ABSTRACT Introduction: Elderly patients are the most frequent patients admitted to dialysis units. Relatively little is known about standards on endof-life (EOL) care among dialysis patients. From international literature, these patients are exposed to more aggressive treatments at EOL than patients with cancer or other chronic diseases and palliative referral is infrequent. This study aims to describe some end-of-life (EOL) practices in patients undergoing hemodialysis in NephroCare clinics in several cities in Portugal. Material and Methods: We designed a prospective, multicenter, observational, cross-sectional study conducted between October 1st, 2020 and September 30th, 2021 in six hemodialysis clinics including 1265 patients (pts). Results: During the year in analysis, 158 pts died (12.4%). Mean age was 76 years (25% of pts were older than 85 years) and dialysis vintage was 84 (2-963) months. Regarding comorbidities, 50% were diabetic, 40% had congestive heart failure, 25% had cancer and 17% had dementia. Mean age-adapted Charlson index was 13 ± 3. For 82 pts (51.9%), nephrologists would not be surprised if they died in the next 6 months. Seventy-three had hospital admissions in the previous three months. Four pts withdrew dialysis. Of those who died, only two had advance care directives, 18 had an opioid prescription in the last month of life and 17 were referred to palliative intervention mostly because of oncologic disorders. Seventy four percent of these pts died at the hospital and 23% died at home/nursing home. Not a single patient died under hospice care. Conclusion: Most of our patients commonly die in hospital, undergo invasive procedures, spend much of their remaining life fulfilling a demanding dialysis schedule and are admitted to the hospital wards without formal palliative intervention or discussion of goals of care, even when death is an expected outcome.

ABSTRACT Supportive care in patients with chronic kidney disease refers to the application of palliative medicine principles and practices in nephrology. The main purpose is to reduce suffering by managing symptoms, helping with decision making and providing holistic support to the patient and family/caregiver in clinical, social, spiritual or nutritional distress, with a multidisciplinary team. We present the case of a 92-year-old male patient with chronic kidney disease of unknown etiology since 2004. In 2019, he was referred to our kidney disease supportive care program, presenting a serum creatinine of 5.44 mg/dL (estimated glomerular filtration rate of 9 mL/min/1.73 m2). During the following three years, the patient lived at home independently, maintained good control of his chronic kidney diseaseassociated symptoms and preserved his functional status. We present this case as a practical example of how to approach an autonomous elderly, chronic kidney disease patient, with few comorbidities, who opted for supportive care.

RESUMO Objetivo Identificar relações entre a utilização sistemática do Microfone Remoto (MR) em sala de aula de estudantes com deficiência auditiva e características das escolas e dos professores. Método Foram analisados 120 sujeitos, entre cinco e 17 anos, com deficiência auditiva que foram adaptados MR em um Serviço de Saúde credenciada pelo Sistema Único de Saúde (SUS). Também foram sujeitos, professores de usuários de MR. Realizou-se uma análise de prontuários e no momento que os sujeitos compareceram para acompanhamento foi realizado entrevista com os pais/responsáveis para caracterizar rotina de utilização do MR na escola. Foram realizados contatos telefônicos e visitas presenciais em algumas escolas. Resultados Quanto ao uso, observa-se que a maioria dos sujeitos utilizava o dispositivo na escola. Aqueles que não utilizavam involuntariamente e voluntariamente consistiu na minoria dos sujeitos. Houve uma similaridade no padrão do Speech Intelligibility Index -SII dos sujeitos que ‘usam’ e ‘não usam involuntariamente’ o MR. Houve diferença significativa entre o tipo de escola e o nível de escolaridade, a maioria dos sujeitos que frequentavam escola regular e estavam matriculados no ensino fundamental I tendem a usar mais o dispositivo. Conclusão A maioria dos sujeitos faz uso do MR na escola. O nível educacional do estudante também foi um fator que interferiu na adesão ao uso dos MR, com maior adesão em estudantes do Ensino Fundamental I. Os dados sugerem que a articulação entre serviço de saúde e escola favorece a utilização do MR, entretanto quando essa relação é intermediada pelos pais, outros fatores acabam interferindo no uso sistemático no cotidiano da escola.

ABSTRACT Purpose To identify relationships between Remote Microphone System (RMS) use in the classroom and the schools’ and teachers’ characteristics. Methods We analyzed 120 subjects aged 5 to 17 years with hearing loss who had received an RMS from a health service accredited by the Unified Health System (SUS). The teachers of RMS users were the other subjects in the study. We analyzed the patients’ medical records and interviewed their parents/guardians at the follow-up visit to verify issues related to the RMS and its use at school. We contacted the schools over the phone and visited some of them. Results Of the students, 54% used the device at school; 22% involuntarily did not use it; and 24% voluntarily did not use it. The Speech Intelligibility Index pattern of those who used the RMS was similar to those who involuntarily did not use it. There was a significant difference between the type of school and educational level – 86% of regular school students and elementary school students tend to use the device more often (62%). Conclusion Most subjects use the RMS at school. The students’ educational level also interfered with the adherence to RMS use, as elementary school students had a higher adherence. The data suggest that the coordination between health services and schools favors RMS use. However, when the parents mediate this relationship, other factors interfere with the systematic RMS use in the school routine.

RESUMO Objetivo Estabelecer relações entre o desempenho em tarefa de reconhecimento de palavras com e sem sentido e grau e configuração da perda auditiva, utilizando valores de Índices de Inteligibilidade de Fala (SII) como indicadores, em crianças com deficiência auditiva. Método Foram estabelecidos os SII para sons de entradas de 55 e 65 Decibéis Nível de Pressão Sonora (dBNPS) de dez crianças com perda auditiva neurossensorial usuárias de aparelho de amplificação sonora individual bilateralmente que têm a linguagem oral como principal modalidade de comunicação. As crianças foram submetidas à tarefa de repetição de palavras com e sem sentido em duas ou três diferentes intensidades. As emissões foram analisadas de acordo com o Protocolo Word Association for Syllable Perception (WASP). Na análise dos dados, o SII foi comparado com os resultados obtidos em cada critério de análise. Resultados Para o desempenho em palavras, houve diferença estatisticamente significante entre os dois tipos de estímulo em 55dBNPS. Para o desempenho em consoantes e ponto de articulação, houve diferença estatisticamente significante entre os tipos de estímulos em 65 e 55dBNPS e também entre as intensidades de 65 e 55 dBNPS nas palavras sem sentido. Conclusão De modo geral, não houve regularidade na relação entre capacidade auditiva e desempenho em tarefas de percepção da fala. Os resultados sugerem que o desempenho nas tarefas de reconhecimento de palavras sem sentido tem maior relação com o índice de inteligibilidade do que as palavras com sentido, possivelmente por limitar as estratégias de fechamento semântico pelo sujeito.

ABSTRACT Purpose To establish the relationship between the performance on word recognition tasks, using words with and without sense and degree, and the configuration of hearing loss, by using Speech Inteligibility Index (SII) values as indicators, in children with hearing loss. Methods SII were established for 55 and 65 Decibel of Sound Pressure Level (dB SPL) input sounds of ten children presenting bilateral sensorineural hearing loss (SNHL), adapted with bilateral hearing aids, and who have oral language as the main mode of communication. The children were submitted to a word and nonsense-word repetition task of two or three intensity degrees. Their productions were analyzed according to the Word Association for Syllable Perception (WASP) Protocol. In the data analysis, the values of SII were compared with the results obtained in each analysis criterion. Results Pertaining to the words, there was statistically significant difference between the two types of stimuli in 55 dBSPL. As for the performance of consonants and point of articulation, there was a statistically significant difference between stimuli types in 65 and 55 dB SPL, and between intensities 65 and 55 dB SPL in nonsense words. Conclusion Overall, there was no regularity in the relationship between hearing ability and performance in speech perception tasks. The results suggest that performance in the nonsense words recognition tasks was more related to intelligibility index than to words with meaning, possibly because it limits semantic closure strategies by the subject.

The influence of common tea preparation procedures (temperature, infusion time, consumption time interval and tea bag/loose-leaf) and the type of water used, on the total phenolic content (TPC), the radical scavenging activity and the α-glucosidase inhibitory activity were assessed. Higher TPC and antioxidant activity were obtained when using lower mineralized waters. Tea bags also evidenced higher antioxidant activity than loose-leaf samples. Under the same conditions (90 ºC and five minutes of infusion time) green tea contains almost twice the quantity of polyphenols and the free radical scavenging ability of black tea. In the α-glucosidase assay all infusions were active (97-100 %). Furthermore, HPLC allowed to identify some of the polyphenols present in both teas and to monitor their composition change with time. After twenty-four hours, the antioxidant activity was maintained without significant changes, but a small decrease in enzyme inhibition was observed, although this activity was still very high.

Resumo A organogênese in vitro das tangerinas Fremont (Citrus clementina x Citrus reticulata), Thomas (Citrus reticulata) e Nules (Citrus clementina) foi avaliada visando a otimizar o protocolo de regeneração para aplicação da transformação genética. O uso de explantes derivados de epicótilo resultou em maior número de explantes responsivos e maior número de brotações desenvolvidas por explante, quando comparado ao uso de explantes derivados de segmentos internodais. A maior eficiência na regeneração de brotos foi observada na presença de 1 mg L-1 de BAP, independentemente do tipo de explante e da cultivar. O protocolo de organogênese in vitro produziu plantas transgênicas das três cultivares de tangerina, expressando o gene attA sob o controle de promotores específicos de floema.

Abstract The in vitro organogenesis of Fremont (Citrus clementina x ), Citrus reticulataThomas (Citrus reticulata), and Nules (Citrus clementina) mandarins was evaluated aiming to optimize a regeneration protocol that could be applied in genetic transformation. The use of epicotyl-derived explants resulted in higher explant responsiveness and number of shoots developed per explant when compared with the use of internodal-derived explants. The highest efficiency in shoot regeneration was observed in the presence of 1 mg L-1 of BAP, regardless of the explant type and cultivar. The in vitro organogenesis protocol produced transgenic plants from three mandarin cultivars expressing attA gene under the control of phloem-specific promoters.

RESUMO Objetivo Comparar valores do índice de inteligibilidade de fala amplificado, para diferentes entradas de sinal de fala (75, 65 e 55 dBNPS), gerados no processo de verificação dos aparelhos de amplificação sonora, nos diferentes graus e configurações de perdas auditiva. Métodos Foram selecionadas 41 crianças com idades entre 3 e 80 meses, totalizando 78 orelhas (quatro orelhas foram excluídas). As perdas auditivas foram classificadas conforme grupos de audibilidade e intervalos do índice de inteligibilidade de fala. Foram analisados 234 valores do índice de inteligibilidade de fala para os estímulos de fala e estes foram também analisados, em relação aos grupos e intervalos de audibilidade. Resultados Os grupos de audibilidade se deslocaram entre os intervalos do índice de inteligibilidade de fala, indicando a variação, conforme o nível de apresentação do sinal de entrada. Duas equações foram geradas para prever valores do índice de inteligibilidade de fala a partir do valor deste índice para o sinal de entrada de 65. Conclusão Menores níveis de pressão sonora do estímulo de fala produzem menores índices de inteligibilidade de fala. Esta diferença é mais acentuada nos deficientes auditivos, que apresentam valores do índice de inteligibilidade de fala de 65 entre 36% e 55%.

ABSTRACT Purpose To compare aided speech intelligibility index (SII) values in different speech signal inputs (75, 65, and 55 dBNPS), generated from the verification process of hearing aids, under different hearing loss severities and configurations. Methods 41 children aged between three and 80 months old were selected, totalizing 78 ears (after exclusion of four ears). Hearing loss was classified according to audibility groups and SII intervals. Two hundred and thirty-four (234) SII values were analyzed as per speech stimulus and with regard to groups and audibility intervals. Results Movement of audibility groups along SII intervals was observed, which indicates variation associated with the input signal intensity, as well as with the distance between the sound source and the hearing aid microphone. Two equations were generated to predict SII values from input signal SII65. Conclusion Lower levels of speech stimulus sound pressure produce lower levels of speech intelligibility. This difference is more pronounced in hearing-impaired persons, who present SII values between 36 and 55%.

Important properties in cementitious materials, such as concrete, are related to the presence of additives that influence the rigidity and the physical and chemical resistances. For the evaluation of the additive effectiveness, known as pozzolanic activity, a simple procedure, the Chapelle test, is commonly used, and it essentially consists in a reaction between the additive with calcium oxide in aqueous medium. However, such procedure presents limitations in terms of processing time and lack of information regarding the reactions kinetics. In this sense, a simple method based on the kinetic and thermodynamic principles of chemical reactions is proposed, which can be performed using conventional electronic pH sensors. The study provides an alternative methodology with many advantages over the traditional procedure, such as energy and time-savings, more robustness and more confidence. In this paper, three types of silica nanoparticles that can be used as low-cost additives were characterized in relation to their morphology and crystallinity by XRD and SEM, the particles average diameters were obtained and the particles were used for studying the chemical process that takes place during the Chapelle test. Results and the semi-empirical analysis provided strong evidence that the process is an acid-base 1:1 reaction and it was verified that the mean reaction times varied from 64 to 195 min. It is a remarkable result, since the proposed analysis can be performed with simple, fast and low-cost instrumentation and needs only a worksheet software, whereas the Chapelle test takes 16 hours and provides no dynamic information. Besides the limitation that the methodology is not able to quantify and to elucidate the effects of the specific surface area of the particles, which needs a complete BET study, the research provides a significant contribution for the understanding of the pozzolanic process, of great importance in both concrete and ceramic research.

RESUMO A utilização de promotores preferencialmente expressos em tecidos vegetais específicos é uma estratégia desejável na busca por resistência a patógenos que colonizam tais tecidos. A bacteria Candidatus Liberibacter asiaticus (Las), associada à doença huanglongbing (HLB) em citros, coloniza o tecido floemático. Alguns promotores, além de conferir expressão específica em certos tecidos, podem também responder diferencialmente à presença de patógenos. O objetivo deste estudo foi verificar se a presença de Las poderia modular a ativação dos promotores de floema AtPP2 (Arabidopsis thaliana phloem protein 2), AtSUC2 (A. thaliana sucrose transporter 2) e CsPP2 ( pCitrus phloemrotein 2), conhecidos por expressão específica em floema de Citrus sinensis. Plantas de laranja ‘Hamlin’ (Citrus sinensis L. Osbeck) transformadas com o gene reporter uidA (GUS) sob controle dos promotores AtPP2, AtSUC2 e CsPP2 foram infectadas para avaliar a interdependência entre a expressão dos transgenes e concentração de Las. Plantas foram inoculadas com Las por meio de Diaphorina citri e dezoito meses após, a concentração de bactéria e expressão do gene uidA foram determinadas por qPCR e RT-qPCR, respectivamente. A expressão do gene reporter dirigida pelo promotor AtSUC2 foi forte e positivamente correlacionada com a concentração de Las. Portanto, este promotor contém características desejáveis quanto à especificidade de expressão e indução específica por patógeno para a produção de plantas transgênicas tolerantes a Las.

ABSTRACT The use of promoters preferentially expressed in specific plant tissues is a desirable strategy to search for resistance for pathogens that colonize these tissues. The bacterium Candidatus Liberibacter asiaticus (Las), associated with huanglongbing disease (HLB) of citrus, colonizes phloem vessels. Some promoters, besides conferring tissue-specific expression, can also respond to the presence of the pathogen. The objective of the present study was to verify if the presence of Las could modulate the activation of the phloem-specific promoters AtPP2 (Arabidopsis thaliana phloem protein 2), AtSUC2 (A. thaliana sucrose transporter 2) and CsPP2 ( pCitrus phloemrotein 2), known to be expressed in Citrus sinensis phloem. ‘Hamlin’ sweet orange plants (Citrus sinensis L. Osbeck) transformed with the uidA (GUS) reporter gene under the control of AtPP2, AtSUC2 and CsPP2 promoters were infected to evaluate the interdependence between transgene expression and the concentration of Las. Plants were inoculated with Las by Diaphorina citri and eighteen months later, bacterial concentration and uidA expression were determined by qPCR and RT-qPCR, respectively. Reporter gene expression driven by AtSUC2 promoter was strongly and positively correlated with Las concentration. Therefore, this promoter combines desirable features of both tissue-specificity and pathogen-inducibility for the production of transgenic plants tolerant to Las.

RESUMO Objetivo estabelecer relações entre valores do índice de inteligibilidade de fala - SII gerados na verificação dos aparelhos de amplificação sonora programados conforme regra prescritiva DSLm[i/o]v5 e uma proposta de classificação de indivíduos que considere a associação de grau e configuração de perdas auditivas. Método foram selecionadas 41 crianças com idades entre 4 e 80 meses, totalizando 78 orelhas para análise. Foram considerados os limiares auditivos nas frequências 250, 500, 1000, 2000 e 4000 Hz e analisados valores de SII para os sinais de entrada 65 dB NPS, obtidos na verificação dos AASI no equipamento Verifit®Audioscan. Resultados as perdas auditivas foram classificadas em cinco grupos homogêneos quanto às características audiológicas (grau e configuração audiométrica) e heterogêneos entre si. A partir dos grupos, determinaram-se três intervalos de valores de SII. Foram determinadas equações para classificação da perda auditiva conforme grupos e equações para determinação de valores de SII ajustado. Conclusão o valor de SII pode ser considerado um indicador da audibilidade para sons de fala para diferentes características de perdas auditivas e nortear avalições de comportamento auditivo. O SII tem relação mais forte com a associação das variáveis grau e configuração audiométrica, quando comparado com sua relação com o grau da perda auditiva isoladamente.

ABSTRACT Purpose To establish the relationship between speech intelligibility index (SII) values generated at the verification of hearing aids programmed according to DSLm [i/o] v5 prescription rule and a proposed individual classification that considers the combination of hearing loss degree and configuration. Methods Forty-one children aged between 4 and 80 months were selected, totaling 78 ears for analysis. We considered hearing thresholds at the frequencies of 250, 500, 1000, 2000 and 4000 Hz; and analyzed values of the Speech Intelligibility Index (SII) for the input signal of 65 dB SPL obtained during the verification of hearing aids using the equipment Verifit®Audioscan. Results Hearing losses were classified into five homogeneous groups regarding audiometric degree and configuration. The groups were heterogeneous when compared to each other. From the groups, three ranges of SII values were determined. Equations were developed for classification of hearing loss according to groups and for determination of the adjusted SII values. Conclusion The SII value is a useful indicator of audibility for speech sounds in different characteristics of hearing losses, and can guide observations of auditory skills. The SII has stronger relationship with the association of the audiometric degree and configuration when compared with degree of hearing loss alone.

RESUMO Objetivo Adaptação semântica e cultural de versão preliminar em português do questionário LittlEars® em famílias de crianças com deficiência auditiva. Métodos Instrumento administrado como entrevista ou questionário, em tradução validada do Questionário LittlEars® em português em pais de crianças com deficiência auditiva com até dois anos de idade auditiva. Comentários, dúvidas, sugestões, exemplos, críticas ou dificuldades dos pais foram utilizadas para elaboração da versão adaptada. Foi então realizada reunião para brainstorming com grupo de especialistas, para validação da versão final. Resultados Trinta e sete sujeitos, pais ou responsáveis de 32 crianças, responderam ao questionário. Somente quatro pais preferiram responder no formato de questionário, enquanto 28, em entrevista. No formato entrevista, houve dificuldade de compreensão em diversos itens. Após análise e discussão dos comentários das famílias, foi proposta uma versão modificada. Conclusão As modificações nos itens do questionário estiveram relacionadas a vocabulário, expressões idiomáticas e exemplos desconhecidos. Os resultados obtidos com o questionário LittlEars® no grupo estudado variaram conforme a idade auditiva e o Índice de Inteligibilidade de Fala (Speech Intelligibility Index - SII), o que sugere que a tradução adaptada tem sensibilidade para medir as habilidades auditivas. São necessários estudos com a versão proposta, em uma população maior, visando sua validação final.

ABSTRACT Purpose Semantic and cultural adaptation of a preliminary Portuguese version of the questionnaire LittlEars® in families of children with hearing loss. Methods Instrument administered as an interview or questionnaire, using a validated translation of LittlEars® Questionnaire in Portuguese with parents of children with hearing loss up to two years of hearing age. Parent’s comments, questions, suggestions, examples, comments or difficulties were used for development of the adapted version. A brainstorming session with a group of experts was held to validate the final version. Results Participants were 37 individuals who answered the questionnaire, parents of 32 children. Only four parents preferred to answer the questionnaire format, while 28 preferred to answer it as an interview. In the interview format, parents had difficulty understanding different items. Upon review and discussion of the comments of the families, a modified version was proposed. Conclusion Changes in the questionnaire items were related to vocabulary, idiomatic expressions and unknown examples. The results of the questionnaire LittlEars® varied depending on hearing age and Speech Intelligibility Index - SII, suggesting that the adapted translation has sensitivity for measuring the hearing abilities. Other studies should apply the new version and final validation.

O objetivo deste trabalho foi avaliar a reação de quatro cultivares de laranjeira doce que expressam o gene atacina A à infecção por 'Candidatus Liberibacter asiaticus' (Las), bactéria associada à doença huanglongbing (HLB). Plantas transgênicas das cultivares Hamlin, Natal, Pêra e Valência, bem como controles não transgênicos, receberam inóculos, por enxertia, de seções de ramos infectados por HLB. A progressão da doença foi avaliada pela observação de sintomas foliares e por análise da reação em cadeia da polimerase (PCR), oito meses após a inoculação. Utilizou-se o delineamento inteiramente casualizado, com quatro experimentos (um para cada cultivar) realizados simultaneamente. A concentração bacteriana foi estimada por PCR quantitativo (qPCR). Sintomas de HLB e Las estavam presentes em plantas não transgênicas e em plantas transgênicas que expressaram o gene atacina A, nas quatro cultivares de laranjeira doce, oito meses após a inoculação. Cinco linhagens transgênicas (eventos de transformação) de laranjeira 'Pêra', que expressam o gene atacina A, apresentam concentrações de Las significativamente inferiores às das plantas não transgênicas desta cultivar.

The objective of this work was to evaluate the reaction of four sweet orange cultivars expressing the attacin A gene to 'Candidatus Liberibacter asiaticus' (Las) infection, a bacterium associated to huanglongbing (HLB) disease. Transgenic sweet orange plants of Hamlin, Natal, Pêra, and Valência cultivars, as well as nontransgenic controls received inocula by grafting budwood sections of HLB-infected branches. Disease progression was evaluated through observations of leaf symptoms and by polymerase chain reaction (PCR) analysis, eight months after inoculation. A completely randomized design was used, with four experiments (one for each cultivar) performed simultaneously. Bacteria title was estimated by quantitative PCR (qPCR). HLB symptoms and Las titers were present in nontransgenic and transgenic plants expressing the attacin A gene of the four sweet orange cultivars, eight months after bacteria inoculation. Five transgenic lines (transformation events) of 'Pêra' sweet orange expressing the attacin A gene have significantly lower Las titers in comparison with nontransgenic plants of this cultivar.

O objetivo deste trabalho foi obter plantas transgênicas de laranja 'Pêra' e 'Valência' por meio do gene D4E1 dirigido pelo promotor Arabidopsis thaliana phloem protein (AtPP2) e quantificar a expressão dos transgenes nos diferentes eventos de transformação. Os experimentos de transformação genética foram realizados com segmentos de epicótilo cocultivados com Agrobacterium tumefaciens. Seis plantas de laranja 'Pêra' e sete plantas de laranja 'Valência' foram confirmadas como distintos eventos transgênicos por meio das técnicas de reação em cadeia da polimerase (PCR) e Southern blot. A expressão dos transgenes foi quantificada por PCR quantitativo em tempo real. Os níveis de expressão do gene D4E1 variam de 5 a 50 vezes entre os diferentes eventos de transformação.

The objective of this work was to produce transgenic 'Pêra' and 'Valência' sweet orange plants using the D4E1 gene driven by the Arabidopsis thaliana phloem protein (AtPP2) promoter and to quantify transgene expression in different transformation events. Genetic transformation experiments were carried out with epicotyl segments co‑cultivated with Agrobacterium tumefaciens. Six plants from 'Pêra' sweet orange and seven plants from 'Valência' sweet orange were confirmed as different transgenic events by means of the polymerase chain reaction (PCR) and the Southern blot techniques. Transgene expression was quantified using real‑time quantitative PCR. D4E1 gene expression levels vary from 5 up to 50 times among different transformation events.

A organogênese in vitro de limão Cravo (Citrus limonia Osbeck) foi estudada em função do tipo de explante e da suplementação do meio de cultura com citocinina. Foram avaliados quatro tipos de explantes coletados da região do epicótilo e do hipocótilo de plântulas germinadas in vitro. Os explantes consistiram em segmentos de epicótilo, segmentos de hipocótilo, segmentos de hipocótilo ligado a um fragmento do cotilédone e segmentos de hipocótilo cortado longitudinalmente. Os explantes foram cultivados em meio de cultura EME suplementado com benzilaminopurina (0; 0,5; 1,0 e 1,5 mg L-1). As avaliações foram realizadas após seis semanas de incubação, e os melhores resultados considerando-se tanto a porcentagem de explantes responsivos como o número de gemas desenvolvidas por explante foram obtidos com o uso de segmentos de epicótilo como explante. Considerando os explantes derivados da região do hipocótilo, não foi detectada diferença na porcentagem de explantes responsivos com o uso de segmentos de hipocótilo ou de segmentos de hipocótilo ligado a um fragmento do cotilédone. A porcentagem de explantes responsivos diminuiu com o uso de segmentos de hipocótilo cortado longitudinalmente como explante. Não foi detectada nenhuma diferença no número de gemas adventícias desenvolvidas por explante considerando-se os três tipos de explantes derivados da região do hipocótilo. A suplementação do meio de cultura com BAP não é essencial para a organogênese in vitro de limão Cravo. No entanto, o desenvolvimento de gemas adventícias foi estimulado com concentrações entre 0,5 - 1,0 mg L-1.

Rangpur lime (Citrus limonia Osbeck) in vitro organogenesis was studied based on explant type and cytokinin culture media supplementation. Four explants types collected from epicotyl or hypocotyl regions of in vitro germinated seedlings were evaluated. The epicotyl-derived explants consisted of epicotyl segments and the hypocotyl-derived explants consisted of the entire hypocotyl segment, the hypocotyl segment attached to a cotyledon fragment, and the hypocotyl segment divided longitudinally. The explants were cultured on EME culture medium supplemented with benzylaminopurine (0, 0.5, 1.0, or 1.5 mg L-1). The evaluation was performed after 6 weeks. Best results considering both the explant responsiveness and number of shoots developed per explants were obtained when epicotyl segments-derived explants were evaluated. Considering the explant responsiveness of hypocotyl segments-derived explants no difference was detected between the entire hypocotyl segment and the hypocotyl segment attached to a cotyledon fragment. Moreover, the percentage of responsive explants decreased when hypocotyl segments divided longitudinally were tested. No difference was detected for the number of shoots developed per explant considering the three types of hypocotyl-derived explants. Culture media supplementation with BAP was not essential for Rangpur lime in vitro organogenesis. However, adventitious shoot development was stimulated in concentrations between 0.5 - 1.0 mg L-1.

O objetivo do trabalho foi estudar um protocolo para o cultivo in vitro de Cucurbita pepo cv. Caserta utilizando como explante um segmento de cotilédone associado a um fragmento do hipocótilo. Explantes foram coletados de plântulas germinadas in vitro com 4 a 6 dias de idade, cultivados em meio de cultura basal MS suplementado com benzilaminopurina (BAP, 4,5 µM), e incubados sob fotoperíodo de 16 h, a 27ºC. Com base nos resultados obtidos, explantes coletados de plântulas germinadas in vitro com 4 dias de idade foram cultivados em meio de cultura MS, suplementados com diferentes concentrações de BAP (0; 1,1; 2,2; 3,3; 4,5 ou 5,5 µM), e incubados sob fotoperíodo de 16 h, a 27ºC. Após 4 semanas de incubação, foi possível detectar o desenvolvimento de gemas adventícias na junção do cotilédone e hipocótilo. As gemas adventícias desenvolvidas foram transferidas para meio de cultura de alongamento e enraizamento e as plantas obtidas foram aclimatizadas para condições de casa-de-vegetação. O processo morfogênico foi caracterizado por análises de microscopia ótica e eletrônica de varredura, confirmando a organogênese. Os resultados obtidos mostram a eficiência do protocolo no cultivo in vitro de abobrinha-de-moita cv. Caserta utilizando este explante alternativo. O protocolo será testado, no futuro, para experimentos de transformação genética desta espécie.

A protocol for the in vitro culture of Cucurbita pepo cv. Caserta was studied, using a cotyledon segment with an attached hypocotyl fragment as an explant. First, to determine the optimal seedling age, explants were collected from 4 to 6-day-old in vitro germinated seedlings and cultured in MS basal medium supplemented with benzylaminopurine (BAP, 4.5 µM), under a 16-h photoperiod at 27ºC. Based on the results obtained, the explants collected from the 4-day-old seedlings were then cultured in MS basal medium supplemented with different concentrations of BAP (0, 1.1, 2.2, 3.3, 4.5, or 5.5 µM) and incubated under a 16-h photoperiod at 27ºC. In vitro organogenesis was most efficient with explants collected from 4-day-old seedlings cultured in medium supplemented with 4.5 µM of BAP. After 4 weeks of incubation the development of adventitious buds at the cotyledon/hypocotyl junction could be observed. These buds were transferred to elongation and rooting medium and the developed plants were acclimatized to greenhouse conditions. The morphogenic process was characterized using light and scanning electron microscopy analyses to confirm the organogenesis. The results showed that this alternate explant is efficient for in vitro culture of zucchini squash cv. Caserta. The protocol will be further examined for future use in genetic transformation experiments in this species.
