ABSTRACT: The domestic and international market is increasingly seeking for foods with reduced chemical additives. The viability using a versatile product as mechanically separate meat (CMS) to produce safety food is necessary to expand the product market. Hansenulawingei strain was selected, a yeast known as a killer toxin for poultry CMs application. The first part of this study consisted of defining the best growth condition for yeast, static or agitation, as well as the time either growth. The result obtained was the medium without agitation and 108 hours of growth at room temperature. The extract was subjected to the atomization drying process and the pulverized extract obtained was evaluated in vitro for minimum inhibitory concentration (MIC) tests for Salmonellasp, Staphylococcus positive coagulase, E. coli and mesophilic bacteria. Proximal composition, which the most relevant values to be analyzed were directed to total proteins that were 4.26g% (± 0.66) and 4.37g% (± 0.01) for agitation and unrestrained system, respectively. In the result of MIC, dry extracts were efficient in controlling the growth of all tested bacteria, including Salmonella spp. In the in situ tests, in which mechanically separated meat was raw materials to determine the antimicrobial factor action against food pathogens the concentration of 0.083g/ml of dry extract in its raw form obtained an inhibition factor as satisfactory as healing salts traditionally applied by the carneous industry. The viability of its growth and concentration for drying, can be considered a viable antimicrobial with good prospects for bio-conservative action.

RESUMO: O mercado nacional e internacional busca cada vez mais por alimentos com reduzido teor de aditivos químicos. Estudar a viabilidade de um novo método de conservação natural de um produto tão versátil como a carne mecanicamente separada (CMS) se faz necessário para expandir o mercado do produto, seguindo as tendências das demandas alimentares dos consumidores atuais. Portanto foi selecionada uma cepa de Hansenula wingei, uma levedura conhecida pela produção de toxina killer para aplicação em CMS de aves. A primeira parte deste estudo consistiu em definir qual a melhor condição de crescimento para a levedura, consistindo este em ser estático ou em agitação, bem como o tempo no qual haveria o maior pico de crescimento da mesma. O resultado obtido foi o meio sem agitação e com 108 horas de crescimento a temperatura ambiente. Na sequência o extrato foi submetido ao processo de secagem por atomização e o pó obtido foi avaliado in vitro para testes de concentração mínima inibitória (MIC) para Salmonella sp, Staphylococcus coagulase positivo, E. coli e Mesofilos aeróbicos e definição de sua composição proximal, a qual os valores mais relevantes a serem analisados eram direcionados para proteínas totais que foram 4.26g% (±0.66) e 4.37g% (±0.01) para sistema com agitação e sem agitação, respectivamente. Como resultado de MIC, os extratos secos mostraram-se eficientes no controle do crescimento de todas as bactérias testadas, inclusive Salmonella spp. Nos testes in situ, no qual a carne mecanicamente separada foi a matéria prima testada para determinar a ação fator antimicrobiano contra patógenos de alimentos a concentração de 0.083g/ml do extrato seco, em sua forma bruta, obteve um fator de inibição tão satisfatório quanto aos sais de cura aplicados tradicionalmente pela indústria cárnea e pela viabilidade de seu crescimento e concentração por secagem, pode ser considerado um antimicrobiano viável e com boas perspectivas para ação bioconservante.

Os consumidores costumam associar os produtos cárneos processados a uma imagem negativa em função dos teores de gordura saturada, sódio e aditivos químicos que apresentam. O trabalho teve por objetivo estudar a influência da adição de farinha de semente de linhaça dourada (Linum usitatissimum L.) como substituto de gordura suína e da utilização de sal com teor reduzido de sódio nas características físicas, químicas, microbiológicas e sensoriais de hambúrgueres de carne bovina. Visando tornar o hambúrguer mais saudável, seis formulações foram desenvolvidas mudando as concentrações de sal light e de farinha de linhaça. Os resultados demonstraram que a adição de 10,0% de farinha de semente de linhaça dourada em substituição a 100,0% do toucinho contribuiu para o aumento (p < 0,05) do teor de proteínas. A adição de 5,0% de farinha de linhaça dourada em substituição a 50,0% do toucinho refletiu em boas notas para todos os atributos sensoriais avaliados. Houve aumento (p < 0,05) dos percentuais de ômega-3 e cinzas, e redução (p < 0,05) dos lipídios totais e valor calórico, independente dos níveis de adição da farinha. A substituição de toucinho por farinha de semente de linhaça contribuiu para o menor encolhimento e melhor rendimento, bem como para maior retenção de umidade e de gordura dos hambúrgueres. O sal light não interferiu negativamente nas características microbiológicas dos hambúrgueres e quando associado com a adição de 5,0% de farinha de linhaça contribuiu para melhor (p < 0,05) sabor e aroma. Os resultados sugeriram que a adição de 5,0% de farinha de semente de linhaça dourada como substituto de 50,0% da gordura suína, bem como a redução de 60,1% do sódio em hambúrgueres de carne bovina, se caracterizou alternativa viável do ponto de vista nutricional e sensorial, contribuindo ainda para o maior rendimento do produto após o cozimento.

Consumers tend to have a negative image of processed meat products, due to their contents of saturated fat, sodium and chemical additives. This work aimed to study the influence of adding golden flaxseed flour (Linum usitatissimum L.) as a substitute for pork fat as well as using reduced sodium salt, on the physical, chemical, microbiological and sensory profiles of beef burgers. With a view to making the burgers healthier, six formulations were developed with different concentrations of light salt and flaxseed flour. The results showed that the total replacement of pork fat by 10.0% of golden flaxseed flour significantly increased (p<0.05) the protein content. The burgers with the addition of 5.0% golden flaxseed flour to replace 50.0% of the pork fat presented good scores for all the sensory attributes evaluated. Independent of the percentage of flour added to the burgers, the omega-3 and ash contents increased (p<0.05), while the total lipids, caloric value and fat content of the dry matter showed a significant decrease (p<0.05). The replacement of pork fat by flaxseed flour contributed to reduced shrinkage and a higher yield, as well as burgers showing greater moisture and fat retention. The use of light salt did not affect the antimicrobial potential and when combined with 5.0% of flaxseed flour significantly improved the flavour and taste (p < 0.05). The results suggested that the addition of 5.0% golden flaxseed flour as a substitute for 50.0% saturated fat and a sodium reduction of 60.1% in the burgers was a viable alternative for the nutritional and sensorial attributes, as well as providing a higher yield after cooking.

Considerando-se a evidente busca por alimentos de rápido e fácil preparo, as comunidades industrial e científica vêm investindo no desenvolvimento de novos produtos que, além de atenderem a essa demanda, possam oferecer benefícios à saúde do consumidor. Dentre os alimentos que não demandam muito tempo para o preparo no domicílio, o hambúrguer merece destaque, em função do seu elevado consumo. No entanto, em razão de conter gordura saturada e por ser submetido a processo de fritura, o consumo demasiado desse produto pode ser prejudicial à saúde humana, podendo causar doenças crônicas, entre as quais, a obesidade e a hipertensão. Estudos têm demonstrado a possibilidade de substituição de ingredientes na formulação de hambúrgueres, com a intenção de incorporar substâncias com propriedades funcionais; portanto, substâncias que possam contribuir para a saúde e o bem-estar dos consumidores. Nesse contexto, esta revisão aborda questões referentes ao hambúrguer, no sentido de conceituar o produto e, à luz da literatura, oferecer alternativas possíveis e que têm sido estudadas para tornar esse alimento mais saudável, com foco na substituição de gordura animal por fibras e na redução do teor de cloreto de sódio.

Considering the evident search for quick and easy food preparation, industrial and scientific communities have been investing in developing new products that fulfill this demand and may offer health benefits to consumer. Among foods that do not require much time for preparation at home, the burger deserves attention due to its high consumption. However, due to its saturated fat contents and being subjected to frying process, the excessive consumption of this product may be harmful to human health and may cause chronic diseases, including obesity and hypertension. Studies have demonstrated the possibility of ingredients substitution in the formulation intending to incorporate substances with functional properties, so that they can contribute to consumers'health and well-being. In this context, this paper describes issues related to the burger, to conceptualize the product and, in the light of the literature, to offer studied alternatives to make it healthier, focusing on replacing animal fat by fibers and on sodium chloride content reduction.

This work evaluated the antagonism of killer positive yeast strains (isolated from 11 samples of different frozen fruit pulps) against the strains of Penicillium expansum and Aspergillus ochraceus. Of the total 41 killer yeasts tested in YM agar, 19 showed antibiosis against P. expansum and A. ochraceus, with inhibition zone ranging from 10 to 18 mm and 10 to 19 mm, respectively. In the following step, the extracellular activity of Kluyveromyces sp. FP4(13) was tested performing the assay in YM broth. The antifungal activity of Kluyveromyces sp. FP4(13) cell-free culture supernatant (25ºC/96 h) was more effective against the conidia germination, showing inhibition rates of 93.33 and 86.44% for P. expansum and A. ochraceus, respectively. The micelial growth inhibition was 28.45 and 21.0%, respectively. The antagonism showed by the selected yeasts could be used as a promising alternative tool to reduce and control the postharvest fungal spoilage of the fruits. However, further studies should be carried out in order to better elucidate the role of innocuous characters in antagonistic microorganisms, as well as the purification and characterization of new killer toxins.

Penicillium expansum é o agente causador da doença em maçã conhecida como mofo azul. O efeito inibitório da capsaicina derivada dos frutos Capsicum spp. e da levedura Hansenula wingei foi avaliado através de ensaios in vitro e in vivo em maçã. A concentração inibitória mínima da capsaicina de 122,16 µg mL-1 foi determinada usando microdiluição. A capsaicina não mostrou capacidade em reduzir a incidência do mofo azul na maçã. Entretanto, um retardo no crescimento do fungo foi observado nos 14 primeiros dias dos ensaios in vivo. Também descrevemos o efeito da levedura Hansenula wingei AM2(-2) isolada e em combinação com tiabendazol em baixa dosagem (40 µg mL-1) no controle da doença de maçãs por P. expansum.Hansenula wingei AM2(-2), em combinação com baixa dosagem de tiabendazol (10% da recomendada pelo fabricante), apresentou 100% de eficácia após 7 dias de estocagem a 21 ºC, seguido de 80,45% e 72,13% de eficácia após 14 e 21 dias, respectivamente(p > 0,05). Estes resultados contribuem com as novas opções de emprego de agentes antifúngicos contra Penicillium expansum.

Penicillium expansum is the causative agent of apple blue mold. The inhibitory effects of the capsaicin derived from Capsicum spp. fruits and yeast Hansenula wingei against P. expansum were evaluated in an in vitro and in in vivo assay using Fuji apples. The minimum inhibitory concentration of capsaicin determined using the broth micro-dilution method was 122.16 µg mL-1. Capsaicin did not reduce blue mold incidence in apples. However, it was able to delay fungal growth in the first 14 days of the in vivo assay. The in vivo effect of the yeast Hansenula wingei AM2(-2), alone and combined with thiabendazole at low dosage (40 µg mL-1), on the incidence of apple diseases caused by P. expansum was also described. H. wingei AM2(-2) combined with a low fungicide dosage (10% of the dosage recommended by the manufacturer) showed the best efficacy (100%) up to 7 days of storage at 21 ºC, later showing a non-statistically different decrease (p > 0.05) after 14 (80.45%) and 21 days (72.13%), respectively. These results contribute providing new options for using antifungal agents against Penicillium expansum.

Considerando o antagonismo e degradação de patulina detectados em Pichia membranifaciens e Sporobolomyces roseus no estudo preliminar, este trabalho avaliou o efeito antagônico de Pichia ohmeri 158 no desenvolvimento de Penicillium expansum e a degradação de patulina "in vitro". O sobrenadante do cultivo de P. ohmeri 158 inibiu 66,17% do desenvolvimento micelial, indicando antibiose relacionada ao fator killer. A concentração inicial de patulina (223 µg) na presença de células íntegras de P. ohmeri foi reduzida em mais de 83% após dois dias de incubação a 25ºC e superior a 99% após 5 dias, com níveis indetectáveis no 15º dia. O decréscimo do pH 4,0 inicial para pH 3,3 sugeriu que a eliminação de patulina é um processo ativo e uma conseqüência do metabolismo da levedura. Os resultados obtidos concluem que P. ohmeri 158 é uma alternativa promissora na inibição do desenvolvimento de P. expansum e na degradação de patulina.

Taking into account the preliminary antagonistic/biodegradation property showed by Pichia membranifaciens and Sporobolomyces roseus, which decreased the initial patulin concentration of 588.4 to 290.0 µg/mL, ability of P. ohmeri 158 in biocontrol against Penicillium expansum and patulin decrease in vitro was performed. The culture supernatant of P. ohmeri 158 was effective against 66.17% micelial growth, indicating antibiosis related with the killer phenomenon. The initial patulin concentration of 223 µg in the presence of P. ohmeri 158 cells was decreased over 83% of the original concentration, when incubated at 25ºC/2 days and > 99% after 5 days incubation time, with undetectable patulin level after 15 days. The initial pH 4.0 decreased to pH 3.3 along 15 days experiment, suggesting that patulin decrease was an active process and a consequence of yeast metabolism. The results suggested that P. ohmeri 158 could be a promising alternative for the inhibition of P. expansum growth and patulin degradation.
