O objetivo deste trablaho foi caracterizar geneticamente o bovino Pantaneiro por meio de sua ancestralidade paterna, pela morfologia de seu cromossomo Y, submetacêntrico ou acrocêntrico, e identificar a ancestralidade materna pelo DNA mitocondrial. O cariótipo e o DNA mitocondrial de 12 touros da raça Pantaneira foram cariotipados e analisados. O cromossomo Y foi analisado por meio das metáfases de linfócitos e o DNA mitocondrial pelo diagnóstico de seu tipo de haplótipo (Bos taurus ou Bos indicus). Entre os Pantaneiros analisados, três apresentaram Y de taurino (submetacêntrico) e nove de zebuíno (acrocêntrico), o que sugere contaminação racial por zebuínos, visto que o Pantaneiro é considerado de origem Européia. O DNA mitocondrial foi exclusivamente de taurinos, o que indica que a participação de zebuínos na formação da raça foi inteiramente de origem paterna.

The objective of this work was to characterize Pantaneiro cattle genetically through its paternal ancestry by the morphology of the Y chromosome, whether submetacentric or acrocentric, as well as to identify the maternal ancestry through mitochondrial DNA. The karyotype and mitochondrial DNA of 12 bulls of Pantaneiro breed were analyzed. The Y chromosome was analyzed in lymphocyte metaphases and the mitochondrial DNA by diagnosing its haplotype (Bos taurus and Bos indicus). Among Pantaneiro animals analyzed three had a taurine (submetacentric) Y and nine had a zebuine (acrocentric) Y chromosome, suggesting breed contamination by Zebu cattle, once Pantaneiro is considered to be of European origin. The mitochondrial DNA was exclusively of taurine origin, indicating that the participation of zebuines in the formation of the breed occurred entirely through the paternal line.

Anteaters belong to the Order Xenarthra / Family Myrmecophagidae and are the only members without teeth. There are three genera with four living species in the family Myrmecophagidae: Myrmecophaga tridactyla (giant anteater), Tamandua tetradactyla (southern lesser anteater), Tamandua mexicana (northern lesser anteater), and Cyclopes didactylus (silky anteater). The karyotypes of M. tridactyla (2n = 60), T. tetradactyla (2n = 54) and C. didactylus (2n = 64) have already been described. In the present paper, three female and two male specimens of giant anteater and one lesser anteater male were analyzed. The results indicate the existence of a new karyotype in the genus Tamandua, with 2n = 56 chromosomes, which can represent a new lesser anteater species. The karyotype of M. tridactyla was also described, supporting previous reports.

Foram analisados os cromossomos de 117 bovinos de diferentes raças para identificação de fusão cêntrica e os cromossomos de 100 éguas jovens da raça Brasileiro de Hipismo para identificação de linhagens 63,X, utilizando a técnica de identificação do X baseada na heterocromatina intersticial do braço longo.

We analyzed the chromosomes of 117 cows of different breeds for the identification of centric fusion and the chromosomes of 100 young Andaluz and Brasileiro de Hipismo mares for the identification of 63,X lines, using the technique of X chromosome identification based on interstitial heterochromatin on the long arm.

Foram cariotipados 95 touros puros de origem, da raça Chianina, ditribuidos em 19 empresas pastoris, em 5 estados brasileiros. O objetivo foi investigar a incidência de indivíduos portadores de cromossomo Y acrocêntrico, típico das raças de Bos taurus indicus, face às especulações de que as raças indianas poderiam ter contribuido para a formação do Chianina. Todos os indivíduos avaliados mostraram o cromossomo Y de Bos taurrus taurus. O índice centromérico obtido foi de 43,91%, o que permitiu classificar o centrômero deste cromossomo como localizado na região mediana. Foram avaliados também 29 touros com o objetivo de verificar a presença do polimorfismo intraracial do cromossomo Y. O índice centromérico e o tamanho relativo do Y foi determinado. O tamanho do cromossomo X serviu como base para estimar o tamano relativo do Y. A análise de variância mostrou diferenças entre touros apenas no tamanho relativo do Y, sendo que o índice centromérico não difereiu entre os mesmos. Concluimos que este polimorfismo indica que a raça Chianina pode ter recebido contribuição de outras raças em passado remoto ou pode também indicar a possibilidade de cruzamentos mais recentes.

There were studied the karyotypes of 95 purebred Chianina bulls, distributed among 19 cattle farms in five Brazilian states. The objective was to investigate the incidence of individuals carring an acrocentric Y chromosome, typical of Bos taurus indicus breeds, in view of speculations that Indian breeds might have contributed to the formation of Chianina. All individual showed the Bos taurus taurus Y chromosome. The centromeric index obtained was 43.91%, which allowed us to classify the centromere of this chromosome as medially located. There were also studied 29 purebred Chianina bulls with the objective of verifying the presence of intraracial polymorphism at the Y chromosome. The centromeric index and relative size of the Y chromosome were determined. The lenght of the X chromosome served as a basis for estimating the relative size of Y. Analysis of variance showed differences among bulls only in the relative size of Y, no differences being detected in the centromeric index. We conclude that this polymorphism indicates the Chianina breed may have received contributions from other breeds in the remote past, or it may also indicate the possibility of more recent crosses.

O presente estudo teve como objetivo desenvolver uma técnica eficiente de estocagem de leucócitos de cervídeos e ovinos para posterior cultivo e obtenção de preparações citogenéticas. Vários meios de cultura, crioprotetores e protocolos de congelação e descongelação foram utilizados. Foram obtidos os melhores resultados utilizando-se meio de cultura McCoy ou VYM, acrescidos de DMSO, soro equino e PVP como crioprotetores. O melhor protocolo de congelação foi submeter as células a 30 minutos em 4°C e 15 minutos em vapor de nitrogênio líquido, e para descongelação, o lançamento do material ainda congelado no meio de cultivo para propagação.

Several culture, cryopreservation, freezing and thawing methods were tested to develop an efficient technique for storing cervid and ovine leukocytes. The best results were obtained with McCoy or Vega y Martinez (VYM) solution with dimethyl sulfoxide (DMSO), horse serum and polyvinylpyrrolidone (PVP) as cryopreservatives. The best protocol for freezing was 4°C for 30 min followed by 15 min in liquid nitrogen vapor. To thaw, the still frozen material was placed onto cultivation medium for propagation.
