Resumo Alguns aspectos comparativos entre hemoglobinas de duas espécies de peixes do rio Amazonas foram analisadas, com a finalidade de estabelecer as bases moleculares de suas propriedades funcionais, bem como, as interações entre estas proteínas com outras moléculas e íons. As espécies estudadas possuem diferentes características fisiológicas de respiração, atividade motora e meio ecológico. A hemoglobina de aruanã por eletroforese em poliacrilamida apresentou um componente único com pronunciado efeito Bohr e Root. Os valores de "n" na equação de Hill, mostram variações apreciáveis com o pH, tanto na presença como na ausência de ATP (10-3M). As hemoglobinas de acari-bodó no hemolisado total não apresentaram efeito Bohr, contudo os quatro componentes isolados mostraram propriedades funcionais diferentes. O primeiro Hbl, mostrou pequeno efeito Bohr reverso e uma acentuada influência de ATP (10-3M) em pH baixo, sobre este efeito tornando-se normal, enquanto que os três outros componentes apresentaram efeito Bohr normal e nenhuma influência de ATP sobre ele. Nossos resultados mostram claramente a capacidade de adaptação molecular das hemoglobinas destes peixes às condições fisiológicas e ambientais a que eles estão sujeitos.

Summary Two species of fish hemoglobin from Amazon river were analized in order to establish the relationship between their molecular and physiological properties as well as their interactions with other molecules and ions. The species were different regarding respiration, activity and habitat. The polyacrylamide eletrophoresis pattern of aruanã hemoglobin showed only one component and pronounced Bohr and Root effect and the Hill coefficient showed significant variation with the pH in the presence or not of 10-3M ATP. The total hemolisate of acari-bodó presented not Bohr effect nevertheless the four isolated components showed different functional properties thus the Hbl component, showed small reverse Bohr effect that becomes normal in the presence of 10-3M ATP at low pH. The other three components presented normal Bohr effect and ATP does not change this effect. These results cleary show the capacity of molecular adaptation of fish hemoglobin to adapt to both the metabolic requirements and environment variations of the animals.

Resumo Têm sido estudados os efeitos de pH e fosfatos orgânicos no equilíbrio e propriedades cinéticas na união de ligantes das hemoglobinas de dois peixes relacionados à Amazônia, Osteoglossum bicirrhosum de respiração branquial obrigatória e Arapaima gigas, de respiração aérea obrigatória. As hemoglobinas de ambos os peixes exibem, um efeito Root, a mínima afinidade pelo oxigênio esta associada com a completa perda da cooper atividade na união do ligante. Nesta baixa afinidade, estado T, há uma grande heterogeneidade das unidades de ambas as hemoglobinas como é evidenciado pelo coeficiente de Hill menor a, unidade e a cinética bifásica na combinação de monóxido de carbono. A maior diferença nas propriedades das hemoglobinas destes dois peixes é encontrada a pH altos (acima de pH 8) onde ambas as hemoglobinas exibem sua mais alta afinidade pelo ligante. Aqui ambas as hemoglobinas diferem em sua afinidade e cooperatividade com a qual elas se unem a seus ligantes, tendo a hemoglobina de Arapaima a menor afinidade porém a mais alta cooperatividade.

Summary The effects of pH and organic phosphate on the equilibrium and kinetic properties of the binding of ligands to the hemoglobins of two related Amazonian fish, Osteoglossum bicirrhosum, an obligate water breather, and Arapaima gigas, an obligate air breather, have been studied. The hemoglobins of both fish exhibit a Root effect, and the minimum oxygen affinity is associated with the complete loss of cooperative ligand binding. In this low affinity, T state, there is great subunit heterogeneity in both hemoglobins as evidenced by Hill coefficients well below unity and biphasic carbon monoxide combination kinetics. The greatest difference in the properties of the hemoglobins of these two fish is found at high pH (above pH 8) where both hemoglobins exhibit their highest ligand affinities. Here the hemoglobins differ both in the affinity and cooperativity with which they bind ligands, Arapaima hemoglobin having the lowest affinity but the highest cooperatively.
