Introducción. Se han detectado asociaciones biológicas huésped-parásito específicas que sugieren que los triatominos del género Rhodnius podrían actuar como filtros biológicos en la transmisión de Trypanosoma rangeli . Objetivo. Estudiar la sensibilidad de cuatro especies de Rhodnius ( Rhodnius robustus , Rhodnius neglectus , Rhodnius nasutus y Rhodnius p ictipes ) frente a la cepa de T. rangeli (SC-58/KP1-). Materiales y métodos. Se seleccionaron treinta ninfas de cada especie después de xenodiagnóstico artificial en sangre infectada con T. rangeli. Se examinaron periódicamente m uestras de heces y hemolinfa. Los insectos con hemolinfas infectadas fueron alimentados en ratones a fin de comprobar la transmisión por picadura y posteriormente disecados para confirmar la infección de las glándulas salivales . Resultados . En Rhodnius pictipes se encontró un mayor porcentaje de infección intestinal que en las otras especies . Se detectaron epimastigotes y tripomastigotes en la hemolinfa de las cuatro especies , y se encontró que el parasitismo fue menor en las especies del linaje R. robustus . Rhodnius robustus y R. neglectus no transmitían T. rangeli a ratones por picadura: después de la disección , sus glándulas no estaban infectadas. Solo un espécimen de R. nasutus y dos de R. pictipes transmitieron el parásito por la picadura . La tasa de infección glandular fue de 16 % para R. pictipes y de 4 % para R. nasutus . Conclusiones . La capacidad infecciosa ( hemolinfática, intestinal y glandular ) y la transmisión de T. rangeli (SC-58/KP1-) fueron mayores y más eficientes en R. pictipes . Estos resultados refuerzan la hipótesis de que estos triatominos actúan como filtros biológicos en la transmisión de T. rangeli .

Introduction: Specific host-parasite a ssociations have been detected experimentally and suggest that triatomines of the genus Rhodnius act as biological filters in the transmission of Trypanosoma rangeli . Objective: To analyze the susceptibility of four Rhodnius species ( Rhodnius robustus , Rhodnius neglectus , Rhodnius nasutus and Rhodnius pictipes ) to a Brazilian strain of T. rangeli (SC-58/KP1-). Materials and methods: We selected t hirty nymphs of each species, which were fed on blood infected with T. rangeli . Periodically, samples of feces and hemolymph were analyzed. Triatomines with T. rangeli in their hemolymph were fed on mice to check for transmission by bites. Later, the triatomines were dissected to confirm salivary gland infection. Results: Specimens of R. pictipes showed higher rates of intestinal infection compared to the other three species. Epimastigotes and trypomastigotes were detected in hemolymph of four species; however, parasitism was lower in the species of the R. robustus lineage. Rhodnius robustus and R. neglectus specimens did not transmit T. rangeli by bite; after dissection, their glands were not infected. Only one specimen of R. nasutus and two of R. pictipes transmitted the parasite by bite. The rate of salivary gland infection was 16% for R. pictipes and 4% for R. nasutus . Conclusions: Both infectivity (intestinal, hemolymphatic and glandular) and transmission of T. rangeli (SC58/KP1-) were greater and more efficient in R. pictipes. These results reinforce the hypothesis that these triatomines may act as biological filters in the transmission of T. rangeli .

The development of strategies for the adequate control of the vector transmission of Chagas disease depends on the availability of updated data on the triatomine species present in each region, their geographical distribution, natural infections by Trypanosoma cruzi and/or T. rangeli, eco-biological characteristics and synanthropic behavioral tendencies. This paper summarizes and updates current information, available in previously published reports and obtained by the authors our own field and laboratory studies, mainly in northwest of Peru. Three triatomine species exhibit a strong synanthropic behavior and vector capacity, being present in domestic and peridomestic environments, sometimes showing high infestation rates: Rhodnius ecuadoriensis, Panstrongylus herreri and Triatoma carrioni The three species should be given continuous attention by Peruvian public health authorities. P. chinai and P. rufotuberculatus are bugs with increasing potential in their role as vectors according to their demonstrated synanthropic tendency, wide distribution and trophic eclecticism. Thus far we do not have a scientific explanation for the apparent absence ofT. dimidiata from previously reported geographic distributions in Peru. It is recommended, in the Peruvian northeastern Amazon region, the development of field studies on species of the genus Rhodnius, as well as of other triatominae, to evaluate their present Trypanosomatidae vector capacity.

El desarrollo de estrategias adecuadas para el control de la transmisión vectorial de la Enfermedad de Chagas depende: de la disponibilidad de datos actualizados de las especies de triatominos presentes en cada región, de su distribución geográfica, infección natural por Trypanosoma cruzi ylo T. rangeli, características eco-biológicas y tendencias de comportamiento sinantrópico. Este trabajo resume y actualiza la información disponible en la literatura y aquella obtenida en nuestros estudios de campo y de laboratorio desarrollados en los últimos años por los autores, predominantemente, en la región Nor-Occidental del Perú. El resultado de esas observaciones es la detección de que, tres especies de triatominos presentan un importante comportamiento sinantrópico y capacidad vectorial, ocupando ambientes intradomiciliares y peridomiciliares: Rhodnius ecuadoriensis, Panstrongylus herreri y Triatoma carrioni. Las tres especies deben recibir constante atención por parte de las autoridades de Salud Pública Peruana. P. chinai y P rufotuberculatus tienen potencial creciente en su papel de vectores, por la comprobada tendencia sinantrópica, distribución geográfica y eclecticismo trófico. Se desconocen las causas de la aparente ausencia actual de Triatoma dimidiata, dada su presencia constatada anteriormente en el Perú. Se recomienda que en la región Nor-Oriental Amazónica Peruana se realicen en forma urgente estudios sobre las especies del genero Rhodnius y de otros triatominos que permitan evaluar la real capacidad vectorial de los Trypano-somatideos.

Artrópodos associados à copa de árvores, principalmente palmeiras, são pouco conhecidos no Cerrado. Para descrever a estrutura da comunidade de artrópodos associada à copa de Mauritia flexuosa (Arecaceae) foram amostradas 150 palmeiras em seis "veredas" do Distrito Federal, de áreas silvestres, rurais e periurbanas na estação chuvosa. Os artrópodos presentes nos ninhos abandonados de aves, refúgios de mamíferos, folhas e matéria orgânica foram coletados manualmente, fixados em etanol 70% e separados em ordem, família, morfoespécie e guildas alimentares. As características das palmeiras medidas foram altura da estipe, diâmetro da copa, número de folhas e de ninhos de aves nas palmeiras. Foram coletados 3.862 indivíduos, pertencentes a 15 ordens, 45 famílias e 135 morfoespécies. As ordens mais abundantes foram Coleoptera (28,6%), Blattodea (21,8%), Collembola (11,4%) e Hemiptera (10,2%). As famílias Blaberidae, Tenebrionidae, Entomobryidae, Reduviidae, Oniscidae, Staphylinidae, Carabidae e Formicidae representaram 82,1% de todos os indivíduos coletados. A maioria das morfoespécies foi pouco abundante, 71 (52,6%) apresentaram uma abundância média igual ou menor que 1 indivíduo/palmeira. Coleoptera compreendeu o maior número de morfoespécies (43,7%) seguida de Araneae (20,0%). A análise das guildas alimentares mostrou prevalência de predadores e hematófagos (36,0%). A riqueza e a abundância de artrópodos foram menores no ambiente periurbano. O número de ninhos de aves apresentou correlação positiva com abundância e riqueza, o que não ocorreu com as medidas das palmeiras. A importância de M. flexuosa para a manutenção da artropodofauna nas "veredas" no bioma Cerrado é discutida.

Canopy arthropods, mainly from palm trees, are little known in the Brazilian Cerrado. In order to describe the arthropod community structure associated with the crown of Mauritia flexuosa (Arecaceae), we sampled 150 palm trees in six "veredas" of the Federal District, Brazil, in wild, rural and periurban areas in the rainy season. The arthropods within abandoned bird nests, mammal refuges, leaves and organic matter were manually collected, preserved in ethanol 70% and separated by order, family, morphospecies and feeding guilds. Stem height and diameter of the palm crowns were measured and leaves and bird nests were counted. We collected 3,862 arthropods, from 15 orders, 45 families and 135 morphospecies. The most abundant orders were Coleoptera (28.6%), Blattodea (21.8%), Collembola (11.4%) and Hemiptera (10.2%). The families Blaberidae, Entomobryidae, Reduviidae, Oniscidae, Staphylinidae, Carabidae and Formicidae, represented 82.1% of all individuals collected. The majority of morphospecies was not abundant, 71 (52.6%) were represented by less than 1 individual/tree. Coleopterans accounted for the highest number of morphospecies (43.7%) followed by Araneae (20.0%). The analysis of the arthropod feeding guilds showed prevalence of predatory/hematophagous ones (36.0%). Arthropod richness and abundance presented smaller values for periurban environment. The number of bird nests presented positive correlation with abundance and richness; this was not found when considering the measurements of the palm trees. The importance of M. flexuosa for the maintenance of the arthropod fauna of the "veredas" in Cerrado biome is discussed.

An electron microscopy study was carried out on the salivary glands of the triatomine bug Rhodnius ecuadoriensis experimentally infected with a Peruvian strain of Trypanosoma (Herpetosoma) rangeli. Epimastigotes forms of T. rangeli were found in large numbers in the haemolymph surrounding the salivary gland. Some of them were closely associated with the basal lamina of the gland epithelium, apparently in a process of cell invasion, in which the flagellum breaking through the basal lamina. Within the cytoplasm of the gland epithelial cells, there were rounded forms of T. rangeli, consisting of tightly coiled sphaeromastigotes. The gland lumen was filled with large numbers of mostly epimastigotes forms of the flagellate; only few paramastigotes and trypomastigotes forms could be unequivocally identified. The presence of paramastigotes in the life cycle of T. rangeli has yet to be fully assessed

Fué realizado un estudio ultraestructural de las glándulas salivares del triatomino Rhodnius ecuadoriensis experimentalmente infectado por el tripanosomatídeo, Trypanosoma (Herpetosoma) rangeli, con la finalidad de documentar los aspectos básicos de la infección glandular por este flagelado. Gran número de formas epimastigotes fueron encontradas en la hemolinfa que baña la parte externa de las glándulas en el hemoceloma. Algunos flagelados parecían íntimamente asociados a la membrana basal del epitelio glandular y varios de ellos parecían estar iniciando el proceso de su invasión, rompiendo la lámina basal con el flagelo. En el interior de las células glandulares observamos que, T. rangeli se presentaba en la forma de esferomastigotes, células redondeadas rodeadas externamente por un flagelo, que envolvía apretadamente el parásito. En la luz glandular también observamos gran número de epimastigotes mezclados con los productos de la secreción glandular; el material presentó apenas pocas formas paramastigotes y trypomastigotes identificadas de manera inequívoca. Descreve-se um caso de calazar grave resistente a dez cursos de antimonial petitavalente (glucantime) à base de 20mg de Sb5/kg/dia, que respondeu favoravelmente ao sulfato de aminosidine intramuscular na dose de 20mg/kg/dia por 20 dias, repetido após 20 dias. O parasitismo esplénico passou de 50 parasitos por campo a 3 parasitos em 10 campos logo após a primeira série de sulfato de aminosidine, tornando-se negativo depois de sete meses. A melhora clínica foi imediata, com redução gradual da hepatoesplenomegalia, e desaparecimento 26 meses após. Neste período aumentou 13 kg. Após o uso de aminosidine a reação de Monténégro tornou-se positiva e as células mononucleares responderam quando estimuladas com antígenos de leishmânia.

A grave kala-azar infection in a 14 years old boy is described. The leishmanial infection failed to respond to ten interrupted courses of glucantime of variable duration (14-56 days) at a dose of 20mg Sb5/kg/day. However a favorable response ocurred to intramuscular aminosidine sulphate (20mg/kg/day) for 20 days. This same regimen was repeated 20 days later. After the first treatment splenic puncture parasite density fell from 50 amastigotes per oil immersion field to 3 amastigotes in 10 fields. A further splenic puncture 7 months after treatment was negative. The marked hepatoesplenomegaly gradually resolved over 26 months follow up and he gained 13 kilogramas in weight. After aminosidine sulphate therapy his Montenegro reaction become positive and his lymphocytes responded to leishmania antigens.

A new parasite species of Leishmania is described, L. (Leishmania) forattinii sp. n., which was isolated from a pooled triturate of liver and spleen of a opossum (Didelphis marsupialis aurita) and from skin samples from a rodent (Proechmys iheringi denigratus), captured in primary forest on the Atlantic Cost of Brazil. Our results on the basis of biological and molecular criteria indicate that this taxonomically distinct parasite ias a new species of the L. mexicana complex, but closely related to L. (L.) aristidesi Laison & shaw, 1979, as revelated by phenetic and phylogenetic numerical analyses of the enzyme data. L. forattinii was clearly distinguishable from other Leishmania species of the genus usisng enzyme electrophoresis, monoclonal antibodies, molecular karyotypes, analysis of restriction enzyme digestion patterns of kinetoplast DNA (kDNA), as well as the use of kDNA hybridization procedures.

Os autores analisam material de biópsias de 378 casos de Leishmaniose Tegumentar, causada por Leishmania braziliensis braziliensis, da localidade endêmica de Três Braços (Estado da Bahia). O parásitos, embora escassos, foram encontrados em 63,7% dos casos da forma cutânea e em 37,5% dos casos da forma mucosa. As alterações dérmicas ou do córion da mucosa permitiram identificar cinco padrões histopatológicos: 1) Reação Exsudativa Celular, constituída por um infiltrado histiolinfoplasmocitário; 2) Reação Exsudativa e Necrótica, na qual ocorre uma necrose no seio do infiltrado inflamatório; 3) Reação Exsudativa e Necrótico-Granulomatosa, que corresponde ao quadro descrito como inflamação crônica granulomatosa com necrose; 4) Reação Exsudativa e Granulomatosa, onde se observa uma reação granulomatosa desorganizada, sem que esteja presente necrose tecidual; 5) Reação Exsudativa e Tuberculóide, caracterizada pelo granuloma tuberculóide. O estudo evolutivo realizado em 49 casos, mostrou que houve uma mudança de padrão histopatológico observada, em biópsias sucessivas, em 63,2% dos casos da forma cutânea e em 45,4% dos casos da forma mucosa. Através desse estudo, é possível afirmar-se que o padrão de Reação Exsudativa Celular constitui o quadro inicial e final da lesão, com os demais padrões aparecendo interposto durante a evolução da doença.

The Authors describe the histopathological findings observed in biopsies from 378 cases of Mucocutaneous Leishmaniasis, caused by Leishmania braziliensis braziliensis. All the patients lived in the locality of Três Braços, State of Bahia, an endemic area of Leishmaniasis in Brazil. Parasites were found in 63.7% of patients with cutaneous form and in 35.7% of cases with mucous lesions, although always in reduced number. Histopathologic findings allowed the caracterization of five different patterns: 1. Exsudative-cellular reaction, due to infiltraction of histiocytes, lymphocytes and plasm cells; 2. Exsudative-necrotic reaction, characterized by the association of necrosis with cellular infiltration; 3. Exsudative and necrotic-granulomatous reaction, corresponding to pattern described as chronic-granulomatous inflammation with necrosis; 4. Exsudativegranulomatous reaction, characterized a disorganized granulomatous reaction, without tissue necrosis; 5. Exsudative-tuberculoid reaction — in which a typical tuberculoid granuloma is formed. The follow-up study in 49 patients showed, in sucessive biopsies, changes in the histopathologic patterns in 63.2% of cutaneous forms and in 45.4% of the mucous forms. It was concluded that the exsudative-cellular reaction represents both the inicial and final pattern of Mucocutaneous Leishmaniasis lesion. The other four histopathologic patterns characterize intermediate forms found during the evolution of the disease.

We have compared differeni media and isolation procedures to cultivate Leishmania braziliensis braziliensis/rom hamster tissues. Wefmd that aspiration of the lesion using a 22 g needle and a five ml syringegives satisfactorv results, and is a convenient sampling method. Difco Blood agar is an efficient medium for isolation. Evans medium gave equal results while Schneiders and AR-103 medium were inferior. In Schneider's medium flagellate growth achieved 0.21 - 2.57 x 10(8) promastigotes/ml bv the fifth day 5-fluorocytosine added to the cultures to avoid fungal contamination does not inhibit the growth of L. b. brasiliensis.

Comparamos diferentes medios artificiales y procedimientos de cultivo in vitro de Leishmania braziliensis braziliensis (Lbb) aislada de lesiones de hamsteres experimentalmente infectados. Observamos que la aspiración de material de la lesión, usando jeringa de 5 mlyaguja 22g(25 x 8mm), mostró resultados satisfactorios siendo un procedimiento conveniente de colecta. El medio de Agar Sangre Difco (DAB) es un medio eficiente para el aislamiento de Lbb. El medio de Evans demostro similares resultados al DAB, mientras que el medio líquido de Schneider y el químicamente definido AR-103 fueron nitidamente inferiores. En la propagación in vitro de Lbb, el medio de Schneider permitió un buen crecimiento del flagelado, alcanzando densidades de 0,21 - 2,57 x 10(8) promastigotes por ml ya, en el 5.° dia de cultivo. La adición de 5-fluorocytosine a los tubos de cultivo (150/jg/ml)para evitar la contaminación por hongos, no inhibió el crecimiento de Lbb.

Em um estudo retrospectivo, indivíduos com a forma mucosa da leishmaniose foram pareados (sexo, idade e ocupação) a outros pacientes com a forma cutânea em atividade, não demonstrando-se diferença significante da freqüência nos grupos raciais. Do mesmo modo, o teste intradérmico de Montenegro não se mostrou diferente nos diversos grupos raciais, entre os 96 pares de pacientes estudados. Uma maior enduração foi observada em pacientes com o acometimento mucoso quando comparado aos cutâneos.

In a retrospective study patients with mucosal leishmaniasis were paired (as regards age sex and occupation) with patients with active cutaneous disease. No signijicant racial dijference could be detected. In the same way the Montenegro cutaneous reaction was not different in 96pairs ofpatients studied as regards different racial groupings. A larger indurated area was observed in patients with mucosal leishmaniasis compared with the cutaneous form.

Três isolados de Leishmania foram obtidos de cinco entre 27 exemplares do roedor Proechimys iheringi denigratus, capturados na região de Três Braços, na mata atlântica do Estado da Bahia, Brasil. O isolamento desse parasito foi feito através de inoculação de triturado de pele, baço e fígado em patas de hamsters. Em pelo menos um dos casos, (MTB-574), o parasito foi isolado da pele. Metas- tase foi observada nos hamsters inoculados, os parasitos cresceram abundantemente em meios artificiais de cultura e um padrão suprapapilario típico foi obtido em Lutzomyia longipalpis, indicando que o parasito pertence ao complexo L. mexicana. Todos os isolados reagiram positivamente com anticorpos monoclonais de L. m. mexicana e L. m. amazonensis. A análise isoenzimática diferenciou o parasito de isolados padrões de L. m. mexicana. L. m. amazonensis. L. m. aristedesi. L. m. pifanoi. L. m. garnhami e L. m. ssp(Goiás-W. Barbosa). O parasito parece ser uma subespécie de L. mexicana muito próxima à L. m. amazonensis, da qual difere pela menor mobilidade eletroforêtica de GPI, PEP e ALAT. Este e o primeiro registro do isolamento de um parasito do gênero Leishmania em um roedor capturado no Estado da Bahia.

Three isolates of Leishmania were recovered from five of 27 specimens of the rodent Proechimys iheringi denigratus Moojen captured near Três Braços in the Atlantic Forest region of Bahia, Brazil. Two of these isolates were recovered from hamsters inoculated with a pooled triturate of liver, spleen and skin tissue from apparently healthy P. i. denigratus. The third isolate was recovered from a triturate of only skin tissue from another. Metastasis was observed in the inoculated hamsters, the parasites grew abundantly in artificial media and a typical suprapylarial pattern of infection in Lutzomyia longipalpis was produced indicating that the parasites belong to the Leishmania mexicana complex. All isolates reacted with Leishmania mexicana mexicana and Leishmania mexicana amazonensis monoclonal antibodies. The isoenzyme analysis differentiated these isolates from standard isolates of L. m. mexicana, L. m. amazonensis, L. m. aristedesi, L. m. pifanoi, L. m. garnhami and L. m. ssp.(Goiás-W. Barbosa). These isolates seem to be a subspecies of L. mexicana very closely related to L. m. amazonensis from which they differ by decreased electrophoretic mobility of GPI, PEP and ALAT. This is the first record of the isolation of a parasite of thegenus Leishmania in a rodent captured in the State of Bahia.

Um inquérito em cães realizado na região de Três Braços, Bahia, mostrou que 3,0% de 98 animais tinham amastigotas em lesões de pele. Parasitos não foram encontrados em pele normal da orelha. De uma amostra selecionada de 13 cães, portadores de lesão cutânea ativa, nove (69,2%) deles estavam comprovadamente infectados. Sete amostras de lesão produziram infecção em hamsters. O estudo biológico (crescimento em meio de cultura, evolução da lesão em hamster e desenvolvimento no tubo digestivo de Lutzomyia longipalpis) identificou o parasito como pertencente ao complexo L. braziliensis. A caracterização bioquímica (mobilidade eletroforética de enzimas em placas de acetato de celulose) e o estudo imunotaxonômico (anticorpos monoclonais) definiram as amostras como L. braziliensis braziliensis. O papel do cão como um possível reservatório de L. b. braziliensis na região de Três Braços é discutido.

During a survey of domestic and hunting dogs conducted in the Três Braços region, State of Bahia, 3,0% of 98 dogs had amastigotes in skin lesions. Parasites were not found in normal ear skin. In a nonrandomly selected sample of 13 dogs with active cutaneous lesions, infection was confirmed in nine (69,2%). Tissue biopsies from seven dog lesions produced infection in hamsters. The biological behaviour of the parasite (rate of growth in culture media, evolution of lesions in hamsters and development in the gut of Lutzomyia longipalpis,) identified it to the Leishmania braziliensis complex. Characterization by biochemical means (eletrophoretic mobility of enzymes in cellulose acetate plates) and immunotaxonomic studies (monoclonal antibodies) defined the strains as L. braziliensis braziliensis. The role of dogs as a possible reservoir host of L. b. braziliensis in Três Braços region is discussed.
