Resumo A cultura da cana-de-açúcar Saccharum spp. (Poales: Poaceae) produz diferentes derivados para o mundo: açúcar, etanol e bioenergia. Apesar da aplicação de pesticidas, os insetos-praga ainda causam prejuízos econômicos, dentre eles a cochonilha rosada da cana-de-açúcar Saccharicoccus sacchari (Cockerell, 1895) (Hemiptera: Pseudococcidae) causando danos diretos e indiretos à planta. Este estudo avaliou a virulência de três espécies de nematoides entomopatogênicos (NEPs) e suas bactérias simbiontes contra a cochonilha rosada da cana-de-açúcar, em condições de laboratório. Quatorze tratamentos representados pelo controle (água destilada), Heterorhabditis bacteriophora Poinar, 1976 (HB EN01) (Rhabditida: Heterorhabditidae), Steinernema rarum (Doucet, 1986) (PAM25) e Steinernema carpocapsae Weiser, 1955 (All) (Rhabditida: Steinermatidae) nas concentrações de 25, 50, 75 e 100 juvenis infectantes (JIs)/inseto, e o produto químico padrão, tiametoxam, foram testados. Em um segundo experimento, a bactéria Photorhabdus luminescens (Thomas e Poinar, 1979), Xenorhabdus szentirmaii Lengyel, 2005 e Xenorhabdus nematophila (Poinar e Thomas, 1965) (Enterobacterales: Morganellaceae) em 3,0 x 109 células/ml foram avaliadas para cada tratamento. Dez repetições foram estabelecidas, cada uma contendo dez fêmeas/cochonilhas dentro de uma placa de Petri de 10 cm, totalizando 100 indivíduos/tratamento. Todos os tratamentos foram mantidos em condições estáveis (25±1 ºC, U 70±10%, no escuro). Todas as espécies de nematoides infectaram S. sacchari. Steinerma rarum (PAM25) proporcionou a maior mortalidade contra a cochonilha rosada da cana-de-açúcar (79,25%), seguida por H. bacteriophora (HB EN01) (58,25%) e S. carpocapsae (All) (42,50%) (P<0,001). As taxas de mortalidade causada por X. szentirmaii, P. luminescens e X. nematophila foram de 40, 45 e 20%, respectivamente. Steinerma rarum (PAM25) tem condições de ser um agente potencial a ser incorporado ao manejo integrado de pragas da cana-de-açúcar.

Abstract Sugarcane crops Saccharum spp. (Poales: Poaceae) produces different derivatives to the world: sugar, ethanol and bioenergy. Despite the application of pesticides, insect pests still cause economic losses, among these the pink sugarcane mealybug Saccharicoccus sacchari (Cockerell, 1895) (Hemiptera: Pseudococcidae) causing direct and indirect damage to the plant. This study assess the virulence of three entomopathogenic nematodes (EPNs) species and their symbiont bacteria against the pink sugarcane mealybug, under laboratory conditions. Fourteen treatments represented by control (distilled water), Heterorhabditis bacteriophora Poinar, 1976 (HB EN01) (Rhabditida: Heterorhabditidae), Steinernema rarum (Doucet, 1986) (PAM25) and Steinernema carpocapsae Weiser, 1955 (All) (Rhabditida: Steinermatidae) at concentrations of 25, 50, 75 and 100 infective juveniles (IJs)/insect, and the standard chemical product, thiamethoxam, were assayed. In a second experiment, the bacteria Photorhabdus luminescens (Thomas and Poinar, 1979), Xenorhabdus szentirmaii Lengyel, 2005 and Xenorhabdus nematophila (Poinar and Thomas, 1965) (Enterobacterales: Morganellaceae) at 3.0 x 109 cells/ml were assessed for each treatment. Ten replications were stablished, each one counting ten females/mealybugs inside a 10 cm Petri dish, amounting 100 individuals/treatment. All treatments were kept under stable conditions (25±1 ºC, H 70±10%, in the dark). All nematodes species infected S. sacchari. Steinerma rarum (PAM25) provided the highest mortality against the pink sugarcane mealybug (79.25%), followed by H. bacteriophora (HB EN01) (58.25%) and S. carpocapsae (All) (42.50%) (P<0.001). The mortality rate caused by X. szentirmaii, P. luminescens and X. nematophila were 40, 45 and 20%, respectively. Steinerma rarum (PAM25) has conditions to be a potential agent to be incorporate into the integrated pest management in sugarcane.

Resumo No estado de São Paulo, principal região produtora de cana-de-açúcar do mundo, duas espécies de cochonilhas têm ocorrido frequentemente, Aclerda takahashii (Kuwana, 1932) (Hemiptera: Aclerdidae) e Saccharicoccus sacchari (Cockerell, 1895) (Hemiptera: Pseudococcidae). Para mapear a distribuição e abundância destas espécies, 17 regiões produtoras de cana-de-açúcar, distribuídas em seis mesorregiões de São Paulo, foram avaliadas em agosto de 2017 e janeiro, fevereiro, junho e julho de 2018, durante a fase de maturação. O estudo da sazonalidade destas espécies, pela presença ou ausência das cochonilhas durante o ciclo fenológico da planta, foi conduzido entre agosto de 2017 e julho de 2018 em duas regiões produtoras de cana-de-açúcar no município de Jaboticabal, São Paulo, Brasil. Saccharicoccus sacchari foi encontrada em todas as localidades analisadas, e A. takahashii em 12. Ambas cochonilhas mostraram diferença significativa de infestação na região dos nós dos colmos durante a fase de maturação em uma das localidades estudadas. O aclerdídeo apresentou diferença significativa pela infestação em uma localidade durante a fase de maturação da planta. O pseudococcídeo infestou um grande número de nós nas seguintes fases de desenvolvimento; perfilhamento, crescimento vegetativo e maturação em ambas áreas estudadas, mas foi em uma localidade durante a fase de maturação que apresentou a maior diferença. Embora a cochonilha rosada da cana-de-açúcar foi mais abundante do que A. takahashii em ambos estudos, não houve padrões de relações entre o número de indivíduos com a localização geográfica e temperatura.

Abstract In the state of São Paulo, the main sugarcane producing region of the world, two species of scale insects have frequently occurred, Aclerda takahashii (Kuwana, 1932) (Hemiptera: Aclerdidae) and Saccharicoccus sacchari (Cockerell, 1895) (Hemiptera: Pseudococcidae). To map the distribution and abundance of these species, 17 sugarcane producing fields, distributed in six mesoregions in São Paulo, were evaluated in August 2017 and, January, February, June and July 2018 during the ripening phase. The study on the seasonality of these species, by the presence or absence of the scale insects during the phenological cycle of the plant, was conducted between August 2017 and July 2018 in two sugarcane producing fields in the municipality of Jaboticabal, São Paulo, Brazil. The presence of S. sacchari was found in all the analyzed locations, and A. takahashii in twelve. Both scale insects showed significant difference of infestation in the node’s region of the stems during the ripening phase in one of the studied locations. The aclerdid presented significant difference by infestation in one site during the ripening phase of the plant. The pseudococcid infested a greater number of nodes in the following phases of development; vegetative, grand growth and ripening in both studied areas, but it was in one site during the ripening phase that presented the greatest difference. Although the pink sugarcane mealybug was more abundant than A. takahashii in both studies, there were no patterns of relationships between the numbers of individuals to geographical locations and temperature.

Abstract Water stress limits the initial growth and development of maize mass and grain, as well as the physiological process for absorbing the amount of mineral elements. The objective was to evaluate the effect of silicon on germination and growth of corn seedlings submitted to water deficit. The experiment was carried out in the laboratory and the experimental design was completely randomized (factorial 3 × 4), with three concentrations of calcium silicate (0.0; 1.0 and 2.0 mM) and 4 solutions of PEG-6000 to simulate different osmotic potentials (0, 0; -0.3; -0.6; -0.9 MPa). Germination percentage, germination speed index (GSI), mean germination time (MGT), percentage of non-germinated and abnormal germinated, length and dry matter of shoot, root and total seedlings were evaluated. Water deficiency reduced the parameters TG, GSI and MGT. The water deficit reduce the MSPA, MSR and MST with more than 80% reduction in mass from seedlings without deficiency to seedlings with deficiency. For CPA, CR and CT there was a reduction of at least 87%, 70% and 77%, respectively, among seeds without deficiency compared to seeds submitted to deficiency. The use of silicon in corn seeds did not attenuate the stress caused by water deficit simulated by PEG-6000.

Resumo O estresse hídrico limita o crescimento inicial e o desenvolvimento da massa e do grão de milho, assim como no processo fisiológico para a absorção da quantidade de elementos minerais. Objetivou-se avaliar o efeito do silício na germinação e no crescimento de plântulas de milho submetidas a deficiência hídrica. O experimento foi desenvolvido em laboratório e o delineamento experimental foi inteiramente casualizado (DIC) (fatorial 3 × 4), sendo três concentrações de Silicato de cálcio (0,0; 1,0 e 2,0 mM) e 4 soluções de PEG-6000 a simular diferentes potenciais osmóticos (0,0; -0,3; -0,6; -0,9 MPa). Foram avaliados a porcentagem de germinação, o índice de velocidade de germinação (IVG), o tempo médio de germinação (TMG), a porcentagem de não germinadas e de germinadas anormais, o comprimento e matéria seca da parte aérea, raiz e total das plântulas. A deficiência hídrica diminuiu os parâmetros TG, IVG e TMG. O déficit hídrico reduz a MSPA, MSR e MST com mais de 80% de redução da massa das plântulas sem deficiência para as plântulas com deficiência. Para CPA, CR e CT houve redução de, no mínimo, 87%, 70% e 77%, respectivamente entre as sementes sem deficiência em comparação as sementes submetidas a deficiência. A utilização de silício em sementes de milho não atenuou o estresse causado pela deficiência hídrica simulada por PEG-6000.

Abstract The water deficit in particular, reduces the productivity of vegetable crops. To minimize these harmful effects on agriculture, several agronomic and physiological practices are being studied, such as the use of bacteria and water stress attenuators, such as brassinosteroids. Considering the socioeconomic relevance of corn culture and its sensitivity when exposed to water deficit, the objective of the present study was to evaluate the action of brassinosteroids and azospirillum on nitrogen metabolism in corn plants subjected to water stress conditions. The experiment was carried out in a greenhouse, in a period of 47 days, with corn plants, using the hybrid K9606 VIP3. The design was completely randomized, in a 2x2x3 factorial scheme, with six replications. The first factor corresponds to two water regimes (presence and absence of water deficit). The second corresponds to inoculation via seed of Azospirillum brasiliense and absence of inoculation. And the third corresponds to the application of three concentrations of brassinosteroids (0, 0.3 and 0.6 μM). Were determined Nitrate; nitrate reductase; free ammonium; total soluble aminoacids; soluble proteins; proline; glycine betaine and glutamine synthetase. The lack of water in plants provided a reduction in the protein and nitrate reductase contents, in leaves and roots. For ammonium, plants with water deficit inoculated at a concentration of 0.3 μM, obtained an increase of 7.16 (70.26%) and 13.89 (77.04%) mmol NH4 + .Kg-1. DM (Dry mass) on the leaf and root respectively. The water deficit in the soil provided significant increases in the concentrations of glycine betaine, nitrate, proline and aminoacids, both in the leaves and in the roots of the corn plants. On the other hand, the contents of glutamine synthetase had a reduction in both leaves and roots.

Resumo O déficit hídrico, em particular, reduz a produtividade das hortaliças. Para minimizar esses efeitos nocivos à agricultura, diversas práticas agronômicas e fisiológicas estão sendo estudadas, como o uso de bactérias e atenuadores de estresse hídrico, como os brassinosteróides. Considerando a relevância socioeconômica da cultura do milho e sua sensibilidade quando expostos ao déficit hídrico, o objetivo do presente trabalho foi avaliar a ação de brassinosteróides e azospirillum sobre o metabolismo de nitrogênio em plantas de milho submetidas a condições de estresse hídrico. O experimento foi conduzido em casa de vegetação, no período de 47 dias, com plantas de milho, utilizando-se o híbrido K9606 VIP3. O delineamento experimental utilizado foi o inteiramente casualizado, em esquema fatorial 2x2x3, com seis repetições. O primeiro fator corresponde a dois regimes hídricos (presença e ausência de déficit hídrico). A segunda corresponde à inoculação via semente de Azospirillum brasiliense e ausência de inoculação. E a terceira corresponde à aplicação de três concentrações de brassinosteróides (0, 0,3 e 0,6 μM). Foram determinados Nitrato; nitrato redutase; amônio livre; aminoácidos solúveis totais; proteínas solúveis; prolina; glicina, betaína e glutamina sintetase. A falta de água nas plantas proporcionou redução nos teores de proteína e nitrato redutase, nas folhas e raízes. Para o amônio, plantas com déficit hídrico, inoculadas na concentração de 0,3 μM, obtiveram aumento de 7,16 (70,26%) e 13,89 (77,04%) mmol NH4 + .Kg-1. MS (Massa seca) na folha e na raiz, respectivamente. O déficit hídrico no solo proporcionou aumentos significativos nos teores de glicina, betaína, nitrato, prolina e aminoácidos, tanto nas folhas quanto nas raízes das plantas de milho. Por outro lado, os teores de glutamina sintetase apresentaram redução tanto nas folhas quanto nas raízes.

Resumo O feijão-caupi (Vigna unguiculata (L.) Walp.) é uma leguminosa amplamente cultivada por pequenos, médios e grandes produtores em diversas regiões brasileiras. No entanto, uma das preocupações para a produção de feijão-caupi no Brasil nos últimos anos é a baixa atividade pluviométrica nessas regiões, o que gera o acúmulo de sais na superfície. O objetivo deste trabalho foi avaliar os efeitos do estresse salino nos parâmetros de crescimento e atividade enzimática em plantas de feijão-caupi em diferentes concentrações de brassinosteróides. O experimento foi desenvolvido em casa de vegetação com delineamento experimental inteiramente casualizado em esquema fatorial 3 x 3. Os tratamentos consistiram de três níveis de brassinosteróides (0, 3 e 6 µM EBL) e três níveis de estresse salino (0, 50 e 100 mM NaCl). Os fatores de crescimento (altura, diâmetro e número de folhas) diminuíram na condição salina. Com a presença do brassinosteroide a altura não aumentou, mas o número de folhas sim, principalmente na dosagem salina de 100 mM NaCl. Na variável integridade de membrana, o brassinosteróide foi eficiente em ambas as dosagens de salinidade, o mesmo não ocorrendo com o teor relativo de água, onde a condição salina não afetou a quantidade de água na hortaliça, com a aplicação de brassino manteve-se elevada, diminuindo apenas na dosagem de NaCl 100 mM. A enzima nitrato redutase foi bastante afetada no sistema radicular mesmo com a aplicação de doses crescentes de brassino. Portanto, o uso de brassinosteroides como promotor de tolerância salina em plântulas de feijão-caupi foi positivo. A concentração de 3µM de EBL proporcionou o efeito mais satisfatório em tolerar os efeitos deletérios da condição salina. O mesmo não pode ser concluído para a concentração de 6µM de EBL que não promoveu tolerância a algumas variáveis.

Abstract Cowpea (Vigna unguiculata (L.) Walp.) is a legume widely cultivated by small, medium and large producers in several Brazilian regions. However, one of the concerns for the production of cowpea in Brazil in recent years is the low rainfall activity in these regions, which generates the accumulation of salts on the surface. The objective of this work was to evaluate the effects of salt stress on growth parameters and enzyme activity in cowpea plants at different concentrations of brassinosteroids. Experiment was developed in a greenhouse using a completely randomized experimental design in a 3 x 3 factorial scheme. The treatments consisted of three levels of brassinosteroids (0, 3 and 6 µM EBL) and three levels of salt stress (0, 50 and 100 mM NaCl). Growth factors (height, diameter and number of leaves) decreased in the saline condition. With the presence of brassinosteroid the height did not increase, but the number of leaves did, mainly in the saline dosage of 100 mM NaCl. In the variable membrane integrity, brassinosteroid was efficient in both salinity dosages, the same not happening with the relative water content, where the saline condition did not affect the amount of water in the vegetable, with the application of brassino it remained high, decreasing only at dosage 100 mM NaCl. The nitrate reductase enzyme was greatly affected in the root system even with the application of increasing doses of brassino. Therefore, brassinosteroids as a promoter of saline tolerance in cowpea seedlings was positive. The concentration of 3µM of EBL provided the most satisfactory effect in tolerating the deleterious effects of the saline condition. The same cannot be concluded for the concentration of 6µM of EBL that did not promote tolerance to some variables.

Resumo Diferentes espessuras de camadas de fotorresiste foram depositadas sobre a lâmina de Ti, de forma a diminuir a corrente inicial durante o processo de síntese e, assim, evitar ou reduzir a formação da camada inicial compacta de TiO2 sobre os nanotubos. Foi realizado um estudo dos estágios iniciais de crescimento dos nanotubos (NTs) para a síntese convencional e com a presença das camadas de fotorresiste. Os NTs de TiO2 foram caracterizados pela técnica de microscopia eletrônica de varredura. Eletrodos de pH foram fabricados com os NTs obtidos e foram caracterizados. Notou-se a total ausência da camada porosa de TiO2 com o uso da camada de fotorresiste e os eletrodos de pH apresentaram melhora em sua sensibilidade e a ausência do efeito de histerese quando comparados aos fabricados com os nanotubos crescidos sem a camada de fotorresiste.

Abstract Different thicknesses of photoresist layers were deposited on the Ti foil in order to decrease the initial current during the anodization process, avoiding or diminishing in this way the formation of the initial compact TiO2 layer. The studies of the initial synthesis stages were performed in both cases, for the conventional synthesis and for that with photoresist layer on top of the Ti foil. TiO2 nanotube pH electrodes were fabricated to study the effect of this change in the anodization process. The nanostructure morphology was analyzed through scanning electron microscopy technique. Total removal of the undesirable layer and a complete release of the nanotube mouth were obtained. The pH electrodes were characterized utilizing a buffer solution and improved pH sensitivity and absence of hysteresis effects were observed for the devices fabricated with TiO2 nanotubes obtained with the optimized process.

Resumo O presente estudo analisou a eficiência do processo de fotoeletrooxidação como metodologia para a degradação e inativação de adenovírus em água. A concepção experimental emprega uma solução preparada a partir de água estéril contendo 5,107 cópias genômicas/L (gc/L) de uma amostra padrão de adenovírus humano tipo 5 (HAdV-5), dividida em duas partes iguais, uma para servir como controle e outra tratada por fotoeletrooxidação (PEO) durante 3 horas e com uma corrente de 5A. As amostras recolhidas durante o processo de exposição foram analisadas por PCR quantitativo em tempo real (qPCR) para identificação e quantificação do genoma viral. Antes da extração de ácidos nucleicos, um passo de tratamento com DNAse paralelo foi realizado para avaliar a integridade das partículas virais. Um ensaio de qPCR integrado à cultura de células (ICC-qPCR) permitiu analisar a viabilidade de infecção em uma cultura de células. O processo mostrou-se eficaz testada para a degradação viral, com uma redução de 7 log10 da carga viral após 60 minutos de tratamento. As amostras tratadas com DNAse exibiram redução completa da carga viral após uma exposição de 75 minutos ao processo, e a análise de ICC-qPCR mostrou partículas virais completamente não-viáveis em 30 minutos de tratamento.

The present study analyzed the efficiency of the photo-electro-oxidation process as a method for degradation and inactivation of adenovirus in water. The experimental design employed a solution prepared from sterile water containing 5.107 genomic copies/L (gc/L) of a standard strain of human adenovirus type 5 (HAdV-5) divided into two equal parts, one to serve as control and one treated by photo-electro-oxidation (PEO) for 3 hours and with a 5A current. Samples collected throughout the exposure process were analyzed by real-time polymerase chain reaction (qPCR) for viral genome identification and quantitation. Prior to gene extraction, a parallel DNAse treatment step was carried out to assess the integrity of viral particles. Integrated cell culture (ICC) analyses assessed the viability of infection in a cell culture. The tested process proved effective for viral degradation, with a 7 log10 reduction in viral load after 60 minutes of treatment. The DNAse-treated samples exhibited complete reduction of viral load after a 75 minute exposure to the process, and ICC analyses showed completely non-viable viral particles at 30 minutes of treatment.

The current world demand for bioethanol is increasing as a consequence of low fossil fuel availability and a growing number of ethanol/gasoline flex-fuel cars. In addition, countries in several parts of the world have agreed to reduce carbon dioxide emissions, and the use of ethanol as a fuel (which produces fewer pollutants than petroleum products) has been considered to be a good alternative to petroleum products. The ethanol that is produced in Brazil from the first-generation process is optimized and can be accomplished at low cost. However, because of the large volume of ethanol that is produced and traded each year, any small improvement in the process could represent a savings of billions dollars. Several Brazilian research programs are investing in sugarcane improvement, but little attention has been given to the improvement of yeast strains that participate in the first-generation process at present. The Brazilian ethanol production process uses sugarcane as a carbon source for the yeast Saccharomyces cerevisiae. Yeast is then grown at a high cellular density and high temperatures in large-capacity open tanks with cells recycle. All of these culture conditions compel the yeast to cope with several types of stress. Among the main stressors are high temperatures and high ethanol concentrations inside the fermentation tanks during alcohol production. Moreover, the competition between the desired yeast strains, which are inoculated at the beginning of the process, with contaminants such as wild type yeasts and bacteria, requires acid treatment to successfully recycle the cells. This review is focused on describing the problems and stressors within the Brazilian ethanol production system. It also highlights some genetic modifications that can help to circumvent these difficulties in yeast.
