Resumo Espécies de plantas que mostram padrão gregário de distribuição e endemismo na Floresta Atlântica, como Dorstenia elata, são particularmente sensíveis aos efeitos da perda da diversidade genética. Considerando isso, objetivou-se com este trabalho quantificar a diversidade genética em populações nativas dessa espécie em um remanescente da Mata Atlântica. A amostra incluiu três subpopulações de indivíduos e a caracterização molecular foi realizada com doze primers ISSR. As análises intrapopulacionais foram baseadas no cálculo do índice de Shannon; a heterozigosidade total esperada e a matriz de distâncias entre pares de indivíduos também foram calculadas. O dendrograma de agrupamento obtido evidenciou a formação de dois grupos. As investigações interpopulacionais foram baseadas na análise de variância molecular e na estimativa do fluxo gênico. Os resultados demonstram que um grupo compreendeu os genótipos de duas subpopulações, e o outro continha exclusivamente os genótipos de uma terceira subpopulação. A maior variabilidade genética foi observada dentro e não entre populações, indicando que a distância geográfica e a estrada que divide as populações estudadas não estão causando perda de diversidade genética.

Abstract Plant species that show gregarious spatial distribution and endemism to the Atlantic Forest, such as Dorstenia elata, are particularly sensitive to the effects of genetic diversity loss. In the present study, we aimed to quantify the genetic diversity in native populations of this species in an Atlantic Forest remnant. The sample included three aggregates of individuals, and molecular characterization was performed with twelve ISSR primers. Intrapopulation analyses were based on the calculation of the Shannon index; total expected heterozygosity and the matrix of distances between pairs of individuals were also calculated. The obtained grouping dendrogram evinced the formation of two groups. Interpopulation investigations were based on the analysis of molecular variance and the estimate of historical gene flow. The results demonstrate that one group comprised the genotypes from two subpopulations, and the other contained exclusively the genotypes of a third subpopulation. The greatest genetic variability was observed within rather than among populations, indicating that the geographical distance and the road that divides the studied populations are not causing loss of genetic diversity.

Resumo Neste trabalho são apresentadas as espécies de Passifloraceae sensu stricto do estado do Espírito Santo, Brasil. A família encontra-se representada por dois gêneros, Mitostemma, com apenas M. glaziovii e Passiflora com 30 espécies, com representantes de quatro subgêneros: duas espécies pertencentes ao subgênero Astrophea, seis ao subgênero Debaloba, uma ao subgênero Deidamioides, e vinte e um ao subgênero Passiflora. Dentre as espécies, P. margaritae é classificada como “em perigo” (EN), de acordo com os critérios da IUCN, e temos o primeiro registro de P. cacao e P. campanulata para o estado. São apresentadas descrições, dados de distribuição geográfica e comentários.

Abstract A treatment of the species of Passifloraceae sensu stricto from the state of Espírito Santo, Brazil is presented. The family is represented by two genera: Mitostemma, with M. glaziovii, and Passiflora with 30 species in four subgenera: two species belonging to subgenus Astrophea, six to subgenus Decaloba, one to subgenus Deidamioides, and twenty one to subgenus Passiflora. Among the species, we highlight P. margaritae, considered endangered (EN), according to the IUCN, and we record the first occurrence of P. cacao and P. campanulata for the state. Descriptions, photos, distribution data and taxonomic comments regarding the species are presented.

The molecular characterization of ten genotypes of the Coffea arabica plants and of seven genotypes of C. canephora having interesting features for coffee breeding programs was carried to select the parents for breeding. A total of 40 SSR and 29 ISSR primers were used. The primers generated a total of 331 (307 polymorphic and 24 monomorphic) bands. Analysis of genetic diversity presented dissimilarity intervals ranging from 0.22 to 0.44 between the Conilon genotypes, from 0.02 to 0.28 between the Arabica genotypes, and from 0.49 to 0.60 between the genotypes of the two species in the joint analysis. Four groups were formed: I = genotypes of C. arabica, II = four progenies of C. canephora, Conilon group, and one non defined C. canephora (Conilon or Robusta), III = one progeny of un-defined C. canephora (Conilon or Robusta) and IV = one progeny of C. canephora of Robusta group. The grouping formed was consistent with the origins of each group. High stabilities of the bifurcations were found by bootstrap analysis. The use of molecular markers of the SSR and ISSR types in the diversity study was efficient in distinguishing genotypes between and within C. arabica and C. canephora.

O biribazeiro é uma planta frutífera nativa das matas Atlântica e Amazônica. Seus frutos têm grande aceitação popular para consumo in natura. Objetivou-se com este estudo a avaliação da diversidade genética de acessos de biribazeiro (Rollinia mucosa [Jacq.] Baill) com a utilização de marcadores moleculares ISSR. Foram analisados 16 acessos com 20 primers ISSR, os quais produziram um total de 118 bandas, sendo 96 polimórficas e 22 monomórficas. Os valores de dissimilaridade genética, calculados de acordo com o complemento do índice de Jaccard, variaram de 0,0909 a 0,5147. O método UPGMA (Unweighted Pair Group Method Average) agrupou os acessos em seis grupos. Os acessos 1 e 5 foram mais dissimilares e 11 e 12 os menos dissimilares. Os marcadores ISSR utilizados neste estudo demonstraram eficiência na detecção de polimorfismos moleculares, revelando variabilidade genética entre os 16 acessos. Diante dos resultados obtidos neste trabalho, é possível inferir que existe considerável variabilidade genética entre os acessos de biribazeiro, demonstrando a importância dos marcadores na análise de variabilidade de espécies pouco estudadas, como Rollinia mucosa [Jacq.]Baill.

The biribazeiro is a fruit plant native from Amazonian and Atlantic forests, in Brazil. Their fruits have great popular acceptance for fresh consumption. The objective of this study was to measure the genetic divergence of biriba genotypes (Rollinia mucosa [Jacq.]Baill) using ISSR molecular markers. Sixteen genotypes of biriba were screened with 20 ISSR primers, which produced a total of 118 bands, with 96 polymorphic and 22 monomorphic fragments. The genetic dissimilarity values ranged from 0.0909 to 0.5147, based on the complement of the Jaccard index. The UPGMA (Unweighted Pair Group Average Method) grouped the accessions into six groups. The genotypes 1 and 5 were most dissimilar and 11 and 12 the most similar. The ISSR markers used in this study demonstrated the efficiency of molecular polymorphisms detection, revealing high genetic variability among the 16 accessions. So, it can be inferred that there is a considerable genetic variation among accessions of the biribazeiro, showing the importance of molecular markers in the analysis of variability of species poorly studied, as Rollinia mucosa [Jacq.]Baill.
