Abstract Studies on metabolic processes and seed germinability of native endangered species are essential for its conservation. The objective of this work was to evaluate the effect of accelerated aging on germination and on the proteomic and polyamine (PAs) profile in seeds of Cariniana legalis. The seeds were aged at 41, 43, 45 and 47ºC for 24, 48, 72 and 96 h, under 100 % relative humidity using a completely randomized factorial design. The data were submitted to analysis of variance followed by Tukey’s test. The temperature of 47ºC was efficient in simulating seed aging, with no germination after 72 h of incubation. At 41ºC, the germination did not change significantly until 96 h. The reduction in the concentration of total free PAs, mainly spermidine, and the down-accumulation of heat shock proteins in seeds aged at 47°C were associated with induction of seed aging. Furthermore, the down-accumulation of the aconitate hydratase protein and the up-accumulation of the alcohol dehydrogenase protein were associated with loss of seed germinability and rapid deterioration in seeds aged at 47°C. The aging of C. legalis seeds is related to alteration in the contents of PAs and accumulation of some proteins involved with the loss of germinability.

Resumo Estudos sobre processos metabólicos e germinabilidade de sementes de espécies nativas ameaçadas de extinção são essenciais para sua conservação. O objetivo deste trabalho foi avaliar o efeito do envelhecimento acelerado na germinação e no perfil proteômico e de poliamina (PAs) em sementes de Cariniana legalis. As sementes foram envelhecidas a 41, 43, 45 e 47°C por 24, 48, 72 e 96 h, sob 100% de umidade relativa, usando um delineamento fatorial inteiramente casualizado. Os dados foram submetidos à análise de variância seguida de teste Tukey. A temperatura de 47°C foi eficiente em simular o envelhecimento das sementes, que não germinaram após 72 h de incubação. Em 41°C, a germinação não mudou significativamente até 96h. A redução na concentração de PAs livres totais, principalmente espermidina e a diminuição no acúmulo de proteínas de choque térmico em sementes envelhecidas à 47°C foram associados à indução do envelhecimento das sementes. Além disso, a diminuição no acúmulo da proteína aconitato hidratase e o aumento no acúmulo da proteína álcool desidrogenase foram associados à perda de germinabilidade das sementes e rápida deterioração em sementes envelhecidas à 47°C. O envelhecimento de sementes de C. legalis está relacionado à alteração no conteúdo de PAs e a alteração de acúmulo de algumas proteínas envolvidas na perda da germinabilidade.

Among commercial fruits, papaya (Carica papaya L.) is notably one of the most common ones around the world. The aim of the current study was to establish a somatic embryogenesis protocol for the hybrid UENF/CALIMAN 01, elucidating the effects of sucrose and 2,4-dichlorophenoxyacetic acid (2,4-D) on the induction of embryogenic cultures and the effect of polyethylene glycol (PEG) on somatic embryo maturation. Immature zygotic embryos of C. papaya hybrid UENF/CALIMAN 01 were cultured in MS culture medium supplemented with different concentrations of sucrose (30 and 60 g L-1) and 2,4-D (0, 20, 40 and 80 μM). The combination of 30 g L-1 sucrose and 20 μM 2,4-D resulted in the highest induction rates and the largest callus diameters. Furthermore, this combination was associated with the greatest potential to form somatic embryos. To promote maturation, cultures were inoculated in MS medium and exposed to different concentrations of PEG (0, 30 and 60 g L-1). The 60 g L-1 PEG treatment group had the highest average for the number, maturation and conversion of somatic embryos that resulted in plantlets at the end of the process. The addition of PEG to MS medium led to increased protein synthesis. These results are unprecedented for C. papaya hybrid UENF/CALIMAN 01 as the current study demonstrates the development of somatic embryogenesis in this high quality hybrid.

O sequenciamento dos genomas de diversos organismos tornou praticável a análise global da expressão gênica, fornecendo numerosas pistas quanto à função biológica e envolvimento de inúmeros genes nos processos biológicos estudados. A proteômica é um dos ramos da biologia molecular e da biotecnologia que passou por expressivo desenvolvimento na era pós-genômica. Apesar dos recentes avanços nas técnicas em proteômica, muito resta a ser aprimorado. Devido às particularidades do reino vegetal, várias abordagens devem ser adaptadas no sentido de aumentar a eficiência e a acurácia de resultados das análises proteômicas em plantas. Nesse sentido, os dados gerados pela proteômica podem contribuir substancialmente para a compreensão e acompanhamento de eventos fisiológicos e para o desenvolvimento de estratégias biotecnológicas. No que tange às espécies tropicais, as dificuldades são ainda maiores, devido ao fato de serem ricas em metabólitos secundários e ainda não terem seus genomas totalmente sequenciados. Esta revisão trata de tópicos atuais em estudos de proteômica de plantas, desafios e perspectivas, com ênfase em espécies tropicais.

Genome sequencing of various organisms allow global analysis of gene expression, providing numerous clues on the biological function and involvement in the biological processes studied. Proteomics is a branch of molecular biology and biotechnology that has undergone considerable development in the post-genomic era. Despite the recent significant advancements in proteomics techniques, still there is much to be improved. Due to peculiarities to the plant kingdom, proteomics approaches require adaptations, so as to improve efficiency and accuracy of results in plants. Data generated by proteomics can substantially contribute to the understanding and monitoring of plant physiological events and development of biotechnological strategies. Especially for tropical species, challenges are even greater, in the light of the abundance of secondary metabolites, as well as of the lack of complete genome sequences. This review discusses current topics in proteomics concerning challenges and perspectives, with emphasis on the proteomics of tropical plant species.

Culturas embriogênicas de Araucaria angustifolia foram estabelecidas em meio de cultura líquido BM suplementado com 2 µM Ácido 2,4 Diclorofenoxiacético, 1 µM 6-Benzilaminopurina e 1 µM Cinetina (BM2) e em meio BM isento de reguladores de crescimento (BM0). Durante 42 dias de cultivo, o padrão de crescimento celular em ambas as culturas foi similar. O pH do meio de cultura BM0 e BM2 sofreu uma progressiva redução durante o período de cultivo. Em ambas as culturas embriogênicas foram observadas um consumo preferencial de glicose no período final da curva de crescimento celular. O nível de proteínas extracelulares foi similar para ambas as culturas embriogênicas. A dupla coloração com carmin acético e azul de Evans revelou diferenças na organização das linhagens celulares embriogênicas, sendo que a presença de proembriões somáticos bipolares foi apenas evidenciada nas culturas embriogênicas mantidas em meio de cultura líquido sem reguladores de crescimento.

Embryogenic cultures of Araucaria angustifolia were established in a BM liquid medium supplemented with 2 µM 2,4-dichlorophenoxyacetic acid, 1 µM 6-benzylaminopurine and 1 µM kinetin (BM2) and in a BM medium free of growth regulators (BM0). During 42 days in culture, the cell growth pattern of both cultures was similar. The pH of the culture medium of both BM0 and BM2 underwent progressive reduction during culture time. For both the embryogenic cultures a preferential uptake of glucose in the late stages of cell growth kinetics was observed. The extracellular protein content was similar for both the embryogenic cultures. Acetocarmine and Evan's blue double stain showed major differences for early somatic embryo organisation, in which only the embryogenic culture grown in a liquid culture medium free of plant growth regulators showed the presence of bipolar somatic pro-embryos.

O objetivo deste trabalho foi determinar os níveis de PAs em tecidos de medula e cultura de calos de plantas haplóides e diplóides de tabaco, obtidas da região apical e basal do caule. Estes explantes foram cultivados em meio RM-64 suplementado com AIA e cinetina, na luz e no escuro, durante vários subcultivos. Nos tecidos medulares, os níveis de PAs apresentam um decréscimo basípeto em diplóides e um aumento em haplóides.Um padrão similar nos níveis de PAs totais (livres+ conjugadas) foi observado em calos haplóides e diplóides mantidos na luz, e haplóides no escuro, enquanto os diplóides cultivados no escuro mostraram um aumento constante até o final do cultivo. O aumento no conteúdo de PAs nos calos diplóides no escuro, foi devido ao aumento do conteúdo de Put livre. Foi observado que a ploidia da planta pode expressar diferentes gradientes de PA ao longo do tecido medular e nas culturas de calos in vitro.

The aim of this work was to determine PAs levels in pith tissues and callus cultures from haploid and diploid tobacco plants, explanted from the apical and basal regions of the stem. These explants were cultured in an RM-64 medium supplied with IAA and kinetin, under light or in the dark, during successive subcultures. PAs levels followed a basipetal decrease in diploid and an increase in haploid, pith tissues. A similar pattern of total PAs (free + conjugated) was observed for the callus of diploid and haploid plants maintained in the light, and for the haploid callus in the dark, whereas the diploid callus in the dark showed a constant increase in total PAs levels until the end of culture. The PA increase in the diploid callus in the dark was related to free Put levels increase. The ploidy status of the plants could express different PA gradients together with the plant pith and in vitro callus cultures.

O objetivo deste trabalho foi a caracterização dos perfis protéicos expressos durante o desenvolvimento da semente de O. catharinensis, uma espécie recalcitrante, via eletroforese bidimensional. A extração de proteínas foi realizada utilizando tampão uréia/tiouréia seguido de uma etapa de precipitação com TCA 10%. A análise comparativa durante o desenvolvimento da semente mostrou um grande número de proteínas exclusivas em cada estádio do desenvolvimento. O estádio cotiledonar, que representa a fase de transição entre a embriogênese e o começo do metabolismo relacionado a maturação, apresentou o maior número de proteínas estádio-específicas. Proteínas relacionadas ao metabolismo oxidativo e a síntese de reservas foram identificadas via MS/MS. Estes resultados contribuem para o melhor entendimento do metabolismo de proteínas durante o desenvolvimento de sementes recalcitrantes, além de prover informações no estabelecimento de parâmetros para protocolos de embriogênese somática.

The aim of the present work was to characterize changes in the protein profile throughout seed development in O. catharinensis, a recalcitrant species, by two-dimensional gel electrophoresis. Protein extraction was undertaken by using a thiourea/urea buffer, followed by a precipitation step with 10% TCA. Comparative analysis during seed development showed that a large number of proteins were exclusively detected in each developmental stage. The cotyledonary stage, which represents the transition phase between embryogenesis and the beginning of metabolism related to maturation, presents the highest number of stage-specific spots. Protein identification, through MS/MS analysis, resulted in the identification of proteins mainly related to oxidative metabolism and storage synthesis. These findings contribute to a better understanding of protein metabolism during seed development in recalcitrant seeds, besides providing information on established markers that could be useful in defining and improving somatic embryogenesis protocols, besides monitoring the development of somatic embryos in this species.

Piper solmsianum é uma espécie herbácea do sudeste brasileiro onde vários compostos biologicamente ativos já foram identificados. O objetivo deste trabalho foi estabelecer suspensões celulares nesta espécie. Para tanto, foram utilizados explantes de pecíolos e folhas, retirados de plântulas cultivadas in vitro, os quais foram submetidos a diferentes combinações de reguladores de crescimento (AIA, ANA, 2,4-D e BAP). Foi obtida a neo-formação de raízes e brotos, estes últimos através do processo de organogênese indireta evidenciada por estudos histológicos. Para a indução e crescimento dos calos, foram avaliados, além das diferentes combinações de reguladores de crescimento, a suplementação ao meio de cultura de carvão ativado (1,5 mg.l-1) e o regime de luz. Culturas mantidas na luz, em meio de cultura suplementado com 0,2 mg.l-1 2,4-D e 2 mg.l-1 BAP e sem carvão ativado, resultaram em maior crescimento (massa fresca) dos calos. A partir destes calos, foram obtidas suspensões celulares, cuja dinâmica de crescimento e acúmulo de metabólitos foi estudado. Os resultados obtidos deverão ser utilizados para a caracterização de rotas biosintéticas em culturas in vitro em P. solmsianum.

Piper solmsianum is a shrub from Southeast Brazil in which many biologically active compounds were identified. The aim of this work was to establish a cell suspension culture system for this species. With this in mind, petiole and leaf explants obtained from in vitro plantlets were cultured in the presence of different plant growth regulator combinations (IAA, NAA, 2,4-D and BA). Root and indirect shoot adventitious formation, detected by histological analysis, was observed. Besides the different combinations of plant growth regulators, light regime and the supplement of activated charcoal (1.5 mg.l-1) were tested for callus induction and growth. Cultures maintained in light, on a 0.2 mg.l-1 2,4-D and 2 mg.l-1 BA supplemented medium, and in the absence of activated charcoal, showed the highest calli fresh matter increment. From a callus culture, cell suspension cultures were established and their growth and metabolite accumulation studied. The achieved results may be useful for further characterization of the activated secondary metabolites pathways in in vitro systems of P. solmsianum.

Culturas embriogênicas de A. angustifolia foram induzidas a partir de embriões zigóticos dominantes e não-dominantes excisados de sementes imaturas provenientes de três diferentes genótipos, em cinco datas de coleta. Os embriões zigóticos foram inoculados em meio de cultura LP e BM suplementados ou não com 2,4-D (5 µM), BA (2 µM) e Kin (2 µM). O genótipo da planta doadora e o estádio de desenvolvimento do explante influenciaram significativamente a freqüência de indução. Na fase de manutenção, as culturas embriogênicas foram mantidas em ciclos contínuos de subcultivo a cada 20 dias em meios semi-sólidos ou líquidos. Na fase de maturação, foram testados fontes e níveis de fitorreguladores, agentes osmóticos, carboidratos e derivados. Meios de cultura suplementados com PEG 3350 (6 e 9%), maltose (6 e 9%), BA e Kin (1 µM cada) foram efetivos no desenvolvimento de embriões somáticos nos estádios globular e torpedo.

Embryogenic cultures of Araucaria angustifolia were induced from dominant and non-dominant zygotic embryos excised from immature seeds proceeding from three different genotypes and five harvest dates. Zygotic embryos were inoculated in inductive culture medium LP and BM supplemented with or without plant growth regulators 2,4-D (5 µM), BA (2 µM) and Kin (2 µM). The genotype of the mother tree and the developmental explant stage affected the induction frequency. In the maintenance phase, embryogenic cultures were maintained at continuous repetitive cell cycles every 20 days in semi-solid or liquid medium. In the maturation phase the culture medium was supplemented with different types and levels of growth regulators, osmotic agents, carbohydrates and derived. Embryogenic cultures inoculated in culture medium supplemented with PEG 3350 (6 and 9%), maltose (6 and 9%), plus BA and Kin (1 µM each) resulted in the progression of somatic embryos to globular and torpedo developmental stages.
