Abstract Ostrea chilensis (Küster, 1844), the flat oyster, is native to Chile and New Zealand. In Chile, it occurs in a few natural beds, from the northern part of Chiloé Island (41 ºS) to the Guaitecas Archipelago (45 ºS). This bivalve is slow growing, broods its young, and has very limited dispersal potential. The Ostrea chilensis fishery has been over-exploited for a number of decades such that in some locations oysters no longer exist. The aim of this study was to study the genetic diversity of the Chilean flat oyster along its natural distribution to quantify the possible impact of the dredge fishery on wild populations. The genetic structure and diversity of Ostrea chilensis from six natural beds with different histories of fishing activity were estimated. Based on mitochondrial (Cytb) and nuclear (ITS1) DNA sequence variation, our results provide evidence that genetic diversity is different among populations with recent history of wild dredge fishery efforts. We discuss the possible causes of these results. Ultimately, such new information may be used to develop and apply new management measures to promote the sustainable use of this valuable marine resource.

ABSTRACT. Given the hybridization ability between species Mytilus edulis platensis andM. galloprovincialis, this research proposed to assess the genetic variability of the hybrid offspring in relation to pure species, using 8 loci microsatellites. The study found differences between hybrid offspring comparing to pure species. The hybrid offspring had the highest observed mean heterozygosity values (Ho = 0,63 ± 0,28) and low values of inbreeding, which tend to significant deviations to negative values of (Fis), demonstrating excess of heterozygote in hybrids comparing with pure species, which displayed significant deviations towards positive values of Fis. The principal component analysis shows three distinct groups among the samples analyzed, identifying the hybrid offspring as an intermediate group between pure species.

RESUMEN. Dada la capacidad de hibridación entre Mytilus edulis platensis y M. galloprovincialis, se planteó evaluar la variabilidad genética de la descendencia híbrida en relación a las especies puras a partir de 8 loci de microsatélites. Se observaron diferencias genéticas entre la descendencia híbrida en comparación a las especies puras, siendo la descendencia híbrida la que presenta los mayores valores de heterocigosidad observada promedio (Ho = 0,63 ± 0,28), con valores de endogamia que tienden mayoritariamente hacia valores de Fis negativos, evidenciando un exceso de heterocigotos en los híbridos en comparación a los individuos puros, quienes presentaron desvíos significativos hacia valores de Fis positivos. El análisis de componentes principales muestra tres grupos claramente diferenciados entre las muestras analizadas, identificando a la descendencia híbrida como un grupo intermedio entre las especies puras.

The sperm ultrastructure has been used to solve several systematic and phylogenetic problems in marine invertebrates. The sperm ultrastructure of the Chilean mussel Mytilus chilensis and Mytilus galloprovincialis corresponds to the ect-aquasperm type. Sperm from both taxa measured 55-60 μm between head (acrosome + nucleus), midpiece (only 5 mitochondria) and the flagellum which in its end piece has a smaller diameter tail. The differences between both taxa are clearly shown, in the structure of the acrosome and nucleus. Therefore, according to our results and those reported in the literature, we indicate that Chilean native mussel sperm is different from other species of the Mytilus complex (M. trossulus, M. galloprovincialis and M. edulis). These differences in sperm ultrastructure found in M. chilensis, are another trait that can be used to validate the taxonomic status of the species. Differences in sperm morphology are related with reproductive isolation, and probably will be useful to understand future data on speciation. Finally, we discussed the finding that Mytilus galloprovincialis sperm from Chile have an acrosome notoriously smaller than those reported for specimens from Europe and Africa, though they have a great similarity with specimens from Japan, as reported in the literature.

La ultraestructura de los espermatozoides se ha utilizado para resolver varios problemas sistemáticos y filogenéticos en invertebrados marinos. La ultraestructura de los espermatozoides del mejillón chileno Mytilus chilensis y Mytilus galloprovincialis corresponde al tipo acuesperma. La medida de los espermatozoides de ambos taxa fue de 55-60 μm entre la cabeza (acrosoma + núcleo), sector medio (solo 5 mitocondrias) y el flagelo, que en su pieza final tiene una cola de menor diámetro. La diferencia entre ambos taxa se mostró claramente en la estructura del acrosoma y el núcleo. Por lo tanto, de acuerdo a nuestros resultados y a los de la literatura, se indica que el mejillón chileno es diferente a los del complejo Mytilus (M. trossulus, M. galloprovincialis y M. edulis). Estas diferencias en la ultraestructura de los espermatozoides que se encontraron en M. chilensis, son otro rasgo que se puede utilizar para validar el estatus taxonómico de las especies. Las diferencias en la morfología de los espermatozoides están relacionadas con el aislamiento reproductivo y probablemente serán de ayuda para entender futuros datos de especiación. Finalmente, se discute que los espermatozoides de Mytilus galloprovincialis que se encuentran en Chile tienen un acrosoma notablemente menor a los reportados en especímenes de Europa y África, aunque tienen una gran similitud con los especímenes de Japón, como se reporta en la literatura.

Adult specimens of M. galloprovincialis from Concepción Bay and M. chilensis from Yaldad Bay, Chile, were transferred to the laboratory to produce crosses of "pure" and "hybrid" species in order to evaluate early larval development and growth. These variables are important for understanding the dynamics of these two mussel species in this potential hybrid zone where they occur sympatrically. The study showed that fertilization occurred in all crosses and significant differences were not detected between pure lines and hybrids in terms of the percentage of eggs that developed into larvae. Hybrid larvae and spat from both reciprocal crosses grew significantly more than those from pure lines, although valve length values were within the ranges reported in the literature.

Ejemplares adultos de M. galloprovincialis de la bahía de Concepción y de M. chilensis de la bahía de Yaldad, Chile, se trasladaron al laboratorio para realizar cruzamientos puros de cada especie e híbridos, para evaluar el desarrollo larval temprano y su crecimiento. Estas variables son importantes para entender la dinámica de estas dos especies de mitílidos en esta potencial zona híbrida donde se encuentran en forma simpátrica. El estudio mostró que la fertilización ocurrió en todos los cruzamientos y no se detectó diferencias significativas entre líneas puras e híbridas en el porcentaje de huevos que se desarrollaron a larvas. Las larvas y juveniles híbridos de ambos cruzamientos recíprocos crecieron significativamente más que las larvas de los cruzamientos de especies puras, aunque los valores de longitud de la valva están dentro de los rangos reportados en la bibliografía.

A qualitative and quantitative analysis was carried out of the gonadal cycle of the bivalve Mytilus chilensis from Chaihuín and Yal bay, southern Chile, between October 2007 and June 2008. Four gametogenic stages were determined using histological analysis of the gonads, and quantitative estimates were made of the Gametic Volume Fraction (VFG), percentage of inter follicular connective tissue, and the Gonadosomatic Index (IG). The quantitative analysis (VFG) was the best indicator of spawning. Two spawning events, one in October and one in March, were observed simultaneously in both sexes of mussels from Chaihuín. However, for specimens from bahía Yal, four spawning events were registered, principally from March to June (autumn), when the water temperature decreased. The relationship between the IG and the gametogenic stages was very low, as was that between the IG and the percentage of mature oocytes. Therefore, the IG is not a good indicator of spawning in this species. A re-evaluation of the ban period established for Mytilus chilensis (1 November to 31 December) is suggested since most individuals from the populations studied mature mainly in October. At both sites, the percentage of connective tissue for the analyzed mussel individuals ranged between 15 and 70% of gonadal coverage. The results obtained in the present study showed differences in the reproductive cycles of Mytilus chilensis between the sites sampled. These differences could be due to environmental differences (e.g. temperature) caused by the latitudinal gradient.

Se analizó de forma cualitativa y cuantitativa el ciclo gonadal del bivalvo Mytilus chilensis en las localidades de Chaihuín y bahía Yal, sur de Chile, entre octubre 2007 y junio 2008. Por medio de análisis histológico gonadal se determinaron cuatro estadios gametogénicos y a su vez se estimó en forma cuantitativa, el Volumen de la Fracción Gamética (VFG), el porcentaje de tejido interfolicular y el índice gonadal. El análisis cuantitativo (VFG) fue el mejor indicador para determinar los desoves. En los ejemplares de Chaihuín se observaron dos eventos de emisión gamética en forma simultánea en ambos sexos, que ocurrieron en octubre y marzo. Sin embargo, en los ejemplares de bahía Yal se registraron cuatro desoves, principalmente de marzo a junio (otoño), cuando la temperatura del agua disminuyó. Se determinó una escasa relación entre el Índice Gonadosomático (IG) y los estadios gametogénicos, al igual que entre el IG y el porcentaje de ovocitos maduros, por ende el IG no sería un indicador apropiado para los desoves en esta especie. Se sugiere la revisión del periodo de veda de Mytilus chilensis (1 noviembre a 31 diciembre), ya que la mayor parte de los individuos de las poblaciones estudiadas, maduran principalmente en octubre. En ambas localidades, el porcentaje de tejido conjuntivo de los especímenes estudiados fluctúo entre 15 y 70% de cobertura gonadal. Los resultados obtenidos mostraron diferencias en los ciclos reproductivos de Mytilus chilensis entre las localidades analizadas, las que se podrían atribuir a diferencias ambientales (e.g. temperatura) causadas por el gradiente latitudinal.

A qualitative and quantitative study of the gametogenic cycle from natural populations of Perumytilus purpuratus at the localities of Taltal and Huasco was carried out between October 2007 and October 2008. By means of histological analysis a gonadal maturation table was constructed according to the microscopic characteristics. Also the Gametic Volume Fraction (GVF) (% of previtelogenic oocytes, vitelogenic oocytes and mature oocytes), percentaje of interfolicular connective tissue and Gonadic Index (GI) were determined. According to the qualitative analysis, P. purpuratus has a semiannual reproductive cycle starting in January with spawning mainly during spring-summer, even do some winter spawning were detected. However, the quantitative analysis of mature oocytes percentajes found that at Taltal population four spawnings were detected (December, March, July and October) while in Huasco population three spawnings were detected (December, February and September). The relationship between GI with the gametogenic stages and the mature oocyte percentajes were significant at the localities studied. Also significant differences of the condition index between males and females were observed. Along the latitudinal gradient the reproductive cycle of P. purpuratus showed variations according to the timing and synchrony.

Se estudió el ciclo gametogénico de forma cualitativa y cuantitativa en poblaciones naturales de Perumytilus purpuratus provenientes de las localidades de Taltal y Huasco, Chile, durante el período entre octubre de 2007 y octubre de 2008. A través del análisis histológico se construyó una tabla de madurez gonadal de acuerdo a las características microscópicas. También se determinó el volumen de la fracción gamética (VFG) (% de ovocitos previtelogénicos, ovocitos vitelogénicos y ovocitos maduros), porcentaje de tejido interfolicular e índice gonadal (IG). De acuerdo al análisis cualitativo P. purpuratus tiene un ciclo reproductivo semi-anual, que comienza en enero con desoves principalmente en primavera-verano, aunque también se detectaron desoves invernales. Sin embargo, el análisis cuantitativo de porcentajes de ovocitos maduros encontró que en Taltal se produjeron cuatro desoves (diciembre, marzo, julio y octubre) mientras que en Huasco tres (diciembre, febrero y septiembre). La relación del IG con los estados gametogénicos y el porcentaje de ovocitos maduros es significativa en las localidades estudiadas. También se observaron diferencias significativas del índice gonadal entre machos y hembras. En un gradiente latitudinal, el ciclo reproductivo de P. purpuratus mostró variaciones de acuerdo al periodo y a la sincronía.

Natural populations of Ostrea chilensis have suffered strong declines during the last two decades due to overfishing that forced in some locations to carry out restocking and culture with juvenile oysters from other local populations. It has been proposed that this type of intervention may have affected the levels of genetic variation among natural populations. Here we analyzed the genetic variation in two natural populations and in three populations affected by restocking of O. chilensis, sampled along its whole natural distribution, by means of Random Amplified Polymorphic DNA (RAPD) analyses. The values of Nei's genetic distance did not reveal significant differences among populations (0.0713 and 0.1549). The Mantel test using 5000 randomizations, showed a significant correlation between genetic and geographic distance. Melinka, the most geographically distant population, was the most genetically differentiated. The levels of gene flow (Nm = 1.587) are strong enough to prevent differentiation due to genetic drift. With these results, the genetic-population structure of O. chilensis, can be characterized a panmictic population, in which, the level of gene flow prevents significant genetic differentiations among populations. However, the population-genetic structure of this species does not depend exclusively on its breeding strategy, since other factors such as: oceanographic and human mediated gene flow probably exert a large influence to the genetic structure of the studied populations.

Las poblaciones naturales de Ostrea chilensis han sufrido fuertes disminuciones debido a la sobrepesca, lo cual ha forzado en algunos sectores a llevar a cabo repoblamiento y cultivo con ostras juveniles provenientes de otras localidades. Se ha planteado que este tipo de intervención podría haber afectado los niveles de variación genética en las poblaciones naturales. Se analizó la variabilidad genética, a través del análisis con ADN Polimórfico Amplificado Aleatoriamente (RAPD), en dos poblaciones naturales y tres poblaciones afectadas por el repoblamiento de O. chilensis, muestreadas a lo largo de su distribución natural. Los valores de distancia genética de Nei no revelaron diferencias significativas entre poblaciones (0,0713-0,1549). La prueba de Mantel con 5000 permutaciones evidenció una correlación significativa entre la distancia geográfica y la genética. La población de Melinka, situada a mayor distancia geográfica mostró la mayor diferenciación genética. Los niveles de flujo génico (Nm = 1,587) son suficientemente fuertes para impedir diferencias genéticas en las poblaciones debido a la deriva génica. Con estos resultados se caracterizó la estructura genético-poblacional de O. chilensis como una población panmíctica, en la cual el flujo génico no permite una significativa diferenciación genética interpoblacional. Sin embargo, la estructura genético-poblacional de esta especie no depende exclusivamente de su estrategia reproductiva, sino que existen otros factores oceanográficos y antropogénicos, que probablemente estarían influenciando la conformación genética de las poblaciones estudiadas.

Genetic differentiation in the Chilean blue mussel Mytilus chilensis (Hupé 1854) was investigated based on the variation in the allozyme frequencies of Pgm, Gpi, Icd, Me, Gsr, Lap and Pep in eight samples collected along 1800 km from Arauco (VIII Region) to Punta Arenas (XII Region). Despite the large geographic separations, values of Neis unbiased genetic distance, D (0.004-0.048) and standardised genetic variation among populations, Fst (0.011-0.055) were small. The levels of gene flow (Nm = 8) found in this study prevent the effect of differentiation among populations by genetic drift. This findings indicate that its long-lived planktotrophic larvae provides this species with considerable dispersal ability throughout its range which is favoured by the ocean currents along the chilean coast. In terms of management of the M. chilensis fishery, the results provide no evidence for discrete stocks, with the possible exception of the Punta Arenas population. Considering the intensive aquaculture activities with this species the present study provide preliminary data which can be used as a baseline for further characterization and /or monitoring these mussel populations.

The oyster parental stock was taken from the 1998 natural spatfall from the wild population of Ostrea chilensis at the Quempillen River Estuary, located at Chiloe Island, southern Chile. From a cohort of 34 months of age 5232 oysters were monitored for the traits shell length and live weight. Selection was carried out applying a selection intensity of +1.7 for the trait live weight, using a control group. The conditioning was carried out in 5 tanks of 120 L, 3 of them containing high selected lines and other 2 control lines of oysters. A highly significant correlation (0.76, p<0.001, N=5232) was found between the traits live weight and shell length. Spat from the 3 selected subgroups and 2 control groups were obtained. Plastic collectors with the spat attached were placed in a common rearing tank. Shell length was monitored after 6 and 16 weeks of age in 200 juveniles of each selected and control lines. The ANDEVA results show a significant difference (p<0.05) between the shell length of the selected and control groups at 6 and 16 weeks of age. Also, a significant response to selection was obtained even when selection was applied for the trait live weight at 34 m-old. These results confirm those obtained in an earlier study using the same natural oyster population from Quempillén

La población base de Ostrea chilensis utilizada en el presente trabajo corresponde a una cohorte obtenida del estuario del río Quempillén, isla de Chiloé, sur de Chile. A los 34 meses de edad y en base a 5232 ostras de la cohorte tomadas al azar, se realizó una curva de distribución para los caracteres peso vivo y longitud de la valva. Como criterio de selección se aplicó una intensidad de selección de +1,7 para el peso vivo, manteniéndose un grupo control. Cada grupo, control y seleccionado (N=400 y N=600 respectivamente), se dividió al azar en subgrupos de 200 ostras, 3 de los cuales contenían líneas seleccionadas y los otros 2, ostras como grupo control. El acondicionamiento se realizó en 5 estanques de 120 L cada uno. El carácter peso vivo mostró tener una alta correlación fenotípica con el carácter longitud de la valva con un valor de 0,76 (N=5.232, p<0,001). Se obtuvieron juveniles en los 5 sub-grupos. Los colectores plásticos con los juveniles adheridos marcados se mantuvieron en crecimiento en un estanque común. Se realizaron dos monitoreos de crecimiento de longitud de valva en 200 juveniles tomados al azar en cada uno de los subgrupos seleccionados. El ANDEVA indica una diferencia significativa (p<0,05) en la longitud de la valva entre juveniles del grupo control y grupo seleccionado, a las 6 y 16 semanas de edad. Ello, aun cuando la selección se basó en el carácter peso vivo a los 34 meses de edad (talla comercial). Estos resultados confirman los obtenidos en un estudio previo utilizando la misma población natural de ostras de Quempillén
