ABSTRACT Secretory cells of the cephalic salivary glands (CSGs) of eusocial bees produce and accumulate lipid-like secretion in the lumens of their alveoli. Correspondingly, secretory cells present typical ultrastructural features of lipid-compound producers. Previous work on bees has revealed inter-specific differences in the chemical composition of secretion, and the production mechanisms and secretory cycle of secretory cells. In this work a comparative analysis of the mechanisms of lipid storage in the CSGs of Apis mellifera (Linnaeus, 1758) and Scaptotrigona postica (Latreille, 1807) workers was carried out. The ultrastructural location of lipids was ascertained using imidazole-osmium (IO), using individuals in different stages of their life cycles. Lipid deposits were identified inside glandular cells and in the alveolar lumens in all individuals, but differences were observed between the species. The glandular cells of A. mellifera workers presented positive reactions to IO as droplets dispersed in the cytoplasm, as vesicles and in the channels formed by apical plasma membrane infolds. In S. postica , lipid compounds were detected inside the mitochondrial matrix and in smooth endoplasmic reticulum cisterns. In both species, forager workers exhibited the largest amounts of lipids stored in the alveolar lumen. The differences between the species are discussed, taking into account specific behavioral differences.

The bee species of the Apinae, in addition to the thoracic salivary glands, possess a pair of cephalic glands originating as branches of the excretory duct that crosses the head. These glands are known as cephalic salivary or labial cephalic glands. The degree of development of these glands in newly emerged, nurse and forager workers and virgin and egg-laying queens of Apis mellifera Linnaeus, 1758 and Scaptotrigona postica Latreille, 1807 were evaluated by measuring the secretory alveolar units. The area of the secretory alveoli, measured in total gland preparations, was used to evaluate differences in size. In both species, gland size was found to increase progressively from newly emerged workers to foragers and from virgin to egg-laying queens. A statistical analysis revealed significant differences (p < 0.05) in the area of gland alveoli of workers in different life phases in both species, and between S. postica virgin and egg-laying queens, but not between A. mellifera queens. In the case of workers, this suggests cephalic salivary gland secretion has a function in forager activity and, in queens, a possible pheromonal function.

Some adult eusocial bees have a pair of cephalic salivary glands (CSG) in addition to the thoracic labial or salivary gland pairs. This paper deals with variations in morphological features and secretion production of the CSG of females and males of Apis mellifera Linnaeus, 1758 and Scaptotrigona postica Latreille, 1807. The following life stages were studied: newly emerged, nurse, and forager workers; newly emerged and egg-laying queens; and newly emerged and sexually mature males. The histological results showed that the CSG differs between the two species in the following features: while alveoli and duct cells are cuboidal in workers and queens of A. mellifera, they change from cuboidal to flat in S. postica as the workers age. The glands of newly emerged males and females of A. mellifera are similar. However, as males become sexually mature, glands degenerate and practically disappear. The secretion from the glands of females of both species is oleaginous and gradually accumulates in the lumen of the alveoli in the beginning of the adult phase. Consequently, forager workers and egg-laying queens exhibit more turgid alveoli than younger individuals. Sudan black and Nile's blue staining indicated that the CSG secretion consists of neutral lipids. The possible role of gland secretion is discussed taking in account tasks performed by the individuals in the particular phases studied.

A influência do hormônio juvenil sobre o desenvolvimento do ovário de larvas de operárias de Apis mellifera foi analisada levando em conta a determinação trófica das castas, segundo a qual a alimentação larval é controlada pelas operárias de maneira a promover uma diferenciação de castas controlada pela produção e disponibilidade desse hormônio. A hipótese testada é que a ação do hormônio juvenil seja capaz de proteger ou prevenir a degeneração nos ovários das larvas de operárias. Foi feita aplicação tópica de 1 ml de hormônio dissolvido em hexano na concentração de 1 mg/ml do segundo até o quinto dia de vida larval, e a morfologia dos ovários avaliada nos dias subseqüentes à aplicação até ao sexto dia de vida larval. Como controles foram utilizadas larvas nas quais se aplicou 1 ml de hexano e larvas que não receberam nenhum tratamento. Constatou-se que o efeito do hormônio juvenil varia conforme a idade larval em que é aplicado e que este efeito foi maior quando a aplicação foi feita no terceiro dia de vida larval.

The influence of juvenile hormone (JH) on the ovarian development of worker larvae of Apis mellifera was analyzed, taking into account the trophic determination of the castes. The workers control the larval feeding in order to promote caste differentiation, which is regulated by the production and availability of this hormone. The hypothesis tested was that the action of juvenile hormone is capable of protecting or preventing the degeneration of the ovaries in worker larvae. A preparation of 1 ml of juvenile hormone dissolved in hexane at a concentration of 1 mg/ml was applied topically to 2- to 5-day-old larvae. The morphology of the ovaries was evaluated on the days following the application, until the larvae were 6 days old. The controls consisted of larvae to which 1 ml of hexane was applied, and larvae that received no treatment. The effect of juvenile hormone varied according to the age of the larvae to which it was applied, and this effect was greatest in 3-day-old larvae.

A ocorrência de reabsorção celular nos ovários de Apis mellifera (Linnaeus, 1758) foi estudada em operárias, em rainhas artificialmente impedidas de se acasalarem e em rainhas em postura utilizando microscopia de luz (LM) e eletrônica de transmissão (MET). São descritos dois tipos de estruturas morfológicas, indicativas da ocorrência de reabsorção. Estas estruturas foram denominadas por analogia com estruturas correspondentes presentes nos ovários dos vertebrados. As resultantes de reabsorção das células foliculares depois da ovulação foram denominadas corpos lúteos e as resultantes da reabsorção de todo o folículo, em qualquer fase do desenvolvimento do ovócito, de corpos atrésicos. As estruturas representativas da reabsorção têm as mesmas características morfológicas e significado fisiológico em ambas as castas. A presença de corpo lúteo indica prévia ovulação e seu número corresponde à taxa de postura da rainha e sua presença nas operárias nutridoras mostra que esta casta pode por ovos nesta fase. A incidência de corpos atrésicos em rainhas diminue com o aumento da taxa de postura indicando que a inibição da sequência normal de maturação de ovócitos no ovário é deletéria. Tanto a incidência do corpo lúteo, como do corpo atrésico pode ser influenciada por fatores ambientais.

The occurrence of cell reabsorption in the ovaries of queens in several rates of laying eggs, artificially impeded of laying, and in nurse workers, of Apis mellifera (Linnaeus, 1758), was studied with light (LM) and transmission electron microscopy (TEM). Two types of structures were described and named by analogy with vertebrates ovarian structures, as corpus luteus, when resulting from the reabsorption of the follicular cells after ovulation, and corpus atresicus when resulting from total follicular reabsorption at any oocyte developmental stage. These structures have the same morphological characteristics and physiological signification in both castes. The corpus luteus occurrence indicates ovulation and its number is correspondent to the queen's rates of oviposition. The presence of this structure in nurse workers ovarioles shows that this caste may lay eggs. The incidence of corpus atresicus in queens decay with the increasing of the oviposition indicating that the inhibition of the normal sequence of oocyte maturation in the ovaries is deleterious. Both, corpus luteus and corpus atresicus incidence may be influenced by environmental factors.

O presente estudo analisou, através de estudos ultra-estruturais a influência do tratamento com hormônio juvenil sobre as glândulas de veneno de operárias de Apis mellifera L. Para tanto, operárias recém-emergidas receberam aplicação tópica de 1µl de hormônio juvenil, na concentração de 2 µg/µl, sendo usado o hexano como veículo. Foram feitos dois controles, um que não recebeu nenhum tipo de tratamento (grupo C1) e o outro que recebeu aplicação tópica de 1 µl de hexano (grupo C2). O aspecto das células glandulares, em operárias recém-emergidas, mostra que estas não estão ainda secretando ativamente. Observa-se que alterações celulares ocorrem de acordo com a idade da operária e da região glandular considerada no controle C1. Assim, a fase de secreção mais ativa da glândula ocorre entre a emergência e os 14 dias de idade; aos 25 dias as células já perderam sua característica secretora, sendo a região distal a primeira a sofrer degeneração. Os tratamentos com hormônio juvenil e com hexano alteram a cronologia do ciclo glandular, antecipando o início da secreção e da degeneração da glândula.

The present study analyzed, the influence of the treatment with juvenile hormone on the ultrastructure of Apis mellifera L. workers' venom glands. Newly emerged workers received topical application of 1 µl of juvenile hormone diluted in hexane, in the concentration of 2 µg/µl. Two controls were used; one control received no treatment (group C1) and other received topical application of 1 µl of hexane (group C2). The aspect of the glandular cells, in not treated newly emerged workers, showed that they are not yet secreting actively. Cellular modifications happened according to the worker age and to the glandular area considered. The most active phase of the gland happened from the emergence to the 14th day. At the 25th day the cells had already lost their secretory characteristic, being the distal area the first to suffer degeneration. The treatment with juvenile hormone and hexane altered the temporal sequence of the glandular cycle, forwarding the secretory cycle and degeneration of the venom gland.

O vaso dorsal é um órgão tubular localizado na região mediano-dorsal do corpo dos insetos, abaixo do tegumento. Fez-se um estudo de microscopia de luz e eletrônica de transmissão da porção abdominal do vaso dorsal, o coração, em uma espécie de abelha indígena. Foram estudadas operárias e rainhas em diferentes idades. O coração está localizado no sinus pericárdico. A parede cardíaca é formada por fibras musculares estriadas e apresenta aberturas ou ostíolos providos de válvulas. A fibra cardíaca contém miofibrilas arranjadas irregularmente, núcleos alongados ou redondos, mitocôndrias grandes e numerosas, e depósitos de glicogênio. Em operárias e rainhas longevas, as fibras encontram-se em degeneração, evidenciada por vacúolos autofágicos, alterações mitocondriais e acúmulo de corpos mielínicos. Em conclusão, o coração de Scaptotrigona postica é semelhante ao de outros insetos estudados. As alterações encontradas estão relacionadas ao processo de envelhecimento e mantêm relação temporal com a expectativa de vida da casta.

The dorsal vessel is a tubular organ located in the mid-dorsal region of the insects body, just below the tegument. This work is a light and electron microscopy study of the abdominal part of the vessel, the heart, in a stingless bee species. Workers and queens of several ages were studied. The heart is located inside the pericardial sinus. Its wall presents openings or ostia provided with valves and is composed by special striated muscular fibers. The cardiac fiber presents myofibrils irregularly arranged, round or elongated nuclei, numerous and large mitochondria, and glycogen deposits. Old workers and queens present fibers degeneration, represented by autophagic vacuoles, mitochondria alterations and accumulation of myelinic bodies. In conclusion, the heart of Scaptotrigona postica Latreille, 1807 is similar to other insects. The changes found are related to aging progress and maintain temporal relationship with the caste life expectancy.

Foram estudadas a ocorrência e a morfologia de glândulas tegumentares presentes no abdome de fêmeas de Melissoptila richardiae. Os resultados mostram que nesta espécie, células glandulares da classe III são encontradas de duas formas: isoladas nos tergitos e esternitos III e IV e formando um aglomerado de unidades glandulares bilateralmente, entre os segmentos III e IV, os quais liberam seu produto de secreção em um reservatório originado a partir da membrana intersegmental. Os resultados sugerem que o produto secretado é lipídico e, provavelmente volátil.

The occurrence and morphology of abdominal tegumentary glands were studied in Melissoptila richardiae. The results showed the existence of class III glandular cells that may appear as isolated units in tergites and sternites III and IV or forming clustered cells in both side of the tergites III and IV. In this late case the secretion is released in reservoirs derived from the tergite intersegmental membranes. The results suggest that this last gland products are lipidic and probably volatile.

A glândula de Dufour é uma glândula acessória do aparelho reprodutivo feminino das abelhas. Nas abelhas neotropicais sem ferrão, tem sido pouca estudada sob todos os aspectos: morfológico, ontogenético e bioquímico. Na tentativa de colaborar com o conhecimento dessa glândula em abelhas sem ferrão, foi realizado um estudo da sua ocorrência, morfologia e desenvolvimento em Scaptotrigona postica Latreille. Os resultados mostraram que ela se encontra ausente nas operárias, como ocorre em muitas outras espécies desse grupo. Nas rainhas, as células glandulares parecem mais ativas nas virgens, possuindo uma desenvolvida rede de retículo endoplasmático liso tubular, grânulos de secreção e polirribossomos dispersos no citoplasma, além de apresentarem núcleos maiores do que os das células glandulares das fisogástricas. Nas rainhas fisogástricas há dois tipos de células glandulares, ambas aparentemente inativas sinteticamente. As glândulas das rainhas fisogástricas são claramente capazes de captar substâncias da hemolinfa, provavelmente lipídios, que não penetram nas células, mas passam pelos espaços intercelulares e, através da cutícula, chegam diretamente à luz da glândula. A bem desenvolvida dupla camada de lâmina basal ao redor da glândula pode atuar no processo de captação de substâncias da hemolinfa. A secreção, e conseqüentemente sua função, pode ser diferente nas duas classes de rainhas.

The Dufour gland is an accessory gland of the bee reproductive female apparatus. In neotropical stingless bees, this gland has been poorly investigated, concerning to developmental, morphological or biochemical characteristics. In order to collaborate with the knowledge of the Dufour gland in stingless bees, a study of its occurrence, morphology and development was performed in Scapatotrigona postica Latreille. The results showed that this gland is absent in workers, although it occurs in workers of many stingless bee species. The glandular cells seem to be very active in virgin queens, presenting a well developed tubular network of smooth endoplasmic reticulum, secretion granules and some polyribosomes in the glandular cell cytoplasm, besides larger nuclei than in physogastric queens. In physogastric queen glands there are two types of cells, both of them apparently not synthetically active. Nevertheless, the physogastric queen glands are clearly able to absorb substances from the haemolymph, probably lipids that do not cross the cells, but pass through the intercellular cell spaces and cuticle, and are released directly to the gland lumen. The very developed double-layered gland basal lamina may actuate in the process of substances uptaking. The secretion, and consequently its function, can be different in these two queen types.

Nas operárias das abelhas eussociais, regiões do epitélio tegumentar do abdome podem hipertrofiar-se e tornarem-se glandulares, sendo responsáveis pela produção de cera usada na contrução dos alvéolos de cria. Nos meliponíneos, epitélio com essas características também foi observado nas rainhas, porém sua função ainda continua desconhecida. Teria esse epitélio função homóloga nas operárias? As rainhas, além da função reprodutiva, desempenhariam funções na colônia até então consideradas exclusivas das operárias? Para tentar colaborar no esclarecimento dessas questões realizou-se um estudo histológico e ultra-estrutural das glândulas epiteliais tegumentares do terceiro tergito em rainhas virgens e fisogástricas de Scaptotrigona postica Latreille. Os resultados morfológicos mostraram que o epitélio glandular existe e é facilmente discernível no terceiro tergito das rainhas, sendo muito mais desenvolvido nas fisogástricas do que nas virgens. A ultra-estrutura mostrou que há diferenças no tipo de organização do retículo endoplasmático liso entre as rainhas, o que, juntamente com os resultados da histologia e morfometria, indicam poder haver diferenças funcionais desse epitélio entre elas. Como já observado na literatura, nas rainhas virgens tal epitélio pode estar envolvido na produção de cera e confecção de alvéolos de cria, porém sua função nas rainhas fisogástricas é totalmente desconhecida. A hipótese lançada no presente estudo é de que esse epitélio pode estar envolvido na produção de feromônios que irão atuar nas interações sociais da rainha poedeira.

In the eusocial worker bees, portions of the epithelium of the abdominal epidermis may become hypertrophied and glandular, and produce the wax used to build the brood alveoli. In meliponine bees, the presence of similar epithelium was already observed in the queens, but its function is still unknown. Would this epithelium have homologous function in the workers? May the queens display other functions, besides reproduction, related to those considered exclusive of workers? In order to contribute to solve these questions, a histological and ultrastructural study of the tegumentary epithelial glands of the third tergite was carried out in virgins and physogastric queens of Scaptotrigona postica Latreille. The glandular tegumentary epithelium was easily discernible in the third tergite of the queens, being much more developed in physogastric than in virgin queens. The ultrastructural differences in the smooth endoplasmic reticulum between the queens, additionally to the histological and morphometrical data, indicate that this epithelium may have functional differences between the queen types. As already known, the virgin queens may produce wax and build brood alveoli, but the role of this epithelium in physogastric queens is totally unknown. A hypothesis considered in the present study is that the epithelium may be involved in production of pheromones, which may play roles in the social interaction of the laying queen.

Workers of Melipona quadrifasciata anthidioides (Lepeletier, 1836) develop their ovaries and lay eggs, therefore the production of vitellogenin is expected. In electrophoretic profiles only fat body extracts from nurse workers and ovary extracts from newly-emerged workers show protein with molecular mass similar to vitellogenin. However, an increase in the protein content was detected in forager fat body. This increase was attributed to storage of vitellogenin or other proteins in the previous phase and not discharged into the hemolymph or to an effect of the increased titre of juvenile hormone in this phase of worker life over the fat body functioning.

As glândulas mandibulares de operárias recém-emergidas, nutridoras e campeiras, rainhas virgens e fisogástricas, e machos maduros de Scaptotrigona postica Latreille foram estudadas histológica e morfometricamente. Estas glândulas apresentam diferenças histológicas características para cada idade e/ou função do indivíduo. De acordo com a morfometria conclui-se que as glândulas mandibulares possuem um ciclo secretor determinado, no qual a secreção começa a ser produzida nas operárias recém-emergidas, e acumula-se no citoplasma e reservatório da glândula das operárias nutridoras. Nas operárias campeiras o citoplasma encontra-se bastante vacuolizado, o que foi interpretado como resultado da eliminação da secreção. Rainhas fisogástricas apresentaram as maiores áreas celulares e nucleares, ao oposto dos machos, os quais apresentaram as menores áreas.

The mandibular glands of newly-emerged, nurse and forager workers, virgin and physogastric queens, and mature males of Scaptotrigona postica Latreille were studied under histological and morphometrical analyses. These glands present differences in their histology, according to the age and/or function of the individuals. The morphometric results showed that the mandibular glands have a very markedly secretory cicle, in which the secretion starts to be produced in newly emerged workers, being accumulated into the cytoplasm and reservoir in the nurses. In forager workers the cytoplasm presents many vacuoles, which are remaining of the secretion releasing. Physogastric queens showed the largest cellular and nuclear areas. On the contrary, the males presented the smallest ones.

Utilizando cromatografia gasosa acoplada à espectrometria de massa, descrevemos as diferenças na composição química da secreção da glândula de Dufour de rainhas virgens e fisogástricas de Melipona bicolor. A secreção da glândula de rainhas virgens consiste de hidrocarbonetos, enquanto a das rainhas fisogástricas contém, além de hidrocarbonetos, uma variedade de outros compostos, tais como ésteres isobutíricos e acéticos. Tais diferenças devem indicar o estado de fecundidade da rainha e os compostos oxigenados da secreção das fisogástricas podem ajudar aumentar sua atratividade.

We describe differences in the chemical composition of the Dufour gland secretion of virgin and physogastric queens of Melipona bicolor through gas chromatography and mass spectrometry. The Dufour gland secretion of virgin queens consists only of hydrocarbons, while that of physogastric queens contains, besides these, a variety of other compounds, such as isobutyrate and acetate esters. Such differences may indicate the queen fecundity condition and the oxygenated compounds of the physogastrics secretion may help to increase their attractiveness.

The anatomy of the internal organs of the male reproductive apparatus (MRA) of adults and pupae was compared among 51 species of bees, including representatives of six families. Four different types of MRA were found. The type I is present in males of the less derived families (Colletidae, Andrenidae, and Halictidae) and is characterized by three seminiferous tubules per testis, which are almost completely enveloped by the scrotal membrane. The type II is an intermediary between types III and I and is present in Melittidae and Megachilidae, as well as in some Apidae studied, being characterized by post-vesicular deferent ducts outside the scrotal membrane and by three or four seminiferous tubules per testis, except for Apis mellifera L., which has a secondarily increased number of tubules. Type III was only found in the Apidae studied and is characterized by separately encapsulated testes and genital ducts (except for the post-vesicular deferent duct). Accessory glands are well developed, and the ejaculatory duct is thickened with fissures in its wall, which may also occur in the type II. Type IV is present exclusively in the tribe Meliponini, and is characterized by the absence of accessory glands.

A anatomia dos órgãos internos do aparelho reprodutor de machos (ARM) adultos e pupas foi comparada em 51 espécies de abelhas, incluindo representantes de seis famílias. Foram obtidos quatro tipos diferentes de ARM. O tipo I está presente em machos das famílias mais basais (Colletidae, Andrenidae e Halictidae) e é caracterizado por três túbulos seminíferos por testículo, o qual é quase totalmente envolvido pela membrana escrotal. O tipo II é um tipo intermediário entre os tipos I e III e está presente em Mellitidae e Megachilidae, como também em alguns Apidae estudados, sendo caracterizado por possuir dutos deferentes pós-vesiculares fora da membrana escrotal e possuir três ou quatro túbulos seminíferos por testículo, exceto Apis mellifera L., a qual possui secundariamente um número aumentado de túbulos. O tipo III foi achado somente nos Apidae estudados e é caracterizado por apresentar os testículos e dutos genitais (exceto o duto deferente pós-vesicular) encapsulados separadamente, as glândulas acessórias são bem desenvolvidas e o duto ejaculador é calibroso, apresentando fissuras em sua parede externa, as quais podem ocorrer também no tipo II. O tipo IV está presente exclusivamente na tribo Meliponini e é caracterizado pela ausência de glândulas acessórias.

Foram pesquisadas variações no padrão eletroforético das proteínas e da atividade da fosfatase ácida contidas em extratos do intestino médio de Apis mellifera L. durante o último estágio larval e pupação com a finalidade de estabelecer um paralelo entre os resultados e os eventos da metamorfose. Verificou-se maior variedade de bandas protéicas durante o estágio de pré-pupa e menor na pupa de olho marrom. A atividade da fosfatase ácida foi maior durante o último estágio larval e menor na pupa de olho branco. A maior variedade de bandas protéicas na pré-pupa coincide com a histólise do epitélio larval e reconstituição do epitélio pupal, enquanto a menor variabilidade na pupa de olho marrom coincide com o fim da diferenciação do intestino médio. A maior atividade fosfatásica no último estágio larval pode ocorrer em razão da sua função na histólise do epitélio.

The electrophoretic pattern of proteins and the activity of acid phosphatase were described in the midgut extracts during the post-embryonic stages of Apis mellifera L. in order to stablish a correlation with the metamorphosis events. The results show the greatest variation in proteic electrophoretical bands during prepupal stage and the smallest variation during the brown eyed pupae stage. The acid phosphatase activity was higher during the larval last instar and lower in white eyed pupae. The greatest variety of bands during the prepupal stage coincides with the histolysis of the larval midgut epithelium and the lowest variability in the brown eyed pupa coincides with the end of the midgut differentiation. In another hand, the greatest phosphatase activity in the last larval stage must reflect this enzyme actuation in the histolysis of the larval epithelium.
