Abstract This work aimed to evaluate the in vitro bioaccessibility of α-carotene, β-carotene, lycopene and phenolic compounds and antioxidant capacity in the pequi almond, pulp and peel flours. Furthermore, it is relevant to interrelate these results with water activity (aw), Total Titratable Acidity (TTA), pH and color parameters through principal component analysis. For this, fruits were collected in the state of Mato Grosso (MT), and they were sanitized, peeled, pulped and the almonds removed. Quantification of total and bioaccessible concentrations of carotenoids and phenolic compounds and antioxidant capacity were performed by UV-Visible spectrophotometry. In vitro bioaccessibility tests were performed using a three-step: salivary; gastric; and intestinal digestions. The highest concentrations of α-carotene (63.81 µg/g), β-carotene (59.83 µg/g) and lycopene (40.66 µg/g) were obtained for pequi pulp flour. However, the highest percentages of bioaccessibility of α-carotene (59.69%), β-carotene (96.14%) and lycopene (44.13%) were found in almond flour. The concentrations of total phenolic compounds were: 210.50; 402.21; and 9475.69 mg GAE/100g, for pequi almond, pulp and peel flour, respectively. The highest percentage of bioaccessibility (86.68%) was found in pulp flour. Pequi peel flour showed the highest antioxidant capacity (EC50) 45.71 g/g of DPPH. The highest results for L* (42.19), a* (4.17), b* (8.39) and TTA (17.83 mL NaOH/100 g) were verified in the pulp flour; whereas the lowest pH (3.34) was verified in the peel flour. Principal component analysis indicated that pH and TTA can influence the in vitro bioaccessibility of phenolic compounds, carotenoids and the antioxidant capacity. Resumo Este trabalho objetivou avaliar a bioacessibilidade in vitro de α-caroteno, β-caroteno, licopeno e de compostos fenólicos, e a capacidade antioxidante em farinhas de amêndoa, polpa e casca do fruto pequi. Além disso, inter-relacionar estes resultados com a atividade de água, acidez e parâmetros de cor, por meio da análise de componentes principais. Os frutos foram coletados no estado de Mato Grosso, sendo higienizados, descascados, despolpados e retiradas as amêndoas. A quantificação das concentrações totais e bioacessíveis dos carotenoides e compostos fenólicos e a capacidade antioxidante foram feitas por espectrofotometria UV-Visível. Os ensaios de bioacessibilidade in vitro foram feitos por meio de três etapas: digestão salivar, gástrica e intestinal. As maiores concentrações de α-caroteno (63,81 µg/g), β-caroteno (59,83 µg/g) e licopeno (40,66 µg/g) foram obtidas para farinha da polpa do pequi. Porém, as maiores porcentagens de bioacessibilidade de α-caroteno (59,69%), β-caroteno (96,14%) e licopeno (44,13%) foram encontradas na farinha da amêndoa. Os teores de compostos fenólicos totais foram: 210,50, 402,21 e 9475,69 mg GAE/100g, para as farinhas de amêndoa, polpa e casca, respectivamente, sendo a maior porcentagem de bioacessibilidade (86,68%) encontrada na farinha da polpa. A farinha da casca do pequi apresentou maior capacidade antioxidante (EC50) 45,71 g/g de DPPH. Os maiores resultados de L* (42,19), a* (4,17), b* (8,39) e acidez titulável (17,83 mL NaOH/100 g) foram verificados na farinha da polpa e o menor pH (3,34), na farinha da casca. A análise de componentes principais indicou que o pH e a acidez titulável podem influenciar na bioacessibilidade de compostos fenólicos e carotenoides, e na capacidade antioxidante. O método empregado para a determinação de sódio por espectrometria de emissão atômica em chama, foi o das adições de analito sendo as intensidades de emissão representadas em função das concentrações de analito adicionadas às várias porções da amostra, numa faixa linear de 0 - 0,300 mg Na L-1, empregando-se 2,1 g K L-1 como supressor de ionização. Foram analisadas 20 amostras comerciais de álcool etílico hidratado combustível (AEHC), coletadas em diferentes postos de abastecimento, na cidade de Araraquara e os resultados obtidos encontram-se no intervalo, desde menor que o limite de detecção (0,0072 mg Na L-1) até 1,55 mg Na L-1. Os limites de detecção referentes à determinação de sódio em cada amostra situam-se entre 0,0026 e 0,0239 mg Na L-1. A porcentagem de recuperação variou no intervalo de 95 a 104 %. Os desvios padrão relativos (n=12) foram <FONT FACE=Symbol>£</FONT> 4,1 % para as três adições de analito, nas 20 amostras de AEHC.

The analyte additions method was applied for sodium determination in fuel ethanol by atomic emission spectrometry. Graphics involving emission intensity versus analyte concentration in the 0 - 0.300 mg Na L-1 interval concentration range contaning 2.1 g K L-1 as an ionisation buffer. Twenty samples of commercial fuel ethanol were collected in differents gas stations located in Araraquara city, analyzed and results obtained varied from 0.0072 to 1.55 mg Na L-1. The limits of detecction (L.O.D.) varied from 0.0026 to 0.0239 mg Na L-1. Recoveries varied in the 95 - 104 % interval. The relative standard deviations (n=12) for three analyte additions in all samples were <FONT FACE=Symbol>£</FONT> 4,1 %.

Simultaneous electrolytic deposition is proposed for minimization of Cu2+ and Pb2+ interferences on automated determination of Cd2+ by the Malachite Green-iodide reaction. During electrolysis of sample in a cell with two Pt electrodes and a medium adjusted to 5% (v/v) HNO3 + 0.1% (v/v) H2SO4 + 0.5 mol L-1 NaCl, Cu2+ is deposited as Cu on the cathode, Pb2+ is deposited as PbO2 on the anode while Cd2+ is kept in solution. With 60 s electrolysis time and 0.25 A current, Pb2+ and Cu2+ levels up to 50 and 250 mg L-1 respectively, can be tolerated without interference. With on-line extraction of Cd2+ in anionic resin minicolumn, calibration graph in the 5.00 - 50.0 µg Cd L-1 range is obtained, corresponding to twenty measurements per hour, 0.7 mg Malachite Green and 500 mg KI and 5 mL sample consumed per determination. Results of the determination of Cd in certified reference materials, vegetables and tap water were in agreement with certified values and with those obtained by GFAAS at 95% confidence level. The detection limit is 0.23 µg Cd L-1 and the RSD for typical samples containing 13.0 µg Cd L-1 was 3.85 % (n= 12).
