Resumo Vigna unguiculata (L.) Walp (feijão-caupi), pertencente à família Fabaceae, é um dos mais importantes vegetais utilizados como alimento em regiões de clima tropical, especialmente em países da África, América do Sul e Ásia. Compostos fenólicos estão associados a importantes propriedades biológicas, e sua ocorrência em plantas comestíveis pode implicar em um alimento altamente funcional. Foi investigada a presença de compostos fenólicos nos extratos brutos metanólicos de duas cultivares de feijão-caupi, cultivar tracuateua (CT) e cultivar caldeirão (CC), utilizando a técnica HPLC-DAD/MS/MS. Os flavonóis quercetina e canferol, e os ácidos fenólicos p-cumárico e protocatecuico foram identificados no feijão-caupi (CC), enquanto no feijão-caupi (CT) foram identificados ácidos fenólicos gálico e protocatecuico. Esses fenólicos ratificam o feijão-caupi como um alimento funcional e bioativo, garantindo uma dieta saudável.

Abstract Vigna unguiculata (L.) Walp (cowpea), Fabaceae family and also known as Leguminosae, is an important vegetable used as food in tropical regions, especially in Africa, South America and Asia countries. Phenolic compounds are associated with important biological properties and their occurrence in edible plants may result in a highly functional food. Chromatographic profiles of phenolic compounds were investigated in two cowpea cultivars, such as tracuateua (CT) and caldeirão (CC), and both were cultivated using High Performance Liquid Chromatography (HPLC) coupled to Mass Spectrometry (MS) (HPLC-DAD/MS/MS). The flavonols quercetin and kaempferol, the phenolic acids, p-coumaric and protocatechuic acid (PCA) were identified in cowpea (CC), while the phenolic acids, gallic and protocatechuic acids, were identified in the cowpea (CT). These phenolic compounds ratify cowpea as a functional and bioactive food, ensuring a healthy diet.

Neolignans are a class of natural products with a wide range of biological effects. These substances are of great synthetic and biological interest, especially in searching for novel anticancer agents. In this paper, we report the synthesis of a new subclass of 8,4’-oxyneolignan analogues (β-ketoethers and β-ketoesters) and their cell viability assay on twenty four different cancer cells, among leukemias and carcinomas. Three compounds inhibited the growth of most human cancer cells. 2-Oxo-2-phenylethyl(2E)-3-[4-(2-oxo-2-phenylethoxy) phenyl]prop-2-enoate showed an antiproliferative activity superior to doxorubicin for U-87, U-138 MG and H1299 cell types and (E)-2-oxo-2-phenylethyl 3-(3-methoxy-4-(2-oxo-2-phenylethoxy)phenyl)acrylate was found to be very selective, demonstrating a growth inhibition of 92.0% against KG-1 cells. Furthermore, 1-oxo-1-phenylpropan-2-yl cinnamate exhibited significant inhibition activity in a range of 52.2 to 91.2% against twelve kinds of leukemia cell lines, revealing excellent results and very comparable to the reference drug.

This research deals with the chemical study and evaluation of the phytotoxic effects of the methanol extract of E. flavescens leaves. The compounds were isolated using classic chromatographic techniques and the identification of the phenolic profile was proposed from the HPLC/DAD/MS-MS data. Structure elucidation of the compounds was performed by 1H and 13C (1D e 2D) NMR techniques and HRMS. The chemical study led to the isolation of betulinic acid (1) and lupeol (2), identified in mixture quercitrin (3) and catechin (4) beyond myricetrin (5) in mixture with (3). In addition, several phenolic compounds were identified, among them, flavonoids and phenolic acids. For bioassays phytotoxic effect were used as test plants two invasive species, M. pudica and S. obtusifolia. The methanol extract showed significant seed germination inhibition. The triterpenoid 1 showed low inhibition effects on seed germination and moderate effects on radicle and hypocotyl growth. The mixture of flavonoids 3/4 showed effective phytotoxic effects, inhibiting 92,1% of the seed germination of M. pudica and 63,0% of S. obtusifolia. Inhibitions of 72,5% and 76,1% of M. pudica radicle and hypocotyl growths were observed, respectively, and 74,5% and 78,0% of S. obtusifolia for the same bioassays, respectively.

Neste trabalho estamos relatando o isolamento por métodos clássicos de cromatografia de seis policetídeos de Penicillium herquei. Os compostos citreoseine ( 1) , emodina ( 2) , janthinona ( 3) , citrinina ( 4) , citrinina H1( 5) e dicitrinol ( 6) foram identificados por métodos espectrais de RMN 1D e 2D e EM. Os compostos 1, 2 e 3 foram testados contra formas promastigotas de Leishmania brasiliensis e 1 e 2 também foram ensaiados contra a Escherichia coli, Pseudomonas aeruginosa e Bacillus subtilis e mostraram boa atividade.

In this work we are reporting the isolation by classical methods of chromatography of six polyketides from Penicillium herquei. The compounds citreorosein ( 1) , emodin ( 2) , janthinone ( 3) , citrinin ( 4) , citrinin H1 ( 5) and dicitrinol ( 6) were identified by spectral methods of 1D and 2D NMR and MS. Compounds 1, 2 and 3 were tested against promastigotes forms of Leishmania brasiliensis and 1 and 2 were also assayed against Escherichia coli, Pseudomonas aeruginosa and Bacillus subtilis and showed good activity.

Em estudos anteriores descrevemos o isolamento de nove substâncias, a partir das folhas de Derris urucu, uma espécie encontrada amplamente na floresta Amazônica, as quais foram identificadas como cinco estilbenos e quatro diidroflavonóis. Neste trabalho, três desses diidroflavonóis [urucuol A (1), urucuol B (2) e isotirumalina (3)] foram avaliados para identificar seus potenciais como aleloquímicos. Estamos relatando também, o isolamento e a determinação estrutural de um novo flavonóide [5,3′-diidroxi-4′-metoxi-(7,6:5″,6″)-2″,2″-dimetilpiranoflavanona (4)]. Investigamos os efeitos dos diidroflavonóis 1-3 sobre a germinação de sementes e desenvolvimento da radícula e do hipocótilo da planta daninha Mimosa pudica, usando soluções a 150 mg.L–1. Urucuol B, isoladamente, foi quem apresentou o maior potencial para inibir a germinação de sementes (26%), por sua vez, isotirulamina exibiu maior capacidade para reduzir o desenvolvimento do hipocótilo (25%), porém nenhuma das três substâncias mostrou potencial para inibir o desenvolvimento da radícula. Quando combinadas aos pares, as substâncias mostraram sinergismo ao desenvolvimento da radícula e do hipocótilo e efeitos, na germinação de sementes, que poderiam ser atribuídos a antagonismo. Quando testadas separadamente, as substâncias apresentaram maior tendência para inibir a germinação de sementes, enquanto que, quando testadas aos pares, observou-se aumento no efeito de inibição do desenvolvimento da planta daninha.

In some previous studies, we described the isolation of nine compounds from leaves of Derris urucu, a species found widely in the Amazon rainforest, identified as five stilbenes and four dihydroflavonols. In this work, three of these dihydroflavonols [urucuol A (1), urucuol B (2) and isotirumalin (3)] were evaluated to identify their potential as allelochemicals, and we are also reporting the isolation and structural determination of a new flavonoid [5,3′-dihydroxy-4′-methoxy-(7,6:5″,6″)-2″,2″-dimethylpyranoflavanone (4)]. We investigated the effects of the dihydroflavonols 1-3 on seed germination and radicle and hypocotyl growth of the weed Mimosa pudica, using solutions at 150 mg.L–1. Urucuol B, alone, was the substance with the greatest potential to inhibit seed germination (26%), while isotirumalin showed greater ability to reduce the development of the hypocotyl (25%), but none of the three substances showed the potential to inhibit radicle. When combined in pairs, the substances showed synergism for the development of root and hypocotyl and effects on seed germination that could be attributed to antagonism. When tested separately, the trend has become more intense effects on seed germination, while for the substances tested in pairs, the intensity of the effect was greater on development of weed.

Neste trabalho estamos reportando o isolamento de cinco compostos de Aspergillus sp EJC08 isolado como endofítico de Bauhinia guianensis uma planta típica da Amazônia. Os compostos ergosterol (1), peróxido de ergosterol (2), mevalolactona (3), monometilsulocrina (4) e tripacidina A (5) foram isolados através de procedimentos cromatográficos e identificados por métodos espectrais de RMN 1D e 2D e EM. Os compostos 3, 4 e 5 foram testados contra Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis e Staphylococcus aureus e mostraram boa atividade.

The compounds ergosterol (1), ergosterol peroxide (2), mevalolactone (3), monomethylsulochrin (4) and trypacidin A (5) were isolated by chromatographic procedures and identified by spectral methods of 1D and 2D NMR and MS. Compounds 3, 4 and 5 were tested against Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis and Staphylococcus aureus and showed good activity.

O fungo Aspergillus flavus isolado como endofítico da planta Paspalum maritimum Trin. foi avaliado quanto ao seu potencial de aplicação em reações de biotransformações. Os compostos chalcona (1), 3,4,5-trimetoxichalcona (2) e 2,3,4,4'-tetrametoxichalcona (3) foram biotransformados, respectivamente, na diidrochalcona (4), 3,4,5-trimetoxidiidrochalcona (5) e 2,3,4,4'-tetrametoxidiidrochalcona (6). As estruturas destes compostos foram determinadas por análises de RMN uni e bidimensionais e por espectrometria de massas. As diidrochalconas 5 e 6 são substâncias inéditas.

The fungus Aspergillus flavus isolated as endophytic of the plant Paspalum maritimum Trin. was evaluated for its potential application in biotransformation reactions. The compounds chalcone (1), 3,4,5-trimethoxychalcone (2) and 2,3,4,4'-tetramethoxychalcone (3) were biotransformed, respectively, in dihydrochalcone (4), 3,4,5-trimethoxydihydrochalcone (5) and 2,3,4,4'-tetramethoxydihydrochalcone (6). The structures were elucidated by spectroscopic methods including 1D and 2D NMR techniques, and MS analysis. The dihydrochalcones 5 and 6 are new compounds.

Um novo derivado de antraquinona, denominado guepinone (1), juntamente com as conhecidas substâncias isossulocrina (2) e cloroissosulocrina (3) foram isolados de uma cultura em arroz de Pestalotiopsis guepinii, um fungo endofitico de Virola michelii. Os compostos foram identificados pela análise de seus dados espectrométricos de RMN 1D e 2D e EM. A atividade antimicrobiana dos compostos isolados foi avaliada e a cloroisossulcrina (3) foi a mais ativa.

A new anthraquinone derivative, named guepinone (1), along with the known substances isosulochrin (2) and chloroisosulochrin (3), were isolated from a rice culture of Pestalotiopsis guepinii, an endophytic fungus of Virola michelii. The compounds were identified by analysis of 1D and 2D NMR and MS spectral data. The antimicrobial activity of these compounds was evaluated and chloroisosulchrin (3) was the most active.

A síntese e a estrutura cristalina por difração de raios-X de dois análogos de neolignanas, 2-(4-clorofenil)-1-feniletanona (20) e 2-[tio(4-clorofenil)]-1-(3,4-dimetoxifenil)propan-1-ona (12) são descritas. O composto 12 apresenta atividade intracelular contra Leishmania donovani e Leishmania amazonensis de amastigotas que causam a leishmaniose tegumentar e visceral. Além disso, a teoria do funcional de densidade (DFT) com o funcional híbrido B3LYP foi empregado para calcular um conjunto de descritores moleculares para dezenove análogos sintéticos de neolignanas com atividades antileishmaniose. Posteriormente, a análise discriminante stepwise foi realizada para investigar possíveis relações entre a estrutura molecular e atividades biológicas. Por meio dessa análise os compostos foram classificados em dois grupos ativos e inativos de acordo com seu grau de atividade biológica, e as propriedades mais importantes foram as cargas de alguns átomos, a afinidade eletrônica e o ClogP.

The synthesis and X-ray crystal diffraction structure of two analogues of neolignans, 2-(4-chlorophenyl)-1-phenylethanone (20) and 2-[(4-chlorophenyl)thio]-1-(3,4-dimethoxyphenyl)propan-1-one (12) is described. The compound 12 presents activity against intracellular Leishmania donovani and Leishmania amazonensis amastigotes that cause cutaneous and visceral leishmaniasis. In addition, the density functional theory (DFT) with the B3LYP hybrid functional was employed to calculate a set of molecular descriptors for nineteen synthetic analogues of neolignans with antileishmanial activities. Afterwards, the stepwise discriminant analysis was performed to investigate possible relationship between the molecular descriptors and biological activities. Through this analysis the compounds were classified into two groups active and inactive according to their degree of biological activities, and the more important properties were charges on some key atoms, electronic affinity and ClogP.

A biodiversidade Amazônica pode ser uma fonte de substâncias capazes de serem utilizadas no controle de plantas daninhas. Neste estudo relatamos o isolamento e a identificação de cinco estilbenos a partir das folhas do "timbó vermelho" (Deguelia rufescens var. urucu): 4-metoxilonchocarpeno (1); 3,5-dimetoxi-4´-hidroxi-3´-prenil-trans-estilbeno (2), lonchocarpeno (3), 3,5-dimetoxi-4´-O-prenil-trans-estilbeno (4) e pteroestilbeno (5). As substâncias 2 e 4 são novos produtos naturais, porém 2 já havia sido citada como produto de síntese. Foi avaliada a potencial atividade alelopática de 1, 2 e 4 sobre a germinação de sementes e o crescimento da planta daninha Mimosa pudica. Os efeitos observados sobre a germinação das sementes de M. pudica não variaram significantemente (p > 0,05) quando a análise da fitotoxidade foi realizada com as substâncias isoladamente, cuja inibição máxima não ultrapassou 20%. A inibição mais intensa, quanto ao desenvolvimento da radícula e do hipocótilo, foi encontrada para o composto 4 (p < 0,05). Isoladamente, 4 causou efeito inibitório significativamente maior (p < 0,05) no desenvolvimento da radícula e do hipocótilo, do que 1 e 2. Quando testados aos pares, apresentaram antagonismo para a germinação de sementes e sinergismo para o desenvolvimento da radícula e hipocótilo.

The Amazon biodiversity may provide plants whose chemical substances are capable of controlling weeds. In this study we report the isolation and identification of five stilbenes from the leaves of "timbó vermelho" (Deguelia rufescens var. urucu): 4-methoxylonchocarpene (1); 3,5-dimethoxy-4´-hydroxy-3´-prenyl-trans-stilbene (2), lonchocarpene (3), 3,5-dimethoxy-4´-O-prenyl-trans-stilbene (4) and pterostilbene (5). Compounds 2 and 4 are new natural products although 2 has been previously cited as synthesis product. Potential allelopathic activity for 1, 2 and 4 was evaluated over seed germination and plant growth of Mimosa pudica weed. The observed effects on seed germination did not vary significantly (p > 0.05) when the analysis of phytotoxicity was performed with the substances alone, the maximum inhibition did not exceed 20%. The most intense inhibitions on radicle and hypocotyl development were found for compound 4 (p < 0.05). When tested in pairs, showed antagonism for seed germination and synergism for radicle and hypocotyl development.

O presente trabalho trata da investigação química das folhas e caule da espécie Ouratea castaneifolia (DC.) Engl., sobre a qual não há registros de estudos químicos ou farmacológicos anteriores. O estudo fitoquímico clássico dos extratos orgânicos do caule e das folhas de O. castaneifolia foi aliado à técnica da cromatografia líquida de alta eficiência (CLAE) e resultou na identificação de dezessete metabólitos: sete triperpenos (friedelina, 3β-friedelinol, α-amirina, β-amirina, lupeol, taraxerol e germanicol), quatro esteróides (sitosterol, estigmasterol e os glucosídeos sitosteril 3-O-β-D-glicopiranosídeo e estigmasteril 3-O-β-D-glicopiranosídeo), uma isoflavona (5,7,4´-trimetoxiisoflavona), uma flavona (5,4´-diidroxi-7,3´,5´-trimetoxiflavona), quatro biflavonas (amentoflavona, 7,7"-O-dimetil-amentoflavona, heveaflavona e tetrametilamentoflavona). A identificação das substâncias foi feita com base na análise de espectros de RMN de ¹H, 13C e técnicas bidimensionais. As classes dos metabólitos identificados estão de acordo com aquelas citadas em estudos químicos do gênero Ouratea.

This paper presents the chemical investigation of the leaves and stems of Ouratea castaneifolia (DC.) Engl.. There are no chemical or pharmacological studies with this species. Classic phytochemical investigation of the organic extracts together with high pressure liquid chromatography (HPLC) procedures lead to the identification of seventeen metabolites: seven triterpenes (friedelin, 3β-friedelinol, α-amyrin, β-amyrin, lupeol, germanicol and taraxerol), four steroids (sitosterol, stigmasterol and the glycosides sitosteryl 3-O-β-D-glucopyranoside and stigmasteryl 3-O-β-D-glucopyranoside), one isoflavone (5,7,4'-trimethoxyisoflavone), one flavone (5,4'-dihydroxy-7,3',5'-trimethoxyflavone) and four biflavones (amenthoflavone, 7,7"-O-dimethylamenthoflavone, heveaflavone and tetramethylamenthoflavone). The structures of the compounds were established by the analysis of ¹H, 13C NMR spectra including bidimensional techniques. The classes of the identified metabolites are in agreement with previous studies of the Ouratea genus.

Derris urucu é uma planta da Amazônia com propriedades inseticida e ictiotóxica. Estudos com esta espécie reportam a presença de flavonóides, principalmente rotenóides, bem como de estilbenos. A partir do extrato etanólico das folhas de Derris urucu (Leguminosae), três novos diidroflavonóides, denominados urucuol A (1), B (2) e C (3) e o diidroflavonol isotirumalina (4), foram isolados e identificados. As estruturas destes compostos foram elucidadas por uma extensiva análise espectroscópica de RMN uni e bidimensional, UV, IV e dados de EM, além de comparação com dados da literatura. Os compostos isolados (1-4) foram avaliados quanto ao seu potencial sequestrador do radical DPPH• e apresentaram baixo poder antioxidante quando comparados ao antioxidante comercial trans-resveratrol.

Derris urucu is an Amazonian plant with insecticide and ichthyotoxic properties. Studies with this species show the presence of flavonoids, mainly rotenoids, as well as stilbenes. The ethanol extract of the leaves of Derris urucu (Leguminosae) afforded three new dihydroflavonols named urucuol A (1), B (2) and C (3), and the dihydroflavonol isotirumalin (4). Their structures were elucidated by extensive analysis of 1D and 2D NMR, UV and IR spectra and MS data and comparison with literature data. The isolated compounds (1-4) were evaluated for DPPH• radical scavenging activity and showed a relatively lower antioxidant ability compared to the commercial antioxidant trans-resveratrol.

O fracionamento do extrato hexânico do caule de um espécime de reflorestamento de Tectona grandis (Verbenaceae), através de procedimentos fitoquímicos clássicos, levou ao isolamento das naftoquinonas lapachol e desidro-a-lapachona e das antraquinonas tectoquinona e obtusifolina. As estruturas das substâncias foram caracterizadas através da análise de métodos espectrométricos de RMN. Este é o primeiro estudo fitoquímico de um espécime de reflorestamento de Tectona grandis, no Brasil, sendo o objetivo principal deste trabalho a comprovação da presença de tectoquinona em espécimes cultivados.

The hexane extract of the bark of Tectona grandis (Verbenaceae) afforded two anthraquinones and two naphtoquinones. Their caracterizations were obtained through NMR spectroscopic techniques. This is the first phytochemical study of the bark of Tectona grandis reforestation specimen in Brazil. The main interest in this work is proving the presence of tectoquinone in reforestation specimen.

O fungo Penicillium janthinellum, obtido de Melia azedarach, produziu ergosterol, ergosterol 5alfa,8alfa-peróxido e uma mistura de esteróides C25 epiméricos de ocorrência restrita na natureza. Os esteróides C25, denominados neociclocitrinóis, possuem o mesmo sistema tetracíclico de anéis presente no ciclocitrinol, o qual foi isolado do fungo Penicillium citrinum, associado a uma esponja, com o mesmo esqueleto biciclo[4:4:1] nos anéis A/B, mas com cadeias laterais diferentes. O P. janthinellum isolado de M. azedarach, foi cultivado sobre milho branco e os três esteróides foram isolados por vários procedimentos cromatográficos e identificados por cuidadosa análise dos dados de RMN, principalmente correlações ¹H - 13C em duas dimensões bem como COSY e TOCSY ¹H - ¹H. A origem biossintética dos ciclocitrinóis é discutida.

A plant-derived fungus, Penicillium janthinellum, obtained from Melia azedarach, produced ergosterol and ergosterol 5alpha,8alpha-peroxide along with a mixture of rare C25 steroid epimers. The C25 steroids, named neocyclocitrinols, shows exactly the same tetracyclic ring system present in cyclocitrinol, which was isolated from a sponge-derived Penicillium citrinum, with the same bicyclo [4:4:1] skeleton at A/B rings, but showing different side chains. The P. janthinellum was cultivated over white corn and the three steroids were isolated by several silica gel based chromatographic procedures and identified by extensive NMR methods, mainly ¹H - 13C correlations and ¹H - ¹H COSY and TOCSY. The biosynthetic origin of the cyclocitrinols is also discussed.

Penicillium janthinellum, isolado como um fungo endofítico dos frutos de Melia azedarach, foi cultivado por 20 dias em milho branco triturado e autoclavado, onde os policetídeos conhecidos citrinina, emodina, 1,6,8-triidróxi-3-hidroximetilantraquinona, e uma nova antraquinona modificada, denominada janthinona, foram produzidos e isolados por procedimentos cromatográficos clássicos e identificados por extensivos estudos espectroscópicos, principalmente RMN 1D e 2D e EM. Essas substâncias foram ensaiadas contra diversas bactérias. Citrinina foi ensaiada pela primeira vez contra Leishmania e inibiu 100% o crescimento de cepas depois de 48h a uma concentração de 40 mig mL-1.

Penicillium janthinellum, isolated as an endophytic fungus from fruits of Melia azedarach, was cultivated over 20 days on ground and autoclaved white corn, where the known polyketides citrinin, emodin, 1,6,8-trihydroxy-3-hydroxymethylanthraquinone, and a new modified anthraquinone, named janthinone, were produced and isolated by classical chromatographic procedures and identified by MS and 1D and 2D NMR spectroscopic data. The antibacterial properties of these polyketides were investigated. Citrinin inhibited 100% of Leishmania growth after 48h at a concentration of 40 mug mL-1.
