Abstract Recent studies have suggested that therapies with stem cells and amniotic membrane can modulate the inflammation following an ischemic injury in the heart. Abstract Muscle overuse and its consequent muscle damage has no cure. Therefore, the present study aimed to investigate the regulatory role of tau-AuNPs on muscle recovery of muscle overuse model. The animals (Male Swiss mice) were randomly divided into four groups: Control (Ctr; n=6); tau-AuNPs (n=6); overuse (n=6); and overuse plus tau-AuNPs (n=6). Exercise sessions were performed for 21 consecutive days, and one exercise model was applied daily in the following sequence: low intensity, moderate intensity, and high intensity. The mice were then sacrificed. The quadriceps muscles were surgically removed for subsequent biochemical analysis (oxidative stress parameters, DNA damage markers and muscle differentiation protein). The overuse group significantly increased the oxidative stress parameters and DNA damage markers, whereas tau-AuNPs significantly decreased the oxidative stress parameters in the overuse animal model. However, there were no significant differences observed between overuse group and overuse plus tau-AuNPs administrated group in relation to DNA damage markers including DNA damage frequency and index levels when compared to control and tau-AuNPs groups. Muscle differentiation protein Myf-5 was increased in the overuse plus tau-AuNPs administration group when compared to control group. In conclusion, tau-AuNPs had significant effect on reducing oxidative stress parameters and increasing myogenic regulatory protein Myf-5 in the overuse group. However, it did not have significant effect on reducing DNA damage.

Abstract The aim of this study was to investigate the effects of yerba mate (Ilex paraguariensis St. Hil.) extract (YME) on oxidative stress parameters and pathological changes in the lungs of mice chronically exposed to hand-rolled cornhusk cigarette (HRC) smoke. Twenty-four male Swiss mice were divided into four groups exposed to the following treatments: control (ambient air), HRC, YME, and HRC plus YME. The animals were exposed to four HRCs per session, with 3 sessions/day, every day for 30 days. Twenty-four hours after the last inhalation, the mice were killed, and the left lungs were removed. The results showed that HRC contains elevated levels of tin and carbon oxide, but less arsenic, cobalt, manganese, and selenium than commercial cigarettes. YME administration reversed fibrosis, alveolar enlargement, and hemorrhage induced by HRC smoke. In addition, the YME and HRC significantly reduced the production of oxidants, oxidative damage and promoted a significant increase in total thiol. In conclusion, exposure to HRC smoke compromised pulmonary histoarchitecture by promoting structural changes and increasing oxidative stress in tissues. However, concomitant treatment with YME regulated the redox state and reduced the harmful effects of HRC smoke exposure in the lungs.

Resumo Fundamento: A obesidade pode ser caracterizada por uma inflamação crônica de baixo grau e está associada à produção excessiva de espécies reativas de oxigênio, fatores que contribuem para doenças coronarianas e outras cardiomiopatias. Objetivo: Verificar os efeitos do treinamento resistido sobre os parâmetros de estresse oxidativo e parâmetro inflamatório em camundongos com obesidade induzida por dieta hiperlipídica (DIO). Métodos: 24 camundongos Swiss foram divididos em 4 grupos: dieta padrão (DP), DP + exercício resistido (DP+ER), obesidade induzida por DIO, DIO + ER. Os animais foram alimentados por 26 semanas com DP ou hiperlipídica realizando treinamento resistido nas 8 semanas finais do estudo. Para avaliar parâmetros clínicos foi realizado o teste de tolerância à insulina (TTI) e monitoramento do peso corporal. No tecido cardíaco foram avaliados parâmetros de estresse oxidativo e inflamação. Dados expressos por média e desvio padrão (p < 0,05). Resultados: O grupo DIO teve um aumento significativo nos níveis espécies reativas e peroxidação lipídica com redução após o exercício. A superóxido dismutase e o sistema glutationa não demonstraram alterações importantes nos animais DIO, com elevação perante DP+ER. Somente a atividade da catalase reduziu tanto com influência da dieta como do exercício. Ocorreu um aumento do fator de necrose tumoral-alfa (TNF-α) no grupo DIO, caracterizando um possível quadro inflamatório, com redução quando expostos ao treino resistido (DIO+ER). Conclusão: A DIO ocasionou um desequilíbrio redox no tecido cardíaco, porém o ER foi capaz de modular estes parâmetros, bem como controlar o aumento do TNF-α.

Abstract Background: Obesity can be characterized by low-grade chronic inflammation and is associated with an excesso production of reactive oxygen species, factors that contribute to coronary heart disease and other cardiomyopathies. Objective: To verify the effects of resistance exercise training on oxidative stress and inflammatory parameters on mice with obesity induced by a high-fat diet (HFD). Methods: 24 Swiss mice were divided into 4 groups: standard diet (SD), SD + resistance exercise (SD + RE), diet-induced obesity (DIO), DIO + RE. The animals were fed SD or HFD for 26 weeks and performed resistance exercises in the last 8 weeks of the study. The insulin tolerance test (ITT) and body weight monitoring were performed to assess the clinical parameters. Oxidative stress and inflammation parameters were evaluated in the cardiac tissue. Data were expressed by mean and standard deviation (p < 0.05). Results: The DIO group had a significant increase in reactive oxygen species levels and lipid peroxidation with reduction after exercise. Superoxide dismutase and the glutathione system showed no significant changes in DIO animals, with an increase in SD + RE. Only catalase activity decreased with both diet and exercise influence. There was an increase in tumor necrosis factor-alpha (TNF-α) in the DIO group, characterizing a possible inflammatory condition, with a decrease when exposed to resistance training (DIO+RE). Conclusion: The DIO resulted in a redox imbalance in cardiac tissue, but the RE was able to modulate these parameters, as well as to control the increase in TNF-α levels.

OBJECTIVES: Progressive decline of physiological processes with aging is normal. Aging is also associated with decreased functional capacity and onset of many diseases. This study evaluated the changes in physical fitness (PF), body composition (BC), and lipid profile (LP) in elderly men completing different training protocols. METHODS: Fifty-five men (age 60-80 years) were randomized into the following groups: without training, aerobic training on dry land, combined training on dry land, and combined training in water. Training was conducted for 8 weeks, and PF, LP, and BC were assessed at the beginning and end of the intervention. RESULTS: Significant improvements were observed in all parameters; however, combined programs on land or in water were more effective at improving strength and aerobic fitness. Combined exercise produced greater effects on BC and LP and some muscle fitness parameters; however, improvements in muscular and aerobic capacities occurred independently of exercise type or model. CONCLUSION: These results indicate that the effects of training occur regardless of training type or model, and are directly associated with training periodization, adherence, and regularity.

Resumo Este estudo teve por objetivo comparar os efeitos do exercício agudo com carga de trabalho relativa em gastrocnêmio de ratos Wistar com três e 18 meses. Os animais foram divididos em quatro grupos (n = 6): controle de três e 18 meses; exercício de três e 18 meses. Os grupos exercitados foram submetidos a uma única sessão de corrida na esteira com 60 minutos em velocidade de 0,8 km/h e 1,2 km/h. Foram analisados parâmetros do metabolismo (níveis de lactato, conteúdo de glicogênio, níveis de succinato desidrogenase, citocromo c oxidase e de estresse oxidativo (SOD, CAT e GPX). Os resultados mostraram um aumento nos marcadores metabólicos após o exercício agudo independentemente da idade dos animais e similaridade nos danos oxidativos apesar de as enzimas antioxidantes apresentarem modulação diferenciada nos animais mais velhos em comparação com animais jovens.

Abstract The effects of exercise on the generation of reactive oxygen species and the response to muscle oxidative stress determines longevity. This study compares the effects of acute exercise with similar relative workload in rats with 3 and 18 months. The animals were divided into four groups (n = 6): control 3-months; exercise 3-months; control 18-months and exercise 18-months. Exercised groups underwent to a single bout of running with 60 minutes in the speed 0.8 km/h (18 months) and 1.2 km/h (3 months). Metabolic parameters (lactate levels, glycogen content, succinate dehydrogenase levels, cytochrome c oxidase) and oxidative stress (activity of superoxide dismutase, catalase and glutathione peroxidase). The results showed an increase in metabolic markers after acute exercise regardless of the age and similarity in oxidative damage even if the antioxidant enzymes are age-dependent. These results suggest that the effects of acute exercise at moderate intensity, affects the metabolism independent of age, and, that the differential response in enzymatic antioxidant system between groups do not promote protection against oxidative damage.

Este estudio ha tenido como objetivo comparar los efectos del ejercicio agudo con carga de trabajo relativamente similar en el gastrocnemio de ratas Wistar con 3 y 8 meses. Se separó a los animales en cuatro grupos (n = 6): control de 3 y 18 meses, y ejercicio de 3 y 18 meses. Se sometió a los grupos de ejercicio a una sola sesión de carrera en la cinta durante 60 minutos con una velocidad de 0,8 km/h y 1,2 km/h. Se analizaron algunos parámetros del metabolismo (niveles de lactato, contenido de glucógeno, niveles de succionato-deshidrogenasa, citocromo c-oxidasa e de estrés oxidativo [superóxido-dismutasa, catalasa y glutatión-peroxidasa]). Los resultados mostraron un aumento de los marcadores metabólicos después del ejercicio agudo independientemente de la edad de los animales y la similitud de los daños oxidativos aunque las enzimas antioxidantes presentan modulación diferenciada en animales viejos en comparación con animales jóvenes.

O presente estudo investigou o efeito de quatro e oito semanas de treinamento físico sobre a atividade dos complexos da cadeia transportadora de elétrons (CTE) e os marcadores de estresse oxidativo em fígado de camundongos. Vinte e um camundongos (CF1, 30-35g) foram distribuídos nos seguintes grupos: não treinado (NT); treinado quatro semanas (T4); treinado oito semanas (T8). Quarenta e oito horas após a última sessão de treinamento os animais foram mortos por decapitação e o fígado foi retirado e estocado em -70ºC para posterior análise. Atividade da succinato desidrogenase (SDH), dos complexos I,II,III e IV da CTE, carbonilação de proteína, conteúdo total de tióis e a atividade da superóxido dismutase foram mensurados. Os resultados demonstram que apenas oito semanas de treinamento aumentam a atividade da SDH, dos quatro complexos da CTE, da superóxido dismutase, e o conteúdo total de tióis em relação ao grupo não treinado. Houve ainda diminuição na carbonilação de proteína no respectivo grupo em relação ao NT. Em conclusão, são necessárias oito semanas de treinamento para que ocorram aumento no funcionamento mitocondrial e melhora nos marcadores de estresse oxidativo em fígado de camundongos.

The present study investigated mitochondrial adaptations and oxidative stress markers after four and eight weeks of running training in liver of mice. Twenty-one male mice (CF1, 30-35g) were distributed into the following groups (n=7): untrained (UT); trained - four weeks (T4); trained - eight weeks (T8). Forty-eight hours after the last training session the animals were killed by decapitation and livers were removed and stored at -70ºC. Succinate dehydrogenase (SDH), complexes I, II, II-III and IV, protein carbonyls (PC), total thiol content and superoxide dismutase activity were measured. The results show that endurance training (8-wk) increases the SDH activity and complexes (I, II, III, IV), superoxide dismutase and total thiol content in liver when compared to untrained animals. Decrease in protein carbonylation in the respective group in relation to UT was also observed. It could be concluded that eight weeks of running training are necessary for mitochondrial respiratory chain enzyme activities increase and improvement in oxidative stress markers in liver of mice.

A atividade física é conhecida por promover saúde e bem-estar. O exercício também é responsável por aumentar a produção de Espécies Reativas de Oxigênio (ERO) pelo acréscimo do consumo de oxigênio mitocondrial nos tecidos. O desequilíbrio entre a produção de EROs e as defesas oxidantes dos tecidos pode provocar danos oxidativos a proteínas, lipídios e DNA. O dano oxidativo cerebral é um mecanismo etiopatológico comum da apoptose e da neurodegeneração. O fator de crescimento cérebro-derivado desempenha um importante papel neste contexto. Nesta revisão, apresentamos os resultados de diferentes modelos de exercício físico no metabolismo oxidativo e neurotrófico do Sistema Nervoso Central (SNC). Também revisamos estudos que utilizaram suplementação antioxidante para prevenir danos oxidativos exercício-induzido ao SNC. Os modelos de exercício físico mais comuns foram as rodas de correr, a natação e a esteira com configurações de treinamento muito diferentes como a duração e a intensidade. Os resultados do treinamento físico no tecido cerebral são muito controversos, mas geralmente demonstram ganhos na plasticidade sináptica e na função cognitiva com exercícios de intensidade moderada e baixa.

Physical activity is known for promoting health and well-being. Exercise is also responsible for increasing the production of Oxygen Reactive Species (ORS) by increasing mitochondrial oxygen consumption causing tissue oxidative stress. The imbalance between ORS production and tissue antioxidant defenses can cause oxidative damage to proteins, lipids and DNA. Brain oxidative damage is a common etiopathology mechanism of apoptosis and neurodegeneration. The brain-derived neurotrophic factor plays an important role in this context. In this review, we showed the results of different models and configurations of physical exercise in oxidative and neurotrophic metabolism of the Central Nervous System (CNS). We also reviewed studies that utilized antioxidant supplementation to prevent exercise-induced oxidative damage to CNS. The commonest physical exercise models were running wheels, swimming and treadmill with very different configurations of physical training such as duration and intensity. The results of physical training on brain tissues are very controversial, but generally show improvement in synaptic plasticity and cognition function with low and moderate intensity exercises.

Estudos têm apontado o exercício físico regular de baixa a moderada intensidade como um importante agente no combate ao estresse oxidativo. O objetivo deste estudo foi investigar o efeito do exercício físico regular na resposta oxidativa pulmonar após a inalação de pó de carvão mineral. Vinte e quatro ratos Wistar machos (200-250g) foram divididos aleatoriamente em dois grupos com respectivos controles (treinado, n = 6 e não-treinado, n = 6). Os animais receberam, por instilação traqueal, pó de carvão mineral (3mg/0,5ml salina, três dias/semana, durante três semanas) ou 0,5ml de solução salina 0,9%. Quarenta e oito horas após a última instilação, o grupo treinado foi submetido a um programa de exercício progressivo em esteira durante 12 semanas (até 17m.min-1, 50min.dia-1, 10% de inclinação). Quarenta e oito horas após a última sessão de treinamento, todos os animais foram mortos por decapitação e os pulmões e sóleo foram cirurgicamente removidos para posterior análise bioquímica. A atividade da citrato-sintase foi determinada no músculo sóleo e os danos em lipídios e proteínas foram avaliados nos pulmões pela concentração de TBARS e pela determinação de grupos carbonil, respectivamente. Os resultados mostram que a prática regular de exercício físico reduz significativamente os níveis presentes de TBARS em ratos treinados e diminui os níveis de oxidação em proteínas em ambos os grupos quando comparados com os respectivos controles. Os resultados nos levam a sugerir que o exercício físico regular em esteira é um agente capaz de amenizar os danos oxidativos pulmonares induzidos pela inalação de partículas de carvão mineral.

Several studies have pointed the regular low to moderate intensity physical exercise an important agent to combat the oxidant stress. The purpose of this study was to investigate the effect of the physical exercise in the pulmonary oxidant response after inhaling mineral coal dust. Twenty-four male Wistar rats (200-250 g) were randomly divided in two groups with their respective controls (trained, n = 6; non-trained, n = 6). All animals received mineral coal dust via tracheal instillation (saline 3 mg/0.5 ml, 3 days/week for 3 weeks), or 0.5 ml of 0.9% saline solution. Forty-eight hours after the last instillation, the trained group was submitted to a progressive exercise program on treadmill for twelve weeks (up to 17 m.min-1, 50 min/day-1, 10% inclination). Forty-eight hours after the last training session, every animal was killed by decapitation, and their lungs and soleus were surgically removed for later biochemical analysis. The citrate synthase activity was determined in the soleus muscle, and the damages on the lipids and proteins were assessed into the lungs by means of the TBARS concentration and by determining the carbonyl groups, respectively. The results show that the regular practice of physical exercises reduces significantly the present TBARS levels in trained rats, decreasing the oxidation levels in proteins of both groups compared to their respective controls. The results lead us to suggest that the regular physical exercise on treadmill is an agent capable to soften the pulmonary oxidant damages caused by inhaling mineral coal particles.

Los estudios han estado apuntando el ejercicio físico regular de intensidad baja a moderada como un agente importante en el combate a la tensión oxidativa. El objetivo de ese estudio era investigar el efecto del ejercicio físico para regular la respuesta oxidativa pulmonar después de la inhalación de polvo de carbón mineral. Veinticuatro ratones varones Wistar (200-250g) fueron aleatoriamente divididos en 2 grupos con los mandos respectivos (, n = 6 y Ningún -, n = 6). Los animales recibieron, por instilación traqueal polvo de carbón mineral (3mg/0,5ml salina, 3 dias/semana, durante 3 semanas) o 0,5ml de solución salina 0,9%. Cuarenta ocho horas después de la última instilación, el grupo especializado fue sometido a un programa de ejercicio progresivo en la cinta durante 12 semanas (a 17m.min-1, 50min.dia-1, 10% de inclinación). Cuarenta ocho horas después de la última sesión de entrenamiento, todos los animales fueron sacrificados por decapitación y los pulmones y el músculo sóleo fueron extirpados por cirugía para el análisis bioquímico subsecuente. La actividad de la citrato-sintetasa fue determinada en los músculos así como los daños y perjuicios en los lipidos y las proteínas se estimaron en los pulmones para la concentración de TBARS y para la determinación de carbonilo de estos grupos, respectivamente. Los resultados mostraron que el ejercicio físico regular reduce los niveles presentes de TBARS significativamente en los ratones especializados y reduce los niveles de la oxidación en las proteínas en ambos grupos cuando se compararon los grupos respectivos. Los resultados permitieron sugerir que el ejercicio físico regular en la cinta es un agente capaz minimizarlos daños y perjuicios oxidativos pulmonares inducidos por la inhalación de partículas de carbón mineral.
