ABSTRACT Visceral leishmaniasis is a zoonosis in which the dog appears as the main source of infection in urban areas. Its diagnosis is complex and the cytopathological exam is a fast and cheap alternative to parasite direct visualization and its sensitivity can be increased by immunocytochemistry, though with a higher cost. The accuracy of such methods is dependent on the microscopist's experience and therefore, this study evaluated the reliability of such techniques between two observers, from bone marrow aspirates of 50 dogs from an endemic area for the disease. The parasitological culture in Novy-MacNeal-Nicolle medium was used as the reference standard. Among the main findings, the sensitivities obtained by observers I and II were respectively 62.5% and 37.5%, while specificities were 81.1% and 100%. On immunocytochemistry evaluation, the sensitivity was 0% for both evaluators and the specificity 97.3% and 100%. The agreement between evaluators was weak (κ = 0.167) for the cytopathological test and it could not be evaluated for immunocytochemistry, for which there was no detection by the evaluator II. The agreements among the diagnostic methods and the standard reference for the observer I were reasonable (κ = 0.364) for cytopathological examination and bad (κ = -0.041) for immunocytochemistry. For observer II, such agreement could be assessed only for the cytopathological test, being moderate (κ = 0.497). The results point to the possible expertise difference between evaluators, with the evaluator II demonstrating greater experience when interpreting the citopathological test. Although there was the expected sensitivity increase with immunocytochemistry, the technique used in this study was not effective for the diagnosis of infection, regardless of the evaluator.

Protetores ou safeners protegem culturas contra a injúria de herbicidas. O propósito deste estudo foi investigar a suscetibilidade diferencial de cinco variedades de trigo (Triticum aestivum) ao herbicida fenoxaprop-p-ethyl e o efeito do protetor mefenpyr-diethyl na minimização da injúria do herbicida e sobre o conteúdo de lipídios. As variedades BRS 49, CD 104, CEP 24, IAPAR 78 e Rubi foram pulverizadas com fenoxaprop-p-ethyl (69 g ha-1), fenoxaprop-p-ethyl + mefenpyr‑diethyl (69 g + 18,75 g ha¹) ou mefenpyr-diethyl (18,75 g ha¹). A injúria das plantas foi avaliada visualmente aos 7 e 14 dias após o tratamento (DAT). A atividade da glutationa S-transferase (GST) foi analisada na parte aérea aos 7 DAT, e o conteúdo de lipídios, aos 14 DAT. As variedades CEP 24, IAPAR 78 e Rubi foram mais tolerantes ao fenoxaprop-p-ethyl do que a BRS 49, e a variedade CD 104 rapidamente se recuperou dos sintomas de intoxicação produzidos pelo herbicida. Mefenpyr-diethyl preveniu a injúria da cultura pelo herbicida. A atividade da GST não foi correlacionada diretamente com a desintoxicação do fenoxaprop-p-ethyl. Contudo, o conteúdo de lipídios esteve relacionado à suscetibilidade de trigo ao tratamento de fenoxaprop-p-ethyl.

Safeners protect crops against herbicide injury. The aim of this study was to examine the differential susceptibility of five wheat (Triticum aestivum) varieties to the herbicide fenoxaprop-p-ethyl, as well as the performance of mefenpyr-diethyl on minimizing herbicide injury and on lipid contents. Varieties BRS 49, CD 104, CEP 24, IAPAR 78 and Rubi were sprayed with fenoxaprop-p-ethyl (69 g ha¹), fenoxaprop-p-ethyl + mefenpyr-diethyl (69 g + 18.75 g ha-1), or mefenpyr-diethyl (18.75 g ha-1). Plants were evaluated visually for injury at 7 and 14 days after treatment (DAT). Glutathione S-transferase (GST) activity was assayed in aerial parts at 7 DAT, and lipid content was measured at 14 DAT. Varieties CEP 24, IAPAR 78 and Rubi were more tolerant to fenoxaprop-p-ethyl than BRS 49, and CD 104 rapidly recovering from the slight phytotoxicity symptoms produced by the herbicide. Mefenpyr-diethyl prevented crop injury associated with the herbicide. GST activity did not correlate directly with fenoxaprop-p-ethyl detoxification. However, lipid content was related to the susceptibility of wheat to fenoxaprop-p-ethyl treatment.

The prevalence of T. gondii infection was investigated in free-range chickens for human consumption in the county of Rio Bonito, Rio de Janeiro, Brazil. Thus, a search for antibodies to T. gondii by indirect fluorescent antibody test was done in the serum of 220 chickens from twenty two farms. Titres equal or greater than 16 were considered positive. The prevalence of toxoplasmic infection found in chickens was 27.6% (61/220). There were positive cases in eighteen (77.2%) farms. The more frequent titre was 16 (12.5%) and the highest was 4096 (0.4%). The high prevalence of T. gondii in chickens and the high percentage of farms with positive cases indicate environmental contamination in the studied area showing the possibility of human and animal infection.

O lactofen é um herbicida difeniléter recomendado para controlar plantas daninhas de folhas largas em campos de soja (Glycine max) e seu mecanismo de ação é a inibição da protoporfirinogênio-IX oxidase (Protox), que atua na biossíntese de clorofilas. Essa inibição resulta em um acúmulo de protoporfirina-IX, o que leva à produção de espécies reativas de oxigênio (ROS), causando estresse oxidativo. Conseqüentemente, podem ocorrer manchas, enrugamento e queima das folhas, o que leva à paralisação temporária do crescimento da cultura. Porém, o óxido nítrico (NO) atua como um antioxidante na eliminação direta das ROS. Assim, o objetivo deste trabalho foi verificar, por meio de avaliações fitométricas e bioquímicas, o efeito protetor do NO em plantas de soja tratadas com o herbicida lactofen. Plantas de soja foram pré-tratadas com diferentes doses de Nitroprussiato de Sódio (SNP), substância doadora de NO, e então pulverizadas com 168 g a.i. ha-1 lactofen. O pré-tratamento com SNP foi benéfico, pois o NO reduziu os sintomas de injúria causados pelo lactofen nos folíolos jovens e manteve baixos teores de açúcares solúveis. Porém, o NO proporcionou crescimento mais lento das plantas. Desta forma, posteriores estudos são necessários para elucidar os mecanismos de ação do NO na sinalização do estresse promovido pelo lactofen na cultura da soja.

Lactofen is a diphenylether herbicide recommended to control broad-leaved weeds in soybean (Glycine max) fields and its mechanism of action is the inhibition of protoporphyrinogen-IX oxidase (Protox), which acts in the chlorophyll biosynthesis. This inhibition results in an accumulation of protoporphyrin-IX, which leads to the production of reactive oxygen species (ROS) that cause oxidative stress. Consequently, spots, wrinkling and leaf burn may occur, resulting in a transitory crop growth interruption. However, nitric oxide (NO) acts as an antioxidant in direct ROS scavenging. Thus, the aim of this work was to verify, through phytometric and biochemical evaluations, the protective effect of NO in soybean plants treated with the herbicide lactofen. Soybean plants were pre-treated with different levels of sodium nitroprusside (SNP), a NO-donor substance, and then sprayed with 168 g a.i. ha-1 lactofen. Pre-treatment with SNP was beneficial because NO decreased the injury symptoms caused by lactofen in young leaflets and kept low the soluble sugar levels. Nevertheless, NO caused slower plant growth, which indicates that further studies are needed in order to elucidate the action mechanisms of NO in signaling the stress caused by lactofen in soybean crop.

RESUMO Este trabalho teve o objetivo de avaliar a eficiência do protetor fluxofenim no tratamento de sementes de trigo, cultivar Ônix, para o aumento da seletividade do herbicida S-metolachlor aplicado em pré-emergência e determinar a atividade da enzima de detoxificação glutationa S-transferase (GST). O trabalho foi realizado em duas etapas: a primeira para a avaliação em campo da eficiência do protetor em reduzir sintomas visuais de fitointoxicação causados pelo herbicida, e a segunda para a determinação da atividade da GST, conduzida em laboratório. Os tratamentos utilizados foram: aplicação ou não do protetor na dose de 40 mL por 100 kg de sementes e a pulverização do herbicida S-metolachlor nas doses de 1.440 e 2.880 mL i.a.ha-1, além de uma testemunha sem aplicação, sendo que, na determinação da atividade da GST, se excluiu o tratamento com aplicação de 2.880 mL i.a.ha-1 de S-metolachlor. Foram comparadas a suscetibilidade ao herbicida por meio da avaliação visual de injúrias aos 3, 7, 15 e 30 dias após a emergência (DAE) e a massa seca de raiz e parte aérea aos 10 DAE. Para a determinação da atividade da GST foram coletadas amostras da parte aérea das plantas aos 15 dias após a aplicação dos tratamentos e aos 10 DAE. Apesar do fluxofenim utilizado no tratamento da cultivar de trigo conferir uma maior tolerância ao herbicida S-metolachlor, ocorreu fitointoxicação visual e redução do estande de plantas. A cultivar testada apresentou aumento na atividade da enzima GST quando da utilização do fluxofenim anteriormente à aplicação do herbicida S-metolachlor na dose de 1.440 mL i.a. ha-1.

ABSTRACT This work aimed to evaluate the effectiveness of fluxofenim used for seed treatment as safener in wheat, Ônix cultivar, treated with the herbicide S-metolachlor applied in pre-emergence. The study was divided in two steps. The first step consisted of an evaluation of fluxofenim’s safener potential for the reduction of visual symptoms of S-metolachlor injury in the field, and the treatments were S-metolachlor at 1,440 and 2,880 mL i.a. ha-1 and fluxofenim at 0, and 40 mL per 100 kg of seeds, and a control without herbicide. The second step was to evaluate glutathione S-transferase activity (GST). Herbicide phytotoxity was measured by way of visual symptoms at 3, 7, 15, and 30 days after emergence (DAE), dry matter from roots and leaves at 10 DAE. For the determination of GST activity, the canopy of plants was collected at 10 DAE and 15 days after treatment application. The wheat presented low tolerance to S-metolachlor at both rates, and fluxofenim increased S-metolachlor selectivity to wheat but not sufficiently, reducing plant population to a nonacceptable level. Gluthationa S-transferase activity for wheat increased when seeds treated with fluxofenim were submitted to S-metolachlor at 1,440 mL a.i. ha-1.

O efeito antibacteriano do óleo essencial da Lippia sidoides (alecrim-pimenta) foi testado contra as bactérias Staphylococcus aureus e Escherichia coli isoladas de queijo minas artesanal, produzido no Brasil. A Concentração Inibitória Mínima (CIM) determinada por Teste de Diluição foi de 13 µL de óleo mL-1 para as duas bactérias, o que caracteriza ação inibitória em caldo durante vinte quatro horas de interação. A Concentração Bactericida Mínima (CBM) determinada através de Teste de Suspensão, com um minuto de contato, foi de 25 µL de óleo mL-1 para ambas as bactérias testadas, obtendo-se a esta concentração o efeito bactericida de 99,9% sobre as células bacterianas viáveis em cada uma das amostras. Os resultados demonstraram atividade bactericida do óleo essencial de Lippia sidoides contra S. aureus e E. coli, sugerindo a possibilidade do uso como agente antibacteriano em alimentos.

The antibacterial effect of Lippia sidoides (rosemary pepper) essential oil was tested against the bacteria Staphylococcus aureus and Escherichia coli isolated from homemade Minas cheese produced in Brazil. The Minimum Inhibitory Concentration (MIC) determined in the Dilution Test was 13 µL oil mL-1 for both bacteria, which characterizes inhibitory action in broth for a 24-hour interaction period. The Minimum Bactericidal Concentration (MBC) determined in the Suspension Test, with one minute of contact, was 25 µL oil mL-1 for both tested bacteria, obtaining at this concentration a bactericidal effect of 99.9% on the viable bacterial cells from each sample. Results demonstrated the bacterial activity of Lippia sidoides essential oil against S. aureus and E. coli, suggesting its use as an antibacterial agent in foods.

A glutationa S-transferase (GST, EC 2.5.1.18) desempenha um papel importante na resposta do estresse causado por herbicidas nas plantas; é considerada uma enzima de desintoxicação, por metabolizar grande variedade de compostos xenobióticos, por meio da conjugação destes com glutationa reduzida, formando substâncias de baixa toxicidade. O milho (Zea mays) foi escolhido neste trabalho por apresentar problemas de injúrias quando submetido ao controle químico de plantas daninhas, por meio do uso de herbicidas. Esta pesquisa teve como objetivo determinar as alterações na atividade desta enzima em plantas de milho submetidas ao tratamento pelo herbicida glyphosate. O delineamento experimental utilizado foi o inteiramente casualizado, em esquema fatorial 4x4, com quatro tratamentos herbicidas (glyphosate nas concentrações de 1.000, 2.500 e 5.000 ppm e as plantas-controle tratadas com água) e quatro estádios de desenvolvimento (9, 16, 23 e 30 dias após a emergência), com cinco repetições. O herbicida foi aplicado na parte aérea das plântulas de milho. A parte aérea foi coletada às 24, 48 e 72 horas após a aplicação do herbicida e utilizada para a determinação da atividade da GST e do teor de lipoperóxidos. Foi verificado que os teores de lipoperóxidos não foram alterados pelo tratamento com o glyphosate, porém a atividade de GST aumentou na maioria dos tratamentos utilizados, indicando ter ação na degradação do herbicida glyphosate em plantas de milho.

Glutathione S-transferase (GST, EC 2.5.1.18) plays an important role in the response to stress caused by herbicides in plants; it is considered to be a detoxifying enzyme since it metabolizes a great variety of xenobiotic compounds, by combining them with reduced glutathione, forming substances of low toxicity. Maize (Zea mays) was chosen in this work for showing injury problems when submitted to weed chemical control. This research aimed to determine the alterations in the activity of this enzyme in glyphosate-treated maize plants. The experimental design was completely randomized, in a 4 x 4 factorial arrangement, with four treatments (glyphosate at concentrations of 1,000, 2,500 and 5,000 ppm and control plant treated with water) and four developmental stages (9, 16, 23 and 30 days after emergence), with five replicates. The herbicide was applied on the maize plant shoots, which were collected at 24, 48 and 72 hours after herbicide application and utilized for determining the GST activity and lipoperoxide content. It was verified that glyphosate does not change the lipoperoxide content, but increases GST activity for most treatments utilized, indicating its action in the degradation of glyphosate in maize plants.

Plantas de aguapé foram cultivadas em solução nutritiva de Hoagland & Arnon n.2, cujo aumento dos níveis de N, P e Cu estabeleceu as diferenças entre os tratamentos. Utilizou-se o delineamento experimental inteiramente casualizado, com quatro repetições. As variáveis fisiológicas avaliadas foram área foliar, peso de matéria seca e taxa de crescimento absoluto, taxa de crescimento relativo, taxa assimilatória líquida, razão de área foliar, peso específico de folha, área foliar específica. Foram determinados também os teores de açúcares totais e redutores e de aminoácidos totais e a atividade das enzimas glutationa S-transferase e superóxido dismutase. Os extratos enzimáticos foram obtidos da matéria fresca da parte aérea das plantas. Após a coleta, foram determinados os pesos de material seco de raízes, pecíolos e folhas, que foram utilizados para a determinação de açúcares solúveis totais e redutores e de aminoácidos. O excesso de nitrogênio causou aumento de açúcares nas folhas e de aminoácidos nas raízes. Já o tratamento com excesso de fósforo levou ao aumento de açúcares nas raízes. Os resultados apresentados sugerem que, entre os nutrientes em excesso avaliados, o cobre (0,12 mg L-1) foi o maior indutor da atividade da GST e SOD, sugerindo que este elemento induziu estresse nas plantas de aguapé.

Water hyacinth plants were cultivated in a Hoagland & Arnon n.2 nutrient solution with increased N, P and Cu levels establishing the differences among the treatments. The experimental design was completely randomized, with four replicates. Physiological evaluations included leaf area, dry matter and growth analysis parameters (absolute growth rate, relative growth rate, net assimilatory rate, leaf area ratio, specific leaf weight, specific leaf area). Biochemistry evaluations were total and reducing sugars, total amino acids and activity of the antioxidant enzymes (glutathione S-transferase and superoxide dismutase). The enzyme extract was obtained from fresh matter of the aerial part of the plants. Root and leaf dry material was used to determine total and reducing sugars, and amino acids. Nitrogen supplementation caused increased levels of sugars in the leaves and of amino acids in the roots. The treatment with excess phosphorus caused increased levels of sugars in the roots. The results suggest that copper (0.12 mg L-1) increased the activity of GST and SOD, inducing stress in the water hyacinth plants.
