ABSTRACT Plant species occurring in the Caatinga biome, when subjected to different water fluctuations, may have their morphophysiological and metabolic performance altered. Depending on the level of stress, the production of quality seedlings is affected. Therefore, the objective of this study was to evaluate the responses to different water availability levels in the morphophysiological and biochemical development of Croton blanchetianus Baill. The experiment was carried out in a greenhouse, evaluating four water regimes: 25%, 50%, 75% and 100% of pot capacity, and conducted in a randomized block design, with a total 5 blocks. The variables analyzed were: branch height and branch diameter; leaf area, number of leaves, root volume; root dry mass, shoot dry mass, total dry mass and root/shoot ratio; relative water content, proline, total soluble proteins, malondialdehyde, hydrogen peroxide, and photo synthetic pigments (chlorophyll ɑ, chlorophyll b, and carotenoids). The species Croton blanchetianus Baill. shows drought tolerance mechanisms, demonstrated by the reductions of growth, leaf area, dry mass of seedlings and photo synthetic pigments. It adjusts osmotically with increments in the production of proline, which acts in osmoprotection when there is water deficit, and in the production of proteins under the lower and higher water regime. RESUMO Espécies vegetais de ocorrência no bioma Caatinga quando submetidas a diferentes flutuações hídricas podem ter seu desempenho morfofisiológico e metabólico alterado. A depender do nível de estresse, a produção de mudas de qualidade é afetada. Logo, objetivou-se avaliar as respostas às diferentes disponibilidades hídricas no desenvolvimento morfofisiológico e bioquímico de Croton blanchetianus Baill. O experimento foi desenvolvido em casa de vegetação e foram avaliados quatro regimes hídricos: 25%, 50%, 75% e 100% da capacidade de vaso e conduzido em delineamento experimental em blocos ao acaso, totalizando 5 blocos. As variáveis analisadas foram: altura e diâmetro dos ramos; área foliar, número de folhas, volume radicular; massa seca da raiz, parte aérea, total e relação raiz/parte aérea; teor relativo de água, prolina, proteínas solúveis totais, malondialdeído, peróxido de hidrogênio e pigmentos fotossintéticos (clorofila ɑ, clorofila b e carotenóides). RESUMO Efluentes salinos da piscicultura em atividades agrícolas vem crescendo, sobretudo nas regiões semiáridas. Neste sentido, objetivou-se avaliar o efluente salino de piscicultura na irrigação de Croton blanchetianus verificando as respostas fisiológicas e bioquímicas. Após o estabelecimento das plantas oriundas de estacas, iniciou-se a irrigação com o efluente, diferenciando-se os tratamentos por quatro níveis de salinidade (controle, 1,5, 2,5 e 3,5 dS m-1). O tratamento controle foi representado pela água de abastecimento (0,56 dS m-1). As variáveis avaliadas foram: crescimento, teor relativo de água, biomassa seca, níveis de proteínas, prolina, peróxido de hidrogênio, malondialdeído e os pigmentos fotossintéticos. Os ajustes fisiológicos e bioquímicos garantiram a manutenção do teor relativo de água e ajuste osmótico em meio à condição salina, sem incremento da peroxidação lipídica. Os níveis de salinidade não afetaram C. blanchetianus, indicando que não houve estresse salino, pois, os efluentes da piscicultura são ricos em matéria orgânica, proveniente da alimentação e excrementos, beneficiado o desenvolvimento da espécie. O reuso do efluente salino de piscicultura na irrigação de C. blanchetianus pode ser uma alternativa ambiental viável, evitando-se o descarte diretamente no ambiente.

ABSTRACT The use of saline effluents from fish farming in agricultural activities has been increasing, especially in semiarid regions. This study was to evaluate the use of saline effluents from fish farming for the irrigation of Croton blanchetianus and to analyze the physiological and biochemical responses. After the cuttings were planted, irrigation with the effluent began. Treatments included four salinity levels (control, 1.5, 2.5, 3.5 dS m-1). The control treatment was the supply water (0.56 dS m-1). The evaluated variables were growth, relative water content, dry biomass, and levels of proteins, proline, hydrogen peroxide, malondialdehyde, and photosynthetic pigments. Physiological and biochemical adjustments ensured the maintenance of relative water content and osmotic adjustment under saline conditions without increasing lipid peroxidation. The salinity levels did not affect C. blanchetianus, indicating the lack of saline stress. The fish farm effluents are rich in organic matter, from food, and excrement, which was beneficial for the development of C. blanchetianus. The reuse of saline effluents from fish farming for the irrigation of C. blanchetianus can be a viable environmental alternative, avoiding direct disposal into the environment.

RESUMO Objetivou-se com este trabalho identificar a resposta antioxidante de variedades de cana-de-açúcar sob condições adversas. Mudas de seis genótipos, procedentes da micropropagação, foram submetidas à avaliação da ação isolada e combinada do estresse hídrico e salino. A pesquisa foi realizada em maio de 2012, em casa de vegetação instalada na Universidade Federal Rural de Pernambuco (UFRPE). O delineamento experimental foi o inteiramente casualizado, com tratamentos dispostos em esquema fatorial 6 x 4 (seis variedades e quatro condições de cultivo) com quatro repetições. Avaliou-se a peroxidação lipídica, o teor de peróxido de hidrogênio (H2O2), o teor relativo de água (TRA) e ação do sistema de defesa antioxidativo mediante a atividade das enzimas superóxido dismutase (SOD), ascorbato peroxidase (APX) e catalase (CAT). As variedades de cana-de-açúcar exibiram maior peroxidação lipídica e/ou maior concentração de peróxido de hidrogênio quando submetidas ao estresse combinado. As enzimas apresentaram respostas diferenciadas de acordo com a variedade de cana-de-açúcar e a intensidade de estresse nos tratamentos isolados, no entanto, podem ter sido inativadas perante o estresse combinado, o que indica que a resposta do estresse combinado é diferente da soma das respostas dos fatores abióticos aplicados isoladamente. Os elevados teores de malondialdeído (MDA) associados ao baixo TRA podem ser indicadores efetivos de sensibilidade a estresse múltiplo em variedades de cana-de-açúcar. As variedades RB99395 e RB867515 apresentaram-se mais eficientes quanto a resposta ao estresse ambiental e a manutenção de conteúdo hídrico celular nos estresses isolados.

ABSTRACT This study aimed to identify the antioxidant responses of sugarcane (Saccharum spp.) varieties subjected to water and saline stress. Sugarcane seedlings of six different varieties obtained through micropropagation were subjected to either water or saline stress, or a combination of water + saline stress. The study was carried out in May 2012, in a greenhouse at the Universidade Federal Rural de Pernambuco (UFRPE). The experimental design was randomized, with treatments arranged in a 6 × 4 factorial scheme (six varieties and four treatments), and four replicates. Lipid peroxidation, hydrogen peroxide (H2O2) concentrations, and relative water content (RWC) were evaluated. Furthermore, we evaluated the plants’ antioxidative defense systems by measuring the activities of superoxide dismutase (SOD), ascorbate peroxidase (APX), and catalase (CAT). The sugarcane varieties had higher lipid peroxidation and/or higher H2O2 concentrations when subjected to the combined water + saline stress. The antioxidant enzymes responded to the water and saline stress treatments differently depending on the sugarcane variety. However, under combined saline + water stress conditions, the enzymes may have become inactivated, which indicates that the response to the combined water + saline stress was different from the sum of the responses to only water stress or only saline stress. High concentrations of malondialdehyde (MDA) associated with low RWC may be an effective indicator of multiple stress sensitivity in sugarcane varieties. The RB99395 and RB867515 sugarcane varieties responded more efficiently to environmental stress, and maintained their cell water content when subjected to either water or saline stress.

RESUMO: Este estudo teve como objetivo avaliar mecanismos fisiológicos e bioquímicos de adaptação de Hyptis suaveolens submetidas ao estresse osmótico decorrente da salinidade inoculadas com espécies de fungos micorrízicos. Sementes de H. suaveolens foram colocadas para germinar em vasos de polietileno contendo substrato associado, ou não, a FMA. Após o estabelecimento das plantas, foram iniciados os tratamentos com diferentes concentrações de sais (0,0; 35; 70 e 105 mM) e fungos (controle e dois tipos de FMA), perfazendo 12 tratamentos com três repetições. O delineamento experimental foi em blocos casualizados em esquema fatorial 4 x 3, perfazendo 12 tratamentos com três repetições cada. Observou-se, através de variáveis fisiológicas e bioquímicas, que a salinidade afetou todas as variáveis analisadas e o estresse promoveu redução no conteúdo de matéria seca. Verificou-se que plantas associadas com FMA apresentaram ganho de matéria seca em relação as não associadas e incremento de respostas bioquímicas e fisiológicas das plantas colonizadas no tratamento 35mM de NaCl. No entanto, o estresse salino afeta negativamente o desenvolvimento de H. suaveolens não sendo observado, portanto, atenuação dos fungos.

ABSTRACT: The objective of this study was to evaluate the physiological and biochemical mechanisms of adaptation of Hyptis suaveolens under osmotic stress due to salinity inoculations with a mycorrhizal fungal species. H. suaveolens seeds were germinated in polyethylene pots containing a substrate associated with or without arbuscular mycorrhizal fungi (AMF). After plant formation, treatments were treated with different salt concentrations (0.0, 35, 70, and 105 mM) and fungi (control and two types of AMF), totaling 12 treatments with three replicates. The experimental design used randomized blocks in a 4 x 3 factorial scheme, totaling 12 treatments with three replicates each. Salinity affected all measured physiological and biochemical variables, and the stress reduced dry matter content. Plants associated with AMF had increased dry matter compared to non-associated plants, and there were increased biochemical and physiological responses of AMF-colonized plants in the 35 mM NaCl treatment. However, saline stress negatively affected the development of H. suaveolens; and therefore, no attenuation of fungi was observed.

RESUMO Varronia leucocephala é uma planta utilizada pela população para fins terapêuticos. Um dos maiores problemas para produção de mudas dessa espécie é o baixo percentual de enraizamento. Objetivou-se estabelecer um protocolo de enraizamento de estacas de V. leucocephala utilizando fitorreguladores e microelementos associados com fungos micorrízicos arbusculares. Sendo a concentração de 1500 mg L-1 do ácido indolbutírico (AIB) a que proporcionou maior percentual de enraizamento, propôs-se associar os microelementos zinco (Zn) e boro (B) com maiores concentrações de AIB. Na presença do Zn, observou-se incremento na percentagem de enraizamento; no entanto, não foi o ideal para aumentar a percentagem de propagação. Testou-se a associação de fungos micorrízicos arbusculares junto à concentração de 1500 mg L-1 de AIB para avaliar o percentual de enraizamento e brotação de parte aérea; biomassa seca de raiz e parte aérea; número de folhas; altura; colonização e dependência micorrízica; densidade de esporos e nutrientes da parte aérea e da raiz. A utilização de zinco juntamente a doses mais elevadas de AIB (1500 mg L-1) e associada com fungo micorrízico arbusculares (Gigaspora albida) tornaram-se eficientes para a propagação vegetativa de V. leucocephala.

ABSTRACT The plant Varronia leucocephala is widely used in Brazil for its therapeutic properties. However, a major problem for the seedlings is the low percentage of root formation. The objective of this study was to establish a rooting protocol for V. leucocephala cuttings, using phytoregulators and microelements associated with arbuscular mycorrhizal fungi. The auxin indole-3-butyric acid (IBA) concentration of 1500 mg L-1 showed the best rooting percentage, and it is proposed associating the microelements zinc and boron with the highest IBA doses. Although an increase in the rooting percentage was observed in the presence of zinc, it was not the most suitable for improving the percentage of propagation. Consequently, association of arbuscular mycorrhizal fungi with 1500 mg L-1 IBA plus zinc was selected to evaluate the rooting percentage and sprouting of the aerial part, dry biomass of roots and aerial part, number of leaves, height, mycorrhizal colonization and dependency, spore density, and nutrients of branches and roots. These results show that using zinc with the highest doses of IBA (1500 mg L-1) in plants inoculated with the arbuscular mycorrhizal fungus (Gigaspora albida) was the most effective at promoting the vegetative propagation of V. leucocephala.

RESUMO: O presente estudo buscou induzir a formação de embriões somáticos em protocormos do híbrido Phalaenopsis Classic Spotted Pink em dois estádios de maturação fisiológica aos 80 e 120 dias após a inoculação das sementes (DAIS). Os protocormos foram inoculados em meio ½ MS suplementado com 0,1 mg L-1 de ANA e 3 mg L-1 de TDZ. Aos 15 dias de cultivo os protocormos, inoculados aos 120 dias após a semeadura, apresentavam estádio mais desenvolvido, apresentando formação de estruturas pró-embriogênicas. Aos 30 dias de cultivo em meio de indução de ES observou-se a formação de embriões somáticos na fase globular e torpedo. Os protocormos inoculados com 80 DAIS não evoluíram para a formação de embriões somáticos, ocorrendo a oxidação destes aos 20 dias após o cultivo em meio de indução de ES. A formação dos embriões somáticos ocorreu diretamente no explante, caracterizando uma embriogênese somática direta. Quando os embriões somáticos foram transferidos para o meio nutritivo sem regulador de crescimento, houve a conversão em plantas. Diante disso, conclui-se que a indução de embriões somáticos a partir de protocormos com 120 dias após a semeadura, foi positiva, em que os embriões obtidos apresentaram competência em converter-se em plantas, apresentando os órgãos vegetativos com características morfológicas satisfatórias.

ABSTRACT: The aim of the present study was to induce the formation of somatic embryos in protocorms from Phalaenopsis Classic Spotted Pink hybrids at two physiological maturation stages, namely: 80 and 120 days after seed inoculation (DASI). Protocorms were inoculated in ½ MS medium supplemented with 0.1 mg L-1 ANA and 3 mg L-1 TDZ. Protocorms were inoculated 120 days after sowing were more developed at the 15th cultivation day due to the formation of pro-embryogenic structures. It was possible seeing the formation of globular- and torpedo-stage somatic embryos at the 30th day of cultivation in somatic embryogenesis (SE) induction medium. The protocorms inoculated at the 80th DASI did not formed somatic embryos; they oxidized 20 days after cultivation in SE-induction medium. The formation of somatic embryos happened directly on the explant, thus characterizing a direct somatic embryogenesis. The embryos converted into plants when the somatic embryos were transferred to the nutrient medium containing no growth regulator. Therefore, it was concluded that the somatic embryos induction from protocorms 120 days after sowing was positive, since the embryos were able to become plants and presented vegetative organs with morphological traits similar to those of the matrix plant.

RESUMO O objetivo deste trabalho foi comparar o desenvolvimento fisiológico em casa de vegetação de plantas de Heliconia bihai cv. Lobster Claw Two provenientes do cultivo in vitro de embriões zigóticos e de propagação convencional. Para tanto, helicônias obtidas a partir de rizomas e de multiplicação in vitro foram avaliadas a cada 30 dias durante dez meses. O delineamento experimental utilizado foi inteiramente casualizado, com dez repetições, sendo o arranjo fatorial 2x10, composto por 2 condições de cultivo e dez avaliações em tempos distintos. Os parâmetros biométricos analisados foram: altura da planta, número de folhas, de perfilhos, área foliar e posteriormente intensidade da cor das brácteas. As plantas obtidas do cultivo in vitro apresentaram diferença significativa no desenvolvimento dos parâmetros fisiológicos avaliados em comparação com as plantas provenientes de rizoma, como também, apresentaram precocidade na floração. Apesar das plantas provenientes do cultivo in vitro procederem de embrião zigótico, não foram observadas alterações morfológicas na parte vegetativa e reprodutiva (inflorescências) das plantas e nem na colorimetria, evidenciando que a técnica de cultivo in vitro de embriões zigóticos, pode ser utilizada para produção de mudas em larga escala, possibilitando incremento no setor da floricultura na região e no país.

ABSTRACT The aim of this study is to compare physiological development of Heliconia bihai cv. Lobster Claw Two plants derived from in vitro culture of zygotic embryos and conventional propagation. Heliconias obtained from rhizomes and from in vitro multiplication were evaluated every 30 days during ten months under greenhouse conditions. The experimental design was completely randomized, with ten repetitions, and the 2x10 factorial arrangement consisted of two plant multiplication methods and ten evaluations performed at different times. The analyzed biometric parameters were plant height, number of leaves, number of tillers, leaf area, and color intensity in the bracts. Plants derived from in vitro culture showed significant differences in the development of the evaluated physiological parameters in comparison to plants derived from rhizomes, and they also showed early flowering. Although the in vitro cultured plants were derived from zygotic embryos, no morphological changes were found in the vegetative and reproductive parts (inflorescence) of the plants or in the colorimetry. It shows that the in vitro cultures of zygotic embryos may be used as a technique to produce seedlings on a large-scale, thus allowing the floriculture sector to grow in the region and all over the country.

RESUMO: A exposição de plantas a condições de estresse múltiplo pode causar uma superprodução de espécies reativas de oxigênio promovendo desequilíbrio no sistema redox. O objetivo deste trabalho foi estudar a resposta antioxidante de variedades de cana-de-açúcar sob condições adversas visando a compreensão de mecanismos de defesa de genótipos tolerantes e sensíveis. Mudas micropropagadas de cana-de-açúcar de seis genótipos, RB966928, RB98710, RB855453, RB99395, RB867515 e RB855156 foram submetidas à avaliação da ação isolada do estresse por alta temperatura e da sua combinação com o estresse hídrico e salino. Avaliou-se a atividade das enzimas superóxido dismutase (SOD), catalase (CAT) e ascorbato peroxidase (APX), bem como a peroxidação lipídica (teor de MDA) e teor de peróxido de hidrogênio (H2O2), indicadores de estresse. A variedade RB867515 destacou-se por apresentar sincronia entre a atividade enzimática na regulação e desintoxicação das ROS, produzidas na célula vegetal durante situações de estresse ambiental, garantindo a manutenção do teor relativo de água sem incremento da peroxidação lipídica e, consequentemente, a manutenção dos processos fisiológicos e o crescimento da planta. Os resultados indicam que a peroxidação lipídica associada ao baixo teor relativo de água podem ser bons marcadores bioquímicos de sensibilidade da cana-de-açúcar à estresses múltiplos.

ABSTRACT: The exposure of plants to multiple stress conditions can cause over production of reactive oxygen species promoting an imbalance in the redox system. The aim of this study was to examine the antioxidant response from varieties of sugarcane under adverse conditions, to understand the defense mechanisms of tolerant and sensitive genotypes. Micropropagated sugarcane of six genotypes, RB966928, RB98710, RB855453, RB99395, RB867515, and RB855156, were subjected to isolated action of high-temperature stress and a combination of water and saline stress, and effects were evaluated. Activities of enzymes such as superoxide dismutase (SOD), catalase (CAT), and ascorbate peroxidase (APX), as well as stress indicators such as lipid peroxidation (MDA content) and hydrogen peroxide (H2O2) content were evaluated. The RB867515 variety showed synchrony between the enzymatic activity and ROS regulation and detoxification, in the plant cell under environmental stress situations, thereby ensuring the maintenance of the relative water content without increment of lipid peroxidation; and consequently, the maintenance of physiological processes and plant growth. These results indicated that lipid peroxidation associated with relative water content can be good biochemical markers of sensitivity to multiple stresses in sugarcane.

RESUMO Este trabalho foi realizado com o objetivo de avaliar o efeito do estresse salino sobre a produção e o metabolismo de defesa antioxidativa de duas cultivares de pimentão (Capsicum annuum L.), cultivadas em sistema hidropônico. O delineamento experimental foi em blocos casualizados, com os tratamentos arranjados em esquema fatorial 5 x 2, com cinco repetições. Os tratamentos consistiram da combinação de cinco níveis de salinidade da solução nutritiva (2,0; 3,5; 4,5; 5,5 e 6,5 dS m-1) com duas cultivares de pimentão (Bruno e Rúbia). A biomassa e a produção foram reduzidas com o incremento da salinidade. Os teores de clorofila a, b e total, diminuíram com o aumento da salinidade. Houve aumento no teor de prolina e na atividade das enzimas superóxido dismutase (SOD), catalase (CAT) e ascorbato peroxidase (APX), evidenciando a ativação do mecanismo de defesa antioxidativo. A maior produção de frutos na cv. Rúbia, bem como a maior atividade da APX e os menores teores de clorofilas refletem a melhor aclimatação desta cultivar ao estresse salino. A produtividade estimada da cv. Rúbia, 39,8 t ha-1, quando cultivada com uma CEs de até 3,5 dS m-1, indica a viabilidade de seu cultivo comercial em condições hidropônicas.

ABSTRACT This study aimed to evaluate the effect of salt stress on production and antioxidative defense metabolism of two bell pepper cultivars (Capsicum annuum L.) grown hydroponically. The experimental design was in randomized blocks, with treatments arranged in a 5 x 2 factorial scheme with five replicates. The treatments consisted of five salinity levels of the nutrient solution (2.0, 3.5, 4.5, 5.5 and 6.5 dS m-1) with two bell pepper cultivars, ‘Bruno’ and ‘Rúbia’. Biomass and yield were reduced with increasing salinity. The contents of chlorophyll a, b and total decreased with increasing salinity. There was an increase in proline content and activity of superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX), showing the activation of the antioxidant defense mechanism. The higher yield in the cv. ‘Rúbia’, as well as the greater activity of the APX and the lower levels of chlorophyll content, reflect the best acclimatization of this cultivar to saline stress. The estimated yield of the cv. ‘Rúbia’, 39.9 t ha-1 when grown with ECs of up to 3.5 dS m-1, indicates the feasibility of its commercial cultivation in hydroponic conditions.

El presente trabajo tuvo como objetivos inducir la formación de embriones somáticos in vitro en el híbrido Phalaenopsis Classic Spotted Pink, utilizando diferentes medios nutritivos, y evaluar la morfología interna de estos embriones mediante análisis histológico e histoquímico. Hojas jóvenes de plantas cultivadas in vitro se utilizaron como explantes para la inducción de embriones somáticos en diferentes medios nutritivos: New Dogashima Medium, contenido de ANA (0.537 mM) y BAP (4.440 μM) además de phytagel y con pH 5.8 (NDM) y el Murashige Skoog con la mitad de la concentración de sales, además de ANA (0.537 μM) y TDZ (13.621 μM), gelificado gelrite y pH 5.2 (½ MS). Se obtuvieron embriones somáticos primarios a los 90 días de cultivo en el medio ½ MS y a estos se les transfirió al mismo medio (½ MS) para la obtención de embriones secundarios. Los embriones somáticos primarios y secundarios fueron subcultivados para MS con la mitad de la concentración de sales, sin reguladores de crecimiento y sometidos a fotoperiodo de 16 horas, lo que estimuló la producción de clorofila tanto en los embriones primarios como en los secundarios, promoviendo el desarrollo de los protocormos y más tarde en las plantas. Los análisis histológicos demostraron que los embriones somáticos fueron formados directamente en las capas epidérmicas de los explantes, sin pasar por la fase de callo, vía embriogénesis somática directa. Los métodos histoquímicos hicieron posible destacar la deposición de almidón y lípidos en las células embriogénicas como resultado de mecanismos fisiológicos, que permiten el desarrollo de los embriones primarios y secundarios en las plantas. Por lo tanto, el medio ½ MS contenido de ANA (0.537 μM) y TDZ (13.621 μM), con gelrite y pH 5.2 permitió obtener embriones primarios y secundarios con capacidad para regenerar plantas con caracteres morfológicos similares a los dela planta matriz.

The present work had as objectives to induce the formation of somatic embryos in vitro on Phalaenopsis hybrid Classic Spotted Pink, using different nutrient medium and assess the internal morphology of these embryos by means of histological and histochemical analysis. Young leaves of plants grown in vitro were used as explants for induction of somatic embryos in different nutrient medium: New Dogashima Medium, containing ANA (0.537 μM) and BAP (4.440 μM) plus phytagel and with pH 5.8 (NDM) and the Murashige & Skoog with half the concentration of salts, plus NNA (0.537 μM) and TDZ (13.621 μM), jellied with gelrite and pH 5.2 (0.5 MS). Primary somatic embryos were obtained to 90 days of cultivation in half MS and have been transferred to the same means for obtaining of secondary embryos. The primary and secondary somatic embryos were subcultived for MS with half the concentration of salts, without fitoregulator subjected to photoperiod of 16 hours, which stimulated the production of chlorophyll in primary embryos as secondary, promoting the development of those in protocorms and later in plants. The histological analysis showed that the somatic embryos were formed directly from the epidermal layers of the explants, without going through the phase of callus, featuring direct somatic embryogenesis. The histochemical methods used made it possible to highlight the deposition of starch and lipids in cells embriogenics as a result of physiological mechanisms, enabling the development of primary and secondary embryos in plants. Therefore, the medium 0.5 MS Plus ANA (0.537 μM) and TDZ (13.621 μM), jellied with gelrite and pH 5.2 promoted to obtain primary and secondary embryos with ability to regenerate plants showing morphological similar the mother plant.

O presente trabalho teve como objetivos induzir a formação de embriões somáticos in vitro no híbrido Phalaenopsis Classic Spotted Pink, utilizando diferentes meios nutritivos e avaliar a morfologia interna desses embriões por meio de análises histológicas e histoquímicas. Folhas jovens de plantas cultivadas in vitro foram utilizadas como explantes para indução de embriões somáticos em diferentes meios nutritivos: New Dogashima Medium, contendo ANA (0,537μM) e BAP (4,440μM), acrescido de phytagel e com pH 5,8 (NDM) e o Murashige & Skoog com a metade da concentração dos sais, acrescido de ANA (0,537μM) e TDZ (13,621μM), gelificado com gelrite e o pH 5,2 (½ MS). Embriões somáticos primários foram obtidos aos 90 dias de cultivo no meio ½MS e foram transferidos para o mesmo meio para obtenção de embriões secundários. Os embriões somáticos primários e secundários foram subcultivados para meio MS com metade da concentração de sais, sem fitoregulador submetidos a fotoperíodo de 16 horas, o qual estimulou a produção de clorofila tanto nos embriões primários como secundários, promovendo o desenvolvimento desses em protocormos e posteriormente em plantas. As análises histológicas demonstraram que os embriões somáticos foram formados diretamente das camadas epidérmicas dos explantes, sem passar pela fase de calo, caracterizando embriogênese somática direta. Os métodos histoquímicos utilizados possibilitaram evidenciar a deposição de amido e lipídeos nas células embriogênicas em decorrência de mecanismos fisiológicos, permitindo o desenvolvimento dos embriões primários e secundários em plantas. Portanto, o meio ½ MS acrescido de ANA (0,537μM) e TDZ (13,621μM), gelificado com gelrite e o pH 5,2 promoveu a obtenção de embriões primários e secundários com capacidade para regenerar plantas apresentando características morfológicas semelhantes a planta matriz.

Neste trabalho foram avaliados dez genótipos diplóides de bananeira (Musa spp) quanto a tolerância à salinidade, estresse abiótico que limita a produtividade da cultura. As plantas foram cultivadas durante 21 dias, em solução acrescida ou não de 100 mol m-3 de NaCl e analisadas variáveis de crescimento que incluem área foliar, biomassa fresca e seca, alocação de biomassa e taxa de crescimento. O experimento foi conduzido em delineamento inteiramente casualizado em fatorial 10 x 2 e três repetições. Na maioria dos genótipos estudados a salinidade provocou reduções em quase todas as variáveis analisadas. O genótipo Lidi destacou-se por apresentar melhor adaptação ao estresse salino, em todas as variáveis biométricas e capacidade de manutenção, sob estresse, da biomassa seca e fresca (limbo, caule, pseudocaule + raiz), área foliar, além de taxa de crescimento absoluto, entre outros. A produção relativa da biomassa seca da parte aérea foi superior a 70%, caracterizando este genótipo como tolerante e promissor para ser integrado a programas de melhoramento. Os genótipos Ouro e Tungia, por sua vez, sofreram grande redução da taxa de crescimento absoluto e a produção relativa da biomassa seca foi inferior a 50% caracterizando este genótipo como sensivel à salinidade.

Ten diploid banana genotypes (Musa spp.) were evaluated with regard to salt tolerance, abiotic stress which limits the productivity of the crop. The plants were grown during 21 days, in a nutrient solution with and without the addition of NaCl (0 and 100 mol m-3). Growth variables including leaf area, fresh and dry biomass, biomass allocation and growth rate were analysed. The experiment was implemented in a completely randomized design in factorial (10 x 2) arrangement and three replicates per treatment. Salinity caused in most genotypes reductions in almost all variables. Genotype Lidi stood out by having better adaptation to saline stress in all measured biometric variables with the ability to maintain, under stress, the fresh and dry biomass (leaf blade, stem, pseudostem + root), leaf area, and absolute growth rate among others. The relative production of dry biomass of shoots was more than 70%, characterizing this as a tolerant genotype and promising to be integrated into breeding programs. Genotypes Ouro and Tungia, in turn, showed reduction of absolute growth rate and relative dry biomass production (less than 50%) were characterized as sensitive to salinity.
