A transformação genética, que consiste na introdução controlada de um gene no genoma de uma célula receptora e em sua posterior expressão, assume adicional significância, pois abre novas perspectivas ao melhoramento genético de espécies florestais, disponibilizando novos genes com características desejáveis para serem incorporados em menor espaço de tempo. Através do uso de estratégias com Agrobacterium e biobalística, já foram obtidas plantas transgênicas com maior produção de biomassa, melhor qualidade de madeira, maior resistência a determinados insetos e com tolerância a herbicidas, entre outras características de interesse. Tais exemplos demonstram a relevância dessas ferramentas para o setor florestal. Esta revisão apresenta, de forma resumida, a importância do estabelecimento de um eficiente sistema de regeneração in vitro, as principais estratégias usadas na transformação genética de espécies florestais e algumas das características agroflorestais que já foram incorporadas nessas espécies.

The genetic transformation which consists in a controlled introduction of a gene in the genome of a receive cell and its subsequent expression, assumes additional significance, giving new perspectives to genetic improvement of the forest species, extending and available new genes with desirable characteristics, to be incorporated in a smaller period of time. Through the strategies Agrobacterium and bioballistic, transgenic plants had already been obtained showing higher biomass production, better wood quality, resistance to some insects, tolerance to herbicides, among other desirable characteristics, showing the relevance of these tools to the forest industry. This revision points out in a summarized way, the importance of establishing an efficient in vitro regeneration system, the strategies used in the genetic transformations of forestry species and some agroforestry characteristics that were previously incorporated in these species.

O objetivo deste trabalho foi avaliar o efeito da idade, da época do ano e da altura de corte sobre a capacidade de rebrota de cepas de Acacia mearnsii De Wild. O trabalho foi conduzido em delineamento de blocos ao acaso e parcelas subdivididas no tempo. Em árvores de quatro e sete anos, foram avaliadas as alturas de corte de 0,15; 0,30; 0,45; 0,60; 1,2 e 2 m aos 15, 30, 45, 60, 75 e 90 dias depois do corte, nas quatro estações do ano. Obteve-se interação significativa entre a altura de corte e a época de avaliação, em ambas as idades avaliadas. As maiores médias, quanto ao número de brotações por cepa, foram constatadas a 1,2 e 2 m de altura, na idade de quatro anos. Elevados porcentuais de mortalidade foram registrados em cepas com sete anos, independentemente da altura de corte. A capacidade de rebrota em cepas de acácia-negra é estimulada, quando o corte é realizado no período entre o outono e a primavera. O verão é a estação menos adequada à rebrota de cepas dessa espécie.

The objective of this work was to evaluate the effect of age, time of the year and cut height on the resprouting capacity of Acacia mearnsii De Wild. stumps. The work was conducted in stands of four and seven years of age, in randomized blocks design and time subdivided plots. Different cut heights (0.15; 0.30; 0.45; 0.60; 1.2 and 2 m), and evaluation times (15, 30, 45, 60, 75 and 90 days after cutting), in the four seasons of the year were evaluated. A significant interaction between cut height and evaluation time was found in both ages. The higher averages due to stumps in each strain were verified in 1.2 and 2 m height, in the age of four years. High death percentages were registered in seven years-old stumps, independently of cut height. The resprouting capacity of black wattle stumps is stimulated when cut is conducted between autumn and spring; summer is the least suitable season to stumps resprouting of this specie.

ABSTRACT The response of explants in a system of tissue culture is dependent on the genotype of the material in culture. The genetic variation among and within open pollination families of a 27 years old population of Eucalyptus grandis, for proliferation of shoots and production of root in vitro was studied. The test was carried out in randomized blocks, with seven treatments (progenies), with five replications and eight explants for pots. Progenies showed highly significant differences in relation to the formation of shoots and roots. Genetic variations occurred among progenies with high variation of shoots formed. Wide genetic variations within progenies also occurred for the capacity of production of shoots. The effects of the genotype and of the enviroment were significant in the formation of shoots and roots. Through selection of families, the production of shoots and of adventitious roots can be increased to optimize the multiplication in vitro that specie.

RESUMO Aresposta de explantes em um sistema de cultura de tecidos depende do genótipo do material em cultura. Estudou-se a variação genética entre e dentro de famílias de polinização aberta de uma população de Eucalyptus grandis, aos 27 anos de idade, para proliferação de gemas e produção de raízes adventícias in vitro. O teste foi em blocos ao acaso, com sete tratamentos (progênies), em cinco repetições e oito explantes por parcela. As progênies apresentaram diferenças significativas quanto à formação de gemas e de raízes. Variações genéticas ocorreram entre progênies com elevada amplitude de gemas formadas. Amplas variações genéticas dentro de progênies também ocorreram para a capacidade de produção de gemas. Os efeitos do genótipo e do ambiente foram significativos na formação de gemas e de raízes. Através da seleção de famílias a produção de gemas e de raízes adventícias pode ser aumentada para otimizar a multiplicação in vitro dessa espécie.

RESUMO A presença de fungos pode reduzir a capacidade germinativa de um lote de sementes, causar a morte de plântulas ou transmitir doenças para plantas adultas. É necessário conhecer os agentes, as causas e as conseqüências decorrentes da contaminação por fungos patogênicos. Desse modo, o presente estudo teve como objetivo identificar os fungos associados às sementes de Acacia mearnsii De Wild, armazenadas a 5°C, por um período de 12 meses. Foram utilizadas sementes de acácia- negra oriundas de plantio comercial, aos 4 anos de idade cuja procedência é África do Sul. As sementes foram colhidas em três épocas distintas: (i) quando com frutos verdes e/ou pigmentados; (ii) quando com frutos negros e início de abertura das vagens; (iii) quando com sementes coletadas no solo, após a dispersão natural, sendo empregadas como testemunha. Os fungos associados às sementes foram: Botryodiplodia sp., Botrytis sp. (família Moniliaceae), Cladosporium sp.(família Dematiaceae), Cylindrocladium sp., Fusarium sp., Penicillium sp., Pestalotia sp., Rhizoctonia sp., Trichoderma sp. e outros fungos não identificados. De maneira geral, a autoclavagem das sementes promoveu maiores taxas de germinação e a eliminação de fungos associados. As sementes, que apresentaram maior contaminação por fungos, foram aquelas oriundas da coleta no solo. Os fungos de solo observados, que poderiam ocasionar danos em plântulas no viveiro e, simultaneamente, estarem associados à gomose em acácia-negra, foram: Botrytis sp., Cylindrocladium sp.

ABSTRACT The presence of fungi can reduce the germination of the seed lot, cause seedling death, or transmit diseases to the trees. Therefore, it is necessary to know the pathogens, the causes and consequences of the contamination of the seeds by fungi. This study has the objetive of identifyng the pathogenic fungi associated with Black Wattle seeds stored at 5ºC for twelve months. The Black Wattle seeds were harvest from a 4-year-old commercial stand originated from a South Africa seed lot. The seeds were collected in three turn: (i) harvest 1, green fruits, with some of them turning black; (ii) harvest 2, all the fruits were black, and the seed pods were beginning to open, and (iii) the seeds collected in soil, after the natural dispersion of the seeds, were used as control. The following fungi were found associated with the seeds: Botryodiplodia sp., Botrytis sp., Cladosporium sp., Cylindrocladium sp., Fusarium sp., Penicillium sp., Pestalotia sp., Rhizoctonia sp., Trichoderma sp., and others non-identified fungi. Generally, breaking the dormancy of the seeds with a heat treatment eliminated the fungi present. The highest contamination level was observed in the control (seeds harvested in soil). The soilborne fungi that can cause nursery diseases and gomosis in black wattle were Botrytis sp., Cylindrocladium sp., Fusarium sp. and Rhizoctonia sp.
