Resumo Ascocotyle (Phagicola) longa é o agente etiológico da fagicolose humana. Os mugilídeos são os segundos hospedeiros intermediários. O primeiro é Heleobia australis e pássaros e mamíferos predadores de mugilídeos, os hospedeiros definitivos. A ocorrência de cistos contendo metacercárias de A. longa é descrita em mugilídeos, com até 100% de prevalência. A ampla distribuição geográfica de A. longa e seus hospedeiros intermediários, acompanhado do aumento do consumo de peixe cru ou mal cozido, pode aumentar o risco de infecção humana, portanto o objetivo do presente estudo foi verificar o padrão de distribuição de cistos, contendo metacercárias de A. longa, em mugilídeos. Amostras de tecidos e órgãos desses peixes foram processadas em liquidificador doméstico e observadas em microscópio estereoscópico à procura de cistos contendo metacercárias do digenético. Todos os 24 (100%) mugilídeos, 12 Mugil curema e 12 Mugil liza examinados apresentaram cistos contendo metacercária de A. longa. Foram observados cistos do digenético nas brânquias, coração, estômago, fígado, intestino, mesentério e tecido muscular de M. curema e de M. liza. Em M. curema, os cistos contendo metacercárias de A. longa estão distribuídos de forma aleatória por praticamente todos os tecidos e órgãos dos mugilídeos examinados.

Abstract Ascocotyle (Phagicola) longa is an etiological agent of human phagicolosis. Mugilids are the second intermediate host, the first being Heleobia australis, and mugilids predatory birds and mammals are its definitive hosts. The occurrence of cysts holding A. longa metacercariae is described in mugilids with a prevalence of up to 100%. The wide geographical distribution of A. longa and its intermediate hosts coupled with the rise in the consumption of raw or poorly cooked fish may elevate the risk of human infection. Therefore, in this study, we aimed to verify the distribution pattern of cysts holding A. longa in mugilids. The tissue and organ samples of these fish were processed in a domestic blender and examined under a stereoscopic microscope to identify the cysts holding the digenetic metacercariae. Of the 24 (100%) fish samples that were analyzed, 12 of Mugil curema and 12 of Mugil liza possessed cysts holding A. longa metacercariae. Digenetic cysts were identified to be present in the gills, heart, stomach, liver, intestines, mesentery, and muscular tissues collected from M. curema and M. liza. Conclusively, in M. curema, the cysts holding A. longa metacercariae were found to be distributed randomly throughout the fish body in almost every tissue and organ that was examined.

Resumo O objetivo deste estudo foi relatar um caso de toxoplasmose felina fatal com coinfecção com o vírus da leucemia felina (FeLV). Um gato doméstico (Felis silvestris catus) apresentou intensa dispneia e morreu três dias depois. Na necropsia, observaram-se pulmões firmes, não colabados e com múltiplas áreas brancas, além de linfoadenomegalia e esplenomegalia moderadas. No exame histopatológico, evidenciaram-se broncopneumonia intersticial necrótica acentuada e hepatite necrótica discreta associada a cistos e taquizoítas de T. gondii intralesionais positivos na imuno-histoquímica (IHC) anti-T. gondii. Evidenciou-se ainda, na medula óssea, leucemia mieloide crônica com IHC anti-FeLV positiva nas células neoplásicas. O DNA extraído dos pulmões foi positivo para T. gondii por meio da PCR-REP-529. T. gondii foi caracterizado por PCR-RFLP e pela técnica de microssatélites. Foi identificado o genótipo ToxoDB-PCR-RFLP #10, i.e., o arquétipo tipo I. A análise por microssatélites mostrou que a cepa era uma variante do tipo I, com dois alelos atípicos. Esta é a primeira vez que T. gondii clonal tipo I foi relacionado com um caso agudo de toxoplasmosis em um hospedeiro no Brasil.

Abstract The objective of the study was to report on a fatal case of feline toxoplasmosis with coinfection with the feline leukemia virus (FeLV). A domestic cat (Felis silvestris catus) presented intense dyspnea and died three days later. In the necropsy, the lungs were firm, without collapse and with many white areas; moderate lymphadenomegaly and splenomegaly were also observed. The histopathological examination showed severe necrotic interstitial bronchopneumonia and mild necrotic hepatitis, associated with intralesional cysts and tachyzoites of Toxoplasma gondii that were positive by anti-T. gondii immunohistochemical (IHC) evaluation. The bone marrow showed chronic myeloid leukemia and the neoplastic cells were positive by anti-FeLV IHC evaluation. DNA extracted from lungs was positive for T. gondii by PCR targeting REP-529. T. gondii was characterized by PCR-RFLP and by the microsatellites technique. ToxoDB-PCR-RFLP #10, i.e. the archetypal type I, was identified. Microsatellite analysis showed that the strain was a variant of type I with two atypical alleles. This was the first time that a T. gondii clonal type I genotype was correlated with a case of acute toxoplasmosis in a host in Brazil.

This study aims to report the incidence of Calodium hepaticum among dogs and cats, pets or stray animals, captured by the Zoonosis Control Center (CCZ) in Lages, Santa Catarina, Brazil. Fecal samples from 108 pet dogs and eight pet cats, and from 357 stray dogs and 97 stray cats, captured by CCZ, were analyzed within the period from July 2010 to November 2012. Coproparasitological exams were performed by techniques of sedimentation, centrifuge-flotation, and simple flotation. Among 465 fecal samples from dogs and 105 from cats, the overall spurious infections for C. hepaticum eggs were 1.05%. For dogs, this positivity was 0.43% and for cats it was 3.81%. The two positive dogs were stray and out of the four cats, three were stray and one was a pet. Although the occurrence of C. hepaticum eggs was low, these data reveal the existence of infected rodents, especially in public places, since, out of the six infected animals, five (83.33%) were stray. These results are discussed and analyzed with an emphasis on the risk to public health.

O presente estudo objetivou determinar a ocorrência de anticorpos anti-Neospora spp. em equinos das regiões serrana e litorânea de Santa Catarina e identificar possíveis fatores de risco para a infecção. Foram analisadas 615 amostras por meio da reação de imunofluorescência indireta (RIFI ≥1:50). Das 615 amostras, 25 (4,1%) foram positivas para Neospora spp. A recíproca dos títulos de anticorpos anti-Neospora spp. nos equinos foi assim distribuída: 1:50 (13), 1:100 (oito), 1:200 (três) e 1:400 (um). Das 311 amostras da Serra Catarinense, oito foram positivas (2,6%). Das amostras da Região Litorânea (304), 17 apresentaram anticorpos anti-Neospora spp., indicando uma prevalência de 5,6%. Não foi observada diferença estatística significante (P=0,06704) entre as soroprevalências para Neospora spp. em equinos das diferentes regiões, porém os animais do litoral de Santa Catarina apresentaram prevalência 2,2 vezes maior que aqueles da região serrana. O contato dos equinos com cães e/ou bovinos (P=0,007596) foi identificado como fator de risco para infecção por Neospora spp.

The aims of this study were to determine occurrences of Neospora spp. IgG antibodies in horses from two geographical regions of the state of Santa Catarina, southern Brazil, and identify risk factors for infection. Analyses were performed on 615 samples using the immunofluorescent antibody test (IFAT ≥ 1:50). Out of the 615 samples, 25 (4.1%) were positive for Neospora spp. The titers for Neospora spp. were distributed as follows: 1:50 (13), 1:100 (eight), 1:200 (three) and 1:400 (one). Out of the 311 samples taken in the mountain region, eight were positive (2.6%). Among the samples from the coastal region (304), 17 had Neospora spp. antibodies, thus indicating occurrence of 5.6%. Although no statistically significant difference was observed (P = 0.06704), the prevalence among animals of the coast was 2.2 times higher than that of the mountain region. Contact with dogs and/or cattle (P = 0.007596) were identified as risk factor for Neospora spp. infection.

A toxoplasmose ovina é uma doença parasitária de elevada importância em medicina veterinária e em saúde pública, acarretando prejuízos na produção animal, gerados pelas perdas reprodutivas e econômicas, além de sua implicação na saúde humana, já que o consumo de carne infectada facilita a transmissão zoonótica. Para determinar a prevalência e identificar fatores de risco para a infecção por T. gondii em ovinos de Lages, Santa Catarina, amostras de sangue (n = 360) foram coletadas em 13 propriedades. Cada criador respondeu a um questionário para permitir a identificação dos fatores de risco da infecção. A pesquisa de anticorpos foi realizada por meio da Reação de Imunofluorescência Indireta (RIFI > 64) e do Ensaio Imunoenzimático Indireto (ELISA). Em 100% das propriedades foram encontrados animais positivos. Pela RIFI, 205 (56,94%) ovinos apresentaram anticorpos contra T. gondii e pelo ELISA, 153 (42,50%). Considerando-se as técnicas sorológicas e a análise estatística, foram fatores de risco pelo ELISA: a idade, a fonte de água e a categoria animal; e pela RIFI, o tipo racial. Foi constatada sensibilidade de 61%, especificidade de 82% e concordância Kappa de 0,41 entre o ELISA e a RIFI (1:64), considerada moderada, permitindo indicar o ELISA como técnica adequada para o diagnóstico de T. gondii na espécie ovina.

Toxoplasmosis in sheep is a disease of great importance in veterinary medicine, which causes economic losses in livestock and has a great impact on human health, since consumption of infected meat facilitates transmission of zoonotic infections. Blood samples from sheep (n = 360) were collected from 13 farm properties in the municipality of Lages, Santa Catarina, to estimate the prevalence of toxoplasmosis and identify risk factors associated with Toxoplasma gondii infection. T. gondii, antibodies were investigated by means of the indirect immunofluorescence assay (IFA) and enzyme-linked immunosorbent assay (ELISA). Animals infected with T. gondii were found on 100% of the farms. IFA detected 56.9% (205/360) and ELISA 42.5% of the infected sheep. Breed was the only risk factor associated with the presence of T. gondii antibodies. ELISA showed sensitivity of 61%, specificity of 82% and kappa of 0.41, which was considered moderate. This allows use of ELISA as an alternative technique for diagnosing T. gondii in sheep.

During 2006-2008, a total of 260 adult ticks were collected from domestic and wild animals in different regions of the state of Santa Catarina (SC), Brazil, including areas where human cases of Brazilian spotted fever have been reported. Collected ticks belonging to nine species (Amblyomma aureolatum, Amblyomma cajennense, Amblyomma dubitatum, Amblyomma longirostre, Amblyomma ovale, Amblyomma tigrinum, Dermacentor nitens, Rhipicephalus microplus and Rhipicephalus sanguineus) were tested by polymerase chain reaction (PCR) for rickettsial infection. Overall, eight (3.1%) ticks were found to be infected with Rickettsia species. After sequencing the PCR products, we determined that the sequences generated from three A. aureolatum, one A. ovale and one R. sanguineus from the municipality of Blumenau, one A. ovale from the municipality of Águas Mornas and one A. ovale from the municipality of Urussanga were identical to the corresponding partial rickettsial ompA gene sequence of Rickettsia parkeri strain Atlantic rainforest. The sequence generated from one A. longirostre from Blumenau was 100% identical to the corresponding partial rickettsial ompA gene sequence of Rickettsia amblyommii strain AL. Because R. parkeri strain Atlantic rainforest was recently shown to have caused two cases of human spotted fever in other states of Brazil, the role of this rickettsial agent as a possible etiological agent of spotted fever in SC is discussed.

INTRODUÇÃO: O aumento no número de casos de leishmaniose tegumentar americana no ano de 2005 foi em decorrência do surto ocorrido nos Municípios de Itapema e Balneário Camboriú. O presente trabalho teve como objetivos determinar a possível presença de infecção por Leishmania sp em cães domésticos residentes em foco endêmico para LTA, no Município de Balneário Camboriú e verificar a existência de correlação entre a resposta imune humoral e celular, presença de lesões sugestivas e positividade no exame parasitológico direto. MÉTODOS: Foram cadastrados 275 animais, os quais foram examinados segundo seus aspectos clínicos, desenvolvimento de hipersensibilidade tardia ao antígeno Immunoleish® e respostas sorológicas à reação de imunofluorescência indireta e ao ensaio imunoenzimático. RESULTADOS: Sete animais apresentaram lesões suspeitas, porém não foram encontradas Leishmanias pelo método parasitológico direto. O resultado da sorologia foi de 5,8% de positividade com a RIFI, 6,2% com o ELISA e 1,8% com a prova intradérmica. Somando os cães positivos pelos exames sorológicos e/ou pela prova intradérmica obteve-se um total de 24 positivos, imunoprevalência de 8,7% para LTA. CONCLUSÕES: Há necessidade de mais estudos para avaliar a participação dos cães na cadeia epidemiológica da LTA no Município de Balneário Camboriú.

INTRODUCTION: The increase in American tegumentary leishmaniasis (ATL) cases in 2005 was due to an outbreak that occurred in the towns of Itapema and Balneário Camboriú. This study aimed to determine the possible presence of infection by Leishmania sp in domestic dogs living in an endemic focus of ATL in the town of Balneário Camboriú and verify the existence of a correlation between the humoral and cellular immune responses, the presence of suggestive lesions and positivity in direct parasitological exams. METHODS: A total of 275 dogs were evaluated, examined according to clinic aspects, the development of delayed hypersensitivity to Immunoleish® antigens and serological responses to the indirect immunofluorescence reaction and immunoenzymatic assay. RESULTS: Seven dogs presented suspect lesions, but Leishmania was not detected using the direct parasitological method. The serological result was 5.8% positivity using the IIFR technique, 6.2% by ELISA and 1.8% using the intradermal test. The total number of dogs positive for ATL using the serological exams and/or intradermal test was 24, indicating prevalence of 8.7% for ATL. CONCLUSIONS: Further studies are required to evaluate the participation of dogs in the epidemiological chain of ATL in Balneário Camboriú.

Amostras de sangue foram obtidas de 300 gatos domiciliados para determinar a prevalência de anticorpos contra Toxoplasma gondii e possíveis fatores de risco. A detecção de anticorpos da classe IgG foi realizada por meio da Reação de Imunofluorescência Indireta (RIFI). Reações positivas com títulos >1:64 foram encontradas em 43 (14,33%) gatos. Os títulos encontrados variaram de 1:64 a 1:4096, sendo que 19 animais (44,19%) tiveram titulação de 1:64, 18 animais (41,86%) com 1:256, cinco animais (11,63%) com 1:1024 e um animal (2,32%) teve titulação de 1:4096. Foi verificada correlação entre soropositividade dos felinos ao T. gondii e idade (p = 0,03758) e com o acesso à rua e/ou zona rural (p = 0,04185). Os resultados indicam que T. gondii está disseminado entre os gatos domiciliados de Lages, Estado de Santa Catarina, Brasil, com uma prevalência de 14,33%.

Sera were collected from 300 domiciled cats from the municipality of Lages, Southern Brazil, to determine the prevalence of Toxoplasma gondii antibodies and risk factors associated. Tests for T. gondii antibodies were performed using indirect immunofluorescent antibody test (IFAT). Positive reactions with titers >1:64 were found in 43 (14.33%) cats. A significant number of seropositive cats were >6 month old (p = 0.03758) and had access to the streets or/and rural areas (p = 0.04185). The results indicate that T. gondii is widespread in cats in Lages with a prevalence of 14.33%.

Os objetivos da realização deste trabalho foram determinar a prevalência de ectoparasitos e parasitos gastrointestinais (GI) em gatos domiciliados, nas regiões central e periférica, da cidade de Lages, SC e também caracterizar as famílias proprietárias de gatos quanto as suas condições sócioeconômicas e culturais. No período de dezembro de 2005 a dezembro de 2006, foram distribuídos 600 questionários a proprietários de animais cidade de Lages, SC e coletados ectoparasitos e amostras de fezes. A prevalência para ectoparasitos foi de 13,8% (28/203) e para parasitos GI, 37,8% (42/111). A prevalência de parasitos em gatos domiciliados da região periférica foi maior que os da região central. Os ectoparasitos observados foram Ctenocephalides felis felis, C. canis e Ctenocephalides híbrido (C. felis felis × C. canis). Os ovos de helmintos intestinais identificados nas fezes de gatos foram Ancylostoma spp., Toxocara sp., Trichuris sp., família Taeniidae e Oncicola sp. Na região periférica, o maior número de animais positivos pertencia a proprietários de baixa renda salarial e pouca escolaridade. As populações residentes nos bairros da região periférica encontram-se mais expostas aos agentes com potencial zoonótico.

The purpose of this research was to determinate the prevalence of ectoparasites and gastrointestinal (GI) parasites in domiciliated cats, from central and peripheral region, of Lages city, Santa Catarina State and also characterize the economic, social and cultural status of families' pet owners. From the period of December 2005 to December 2006, 600 questionnaires were distributed to pet owners of Lages city and ectoparasites and faeces were collected. The prevalence of ectoparasites was 13.8% (28/203) and for GI parasites was 37.8% (42/111). The prevalence of parasites in domiciliated cats from the peripheral region were bigger than in the central region. The ectoparasites observed were Ctenocephalides felis felis, C. canis and Ctenocephalides hybrid (C. felis felis x C. canis). The intestinal helminths eggs observed in cats faeces were Ancylostoma spp., Toxocara sp., Trichuris sp., Taeniidae family and Oncicola sp. In the peripheral region, the largest number of positive animals belonged to owners of low-income wage and low education The peripheral region population are over exposed to potencial zoonotic agents.
