Abstract Introduction Perianal fistula is a common colorectal disease which is caused mainly by cryptoglandular disease. Although most cases are treated successfully by surgery, management of complex perianal fistulas (CPAF) remains a challenge with limited results in recurrence and sometimes associated with fecal incontinence. The CPAF treatment with autologous adipose-derived mesenchymal stem cells (ASCs) had become a research hotspot. The technique started to be used in the treatment of Crohn's disease (CD) fistulas, where the studies showed safe and goods result from the procedure. Cultured ASCs have been used but this approach requires the preceding collection of adipose tissue, time for isolation of ASCs and subsequent in vitro expansion, need for laboratory facilities, and expertise in cell culturing. These factors have been getting over by using the commercially available alternative, allogenic ASCs. Treatment with allogeneic ASCs has shown good results in patients with CD fistulas, however with the disadvantage of being expensive. Objective To show that the injection with freshly collected adipose tissue is an alternative to treatment with autologous or allogenic ASCs with several advantages. Methods: In this case report, we show our first experience in the treatment of CPAF with the application of collected adipose tissue in a tertiary referral hospital from Belo Horizonte, Brazil. RESUMO Contexto: Aproximadamente 71 milhões de pessoas estão infectadas pelo vírus da hepatite C em todo o mundo. Um número significativo desses indivíduos desenvolverá cirrose hepática e/ou carcinoma hepatocelular. Além do fígado, há evidências científicas que associam doenças cardiovasculares e hepatite C crônica; no entanto, os mecanismos biológicos implicados na ocorrência dessas condições ainda não foram completamente esclarecidos. Objetivo: Avaliar a associação entre histologia hepática, comorbidades clínicas e perfil lipídico em pacientes com hepatite C crônica. Investigar associações entre histologia hepática e parâmetros demográficos, nutricionais, bioquímicos e virológicos. Métodos: Oitenta e cinco pacientes com hepatite C crônica foram prospectivamente submetidos à biópsia hepática. Biópsias hepáticas foram obtidas de cada paciente por via percutânea com agulha de Menghini. A fibrose foi avaliada de acordo com o sistema de pontuação METAVIR, como segue: F0, sem fibrose; F1, expansão portal fibrosa; F2, alargamento portal fibroso com poucos septos; F3, fibrose em ponte com distorção arquitetônica; e F4, cirrose hepática. A atividade foi classificada com base no grau de infiltração de linfócitos e necrose de hepatócitos, de A0 a A3. O diagnóstico da doença hepática foi baseado em métodos clínicos, bioquímicos, histológicos e radiológicos. Os dados foram analisados por modelos de regressão logística. Resultados: Neste estudo transversal, realizado em um ambulatório do hospital universitário, foram incluídos 85 pacientes que tinham média de idade de 57,2±10,7 anos, sendo 45 (52,9%) do sexo feminino. Havia 10 pacientes com cirrose. Os pacientes com METAVIR F3-F4 eram significativamente mais velhos (P=0,02) e tinham níveis mais elevados de ALT (P=0,0006), AST (P<0,0001), γ-GT (P=0,03) e bilirrubina (P=0,001) e, maior tempo de protrombina do que pacientes com escore F0-F2. Os níveis de albumina (P=0,01) foram significativamente mais baixos naqueles classificados como METAVIR F3-F4. Idade (OR=1,09; IC95%=1,02-1,16; P=0,02), esteatose (OR=4,03; IC95%=1,05-15,45; P=0,04) e HDL-C <60 mg/dL (OR=7,67; 95%IC=1,71-34,49; P=0,008) foram independentemente associados à fibrose. ABSTRACT Background: Approximately 71 million people are chronically infected with hepatitis C virus (HCV) worldwide. A significant number of these individuals will develop liver cirrhosis and/or hepatocellular carcinoma. Beyond the liver, there is a sizeable body of scientific evidence linking cardiovascular disease and chronic hepatitis C (CHC); however, the biological mechanisms behind the concurrence of these conditions have not been completely clarified yet. Objective: To evaluate associations between hepatic histology, clinical comorbidities and lipid profile in patients with CHC. To investigate associations between liver histology and demographic, nutritional, biochemical and virological parameters. Methods: Eight-five patients with CHC prospectively underwent hepatic biopsy. Liver fragments were obtained from each patient by percutaneous route using a Menghini needle. Fibrosis was evaluated according to the METAVIR scoring system, as follows: F0, no fibrosis; F1, fibrous portal expansion; F2, fibrous portal widening with few septa; F3, bridging fibrosis with architectural distortion; and F4, liver cirrhosis. The activity was classified based on the degree of lymphocyte infiltration and hepatocyte necrosis, from A0 to A3. The diagnosis of liver disease was based on clinical, biochemical, histological, and radiological methods. The data were analyzed by logistic regression models. Results: This cross-sectional study included 85 outpatients followed at the tertiary care ambulatory centre with a mean age of 57.2±10.7 years and 45 (52.9%) were females. There were 10 patients with cirrhosis. Patients with a METAVIR F3-F4 were significantly older (P=0.02) and had higher levels of ALT (P=0.0006), AST (P<0.0001), γ-GT (P=0.03) and bilirubin (P=0.001) and higher prothrombin time than patients with F0-F2 score. Albumin levels (P=0.01) were significantly lower in METAVIR F3-F4. Age (OR=1.09; 95%CI=1.02-1.16; P=0.02), steatosis (OR=4.03; 95%CI=1.05-15.45; P=0.04) and high-density lipoprotein cholesterol (HDL-C) <60 mg/dL (OR=7.67; 95%CI=1.71-34.49; P=0.008) were independently associated with fibrosis. Hypertension (OR=6.36; 95%CI=1.31-30.85; P=0.02) and HDL-C <60 mg/dL (OR=9.85; 95%CI=2.35-41.39; P=0.002) were independently associated with necroinflammatory activity. RESUMO O café é uma das principais commodities do agronegócio mundial e de grande relevância econômica e social para o Brasil. A falta de mão de obra e seu alto custo são fatores que preocupam os cafeicultores, principalmente na colheita do café conilon, feita manualmente. O objetivo deste trabalho foi avaliar a eficiência de uma colhedora automotriz em diferentes condições de ajuste da máquina e condução da lavoura de café conilon e medir sua influência no custo de colheita comparada a colheita manual. Foram avaliados os fatores velocidade de colheita (800 a 1600 m h-1), rotação do cilindro vibrador de varetas (1,0 e 1,5 RPM), número de ramos ortotrópicos (um, dois e três) e plantas com e sem os ramos plagiotrópicos no terço inferior. O aumento da velocidade de colheita reduziu as eficiências de derriça e de colheita e a desfolha. O aumento de um para três ramos ortotrópicos por planta aumentou as eficiências de colheita e de derriça, a perda de chão e a desfolha e reduziu a carga pendente. O manejo sem os ramos plagiotrópicos apresentou maior eficiência de colheita, menor perda de chão e menor desfolha. As velocidades de colheita de 800 a 1600 m h-1 reduziram os custos totais e unitários em até 62% em relação à colheita manual. O aumento da eficiência de colheita acima de 70% reduziu os custos de colheita em até 79% em relação à colheita manual.

ABSTRACT Coffee is one of the main commodities of global agribusiness and of outstanding economic and social relevance for Brazil. The lack of labor and its high cost are factors that worry coffee producers, mainly during the conilon coffee harvesting, which is performed manually. This study aimed to evaluate the efficiency of a self-propelled harvester under different conditions of machine adjustment and conduction of the Coffea canephora crop and measure its influence on the cost of harvesting compared to manual harvesting. Harvesting speed (800 to 1600 m h-1), rotation of the vibrating rod cylinder (1.0 and 1.5 RPM), number of orthotropic branches (one, two, and three), and plants with and without plagiotropic branches in the lower third were assessed. The increase in harvesting speed reduced the efficiencies of stripping and harvesting and defoliation. Increasing from one to three orthotropic branches per plant increased harvesting and stripping efficiencies, fruit loss on the ground, defoliation, and reduced pending load. The management without plagiotropic branches showed higher harvesting efficiency, lower loss on the ground, and lower defoliation. Harvesting speeds from 800 to 1600 m h-1 reduced the total and unit costs up to 62% compared to manual harvesting. Increasing harvesting efficiency above 70% has reduced harvesting costs by up to 79% compared to manual harvesting.

Este estudo identificou aglomerados espaciais de diabetes mellitus tipo 2 entre participantes do Estudo Longitudinal de Saúde do Adulto no Brasil (ELSA-Brasil) em duas cidades e verificou características socioeconômicas ambientais individuais e de vizinhança associadas aos aglomerados espaciais. Se trata de um estudo transversal com 4.335 participantes. Diabetes mellitus tipo 2 foi definido com base em glicemia de jejum ≥ 126mg/dL (7,0mmol/L); teste oral de tolerância à glicose ≥ 200mg/dL (11,1mmol/L); hemoglobina glicada ≥ 6,5% (48mmol/L); uso de drogas antidiabéticas; ou pelo autodiagnóstico médico de diabetes mellitus tipo 2. As características socioeconômicas do bairro foram obtidas a partir do censo brasileiro de 2011. A análise dos dados espaciais foi realizada pelo método SaTScan para detectar os aglomerados espaciais. Os modelos de regressão logística foram ajustados para estimar a magnitude das associações. Um total de 336 e 343 participantes apresentaram diabetes mellitus tipo 2 em Belo Horizonte, Minas Gerais (13,5%) e Salvador, Bahia (18,5%), respectivamente. Foram identificadas duas áreas de aglomerados com alta probabilidade de diabetes mellitus tipo 2 em Belo Horizonte e Salvador. Em ambas as cidades, os participantes residentes nos aglomerados com alta taxa de diabetes mellitus tipo 2 tinham maior probabilidade de relatar cor de pele parda ou preta, baixa escolaridade e ocupação de trabalho manual; essas áreas também foram consideradas de baixa renda. Por outro lado, os participantes do aglomerado com baixa taxa de diabetes mellitus tipo 2 de Salvador tinham menor probabilidade de serem negros e maior probabilidade de terem diploma universitário, além de morarem em áreas de alta renda. Características socioeconômicas individuais e de vizinhança mais vulneráveis estavam associadas à residência em aglomerados de maior ocorrência de diabetes mellitus tipo 2, enquanto o oposto foi observado para perfis contextuais melhores.

El análisis de datos espaciales se realizó utilizando el método SaTScan para detectar grupos espaciales. Los modelos de regresión logística se ajustaron para estimar la magnitud de las asociaciones. Se identificaron dos áreas de grupos con alta probabilidad de diabetes mellitus tipo 2 en Belo Horizonte y Salvador. En ambas ciudades, los participantes que residían en las áreas del grupo con una alta tasa de diabetes mellitus tipo 2 tenían más probabilidades de informar el color de piel pardo o negro, la baja educación y la ocupación del trabajo manual; estas áreas también se consideraron de bajos ingresos. Por el contrario, los participantes en el área del grupo con baja tasa de diabetes mellitus tipo 2 de Salvador tenían menos probabilidades de ser negros y más probabilidades de tener un título universitario, además de vivir en áreas de altos ingresos. This study identified spatial clusters of type 2 diabetes mellitus among participants of the Brazilian Longitudinal Study of Adult Health (ELSA-Brasil) residing in two cities and verified individual and neighborhood socioeconomic environmental characteristics associated with the spatial clusters. A cross-sectional study was conducted with 4,335 participants. Type 2 diabetes mellitus was defined as fasting blood glucose ≥ 126mg/dL (7.0mmol/L), oral glucose tolerance test ≥ 200mg/dL (11.1mmol/L), or glycated hemoglobin ≥ 6.5% (48mmol/L); by antidiabetic drug use; or by the self-reported medical diagnosis of type 2 diabetes mellitus. Neighborhood socioeconomic characteristics were obtained from the 2011 Brazilian census. A spatial data analysis was conducted with the SaTScan method to detect spatial clusters. Logistic regression models were fitted to estimate the magnitude of associations. Two cluster areas showing a high chance of type 2 diabetes mellitus were identified in Belo Horizonte and Salvador. In both cities, participants living in the high type 2 diabetes mellitus cluster area were more likely to be mixed-race or black and have a low schooling level and manual work; these were also considered low-income areas. On the other hand, participants in the low type 2 diabetes mellitus cluster area of Salvador were less likely to be black and have low schooling level (university degree) and live in a low-income area. More vulnerable individual and neighborhood socioeconomic characteristics were associated with living in clusters of higher type 2 diabetes mellitus occurrence , whereas better contextual profiles were associated with clusters of lower prevalence.

Resumo Objetiva-se, neste artigo, realizar uma análise da formação e das transformações do patrimônio da Câmara Municipal de Desterro ao longo do século XIX. A partir da utilização de fontes primárias e secundárias, busca-se debater o cadastramento de ocupantes de 1817, o tombamento do patrimônio de 1823 e a dinâmica de posses e aforamentos realizada pela Câmara Municipal. À luz de debates sobre a historiografia da propriedade da terra no Brasil, identifica-se localmente o aumento do controle estatal sobre o rossio e a baixa influência da Lei de Terras durante o período estudado. Destaca-se, finalmente, a importância de ampliar o conhecimento acerca da alienação do patrimônio fundiário municipal, seus efeitos na configuração da cidade e na formação do patrimônio imobiliário privado.

Abstract The objective of this article is to carry out an analysis of the formation and transformations of the patrimony of Desterro’s City Council throughout the 19th century. From the use of primary and secondary sources, we sought to discuss the registration of occupants from 1817, the patrimony demarcation of 1823 and the dynamics of possessions and leases carried out by the City Council. In the light of debates on the historiography of land ownership in Brazil, we identified the increase in state control over the rossio and the low influence of the Land Law during the period studied. Finally, it is worth highlighting the importance of expanding knowledge about the alienation of municipal land patrimony, its effects on the configuration of the city and on the formation of private real estate patrimony.

Este estudio identificó grupos espaciales de diabetes mellitus tipo 2 entre los participantes del Estudio Longitudinal de Salud del Adulto en Brasil (ELSA-Brasil) en dos ciudades y verificó las características socioeconómicas ambientales individuales y de vecindario asociadas con los grupos espaciales. Se trata de un estudio transversal con 4.335 participantes. RESUMO Introdução: A sobreposição de modelos 3D é uma necessidade iminente. No entanto, os métodos atuais dependem da marcação de múltiplos pontos na maxila e na mandíbula, o que pode aumentar a incorporação de erros no processo de sobreposição. Objetivo: O objetivo desse estudo foi desenvolver um método para sobrepor modelos 3D das arcadas superior e inferior utilizando o software de engenharia Autodesk Inventor®, por meio da marcação de um único ponto em cada arcada, usando o sistema de coordenadas universal (UCS). Métodos: No total, 104 modelos STL das arcadas superior e inferior exportados da plataforma My iTero® foram selecionados retrospectivamente, onde T0 foi o período inicial e T1, o de refinamento (n=26 por grupo). As coordenadas X, Y e Z associadas a um único ponto em cada arcada foram inseridas nos modelos usando o software SlicerCMF®. Os modelos com os pontos UCS demarcados foram transferidos para o software Autodesk Inventor® para realizar a sobreposição e medir os movimentos dentários realizados durante o tratamento com Invisalign®. Os movimentos de expansão, intrusão e rotação foram analisados por dois examinadores. A análise estatística foi realizada usando os coeficientes de correlação intra-classe (ICC), fórmula de Dahlberg e teste t (p<0,05). Resultados: Foi desenvolvido um método confiável de sobreposição de modelos digitais 3D usando um único ponto UCS na maxila e mandíbula. O ICC apresentou excelente correlação intra e inter-avaliadores (ICC>0,90). Não foi encontrado erro sistemático nas medidas lineares e angulares (<1mm e <1,5°, respectivamente). ABSTRACT Introduction: Superposing 3D models is an imminent need. However, current methods rely on marking multiple points on the maxilla and mandible, which could increase point marking and overlapping errors. Objective: This study aimed at developing a method for superimposing 3D models of the maxillary and mandibular arches with Autodesk Inventor® engineering software, using a single universal coordinate system (UCS) point superposition. Methods: A total of 104 STL (stereolithography) models of the maxillary and mandibular arches exported from My iTero® platform were retrospectively selected, in which T0 and T1 were the initial and refinement periods, respectively (n=26 per group). The X, Y, and Z coordinates associated with a single point in each arch were inserted into the models with SlicerCMF® software for model orientation. The arch models with UCS registration were transferred to Autodesk Inventor® for superimposition and to measure tooth movements performed during Invisalign® treatment. Arch expansion, intrusion and rotation were analyzed by two examiners. The statistics were performed using intraclass correlation coefficients (ICC), Dahlberg’s formula, and t-test (p<0.05). Results: A reliable method of superimposing 3D digital models using a single UCS point in the maxilla and mandible was developed. ICC showed excellent intra- and inter-examiner correlation (ICC>0.90). A systematic error was not found concerning linear and angular measurements (<1mm and <1.5°, respectively). Crotalaria ochroleuca (Fabaceae) is rich in bioactive compounds used for nematode control. Transformed root culture allows the production of a large amount of hairy root biomass, conditioned by auxin biosynthesis-related genes integrated via the process of Agrobacterium rhizogenes infection, which is known to cause a hairy root phenotype. We transformed A. rhizogenes to increase the biomass of hairy roots and optimized the process of production of bioactive compounds. The transformed nature of hairy roots was confirmed by polymerase chain reaction, which confers kanamycin resistance. The transformed roots were cultured in full-strength in liquid media to realize the growth of hairy roots and the production of bioactive compounds. The transformed roots were grown in a culture medium supplemented with elicitors to produce total phenols, and flavonoids. Chromatographic analysis of transformed roots revealed the presence of flavonoids apigenin-6,8-C-diglucoside and luteolin-6-C-glucoside. The results were obtained by conducting physiological and biochemical studies with the Flavonoids and studying the pathways that led to the production of large amounts of bioactive compounds from the hairy roots of C. ochroleuca. Resumo As bibliotecas públicas são espaços sociais e políticos destinados ao atendimento de todos da comunidade, assim elas ancoram-se no discurso inclusivo e democrático do acesso à informação, serviços e produtos. Contudo, há ainda um caminho a ser percorrido em direção a diversidade e a multiculturalidade em seus espaços, a começar pelos acervos. Discutiremos neste texto, a partir da literatura da área da Biblioteconomia e Ciência da Informação, a relação entre as bibliotecas públicas e a formação e o desenvolvimento de coleções, tendo como foco a literatura afro-brasileira. Consideramos o papel social das bibliotecas públicas e de seus profissionais, os bibliotecários, como agentes de mudanças, ressaltando a importância de ambos, com vistas à construção de uma sociedade justa e democrática. Compreendemos que esses profissionais devem conscientizar-se da relevância de formar e desenvolver acervos que propiciem o acesso à leitura de livros de autoras e autores de literatura afro-brasileira, visando tanto o fortalecimento das identidades dos sujeitos quanto do pensamento afrocentrado nas comunidades. Em suma, bibliotecas mais democráticas requerem práticas de decolonização dos acervos por meio de ações e políticas de reparação das coleções.

Abstract Public libraries are social and political spaces intended to serve all members of the community, and are thus anchored in the discourse of inclusion and democracy in the access to information, products and services. However, there is still a long way to go to achieve diversity and multiculturalism in its spaces, starting with collections. Based on literature from Library Science and Information Science, this article discusses the relationship between public libraries and the construction and development of collections, with a focus on Afro-Brazilian literature. The social role of public libraries is considered, as well as the role of their professionals, librarians, who are seen as agents of change, emphasizing the importance of both, with a view to building a just and democratic society. As such, these professionals should be aware of the relevance of forming and developing collections that provide ready access to books and especially to books by authors of Afro-Brazilian literature, aiming at strengthening both the identity of subjects as well as the Afro-centered thinking in the communities. In short, more democratic libraries require practices of decolonization of their collections through actions and policies which repair these collections.

Atualmente, a DCJ não possui cura e muitos pacientes morrem no primeiro ano de doença, mas alguns medicamentos vêm sendo estudados como opções no manejo desta condição. Objetivo Avaliar a eficácia dos tratamentos farmacológicos oferecidos aos pacientes com DCJ no aumento de sobrevida e na redução da deterioração cognitiva. Métodos Foi realizada uma revisão sistemática da literatura utilizando 4 revisores independentes e 1 extra para resolver divergências eventuais na busca e na análise de trabalhos indexados nas bases de dados MedLINE (via PubMed), SciELO e Lilacs. Os termos Medical Subjects Heading (MeSH) utilizados foram: prion diseases, creutzfeldt jakob disease, pharmacologic therapy, therapeutics, quinacrine, doxycycline, flupirtine e pentosan polysulfate, com os operadores booleanos AND e OR. Essa pesquisa incluiu ensaios clínicos controlados, não controlados e séries de casos, publicados a partir do ano 2000 no idioma inglês. Resultados Ao todo, foram encontrados 85 trabalhos através dos descritores utilizados. Ao final das análises de seleção, restaram 9 artigos, que foram analisados na íntegra individualmente. Conclusões Nenhuma das drogas avaliadas se mostrou significativamente eficaz no aumento da sobrevida dos pacientes com DCJ. A flupirtina parece ter um efeito benéfico na redução da deterioração cognitiva dos pacientes com DCJ. Entretanto, estudos adicionais são necessários para o estabelecimento de melhores evidências e opções terapêuticas para o manejo dos pacientes com DCJ. Abstract Background The Creutzfeldt-Jakob disease (CJD) is a spongiform encephalopathy that manifests as a rapidly progressive dementia syndrome. Currently, CJD has no cure, and many patients die within the first year, but some drugs are being studied as options for managing this condition. Objective To evaluate the effectiveness of pharmacological treatments offered to patients with CJD as a means to increase survival and reduce cognitive deterioration. Methods A systematic review of the literature was performed using 4 independent reviewers and 1 extra reviewer to resolve possible divergences in the search and analysis of papers indexed in MedLINE (PubMed), SciELO and Lilacs databases. The Medical Subject Heading (MeSH) terms used were: prion diseases, Creutzfeldt-Jakob disease, pharmacologic therapy, therapeutics, quinacrine, doxycycline, flupirtine, and pentosan polysulfate, with the Boolean operators AND and OR. Abstract Introduction: Urothelial carcinoma is a significant public health problem. Transitional cell carcinoma (TCC) is the most common subtype, accounting for approximately 90 % of all bladder cancers. Chemotherapeutic protocols have been studied, but some present high toxicity and low tolerability. Naringin is a polyphenolic compound found mainly in citrus fruits, which antitumor activity has been studied in several types of cancer. However, there is little information about naringin effects on bladder cancer. This study aimed to evaluate the antitumor potential of naringin in silico and in vitro using two bladder cancer cell lines Method: In silico analysis was carried out by PASS Online software. In vitro , the effects of naringin treatment (12.5 - 400 µM) were evaluated regarding its cytotoxicity, clonogenic survival, morphological alterations, cell cycle progression, migration, and mutagenicity Results: In silico analyses predicted antitumor activity through several mechanisms of action. In vitro results showed naringin presented cytotoxic effects, reduced the number of colonies, inhibited cell migration, and changed the morphology and cell cycle progression of the two cell lines evaluated. However, naringin did not present mutagenic effects. Conclusions: Naringin has antiproliferative activity and is a promising candidate for bladder cancer treatment.

Resumen Introducción: el carcinoma urotelial es un problema de salud pública importante. El carcinoma de células de transición es el subtipo más común y representa aproximadamente el 90 % de todos los cánceres de vejiga. Se han estudiado protocolos quimioterapéuticos, pero algunos presentan alta toxicidad y baja tolerabilidad. La naringina es un compuesto polifenólico que se encuentra principalmente en los cítricos, cuya actividad antitumoral se ha estudiado en varios tipos de cáncer. Sin embargo, hay poca información sobre los efectos de la naringina en el cáncer de vejiga. Este estudio tuvo como objetivo evaluar el potencial antitumoral de la naringina in silico e in vitro utilizando dos líneas celulares de cáncer de vejiga. Método: el análisis in silico se llevó a cabo mediante el software PASS Online. In vitro, se evaluaron los efectos del tratamiento con naringina (12,5 - 400 µM) en cuanto a su citotoxicidad, supervivencia clonogénica, alteraciones morfológicas, progresión del ciclo celular, migración y mutagenicidad. Resultados: los análisis in silico predijeron la actividad antitumoral a través de varios mecanismos de acción. Los resultados in vitro mostraron que la naringina presentó efectos citotóxicos, redujo el número de colonias, inhibió la migración celular y cambió la morfología y la progresión del ciclo celular de las dos líneas celulares evaluadas. Sin embargo, la naringina no presentó efectos mutagénicos. Conclusiones: la naringina tiene actividad antiproliferativa y es un candidato prometedor para el tratamiento del cáncer de vejiga.
