ABSTRACT During experiment planning, determining the number of replicates for tested treatments is important because it directly affects the accuracy of the obtained results. This study was conducted to determine the number of measurements (repetitions) necessary to evaluate the yield traits and soluble solids in Cantaloupe and Gália melon hybrid trials. The study comprised twenty-one experiments, nine for evaluating eight Cantaloupe melon hybrids, and twelve for evaluating nine Galia melon hybrids, conducted in a randomized complete block design with three replicates each. Analysis of variance was performed, and repeatability and genotypic determination coefficients were estimated for each experiment. The use of three repetitions allowed identification of superior genotypes with 83.6 and 80.7% predictions of the real values for the yield and soluble solids, respectively, for Cantaloupe melons. Evaluating the trials with Galia melon using three repetitions allowed prediction of the true value of the genotypes with 86.1 and 98.6% accuracy for fruit yield and soluble solids, respectively. Therefore, the use of three replicates was determined to be sufficient for detecting superior genotypes both for fruit yield and soluble solid content, with more than 80% certainty for their true values.

RESUMO: Objetivou-se com este trabalho estudar o controle genético de caracteres relacionados à qualidade do fruto do melão. Foram avaliadas as gerações F1, F2, RC1, RC2 dos cruzamentos OL x A-16 e OL x PV em dois ensaios separados conduzidos em blocos casualizados com três repetições. Os caracteres avaliados foram: peso médio do fruto, índice de formato, espessura da polpa, firmeza da polpa, sólidos solúveis e porcentagem de rachadura. As análises foram feitas por meio de estudo clássico de gerações envolvendo modelos mistos. Foram estimados os parâmetros de herdabilidades nos sentidos amplo e restrito e número de loci que controlam o caráter. A herança dos caracteres estudados é complexa com a presença de gene de efeito maior e poligenes com efeitos aditivos e de dominância.

ABSTRACT: This research studied the genetic control of the traits related to melon fruit quality. The F1, F2, BC1, BC2 generations from the OL x A-16 and OL x PV crossings were evaluated in two separate trials conducted in randomized blocks with three replications. The evaluated traits were: average fruit weight, shape index, pulp thickness, pulp firmness, soluble solids content and cracking rate. The analyses were accomplished through a classic study of generations involving mixed models. The parameters on heritability and number of loci controlling the traits were evaluated in a broad and narrow sense. The inheritance of the evaluated traits is complex, presenting one gene of greater effect and polygenes with additive and dominant effects.

ABSTRACT The objective of the present work was to evaluate the influence of environmental variables on the interaction between genotypes and environments and to identify adapted and stable genotypes for grain seed yield. Twenty-one cultivars were evaluated in randomized blocks with four replications in the years 2016, 2017, and 2018 in the northeastern semi-arid region of Brazil, for seed yield and oil content. Factor regression methodologies and principal component analysis were used with predictions of the sum of the genotypic effects and the interaction to quantify the role of five environmental covariates in the genotype x environment interaction; the Harmonic Mean Method of Relative Performance of Genotypic Values was used for identifying adapted and stable genotypes. The covariance biplot model is useful for relating important environmental factors and indicating their relative effect on seed yield and oil content. Rainfall, relative humidity and maximum temperature contribute positively to increasing oil content while minimum temperature and solar radiation reduce it. Within the limits of the work, the maximum temperature positively influences grain production while the minimum reduces it. The most stable genotypes and those adapted for grain seed yield and oil content are BMX OPUS IPRO, P 98Y70 RR, BRS 333 RR, BRS 9280 RR, M 8644 IPRO, M 8372 IPRO, and ST 920 RR.

RESUMO O objetivo do presente trabalho foi avaliar a influência de variáveis ambientais na interação entre genótipos e ambientes e identificar genótipos adaptados e estáveis para a produção de sementes. Vinte e uma cultivares foram avaliadas em blocos ao acaso com quatro repetições nos anos de 2016, 2017 e 2018 no semiárido nordestino do Brasil, para rendimento de sementes e teor de óleo. Metodologias de regressão fatorial e análise de componentes principais foram utilizadas com previsões da soma dos efeitos genotípicos e a interação para quantificar o papel de cinco covariáveis ambientais na interação genótipo x ambiente; o Método da Média Harmônica de Desempenho Relativo de Valores Genotípicos foi usado para identificar genótipos adaptados e estáveis. O modelo biplot de covariância é útil para relacionar fatores ambientais importantes e indicar seu efeito relativo no rendimento de sementes e no teor de óleo. A precipitação, a umidade relativa e a temperatura máxima contribuem positivamente para aumentar o teor de óleo, enquanto a temperatura mínima e a radiação solar o reduzem. Dentro dos limites do trabalho, a temperatura máxima influencia positivamente a produção de grãos enquanto a mínima a reduz. Os genótipos mais estáveis e adaptados para rendimento de sementes de grãos e teor de óleo são BMX OPUS IPRO, P 98Y70 RR, BRS 333 RR, BRS 9280 RR, M 8644 IPRO, M 8372 IPRO e ST 920 RR.

RESUMO O uso de cultivares resistentes é um método eficiente e recomendável para manejo da mosca minadora, principal problema fitossanitário na cultura do meloeiro. Nesse contexto, os objetivos deste trabalho foram identificar fontes de resistência à mosca minadora em acessos de melão amarelo e determinar sua herança genética. Para tanto, foram realizados dois experimentos em campo nos municípios de Baraúna - RN e Icapuí - CE para identificar fontes de resistência. O delineamento adotado foi em blocos completos casualizados com 22 tratamentos e quatro repetições. Na avaliação foi quantificado o número de minas por folha. A heterogeneidade dos materiais estudados possibilitou identificar os acessos AM-RT e AM-TM como fontes de resistência, considerando que apresentaram número de minas igual a zero nos dois ambientes de avaliação. O acesso AM-RT foi selecionado e utilizado para obter a população S1 (autofecundação), população derivada de S1:2 e o cruzamento entre AM-RT e ‘Goldex’, que foram avaliadas em um terceiro ensaio de laboratório visando determinar o controle genético da resistência em AM-RT. Portanto, por meio do padrão de segregação das populações S1, S1:2 e do cruzamento (AM-RT’ and ‘Goldex’) avaliados, estimando-se os valores de qui quadrado (χ2), que foram de 1,33, 3,14 e 0,36, respectivamente, foi determinado que a herança da resistência é controlada por apenas um gene com dominância completa. Então, nesse estudo foram identificadas duas fontes de resistência à mosca minadora e um gene com dominância completa condiciona à resistência no acesso AM-RT.

ABSTRACT The use of resistant cultivars is an efficient and recommended method for the management of leafminers, which are the main phytosanitary problem in melons. The objectives of this study were to identify the sources of resistance to the leafminer in yellow melon accessions and to determine the resistance inheritance in acession AM-RT. Two field experiments were conducted in the municipalities of Baraúna, RN and Icapuí, CE, Brazil, to identify the sources of resistance. The design adopted was completely randomized blocks with 22 treatments and four replications. In this evaluation, the number of mines per leaf was quantified. The heterogeneity of the studied materials allowed for the identification of the accessions AM-RT and AM-TM as sources of resistance, considering that they revealed zero mines in the two evaluation environments. The accession AM-RT was selected and used to obtain the S1 population (by self-fertilization), S1:2 population derived from S1, and crossing between AM-RT and ‘Goldex’, which were evaluated in a third laboratory trial to determine the genetic control of resistance in that material. By the segregation pattern of the populations S1, S1:2, and the crossing (AM-RT and ‘Goldex’) and the estimation of the chi-squared (χ2) values, which were 1.33, 3.14, and 0.36, respectively, it was determined that the inheritance of resistance was controlled by only one gene with complete dominance. Therefore, in this study, two sources of resistance to the leafminer were identified, and resistance was conditioned by a gene with complete dominance in the accession ‘AM-RT’.

RESUMO Macrophomina (Botryosphaeriaceae) é um dos principais gêneros de fungos fitopatogênicos de solo, que causam apodrecimento de raízes e sementes em mais de 800 plantas hospedeiras em todo o mundo. Estudos filogenéticos recentes identificaram as espécies M. phaseolina e M. pseudophaseolina em Trianthema portulacastrum e Boerhavia diffusa em áreas de produção de melão e melancia no Nordeste do Brasil. Portanto, o objetivo deste estudo foi: i) verificar o efeito da temperatura e salinidade sobre o crescimento micelial de M. phaseolina, M. pseudophaseolina e M. euphorbiicola, ii) avaliar sua patogenicidade em mudas de melão e melancia, e iii) determinar suas sensibilidades ao fungicida carbendazim. A temperatura ótima para taxa de crescimento micelial (MGR) para Macrophomina spp. variou de 27,1 ºC (CMM4771 – M. pseudophaseolina) a 31,8 ºC (CMM4763 – M. phaseolina). Para o efeito da salinidade no crescimento micelial de isolados de Macrophomina, a EC50 variou de 103,76 (CMM4868 – M. euphorbiicola) a 315,25 mM (CMM4801 – M. pseudophaseolina). O teste de patogenicidade demonstrou que M. phaseolina, M. pseudophaseolina e M. euphorbiicola são patogênicas em melão com M. phaseolina apresentando maior virulência. Isolados de Macrophomina euphorbiicola não causaram doenças em melancia. Os isolados mais sensíveis ao fungicida carbendazim foram CMM4868, CMM4867 (M. euphorbiicola) e CMM1531 (M. phaseolina) com EC50 de 0,003; 0,012 e 0,012 mg.L-1 i.a., respectivamente. Todas as espécies de Macrophomina spp. utilizados nestes experimentos foram patogênicos para as cultivares de melão e melancia testadas com exceção dos isolados de M. euphorbiicola que não causaram danos à melancia.

ABSTRACT Macrophomina (Botryosphaeriaceae) is one of the main genera of soilborne phytopathogenic fungi, which causes root and seed rot in more than 800 host plants worldwide. Recent phylogenetic studies identified the species M. phaseolina and M. pseudophaseolina in Trianthema portulacastrum and Boerhavia diffusa in melon and watermelon production areas in northeastern Brazil. Therefore, the objective of this study was: i) to verify the effect of temperature and salinity on the mycelial growth of M. phaseolina, M. pseudophaseolina and M. euphorbiicola, ii) to assess their pathogenicity on melon and watermelon seedlings, and iii) to determine their sensitivity to the fungicide carbendazim. The optimal temperature for mycelial growth rate (MGR) for Macrophomina spp. ranged from 27.18 ºC (CMM4771 – M. pseudophaseolina) to 31.80 ºC (CMM4763 – M. phaseolina). For the effect of salinity on mycelial growth of Macrophomina isolates, the EC50 ranged from 103.76 (CMM4868 – M. euphorbiicola) to 315.25 mM (CMM4801 – M. pseudophaseolina). The pathogenicity test demonstrated that M. phaseolina, M. pseudophaseolina and M. euphorbiicola are pathogenic on melon with M. phaseolina exhibiting a higher level of virulence. Macrophomina euphorbiicola isolates did not cause disease in watermelon. The most sensitive isolates to the fungicide carbendazim were CMM4868, CMM4867 (M. euphorbiicola) and CMM1531 (M. phaseolina) with EC50 of 0.003, 0.012 and 0.012 mg.L-1 a.i., respectively. All Macrophomina spp. used in these experiments were pathogenic to the tested melon and watermelon cultivars with the exception of the M. euphorbiicola isolate that did not cause damage to watermelon.

RESUMO A acidez é um dos principais determinantes do sabor e qualidade dos frutos do meloeiro juntamente com açúcares e compostos voláteis. O objetivo deste trabalho foi estudar a herança do potencial hidrogeniônico (pH) e da acidez titulável dos frutos do meloeiro. Foram avaliadas em um delineamento em blocos casualizados com três repetições as gerações F1, F2 e os retrocruzamentos (RC1 e RC2) a partir dos genitores Védrantais (Cucumis melo subsp. melo var. reticulatus), com alto pH e baixo teor acidez total titulável, e AC-16 (C. melo subsp. melo var acidulus), com baixo pH e elevado teor de acidez titulável. A herança do pH e da acidez titulável em melão são complexas e cada uma envolve um gene maior com efeitos aditivos e de dominância associado a poligenes com efeitos aditivos.

ABSTRACT Acidity is one of the main determinants of the taste and quality of melon fruit, together with sugars and volatile compounds. The aim of this study was to investigate the inheritance of hydrogen potential (pH) and titratable acidity in melon fruit. A randomized block design with three replications was used to evaluate the F1 and F2 generations and backcrosses (RC1 and RC2) of the Védrantais melon (Cucumis melo subsp. melo var. reticulatus) with high pH and low titratable acidity, and of the AC-16 melon (C. melo subsp. melo var acidulus), with low pH and high titratable acidity. Inheritance of pH and titratable acidity in the melon is complex, and each involves a major gene with additive and dominance effects associated with polygenes with additive effects.

Resumo O objetivo deste trabalho foi determinar a herança do teor de betacaroteno em melão (Cucumis melo). O acesso AC-16 (Cucumis melo subsp. melo var. acidulus) – com baixo teor de betacaroteno e mesocarpo branco – foi cruzado com a cultivar Vedrantais (C. melo subsp. melo var. cantalupensis) – com alto teor de betacaroteno e mesocarpo de cor salmão –, para obtenção das gerações F1, F2, RC1 e RC2. Avaliaram-se os genitores AC-16 e 'Vedrantais', as gerações F1 e F2, e os retrocruzamentos de cada genitor RC1 e RC2. A quantificação do betacaroteno foi realizada em sistema de cromatografia líquida de alto desempenho. Foram estimados os componentes de média relacionados aos efeitos aditivos e de dominância, as variâncias aditiva e de dominância e a herdabilidade. O teor de betacaroteno foi alto (17,78 µg g-1) em 'Vedrantais' e baixo em AC-16 (0,34 µg g-1). Observaram-se os seguintes resultados: efeito aditivo e de dominância no controle genético do caráter, dominância de caráter incompleta, número estimado de loci próximo de dois, maior variância para populações segregantes (F2 e retrocruzamentos), e valores de herdabilidade nos sentidos amplo (87,75%) e restrito (64,19%). O teor de betacaroteno em melão é controlado por um gene de efeito maior, com efeitos aditivos e de dominância associados a poligenes com efeitos aditivos.

Abstract The objective of this work was to determine the inheritance of beta-carotene content in melon (Cucumis melo). The AC-16 accession (Cucumis melo subsp. melo var. acidulus) – with a low beta-carotene content and white mesocarp – was crossed with the Vedrantais cultivar (C. melo subsp. melo var. cantalupensis) – with a high beta-carotene content and salmon colored mesocarp –, to obtain the F1, F2, BC1, and BC2 generations. The AC-16 and 'Vedrantais' parents, the F1 and F2 generations, and the BC1 and BC2 backcrosses of each parent were evaluated. The quantification of beta-carotene was carried out in a high-performance liquid chromatography system. Mean components related to the additive and dominance effects, additive and dominance variances, and heritability were estimated. The beta-carotene content was high in 'Vedrantais' (17.78 µg g-1) and low in AC-16 (0.34 µg g-1). The following results were observed: additive and dominance effects on the genetic control of the character, incomplete character dominance, estimated number of loci close to two, greater variance for segregating populations (F2 and backcrosses), and heritability values in the broad (87.75%) and narrow (64.19%) senses. The beta-carotene content in melon is controlled by a major effect gene, with additive and dominance effects associated with polygenes with additive effects.

RESUMO O objetivo deste trabalho foi avaliar genótipos de abóbora de leite e jerimum caboclo para resistência ao fungo Macrophomina phaseolina (Taissi) Goid. Foram utilizados 17acessos de Cucurbita moschata Duchesne and 20 de Cucurbita máxima Duchese. Também foram utilizadas quatro testemunhas comerciais, duas para cada espécie. Em ambos os trabalhos, inoculou-se três isolados de M. phaseolina que estão mantidos na Micoteca da UFERSA: MM1531 (GenBank: MN136199), ME249 e ME250. As plantas receberam inóculo do patógeno em pedaços de palito de dente colonizado, e foram avaliadas quanto a incidência e severidade da doença. Ambas as espécies apresentaram plantas resistentes, porém com frequências variadas para os acessos. Quando consideradas as espécies, as frequências verificadas para C. Moschata foram superiores às constatadas para C. maxima. Os acessos de C. moschata ABPUN 206 F1, P114-1, P160-2, P11-2, P114-6, P114-02 e P97-1 apresentam diferentes níveis de resistência a Macrophomina phaseolina para os isolados MM1531, ME-249 e ME-250. Assim, para a seleção eficiente em programa de melhoramento para a característica, as estratégias de seleção adotadas devem considerar cada fonte de resistência isoladamente e não cada espécie.

ABSTRACT The objective of this study was to evaluate genotypes of Cucurbita moschata and Cucurbita maxima for resistance to the fungus Macrophomina phaseolina (Taissi) Goid. The experiment evaluated 17 accessions of Cucurbita moschata Duchesne and 20 accessions of Cucurbita maxima Duchesne preserved in the Cucurbit Germplasm Collection of the Federal Rural University of the Semi-Arid Region (UFERSA). Four commercial controls were also used, two for each species. In both studies, three isolates of M. phaseolina were inoculated, which are kept at the Fungal Culture Collection of UFERSA: MM1531 (GenBank identificatio: MM1531 (GenBank identification: MN136199), ME249 and ME250. Plants received inoculum of the pathogen in pieces of colonized toothpick and were evaluated for the incidence and severity of the disease. Both species showed resistant plants, but with varied frequencies for the accessions. Considering the species, the frequencies observed for C. moschata were higher than those observed for C. maxima. The accessions of C. moschata ABPUN 206 F1, P114-1, P160-2, P11-2, P114-6, P14-02 and P97-1 show different levels of resistance to the Macrophomina phaseolina isolates MM1531, ME-249 and ME-250.Thus, for an efficient selection in a breeding program for the characteristic, the selection strategies adopted must consider each source of resistance in isolationand not each species.

RESUMO: O melão amarelo (Cucumis melo L.) é uma hortaliça de grande importância econômica. Atualmente as cultivares plantadas, em quase sua totalidade, são híbridos. A heterose tem sido explorada no meloeiro, sendo o emprego de híbridos uniformes uma das razões do sucesso da cultura. O objetivo do presente trabalho foi avaliar o potencial agronômico de linhagens e híbridos de melão amarelo. Foi conduzido um ensaio em blocos casualizados com três repetições para avaliar 24 híbridos gerados pelos cruzamentos entre três linhagens elites utilizadas como genitores femininos (AF-01, AF-02 e AF-03), e oito linhagens utilizadas como genitores masculinos (LAM-01; LAM-02; LAM-03; LAM-04; LAM-05; LAM-06; LAM-07 e LAM-08). Foram avaliados caracteres a produção e a qualidade do fruto. Realizou-se a análise dialélica parcial para estimação das capacidades geral e específica de combinação. Observou-se efeitos aditivos e não aditivos para os caracteres produtividade, peso médio do fruto e espessura da polpa. As linhagens AF-02, LAM-02 e LAM03 são as mais promissoras por possuir maior frequência de alelos favoráveis. Os híbridos de maior destaque foram AF-02 x LAM-02, AF-02 x LAM-03, AF-02 x LAM-04 e AF-03 x LAM-06.

ABSTRACT: Melon or muskmelon (Cucumis melo L.) is of considerable economic importance in Brazil. Nearly all the cultivars currently grown are hybrids. Heterosis has been used to advantage in the melon plant, and the use of uniform hybrids is one of the reasons for successful cultivation. The aim of the present study was to evaluate the agronomic potential of yellow melon lines and hybrids. An experiment was conducted in randomized blocks with three replications to evaluate 24 hybrids generated by crosses among three elite lines used as female parents (AF-01, AF-02, and AF-03) and eight lines used as male parents (LAM-01, LAM-02, LAM-03, LAM-04, LAM-05, LAM-06, LAM-07, and LAM-08). Yield and fruit quality traits were evaluated. Partial diallel analysis was carried out to estimate general combining ability and specific combining ability. Additive and non-additive effects were observed for the yield, mean fruit weight, and pulp thickness traits. The lines AF-02, LAM-02, and LAM-03 are the most promising as parents as they more frequently have favorable alleles. The most prominent hybrids in diallel analysis were AF-02 x LAM-02, AF-02 x LAM-03, AF-02 x LAM-04, and AF-03 x LAM-06.

Abstract The aim of this study was to evaluate the genotypic performance of twelve Piel de Sapo melon hybrids in the Mossoró-Assú agricultural region, state of Rio Grande do Norte, Brazil, using mixed models. Adaptability and stability of the predicted genotypic values were studied by the Harmonic Mean of the Relative Performance of Genotypic Values (HMRPGV) procedure. The traits evaluated were yield (Mg ha-1) and soluble solids content (ºBrix). The genotype x environment interaction was significant for the two variables in all the hybrid groups evaluated, with predominance of the complex part of the interaction, and this makes the selection process more difficult. Considering the genotypic values predicted, the HMRPGV method allowed identification of hybrids with greater genotypic adaptability and stability. The experimental hybrids HP-09 and HP-06 were more promising for growing in the Mossoró-Assú agricultural region since they have high stability and adaptability, as well as high yield and soluble solids.

RESUMO O estudo de genótipos, em diferentes ambientes ou épocas de cultivo, tem grande importância para a cultura da alface, cuja expressão do potencial produtivo é influenciada pelas condições ambientais. Deste modo, a aplicação da metodologia de modelos mistos (REML/BLUP) tem sido utilizada para avaliar os efeitos da interação genótipo x ambiente. Assim, o objetivo do presente trabalho foi estimar os parâmetros de adaptabilidade e estabilidade fenotípica que estão relacionados a produção de cultivares de alface em região de clima semiárido utilizando modelo misto. Avaliaram-se doze cultivares de alface dos grupos Americana (Angelina, Amélia, Tainá), Crespa (Scarlet, Vera, Isabela, Vanda), Mimosa (Mila, Mimosa, Lavínia) e Lisa (Elisa e Regiane). O delineamento utilizado foi em blocos casualizados completos, com quatro repetições. Os parâmetros genéticos foram estimados pela metodologia REML/BLUP e a seleção simultânea da produtividade, estabilidade e adaptabilidade dos genótipos, baseou-se no método da média harmônica da performance relativa dos valores genotípicos preditos (MHPRVG). As cultivares Amélia, Angelina e Tainá sobressaíram-se para a produtividade. As cultivares Regiane e Elisa foram responsivas para o número de folhas e produtividade, sendo as mais indicadas para o cultivo nas condições de Mossoró. Os grupos Crespa e Mimosa não foram adaptados e estáveis nas condições ambientais em que foram conduzidos os ensaios. Recomenda-se que mais genótipos de todos os grupos de alface sejam testados e que, sobretudo, programas, para obter genótipos com maior resistência ao calor, produtivos e com boa qualidade, sejam desenvolvidos em regiões semiáridas.

ABSTRACT Studies on lettuce genotypes growing in different environments and planting seasons are important for this crop because its productive potential is greatly affected by environmental conditions. The mixed model methodology (REML/BLUP) have been used to evaluate the effects of the genotype-environment interaction on crops. Thus, the objective of the present work was to estimate parameters of adaptability and the phenotypic stability in lettuce cultivars grown in a semiarid region of Brazil, using a mixed model. Twelve lettuce cultivars from the Americana (Angelina, Amélia, and Tainá), Crespa (Scarlet, Vera, Isabela, and Vanda), Mimosa (Mila, Mimosa, and Lavínia), and Lisa (Elisa and Regiane) groups were evaluated. The experiments were conducted in a randomized complete block design, with four replications. The genetic parameters were estimated by the REML/BLUP method; and the simultaneous selection of productivity, stability, and adaptability of the genotypes was based on harmonic means of relative performances of predicted genotypic values (HMRPPGV). The Amélia, Angelina, and Tainá cultivars stood out for yield. The Regiane and Elisa cultivars presented significant responses in yield and number of leaves and are the most suitable for growing in the environmental conditions of Mossoró, RN, Brazil. The Crespa and Mimosa groups presented no stability and adaptability to the environmental conditions in which the experiments were conducted. More genotypes of all lettuce groups should be evaluated, and breeding programs should be developed to obtain genotypes with higher productivity, quality, and resistance to heat for semiarid regions.

RESUMO A podridão-do-colo do meloeiro, causada por Fusarium solani (Mart.). e a murcha do Sclerotium, causada pelo patógeno Sclerotium rolfsii Sacc., são importantes doenças que acometem o meloeiro. Metodologias adequadas de inoculação destes patógenos nas culturas são de grande importância para diversos estudos envolvendo o controle destas doenças, como a resistência de acessos de meloeiro. Desta forma, este trabalho teve como objetivo determinar métodos de inoculação de F. solani e S. rolfsii em meloeiro, com intuito de verificar uma metodologia de inoculação adequada e eficiente para estudos com os referidos patógenos. Dois experimentos foram conduzidos em casa de vegetação. No primeiro experimento estudou-se 4 métodos de inoculação para F. solani em 5 acessos de meloeiro. O segundo, testou-se 5 métodos de inoculação para S. rolfsii em 4 acessos da mesma cultura. O delineamento experimental foi em blocos casualizados com 5 repetições. Os métodos de inoculação utilizados foram: inoculação com palito de dente colonizado com estruturas do patógeno, discos de BDA com estruturas do fungo, infestação do solo com arroz colonizado com estruturas do fungo e infestação do solo com milho colonizado com estruturas do fungo. Para o S. rolfsii foi acrescentado o método dos escleródios. Em ambos os experimentos foram utilizadas sementes de melão das cultivares Iracema, Olimpic, A-16, A-50 e A-15, exceto A-15 para o fungo S. rolfsii. Utilizou-se a macro SAS F1_LD_F1 para realizar a análise de variância. Para o patógeno F. solani, as metodologias do palito e arroz infestado foram as mais eficientes. Já o método do arroz infestado foi o mais eficiente para o fungo S. rolfsii.

ABSTRACT Melon rot, caused by Fusarium solani (Mart.), and Sclerotium wilt, caused by the pathogen Sclerotium rolfsii Sacc, are important diseases that affect melon plants. Suitable methodologies for the inoculation of these pathogens in plants are of great importance to studies involving the control of these diseases, such as resistance of different melon accessions. Thus, the present study aimed to determine methods for the inoculation of F. solani and S. rolfsii in melon plants in order to verify a suitable and efficient inoculation methodology for studies with these pathogens. Two experiments were conducted in a greenhouse. In the first experiment, 4 inoculation methods were studied for F. solani in 5 melon accessions. In the second experiment, 5 inoculation methods were tested for S. rolfsii in 4 accessions of the same crop. Experimental design was in randomized blocks with 5 replicates. The used inoculation methods were: inoculation with toothpicks colonized with structures of the pathogen, PDA discs with structures of the fungus, soil infestation with rice colonized with structures of the fungus, and soil infestation with corn colonized with structures of the fungus. For S. rolfsii, the method of sclerotia was added. In both experiments, melon seeds of the cultivars Iracema, Olimpic, A-16, A-50 and A-15 were employed, except A-15 for S. rolfsii. The SAS F1_LD_F1 macro software was adopted to perform the analysis of variance. For F. solani, toothpick and infested rice were the most efficient methodologies. Infested rice was the most efficient method for the fungus S. rolfsii.

Resumo A adoção de híbridos de melão pelo setor produtivo requer uma prévia avaliação quanto à produtividade, qualidade e vida útil pós-colheita de seus frutos. Devido as diferentes condições ambientais em que os híbridos são avaliados, espera-se que ocorra acentuada interação genótipos x ambientes e que a mesma tenha um importante papel na manifestação fenotípica. Diante disso, o presente trabalho teve como objetivo avaliar híbridos de melão no Estado do Rio Grande do Norte. Estimou-se a adaptabilidade e estabilidade dos valores genotípicos preditos pelo método MHPRVG (Média Harmônica da Performance Relativa dos Valores Genéticos) para os caracteres: produtividade e teor de sólidos solúveis (SS) no período de 2010 a 2015. Em 2017, avaliou-se os híbridos mais promissores em condições de cultivo comercial para produtividade, qualidade e vida útil pós-colheita. Verificou-se interação genótipos x ambientes para produtividade e SS com predomínio da parte complexa da interação. Os híbridos HC-02 e HC-11 são promissores para cultivo nas condições de semiárido do Agropolo Mossoró-Assu por possuir elevadas estabilidade, adpatabiliadade, produtividade e qualidade dos frutos. Os híbridos HC-02, HC-11 e ‘Hy Mark’ possuem vida útil pós-colheita que permitem sua comercialização no mercado externo.

Abstract The adoption of melon hybrids by the productive sector necessitates a prior evaluation of the productivity, quality and shelf life of their fruits. Due to the different environmental conditions under which the hybrids are evaluated, an accentuated genotype-environmental interaction is expected to become apparent and likewise play important role in manifestation of phenotypic traits. The objective of the present work was to evaluate melon hybrids in the state of Rio Grande do Norte. The adaptability and stability of the genetic values were estimated by the HMRPGV method (Harmonic Mean of the Relative Performance of Genetic Values) for the following traits: yield, and soluble solids (SS) content in the period between 2010 to 2015. In 2017, the more promising melon hybrids from previous trials were evaluated under commercial conditions to determine the fruit’s yield, quality and shelf life. The predominance of the complex part of the genotype-environmental interaction was verified for both yield and SS content. Hybrids HC-02 and HC-11 show promising traits when grown under the conditions of the Mossoró-Assu Agricultural Complex owing their high stability, adaptability, yield, and fruit quality. Hybrids HC-02, HC-11, and ‘Hy Mark’ are suitable as international exports due to their long shelf life.

Resumo No meloeiro (Cucumis melo), a resistência ao Cucumber mosaic virus (CMV) e o conhecimento sobre características reprodutivas são importantes para escolha de genitores de populações de melhoramento. Desta forma, objetivou-se verificar se diferentes acessos de meloeiro em um banco de germoplasma brasileiro possuem resistência ao CMV (estirpe FNY) e polimorfismo, em locos marcadores de resistência, bem como se há diversidade genética quanto à resistência e aos descritores florais e reprodutivos, visando ao melhoramento genético. Os sintomas ocasionados por CMV foram avaliados em 52 genótipos de meloeiro, os quais foram genotipados com três marcadores de polimorfismos de nucleotídeos únicos, relacionados com resistência ao CMV. Uma amostra de 24 genótipos foi caracterizada quanto a descritores florais e reprodutivos. A variação significativa nos níveis de resistência possibilitou classificar os acessos em cinco grupos. Os polimorfismos detectados por meio dos marcadores distribuíram-se de forma independente nesses grupos, consistente com a natureza quantitativa oligogênica da resistência. Houve diferenças significativas entre acessos nos descritores florais e reprodutivos. Por meio da estratégia multivariada Ward-MLM, esses acessos distribuíram-se em sete grupos com características reprodutivas distintas, incluindo-se variabilidade em viabilidade polínica. Portanto, há acessos brasileiros de meloeiro que possuem resistência ao CMV, bem como variabilidade nas características reprodutivas.

Abstract In melon (Cucumis melo) the resistance to Cucumber Mosaic Virus (CMV) and the knowledge about reproductive characteristics are important for selection of genitors for breeding population. So, the objective was to investigate if different accessions of melon from a Brazilian germplasm bank have variability in resistance to CMV (FNY strain) and polymorphism in resistance loci markers, as well as whether there is genetic diversity on resistance and floral morphology descriptors and reproductive biology, aiming at genetic improvement. The symptoms induced by CMV were evaluated in 52 melon genotypes which were genotyped with three markers of single nucleotide polymorphisms related to CMV resistance. A sample of 24 genotypes was characterized for floral and reproductive descriptors. The significant variation in resistance levels allows classifying the accessions in five groups. The polymorphisms detected by the markers were independently distributed in these groups, consistent with the oligogenic quantitative expression of the melon’s resistance. There were significant differences between accessions in floral and reproductive descriptors. Through the Ward-MLM multivariate strategy, these accesses were distributed into seven groups with distinct reproductive characteristics, including variability in pollen viability. Therefore, there are Brazilian accessions of melon that have resistance to CMV, as well as variability in reproductive characteristics.

RESUMO Muitos fungos habitantes do solo acometem a lavoura meloeira, dentre estes, destaca-se o Myrothecium roridum que prejudica várias partes da planta, inclusive a semente. O presente trabalho estudou o efeito da inoculação de M. roridum sobre a germinação de sementes e crescimento de plântulas de acessos/cultivares de meloeiro, visando identificar genótipos com menor sensibilidade. O experimento foi realizado em delineamento inteiramente casualizados com quatro repetições em fatorial 38 x 2, em que o primeiro fator correspondeu aos acessos/cultivares, com 38 níveis, e o segundo, a inoculação, com dois níveis: com e sem inoculação. Foram avaliadas as características relativas à germinação (porcentagem e velocidade de emergência) e ao crescimento da plântula (altura, diâmetro do colo, peso seco da plântula e índice de área foliar). O fungo M. roridum afetou de forma negativa, a germinação, a velocidade de emergência e o desenvolvimento de plântulas dos acessos de meloeiro. Os acessos com menor sensibilidade ao fungo M. roridum foram A-6, A-8, A-19, A-22, A-39 e A-45.

ABSTRACT A large number of soilborne fungi affect melon crop, among which the species Myrothecium roridum is highlighted for attacking various parts of the plant, including the seed. The present study investigated the effect of Myrothecium roridum inoculation on seed germination and seedling growth of melon accessions/cultivars in order to identify the least sensitive genotypes. The experiment was carried out in completely randomized design with four replicates in 38 x 2 factorial arrangement, in which the first factor corresponded to the accessions/cultivars, with 38 levels, while the second factor, inoculation, had two levels: with and without inoculation. The evaluated traits were related to germination (emergence percentage and rate) and seedling growth (height, collar diameter, seedling dry weight and leaf area index). The fungus M. roridum negatively affected the germination, emergence rate and seedling development of melon accessions. The accessions with least sensitivity to the fungus M. roridum were: A-6, A-8, A-19, A-22, A-39 and A-45.
