RESUMO: O mormo é uma enfermidade infectocontagiosa e frequentemente letal, aguda ou crônica, caracterizada por lesões respiratórias, linfáticas e cutâneas em equídeos, porém de caráter zoonótico. Objetivou-se acompanhar o desenvolvimento dos sinais clínicos e de lesões em animais infectados naturalmente por B. mallei em diferentes propriedades de criação de equídeos, e isolar e caracterizar a bactéria causadora da doença. Foram estudados dois focos oficiais para o mormo no estado de Alagoas. Realizou-se o acompanhamento clinico e imunológico dos animais nos focos, eutanásia e exame anatomopatológico dos animais, além de colheita de material para diagnóstico através de técnicas de isolamento, fenotípicas, moleculares e prova biológica. Neste estudo não se observou um padrão clínico para o diagnóstico do mormo em animais infectados com a mesma cepa de B. mallei. O diagnóstico precoce da infecção deve ser realizado por meio de testes imunológicos de elevada sensibilidade e especificidade. Nos casos assintomáticos e positivos nos testes oficiais é importante a realização do exame anatomopatológico, microbiológico e molecular.

ABSTRACT: Glanders is an infectious and often lethal zoonotic disease of equines caused by the bacterium, Burkholderia mallei. This condition is characterized by respiratory, lymphatic, and cutaneous lesions. In this study, we monitored the development of clinical symptoms in animals naturally infected with B. mallei across different equine breeds and also isolated and characterized the disease-causing bacteria. We studied two official glanders outbreaks in the state of Alagoas, Brazil. During the outbreaks, we performed clinical and immunological follow-up of the animals, as well as euthanasia and anatomopathological examination. We also collected diagnostic materials for isolative, phenotypic, molecular, and biological testing. We did not observe any clinical patterns of glanders among animals infected with the same strain of B. mallei. Based on our results, we suggest that early diagnosis of infection should be made through highly sensitive and specific immunoassays. In asymptomatic but positive test cases, we confirmed the importance of conducting pathological, microbiological, and molecular examinations.

RESUMO: Objetivando conhecer a situação da tuberculose ovina no Maranhão, avaliaram-se 343 ovinos da região sudoeste do estado, 247 fêmeas e 96 machos, com idade acima de 6 meses, por meio do teste cervical comparativo (TCC), com inoculação de tuberculinas na região cervical direita. Cranialmente, injetou-se derivado proteico purificado (PPD) aviário; e caudalmente, PPD bovino, em doses individuais de 0,1 mL. Todos os animais apresentaram resultado negativo para tuberculose, entretanto, 65 ovinos apresentaram reação à tuberculina bovina superior à aviária. Para investigar a possibilidade de baixa sensibilização ou anergia em animais com tuberculose avançada, foi necropsiado um ovino caquético, idade acima de quatro anos, com resultado negativo ao TCC, porém, com reação à tuberculina aviária e linfonodos aumentados com consistência endurecida. Os resultados indicaram infecções por C. pseudotuberculosis, Actinomyces pyogenes, Staphylococcus aureus e Mycobacterium sp. A cultura e o isolamento resultaram em Pseudomonas aeruginosas e Bacillus sp. Não foram visualizadas estruturas bacterianas compatíveis com bacilo álcool ácido resistente (BAAR). Os resultados confirmam ausência de animais reagentes ao M. bovis, pressupondo inexistência da tuberculose em ovinos na região estudada.

ABSTRACT: In order to know the situation of ovine tuberculosis in Maranhão, Brazil, we evaluated 343 sheep in the southwest region of the state, of both sexes (247 females and 96 males), aged over six months, through the comparative cervical test (TCC), with tuberculin inoculation in the right cervical region. Cranially, avian purified protein derivate (PPD) was injected; and caudally, beef PPD was injected, in individual doses of 0.1 mL. All animals were negative for tuberculosis; however, 65 sheep had higher reactions to bovine tuberculin than to the avian one. To investigate the possibility of low awareness or anergy in animals with advanced tuberculosis, one cachectic sheep was necropsied, aged four years, with negative results for TCC, but with reaction to avian tuberculin and enlarged hardened lymph nodes. The results indicated C. pseudotuberculosis, Actinomyces pyogenes, Staphylococcus aureus and Mycobacterium sp infections. Culture and isolation resulted in Pseudomonas aeruginosa and Bacillus sp. No bacterial structures compatible with acid-fast bacteria were found. The results confirmed the absence of animals reactive to M. bovis, assuming no tuberculosis in sheep in the study area.

RESUMO: Maedi-Visna (MV) é uma enfermidade causada por lentivírus de pequenos ruminantes com evolução crônica e em grande parte dos casos sinais clínicos inaparentes. O diagnóstico da doença é baseado em sinais clínicos e dados epidemiológicos, sendo a imunodifusão em gel de ágar (IDGA) o método padrão para a detecção sorológica de anticorpos contra o lentivírus. Sabendo que o estado do Tocantins possui potencial para o desenvolvimento da ovinocultura e que grande parte dos produtores de Colinas do Tocantins, no referido estado, possui interesse em estabelecer criação racional, esta pesquisa teve por objetivo a realização de um estudo acerca da soroprevalência da doença. Foram coletadas 369 amostras de sangue de ovinos, independentemente de raça, sexo e idade, de diferentes propriedades rurais do município para diagnóstico de MV utilizando a técnica de IDGA. Após as análises laboratoriais, para avaliação dos resultados no tocante às categorias, foi utilizado o teste exato de Fisher e também foi calculado o odds ratio , com intervalo de confiança de 95% para verificação da idade como possível fator de risco ou de proteção. Constatou-se que 6 animais (1,62%) se apresentaram positivos no IDGA. Diante desses resultados, foi possível concluir que a frequência de ovinos soropositivos no município é baixa.

ABSTRACT: Maedi-Visna is a disease caused by a small ruminant lentivirus, with a chronic evolution and, in most cases, unapparent signs. Its diagnosis is based on clinical signs and epidemiological data, with the agar gel immunodiffusion (AGID) test being the classical method for detecting antibodies against lentiviruses. Considering that the state of Tocantins has the potential to develop sheep breeding and that the majority of producers from Colinas do Tocantins city has shown an interest in establishing a rational breeding, this study aimed to determine the prevalence of such disease in that region. A total of 369 blood samples was drawn from sheep bred in several farms of that town, regardless of breed, gender, and age. Every sample underwent an AGID to test for Maedi-Visna. After the laboratory analyses were concluded, the Fisher's exact test was used to evaluate the results against the categories. Also, the odds ratio, with 95% confidence interval, was calculated to check whether age played a role as either a risk or protective factor in these results. It was found that six animals (1.62%) were positive in AGID, therefore concluding that the frequency of seropositivity in that region is low.

RESUMO: A soroprevalência da infecção por lentivírus de pequenos ruminantes (LVPR) foi determinada em amostras de soros sanguíneos de caprinos e ovinos de aptidão cárnea provenientes de abatedouros de dez municípios do estado de Pernambuco, Brasil. O diagnóstico sorológico ocorreu por meio da imunodifusão em gel de agarose (micro-IDGA) com antígenos dos vírus artrite encefalite caprina (CAE)/Maedi-Visna. Entre as 369 amostras de caprinos, 7(1,89%) (0,8-3,9%) eram soropositivas, e, entre as 383 de ovinos, 1 (0,26%) (0,0-1,4%) estava infectada. Os 7 caprinos soropositivos procederam dos abatedouros públicos dos municípios de Gravatá (n=2), Sertânia (n=4) e Timbaúba (n=1), e o ovino soropositivo veio do abatedouro público de Serra Talhada. A soroprevalência da infecção por LVPR em pequenos ruminantes oriundos de abatedouros do estado de Pernambuco, de 1,06% (8/752), é considerada baixa.

ABSTRACT: The prevalence of lentivirus infection of small ruminants (LVPR) was determined in samples of serum from goats and sheep in slaughterhouses from ten districts of Pernambuco State. The serological test was used in agarose gel immunodiffusion (AGID) with antigen caprine arthritis and encephalitis virus (CAE)/Maedi Visna virus. Among the 369 blood serum samples of goats examined, seven (1.89%) (0.8-3.9%) were seropositive, and among the 383 sheep samples examined, just one (0.26%) (0.0-1.4%) was infected. The seven seropositive goats came from public slaughterhouses from Gravatá (n=2), Sertânia (n=4) and Timbaúba (n=1), and the soropositive sheep was from a public slaughterhouse of Serra Talhada. The soroprevalence of LVPR infection in small ruminants from Pernambuco's slaughterhouses, of 1.06% (8/752), is considered low.

A particularidade da secreção láctea caprina, do tipo apócrina, diferente da secreção merócrina da vaca, leva a erros de interpretação durante a realização de técnicas de avaliação da celularidade do leite de fêmeas desta espécie. Portanto, o presente trabalho teve o objetivo de determinar a contagem de células somáticas pelo método indireto California Mastitis Test (CMT), e por métodos diretos, incluindo a contagem por citometria de fluxo e a contagem microscópica direta, através da coloração de verde de metil e pironina-Y, além de comparar os métodos de contagem celular. Foram analisadas 102 amostras de 51 fêmeas caprinas, das raças Saanen, Parda Alpina e Toggenburg, criadas no Estado de São Paulo. Os animais foram categorizados segundo a fase da lactação, exame físico da glândula mamária e exame do leite. As amostras foram colhidas, após a realização do exame Califórnia Mastitis Test, em duas alíquotas, uma destinada à contagem celular automática e a outra, a contagem microscópica direta, utilizando-se o corante verde de metil e pironina- Y. De acordo com os diferentes escores do CMT, observou- se 74,5% de amostras negativas, 8,8% de amostras com escore traços, 8,8% de amostras ligeiramente positivas (+), 6,8% de amostras fracamente positivas (++) e 0,9% de amostras fortemente positivas (+++). Os valores medianos das contagens de células somáticas presentes no leite de cabras, avaliadas através de contador automático e microscopia direta, e analisadas de acordo com os diferentes escores do CMT, foram, respectivamente, 181.000, 578.000, 628.000, 1.421.500 e 5.542.000 células/mL de leite e 74.991, 271.396, 71.420, 640.995 e 5.049.394 células/ mL de leite, nos escores negativo, traços, +, ++ e +++. Os valores medianos obtidos através da contagem de células somáticas pelo método automático e microscópico direto, de acordo com as fases de lactação foram de 159.500, 508.000 e 277.500 células/mL de leite, e 62.493, 89.275 e 146.411. A correlação obtida entre a contagem celular automática e microscópica direta foi de 88%. A partir dos resultados observados pode-se concluir que existe diferença na contagem celular determinada através do método automático e microscópico sendo este último o mais adequado para a determinação da celularidade no leite de cabras.

The particular apocrine secretion of goat milk different from the merocrine one observed in cows, may lead to errors in interpreting cellularity evaluations in the milk of this species. Thus, the objective of the present trial was to determine Somatic Cell Counts by means of one indirect methods, the California Mastitis Test (CMT), and direct methods, flow cytometry and direct microscopic count using methyl green-pyronine-Y stain, beyond comparing the methods of cellular counting. A total of 102 samples from 51 Saanen, Brown Alpine and Toggenburg female goats, bred in the state of São Paulo, were analyzed. Goats were separated in groups according to the phase of lactation and to physical examination of the mammary gland, and milk examination. Samples were divided into two aliquots, and were collected after California Mastitis Test evaluation. One aliquot was used in automatic cell counts, and the other, in direct microscopic count using methyl green-pyronine-Y stain. CMT results were as follows: 74.5% of the samples were negative, 8.8% yielded traces, 8.8% were weak positive (1), 6.8% were distinct positive (2) and 0.9% were strong positive (3). Medians of somatic cell counts in goat milk as evaluated by means automatic cell counter and direct microscopy, and grouped according to the different CMT scores, were as follows: 181,000, 578,000, 628,000, 1,421,500, and 5,542,000 cells/mL of milk and 74,991, 271,396, 71,420, 640,995, and 5,049,394 cells/mL of milk in scores negative, traces, 1, 2 and 3, respectively. Medians obtained in automatic cell counts and direct microscopic counts, grouped according to the phase of lactation were 159,500; 508,000; and 277,500 cells/mL of milk and 62,493; 89,275; and 146,411 cells/ml of milk, respectively. The correlation between the automatic and microscopic methods for somatic cell counts was 88%. Based on the results obtained, it could be concluded that there were differences between the automatic and microscopic methods for somatic cell counts, being this most adequate for the determination of the celularidade in the cellularity of goat milk.

Dois surtos de agalaxia contagiosa causada por Mycoplasma agalactiae são descritos no Estado da Paraíba, região Nordeste do Brasil. A doença caracterizou-se por mastite, agalaxia e poliartrite em cabras e poliartrite e cerato-conjuntivite em cabritos e cordeiros. Febre e anorexia também foram observadas. A morbidade variou de 26,1% a 100% nas cabras, 36,5% a 100% em cabritos e 49,0% em cordeiros. Na primeira fazenda, 14,3% das cabras em lactação e 6,4% dos cabritos morreram ou foram sacrificados. Na outra propriedade, 3,3% dos caprinos adultos, 36,5% dos cabritos e 22,9% dos cordeiros morreram e outros 84 caprinos foram sacrificados para controle da doença. M. agalactiae foi isolado a partir de leite, líquido articular, suabe nasal e lavado do conduto auditivo externo. Colônias características de Mycoplasma e que não fermentaram a glicose e arginina foram observadas. A identificação de M. agalactiae foi realizada por imunoperoxidase indireta e PCR. Sendo assim, M. agalactiae é descrito pela primeira no Brasil, mas a origem da infecção permanece desconhecida.

Two outbreaks of contagious agalactia by Mycoplasma agalactiae occurred in Paraíba State, Northeastern Region of Brazil are reported. The disease was characterized by mastitis, agalactia and polyarthritis in does and polyarthritis and conjunctivitis in kids and lambs. Fever and anorexia were also observed. Morbidy was from 26.1% to 100% in does, 36.5 to 100% in kids and 49% in lambs. In one farm 14.3% of the lactating goats and 6.4% of the kids died or were euthanized. In the other, 3.3% of the does, 36.5% of the kids and 22.9% of the lambs died and 84 affected goats were euthanized to control the disease. M. agalactiae was isolated from milk, joint exudates, nasal swabs and ear washings. The colonies were characteristic of Mycoplasma and the agent did not ferment both glucose and arginin. It was typed as Mycoplasma agalactiae by immunoperoxidase and PCR. This is the first report of M. agalactiae infection in Brazil, but the source of the infection remains unknown.

Descrevem-se 24 surtos de raiva em bovinos (25 casos), 4 em eqüinos (5 casos), 2 em caprinos (2 casos) e 2 em ovinos (4 casos). Todos os surtos ocorreram na Paraíba, exceto um em eqüinos que ocorreu no Rio Grande do Norte. Todos os surtos, com a exceção de um em ovinos, foram transmitidos provavelmente por morcegos hematófagos; no entanto, não se descarta a possibilidade de transmissão por raposas (Dusicyon vetulus). Os sinais clínicos foram representativos da localização das lesões no sistema nervoso central (SNC). Em bovinos os sinais eram, principalmente, da forma paralítica, causados por lesões da medula, tronco encefálico e cerebelo; mas alguns animais apresentaram depressão, excitação e outros sinais associados a lesões cerebrais. Três dos 5 eqüinos apresentaram sinais de lesões cerebrais e 2 apresentaram a forma paralítica. De 4 ovinos e 2 caprinos, 4 apresentaram a forma paralítica, mas em um caprino e um ovino os sinais foram predominantemente de lesões cerebrais. Todos os animais afetados, exceto um caprino, tiveram um curso clínico de 2-8 dias. As únicas lesões macros-cópicas observadas foram a dilatação da bexiga em 4 bovinos e a dilatação da ampola retal em 2. Dois cavalos apresentaram lesões da pele causadas por traumatismos. As lesões histológicas foram de encefalomielite e meningite difusa não supurativa. Nos eqüinos e em um caprino com evolução clínica de 35 dias as lesões foram mais severas, observando-se necrose neuronal, neuronofagia e presença de esferóides axonais. Corpúsculos de Negri foram observados em 87% (20/23) dos bovinos examinados e em 83% (5/6) dos ovinos e caprinos. Nessas 3 espécies os corpúsculos foram mais freqüentes no cerebelo, mas ocorreram também no tronco encefálico, medula e cérebro. Em cavalos, corpúsculos de Negri foram menos numerosos, sendo observados somente no córtex de um animal e no córtex e hipocampo em outro. No gânglio trigeminal, as lesões histológicas e os corpúsculos de Negri foram menos freqüentes que no SNC. Esses resultados mostram que na raiva dos herbívoros os sinais clínicos e a distribuição das lesões histológicas do SNC são variáveis, e que para o correto diagnóstico da enfermidade são necessários um bom exame clínico e o estudo histológico das diferentes regiões do SNC. Sugere-se, também, que perante resultados negativos nos testes de imunofluo-rescência e inoculação em camundongos, estes devam ser repetidos com amostras de diferentes regiões do SNC. Utilizando-se dados sobre a freqüência de doenças, de 4 laboratórios de diagnóstico, foi realizada uma estimativa das mortes de bovinos causadas pela raiva, anualmente, em 3 Estados. Na Paraíba, com uma população de 918.262 bovinos, o número de mortes é estimado em 8.609 cabeças por ano. No Mato Grosso do Sul, com uma população de 23 milhões de bovinos, as perdas por raiva são estimadas em 149.500 cabeças e, no Rio Grande do Sul, com uma população de 13 milhões de bovinos, as mortes são estimadas em 13.000 a 16.250 animais por ano. Se estas estimativas forem extrapoladas para todo o Brasil, com 195 milhões de bovinos, as mortes podem ser estimadas em 842.688 cabeças por ano.

Twenty four outbreaks of rabies in cattle, 4 in horses, 2 in sheep, and 2 in goats are reported in northeastern Brazil. All outbreaks occurred in the state of Paraíba, except one in horses that occurred in the state of Rio Grande do Norte. All outbreaks, except one in sheep, were probably transmitted by vampire-bats, but the transmission by foxes (Dusicyon vetulus) is also possible. Clinical signs were characteristic for distribution of the lesions in the central nervous system (CNS). In cattle, signs were mainly of the paralytic form of rabies, caused by lesions on the spinal cord, brain stem and cerebellum; but some animals showed also depression, excitation and other signs due to cerebral lesions. In 3 out of 5 horses, the main clinical signs were due to lesions in the cerebrum, and 2 had the paralytic form. From 4 sheep and 2 goats affected, 4 showed clinical signs of the paralytic form; but in 1 goat and 1 sheep the main clinical signs were caused by cerebral lesions. All affected animals, except 1 goat, had a clinical manifestation period of 2-8 days. The only gross lesions were distention of the urinary bladder in 4 cattle and distention of the rectum in 2 others. Two horses had skin lesions due to traumatic injury. Histologic lesions were diffuse non-suppurative encephalomyelitis and meningitis. In the horses, and in one goat with a clinical manifestation period of 35 days, the lesions were more severe, with neuronal necrosis, neuronophagia, and presence of axonal spheroids. Negri bodies were found in 87% (20/23) of the cattle cases examined histologically. In small ruminants Negri bodies were found in 83% (5/6) of the cases. In sheep, goats and cattle, Negri bodies were more frequent in the cerebellum, but they were found also in brain stem, spinal cord and cerebrum. In horses, Negri bodies were found in small amounts only in the cortex of one animal, and in the cortex and hippocampus of another. Histologic lesions and Negri bodies in the trigeminal ganglia were less frequent than in the CNS. These results show that in rabies of herbivores, clinical signs and distribution of lesions in the CNS are variable, so that for the diagnosis and adequate clinical evaluation and the histologic study of different areas of the CNS are necessary. This also suggests that when the fluorescent antibody test and mouse inoculation test are negative, they should be repeated with samples from different areas of the brain and spinal cord. Frequency data of diseases from 4 diagnostic laboratories were used to estimate cattle deaths due to rabies in 3 Brazilian states. In Paraíba, with a population of 918,262 cattle, the annual death rate is estimated in 8,609 heads. In Mato Grosso do Sul, with a population of 23 millions cattle, deaths caused by rabies are estimated in 149,500 heads, and in Rio Grande do Sul, with a cattle population of 13 millions, cattle deaths due to rabies are estimated in 13,000 to 16,250 heads. If these data are used to estimate cattle losses in Brazil, with a cattle population of 195 millions, it can be estimated that 842,688 deaths are caused annually by rabies.

Os lentivírus de pequenos ruminantes (SRLV), cujos protótipos são os vírus da Artrite-Encefalite Caprina (CAEV) e Maedi-Visna, são patógenos amplamente distribuidos, os quais causam doenças degenerativas progressivas lentas em caprinos e ovinos, determinando importantes perdas econômicas. Estes vírus causam infecções persistentes com período de incubação longo e causam inflamatórias e degenerativas. As lesões são induzidas em tecidos específicos do hospedeiro como articulações, pulmões, CNS e glandulas mamárias devido à replicação viral em células da linhagem monocítico-fagocitária que são as principais células-alvo. A infecção ocorre principalmente durante os primeiros meses de vida, através da ingestão de vírus no leite ou colostro de cabras ou ovelhas infectadas. A indução da resposta imunológica é variável e não protege contra a infecção. O diagnóstico é baseado primariamente na detecção de anticorpos para SRLV, geralmente por imunodifusão em gel de agar (AGID) e enzyme linked immunosorbent assay (ELISA). O diagnóstico e separação ou descarte dos animais soropositivos associado ao uso de certas práticas de manejo, especialmente das crias, são os principais meios implementados para prevenir a disseminação de SRLV, uma vez que ainda não existe vacina contra o vírus. As estratégias adotadas pelos SRLV para enfrentar o sistema imune dificultam o diagnóstico da infecção, controle ou prevenção da disseminação de SRLV. Esta revisão apresenta alguns aspectos das lentivíroses de pequenos ruminantes baseadas em estudos filogenéticos de amostras isoladas, aspectos clínicos e imunopatológicos.

Small ruminant lentiviruses (SRLV), whose prototypes are Caprine Arthritis-Encephalitis virus (CAEV) and Maedi-Visna virus, are the causative agents of slow progressive degenerative diseases of goats and sheep (infected animals), responsible for significant economic losses. These viruses cause persistent infections with long periods of incubation and induce inflammatory and degenerative lesions. The lesions are induced in target organs of the host such as joints, CNS, lungs and mammary glands due to viral replication in cells of the monocyte/macrophage lineage which is the main target cell. Infections occur particularly in the young and are acquired through ingestion of virus in milk or colostrum from infected does or ewes. The induction of immune response is variable and does not protect against the infection. Diagnosis is primarily based on the presence of SRLV antibodies usually detected by agar gel immunodiffusion (AGID) or enzyme linked immunosorbent assays (ELISA). As no vaccine is available, most often employed schemes to prevent spread of SRLV are based on segregation or/and culling of positive animals associated with management practices, especially the offspring. The strategies of SRLV for dealing with the immune system make difficult to accomplish diagnosis of infection, control or prevention of the viral spread. This review shows aspects of SRLV based on their phylogenetic studies of fields isolates, clinical, and immuno-pathological features.
