No presente trabalho, objetivou-se avaliar a soroprevalência da infecção por Toxoplasma gondii, em 708 matrizes suínas dos municípios de Nova Mutum e Diamantino do Estado de Mato Grosso, Brasil. As amostras de soro foram examinadas por meio da reação de imunofluorescência indireta (RIFI). Foi encontrada a frequência de 12,8% de soros positivos, com diluições iguais ou superiores a 64. Portanto, os dados obtidos reforçam a necessidade de um manejo de criação adequado, visando à minimização do risco de infecção de suínos por T. gondii.

This report aimed to assess the seroprevalence of Toxoplasma gondii infection in 708 swine matrices in Nova Mutum and Diamantino in the state of Mato Grosso, Central-West Brazil. Serum samples were examined by indirect fluorescent antibody test (IFAT). It was found a seroprevalence of 12.8%, considering titers >64. Therefore, the data reinforce the need for appropriate management of swine raising to minimize the risk of infection of pigs with T. gondii.

Toxoplasmosis is a worldwide zoonosis that generally produces an asymptomatic infection. In some cases, however, toxoplasmosis infection can lead to ocular damage. The immune system has a crucial role in both the course of the infection and in the evolution of toxoplasmosis disease. In particular, IFN-³ plays an important role in resistance to toxoplasmosis. Polymorphisms in genes encoding cytokines have been shown to have an association with susceptibility to parasitic diseases. The aim of this work was to analyse the occurrence of polymorphisms in the gene encoding IFN-³ (+874T/A) among Toxoplasma gondii seropositive individuals, including those with ocular lesions caused by the parasite, from a rural population of Santa Rita de Cássia, Barra Mansa, state of Rio de Janeiro, Brazil. Further, we verified which of these polymorphisms could be related to susceptibility to the development of ocular toxoplasmosis. This study included 34 individuals with ocular toxoplasmosis (ocular group) and 134 without ocular lesions (control group). The differences between A and T allele distributions were not statistically significant between the two groups. However, we observed that a higher frequency of individuals from the ocular group possessed the A/A genotype, when compared with the control group, suggesting that homozygocity for the A allele could enhance susceptibility to ocular toxoplasmosis in T. gondii infection.

Achados de necropsia em três casos de toxoplasmose ocorridos naturalmente em macacos-de-cheiro (Saimiri sciureus) obtidos de capturas na região Amazônica foram descritos. A avaliação histopatológica do fígado, baço, rins e linfonodos mesentéricos mostrou múltiplos focos de inflamação e necrose. Cistos morfológicos típicos e bem definidos de Toxoplasma gondii e formas livres foram detectados pelo método da imunoperoxidase com infiltrados inflamatórios. A presença de T. gondii na colônia de primatas estudada pode influenciar severamente os resultados de experimentos realizados nos animais.

Necropsy findings in three cases of naturally occurring toxoplasmosis in squirrel monkeys (Saimiri sciureus) obtained as wild catches from the Amazon region were described. Histopathological evaluation of the liver, spleen, kidneys and mesenteric lymph nodes showed multiple foci of inflammation and necrosis. Typical, well-defined morphologic Toxoplasma gondii cysts and free forms of the parasite were found by immunoperoxidase staining within inflammatory infiltrates. The presence of T. gondii in the studied colony could severely influence the results of experiments performed in the animals.

O risco de infecção por Toxoplasma gondii em trabalhadores de um frigorífico de suínos em Palmas, Paraná, Brasil, foi estudado comparativamente a outros indivíduos que não mantinham contato com animais e suas carcaças. As 174 amostras de soros foram divididas em dois grupos: grupo 1 - funcionários de um matadouro-frigorífico de suínos (133) e grupo 2 - controle (41). No ato da coleta, todos os participantes responderam a um questionário epidemiológico com as seguintes variáveis: sexo, idade, contato com gatos e solo, hábito de ingerir carne crua ou mal cozida, embutidos crus e leite cru. Todas as amostras de soros foram submetidas aos testes ELISA (Enzyme linked immunosorbent assay) e RIFI (Reação de imunofluorescência indireta) - IgG anti-T. gondii; por sua vez, para IgM anti-T. gondii, foi utilizada a RIFI. Todos os 174 indivíduos eram IgM soro não-reagentes. No grupo 1, 48,1% eram IgG sororreagentes na RIFI e 58,6% na ELISA. No grupo 2, 39,0% eram IgG sororreagentes na RIFI e 51,2% na ELISA. Não foram observadas diferenças estatisticamente significativas entre os grupos 1 e 2 em todas as variáveis, com exceção de "contato com gatos", para o grupo 1, e de "ingestão de leite cru", para o grupo 2, que foram estatisticamente significativas. Os resultados sugerem que a manipulação de carcaças (cistos) não deve ter sido o único mecanismo de infecção envolvido na alta freqüência de sororeagentes na população estudada, embora possa ter contribuído na transmissão do parasita como fator de risco, assim como o contato com solo contaminado ou com gatos (oocistos).

The risk of Toxoplasma gondii infection in workers of a swine slaughterhouse in Palmas, Paraná, Brazil was compared to other individuals not exposed to these animals and its carcasses. The 174 serum samples were divided in two groups: group 1 - slaughterhouse workers (133), and group 2 - control (41). During blood sampling, workers answered an epidemiological questionnaire with the following variables: sex, age, contact with cats and soil, habit of eating raw or poorly cooked meat, sausages and raw milk. Serum samples were tested for IgG antibodies anti-T. gondii by enzyme-linked immunosorbent assay (ELISA) and indirect fluorescent antibody test (IFAT) and to the investigation of IgM antibodies anti-T. gondii by IFAT. All the 174 individuals examined were not IgM reacting. In the first group, 48.1% were IgG seroreacting by IFAT and 58.6% by ELISA. In the second group, 39.0% were IgG seroreacting in the IFAT and 51.2%. by ELISA. No statistically significant difference was found between groups 1 and 2 among the majority of the variables, except for the "contact with cats" in group 1 and "raw milk ingestion" in group 2. The results suggest that ingestion of cysts as a consequence of carcass manipulation probably was not the only mechanism involved in the high frequency of reactive serum in the studied population, although it should have contributed in the transmission of the parasite as risk factor, as well as contact with contamined soil or with cats (oocysts).

Visando avaliar a participação da carne bovina na epidemiologia da toxoplasmose, foram coletadas amostras de soro de 348 bovinos e de 64 funcionários em quatro matadouros da microrregião de Pato Branco, Estado do Paraná, Brasil. Os soros dos bovinos foram avaliados pela reação de imunofluorescência indireta (RIFI) e apresentaram soropositividade (IgG) em 41,4% das amostras examinadas. O título mais freqüentemente encontrado foi o de 64 (92,4%). Nenhum animal apresentou título superior a 1024. Não foram encontradas diferenças significativas em relação ao sexo, idade e procedência dos animais. Os soros humanos, avaliados pelos testes de RIFI e de ELISA (imunoensaio enzimático) para IgG, apresentaram 67,2% e 84,4% de positividade, respectivamente. Não foram encontradas diferenças significativas com relação às variáveis idade, sexo, tempo de serviço no abatedouro, contato com gatos e hábito de ingerir carne crua ou mal cozida. Os resultados sugerem que a carne bovina pode desempenhar importante papel na manutenção da toxoplasmose na região.

In order to determine the role of bovine meat in the epidemiology of toxoplasmosis, 348 serum samples were collected from cattle and 64 serum samples from slaughterhouse workers at four plants in the region of Pato Branco, Paraná state, southern Brazil. Cattle sera were examined by the indirect fluorescent antibody test (IFAT), showing an IgG-seropositivity of 41.4%. The most frequently titer found was 64 (92.4%). Maximum titer found was 1024. There was no significant difference concerning sex, age and origin of positive sera. Human sera were tested by the IFAT and enzyme-linked immunosorbent assay (ELISA), showing 67.2% and 84.4% of IgG-seropositivity, respectively. No significant difference was observed between prevalences and age, sex, duration of employment at slaughterhouse, contact with cats and eating raw or undercooked meat. Results suggest that bovine meat may be a possible source of toxoplasmosis in the region.
