RESUMO As espécies vegetais apresentam diferentes mecanismos de adaptação no que diz respeito à sobrevivência das sementes à dessecação, prevenindo a destruição celular durante a perda de água. O conhecimento desses mecanismos é de grande importância para o entendimento de como ocorre os processos de formação/maturação de sementes e dos processos envolvidos na germinação. Dessa forma, o objetivo deste trabalho foi determinar as alterações fisiológicas e enzimáticas durante o processo de maturação das sementes de melão da cultivar “BRS Anton”, obtidas de frutos em diferentes estádios de maturação e submetidas ao armazenamento pós-colheita, por meio da ação das proteínas totais e das enzimas catalase, peroxidase e superóxido dismutase. Os frutos de melão foram cultivados em casa de vegetação na Embrapa Hortaliças e colhidos em cinco épocas distintas: aos 30, 45, 60, 75 e 90 dias após a antese (DAA). Em cada época, foram colhidos 30 frutos, sendo que 15 frutos tiveram suas sementes extraídas imediatamente após a colheita e 15 frutos foram armazenados por 15 dias. Após o beneficiamento e secagem, as sementes foram submetidas às seguintes análises: germinação, primeira contagem, análise do perfil de proteínas e atividade de enzimas antioxidantes (proteínas totais, peroxidase, catalase e superóxido dismutase). Observou-se que as atividades das enzimas antioxidativas a partir dos 60 DAA sob armazenamento, apresentou um comportamento mais ordenado reafirmando os resultados dos testes fisiológicos. Demostrando que as sementes neste ponto de maturação fisiológica, encontra-se bem formadas e que a secagem não provocou danos em suas membranas celulares.

ABSTRACT Plant species have different adaptation mechanisms regarding seed survival to desiccation, preventing cellular destruction during the water loss. The knowledge of these mechanisms is of great importance for the understanding of how the seed formation/maturation processes and the processes involved in germination occur. Thus, this study aimed to determine physiological and enzymatic changes during the maturation process of melon seeds of the cultivar BRS Anton, obtained from fruits at different maturation stages and subjected to post-harvest storage, by the action of total proteins and the enzymes catalase, peroxidase, and superoxide dismutase. Melon fruits were grown in a greenhouse at Embrapa Vegetables and harvested at five different times: 30, 45, 60, 75, and 90 days after anthesis (DAA). Thirty fruits were harvested in each season and 15 fruits had their seeds extracted immediately after harvest and the other 15 fruits were stored for 15 days. After processing and drying, the seeds were submitted to the following analyses: germination, first count, protein profile analysis, and activity of antioxidant enzymes (total proteins, peroxidase, catalase, and superoxide dismutase). The activity of the antioxidant enzymes from 60 DAA under storage showed a more orderly behavior, reaffirming the results of physiological tests. It shows that the seeds at this point of physiological maturation are well-formed and drying caused no damage to their cell membranes.

RESUMO A ‘BRS Paranoá’ foi obtida pelo programa de melhoramento genético de cenoura da Embrapa Hortaliças, visando obter uma cultivar adaptada à maioria das regiões produtoras de cenoura em sistema orgânico. Essa cultivar foi desenvolvida através de seleção de progênies meias-irmãs e validada no Brasil. A ‘BRS Paranoá’ é indicada para semeadura nas estações primavera/verão. Essa cultivar apresenta elevada resistência à queima das folhas causada pela associação fungo e bactéria (Alternaria dauci, Cercospora carotae e Xanthomonas hortorum pv. carotae) com desfolhamento entre 20 a 25% aos 100 dias após o plantio (DAP). Além disso, apresenta elevada estabilidade de produção e elevado teto produtivo, superando 32 t ha-1. A ‘BRS Paranoá’ possui ciclo precoce, com a colheita podendo ser realizada a partir dos 85 DAP. As raízes apresentam comprimento variando de 16 a 22 cm, diâmetro de 3 cm, coloração alaranjada intensa, formato cilíndrico e ausência de defeitos fisiológicos externos ou internos. A qualidade pós-colheita foi avaliada e a acidez titulável encontrada foi de 0,20%, sólidos solúveis de 6,9oBrix, carotenoides totais de 78,40 µg g-1 e ß-caroteno (pró-vitamina A) de 22,69 µg g-1. A ‘BRS Paranoá’ foi registrada e protegida pelo Ministério da Agricultura, Pecuária e Abastecimento e suas sementes têm sido comercializadas pelas principais empresas de sementes de hortaliças no Brasil.

ABSTRACT ‘BRS Paranoá’ was obtained by the carrot breeding program of Embrapa Hortaliças, aiming to obtain a cultivar adapted to most carrot-producing regions in an organic system. It was developed through selection of half-sib progenies and validated in Brazil. ‘BRS Paranoá’ is indicated for sowing in the spring/summer seasons especially because it displays high resistance to leaf blight caused by the fungal and bacterial association (Alternaria dauci, Cercospora carotae, and Xanthomonas hortorum pv. carotae) with defoliation between 20 to 25% at 100 days after planting (DAP), eliminating the need to control this disease with chemical pesticides. In addition, it shows high production stability and an outstanding production exceeding 32 t ha-1. ‘BRS Paranoá’ is an early maturity cultivar, and the harvest can be carried out from 85 DAP when the commercial roots range from 16 to 22 cm, 3 cm diameter, intense orange color, cylindrical shape, and no external or internal physiological defects. The postharvest quality was evaluated and the titratable acidity found was 0.20%, 6.9oBrix soluble solids, 78.40 µg g-1 total carotenoids, and 22.69 µg g-1 ß-carotene (provitamin A). ‘BRS Paranoá’ was registered and protected by the Ministry of Agriculture, Livestock and Food Supply and its seeds have been marketed by the main vegetable seeds companies in Brazil.

ABSTRACT. Chickpea is a legume with nutrient-rich grains important for human feeding. Tobacco budworm, Chloridea virescens (Lepidoptera: Noctuidae), is one of the most major pests of chickpea (Cicer arietinum) in Brazil. This pest damages leaves, flowers, pods, and grains. Plant resistance to insects is an important tactic of pest management, which usually facilitates and reduce costs of implementing an Integrated Pest Management for farmers. Thus, this study aimed to evaluate the resistance in chickpea cultivars to C. virescens. Six chickpea cultivars were evaluated for antixenosis, initial antibiotic parameters, and behavior under field conditions. The cultivars BRS Kalifa and BRS Cícero were less attractive in a free-choice test, while Jamu 96 and BRS Kalifa were less attractive in a no-choice test. BRS Kalifa and BRS Toro leaves had a higher trichome density. Jamu 96 and BRS Toro had higher contents of oxalic and malic acids. C. virescens larvae in BRS Cícero, BRS Toro, and BRS Kalifa showed the lowest weights. Jamu 96 pods were the least damaged, and BRS Aleppo and Jamu 96 had the highest yields. The chickpea cultivars Jamu 96 and BRS Aleppo, which had resistance levels and mechanisms, can be used in integrated pest management programs to control C. virescens.

Resumo: A definição de metodologias que permitam avaliar o potencial fisiológico das sementes de grão-de-bico é importante para assegurar a comercialização de lotes com alto desempenho em campo. Assim, objetivou-se determinar as condições mais adequadas para a condução do teste de condutividade elétrica em sementes de grão-de-bico visando a classificação de lotes quanto ao vigor. Para tanto, sementes de cinco lotes da cultivar BRS Aleppo e de quatro lotes da cultivar Cícero foram submetidas a diferentes testes para a caracterização da sua qualidade inicial e os resultados comparados com os obtidos no teste de condutividade. Para avaliar a condutividade elétrica, 5 períodos de embebição (2, 4, 8, 12 e 24 horas) foram combinados a 4 volumes de água (75, 100, 150 e 250 mL) sob a temperatura de 25 °C. O teste de condutividade elétrica é eficiente para a avaliação do potencial fisiológico de sementes de grão-de-bico fornecendo informações equivalentes a outros testes de vigor. Para a condução do teste de condutividade elétrica recomenda-se a utilização de 50 sementes imersas em 150 mL de água, por 24 horas, a 25 °C.

Abstract: The definition of methodologies that allow evaluating the physiological potential of chickpea seeds is important to ensure the commercialization of lots with high performance. Thus, this research aimed to determine the most suitable conditions for conducting the electrical conductivity test in chickpea seeds to classify lots according to vigor level. Therefore, seeds from five lots of cultivar BRS Aleppo and four lots of cultivar Cícero were subjected to tests to characterize their initial quality and the results were compared with those obtained in the electrical conductivity test. To assess electrical conductivity, five soaking periods (2, 4, 8, 12, and 24 hours) were combined with four volumes of water (75, 100, 150, and 250 mL) at 25 °C temperature. The results show that the electrical conductivity test is efficient in evaluating the physiological potential of chickpea seeds, providing information equivalent to other vigor tests. For the electrical conductivity test, it is recommended to use 50 seeds soaked in 150 mL of water, for 24 h, at 25 °C.

RESUMEN El objetivo de esta investigación fue evaluar la dinámica poblacional de los nematodos Heterodera glycines raza 5 y Pratylenchus brachyurus en cultivos sucesivos de soya y garbanzo. El experimento se llevó a cabo en Campos Novos dos Parecis, estado de MT, Brasil, de febrero a mayo de 2017. Se sembraron seis cultivares de garbanzo en febrero y se evaluaron en un área naturalmente infestada de 60 ha. Se recolectaron cinco muestras de suelo al azar en ubicaciones georreferenciadas y equidistantes, formando una muestra compuesta por replicación. Se recolectaron cinco plantas por punto georreferenciado y también nuevas muestras de suelo a intervalos regulares de aproximadamente 30 d. Los nematodos se extrajeron, identificaron y contaron. Los datos fueron sometidos a análisis de varianza y regresión. Se observó una reducción en los niveles poblacionales de H. glycines raza 5 a lo largo del ciclo del garbanzo, lo que indica que es posible cultivar esta especie en sucesión con soya en presencia de este nematodo. Sin embargo, debido al mantenimiento en la población de P. brachyurus en las raíces, no se recomienda la rotación de soya con garbanzos en campos naturalmente infestados con este nematodo.

ABSTRACT The objective of this research was to evaluate the population dynamics of the nematodes Heterodera glycines race 5 and Pratylenchus brachyurus in a succession crop of soybeans and chickpeas. The experiment was carried out in Campos Novos dos Parecis, MT State, Brazil, from February to May 2017. Six chickpea cultivars were planted in February and evaluated in a naturally infested area of 60 ha. Five soil samples were collected at random in georeferenced and equidistant locations, forming a composite sample by replication. Five plants per georeferenced point and new soil samples were collected at regular intervals of about 30 d. The nematodes were extracted, identified, and counted. Data were subjected to variance and regression analyses. A reduction in the population levels of H. glycines race 5 was observed throughout the chickpea cycle, indicating that this species can be cultivated in succession with soybeans in the presence of this nematode. However, due to the maintenance in the population of P. brachyurus in the roots, rotation of soybean with chickpeas is not recommended in fields naturally infested with this nematode.

RESUMO A produção de lentilha apresenta tendência de crescimento no Brasil devido a uma forte demanda do mercado nacional e internacional por esta hortaliça leguminosa. Apesar da importância econômica, existem poucas cultivares disponíveis no mercado brasileiro. O objetivo deste estudo foi avaliar 48 linhagens de lentilha e uma cultivar comercial (Silvina), com base em características morfoagronômicas, e identificar genótipos que possam ser empregados no melhoramento genético. Foram utilizados 21 descritores morfoagronômicos (nove qualitativos e doze quantitativos) que evidenciaram a existência de variabilidade genética entre os genótipos estudados. A associação das técnicas de agrupamento com os dados de caracterização possibilitou a identificação de genótipos geneticamente divergentes e, ao mesmo tempo, superiores quanto a características agronômicas: FLIP2010-8L e FLIP2010-12L (grupo de similaridade I), FLIP2010-99L, FLIP2010-20L e FLIP2010-106L (grupo II), FLIP90-25L e 6031 (grupo III), FLIP2007-16L (grupo IV) e a cultivar comercial Silvina (grupo V). Esses genótipos apresentam potencial para serem utilizados em programas de melhoramento genético da espécie.

ABSTRACT Lentil production has been increasing in Brazil, due to strong national and international demand. Despite the economic importance, few cultivars are available on the Brazilian market. The aim of this study was to evaluate 48 lentil inbred lines and one commercial cultivar (Silvina), based on morpho-agronomic traits and to identify genotypes which can be used in a plant breeding program. Twenty-one morpho-agronomic descriptors (nine qualitative and twelve quantitative) were used. The descriptors showed genetic variability among the genotypes. Clustering techniques using characterization data allowed to identify genetically divergent genotypes as well as identify superior genotypes in relation to agronomic traits: FLIP2010-8L and FLIP2010-12L (similarity group I), FLIP2010-99L, FLIP2010-20L and FLIP2010-106L (group II), FLIP90-25L and 6031 (group III), FLIP2007-16L (group IV) and the commercial cultivar Silvina (group V). We concluded that these genotypes have the potential to be used in lentil breeding programs.

RESUMO O estádio de maturação das sementes influencia a sua qualidade fisiológica logo após a colheita. O objetivo do trabalho foi avaliar as alterações anatômicas, histoquímicas e fisiológicas em sementes de grão-de-bico ‘BRS Aleppo’ obtidas de vagens em diferentes estádios de maturação. Foram colhidas vagens em cinco estádios de maturação: verde, verde-amarelo, amarelo, amarelo-dourado e marrom. Para caracterização anatômica e histoquímica os cortes foram corados com Azul de Toluidina, Lugol, Xylidine Ponceau (XP), Vermelho de Rutênio. Para avaliação das alterações fisiológicas, as sementes foram secas até 12% de umidade. Foram realizados os seguintes testes: grau de umidade, germinação, porcentagem e índice de velocidade de emergência e condutividade elétrica. Utilizou-se o delineamento inteiramente casualizado com quatro repetições. Verificou-se que com o desenvolvimento das sementes de grão-de-bico as camadas mais internas do tegumento sofrem colapso. O acúmulo de amido e proteína já está em andamento nas sementes de vagens verdes. A pectina se destaca dentre os compostos constituintes de parede celular dos esclereídes do tegumento. Em geral, sementes obtidas de vagens com coloração externa amarela, amarelo-dourado e marrom apresentam maior germinação e vigor.

ABSTRACT The maturation stage of seeds affects their physiological quality soon after harvest. The aim of this study was to evaluate the anatomical, histochemical, and physiological changes in chickpea ‘BRS Aleppo’ seeds obtained from pods in different maturity stages. Pods were collected in five maturity stages: green, yellow-green, yellow, golden-yellow, and brown. For anatomical and histochemical characterization, sections were stained with toluidine blue, Lugol’s solution, Xylidine-Ponceau (XP), and ruthenium red. For evaluation of physiological changes, seeds were dried to 12% (wet basis) water content. The following tests were performed: seed water content, germination, percentage and speed of seedling emergence, and electrical conductivity. A completely randomized experimental design was used, with four replications. As the chickpea seeds develop, the innermost layers of the seed coat collapse. Accumulation of starch and protein were already observed in the seeds of green pods and pectin was prominent compound of the seed coat sclerids. In general, chickpea seeds from yellow, golden-yellow and brown pods have greater germination and vigor.

Resumo: O cultivo de grão-de-bico vem se expandindo no Brasil e, com isto, a demanda por sementes de alta qualidade também aumenta. Neste trabalho, objetivou-se adequar metodologia para avaliação do vigor de sementes de grão-de-bico pelo teste de envelhecimento acelerado, bem como analisar se a atividade de enzimas antioxidantes apresenta relação com o potencial fisiológico das sementes. Sementes de cinco lotes, cv. BRS Aleppo, e de quatro lotes da cv. Cícero, foram inicialmente submetidas a testes para a caracterização do seu potencial fisiológico. O teste de envelhecimento acelerado foi conduzido pelo método tradicional (100% UR) e com solução saturada de NaCl (76% UR), ambos a 41 °C, por 24, 36, 48 e 72 h. Foram determinadas também a atividade das enzimas SOD, CAT e APX. Para o teste de envelhecimento acelerado, recomenda-se a utilização de 41 °C/48 h com solução saturada NaCl para a avalição do vigor das sementes de grão-de-bico. A atividade das enzimas CAT e APX pode ser relacionada ao potencial fisiológico das sementes, o que não ocorre para a SOD.

Abstract: The chickpea crop has been expanding in Brazil and, thus, the demand for high quality seeds along with it. The aim of this study was to adapt the methodology for evaluation of chickpea seed vigor by the accelerated aging test and to analyze whether antioxidant enzyme activity is related to the physiological potential of these seeds. Seeds from five seed lots of the cv. BRS Aleppo and from four seed lots of the cv. Cícero were initially tested to characterize their physiological potential. The accelerated aging test was conducted by the traditional method (100% RH) and with a saturated NaCl solution (76% RH), both at 41 °C, for 24, 36, 48, and 72 h. The activity of the SOD, CAT, and APX enzymes were also determined. The accelerated aging test using 41 °C/48 h with a saturated NaCl solution is recommended for assessment of chickpea seed vigor. The activity of CAT and APX enzymes may be related to the physiological potential of these seeds, but this is not the case with SOD.

Resumo O objetivo deste trabalho foi avaliar a resistência de genótipos de grão-de-bico (Cicer arietinum) aos nematoides da galha (Meloidogyne javanica) e das lesões (Pratylenchus brachyurus), bem como verificar como estas espécies afetam essa cultura em condições de campo. O experimento foi realizado no bioma Cerrado, no estado de Goiás, Brasil, em área naturalmente infestada. Foram avaliados as cultivares BRS Aleppo, Cícero, BRS Cristalino, BRS Toro e BRS Kalifa e o genótipo Jamu 96. As variáveis avaliadas foram: fator de reprodução dos nematoides, massa fresca da parte aérea e do sistema radicular, matéria seca da parte aérea, matéria seca total das plantas, índice de clorofila, altura das plantas e rendimento de grãos. Pratylenchus brachyurus se multiplica mais do que M. javanica e diminui massa fresca da raiz, altura da planta e rendimento de grãos. Todos os genótipos são resistentes a M. javanica, principalmente Jamu 96 e a cultivar BRS Kalifa. Jamu 96 e a cultivar Cícero são suscetíveis a P. brachyurus. BRS Kalifa é a cultivar mais resistente a ambos os nematoides e apresenta maior rendimento de grãos.

Abstract The objective of this work was to evaluate the resistance of chickpea (Cicer arietinum) genotypes to the root-knot (Meloidogyne javanica) and root-lesion (Pratylenchus brachyurus) nematodes, as well as to verify how these species affect this crop in field conditions. The experiment was carried out in the Cerrado biome in the state of Goiás, Brazil, in a naturally infested area. The BRS Aleppo, Cícero, BRS Cristalino, BRS Toro, and BRS Kalifa cultivars and the Jamu 96 genotype were evaluated. The assessed variables were: nematode reproduction factor, fresh mass of the aerial part of the plant and of the root system, dry matter of the aerial part of the plant, total dry matter of the plants, chlorophyll index, height of the plants, and grain yield. Pratylenchus brachyurus multiplies more than M. javanica, decreasing root fresh matter, plant height, and grain yield. All genotypes are resistant to M. javanica, especially Jamu 96 and cultivar BRS Kalifa. Jamu 96 and cultivar Cícero are susceptible to P. brachyurus. BRS Kalifa is the cultivar most resistant to both nematodes and shows the highest grain yield.

RESUMO A análise de imagens tem sido usada recentemente para avaliar a qualidade das sementes e elucidar alguns aspectos da fisiologia das sementes, incluindo testes de raios X e outros recursos. O objetivo deste estudo foi examinar se o raios X pode ser utilizado para detectar anormalidades físicas e relacionar sua ocorrência com o potencial fisiológico de sementes de abóbora tipo tetsukabuto. Os frutos foram colhidos em diferentes estádios de maturação (15, 30, 45, 60 e 75 dias após a antese - DAA) e armazenados por 20 dias. As sementes foram radiografadas com intensidade de 25 kV por 40 segundos e em seguida foi realizado um teste de germinação em alternância de 20-30 ºC. Após quatro e oito dias, foram registrados percentuais de plântulas normais e anormais e sementes mortas. Os resultados de ambas as contagens foram avaliados comparativamente com imagens da morfologia interna de cada semente, obtidas por meio de teste de raios X. A análise radiográfica é eficaz na detecção de sementes de abóbora inteiras, malformadas e vazias. A utilização da técnica de raios X é eficiente na detecção de sementes cheias, mal formadas e vazias de abóbora, e ainda confirma os resultados obtidos nos testes fisiológicos, onde as sementes provenientes de frutos colhidos aos 60 DAA e armazenados por 20 dias apresentaram uma qualidade superior.

ABSTRACT Analysis of images has been recently used in order to both assess seed quality and elucidate some aspects of seed physiology, including X-ray tests and other resources. The objective of this study was to examine whether the X-ray may be successfully used to detect physical abnormalities, and relate their occurrence with physiological potential of tetsukabuto type squash seeds. Fruits were harvested at different maturation stages (15, 30, 45, 60 and 75 days after anthesis - DAA) and temporarily or not stored for 20 days. Seeds were radiographed using an intensity of 25 kV for 40 seconds and then germinated under alternating temperatures of 20-30 ºC. After four and eight days, percentages of normal and abnormal seedlings and dead seeds were recorded. Results of both counts were evaluated comparatively with images of the internal morphology of each seed, obtained by means of X-ray test. The radiographic analysis is effective in detecting full, malformed and empty squash seeds. This confirmed information provided by the germination test, in which seeds harvested from 60 DAA and stored for 20 days were better in physiological potential.

Resumo: O objetivo deste trabalho foi avaliar o método de inoculação de sementes de cenoura com meio batata-dextrose-ágar (BDA) acrescido de manitol. O fungo escolhido para testar a metodologia foi o Alternaria radicina. O restritor hídrico foi adicionado ao meio BDA de modo a obter os potenciais osmóticos de 0,6 MPa, 0,8 MPa, 1,0 MPA e 1,2 MPa. O crescimento micelial foi avaliado, e não foi observado inibição do crescimento em nenhum dos potenciais osmóticos, em relação a testemunha. As sementes foram distribuídas sobre as colônias fúngicas para a infecção, onde permaneceram por 48, 72, 96, 120 e 144 horas nos potenciais osmóticos 0,0 MPa (controle), 0,6 MPa, 0,8 MPa, 1,0 MPa e 1,2 MPa, respectivamente. Para avaliar os efeitos da colonização foram realizados testes de germinação, sanidade e emergência. Os potenciais osmóticos avaliados apresentaram porcentagens semelhantes de plântulas anormais e sementes mortas no teste de germinação. No teste de sanidade, maiores porcentagens de infecção foram observadas nos potenciais osmóticos 1,0 MPa e 1,2 MPa. O potencial -1,2 MPa apresentou a menor porcentagem de emergência. Assim, o uso de manitol -1,2 MPa é sugerido como um método eficaz para a infecção de sementes de cenoura com para fins de pesquisa.

Abstract: This study aimed at evaluating the method of inoculation of carrot seeds in a potato-dextrose-agar medium (PDA) added of mannitol. Alternaria radicina was the fungus chosen to test such methodology. The osmosis-controlling agent was employed to obtain the osmotic potentials of 0.6 MPa, 0.8 MPa, 1.0 MPa and 1.2 MPa. In comparison with the control treatment, no significant mycelial growth inhibition was noticed at any of the osmotic potentials considered. The seeds were distributed over the fungal colonies for infection, where they remained for 48, 72, 96, 120 and 144 hours, at the osmotic potentials of 0.0 MPa (control), 0.6 MPa, 0.8 MPa, 1.0 MPa and 1.2 MPa, respectively. Tests of germination, sanitization and emergence were performed to evaluate the effects of colonization. Regarding the germination, the different osmotic potentials resulted in similar percentages of abnormal seedlings and dead seeds. As for the sanitization testing, higher infection rates were observed at the osmotic potentials of 1.0 MPa and 1.2 MPa. The lowest percentage of emergence was found at -1.2 MPa, therefore suggesting that this osmotic potential (produced with mannitol) might be used for infecting carrot seeds for research purposes.

RESUMO A antracnose causada pelo fungo Colletotrichum gloeosporioides f. sp. cepae é umas das principais doenças na cultura da cebola. A principal estratégia de controle deste fungo é o controle químico com fungicida sendo esta técnica utilizada na maior parte dos cultivos. Atualmente vem sendo estudados métodos de controle de doenças que sejam menos agressivos ao ambiente, mostrando-se como alternativa o uso de óleos essenciais. Diante do exposto, objetivou-se avaliar o efeito direto dos óleos essenciais de manjericão (Ocimum basilicum), capim-limão (Cymbopogon citratus (DC) Stapf.), sálvia (Salvia officinalis), tomilho (Thymus vulgaris) e citronela (Cymbopogon winterianus) como alternativa no controle do fungo C. gloeosporioides f. sp. cepae, além de determinar seu efeito sobre a qualidade fisiológica de sementes de cebola. Os óleos essenciais de capim-limão e sálvia não afetaram a qualidade fisiológica das sementes de cebola e todos os óleos essenciais utilizados promoveram a inibição da germinação de esporos de C. gloeosporioides f. sp. cepae. Os óleos essenciais de tomilho, capim-limão e citronela, na concentração de 2.000 ppm, inibiram totalmente o crescimento micelial do fungo. O tratamento com o óleo essencial de capim-limão em sementes inoculadas com patógeno proporcionou maior proteção contra o ataque de fungo.

ABSTRACT Anthracnose, one of the major diseases in the onion, is caused by the fungus Colletotrichum gloeosporioides f. sp. cepae. The main strategy for controlling this fungus is by the use of fungicide, with this technique being used on the majority of crops. Currently, methods of disease control are being studied that are less aggressive to the environment, with the use of essential oils proving to be an alternative. As such, the aim of this study was to evaluate the direct effect of the essential oils of basil (Ocimum basilicum), lemongrass (Cymbopogon citratus (DC) Stapf.), sage (Salvia officinalis), thyme (Thymus vulgaris) and citronella (Cymbopogon winterianus) as an alternative in the control of the fungus C. gloeosporioides f. sp. cepae, in addition to determining its effect on the physiological quality of onion seeds. The essential oils of lemongrass and sage did not affect the physiological quality of the onion seeds, and all the essential oils used helped to inhibit spore germination in C. gloeosporioides f. sp. cepae. The essential oils of thyme, lemongrass and citronella at a concentration of 2000 ppm completely inhibited mycelial growth in the fungus. Treatment with the essential oil of lemongrass in seeds inoculated with the pathogen gave greater protection against fungal attack.

Resumo: A alface é uma das principais espécies de hortaliças utilizadas na produção orgânica, porém, os estudos e pesquisas na área de orgânicos são considerados escassos. O objetivo do estudo foi avaliar a produtividade e qualidade de sementes de genótipos de alface produzidas em dois anos consecutivos no sistema de manejo orgânico. Foram instalados dois plantios em condições de campo aberto correspondendo às estações de outono/inverno em 2016 e 2017. Avaliaram-se 10 genótipos de alface, em quatro repetições, utilizando biofertilizante suíno no manejo e colheita manual das sementes. As avaliações efetuadas foram: produtividade das sementes em kg.ha−1; teste físico (peso de 1000 sementes); teste de germinação e de vigor (primeira contagem de germinação, índice de velocidade de germinação e massa seca de plântulas). O genótipo Cubana e os dois genótipos Grand Rapids apresentaram produtividades superiores nas condições testadas. Em 2017 foram registrados maiores pesos de sementes. Os dois genótipos Grand Rapids atingiram o padrão mínimo de germinação estabelecido para a comercialização (80%). Vanda e AC 5058 apresentaram melhores desempenho para IVG e primeira contagem, e, Cinderela para massa seca de plântulas. Tais variedades mostraram ser promissoras para o cultivo orgânico.

Abstract: is one of the main vegetable species used in organic production. However, studies and research in the organic area are scarce. This study aimed to evaluate the yield and quality of seeds of lettuce genotypes produced in two consecutive years under an organic management system. Two plantations were installed under open field conditions corresponding to autumn/winter in 2016 and 2017. Ten lettuce genotypes were evaluated in four replications using a biofertilizer made from swine manure in the management and manual harvesting of seeds. Evaluations consisted of seed yield (in kg.ha−1), physical test (1000-seed weight), and germination and vigor test (first germination count, germination rate index, and seedling dry matter). The genotype Cubana and two Grand Rapids had higher yields under the tested conditions. Higher seed weights were recorded in 2017. Both genotypes Grand Rapids reached the minimum germination standard established for commercialization (80%). Vanda and AC 5058 showed a better performance for germination rate index and first germination count, while Cinderela had a better seedling dry matter. These varieties were promising for organic cultivation.

Resumo: O potencial fisiológico das sementes está diretamente relacionado à normalidade do processo de formação delas, sob ponto de vista morfológico, estrutural e funcional. Deste modo, o objetivo deste trabalho foi avaliar a morfologia interna de sementes de tomate, híbrido BRS Nagai, em diferentes estádios de maturação, por meio da análise de imagens de raios-X, relacionando com a germinação e o vigor das sementes. As sementes foram radiografadas tendo sido suas imagens analisadas para a classificação da morfologia do embrião e mensuração do espaço livre entre o embrião e endosperma, com auxílio do programa de análise de imagens ImageJ. Estes resultados foram comparados com a qualidade fisiológica através do teste de germinação. As correlações mostraram que as sementes possuidoras de embriões classificados como normais tendem a apresentar maior germinação, e produzir plântulas normais. E àquelas que possuem maior área livre interna tendem a apresentar ausência de germinação ou originarem maiores percentuais de plântulas anormais. A análise de imagens utilizando raios-X é eficiente para o estudo da maturidade fisiológica de sementes de tomate. A análise de raios-X pode predizer o desempenho das sementes por meio da morfologia do embrião, permitindo a seleção de sementes de alta qualidade. Sementes de tomateiro de frutos com até 40 dias após a polinização apresentam baixa qualidade morfofisiológica quando comparadas com sementes extraídas de frutos totalmente maduros, a partir dos 60 dias após a polinização.

Abstract: The seed physiological potential has been directly related to normal formation process under morphological, structural and functional point of view. The objective of this study was to evaluate the internal morphology of ‘BRS Nagai’ tomato hybrid seeds at different stages of development and maturation, through the X-ray image analysis and relate the results with the seed germination and vigor. Seeds were extracted from fruits harvested at 30, 40, 50, 60 and 70 days after pollination (DAP). Subsequently, seeds were x-rayed with their images analyzed visually for the embryo morphology classification; also, the measurement of clearance between the embryo and endosperm was performed with the help of the ImageJ software. These results were compared with the seed physiological quality by germination. The correlations involving the analysis of X-ray showed that the seeds have embryos classified as normal tend to have higher vigor and produce normal seedlings. On the other hand, seeds with higher internal free area tend to have low vigor and higher percentage of abnormal seedlings. The image analysis using X-rays is efficient for the study of physiological maturity of tomato seeds. The analysis of X-rays may predict seed performance by means of the embryo morphology, permitting the selection of high quality seeds. Tomato seeds of fruits with up to 40 days after pollination have low morphophysiological quality when compared to seeds extracted from fully mature fruits, from 60 days after pollination.

Resumo: Testes rápidos são ferramentas imprescindíveis na avaliação da qualidade de sementes. O objetivo deste estudo foi adequar a metodologia do teste de tetrazólio para avaliar a qualidade fisiológica de sementes de grão-de-bico, visando a identificação dos melhores tempos e temperaturas de pré-condicionamento, e concentrações das soluções de tetrazólio. Além disso, foi proposta uma separação em classes quanto à viabilidade e vigor das sementes. Foram avaliados três lotes de sementes de grão-de-bico (lote 1 - ‘BRS Aleppo’ C1, lote 2 - ‘BRS Aleppo’ básico e lote 3 - ‘Cícero’ básico), três binômios tempo-temperatura de embebição (41 °C por 4 horas, 41 °C por 6 horas e 30 °C por 18 horas) e duas concentrações de tetrazólio (0,1% e 0,5%). As condições de embebição de 41 °C por 4 ou 6 horas e de 30 °C por 18 horas permitiram visualizar de forma clara os danos ocorridos nas sementes quando imersas em solução 0,1% de tetrazólio. A coloração intensa formada nas sementes submetidas à concentração de 0,5% do sal impediu a diferenciação dos tipos de danos. Além disso, foi possível realizar a separação em quatro classes, sendo viáveis as sementes das classes I, II e III, vigorosas as das classes I e II e não viáveis as da classe IV.

Abstract: Quick tests are essential tools to evaluate seed quality. The objective of this study was to adapt the methodology of tetrazolium test, by identifying the most suitable pre-conditioning parameters of imbibition temperature and duration, and tetrazolium solution concentrations, in order to assess the physiological quality of chickpea seeds. Also, this work proposed the separation of lots in classes, according to the viability and vigor of the seeds. Three lots of chickpeas (lot 1 - BRS Aleppo C1, lot 2 - BRS Aleppo basic, and lot 3 - Cicero) were evaluated. They were analyzed according to three combinations of imbibition temperature and duration (41 °C for 4 hours, 41 ºC for 6 hours, and 30 ºC for 18 hours) and two tetrazolium concentrations (0.1% and 0.5%). The imbibition at 41 °C for 4 or 6 hours, and at 30 ºC for 18 hours allowed clear visualization of injuries in the seeds after immersion in 0.1% tetrazolium solution. The intense coloration formed by the 0.5% solution of this salt prevented the differentiation of the types of damage. In addition, it was possible to separate the seeds into four classes: class I (viable and vigorous, without damages), class II (viable and vigorous, with superficial injuries), class III (viable and non-vigorous), and class IV (non-viable).
