RESUMO Neste estudo, os efeitos tóxicos da melitina em células Madin-Darby Bovine Kidney (MDBK) foram analisados quanto à funcionalidade mitocondrial, por redução de MTT e citometria de fluxo, potencial de apoptose, necrose, produção de espécies reativas de oxigênio (ROS), peroxidação lipídica e fragmentação de DNA, utilizando-se citometria de fluxo e desestabilização da membrana celular, por microscopia confocal. A toxicidade apresentou características dose-dependentes e a atividade mitocondrial foi inibida até 78,24±3,59% (P<0,01, n = 6) em células MDBK expostas à melitina (10μg/mL). Análises por citometria de fluxo revelaram que a melitina a 2μg/mL apresentou o maior índice necrótico celular (P<0,05). A maior lipoperoxidação de membranas também foi na concentração de 2μg/mL de melitina (P<0,05), o que foi posteriormente confirmado por microscopia confocal. A maior produção de ROS aconteceu quando as células foram expostas a 2,5μg/mL de melitina (P<0,05), e essa concentração também aumentou a fragmentação de DNA (P<0,05). Houve uma significativa correlação positiva entre a lipoperoxidação de membranas e a produção de ROS (R=0,4158), funcionalidade mitocondrial (R=0,4149) e apoptose (R=0,4978). Portanto, o estresse oxidativo gerado pela melitina culminou na elevação de ROS intracelular, que inicia uma cascata de eventos tóxicos nas células MDBK.

ABSTRACT In this study, the toxic effects of melittin on Madin-Darby Bovine Kidney cells (MDBK) were analyzed with respect to mitochondrial functionality by reduction of MTT and flow cytometry, apoptosis potential, necrosis, oxygen reactive species (ROS) production, lipid peroxidation, and DNA fragmentation using flow cytometry and cell membrane destabilization by confocal microscopy. The toxicity presented dose-dependent characteristics and mitochondrial activity was inhibited by up to 78.24 ±3.59% (P<0.01, n = 6) in MDBK cells exposed to melittin (10μg/mL). Flow cytometry analysis revealed that melittin at 2μg/mL had the highest necrosis rate (P<0.05) for the cells. The lipoperoxidation of the membranes was also higher at 2μg/mL of melittin (P<0.05), which was further confirmed by the microphotographs obtained by confocal microscopy. The highest ROS production occurred when the cells were exposed to 2.5μg/mL melittin (P<0.05), and this concentration also increased DNA fragmentation (P<0.05). There was a significative and positive correlation between the lipoperoxidation of membranes with ROS (R=0.4158), mitochondrial functionality (R=0.4149), and apoptosis (R=0.4978). Thus, the oxidative stress generated by melittin culminates in the elevation of intracellular ROS that initiates a cascade of toxic events in MDBK cells.

RESUMO Das células do sistema imunológico, macrófagos desempenham um papel fundamental. Apamina, constituinte do veneno de abelhas, é importante na defesa destas. Objetivou-se avaliar o metabolismo da linhagem de macrófagos J774 1.6 expostos à apamina isolada e purificada, avaliando-se citotoxicidade por redução de MTT e análise por citometria de fluxo (apoptose / necrose, produção de espécies reativas de oxigênio (EROs), lipoperoxidação membranosa (LPO), potencial elétrico da membrana mitocondrial (MMP) e fragmentação do DNA). Nenhuma concentração testada (10 a 100μg / mL) foi citotóxica. As taxas de apoptose diminuíram nas concentrações 2,5, 5,0 e 10,0μg / mL (P<0,05), enquanto as de necrose aumentaram (P<0,05). Entretanto, as taxas de células saudáveis na maior concentração testada (10μg / mL) não diferiram do controle (P>0,05). A apamina não alterou as ERO, a LPO nem a fragmentação do DNA. Portanto, todas as concentrações analisadas (1,25 a 10μg / mL) diminuíram a mMP. Tal diminuição na apoptose pode ser por uma supressão de mensageiros pró-apoptóticos mitocondriais, já que este peptídeo não causa estresse oxidativo, peroxidação lipídica nem dano ao DNA. Técnicas altamente sensíveis são importantes para adequada interpretação dos mecanismos de citotoxicidade.

ABSTRACT Among the immune system cells, macrophages have an important role. Apamin, a bee venom constituent, is important in the defense of these insects. Thus, we aimed to evaluate the metabolism of J774 1.6 macrophage cell line when exposed to isolated and purified apamin, using cytotoxicity tests by MTT reduction and analysis by flow cytometry (apoptosis / necrosis, production of reactive oxygen species (ROS), membranous lipoperoxidation (LPO), electrical potential of the mitochondrial membrane (mMP) and DNA fragmentation). None of the tested concentrations (10 to 100μg/mL) were cytotoxic according to MTT reductions. Apoptosis rates decreased at concentrations of 2.5, 5.0, and 10.0μg/mL (P<0.05), while necrosis rates increased (P<0.05). However, rates of healthy cells at the highest tested concentration (10μg/mL) did not differ from control (P>0.05). Apamin did not alter ROS, LPO, or DNA fragmentation. Therefore, all analyzed concentrations (1.25 to 10μg/mL) decreased mMP. Such decrease in apoptosis might be due to a suppression of mitochondrial pro-apoptotic messengers, as this peptide causes no oxidative stress, lipid peroxidation, and DNA damage. Highly sensitive techniques are majorly important for proper interpretation of cellular toxicity mechanisms, combined with routine laboratory methods.

RESUMO Apesar dos bovinos serem considerados os hospedeiros naturais do BoHV-1, estudos sorológicos têm sugerido que búfalos podem ser suscetíveis ao BoHV-1 e a outros alfa-herpesvírus geneticamente relacionados. O objetivo deste estudo foi detectar a presença de DNA viral de BoHV-1 em 202 amostras de gânglios trigêmeos de búfalos, pela técnica de semi-nested PCR, para detecção de um segmento do gene codificante da glicoproteína D (gD) do BoHV-1. Além disso, 242 amostras de soro foram analisadas pela técnica de soroneutralização (SN) para a detecção de anticorpos neutralizantes contra BoHV-1, BoHV-5 e BuHV. Todas as amostras clínicas foram coletadas em um matadouro na cidade de Pelotas, RS, Brasil. O DNA de BoHV-1 foi detectado em 61 (30,1%) gânglios, e os resultados da SN demonstraram que 27,6% dos animais apresentaram anticorpos contra, pelo menos, um dos vírus testados. O sequenciamento genômico e a análise de 14 amplicons confirmaram a presença do DNA do BoHV-1 nos tecidos analisados. Em resumo, os resultados indicam que o BoHV-1 está distribuído em rebanhos bubalinos provenientes da região Sul do Brasil. Entretanto, são necessárias investigações adicionais, no sentido de elucidar o papel exato dos búfalos na epidemiologia das infecções pelo BoHV-1.

ABSTRACT Although bovines are natural hosts for BoHV-1, serologic studies in several countries have suggested that buffaloes (Bubalus bubalis) may be susceptible to BoHV-1 and other genetically related alphaherpesvirus. This study aimed to investigate the presence of BoHV-1 DNA in trigeminal ganglia from 202 buffaloes by a semi-nested PCR to amplify partially the glycoprotein D (gD) gene of BoHV-1. Additionally, 242 serum samples were tested by serum neutralization (SN) for the detection of antibodies against BoHV-1, BoHV-5 and BuHV. All clinical samples were collected in a slaughterhouse located in Pelotas, RS, Brazil. BoHV-1 DNA was detected in 61 (30.1%) of the samples and SN revealed 27.6% of the animals with neutralizing antibodies against at least one of the tested viruses. Nucleotide sequencing of 15 amplicons followed by BLAST analysis confirmed the presence of BoHV-1 DNA in the analyzed tissues. Taken together, these data indicate that BoHV-1 infection is distributed in buffaloes in southern Brazil. However, the role of buffaloes in the BoHV-1 epidemiology needs further investigation.

RESUMO Neste estudo, descreveu-se a atividade antifúngica de três extratos de própolis brasileiras: marrom, verde e de abelhas jataí (Tetragonisca angustula), contra Sporothrix brasiliensis. Os extratos foram obtidos de extração etanólica, e a sua composição química foi determinada por cromatografia líquida de alta eficiência, acoplada à espectrometria de massa. A toxicidade celular foi medida em células MDBK (Madin-Darby Bovine Kidney), avaliada por observação microscópica e quantificada pelo ensaio MTT (3- (4,5-dimetiltiazol-2-ilo -2,5-difenil-2H bromo tetrazolato). Para a atividade antifúngica, determinou-se a concentração inibitória mínima (CIM) e a concentração fungicida mínima (CFM) por meio de microdiluição em caldo. Os resultados mostraram que a toxicidade celular não foi observada em concentrações menores (0,097 a 0,39μg/ml). Entre os compostos químicos identificados, foram quantificados o ácido cafeico, ácido p-cumárico, ácido clorogênico, ácido ferúlico e a rutina. Na atividade antifúngica, as própolis verde e jataí não apresentaram atividade contra os isolados (CIM e CFM maior que 0,78mg/ml), porém todos os isolados de S. brasiliensis foram sensíveis à própolis marrom (CIM de 0,09 a 0,78mg/ml), incluindo a cepa padrão (P<0,001). Entre as própolis brasileiras estudadas, a marrom mostrou atividade contra S. brasiliensis, e mais estudos devem ser realizados para avaliar seu uso promissor no tratamento da esporotricose.

ABSTRACT In this study, we described the antifungal activity of three Brazilian propolis extracts: brown, green and from jataí bees against Sporothrix brasiliensis. The extracts were obtained from ethanolic extraction and their chemical composition was determined by high-performance liquid chromatography coupled to mass spectrometry. The cellular toxicity was measured in MDBK (Madin-Darby Bovine Kidney) cells and quantified by the MTT assay (3- (4,5 dimethylthiazol-2yl -2,5-diphenyl-2H bromine tetrazolato). For antifungal activity, the minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) were determined by broth microdilution. The results showed that cell toxicity was not observed at lower concentrations (0.097 to 0.39μg/ml) for all extracts in comparison to cell control. Among the chemical compounds identified, caffeic acid, p-coumaric acid, chlorogenic acid, ferulic acid and rutin were quantified. In antifungal activity, green and jataí did not exhibit activity against the isolates (MIC and MFC greater than 0.78mg/ml). However, all isolates of S. brasiliensis were sensitive to brown propolis (MIC of 0.09 to 0.78mg/ml), including the standard strain (P<0.001). Among the Brazilian propolis studied, the brown propolis showed activity against the S. brasiliensis isolates and more studies should be undertaken in order to evaluate its promising use in the treatment of sporotrichosis.

RESUMO Esforços significativos têm sido realizados para o desenvolvimento de vacinas mais eficientes em saúde animal. Uma estratégia que pode ser usado para melhorar a eficácia da vacina é o uso de probióticos para melhorar a resposta imune do hospedeiro, conduzindo ao aumento da imunogenicidade de preparações de antígenos. Herpesvírus bovino 5 (BoHV-5) é um exemplo de um importante patógeno animal para os quais vacinas têm fornecido apenas uma protecção limitada. Neste estudo, examinou-se o uso do probiótico Saccharomyces boulardii (Sb) como um adjuvante potencial para melhorar a eficiência da vacina. Verificou-se que os animais suplementados apresentaram uma produção de anticorpos IgG superior e com desvio para Th1 em favor de IgG2, além do aumento dos níveis de expressão de mRNA para as citocinas IFN-γ, IL-12, IL-17 e IL-10. Esses resultados sugerem que a suplementação de Sb pode fornecer um meio promissor para melhorar a eficiência de vacinas, particularmente aquelas que dependem de uma resposta imune mediada por células.

ABSTRACT There have been significant efforts towards the development of more efficient vaccines for animal health. A strategy that may be used to improve vaccine efficacy is the use of probiotics to enhance the immune response of the host, leading to increased immunogenicity of antigen preparations. Bovine herpesvirus 5 (BoHV-5) is an example of an important animal pathogen for which vaccines have provided only limited protection. In this study, we examined the use of the probiotic Saccharomyces boulardii (Sb) as a potential adjuvant to improve vaccine efficiency. We found that the supplemented animals exhibited an enhanced systemic IgG antibody response toward a Th1 response in favor of IgG2a and increased mRNA expression levels of the cytokines IFN-y, IL-12, IL-17 and IL-10 in the spleen. These results suggest that Sb supplementation may provide a promising means for improving the efficiency of vaccines, particularly those that rely on a cell-mediated immune response.

ABSTRACT The pepper tree (Schinus molle L.) has secondary metabolites with insecticidal activity. The objective of this study was to assess under laboratory conditions the insecticidal and insectistatic activity of fruit powder and essential oil of S. molle against Sitophilus zeamais. In contact toxicity bioassays, the highest mortality was observed with essential oil at a concentration of 8.0%, recording 96.3% of dead insects with an LC50 of 3.82 mL essential oil 100 g-1 maize (3.82%). Powder at the same concentration showed a 63.8 % of mortality with an LC50 of 7.95 g powder 100 g-1 maize (7.95%). None of the treatments affected germination or grain weight loss. The essential oil at a concentration of 8.0% caused a complete inhibition of F1 resulting in 0% of adult insects. All treatments had repellent effects. The highest antifeeding effect observed with the essential oil was recorded at a concentration of 16.0% with a 72.2% of inhibition. It was concluded that both powder and essential oil of S. molle show contact insecticidal toxicity as well as repellent and antifeeding effects against S. zeamais.

RESUMEN El pimiento boliviano (Schinus molle L.) es un árbol del cual se han identificado metabolitos secundarios con actividad insecticida. El objetivo fue evaluar, en condiciones de laboratorio, la actividad insecticida e insectistática del polvo y aceite esencial de frutos de S. molle sobre Sitophilus zeamais. En los bioensayos de toxicidad por contacto la mayor mortalidad se obtuvo con el aceite esencial al 8,0 % con 96,3% de insectos muertos con una CL50 de 3,82 mL aceite esencial 100 g-1 maíz (3,82%). A la misma concentración el polvo mostró un 63,8% de mortalidad y una CL50 de 7,95 g polvo 100 g-1 maíz (7,95%). Ambos tratamientos no afectaron significativamente la germinación ni la pérdida de peso del grano. El aceite esencial en la concentración de 8,0% provocó una completa inhibición en la emergencia mostrando una F1 de 0% de insectos adultos. Todos los tratamientos fueron repelentes y el mayor efecto antialimentario del aceite esencial se registró con la concentración de 16% con 71,2% de inhibición. Se concluyó que el polvo y aceite esencial de S. molle presentan toxicidad insecticida por contacto y efecto repelente e inhibidor de la alimentación sobre S. zeamais.

RESUMO P34 é um peptídeo antimicrobiano produzido pelo Bacillus sp. P34, isolado do conteúdo intestinal de um peixe na Bacia Amazônica. Esse peptídeo demonstrou propriedades antibacterianas contra bactérias Gram-positivas e Gram-negativas e foi caracterizado como uma substância do tipo bacteriocina. Foi demonstrado que o peptídeo P34 exibiu atividade antiviral contra o herpesvírus felino tipo 1 in vitro. O objetivo deste trabalho foi avaliar o P34 in vitro, em cultivo de células RK 13, contra o vírus da arterite equina, uma vez que não há tratamento específico e uma variável proporção de garanhões pode permanecer persistentemente infectada. Os resultados obtidos mostram que o peptídeo exerce atividade antiviral e virucida contra o vírus da arterite equina, agindo provavelmente no envelope viral. Os ensaios antivirais realizados demonstraram que o P34 reduz significativamente os títulos do vírus nas células infectadas e tratadas. O mecanismo de ação do P34 parece ser tempo/temperatura dependente. Esse peptídeo tende a ser um antiviral promissor para o tratamento e a prevenção das infecções por arterivírus, tendo em vista que ele possui um alto índice terapêutico. Contudo, estudos mais detalhados devem ser realizados para precisar a etapa exata da infecção viral em que o P34 age, para que ele possa ser usado como antiviral in vivo no futuro.

ABSTRACT P34 is an antimicrobial peptide produced by Bacillus sp. P34, isolated from the intestinal contents of a fish from the Amazon basin. This peptide showed antibacterial properties against Gram-positive and Gram-negative bacteria and was characterized as a bacteriocin like substance. It was demonstrated that the peptide P34 exhibited antiviral activity against feline herpesvirus type 1 in vitro. The aim of this work was to evaluate P34 for its antiviral properties in vitro, using RK 13 cells, against the equine arteritis virus, since it has no specific treatment and a variable proportion of stallions may become persistently infected. The results obtained show that P34 exerts antiviral and virucidal activities against equine arteritis virus, probably in the viral envelope. The antiviral assays performed showed that P34 reduces significantly the viral titers of treated cell cultures. The mechanism of action of P34 seems to be time/temperature-dependent. This peptide tends to be a promising antiviral compound for the prevention and treatment of arteriviral infections since it has a high therapeutic index. However, more detailed studies must be performed to address the exact step of viral infection where P34 acts, in order to use this peptide as an antiviral drug in vivo in the future.

El crecimiento del fruto de la feijoa es principalmente estimulado por la temperatura, pero también es afectado por otros factores climáticos como la altitud. El objetivo de este estudio es proponer un modelo de crecimiento del fruto de la feijoa en función del tiempo térmico (GDC, grados día de crecimiento) y de la altitud (H) de la zona de producción, para lo cual se marcaron veinte árboles por finca en dos localidades del departamento de Cundinamarca (Colombia), durante los años 2012 a 2014. Las mediciones se realizaron cada 7 días a partir del día 96 y 99 después de antesis hasta la cosecha, para las localidades de Tenjo (2580 m.s.n.m.) y San Francisco de Sales (1800 m.s.n.m.) respectivamente. Se obtuvo un modelo de crecimiento para el peso en función de la longitud y el diámetro del fruto, así como para el peso del fruto en función de GDC y H, ajustándose esta última a un modelo de crecimiento sigmoidal logístico. Los parámetros del análisis de regresión mostraron que los modelos predijeron satisfactoriamente el crecimiento del fruto para las dos localidades, con alto coeficiente de determinación. La validación cruzada mostró buen ajuste estadístico entre valores pronosticados y observados; la intercepción no fue significativamente diferente de cero y la pendiente fue estadísticamente igual a uno.

The growth of the pineapple guava fruit is primarily stimulated by temperature but is also influenced by other climactic factors, such as altitude. The goal of this study was to develop a growth model for the pineapple guava fruit as a function of thermal time (GDD, growing-degree day) and altitude (H) of the production area. Twenty trees per farm were marked in two sites in the Cundinamarca region (Colombia) during the 2012 and 2014 seasons. Measurements were performed every seven days after day 96 and 99 post-anthesis until harvest in the sites of Tenjo (2580 m.a.s.l.) and San Francisco de Sales (1800 m.a.s.l.), respectively. A growth model was produced for weight as a function of fruit length and diameter as well as for the weight of the fruit as a function of GDD and H, with this last measure adjusted to a sigmoidal logistic growth model. The parameters for the regression analysis showed that the models satisfactorily predicted fruit growth for both of the sites, with a high determination coefficient. The cross-validation showed good statistical fit between the predicted and observed models; the intercept was not significantly different than zero, and the slope was statistically equal to one.

No Brasil existem poucos estudos sobre a ocorrência da infecção pelo vírus da imunodeficiência felina (FIV), assim como a determinação dos subtipos circulantes, o que é indispensável para o desenvolvimento de vacinas e novos testes diagnósticos. O presente trabalho investigou a ocorrência da infecção pelo FIV entre os anos de 2010 e 2011 em gatos domésticos submetidos a atendimento clínico na cidade de Pelotas e região. Amostras de sangue total de 70 animais, incluindo suspeitos (28) ou não suspeitos (42) da infecção pelo FIV, foram submetidas à reação de PCR nested. Os resultados indicaram uma frequência de infecção de 15,7% (11/70) e a análise dos fatores associados (sexo, idade e condição clínica) evidenciou uma maior ocorrência em gatos com idade superior a 10 anos e acometidos por infecções crônicas e recidivantes. Oito amostras positivas na PCR nested foram submetidas a sequenciamento genômico e somente o subtipo B foi detectado na região estudada.

In Brazil there are few studies on the occurrence of the feline immunodeficiency virus (FIV) infection and its subtypes, which are essential for the development of vaccines and new diagnostic tests. The present study investigated the occurrence of the FIV infection between 2010 and 2011 in domestic cats submitted to medical attendance in the city of Pelotas and nearby area. Total blood samples of seventy cats, suspected (28) or not (42) of infection by FIV were analyzed by nested PCR in order to perform a diagnosis. The results pointed to a FIV infection frequency of 15.7% (11/70) and the analysis of the risk factors related to infection (sex, age and clinical condition) evidenced a greater occurrence in cats up to 10 years of age with chronic and recurrent infections. Eight samples found positive by nested PCR were submitted to DNA sequencing indicating that only the subtype B was detected in the studied region.

A mastite bovina é uma doença importante na exploração leiteira, não apenas pelas perdas econômicas diretas que promove, mas também pelas perdas indiretas e o potencial risco à saúde pública. Dentre as principais causas de infecções intramamárias, destacam-se as bactérias do gênero Staphylococcus spp., sendo que Staphylococcus aureus é o agente etiológico predominante em mastite subclínica. O objetivo desse trabalho foi verificar a frequência de mastite subclínica em oito rebanhos localizados na região Sul do Rio Grande do Sul (Brasil) e a relação da enfermidade com a presença de S. aureus. Adicionalmente, pesquisou-se a presença de S. intermedius e S. hyicus nas amostras de leite obtidas. Para identificação da doença, utilizou-se o California Mastitis Test (CMT). A identificação da espécie de Staphylococcus spp. foi feita em meio de cultura ágar Baird-Parker, com posterior confirmação das colônias suspeitas em coloração de gram, prova de catalase, pesquisa de coagulase livre e pesquisa de termonuclease. A mastite subclínica foi constatada em 53,6% dos animais testados. A presença de Staphylococcus coagulase positiva foi identificada em 12,6% dos animais com mastite subclínica. Nesses mesmos animais, a bactéria identificada como S. aureus foi o agente etiológico presente em 17,6% dos casos. Adicionalmente, pode-se perceber que, dentre o grupo identificado como coagulase positiva, 85,7% corresponderam a S. aureus, enquanto 8,5% mostraram características bioquímicas compatíveis com S. intermedius e 5,8% foram consideradas S. hyicus.

Bovine mastitis is an important disease in dairy farming, not only by promoting direct economic losses, but also for indirect losses and the potential risk to public health. The main causes of intramammary infections include the bacteria of the genus Staphylococcus spp., Staphylococcus aureus being the predominant etiologic agent in subclinical mastitis. The aim of this study was to determine the frequency of subclinical mastitis in eight herds from southern Rio Grande do Sul (Brazil) and the relationship of the disease with the presence of S. aureus. In addition, we checked for the presence of S. intermedius and S. hyicus in the milk samples obtained. For identification of the disease, we used the California Mastitis Test (CMT). Identification of Staphylococcus spp. species was made in Barid-Parker agar culture medium, with subsequent confirmation of suspected colonies by way of Gram stain and catalase test along with free-coagulase and thermonuclease research. Subclinical mastitis was identified in 53.6% of animals tested. The presence of coagulase-positive Staphylococcus was identified in 12.6% of animals with subclinical mastitis. In these same animals, bacteria identified as S. aureus were the etiologic agent present in 17.6% of cases. Additionally, it was revealed that among the group identified as coagulase positive, 85.7% corresponded to S. aureus, while 8.5% had biochemical characteristics consistent with S. intermedius and 5.8% were considered S. hyicus.

Durante o processo de resfriamento dos ovos embrionados, há um fluxo natural de ar da superfície para o interior dos ovos carreando contaminantes como bactérias e fungos, por meio dos poros da casca, infectando o embrião e resultando na inabilidade para eclodir e pintinhos de má qualidade. O formaldeído que é um produto tóxico ainda é o desinfetante mais utilizado para a desinfecção de ovos embrionados pela indústria avícola. Para avaliar a atividade antimicrobiana do extrato etanólico da própolis verde, como alternativa ao formaldeído, foram coletados 140 ovos de ninhos de matrizes de frango de corte submetidos à desinfecção com cinco tratamentos: T1 - sem desinfecção; T2 - ovos fumigados com formaldeído; T3, T4 e T5 desinfetados por imersão com solução de própolis nas concentrações de 2.400 µg, 240 µg e 24 µg, respectivamente. Os níveis de contaminação da casca dos ovos por mesófilos totais e fungos (Aspergillus e outros bolores), após a desinfecção com própolis, foram menores quando comparados ao controle. Na comparação ao tratamento com formaldeído as concentrações de própolis com 240 µg e 24 µg não diferiram para atividade antibacteriana, já para atividade antifúngica, 2,4 mg e 240 µg foram superiores. Com relação à eclodibilidade dos ovos, após 21 dias de incubação, os tratamentos de própolis (2,4 mg e 240 µg) apresentaram as maiores taxas com 94,11% superando o tratamento com formaldeído. Portanto, o extrato etanólico de própolis verde apresenta atividade antibacteriana e antifúngica em ovos embrionados podendo ser um novo produto natural desinfetante em substituição ao formaldeído.

During the cooling process of embryonated eggs, there is a natural air flux from the surface to the inner part of the eggs, carrying contaminants such as bacteria and fungi through the shell's pores, infecting embryos and resulting in the inability to hatch or poor chick quality. Formaldehyde, a toxic product, is still the most used disinfectant for embryonated eggs in the aviculture industry. In order to evaluate the antimicrobial activity of the green propolis ethanolic extract as an alternative to formaldehyde, 140 hatching eggs from laying hens were collected and submitted to disinfection with five different treatments: T1 - without disinfection; T2 - formaldehyde fumigated eggs; T3, T4 and T5 disinfection by immersion in propolis solution in the concentrations of 2,400 µg, 240 µg and 24 µg, respectively. The contamination levels by total mesophiles and fungi of the egg shells (Aspergillus sp. and other moulds) after disinfection with propolis were lower than when compared to the control without disinfection. In comparison with formaldehyde, the 240 µg and 24 µg propolis concentrations did not differ regarding antibacterial activity, but for antifungal activity the 2,400 µg and 240 µg concentrations were more efficient. The 2,400 µg and 240 µg propolis treatments presented a hatching rate of 94.1%, compared to only 84.6% for the formaldehyde treatment. The green propolis ethanolic extract presented antibacterial and antifungal activities in embryonated eggs showing that it can be a new natural disinfectant product substituting formaldehyde.

In stored grains, the main agents diminishing production are insects, which can produce losses between 20% and 80% before harvest or under storage. The insecticidal properties of the essential oil of fresh leaves of Peumus boldus Molina against maize weevil (Sitophilus zeamais Motschulsky) adults were determined under laboratory conditions. The highest mortality (100%) was achieved at 4% concentration by contact with a treated glass surface. The same concentration in impregnated corn (Zea mays L.) grain, resulted in 98.7% mortality. Mortality by fumigant action at 6 h was 100% with 35 µL oil in 0.15 L (air volume). Concentrations 1, 2 and 4% of essential oil produced 0% F1 adult emergence. At 10 d of residual effect, the 4% concentration reached 63.7% mortality. All treatments were repellent to adults of S. zeamais and corn grain germination was not affected by any treatment.

Los principales agentes que disminuyen la producción en los granos almacenados son los insectos, antes de la cosecha y en el almacenamiento pueden causar pérdidas de 20 a 80%. Se evaluaron las propiedades insecticidas del aceite esencial de hojas frescas de Peumus boldus Molina para el control de adultos de gorgojo del maíz (Sitophilus zeamais Motschulsky) en laboratorio. La mayor mortalidad (100%) por contacto con una superficie de vidrio tratada se obtuvo con la concentración de 4%. Esta misma concentración produjo 98,7% de mortalidad en exposición a grano de maíz (Zea mays L.) tratado. El efecto fumigante a las 6 h de exposición fue 100% con 35 µL de aceite en 0,15 L (volumen de aire). Con las concentraciones de 1, 2 y 4% de aceite esencial, el porcentaje de emergencia de la F1 fue 0%. A los 10 d de efecto residual se alcanzó 63,7% de mortalidad con la concentración de 4%. Todos los tratamientos fueron repelentes para adultos de S. zeamais y ningún tratamiento afectó la germinación de los granos.

RESUMO A própolis, uma substância resinosa produzida pelas abelhas melíferas a partir de exsudatos coletados em diferentes partes das plantas, tem sido utilizada há séculos na medicina popular devido as suas propriedades terapêuticas. Esta substância apresenta atividade antiinflamatória, antitumoral, antioxidante e imunomoduladora, embora muitos dos seus mecanismos de ação sejam desconhecidos. A imunomodulação pela própolis pode estar associada tanto com a estimulação quanto com a supressão de determinados eventos da resposta imune. Os efeitos antagônicos e muitas vezes não complementares sobre o sistema imunológico podem ser decorrentes da grande variabilidade química existente entre diferentes amostras de própolis, bem como das diferentes metodologias adotadas nos estudos científicos, resultando em variada ação farmacológica. Este artigo teve como objetivo revisar e discutir alguns aspectos relacionados à ação da própolis sobre o sistema imunológico.

ABSTRACT Propolis is a resinous substance produced by honeybees from exudates collected from different parts of the plants. It has been used for centuries in popular medicine due to its therapeutic characteristics. This substance presents antiinflammatory, anti-tumoral, anti-oxidant and immunomodulator activities, even though many of its mechanisms of action are still unknown. Immunomodulation induced by propolis can be associated with stimulation as well as suppression of certain events of the immune response. The antagonic and sometimes noncomplementary effects on the immunological system can be attributed to the wide chemical variability present among different propolis samples, as well as to the different methodologies used in scientific studies, resulting in a varied pharmacological action. This work aims at reviewing and discussing some aspects related to the mechanism of action of propolis on the immunological system.

Antibody titres to canine distemper virus (CDV) and canine parvovirus (CPV) were measured in 132 dogs: 80 had been vaccinated at least once, 22 had not been vaccinated, and 30 had unknown vaccination history. Serum antibody titers were measured by means of serum neutralization (CDV) or hemagglutination inhibition (CPV). Serum CDV titers >20 and serum CPV titers >80 were considered protective. Protective antibodies to CDV were present in 40.1% of the population: 39.8% of the vaccinated dogs, 31.8% unvaccinated, and in 46.6% of the dogs with unknown vaccination history. Protective antibodies to CPV were present in 90.9% of the dogs: 93.7% of the vaccinated dogs, 90.9% of the unvaccinated, and 83.3% of the dogs with unknown vaccination history.
