Este trabalho foi desenvolvido com o objetivo de realizar triagem primária para caracterização da tolerância de variedades SP e CTC de cana-de-açúcar à aplicação de dez herbicidas comumente recomendados para manejo de plantas daninhas nesta cultura. Dois experimentos foram desenvolvidos com esquema fatorial entre variedades de cana-de-açúcar e onze tratamentos herbicidas. No primeiro experimento utilizaram-se sete variedades SP (fatorial 7 x 11): SP80-1842, SP80-3280, SP83-2847, SP87-344, SP87-396, SP89-1115 e SP90-3414. No segundo experimento, foram avaliadas seis variedades CTC (fatorial 6 x 11): CTC1, CTC2, CTC3, CTC4, CTC5 e CTC6. Os tratamentos herbicidas aplicados em pós-emergência inicial foram: ametryn, ametryn + trifloxysulfuron-sodium, clomazone, diuron + hexazinone, isoxaflutole, imazapic, 2,4-D, tebuthiuron, sulfentrazone, MSMA e testemunha sem aplicação. Avaliou-se massa fresca, altura, teor de clorofila (índice SPAD) e perfilhamento, todos em termos percentuais relativos à testemunha sem aplicação. Constatou-se tolerância diferencial das variedades de cana-de-açúcar aos herbicidas comumente utilizados na cultura. As variedades SP80-3280 e CTC2 foram as mais tolerantes aos tratamentos herbicidas, podendo ter esta característica explorada em programas de melhoramento genético.

This work was carried out with the objective of screening for tolerance response of SP and CTC sugarcane varieties to application of ten herbicides commonly recommended to weed management in this crop. Two trials were evaluated with factorial design between the sugarcane varieties and eleven herbicide treatments. In the first experiment, seven SP varieties were used (factorial 7 x 11): SP80-1842, SP80-3280, SP83-2847, SP87-344, SP87-396, SP89-1115 and SP90-3414. In the second experiment, six CTC varieties were evaluated (factorial 6 x 11): CTC1, CTC2, CTC3, CTC4, CTC5 and CTC6. The post-emergence-applied herbicide treatments were: ametryn, ametryn + trifloxysulfuron-sodium, clomazone, diuron + hexazinone, isoxaflutole, imazapic, 2,4-D, tebuthiuron, sulfentrazone, MSMA and check without application. The variables evaluated were: fresh mass, height, SPAD index and tillering: Valves here considered as relative percentage to the untreated control. Differential tolerance of sugarcane varieties was observed regarding to application of the tested herbicides. SP80-3280 and CTC2 were the most tolerant varieties to herbicide application, and might have this characteristic exploited by sugarcane breeding programs.

Seis linhagens S5 de milho, com diferenças para conteúdos de óleo e de lisina nos grãos, foram usadas para realizar uma série completa de cruzamentos dialélicos. Os 15 híbridos resultantes foram cultivados em campo com dois níveis de nitrogênio (10 e 130 kg N ha-1). A capacidade geral de combinação (CGC) e a capacidade específica de combinação (CEC) foram obtidas com o método 4, modelo I de Griffing para produtividade de grãos e concentração de lisina e óleo nos grãos. Interação significativa (p < 0.001) foi observada entre CGC e níveis de N para produtividade de grãos, indicando seleção de diferentes linhagens para cada nível de N. Esta interação não foi significativa para conteúdo de lisina, mas existiu efeito significativo (p < 0.1) da CGC nos dois níveis de N para esta variável. Os efeitos de CGC e CEC foram não significativos para conteúdo de óleo, contudo, foi observada correlação positiva entre conteúdos de lisina e óleo nos híbridos, nas linhagens e mesmo nas cultivares utilizadas como controle. Os resultados indicam a efetividade da seleção para conteúdo de lisina nas linhagens não opaco de milho estudadas, independente dos níveis de N, e a possibilidade de atingir concomitantemente maiores níveis de óleo e lisina nos grãos.

Six S5 lines of maize, with differences for lysine and oil contents in grains, were used to carry out a complete series of diallelic crosses. The resulting 15 hybrids were grown in a field at two nitrogen levels (10 and 130 kg N ha-1). The general combining ability (GCA) and specific combining ability (SCA) were obtained by using the method 4, model I of Griffing for grain yield, and grain lysine and oil contents. Significant (p < 0.001) interaction was observed between GCA and N levels for grain yield, indicating the selection of different lines for each N level. This interaction was not significant for lysine content, but there were significant effects of GCA for this trait at both N levels (p < 0.1). Significant effects were not observed for GCA or SCA for oil content, however a positive correlation was observed between lysine and oil contents in the hybrids, in the lines and even in the control cultivars. The results indicate the effectiveness of selection for lysine content, irrespective of N level, in the studied non-opaque maize lines, and the possibility of achieving both high lysine and oil content in grains.

A seletividade dos herbicidas é uma tecnologia agrícola que tem sido vastamente explorada nas estratégias de controle químico de plantas daninhas. É resultado da ação conjunta de diversos mecanismos que protegem a cultura da fitotoxicidade dos tratamentos herbicidas, mantendo-a com níveis de injúrias aceitáveis agronomicamente, ou mesmo na ausência destas. O principal mecanismo de seletividade dos herbicidas é o metabolismo diferencial desses produtos entre plantas daninhas e cultivadas, em que, nas situações de recomendação agronômica, as plantas daninhas são menos hábeis em realizá-lo. Neste caso, a fitotoxicidade pode ser entendida como a suplantação da capacidade máxima de proteção oferecida pelos mecanismos de seletividade ou, considerando o metabolismo como o principal mecanismo, como a superação da capacidade intrínseca da espécie em detoxificar determinada molécula. Considerando-se que o metabolismo de herbicidas envolve gasto de energia, os sintomas de fitotoxicidade caracterizam um segundo gasto energético que não deve ser aceito como uma resposta fisiológica natural, portanto pode resultar em perdas de rendimento das culturas. Para evitar ou minimizar as perdas ou injúrias às culturas, é necessário que as recomendações de herbicidas sejam baseadas em trabalhos de seletividade conduzidos com adequado rigor experimental; bem como é importante a conscientização dos agricultores quanto a melhor forma de utilizar cada produto.

Herbicide selectivity is an agricultural technology largely exploited in chemical strategies of weed control. The joint action of several protection mechanisms avoids phytotoxicity from herbicide treatment, maintaining the level of agronomically accepted damage to a minimum, or even totally avoiding them. The major mechanism of herbicide selectivity derives from the differential metabolism between weed and crop plant species, with weeds presenting a limited ability to perform it under agronomically recommended conditions. In this case, phytotoxicity can be interpreted as an overcoming of the maximum protection capacity offered by the mechanisms of selectivity, or when considering metabolism as the main factor, the overcoming of the inherent plant ability to detoxify a particular molecule. Considering that herbicide metabolism requires energy disposal, symptoms of phytotoxicity characterize an additional waste of energy that should not be accepted as a natural physiologic response; therefore it might result in yield losses. To avoid or minimize crop losses or damages, it is required that herbicide application recommendations are based on results from rigorously conducted selectivity experiments, as well as that there is an increase in the awareness of growers about the best use of each product.

Plants not only evolve but also reduce oxygen in photosynthesis. An inevitable consequence of this normal process is the production of reactive oxygen species (ROS). Plants are adequately protected by the presence of multiple antioxidative enzymes in the cytosol and also in the different cell organelles such as chloroplasts, mitochondria, and peroxisomes. Traditionally, ROS were considered to be only a toxic byproduct of aerobic metabolism. However, recently it has become apparent that plants actively produce these molecules which may control many different physiological processes such as abiotic and biotic stress response, pathogen defense and systemic signaling. The search results using the Citrus Genome Program in Brazil (CitEST) for oxidative stress and the antioxidant enzyme system in Citrus Sinensis variety ‘Pera IAC’ indicated that the multiple ROS-scavenging enzymes were expressed throughout all citrus tissues. The analyses demonstrated the ubiquitous expression of metallothioneins, probably indicating a constitutive expression pattern. Oxalate oxidase has been identified as the most abundant expressed gene in developing fruits, which suggests a specific function in the ripening of citrus fruit. Moreover, infected leaves with Xylella fastidiosa and Leprosis citri showed a massive change in their ROS gene expression profile which may indicate that the suppression of ROS detoxifying mechanisms may be involved in the induction of the diseases.

Lisina, treonina, metionina e isoleucina são derivados do aspartato. Aspartato quinase tem um papel chave no controle da via metabólica do aspartato. A enzima é muito sensível à manipulação e armazenagem e o ensaio enzimático do hidroxamato que é normalmente utilizado para determinar a atividade da aspartato quinase deve ser alterado de acordo com a espécie de planta e tecido vegetal a ser analisado. Nós otimizamos o ensaio para a determinação da atividade da aspartato quinase em culturas celulares de calos de milho. Entre os vários parâmetros testados do ensaio, a concentração de ATP/Mg e temperatura foram críticos para atividade enzimática. No caso da temperatura, 35°C foi a temperatura ótima para atividade da aspartato quinase.

Lysine, threonine, methionine and isoleucine are synthesized from aspartate in a branched pathway in higher plants. Aspartate kinase plays a key role in the control of the aspartate pathway. The enzyme is very sensitive to manipulation and storage and the hydroxamate assay normally used to determine aspartate kinase activity has to be altered according to the plant species and tissue to be analyzed. We have optimized the assay for the determination of aspartate kinase in maize plants callus cell cultures. Among all the assay parameters tested, the concentration of ATP/Mg and temperature were critical for enzyme activity. In the case of temperature, 35°C was shown to be the optimum temperature for aspartate kinase activity.

As sementes dos cereais são pobres em aminoácidos essenciais, principalmente lisina, triptofano e treonina. Os aminoácidos lisina e treonina são sintetizados na via metabólica do aspartato. Apesar de a maioria das enzimas da via do aspartato ter sido isolada e caracterizada em várias espécies de plantas, o metabolismo de lisina e treonina é totalmente desconhecido em sorgo. Isolamos duas enzimas, aspartato quinase (AK) e homosserina desidrogenase (HSDH) de sorgo, estabelecendo condições ótimas de ensaio para determinação da atividade de AK e HSDH. Observaram-se as atividades mais elevadas de AK e HSDH em sementes imaturas. A atividade de AK foi inibida por treonina e lisina, indicando a existência de duas isoenzimas, uma sensível à inibição por treonina e, a outra, sensível à inibição pela lisina, predominando, esta última, nas sementes de sorgo. A atividade de HSDH foi inibida por treonina, indicando a existência de uma isoenzima sensível à treonina, a maioria da atividade, entretanto, não foi inibida por treonina, sugerindo a existência de uma segunda isoenzima de HSDH predominante resistente à inibição pela treonina.

Cereal seeds are poor in essential amino acids, particularly lysine, tryptophan and threonine. The amino acids lysine and threonine are synthesized in the aspartate pathway. Although most of the enzymes of the aspartate pathway have been isolated and characterized in higher plant species, the metabolism of lysine and threonine is totally unknown in sorghum. We have isolated two enzymes, aspartate kinase (AK) and homoserine dehydrogenase (HSDH) from sorghum. Optimum assay conditions were established for the determination of AK and HSDH activities. The highest level of activity was observed in immature seeds. AK was shown to be inhibited by threonine and lysine indicating the existence of at least two isoenzymes, one sensitive to threonine inhibition and the other sensitive to lysine inhibition with the latter being predominant in sorghum seeds. HSDH was shown to be inhibited by threonine indicating the existence of a threonine-sensitive HSDH, however, most of the activity was not inhibited by threonine, suggesting the existence of a second predominant isoenzyme of HSDH resistant to threonine inhibition.

A lisina é um aminoácido essencial, sintetizado na via metabólica do ácido aspártico. O catabolismo da lisina é realizado pela ação das enzimas consecutivas, lisina 2-oxoglutarato redutase (LOR) e sacaropina desidrogenase (SDH). A concentração final de lisina solúvel em plantas, incluindo cereais, é controlada tanto pela taxa de síntese quanto pela de catabolismo. Nos últimos 40 anos, as enzimas envolvidas na biossíntese de lisina foram isoladas, purificadas e caracterizadas em várias espécies vegetais, incluindo cenoura, milho, cevada, arroz e coix. Relatos mais recentes revelaram que o catabolismo da lisina pode estar envolvido em vários processos fisiológicos como crescimento, desenvolvimento, resposta a mudanças e estresses ambientais. A manipulação das enzimas dessa via metabólica é imprescindível para obtenção de plantas que acumulem altos níveis de lisina solúvel, bem como um melhor conhecimento desse metabolismo.

Lysine is an essential amino acid, synthesized in plants in the aspartic acid pathway. The lysine catabolism is performed by the action of two consecutive enzymes, lysine 2-oxoglutarate reductase (LOR) and saccharopine dehydrogenase (SDH). The steady state of lysine is controlled by both, synthesis and catabolism rates, with the final soluble lysine concentration in cereal seeds a direct result of these processes. In the last 40 years, the enzymes involved in lysine biosynthesis have been purified and characterized from some plant species such as carrot, maize, barley, rice, and coix. Recent reports have revealed that lysine degradation might be related to various physiological processes, for instance growth, development and response to environmental changes and stress. The understanding of the regulatory aspects of the lysine biosynthetic and catabolic pathways and manipulation of related enzymes is important for the production of high-lysine plants.
