Summary: Chitosan nanoparticles (NPsCS) are used as natural biostimulants in sustainable agriculture since they increase crop productivity and induce the synthesis of enzymatic and non-enzymatic antioxidants, protecting the plant from stress. The present study was developed to determine the effect of foliar application of NPsCS on yield, enzymatic activity, and content of bioactive compounds in tomato fruits. The trial was established completely randomized design with six escalating doses of NPsCS 0, 0.05, 0.1, 0.2, 0.4, and 0.8 mg mL-1. The foliar spray of 0.2 mg mL-1 increases the fruits’ yield, size, and firmness. High doses increase bioactive compounds and enzymatic activity. The foliar-applied NPsCS has excellent potential to be used as biostimulants to improve performance and obtain functional foods.

Resumen: Las nanopartículas de quitosán (NPsCS) son utilizadas como bioestimulantes naturales en la agricultura sustentable, ya que incrementan la productividad de los cultivos e inducen la síntesis de antioxidantes enzimáticos y no enzimáticos, protegiendo a la planta del estrés. El presente estudio se desarrolló con el objetivo de determinar el efecto de la aplicación foliar de las NPsCS sobre el rendimiento, actividad enzimática y contenido de compuestos bioactivos en frutos de tomate. El ensayo se estableció en un diseño completamente al azar con seis dosis crecientes de NPsCS: 0, 0.05, 0.1, 0.2, 0.4 y 0.8 mg mL-1. La aspersión foliar de 0.2 mg mL-1 aumentó el rendimiento, tamaño y firmeza de los frutos; en cambio dosis alta incrementan los compuestos bioactivos y la actividad enzimática. El uso de NPsCS aplicadas de forma foliar presentan un gran potencial para utilizarse como bioestimulantes para mejorar el rendimiento y obtener alimentos funcionales.

Abstract The use of chitosan nanoparticles (NPs CS) has become a promising alternative in modern agriculture as an inducer in the biosynthesis of secondary metabolites. The objective of this research was to evaluate the effect of NPs CS on the nutraceutical quality of triticale sprouts (x Triticosecale Wittmack). Increasing doses of NPs Cs: 0, 0.1, 0.2, 0.4 and 0.8 mg ml-1 were applied only once at the imbibition stage, then they were left in Petri dishes for 7 days at 25 ±2 °C temperature. NPs CS did not affect germination or fresh root weight at the tested concentrations, and the concentration of 0.1 mg ml-1 increased the fresh weight of the shoots up to 83.3%. In the presence of 0.8 mg ml-1 of NPs CS phenolic compounds decrease by 7% and flavonoids increase by 29%. The results confirm a promoter effect of NPS CS on sprouts, opening the possibility of being used as inducers in the biosynthesis of bioactive compounds in triticale sprouts.

Resumen El uso de nanopartículas de quitosán (NPs CS) se ha vuelto una alternativa prometedora en la agricultura moderna como un inductor en la biosíntesis de metabolitos secundarios. El objetivo del presente trabajo fue evaluar el efecto de NPs CS en la calidad nutraceútica de germinados de triticale (x Triticosecale Wittmack). Dosis creciente de NPs CS: 0, 0.1, 0.2, 0.4 y 0.8 mg ml-1 se aplicaron una sola vez en la etapa de imbibición, después se dejaron en cajas Petri durante 7 días a 25 ±2 °C de temperatura. Las NPs CS no afectaron la germinación ni el peso freso de la raíz a las concentraciones probadas, y a la concentración de 0.1 mg ml-1 aumentó el peso fresco del brote hasta 83.3%. En presencia de 0.8 mg ml-1 de las NPs CS los compuestos fenólicos disminuyen un 7% y aumentan 29% los flavonoides. Los resultados confirman un efecto promotor de las NPS CS en los germinados, abriendo la posibilidad de ser utilizadas como inductores en la biosíntesis de compuestos bioactivos en germinados de triticale.

Abstract: The immobilization of Trichoderma harzianum on chitosan hydrogels and their effects on greenhouse tomato plants (Solanum lycopersicum) was documented. The experiment was conducted in two cycles. In July-August 2009 and March-April 2010. This consisted in the germination of tomato seeds in pots with turb as substrate. Six treatments were established: two treatments of T. harzianum immobilized on chitosan hydrogels applied to two doses (6 and 3 g kg-1 of substrate), two treatments with chitosan hydrogels applied to the same doses without the fungus, a treatment where the fungus was applied to the substrate as a suspension, and a control (only substrate). In both cycles the viability of T. harzianum in the substrate samples used for treatments, at the time of immobilization and 30 and 51 days after sowing was tested. The results show that the growth of fungi immobilized in the first cycle was similar in all treatments, in the second cycle, when the high concentration of T. harzianum (Th) was maintained for a longer time, the superiority of the immobilized fungus was evident. In terms of effects on tomato plants of the variables studied, we observed that treatments with the fungus immobilized in the hydrogels produced less plant growth and plant biomass, but the P, Ca, and Cu mineral content was higher in treatments with the presence of fungus. There was no effect on the leaf epidermis, vascular tissue of leaves, stems and roots, or CO2 assimilation (P ≤ 0.05).

Resumen: Fue documentada la inmovilización de Trichoderma harzianum en hidrogeles de quitosano y sus efectos en las plantas de tomate (Solanum lycopersicum) bajo condiciones de invernadero. El experimento se realizó en dos ciclos. En los meses de julio-agosto de 2009 y marzo-abril de 2010. Esta consistió en la germinación de semillas de tomate en macetas con turba como sustrato. Se establecieron 6 tratamientos, estos fueron: dos tratamientos con el T. harzianum inmovilizado en los hidrogeles de quitosano aplicado al sustrato a dos dosis (6 y 3 g por kg de sustrato), dos tratamientos con los hidrogeles de quitosano aplicado a las mismas dosis pero sin el hongo, un tratamiento donde se aplicó el hongo en forma de suspensión al sustrato, y un testigo absoluto (solo sustrato). En ambos ciclos se realizaron pruebas de viabilidad de T. harzianum en las muestras de sustrato usadas de los tratamientos correspondientes; al momento de la inmovilización y a los 30 y 51 días después de la siembra. Los resultados obtenidos muestran que el crecimiento de los hongos inmovilizados en el primer ciclo fue similar en todos los tratamientos correspondientes, en el segundo ciclo sobresale la superioridad del hongo inmovilizado al mantener por más tiempo una concentración alta de T. harzianum (Th). En cuanto a los efectos sobre las plantas de tomate correspondientes a las variables estudiadas, se observó que los tratamientos con el hongo inmovilizado en los hidrogeles producían plantas con menor crecimiento y biomasa vegetal, sin embargo, el contenido mineral de P, Ca y Cu fue mayor en los tratamientos con la presencia del hongo. No se observó ningún efecto sobre la epidermis foliar, tejido vascular de las hojas, tallos y raíces, y la asimilación de CO2 (P ≤ 0.05).

A atividade antimicrobiana de quitosana e complexos interpolieletrolíticos hidrossoluvéis de poli(ácido acrílico)-quitosana foi estudada. Quitosanas de dois diferentes pesos moleculares foram testados em diferentes concentrações, 0,5 a 5 g • L-1, como agentes antimicrobianos nas P. aeruginosa e P. oleovorans. Em ambos os casos, obteu-se a melhor inibição microbiana com a concentração de 5 g • L-1, no entanto os complexos interpolieletrolíticos de poli (ácido acrílico)-quitosana com composição φ = 2 apresentaram maior atividade antibacteriana do que os dois quitosans na concentração de 0,5 g • L-1. O complexo NPEC2 foi mais eficaz do que as quitosanas, sendo que o resultado pode ser atribuído ao número de moles dos grupos aminos da quitosana e aos grupos carboxílicos dos complexos de poli(ácido acrílico).

The antimicrobial activity of chitosan and water soluble interpolyelectrolyte complexes of poly(acrylic acid)-chitosan was studied. Chitosans of two different molecular weights were tested at different concentration for 0.5 to 5 g·L-1 as antimicrobial agents against P. aeruginosa and P. oleovorans. In both cases, the best microbial inhibition was obtained with the concentration of 5 g·L-1. However, the interpolyelectrolyte complexes of poly(acrylic acid)-chitosan with composition φ =2 produced higher antibacterial activity than the two chitosans at the concentration of 0.5 g·L-1. The NPEC2 complex was more effective than chitosans. This could be attributed to the number of moles of the amino groups of chitosan and the carboxylic acid groups of the interpolyelectrolyte complexes poly(acrylic acid).

The objective of this work was the evaluation of the peroxidase and catalase level of activity in the protein extract of tomato in response to different treatments with chemical elicitors in different stages of tomato development. The elicitor compounds chitosan at 0.1 % (w/v), 0.1 mM salicylic acid and 0.1 mM benzoic acid were sprayed on tomato fruits without applying them in the leaves and stems of the plant. The elicitors were sprayed at one of the following stages: fruit set, fruit growth and phase 3 of fruit ripening. The activity of the antioxidant enzymes catalase and peroxidase was determined in the phase 4 of fruit ripening, being found an increase in the activity of both enzymes upon applying the elicitors in certain stages of the fruit development. The chitosan applied during fruit growing, and the salicylic acid applied during fruit set increased the activity of catalase. The peroxidase activity increased significantly upon applying chitosan during fruit set and fruit growing, finding a smaller effect for the salicylic acid. Benzoic acid did not modify the peroxidase or catalase activity.

El objetivo de este trabajo fue la evaluación del nivel de la actividad de peroxidasa y catalasa en extracto proteínico de tomate en respuesta a los diferentes tratamientos con inductores químicos en diferentes estados de desarrollo del tomate. En forma exógena se aplicaron los inductores de tolerancia quitosano al 0.1 %, ácido salicílico 0.1 mM y ácido benzoico 0.1 mM en frutos de tomate sin aplicarlos en las hojas y tallos de la planta. Los compuestos inductores se aplicaron mediante aspersión a los racimos fructificados en alguna de las siguientes etapas: amarre, llenado y etapa 3 de la maduración del fruto. La actividad de las enzimas antioxidantes catalasa y peroxidasa se determinó en la etapa 4 de maduración, encontrándose aumento en la actividad de ambas enzimas al aplicar los inductores en ciertas etapas del crecimiento del fruto. El quitosano aplicado en el llenado de fruto y el ácido salicílico durante el amarre incrementaron la actividad de catalasa. La actividad de peroxidasa aumentó significativamente al aplicar quitosano en el amarre y llenado de fruto encontrando un efecto menor para el ácido salicílico. El ácido benzoico no modificó la actividad de catalasa o peroxidasa.
