and different polar parts from Blaps rynchopetera Fairmaire. Methods: Phagocytic activity was evaluated by neutral red assay, and the effect of the immune function was investigated by normal and immunocompromised mice models. Results: In vitro, total extract, as well as chloroform, ethyl acetate, n-butanol and water fractions could individually enhance the phagocytic ability of mouse peritoneal macrophages; in addition, chloroform and ethyl acetate fractions had an increasing tendency when combined stimulation with lipopolysaccharide (LPS). In vivo, ethyl acetate fraction (EAF) could enhance the immune organ index, increase the serum hemolysin level and peripheral blood immune cells of immunocompromised mice, while for normal mice, the effect was inconspicuous. Conclusions: Blaps rynchopetera extracts had noteworthy immunomodulatory effect, especially for individuals with immune disorders.

Abstract In this study, through animal experiments, we investigated the effects of Qingli Gandan decoction on the IL-23/IL-17 signalling pathway and the expression of CD4+/IL-17A+ cells in EAU rats. We then predicted the potential mechanisms of the action. First, Lewis rats were divided using the random number table method into a (1) control group, (2) model group and (3) TCM group. The TCM group was given a Qingli Gandan prescription by gavage after an intervention model. Next, blood, aqueous humour and vitreous were collected from each group on days 4, 8, 12, 16 and 20 after immunisation. An enzyme linked immunosorbent assay (ELISA) was used to detect the protein concentrations of IL-17 and IL-23 in a serum, aqueous humour and vitreous of rats on different immunisation days, and the percentage of CD4+/IL-17A+ cells in rat spleen was detected by flow cytometry. The peak concentration of serum IL-17 protein in group 3 was statistically lower than that in group 2 (P < 0.05) on days 4, 8 and 12 of immunisation. However, the peak concentration of serum IL-23 protein in group 3 was statistically lower than in group 2 (P < 0.05) on days 8 and 12 of immunisation; in addition, on days 8 and 12, the protein concentrations of IL-23 and IL-17 in the aqueous humour group 3 were all lower than those in group 2. On days 4, 8 and 12, the protein concentrations of IL-23 and IL-17 in vitreous in group 3 were significantly lower than those in group 2. We found that the proportion of CD4+/IL-17A+ cells in the groups 2 and 3 increased on day 8 compared with group 1 (P < 0.05). The proportion of CD4+/IL-17A+ cells decreased in group 3 compared with group 2 (P < 0.05).At last, we think Qingli Gandan can reduce the onset of EAU in rats and treat it, presumably because of inhibiting the IL-23/IL-17 axis. It may inhibit the expression of CD4+/IL-17A+ cells and play a role in inhibiting the IL-23/IL-17 signalling pathway.

Abstract Due to the good functional characteristics and nutritional value, ovalbumin (OVA) has a widely ranges of applications in food industry. But the characteristics of being susceptible to external conditions, such as processing, storage conditions and food ingredients, makes it display a relative lower functional property and limits its applications. Glycosylation is a natural, green approach of protein modifications that has an impact on its functional properties. In this work, we prepared the OVA-pectin conjugates through wet-heating and ultrasound glycosylation to investigate the effects on the functional and structural properties of OVA. Both wet-heating and ultrasound treated OVA at present of pectin showed higher emulsify and foam properties than untreated OVA. The results of particle size, SDS-PAGE, FT-IR, and SEM indicated that the covalent binding of pectin with OVA molecule. This study suggests that wet-heating and ultrasound glycosylation with pectin are promising method to improve the functional properties of OVA.

Abstract Objective: Long Non-Coding RNAs (LncRNAs) act as an indispensable role in cancer development. The study aimed to investigate the role and mechanism of lncRNA Small Nucleolar RNA Host Gene 1 (SNHG1) in Bladder Cancer (BC) progression. Method: The expression, prognostic value, diagnostic value, and correlation of SNHG1, Enhancer of Zeste 2 polycomb repressive complex 2 subunit (EZH2), and Kruppel Like Factor 2 (KLF2) were analyzed through bioinformatics analysis. The expression was also validated in BC tissues and cell lines. Besides, their regulation and binding were tested via qPCR, Western blot, Dual-Luciferase Reporter Assay (DLRA), Argonaute RISC catalytic component 2-RNA Immunoprecipitation (AGO2-RIP), and Chromatin Immunoprecipitation (ChIP). A xenograft model in nude mice was also established. Results: SNHG1 was significantly overexpressed in BC tissues and cells. Importantly, SNHG1 was associated with poor survival, and ROC curves revealed high diagnostic values. Moreover, by CCK8, wound healing, transwell, and Western blot analysis, SNHG1 knockdown significantly inhibited the proliferation, migration, invasion, and epithelial-mesenchymal transition of BC cells. Additionally, in vivo experiments showed that silencing SNHG1 hindered tumorigenesis and tumor growth. Regarding mechanism, the results of AGO2-RIP, ChIP or DLRA showed that SNHG1 played different roles at diverse subcellular sites. In the cytoplasm, SNHG1 acted as a competing endogenous RNA for miR-137-3p to promote EZH2 expression. In the nucleus, SNHG1 could interact with EZH2 to inhibit KLF2 transcription. Conclusion: Our study elucidated that SNHG1 formed a regulatory network and played an oncogenic role in BC, which provided a novel therapeutic target for BC treatment.

Abstract INTRODUCTION: Artemisinin-based combination therapy (ACT), such as artemisinin-piperaquine (AP), dihydroartemisinin-piperaquine (DP), and artemether-lumefantrine (AL), is the first-line treatment for malaria in many malaria-endemic areas. However, we lack a detailed evaluation of the cardiotoxicity of these ACTs. This study aimed to analyze the electrocardiographic effects of these three ACTs in malaria patients. METHODS: We analyzed the clinical data of 89 hospitalized patients with falciparum malaria who had received oral doses of three different ACTs. According to the ACTs administered, these patients were divided into three treatment groups: 27 treated with AP (Artequick), 31 with DP (Artekin), and 31 with AL (Coartem). Electrocardiograms and other indicators were recorded before and after the treatment. The QT interval was calculated using Fridericia’s formula (QTcF) and Bazett’s formula (QTcB). RESULTS: Both QTcF and QTcB interval prolongation occurred in all three groups. The incidence of such prolongation between the three groups was not significantly different. The incidence of both moderate and severe prolongation was not significantly different between the three groups. The ΔQTcF and ΔQTcB of the three groups were not significantly different. The intra-group comparison showed significant prolongation of QTcF after AL treatment. CONCLUSIONS: Clinically recommended doses of DP, AL, and AP may cause QT prolongation in some malaria patients but do not cause torsades de pointes ventricular tachycardia or other arrhythmias.

OBJECTIVES: To investigate the clinical utility of serum microRNA levels (miR-9-5p and miR-128-3p) in the diagnosis and prognosis of early-stage acute ischemic stroke (AIS). METHODS: We compared the differences in serum miR-9-5p and miR-128-3p levels between patients with AIS and healthy individuals (controls). The serum levels of miR-9-5p and miR-128-3p were quantified using quantitative real-time PCR, and the association of each miRNA with AIS was determined using receiver operator characteristic curve analysis. The predictive value of these indices in the diagnosis of early-stage AIS was evaluated in conjunction with that of computed tomography findings and neuron-specific enolase levels. The prognosis of patients with AIS was evaluated three months after their discharge from hospital using the modified Rankin scale, which classifies the prognosis as either favorable or poor. Logistic regression analysis was used to analyze the correlation between miR-9-5p and miR-128-3p levels and patient prognosis. RESULTS: The serum levels of miR-9-5p and miR-128-3p were upregulated in patients with AIS relative to those in healthy individuals. A pronounced correlation was identified between serum miR-9-5p and miR-128-3p levels and patient prognosis, with high levels of both miRNAs being associated with poor patient outcomes. CONCLUSION: Assessment of serum miR-9-5p and miR-128-3p levels is important for the early diagnosis and prognosis of AIS.

T0001 is the first mutant of etanercept with a higher affinity to tumor necrosis factor α (TNF-α) than etanercept. In order to investigate the safety and tolerability of T0001, a study was carried out in healthy Chinese subjects. A first-in-human, dose escalation study was conducted in healthy Chinese subjects. Fifty-six subjects were divided into six dose cohorts (10 mg, 20 mg, 35 mg, 50 mg, 65 mg and 75 mg) to receive a single subcutaneous injection of T0001. Safety and tolerability assessment were based on the records of vital signs, physical examinations, clinical laboratory tests, 12-lead electrocardiograms and adverse events (AEs). All subjects were in good compliance and none withdraw due to AEs. No serious AEs occurred. A total of twenty-three AEs in sixteen subjects were recorded, and eighteen of these AEs were believed to be related to T0001. The most frequently reported AEs were injection site reactions and white blood cell count increase. All these AEs were of mild to moderate intensity and most of them recovered spontaneously within 14 days. In this study, no dose-limiting toxicity was observed, and the maximum tolerated dose was identified as 75 mg. T0001 was considered safe and generally well tolerated at doses up to 75 mg in healthy Chinese volunteers.

Abstract The compressive and tensile behaviors in a Ti nanopillar with a biphasic hexagonal close-packed (HCP) /face-centered cubic (FCC) phase boundary are theoretically researched using classic molecular dynamic simulation. The results indicate that the HCP/FCC interface and free surface of the nanopillar are the sources of dislocation nucleation. The plastic deformation is mainly concentrated in the metastable FCC phase of the biphasic nanopillar. Under compressive loading, a reverse phase transformation of FCC to the HCP phase is induced by the dislocation glide of multiple Shockley partial dislocations 1 2 < 1 ¯ 21 >under compressive loading. However, for tensile loading a large number of Lomer-Cottrell sessile dislocations and stacking fault nets are formed when the partial dislocations react, which leads to an increase in stress. The formation mechanism of a Lomer-Cottrell sessile dislocation is also studied in detail. Shockley partial dislocations are the dominant mode of plastic deformation behaviors in the metastable FCC phase of the biphasic nanopillar.

Abstract We report on the genetic analysis of a Chinese family in which four male patients presented with postlingual progressive hearing loss, associated with distal muscle wasting and unsteady ataxic gait. Using whole exome sequencing, we identified a new pathogenic variant (c.1463C>T, p.Pro488Leu) in the AIFM1 gene, which encodes the apoptosis-inducing factor mitochondrion-associated 1 precursor. AIFM1 is involved in the mitochondrial respiratory chain and cellular caspase-independent apoptosis pathway and has been reported to cause multiple phenotypes including hearing loss. The p.Pro488Leu missense variant segregated with symptoms in the pedigree. It was not found in the dbSNP database, databases of genomes and SNPs in the Chinese population, in 74 patients with sporadic hearing loss, or in 108 normal individuals.We also verified that this AIFM1variant enhanced cell apoptosis rates compared in 293T cells transfected with wild-type AIFM1. Different variations of AIFM1 give rise to different phenotypes in patients, and this is the second reported family with a variant in the C-terminal domain of AIFM1 showing the phenotype of hearing loss and peripheral neuropathy.
