ABSTRACT: Desiccation interrupts the acquisition of physiological quality in soybean seeds, as this quality peaks at R9. However, this interruption can be mitigated by the stage in which herbicide is applied and which herbicide is used. The aim of this study was to characterize the acquisition of physiological quality in soybean seeds from plants desiccated by herbicides at different stages. A completely randomized experimental design was used in a 3×6 factorial arrangement, consisting of three phenological stages (R6, R7.1, R7.3) and six different desiccant treatments (Diquat 1.5 L.ha-1; Glufosinate 1.5 L.ha-1 and 2.0 L.ha-1; Diquat 1.0 L.ha-1 + Glufosinate 1.0 L.ha-1; Saflufenacil 140 g.ha1 + Glufosinate 1.0 L.ha-1; and Saflufenacil 140 g.ha-1 + Diquat 1.0 L.ha-1), with four replications. The following evaluations were made: desiccation rate, germination, desiccation tolerance (DT), longevity (P50), viability after one year of storage, and thousand seed weight. Seeds are able to germinate and have DT when plant desiccation through application of Diquat, Diquat + Glufosinate, Saflufenacil + Diquat, and Saflufenacil + Glufosinate occurs at R6, and for Glufosinate, at R7.1. The use of Diquat + Glufosinate and their combination with Saflufenacil showed less impact on acquisition of soybean seed longevity, which was better when application was made at R7.3. RESUMO: A dessecação interrompe a aquisição da qualidade fisiológica de sementes de soja, que atinge seu máximo em R9. Contudo, isto pode ser mitigado pelo estádio de aplicação e o herbicida utilizado. Objetivou-se caracterizar a aquisição da qualidade fisiológica de sementes de soja de plantas dessecadas por herbicidas em estádios distintos. ABSTRACT. Water and salt stress conditions affect germination and seedling emergence. It is known that physiological priming can not only trigger different mechanisms to increase the speed and uniformity of germination, but also influence the antioxidant defence system of the seeds, especially in adverse conditions. In this context, the objective of this work was to evaluate the physiological seed quality and the activity of antioxidant enzymes in seeds of Petroselinum crispum induced to tolerate saline stress through osmo-priming. Seeds were placed in polyethylene glycol solutions with three osmotic potentials (-0.5, -1.0 or -1.5 MPa) for 2, 4 or 6 days. Subsequently, they were placed on substrates moistened with sodium chloride solution (NaCl), in concentrations that generated osmotic potentials of -0.2, -0.4, -0.6, -0.8 or -1.0 MPa, to germinate. The germination percentage, germination speed index, seedling length, fresh weight, dry weight and activity of the antioxidant enzymes superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were evaluated. Germination and germination speed index decreased linearly with the increase in NaCl concentrations, indicating the species sensitivity. As for length, fresh weight and dry weight of seedlings, this effect was more expressive from the potential of -0.4 MPa. The vigour and activity of SOD, POD, and CAT enzymes were reduced in unprimed seeds, emphasising the effectiveness of the technique. The induction of tolerance to saline stress can be related to the activity of the antioxidant system observed in seeds.

RESUMO O desenvolvimento de novos genótipos de mamoneira com alta produtividade de grãos e qualidade fisiológica de sementes é fundamental para o sucesso econômico da cultura. Objetivou-se avaliar o desempenho agronômico e a qualidade fisiológica de sementes de doze linhagens de mamoneira, para determinar genótipos superiores. Foram avaliados a altura de planta, altura de inserção e número de racemos, rendimento de grãos, teor de óleo, porcentagem de germinação e vigor. As linhagens anãs de mamoneira H4, H5 e H11 foram responsáveis por produtividade de grãos acima de 1.400 kg ha-1 e, portanto, são promissoras para a obtenção de novos híbridos. A seleção indireta de linhagens de mamoneira anã pode ser realizada por meio de características morfológicas. As linhagens H4, H6, H9, H11 e H12 produziram sementes com vigor e qualidade germinativa superiores.

ABSTRACT The development of new castor bean genotypes with high grain yield and physiological seed quality is fundamental to the economic success of the crop. This study aimed to evaluate the agronomic performance and physiological quality of seeds of twelve castor bean lines, in order to determine superior genotypes. Plant height, insertion height and number of racemes, grain yield, oil content, germination percentage and vigor were evaluated. The dwarf castor bean lines H4, H5 and H11 were responsible for a grain yield above 1,400 kg ha-1 and are, therefore, promising for getting new hybrids. The indirect selection of dwarf castor bean lines can be performed through morphologic traits. The lines H4, H6, H9, H11 and H12 produced seeds with superior vigor and germinative quality.

RESUMO Espécies que produzem sementes com tegumento rígido representam um sério problema, pois esta estrutura restringe a entrada de água, o que dificulta a germinação. O heteromorfismo do tegumento em Bowdichia virgilioides faz com que a embebição de água varie substancialmente e, portanto, o objetivo deste trabalho foi estudar a relação da cor do tegumento com a permeabilidade e viabilidade em Bowdichia virgilioides. As sementes foram separadas em cinco cores diferentes (amarelado/alaranjado, alaranjado, alaranjado/ avermelhado, avermelhado e vermelho ferrugíneo) e os seguintes parâmetros foram analisados: absorção de água, germinação, mortalidade, viabilidade, sincronia e potencial máximo de germinação. A cor foi um fator determinante para a maioria dos parâmetros avaliados. As sementes com camadas amareladas, alaranjadas e alaranjados/avermelhado tendem a ser pouco permeáveis à água, enquanto as sementes com camadas avermelhadas ou vermelho ferrugíneo são altamente permeáveis. Os resultados indicaram que sementes com coloração avermelhada apresentaram maior potencial fisiológico indicando provável relação com o ponto de maturação da espécie. Porém, nas sementes da espécie estudada, independentemente da coloração, a germinação foi tardia, com baixa sincronia e baixa contagem diária de germinação, características típicas de dormência.

ABSTRACT Species that produce seeds with a rigid integument represent a serious problem because their impermeable coat restricts the entry of water, which makes germination difficult. The heteromorphism of the integument in Bowdichia virgilioides causes water imbibition to vary substantially and, therefore, the objective of this work was to study the relation of seed coat color with permeability and viability in Bowdichia virgilioides. Seeds were separated into five different colors (yellowish-orange, orange, orange/reddish, reddish, and rusty red) and the following parameters were analyzed: water absorption, germination, mortality, viability, synchrony, and maximum potential of germination. The color was a determining factor for most of the evaluated parameters. The seeds with yellowish or orange/reddish coats tend to be little permeable to water while seeds with reddish or rusty red coats are highly permeable. The results indicated that seeds with reddish color had greater physiological potential indicating a probable relationship with the maturation point of the species. However, in the seeds of the species studied, irrespective of coloring, germination was late with low synchrony and low daily seed germination count, typical characteristics of dormancy.

Resumo: A análise da longevidade de um lote de sementes pode auxiliar na decisão de por quanto tempo armazenar um lote de sementes até a sua comercialização, uma vez que esta característica implica na manutenção da viabilidade ao longo do tempo. A longevidade é analisada pelo teste de p50, o qual representa o tempo para perda de 50% da viabilidade inicial. Sementes com elevada germinação e vigor têm maior potencial fisiológico e, assim, maior capacidade de manter a qualidade ao longo do período de armazenamento. No entanto, pouco tem sido estudado quanto às correlações entre a análise do p50 (longevidade) com os testes utilizados para mensurar o potencial fisiológico de lotes (germinação e vigor), o que pode ser utilizado como ferramenta para obter inferências sobre a longevidade por meio dos testes mais comumente utilizados. Assim, objetivou-se investigar quais testes utilizados para mensurar o potencial fisiológico de lotes pode ser utilizado para estimar o p50. Para isso, foram avaliados a germinação e o vigor, utilizando-se testes tradicionais e, em paralelo, a longevidade em oito lotes de sementes de soja. As correlações e a regressão linear para as variáveis tradicionais versus p50 também foram testadas. Foi verificado que o uso dos testes de envelhecimento acelerado, condutividade elétrica e tempo para 50% de protrusão da radícula tem elevado potencial para estimar a longevidade mensurada pelo p50.

Abstract: The analysis of longevity can support decisions about the length of seed lot storage until commercialization, since this characteristic implies the maintenance of viability over time. Seed longevity is analyzed by the p50 test, which expresses the time to lose 50% of the initial viability. Seeds with high vigor and germination have greater physiological potential and, thus, a greater capacity to maintain quality throughout the storage period. However, there has been little research on the correlations between the analysis of p50 (longevity) and the tests used to measure physiological potential (germination and vigor) of lots, which can be used as a tool to make inferences about longevity using the most traditional tests. Thus, the objective of this study was to investigate which tests used to measure the potential of lots can estimate p50. To this end, germination and vigor were evaluated using traditional tests while longevity was assessed in eight soybean seed lots. Correlations and linear regression were tested for the traditional variables versus p50. It was found that the use of accelerated aging, electrical conductivity, and time to 50% radicle protrusion has high potential to estimate longevity as measured by p50.

Resumo: A germinação da semente do café (Coffea arabica L.) é resultado de eventos que ocorrem simultaneamente no embrião e endosperma. Para entender os mecanismos moleculares responsáveis por estes eventos, nós realizamos uma análise transcricional do embrião e dos endospermas micropilar e lateral de sementes embebidas por 10 dias. O sequenciamento obteve contigs que codificam 16.813 proteínas. Destas, 14.005 (~ 83%) foram altamente similares a pelo menos uma sequência de proteína na base de dados nr. 162 genes foram significativamente expressos em embrião, 36 em endosperma micropilar e 72 em endosperma lateral. A análise de tecido-especificidade dos genes significativamente expressos mostrou que a maior proporção de genes específicos está no embrião (113/162, ~70%), enquanto 11 genes foram expressos nos endospermas lateral e micropilar. No embrião, os genes foram associados principalmente com estresse abiótico, crescimento celular e comunicação intercelular. Nos endospermas micropilar e lateral, eles foram associados com estresse abiótico e degradação da parede celular. A acurácia da análise de RNA-seq foi confirmada por RT-qPCR. Este trabalho acrescenta novas informações sobre os mecanismos moleculares envolvidos na germinação de sementes de café.

Abstract: Germination of the coffee (Coffea arabica L.) seed is the result of events that occur simultaneously in the embryo and endosperm. To understand the molecular mechanisms responsible for these events, we undertook a transcriptome analysis of embryo, micropylar and lateral endosperms from 10-day-imbibed seeds. The sequencing yielded contigs coding for 16,813 proteins. From those, 14,005 (~ 83%) were highly similar to at least one protein sequence in the nr database. 162 genes were significantly expressed in the embryo, 36 in the micropylar endosperm and 72 in the lateral endosperm. The tissue specificity analysis of the significantly expressed genes showed that the embryo had the highest proportion of specific genes (113/162, ~70%), while 11 were expressed in the micropylar and lateral endosperms. In the embryo, genes were mainly associated with abiotic stress, cell growth, and intercellular communication. In the micropylar and lateral endosperms, they were associated with abiotic stress and cell wall degradation. The accuracy of RNA-seq data was confirmed by RT-qPCR. This work adds new information about the molecular mechanism involved in coffee seed germination.

Resumo: A sobrevivência durante o armazenamento ou longevidade é pré-requisito para evitar a deterioração, que leva à perda de vigor. A longevidade é rotineiramente avaliada pela habilidade da semente em germinar depois do armazenamento, que aumenta progressivamente durante a maturação depois da aquisição da tolerância à dessecação. No entanto, a germinação representa somente uma parte do processo de estabelecimento da planta. Foi avaliado como a maturação afeta a resistência de várias características, como vigor, relacionadas ao estabelecimento das plântulas em sementes de soja da variedade BRS 284. Três novos estádios entre 7.1 e 7.2 (7.1.1, 7.1.2 e 7.1.3) foram introduzidos para capturar o rápido aumento na longevidade das sementes. O armazenamento induziu uma queda na velocidade de germinação a partir de 7-14 dias de armazenamento, dependendo do estádio, e essa queda diminuiu com a maturação, a partir do estádio 7.1.3. O tempo para perda de 50% na capacidade de alongamento de ambos os órgãos foi similar àquele observado para capacidade de germinação. Além disso, esse tempo aumentou de forma constante durante o período de maturação, demonstrando a importância das fases finais de maturação da semente na construção do vigor.

Abstract: Long-term survival during dry storage or longevity is a pre-requisite to avoid deterioration, leading to loss of vigor. Longevity is routinely evaluated by the ability to germinate after storage. It increases progressively during seed maturation, after the acquisition of desiccation tolerance. However, the capacity to germinate represents only a part of the success of crop establishment. How seed maturation affects the resistance of several traits, as vigor, associated with seedling establishment, against deterioration was evaluated during seed filling and post-abscission phase of soybean BRS 284 seeds. Three new phenological stages between 7.1 and 7.2 (7.1.1, 7.1.2 and 7.1.3) were introduced to capture the rapid increase in seed longevity. Germination speed started to be affected at 7-14 days after storage depending on the stages. The delay on germination increased with maturation from 7.1.3 to dry mature seeds. The time to 50% loss of elongation capacity of both organs during storage was similar to that of loss of germination. Also, it increased steadily during seed maturation after mass maturity and harvest maturity stages, highlighting the importance of the late phase of seed maturation for building seed vigor.

RESUMO Objetivou-se com esse trabalho avaliar a integridade do DNA e identificar mudanças ultra-estruturais em raiz primária de sementes de Peltophorum dubium submetidas a secagem. Para tanto, sementes foram posta a germinar e ao atingirem 1 mm de comprimento da raiz primária, foram desidratadas, até atingirem diferentes graus de umidade, sendo em seguida reidratadas e avaliadas quanto à sobrevivência. Após esse procedimento foram selecionados diferentes pontos de interesse (controle, 20% e 10% de umidade) para avaliação da integridade do DNA e da ultraestrutura celular no intuito de detectar possíveis mudanças celulares nas raízes primárias após desidratação. Quando secas até 32% de umidade não houve influência na retomada do crescimento das sementes germinadas, mas ao serem desidratadas a 20% de umidade, a sobrevivência foi reduzida a 67% e a secagem a 8% de umidade concorreu para nulidade na sobrevivência. Pela avaliação de mudanças ultraestruturais em células da raiz observou-se que secagem a aproximadamente 20% de umidade marcou o início dos danos por dessecação, tanto fisiológica quanto ultraestruturalmente. Secagem a níveis próximos a 10% de umidade concorreu não somente para total desconfiguração e ruptura das membranas como também promoveu colapso intracelular. Foi observada perda da integridade do DNA genômico no decorrer da secagem.

ABSTRACT The aim of this study was to evaluate DNA integrity and to identify ultrastructural changes in the primary root of Peltophorum dubium seeds subjected to drying, since these changes may reflect the germination capacity. Seeds were germinated and were dehydrated when reached 1 mm in length of the primary root, until reaching different moisture contents, being then rehydrated and evaluated in relation to survival. After this procedure, different points of interest (control, 20% and 10% moisture content) were selected to evaluate DNA and cell ultrastructure integrity in order to detect possible cellular changes in primary roots after dehydration. When dried down to 32% moisture, there was no influence on the growth resumption of the germinated seeds, but when they were dehydrated to 20% moisture, the survival was reduced to 67%, reaching nullity in the survival with drying to 8% moisture. By the evaluation of ultrastructural changes in root cells, it was observed that drying to approximately 20% moisture content marked the onset of damages by desiccation, both physiologically and ultrastructurally. Drying to levels close to 10% moisture content was not only responsible for total disorganization and rupture of membranes, but also promoted intracellular collapse. Loss of genomic DNA integrity was observed during drying.

RESUMO A uvaieira (Eugenia pyriformis Cambess.), espécie arbórea e frutífera, com potencial farmacológico e gastronômico, apresenta sementes sensíveis à dessecação. O objetivo deste trabalho foi analisar se o grau de tolerância à dessecação de sementes de uvaieira depende do grau de maturidade das sementes no momento de sua dispersão. Este, por sua vez, depende das condições ambientais nas quais as sementes se desenvolvem, incluindo-se o acúmulo de graus-dia e a chuva acumulada no período. As sementes foram coletadas de frutos maduros de matrizes localizadas em São Paulo e Minas Gerais, submetidas à secagem e analisadas quanto ao teor de água e germinação. Utilizou-se o delineamento inteiramente casualizado, em esquema fatorial 20 x 3 (origem do material x níveis de secagem). O grau de tolerância à dessecação difere entre as regiões e épocas de coleta, inclusive dentro da mesma matriz, quando as sementes são coletadas em anos distintos. As condições hídricas e térmicas do ambiente, durante o desenvolvimento das sementes, condicionam o ciclo de maturação, a qualidade fisiológica e a aquisição da tolerância à dessecação. A tolerância à dessecação de sementes de uvaieira depende da maturidade fisiológica das sementes no momento de sua dispersão, a qual está associada às condições ambientais.

ABSTRACT Eugenia pyriformis Cambess., known locally as uvaieira, a species of fruit-bearing tree with both pharmacological and gastronomic potential, has seeds which are sensitive to desiccation. The aim of this study was to analyse whether the degree of tolerance to desiccation of uvaieira seeds depends on the stage of maturation of the seeds at shedding. This, in turn, depends on the environmental conditions in which the seeds develop, including the accumulation of degree-days and rainfall in the period. Seeds were collected from the ripe fruit of parent plants located in the states of São Paulo and Minas Gerais, Brazil, submitted to drying and analysed for water content and germination. A completely randomised design was used in a 20 x 3 factorial scheme (source of material x level of drying). The degree of desiccation tolerance differs between region and period of collection, even for the same parent plant when the seeds are collected in different years. The water and thermal conditions of the environment during seed development modify the maturation cycle, the physiological quality and the acquisition of desiccation tolerance. In uvaieira seeds, desiccation tolerance depends on the physiological maturity of the seeds at the time of dispersal, which is associated with the environmental conditions.

RESUMO: A cultura da soja apresenta importância comercial extremamente relevante para a economia brasileira. O condicionamento fisiológico é uma técnica que permite melhorar a qualidade das sementes, sobretudo daquelas de baixo vigor. O vigor é definido como a capacidade das sementes em apresentar desempenho adequado, quando expostas a diferentes condições ambientais. Sementes de alto vigor produzem plantas com maior velocidade de emergência e maiores níveis nos componentes de rendimento, desdobrando assim em uma maior produtividade de sementes no final do seu ciclo. O objetivo foi avaliar o comportamento agronômico em plantas, em condições de campo, oriundas de sementes de soja condicionadas sob diferentes potenciais osmóticos, em função dos níveis de vigor. Foram avaliados os componentes de produção e a produtividade da soja. O condicionamento fisiológico em sementes de médio vigor influencia positivamente nos componentes (emergência de plântulas, altura de inserção da primeira vagem, número de vagens/planta, número de sementes/planta, número de sementes/vagem) e na produtividade da cultivar 'M7211RR' de soja. Os diferentes níveis de vigor das sementes de soja ocasionam diferenças no número de vagens/planta.

ABSTRACT: The Soybean crop is extremely relevant to the Brazilian economy. Priming is a technique that allows improvement of seed quality, especially those of low vigor. Vigor is defined as the ability of the seed to perform adequately when exposed to different environmental conditions. High vigor seeds generate plants with higher emergency speed and superior yield components, favoring into a higher seed yield. The objective of this work was to evaluate the agronomic behavior of soybean plants under field conditions originated from primed seeds with different level of vigor. Yield components and soybean yield were also evaluated. Seed priming in seed with medium vigor positively influences the components (seedling emergence, first pod height, number pods/plant, number of seeds/plant, number of seeds/pod) and favored productivity of soybean cultivar 'M7211RR'. Different levels of vigor of soybean seeds cause differences in the number of pods/plant.

O objetivo geral deste trabalho foi avaliar o uso da técnica de PCR (Polimerase Chain Reaction) na caracterização de cultivares de soja. O estudo foi realizado no Departamento de Produção Vegetal da Faculdade de Ciências Agronômicas / UNESP e do Instituto de Biociências de Botucatu-SP. Foram utilizadas quatorze cultivares de soja comerciais, das quais seis foram selecionadas como reação positiva à peroxidase (BRS 320, BRS 284, BRS 232, BRS 7860RR, BRSMG 760SRR, BRS295RR), quatro como reação negativa (BRS 326, BRS 8160RR, BRSMG 800A (NutriSoy), BRS Valiosa RR) e quatro como reação positiva e negativa (BRS 8060, BRS 270RR, FTS Campo Mourão e BRS 239). Assim, as 14 cultivares foram submetidas ao ensaio bioquímico colorimétrico tradicional e os resultados obtidos foram comparados com o ensaio de PCR convencional. Para a análise de PCR, o DNA foi extraído de sementes inteiras, sendo que os primers foram testados por PCR e visualizados por eletroforese em gel de agarose. A combinação dos primers prx9 + prx10 confirmou a utilização da reação de PCR para caracterizar as cultivares de soja considerada duvidosa por teste convencional colorimétrico.

The aim of this work was to evaluate the use of the molecular biology technique of PCR (Polimerase Chain Reaction) in the characterization of soybean cultivars. The study was performed at the Department of Plant Production, Faculty of Agricultural Sciences/ UNESP and Institute of Bioscience, Botucatu-SP. Fourteen commercial soybean cultivars were used, of which six were selected as positive reaction to peroxidase (BRS 320, BRS 284, BRS 232, BRS 7860RR, BRSMG 760SRR, BRS295RR), four as negative reaction (BRS 326, BRS 8160RR, BRSMG 800A (NutriSoy), BRS Valiosa RR) and four as double reaction (BRSGO 8060, BRS 270RR, FTS Campo Mourão and BRS 239). Thus, the results attained by the traditional biochemical colorimetric test for the 14 cultivars were compared with the conventional PCR assay. For PCR analysis, DNA was extracted from whole seeds and the primers were tested, and subsequently PCR and agarose gel electrophoresis were performed. The combination of primers prx9 + prx10 confirmed the use of the PCR reaction to characterize soybean cultivars considered doubtful by conventional colorimetric text.

(Burseraceae) foram coletadas e transportadas ao Laboratório de Sementes Florestais, aonde foram beneficiadas e submetidas a dois tipos de secagem, rápida e lenta. Em seguida, foram armazenadas em freezer ou em câmara fria e avaliadas quanto à viabilidade. Após a classificação fisiológica, verificou-se que sementes de C. sylvestris e E. incanus são ortodoxas. Em contra-partida, sementes de G. kunthiana e P. heptaphyllum foram classificadas como recalcitrantes e Q. grandiflora como intermediárias, pois não toleram a secagem a baixos conteúdos de água.

Seeds of five forest species were classified according to their physiological storage behavior. Seeds of Casearia sylvestris Swart (Salicaceae), Qualea grandiflora Mart. (Vochysiaceae), Guarea kunthiana A. Juss. (Meliaceae), Eremanthus incanus Less. (Asteraceae), Protium heptaphyllum March. (Burseraceae) were collected and taken to the laboratory, where they were processed and submitted to both rapid and slow drying, storage and assayed for viability. After physiological classification regarding storage behavior, it was observed that seeds of C. sylvestris and E. incanus presented orthodox behavior. Seeds of G. kunthiana and P. heptaphyllum were classified as recalcitrant and Q. grandiflora as an intermediate, which did not tolerate low moisture content.

No presente trabalho, objetivou-se avaliar os efeitos da dessecação e identificar lesões por predação por insetos, em diásporos de canela-batalha (Cryptocarya aschersoniana Mez.), utilizando-se testes de raios X. Os danos provocados pela dessecação foram dimensionados nas imagens e associados à formação de plântulas. Diásporos recém-beneficiados (45 % de umidade e 37 % de germinação) foram colocados para secar em sala climatizada (20 ºC e 60 % UR), dentro de bandejas plásticas em camada única. Posteriormente, com o intuito de acelerar o processo de secagem, foram colocados em caixas de secagem com solução saturada de hidróxido de sódio (28 % UR) e amostrados com 45, 37, 35, 31 e 26 % de umidade. Para as radiografias, utilizou-se a intensidade de radiação de 40 kVp e tempo de exposição de 1,5 minutos. Posteriormente, as radiografias foram fotografadas e as imagens analisadas em computador, sendo medido o afastamento entre o endocarpo e a semente. As sementes foram classificadas em sementes intactas, sementes com afastamento parcial, sementes com afastamento total e sementes predadas. Os testes de germinação foram realizados sobre areia, em germinadores tipo Mangelsdorf a 25 ºC e luz branca constante. Pelos resultados, observa-se que a germinação é comprometida quando o teor de água das sementes fica abaixo de 26 %. Nesse ponto, o afastamento entre o endocarpo e a semente é de 0,65 mm. Houve uma correlação positiva entre a viabilidade das sementes, avaliada pelo teste de germinação, e o afastamento entre o endocarpo e a semente observado nas radiografias. A análise radiográfica possibilita identificar danos provocados por predação após infestação por insetos.

This study evaluated the effect of desiccation, and identified injuries and predation by insects in diaspores of "canelabatalha" (Cryptocarya aschersoniana Mez.) by using X-rays. The damages caused by desiccation were measured in the images and related to the formation of seedlings. Right after fruit depulping, the diaspores (endocarp plus seed) (45 % moisture content-MC and 37 % germination) were dried in acclimatized room (20 ºC and 60 %% RH), inside of plastic trays in a single layer. Later, in order to speed up the drying process, they were placed in drying boxes with saturated solution of sodium hydroxide (28 %% RH) and sampled with 45, 37, 35, 31 and 26 % MC. For the X-rays, an intensity of radiation of 40 KVp was used and the time of exposition of 1.5 min. The radiographies were then photographed and the images analyzed in computer, being measured the separation between endocarp and the seed. The diaspores were classified as intact; with partial separation; with total separation and predated. The germination tests were carried out on sand, in incubators Mangelsdorf at 25 ºC and constant white light. The results showed that the germination is affected when the diaspore MC reaches values below 26 %. In this point, the separation between endocarp and the seed is 0.65 mm. There was a positive correlation between the viability of the seeds, evaluated through germination tests and the separation between endocarp and the seed, observed through X-rays. The radiographic analysis helps identify damage caused by predation after insect infestation.

O objetivo deste trabalho foi avaliar a sensibilidade à dessecação de sementes de Tapirira obtusa (Benth.) J. D. Mitchell provenientes de três ambientes e submetidas a duas velocidades de secagem. As sementes foram coletadas em área de campo rupestre, Cerrado e em mata ciliar, da região de Lavras, MG. As sementes foram submetidas à secagem com cloreto de magnésio (secagem lenta) ou sílica gel (secagem rápida), em ambiente fechado, até os graus de umidade de 40%, 30%, 20% e 10%, considerando como controle o percentual de germinação na umidade inicial em cada ambiente, a qual variou de 47% a 50%. Foram avaliados os percentuais de germinação e de plântulas normais e também o índice de velocidade de germinação. Nos três ambientes analisados, não houve efeito da secagem lenta sobre a germinação das sementes. Sementes oriundas do Cerrado apresentaram pequena redução da germinação, quando submetidas à secagem rápida. Sementes oriundas de áreas rupestres apresentaram aumento na germinação quando submetidas à secagem rápida. Sementes oriundas de mata ciliar não apresentaram redução na germinação, independente da velocidade de secagem. Os resultados sugerem que sementes de T. obtusa não apresentam sensibilidade à dessecação.

This study was aimed at evaluating the desiccation sensitivity in seeds of the tree Tapirira obtusa (Benth.) J. D. Mitchell collected from three different environments and subjected to two distinct drying speeds. Seeds were collected from a rocky area, in the "Cerrado", and in a riparian forest area, in the region of municipality of Lavras, State of Minas Gerais. The seeds were subjected to drying with magnesium chloride (slow drying) or silica gel (fast drying), into closed environment, until moisture contents of 40%, 30%, 20% and 10%, considering as control, the percentage of germination at the initial moisture content in each environment, which varied from 47% to 50%. Percentages of germination and normal seedlings as well as germination speed index were assessed. For the three environments studied, there was no effect of slow drying on seed germination. Seeds from area of Cerrado, however, have shown a slight reduction on germination when subjected to fast drying. Oppositely, seeds from rocky area had germination increased when subjected to fast drying. Seeds from riparian forest area had no reduction on germination percentage, independent of drying speed. Results suggest that seeds of T. obtusa are not sensitive to desiccation.

Conduziu-se este trabalho, com os objetivos de desenvolver métodos de beneficiamento e análise de raios X, e estudar o padrão de embebição de sementes de candeia (Eremanthus incanus). As sementes foram colhidas em 2001 e 2002, respectivamente, em Morro do Pilar e Lavras, beneficiadas e guardadas em câmara fria (5ºC/60%UR) até o início dos experimentos. Para identificar sementes vazias, foi desenvolvido um protocolo utilizando raios X combinando potência de radiação e tempo de exposição. Para a eliminação das sementes vazias foi realizado um experimento utilizando soprador tipo South Dakota, combinando aberturas do aparelho e tempo de ventilação. Os lotes originais e as categorias encontradas pelas radiografias foram testadas por testes de germinação, no qual as sementes foram lavadas em hipoclorito de sódio e distribuídas sobre papel, em gerbox, levadas para germinadores com temperatura alternada 20-30ºC, com 10 horas de luz, por até 15 dias. Foram determinadas as curvas de embebição, nas condições de 30ºC, 20-30ºC/10 horas de luz e 30-20ºC/10 horas de luz. O protocolo de raios X que melhor permitiu a visualização das estruturas internas da semente foi 30Kv por 45 segundos. A separação das sementes no soprador na abertura 3,0 por 30 segundos elevou o número de sementes com embrião para valores superiores a 99%, tendo como consequência, a elevação das taxas de germinação. O padrão de embebição é trifásico e o processo de germinação se completa em 72 horas. Sob temperatura alternada, o crescimento da raiz primária é favorecido.

The objective of this work was to develop a methodology for seed processing and X-ray analysis and to study imbibition pattern in seeds of candeia (Eremanthus incanus). Seeds were gathered in 2001 and 2002 in Morro do Pilar and Lavras respectively, processed and stored in a cold chamber (5ºC/60%RH) until the start of experiments. In order to identify unfilled seeds, a protocol was developed for X-ray use combining radiation energy (Kv) and exposure time (seconds). For elimination of unfilled seeds, an experiment was conducted using a South Dakota seed blower whereby different opening settings and ventilation times were combined. Original seed lots and categorized lots following radiograph viewing were tested by germination tests, with seeds being cleansed in sodium hypochlorite and scattered over blotting paper in 'gerboxes' and then taken to germinators with alternating temperatures of 20º-30ºC and 10 hours of light, for up to 15 days. Imbibition curves were determined under the conditions 30ºC, 20º-30ºC/10 hours of light and 30º-20ºC/10 hours of light. The X-ray protocol that best allowed viewing of internal seed structures was 30Kv for 45 seconds. Seed separation using the blower setting at a 3.0 opening for 30 seconds raised the number of embryonic seeds to values exceeding 99%, also raising the germination rates as a consequence. The imbibition pattern is three-phased and the germination process is completed in 72 hours. Under alternate temperatures, primary root growth is favored.
