RESUMO Objetivou-se testar a vitrificação de ovários de camundongos do ICTB/Fiocruz. Inicialmente, fez-se coleta e maturação in vitro dos oócitos de ovários a fresco e vitrificados, bem como avaliação de estruturas no cultivo embrionário, pós-fertilização in vitro. Fêmeas B6D2F1 foram eutanasiadas para remoção dos ovários (n=60) e divididas em três grupos: grupo 1 (n=30 animais) - oócito de ovários vitrificados, maturados e fertilizados in vitro (120 fragmentos); grupo 2 (n=15) (controle 1) - oócitos coletados a fresco, maturados e fertilizados in vitro; e grupo 3 (n=15) (controle 2) - oócitos maturados in vivo e fertilizados in vitro. A técnica foi verificada no desenvolvimento embrionário in vitro, que foi avaliado pelo teste de qui-quadrado (BioStat 5.0). Recuperaram-se 123, 224 e 328 oócitos nos G1, G2 e G3, respectivamente. Observaram-se diferenças significativas nas taxas de clivagem às 24 horas (embriões ≥ 2 células) entre G1 (8%) e G2 (32%) (P<0,1) e G1 e G3 (49%) (P<0,05), mas não entre G2 e G3 (P>0,05). Para blastocistos, às 96 horas, os grupos G1, G2 e G3 apresentaram, respectivamente, 6%, 11% e 46%, diferindo significativamente entre eles (P<0,05). A vitrificação de ovários, a maturação oocitária e a fertilização in vitro são alternativas para a produção de embriões de camundongos in vitro.

ABSTRACT This work aimed test ovarian vitrification of hybrid mouse from ICTB/Fiocruz. Protocol collection and oocyte in vitro maturation from fresh and vitrified ovaries was established and embryos were evaluated after fertilization. B6D2F1 females were euthanized for ovarian removal (n= 60) and divided into 3 groups: G1 (n= 30) - ovaries fragmented (n= 120), vitrified, matured and fertilized; G2 (n= 15) - in vitro fertilization of oocytes matured in vitro from fresh ovaries; G3 (n= 15) - ampulla region oocytes in vitro fertilizated. Viability was verified by thawing, oocyte in vitro maturation and fertilization. In vitro embryo development of each group was evaluated by Chi-square test (BioStat 5.0). 123, 224 and 328 oocytes were recovered from G1, G2 and G3, respectively. Significant differences were observed in cleavage rates at 24 hours (embryos with 2 cells or more) between G1 (8%) and G2 (32%) (P< 0.1) and G1 and G3 (49%) (P< 0.05) but not between G2 and G3 (P> 0.05). Blastocysts at 96 hours presented 6%, 11% and 46%, respectively for G1, G2 and G3, differing significantly (P< 0.05). Ovary vitrification, oocyte in vitro maturation and in vitro fertilization were available for the production of in vitro mouse embryos.

RESUMO O presente estudo avaliou o perfil hormonal e a expressão gênica de receptores de prostaglandina F2α (PGF2α), ocitocina e estrógeno no endométrio de vacas pós-parto tratadas com cloprostenol. Vinte vacas mestiças Holandês-Zebu foram tratadas com solução salina (tratamento CONT, n = 10) ou cloprostenol (tratamento CLO, n = 10), ambos administrados dois e cinco dias após o parto. Amostras de sangue foram coletadas nos dias dois, sete, 14, 21 e 28 pós-parto para mensuração de progesterona, de metabólito de PGF2α (PGFM) e de estradiol, e foram obtidas biópsias endometriais para quantificar a expressão de PTGFR, OXTR e ESR1. No tratamento CLO, a expressão gênica de receptores de ocitocina foi menor (P<0,05). As concentrações de estrógeno aumentaram progressivamente até o dia 14 (P<0,05). A maior expressão de OXTR foi observada no dia 14 (P<0,05). A expressão de ESR1 foi semelhante entre os tratamentos (P>0,05). Os níveis de PGFM foram altos durante todo o estudo. Conclui-se que a administração de cloprostenol nos dias dois e cinco pós-parto parece diminuir a expressão de OXTR no endométrio em vacas mestiças.

ABSTRACT The present study investigated the hormonal profile and expression of prostaglandin F2α (PGF2α), oxytocin and estrogen receptors in uterine tissues of postpartum cows treated with cloprostenol. Twenty Holstein-Zebu crossbred cows were treated with saline solution (treatment CONT) or cloprostenol (treatment CLO), both administered two and five days postpartum. Blood samples were collected on days two, seven, 14, 21 and 28 postpartum for progesterone, PGF2α metabolite (PGFM) and estradiol determination, and endometrial biopsy was performed in order to quantify the expression of oxytocin receptor (OXTR), prostaglandin F receptor (PTGFR) and estrogen receptor 1 (ERS1) genes. In the CLO treatment, expression of OXTR was reduced (P<0.05) but no difference (P>0.05) between treatments was found for PTGFR and ERS1 expression. Estrogen concentrations increased progressively until day 14 (P<0.05) and the highest OXTR expression and lowest PTGFR expression were observed on day 14 (P<0.05) in both treatments. Serum PGFM concentrations were high throughout the experiment. In conclusion, cloprostenol administration at days two and five of postpartum seems to reduce OXTR expression in the endometrium in crossbred cows.

RESUMO Este estudo avaliou a integridade da membrana plasmática, da membrana acrossomal e o potencial da membrana mitocondrial de espermatozoides de touros da raça Nelore da puberdade até à maturidade sexual e suas correlações com motilidade, vigor, turbilhão dos espermatozoides, circunferência escrotal e testosterona. Sessenta touros da raça Nelore, com idade entre 18 e 19 meses, foram divididos em quatro lotes (n=15 touros / lote) e foram avaliados por 280 dias. Coletaram-se as amostras de sêmen a cada 56 dias por eletroejaculação e, logo após, foram determinados a motilidade, o vigor e o turbilhão dos espermatozoides por microscopia óptica. A integridade da membrana de esperma, da membrana acrosomal e da atividade mitocondrial foi avaliada por microscopia de fluorescência, utilizando-se associação das sondas (FITC-PSA, PI, JC-1, H342). Os espermatozoides foram classificados em oito categorias de integridade, dependendo se eles exibiram membranas plasmática e mitocondrial intactas ou danificadas e potencial mitocondrial elevado ou baixo. Os resultados mostram que os touros têm uma baixa quantidade de espermatozoides com membranas íntegras na puberdade, com baixa motilidade, vigor e turbilhão. No entanto, nos touros na maturidade sexual precoce, a integridade da membrana dos espermatozoides aumentou significativamente. A taxa de dano à membrana espermática foi negativamente correlacionada com a motilidade, o vigor, o movimento das ondas e a concentração de testosterona nos touros, e uma correlação positiva existiu entre a integridade da membrana plasmática e a circunferência escrotal. A integridade da membrana acrossomal não foi influenciada pela puberdade.

ABSTRACT This study evaluated the plasma membrane integrity, acrosomal membrane integrity, and mitochondrial membrane potential of Nelore bull sperm from early puberty to early sexual maturity and their associations with sperm motility and vigor, the mass motility of the spermatozoa (wave motion), scrotal circumference, and testosterone. Sixty Nelore bulls aged 18 to 19 months were divided into four lots (n=15 bulls/lot) and evaluated over 280 days. Semen samples, collected every 56 days by electroejaculation, were evaluated soon after collection for motility, vigor and wave motion under an optical microscope. Sperm membrane integrity, acrosomal integrity, and mitochondrial activity were evaluated under a fluorescent microscope using probe association (FITC-PSA, PI, JC-1, H342). The sperm were classified into eight integrity categories depending on whether they exhibited intact or damaged membranes, an intact or damaged acrosomal membrane, and high or low mitochondrial potential. The results show that bulls have a low amount of sperm with intact membranes at puberty, and the sperm show low motility, vigor, and wave motion; however, in bulls at early sexual maturity, the integrity of the sperm membrane increased significantly. The rate of sperm membrane damage was negatively correlated with motility, vigor, wave motion, and testosterone in the bulls, and a positive correlation existed between sperm plasma membrane integrity and scrotal circumference. The integrity of the acrosomal membrane was not influenced by puberty. During puberty and into early sexual maturity, bulls show low sperm mitochondrial potential, but when bulls reached sexual maturity, high membrane integrity with high mitochondrial potential was evident.

A coloração pela prata das regiões organizadoras de nucléolos (NORs) é caracterizada por marcar proteínas ligadas ao ácido ribonucleico ribossômico, avaliando a proliferação em células normais ou neoplásicas. Objetivou-se estudar, em testículos de ovinos obtidos em matadouro, a validade do uso da técnica de coloração pela prata (AgNOR) na identificação das regiões organizadoras de nucléolo (NORs) em células saudáveis da linhagem espermatogênica. Utilizaram-se 43 pares de testículos de ovinos mestiços entre seis e 10 meses de idade. Testes de Wilcoxon e Spearman foram empregados, com nível de 5%. As médias das NORs nas células das gônadas direita e esquerda foram, respectivamente: espermatogônia (8,77±1,14 e 9,04±0,96), espermatócitos (4,99±2,00 e 6,20±2,07; P<0,05), Leydig (8,05±2,82 e 7,89±2,29) e Sertoli (8,07±1,88 e 7,61±2,16; P<0,05). Houve correlação (P<0,05) entre os lados para o número de NORs: espermatócitos x Leydig (0,49); espermatócitos x Sertoli (0,49) e Leydig x Sertoli (0,96). Conclui-se ser válido o emprego da técnica AgNOR para avaliar o potencial proliferativo das células saudáveis em testículos de ovinos com prática execução e baixo custo.

The silver staining technique for AgNOR nucleolar organizer regions (NORs) is characterized by marking proteins linked to the ribosomal ribonucleic acid, evaluating cell proliferation. The aim was to study the validity of the AgNOR staining technique in the testicular cell proliferation in crossbred ovine. Forty-three pairs of ovine testicles between 6 and 10 months old were collected. Wilcoxon and Spearman tests were used with a significance level of 5%. The mean NORs count in cells of the right and left gonads were respectively: spermatogonia (8.77±1.14 and 9.04±0.96), spermatocytes (4.99±2.00 and 6.20±2.07, P<0.05), Leydig (8.05±2.82 and 7.89±2.29) and Sertoli cells (8.07±1.88 and 7.61±2.16; P<0.05). There was a correlation between the mean values for the right and left sides for the number of NORs (P<0.05) between Leydig x spermatocytes (0.49); spermatocytes x Sertoli (0.49) and Sertoli x Leydig (0.96). The study demonstrates that the AgNOR staining technique is indicated to evaluate the cell proliferative potential in ovine testis with practical implementation and low cost.

Estudou-se a influência das quatro estações do ano nas características do sêmen e nas concentrações de testosterona e cortisol em touros. Cinco touros Nelore e cinco Simental entre 48 e 72 meses de idade, criados extensivamente, foram avaliados andrologicamente por meio de exames físicos das características morfológicas do sêmen e das concentrações séricas de testosterona e cortisol. Houve redução na motilidade e no vigor do sêmen no inverno (P<0,05) na raça Simental. Observou-se correlação (P<0,01) entre testosterona x motilidade (0,69) e testosterona x vigor (0,57) na raça Simental, e cortisol x motilidade (0,68) e cortisol x vigor (0,65) na raça Nelore. O efeito das estações do ano modificou a qualidade do sêmen com aumento da motilidade e vigor espermáticos na primavera e verão nos touros Simental. A concentração de cortisol diminuiu no outono nos touros Nelore.

The influence of four seasons per year on semen characteristics and levels of testosterone and cortisol in Nelore and Simental bulls was studied. Five Nelore and five Simental bulls extensively managed, at 48-72 months old were evaluated for sexual soundness using physical and morphological characteristics of semen and serum levels of testosterone and cortisol. In Simental bulls there was decreased motility and vigor of semen (P<0.05) during winter. There was a correlation (P<0.01) between testosterone x motility (0.69) and testosterone x vigor (0.57) in the Simental breed and cortisol x motility (0.68) and cortisol x vigor (0.65) in Nelore the breed. The effect of seasons during the year changed the semen quality with increased sperm motility and vigor in spring-summe r in Simental bulls. The cortisol level decreased in autumn in Nelore bulls.

Avaliou-se o efeito do citrato em meio CR2aa suplementado com soro fetal bovino (SFB) ou livre de proteínas séricas e sua associação com taurina no desenvolvimento de embriões bovinos fecundados in vitro. Embriões foram cultivados em CR2aa contendo 0, 0,5, 1,0 e 3,0mM citrato, suplementado com 10% SFB (experimento 1) ou com álcool polivinil (PVA; experimento 2). No terceiro experimento, embriões foram cultivados em meio com 0,5mM citrato, ou 7mM taurina, ou com a associação de ambos, suplementado com SFB. Os cultivos foram realizados com células do cumulus em ambiente a 38,8ºC com 5% de CO2 em ar atmosférico. Melhora no desenvolvimento embrionário foi observado no cultivo de embriões em CR2aa com 0,5 e 1,0mM citrato na ausência de SFB (P<0,05), 8,6% e 11,3% de blastocistos, respectivamente, porém com valores mais baixos (P<0,05) que embriões cultivados em CR2aa com SFB (31,9%). Associação de citrato com taurina em meio com SFB não influenciou (P>0,05) a produção de embriões ou o número de células. Citrato em meio CR2aa pode ser uma alternativa para cultivo embrionário em condições atmosféricas com 5% de CO2 em ar na ausência de proteína sérica.

The effect of citrate added to CR2aa medium supplemented with fetal calf serum (FCS) or serum-proteinfree and its association with taurine on the development of in vitro-fertilized bovine embryos was evaluated. Embryos were cultured with 0, 0.5, 1.0, and 3.0mM citrate, in CR2aa supplemented with 10% FCS (experiment 1), or polyvinyl alcohol (PVA; experiment 2). In experiment 3, embryos were cultured with 0.5mM citrate, 7.0mM taurine or with association of both, in medium supplemented with FCS. Embryo culture was performed with cumulus cells at 38.8ºC in 5% CO2 under air for all experiments. Positive effect on embryo development was only observed with 0.5 and 1.0mM citrate in FCS-free CR2aa (P<0.05; 8.6% and 11.3% blastocyst, respectively), however with lower embryo rate than CR2aa with FCS (31.9%). Association between citrate and taurine in medium supplemented with FCS did not affect (P>0.05) embryo rate nor total cell number. Citrate in CR2aa medium can be an alternative for serumfree embryo culture under 5% CO2 in air, absence of serum protein.

Determinaram-se as características andrológicas de touros da raça Gir, classificando-os quanto ao seu potencial reprodutivo, utilizando-se o sistema de classificação andrológica por pontos (CAP). Os animais foram separados em três grupos: G1= animais de 18-24 meses (n=33), G2 = 25-31 meses (n=24) e G3 = 32-38 meses (n=12). A média da circunferência escrotal no G1 foi menor (P<0,05) do que nos G2 e G3. A motilidade (%) e o vigor não diferiram entre os grupos. As taxas (%) de defeitos maiores e totais foram maiores (P<0,05) no G1 do que nos G2 e G3. Para as características andrológicas não houve diferenças entre G2 e G3. Com relação ao CAP, G1, G2 e G3 apresentaram, respectivamente, 25,7%, 58,3% e 61,5% de animais aptos à reprodução; 11,4%, 20,8% e 15,4% questionáveis e 57,1%, 12,5% e 15,4% inaptos ou imaturos. A idade dos touros Gir influenciou a circunferência escrotal e os defeitos maiores e totais, mas não a motilidade e o vigor. Maior proporção de animais aptos à reprodução ocorreu após 24 meses de idade.

Gyr bulls were ranked regarding their reproductive potential, following evaluations through the score-based andrological classification system (ACP). For the examinations, three animal age groups were considered: 18 to 24 month-old (G1, n=33), 25 to 31 month-old (G2, n=24), and 32 to 38 month-old (G3, n=12). Scrotal circumference was lower in G1, as compared to G2 and G3 (P<0.05). Percent motility and vigor were not affected by the age groups. Percent rates of major and total defects were higher (P<0.05) in G1 than in G2 or G3. No significant differences were observed when G2 and G3 were compared. Regarding ACP data, G1, G2 and G3 yielded, respectively, 25.7, 58.3 and 61.5% of adequate breeders; 11.4, 20.8 and 15.4% of bulls with undefined breeding capacity, and 57.1, 12.5 and 15.4% of inapt breeders or immature animals. Age group affected scrotal circumference and major and total defects, but did not influence motility or vigor. The highest proportion of adequate breeders was obtained with 24-month-old bulls and older.

Avaliaram-se os efeitos de dois protocolos de punção folicular na quantidade/qualidade dos oócitos e na produção in vitro de embriões, em vacas da raça Gir, não-lactantes. O ciclo estral foi sincronizado com cloprostenol e ao longo do experimento os animais receberam implantes auriculares de norgestomet, renovados a cada 14 dias. Os animais foram submetidos aos protocolos I (sem estimulação hormonal, com punção folicular duas vezes/semana) e II (pré-tratamento com 250 UI de FSH em doses decrescentes, três dias antes da punção folicular). Os oócitos recuperados foram levados ao laboratório em meio TALP-Hepes e submetidos à maturação. Na fecundação in vitro utilizou-se sêmen de touro Gir, previamente capacitado. Após 22 horas de fecundação, os prováveis zigotos foram co-cultivados com células da granulosa em CR2aa acrescido de 10% de soro fetal bovino. A taxa de clivagem foi avaliada 72 horas pós-fecundação e a de blastocisto 192 horas pós-fecundação. O número total de folículos foi maior (P<0,05) no protocolo II, assim como o número de folículos grandes e médios (P<0,05) e o diâmetro do maior folículo (P<0,05), sendo o número de folículos pequenos (P<0,05) menor. O total de oócitos recuperados por sessão não foi diferente entre os protocolos I e II (P>0,05). O número de oócitos de grau I e a taxa de clivagem foram maiores (P<0,05) e o número de degenerados menor protocolo II. A pré-estimulação ovariana com FSH pode melhorar a qualidade e a taxa de clivagem dos oócitos recuperados por punção folicular em animais Gir.

The effect of two ovum pick-up protocols on amount/quality of oocytes retrived and on in vitro embryo production of Gyr cows was evaluated in non-lactating cycling cows with good body and reproductive conditions. Estrous cycles were synchronized with cloprostenol. During the experiment, animals received norgestomet ear implants, replaced every 14 days. Animals were submitted to protocol I (without hormonal estimulation) and protocol II (pre-stimulation with 250 IU of FSH three days before aspiration). Recovered oocytes were transported in Talp-Hepes to the laboratory, classified and maturated. Twenty two hours after fertilization, presumptive zygotes were co-culture with granulosa cells in CR2aa supplemented with 10% of fetal calf serum. Cleavage rate was assessed at 72 hours post-fertilization, and blastocyst production 192 hours post-fertilization. The total number of follicles, the number of large and medium follicles and the diameter of the largest follicle were higher (P<0.05) in protocol II, and the number of small follicles (P<0.05) was lower than in protocol I. No difference between protocols I and II was observed for oocytes recovered per aspiration section (P>0.05). The number of grade I oocytes and cleavage rate were higher (P<0.05) in protocol II and the number of degenerated oocytes was lower (P<0.05). The pre-stimulation with FSH can improve the quality and cleavage rate of oocytes recovered by ovum pick-up in zebu animals.

Estudou-se o efeito da concentração espermática e período de incubação e da interação dessas características sobre a fecundação in vitro (FIV) usando-se sêmen de touros Guzerá. Oócitos (n=1146) maturados in vitro foram divididos em tratamentos objetivando a FIV, em esquema fatorial 3×2×2 (três touros - A, B e C, duas concentrações espermáticas - 2 e 4×10(6) espermatozóides/ml e dois tempos de incubação 12 e 18 horas). Utilizaram-se espermatozóides viáveis obtidos por swin-up. A FIV foi realizada em meio fert-talp com heparina, em incubadora com 5% de CO2 e 95% de umidade, a 38,5°C. Após incubação, 50% dos oócitos foram fixados e corados para determinação das taxas de penetração, fecundação monoespermática e poliespermia. O restante foi co-cultivado com células da granulosa em meio CR2aa por oito dias, avaliando-se a taxa de clivagem e a produção de blastocisto. Houve maior taxa de penetração (P<0,05) na concentração de 4×10(6) espermatozóides/ml por 18 horas (64%), e não se observou diferença (P>0,05) entre os demais tratamentos. A taxa de poliespermia foi maior (P<0,05) na concentração espermática de 4×10(6), independente do tempo de incubação. Na concentração espermática mais alta, a taxa de poliespermia foi maior no tempo de incubação de 18 horas (P<0,05). O touro A apresentou menor (P<0,05) taxa de poliespermia em relação aos demais. Ainda, no touro A a taxa de clivagem foi maior (P<0,05) que no touro B, enquanto o C mostrou-se semelhante tanto ao A quanto ao B. O tempo de incubação, a concentração espermática e a interação das variáveis influenciaram as taxas de penetração e poliespermia, sem interferir na taxa de fecundação monoespermática e na produção de blastocistos.

The effects of sperm concentration, incubation period and their interaction on in vitro fertilization (IVF) using sperm of Guzera bulls were evaluated. In vitro matured oocytes (n=1146) were allotted to IVF treatments in a factorial scheme 3×2×2 - three bulls (A, B and C), two sperm concentrations (2 and 4×10(6) spermatozoa/ml) and two incubation periods (12 and 18h). Viable spermatozoa were obtained by swim-up. The IVF was performed using in Fert-Talp medium with heparin, on incubator with 5% CO2 and 95% humidity, at 38.5ºC. After the incubation, 50% of oocytes were fixed and stained to determine penetration, monospermic fecundation and polyspermy rates. The remainder was co-cultured with granulosa cells in CR2 medium for eight days to evaluate cleavage and embryo production rates. The penetration rate was higher (P<0.05) for sperm concentration of 4×10(6) spermatozoa/ml incubated for 18 hours (64%), and no differences (P>0.05) among remainder treatments were observed. The polyspermic rate (P<0.05) was higher for higher sperm concentration and incubation period. Bull A showed the lowest (P<0.05) polyspermy rate. Bulls A and C showed higher (P<0.05) cleavage than bull B. Incubation period, sperm concentration, and interaction of these two factors influenced penetration and polyspermy rates, without any effect on monospermic fecundation rates and embryos production.

Estudaram-se os efeitos da concentração espermática e do tempo de incubação do sêmen com ovócitos, durante a fecundação in vitro, sobre as taxas de penetração espermática, de fecundação monoespermática e de clivagem, utilizando-se sêmen de touros da raça Gir. Ovócitos (n=817) maturados in vitro foram distribuídos em tratamentos visando à fecundação in vitro (FIV), em um delineamento fatorial 2×2×2, com duas concentrações espermáticas (2 e 4×10(6) espermatozóides/ml), dois períodos de incubação (12 e 18h) e dois touros (A e B). Espermatozóides viáveis foram obtidos pela técnica de swin-up. A FIV foi realizada em meio fert-talp com heparina, em incubadora com 5% de CO2 em ar atmosférico e 95% de umidade, a 39ºC. Após inseminação, 359 ovócitos foram fixados e corados para determinação das taxa de penetração e poliespermia. O restante foi co-cultivado com células da tuba uterina e TCM-199 por 72h, avaliando-se a clivagem. As taxas de penetração, fecundação monoespermática e clivagem não foram influenciadas (P>0,05) pela concentração espermática e pelo período de incubação. O touro B produziu maiores taxas (P<0,05) de penetração e de clivagem (83,3 e 81,0%, respectivamente) do que o touro A (66,5 e 64,0%). Houve tendência do touro B apresentar maior taxa de polispermia (P=0,067) do que o touro A (16,4 e 6,2 %, respectivamente). As interações entre tratamentos não foram significativas (P>0,05). O efeito touro sobre a capacidade de fecundação dos espermatozóides deve ser considerado quando da fecundação in vitro.

The aim of this study was to evaluate the effects of differents sperm concentration and incubation time during in vitro fecundation on sperm penetration, monospermic fecundation and cleavage rates, using sperm of Gyr bulls. In vitro matured oocytes (n=817) were distributed into treatments to in vitro fertilization (IVF) in a factorial design, with two sperm concentration (2 and 4×10(6) spermatozoa/ml), two incubation time (12 and 18h) and two bulls (A and B). Viable spermatozoa were obtained by swin-up. The IVF was realized in Fert-Talp medium with heparin, on incubator with 5% CO2 and 95% humidity in air, at 39ºC. After insemination, 359 oocytes were fixed and stained to determine penetration and polyspermy rates. The remainder were co-cultivated in bovine oviduct epithelial cells and TCM-199, for 72h, to evaluate cleavage rate. The penetration, monospermic fecundation, and cleavage rates were not affected (P>0.05) by sperm concentration and incubation time. Higher (P<0.05) penetration and cleavage rates were observed for bull B (83.3 and 81.8%, respectively) than for bull A (66.3 and 64%, respectively). Bull B showed a tendency to present higher (P<0.07) polyspermy than bull A (16.4 and 6.25, respectively). No interactions among treatments were observed.

Utilizaram-se quinze vacas da raça Holandesa, submetidas a três tratamentos: T1-controle, T2-monensina sódica 300 mg/animal/dia, T3-monensina sódica 225 mg/animal/dia. A produção de leite, consumo de ração e o peso dos animais foram mensurados, juntamente com a avaliação da condição corporal Após o período experimental de 112 dias, evidenciou-se aumento na produção de leite de 13,5% e 10% (T2) em relação a TI e T3 respectivamente. Houve redução de consumo de ração de 4,5% (T2) e 3% (T3) em relação ao controle.

Milk and feed consumption were monitored in fifteen Holstein cows divided in 3 groups: T1 (control), T2 (sodium monensin - 300 mg/head) T3 (sodium monensin - 225 mg/head). After an experimental period of 112 days, there were 13.5% and 10% (T2) increase in milk production as compared to the control and T3 respectively, and also a feed intake reduction 4.5% (T2) and 3% (T3) as compared to the control.
