Abstract Angular leaf spot (ALS) and powdery mildew (PWM) are two important fungi diseases causing significant yield losses in common beans. In this study, a new genetic linkage map was constructed using single sequence repeats (SSRs) and single nucleotide polymorphisms (SNPs), in a segregating population derived from the AND 277 x SEA 5 cross, with 105 recombinant inbred lines. Phenotypic evaluations were performed in the greenhouse to identify quantitative trait loci (QTLs) associated with resistance by means of the composite interval mapping analysis. Four QTLs were identified for ALS resistance. The QTL ALS11AS, linked on the SNP BAR 5054, mapped on chromosome Pv11, showed the greatest effect (R2 = 26.5%) on ALS phenotypic variance. For PWM resistance, two QTLs were detected, PWM2AS and PWM11AS, on Pv2 and Pv11, explaining 7% and 66% of the phenotypic variation, respectively. Both QTLs on Pv11 were mapped on the same genomic region, suggesting that it is a pleiotropic region. The present study resulted in the identification of new markers closely linked to ALS and PWM QTLs, which can be used for marker-assisted selection, fine mapping and positional cloning.

A infecção de grãos de milho por Fusarium verticillioides, agente causal da podridão da espiga, pode resultar na produção de micotoxinas do grupo das fumonisinas. A resistência genética é a forma de controle mais eficiente dessa enfermidade. Assim, o objetivo do trabalho foi buscar fontes de resistência em linhagens de milho tropical à F. verticillioides e à produção de fumonisinas. Seis linhagens tropicais de milho, três, pré-classificadas como resistentes e três, pré-classificadas como suscetíveis à F. verticillioides, foram submetidas à inoculação do patógeno e posteriormente, avaliadas quanto à severidade da podridão de espiga, incidência de grãos sintomáticos e concentração de fumonisinas. Os resultados mostraram que as linhagens R1 e R3 apresentaram alta resistência à infecção do patógeno. No entanto, apenas a R3 foi resistente ao acúmulo de fumonisinas. Dessa forma, sugere-se que a ausência de relação entre intensidade da doença e níveis de fumonisinas seja fator inerente desse patossistema. Assim, não é possível assegurar que grãos assintomáticos quanto à infecção por F. verticillioides, estejam livres de contaminação por fumonisinas.

Maize infection by Fusarium verticillioides, the causal agent of Fusarium ear rot, can result in the production of mycotoxins of the fumonisin group. Genetic resistance is the most effective way to control this disease. Thus, the aim of this study was to evaluate tropical maize inbred lines as potential sources of resistance to F. verticillioides and production of fumonisins. Six maize inbred lines, three previously classified as resistant and three as susceptible to F. verticillioides, were inoculated with the pathogen and subsequently evaluated for ear rot severity, incidence of symptomatic grain and fumonisin concentration. Results showed that the lines R1 and R3 exhibited high resistance to the pathogen infection. However, only R3 was resistant to the accumulation of fumonisins. Therefore, it is suggested that absence of relationship between disease intensity and fumonisin production is inherent to this pathosystem. Thus, asymptomatic grains infected by F. verticillioides cannot be ensured to be free of contamination by fumonisins.

Zucchini yellow mosaic virus (ZYMV) causes substantial economic losses in cucurbit crops. Although ZYMV has been present in Brazil for more than 20 years, there is little information about the biological and molecular characteristics of the isolates found in the country. This study aimed to characterize the experimental hosts, pathotypes and genetic diversity of a collection of eleven Brazilian ZYMV isolates within the coat protein gene. For biological analysis, plant species from Amaranthaceae, Chenopodiaceae, Cucurbitaceae, Fabaceae, Solanaceae, and Pedaliaceae were mechanically inoculated and pathotypes were identified based on the reaction of a resistant Cucumis melo, accession PI414723. All of the cucurbit species/varieties and Sesamum indicum were systemically infected with all isolates. The nucleotide sequence variability of the coat protein gene ranged from 82 % to 99 % compared to the corresponding sequences of ZYMV isolates from different geographical locations. No recombination event was detected in the coat protein gene of the isolates.

O fungo Podosphaera xanthii afeta a cultura do meloeiro, apresenta diversas raças e é controlado por genes de resistência raça-específica. O acesso PI414723 é resistente às raças 1, 3 e 5 e é uma fonte adequada de genes de resistência. A herança da resistência a essas raças foi analisada em 87 plantas F2 do cruzamento PI414723 × Védrantais. A resistência às três raças pode ser explicada pela segregação de um gene dominante, embora um modelo digênico também possa ser aceito. Um mapa genético foi obtido com 206 marcadores e análise de co-segregação de fenótipos resistentes indicou a existência de dois locos ligados, posicionados no grupo de ligação II, um conferindo resistência às raças 1 e 5 (denominado Pm-x1,5) e o segundo à raça 3 (denominado Pm-x3), localizado a 5,1cM. Este estudo relata pela primeira vez a existência de Pm-x3 e as localizações genéticas desses genes de resistência de PI414723.

The fungus Podosphaera xanthii affects melon crops and presents several races controlled by race-specific resistance genes. The accession PI 414723 is resistant to races 1, 3 and 5 and it is a suitable source of resistance genes. The inheritance of resistance to these races was analyzed on 87 F2 plants from the cross of PI 414723 × Védrantais, and resistance to all three races could be explained by the segregation of a single dominant gene, although a digenic model could also be accepted. A genetic map was assembled with 206 markers, and co-segregation analysis of resistance phenotypes indicated the existence of two linked loci in linkage group II, one conferring resistance to races 1 and 5 (denominated Pm-x1,5), and the second to race 3 (denominated Pm-x3), located 5.1 cM apart. This study reports for the first time the existence of Pm-x3 and the genetic locations of these resistance genes from PI 414723.

Análise de médias foi usada para estudar o modo de herança da resistência a antracnose do colmo em milho tropical. Cada população foi composta de seis gerações que foram avaliadas para resistência em dois ensaios no delineamento de blocos ao acaso. As inoculações foram realizadas usando uma suspensão de 10(5) conídios mL-1 aplicados dentro do colmo. O comprimento interno de lesão foi medido diretamente após abertura do colmo 30 dias após a inoculação. Os resultados indicaram modos contrastantes de herança. Em uma população, os efeitos gênicos dominantes predominaram. Embora as interações, aditivo x dominante e aditivo x aditivo, também foram encontradas. Valores intermediários de herdabilidade indicaram herança da resistência complexa, provavelmente condicionada por vários genes de pequeno efeito. O modelo genético aditivo-dominante foi suficiente para explicar a variação na segunda população, onde os efeitos aditivos predominaram. Poucos genes de grande efeito controlam a resistência neste cruzamento. Heterose diferiu amplamente entre as populações o que pode ser explicado pelo background genético das linhagens genitoras resistentes.

Generation means was used to study the mode of inheritance of resistance to anthracnose stalk rot in tropical maize. Each population was comprised of six generations in two trials under a randomized block design. Inoculations were performed using a suspension of 10(5) conidia mL-1 applied into the stalk. Internal lesion length was directly measured by opening the stalk thirty days after inoculation. Results indicated contrasting modes of inheritance. In one population, dominant gene effects predominated. Besides, additive x dominant and additive x additive interactions were also found. Intermediate values of heritability indicated a complex resistance inheritance probably conditioned by several genes of small effects. An additive-dominant genetic model sufficed to explain the variation in the second population, where additive gene effects predominated. Few genes of major effects control disease resistance in this cross. Heterosis widely differed between populations, which can be attributed to the genetic background of the parental resistant lines.

O meloeiro (Cucumis melo L.) é uma frutífera largamente cultivada no Brasil, principalmente no nordeste brasileiro, onde é produzida principalmente para a exportação. Plantas da família do meloeiro, como pepino e abóbora, podem ser severamente afetadas pelo oídio, causado por Podosphaera xanthii.. Este fungo apresenta diversas raças fisiológicas cuja correta identificação é importante para o manejo da doença, já que o uso de variedades resistentes é o método mais eficaz de seu controle. No entanto, a identificação destas raças por meio da prática tradicional de inoculações em uma série diferenciadora de variedades de meloeiro é laboriosa e passível de erros. Devido a isso, um método alternativo seria o uso de marcadores moleculares para determinar de forma rápida a identidade das raças. O objetivo deste estudo foi o de analisar a variabilidade entre isolados de P. xanthii previamente classificados em raças através da técnica de AFLP e do seqüenciamento da região ITS 5.8S do rDNA. A partir dos marcadores AFLP obteve-se um dendrograma no qual não houve separação dos isolados quanto às suas raças, origem geográfica ou hospedeiro de origem. Com esta técnica verificou-se alta variabilidade entre isolados, com similaridade genética máxima de 69% e similaridade mínima de 23%. Ao contrário da informação gerada por AFLP, não foi observada variação na sequência da região ITS 5.8S entre isolados. Desta forma, a análise por AFLP indicou que os isolados tem composição genética heterogênea muito embora este fato não tenha sido evidenciado pelo sequenciamento da região ITS.

Melon crop (Cucumis melo L.) is a fruit largely cultivated in Brazil, especially in the Northwestern region, where it is produced mainly for export. Plants of the melon family, such as cucumber and pumpkin, can be severely affected by powdery mildew, caused by Podosphaera xanthii. This fungus has several physiological races and their correct identification is important for managing the disease, since the use of resistant cultivars is the most effective method for its control. However, identification of races by the traditional method of inoculation in a differential series of melon varieties is laborious and error prone. Due to this, an alternative method would be the use of molecular markers to quickly determine the race identity. The objective of this study was to analyze the variability among P. xanthii isolates previously classified into races through the AFLP technique and sequencing of the rDNA's ITS 5.8S region. With the AFLP markers a dendrogram was generated, where the isolates could not be grouped according to their races, geographic region or host. With this technique, a high variability among isolates was verified, with maximum genetic similarity of 69% and minimum similarity of 23%. In contrast to the AFLP data, there was no sequence variation in the ITS 5.8S region among isolates. Thus the AFLP data indicated that the isolates have a heterogeneous genetic composition even though this was not evidenced by the sequencing of the ITS region.

Estudos foram realizados para analisar a caracterização genética usando marcadores de RFLP-ITS e ISSP e a amplicação do gene de patogenicidade cap20 por PCR em isolados de Colletotrichum gloeosporioides de diferentes hospedeiros. Primers para o gene cap20 foram construídos a partir de seqüências selecionadas do GenBank (National Center of Biotechnology Information, http://www.ncbi.nlm.nih.gov) com o programa Primer 3. A análise dos dendrogramas revelou que o marcador RFLP-ITS foi mais informativo em separar as espécies de Colletotrichum, e que o primer EI1 evidenciou maior diversidade genética. A amplificação do DNA dos isolados de Colletotrichum para o gene cap20 com os primers P1 e P2 indicou que este gene pode apresentar variações dentro de C. gloeosporioides relacionada ao hospedeiro, e que também está presente em outras espécies de Colletotrichum.

Studies were performed to analyze the genetic characterization using RFLP-ITS and Intron (primer EI1) markers and the amplification of the cap20 pathogenicity gene by PCR in Colletotrichum gloeosporioides isolates of different hosts plant. The genetic variability was accessed using RFLP-ITS and Intron markers and grouping by UPGMA method. Primers to cap20 gene were constructed using selected sequences of the GenBank (National Center of Biotechnology Information, http://www.ncbi.nlm.nih.gov) with the Primer 3 program. The dendrograms analysis showed that the RFLP-ITS marker was more informative to separate the Colletotrichum sp, and that primer EI1 demonstrated greater genetic diversity. The amplification of the DNA of the Colletotrichum isolates to the cap20 gene with primers P1 and P2 indicated that this gene could present variations into C. gloeosporioides related with the host, and also that it was present in other Colletotrichum sp.

Na cultura do milho as podridões de espigas causadas por fungos destacam-se como uma das principais responsáveis pelas perdas em produção e qualidade, principalmente devido a formação dos chamados grãos ardidos. O objetivo deste trabalho foi o de identificar a micoflora presente em grãos e sementes produzidos em difer entes regiões e zonas macro-climáticas do Brasil, nos períodos de safra e safrinha. A determinação da incidência dos gêneros fúngicos foi feita pelo método do papel de filtro com congelamento. Foram a nalisa dos duzentos grãos/ sementes por amostra em um total de 44 amostras de grãos ardidos e 12 de sementes. As análises foram efetuadas sob mi croscópio estereoscópico e microscópio ópt ico. Os principais fungos encontrados neste levantamento, tanto em sementes como em grãos ardidos, foram Penicillum sp., Fusarium spp., Aspergillus spp., Cladosporium sp., Cephalosporium sp. e Stenocarpella spp. Dentre os patógenos principais, Fusarium spp. e Penicillium sp. foram os gêneros encontrados em maior incidência tanto nos grãos como nas sementes nas duas safras. Não houve diferenças significativas entre os diferentes climas e regiões na safra verão para a incidência de Fusarium spp. e Penicillium sp. Porém, durante a safrinha, foi observada uma maior incidência de Fusarium spp. na região CO. Cladosporium sp. se destacou entre os fungos sobretudo durante a safrinha.

In maize crop, ear rot caused by fungi is one of the main responsible for quality and production losses, mainly because of the development of the so called discolored grains. The purpose of this study was to identify the mycoflora present in grains and kernels produced in different regions and climatic zones of Brazil during summer and winter harvest and relate the occurrence of fungi with such parameters. The identification of the fungi was done by the filter paper with freezing method. Two hundred grains/kernels for sample totaling 44 samples of discolored grains and 12 of kernels were analyzed. The tests were performed under a stereomicroscope and an optical microscope. The main fungi found in this survey, both in kernel and discolored grains were Penicillium sp., Fusarium spp., Aspergillus spp., Cladosporium sp. Cephalosporium sp. and Stenocarpella spp. Among the major pathogens, Fusarium spp. and Penicillium spp. were found in higher incidence in grains and kernels in both seasons. There were no significant differences between regions and climates in the summer season for Fusarium spp. and Penicillium sp. incidence. However, during the winter season there has been observed a higher incidence of Fusarium spp. in the region CO. Cladosporium sp. stood out among fungi especially during the winter season.

O raquitismo-da-soqueira (RSD), causado pela bactéria Leifsonia xyli subsp. xyli, é uma das mais importantes doenças da cana-de-açúcar nas regiões produtoras do mundo. O presente trabalho teve como objetivo avaliar o grau de resistência de 10 variedades comerciais RB de cana-de-açúcar mais a variedade CB 49-260, a qual foi incluída como padrão suscetível. O trabalho visou avaliar os danos amostrados em campo pelo peso das parcelas em função da relação entre a produtividade de parcelas inoculadas e não inoculadas artificialmente com a bactéria. Os resultados mostraram que as variedades RB 72 454, RB 83 5486, RB 86 7515, RB 92 5211, RB 92 5268, RB 92 5345, RB 92 8064 e a variedade padrão CB 49-260 comportaram-se como variedades suscetíveis. A variedade RB 85 5156 comportou-se como de resistência intermediária e apenas as variedades RB 85 5453 e RB 85 5536 comportaram-se como tolerantes.

Ratoon stunting disease (RSD) caused by bacteria Leifsonia xyli subsp. xyli is one of the most economically important disease of sugarcane worldwide. The present survey had objective to evaluate the resistance of 10 RB commercial cultivars of sugarcane including CB 49-260 as a susceptible standard. The study evaluated the correlating the damages presented for overhauls productivity of inoculate and non inoculate parcels with the bacteria. The results showed that varieties RB 72 454, RB 83 5486, RB 86 7515, RB 92 8064, RB 92 5211, RB 92 5345 and RB 92 5268 were susceptible. RB 85 5156 had a intermediary resistance and varieties RB 85 5453 and RB 85 5536 both showed tolerant behavior.

Plantas de Capsicum annuum cv. Magali R, resistentes ao Pepper yellow mosaic virus (PepYMV), exibindo sintomas severos de mosaico amarelo, malformação foliar e subdesenvolvimento foram encontradas em plantios na região de Lins, SP, Brasil, em 2003/04. Partículas semelhantes àquelas do gênero Potyvirus foram observadas em extrato foliar de planta infectada examinado em microscópio eletrônico de transmissão. O extrato foliar também reagiu com anti-soro contra o PepYMV em PTA-ELISA. Além de C. annuum cv. Magali R, esse potyvirus também infectou sistemicamente C. annuum cv. Rubia R, que é resistente ao PepYMV. A seqüência de nucleotídeos de parte do gene da proteína capsidial (CP) desse potyvirus apresentou 96-98% de identidade com a de outros isolados do PepYMV. A seqüência parcial de nucleotídeos da região 3' não traduzida (3' NTR) apresentou 94-96% de identidade com a do PepYMV. Esses resultados são indicativos de que o potyvirus que quebrou a resistência em pimentão é um isolado do PepYMV.

Plants of Capsicum annuum cv. Magali R, resistant to Pepper yellow mosaic virus (PepYMV), which showed severe yellow mosaic, leaf malformation and stunting were observed during the 2003/04 growing season in Lins, São Paulo State, Brazil. Potyvirus-like particles observed in leaf sap from infected plants under the electron microscope reacted with an antiserum against PepYMV in PTA-ELISA. In addition to C. annuum cv. Magali R, this potyvirus also infected systemically the resistant C. annuum cv. Rubia R. The nucleotide sequence of part of the CP gene of this potyvirus shared 96-98% identity with that of other PepYMV isolates. The partial nucleotide sequence of the 3' NTR showed 94-96% identity with that of PepYMV. These data indicate that this potyvirus is a resistance-breaking isolate of PepYMV.

Brazil produced 330,000 metric tons of melons in 2005, principally in the Northeast region where one of the most important melon pathogens is the powdery mildew fungus Podosphaera xanthii. The disease is controlled mainly by incorporating single dominant resistance genes into commercial hybrids. We report on linkage analysis of the Pm-1 resistance gene, introgressed from the AF125Pm-1 Cantalupensis Charentais-type breeding line into the yellow-fleshed melon (Group Inodorus) breeding line AF426-S by backcrossing to produce the resistant line AF426-R, and the amplified fragment length polymorphism (AFLP) marker M75/H35_155 reported to be polymorphic between AF426-S and AF426-R. Segregation analysis of M75/H35_155 using a backcross population of 143 plants derived from [AF426-R x AF426-S] x AF426-S and screened for resistance to P. xanthii race 1 produced a recombination frequency of 4.9%, indicating close linkage between M75/H35_155 and Pm-1. Using the same segregating population, the M75/H35_155 marker had previously been reported to be distantly linked to Prv¹, a gene conferring resistance to papaya ringspot virus-type W. Since M75/H35_155 is linked to Prv¹ at a distance of 40.9 cM it is possible that Pm-1 and Prv¹ are also linked.

A cachaça é uma bebida típica brasileira produzida a partir da destilação do caldo de cana-de-açúcar fermentado principalmente por Saccharomyces cerevisiae. Grande parte da produção nacional é artesanal, e não há uma preocupação por parte dos produtores quanto ao controle microbiológico da fermentação. Este trabalho objetivou caracterizar a comunidade bacteriana contaminante do fermento utilizado na produção de cachaça em um alambique artesanal. Foram coletadas quatro amostras, sendo uma (NA) utilizada como efeito comparativo e coletada um ano anterior às demais. As restantes foram coletadas em três diferentes períodos: ao final do primeiro dia de fermentação (NP), após quinze dias (NS) e trinta dias após a utilização do mesmo fermento (NT). Foram analisadas, a partir do seqüenciamento parcial do gene 16S rDNA, 587 seqüências. As análises das seqüências revelaram a presença de 170 unidades taxonômicas operacionais. Destas, 152 foram identificadas uma única vez em diferentes amostras, dezessete foram comuns em pelo menos duas amostras e somente uma foi identificada em três amostras, evidenciando uma grande dinâmica populacional bacteriana durante o processo fermentativo. Análises estatísticas revelaram diferenças na composição bacteriana entre as amostras. Foram encontradas espécies ainda não descritas na literatura em fermentos para a produção de cachaça, como Weissella cibaria, Leuconostoc citreum e algumas espécies de Lactobacillus, além de bactérias não conhecidas. Os resultados revelaram que a comunidade de bactérias contaminantes do processo fermentativo é muito mais complexa do que se conhecida. Não há conhecimento de relato anterior sobre a utilização desta técnica para determinar contaminantes bacterianos em fermentos de cana-de-açúcar para produção de cachaça.

Cachaça is a typical Brazilian liquor produced from the distillation of fermented sugarcane juice mainly by Saccharomyces cerevisiae. Most of the domestic production is artisanal, and producers usually are not concerned regarding microbiological control of the fermentation. This study aimed to characterize the contaminant bacterial community of the yeast used in the production of cachaça in an artisanal still. Four samples were collected, of which one (NA) was used for comparison purposes and was collected one year earlier. The remaining samples were collected at three different periods: at the end of the first day of fermentation (NP), after fifteen days (NS), and thirty days after the same yeast was used (NT). Five hundred and eighty-seven sequences were analyzed from the partial sequencing of the 16S rDNA gene. Sequence analyses revealed the presence of 170 operational taxonomic units (OTUs). Of these, only one was shared among three samples and seventeen were shared between two samples. The remaining 152 OTUs were identified only once in distinct samples indicating that the contaminant bacterial population is highly dynamic along the fermentation process. Statistical analyses revealed differences in bacterial composition among samples. Undescribed species in the literature on yeasts of cachaça were found, such as Weissella cibaria, Leuconostoc citreum, and some species of Lactobacillus, in addition to some unknown bacteria. The community of bacteria in the fermentation process is much more complex than it was previously considered. No previous report is known regarding the use of this technique to determine bacterial contaminants in yeast for the production of cachaça.

A mancha-de-Phaeosphaeria, de ampla ocorrência no Brasil, tem causado expressiva redução na produtividade da cultura do milho no país. Por isso, o desenvolvimento de híbridos resistentes à doença é um dos principais objetivos dos programas de melhoramento genético da cultura. Informações sobre o controle genético da resistência à doença são necessárias para que os programas de melhoramento sejam eficientes. O objetivo deste estudo foi determinar o controle genético da resistência à mancha-de-Phaeosphaeria em milho a partir da avaliação das médias de gerações oriundas de dois cruzamentos entre linhagens resistentes (DAS95 ou DAS72) com uma suscetível (DAS21), sob condições de infecção natural da doença. Os ensaios foram conduzidos em Indianópolis (MG), em duas épocas de semeadura, outubro e novembro de 2000. Foi utilizado o delineamento experimental blocos ao acaso com três repetições. A avaliação para resistência foi realizada 30 dias após o florescimento com auxílio de uma escala diagramática de porcentagem de tecido foliar total da planta afetado pela doença. As médias da severidade da doença dos genitores e das gerações F1, F2, RCP1 e RCP2 foram analisadas segundo o modelo de MATHER & JINKS (1971). A variação genética devida a efeitos aditivos foi de 73 a 84%, enquanto, devido aos efeitos dominantes, foi de 13 a 23%. Foi evidenciada a predominância dos efeitos gênicos aditivos sobre dominantes em ambas as populações estudadas. Os valores da herdabilidade foram altos, variando de 61 a 88%. Os resultados indicaram condições favoráveis ao melhoramento genético visando àresistência à mancha-de-Phaeospheria nas populações estudadas.

The Phaeosphaeria leaf spot, of ample occurrence in Brazil, has been causing an expressive reduction in the corn yield in the country. Thus, the development of resistant hybrids to this disease is one of the main objectives of corn breeding programs. Information about the genetic control of resistance to the disease is necessary so that the programs can be efficient. The main goal of this study was to determine the genetic control of resistance to the Phaeosphaeria leaf spot in maize through the assessment of the generation means from two crossing between a resistant inbred line (DAS95 or DAS72) with a susceptible line (DAS21) under natural infection conditions of the disease. The experiment was carried out in Indianópolis (MG) in two sowen dates, October and November, 2000. The randomized blocks design with three repetitions was utilized. The evaluation to resistance was performed thirty days after the flowering using a diagrammatic scale of percentage of the total foliar tissue of the plant affected by the disease. Disease severity means of parental lines and generations F1, F2, RCP1 and RCP2 were analysed according to the model by MATHER & JINKS (1971). Genetic variation due to additive effects varied from 73% to 84% whilst dominant effects ranged from 13% to 23%. In both studied population, the predominance of gene additive over dominant effects was evidented. The inheritance values were high, varying from 61% to 88%. These results indicate favourable conditions to develop new resistance lines to Phaeosphaeria leaf spot with the studied populations.

We used 125 microsatellite markers to genotype the maize (Zea mays L.) near isogenic lines (NIL) L30HtPHtPRtRt and L30htphtpRtRt and the L40htphtprtrt line which contrast regarding the presence of the recently described dominant HtP and the recessive rt genes that confer resistance to Exserohilum turcicum. Five microsatellite markers revealed polymorphisms between the NIL and were considered candidate linked markers for the HtP resistance gene. Linkage was confirmed by bulked segregant sample (BSS) analysis of 32 susceptible and 34 resistant plants from a BC1F1 population derived from the cross (L30HtPHtPRtRt x L40htphtprtrt) x L40htphtprtrt. The bnlg198 and dupssr25 markers, both located on maize chromosome 2L (bin 2.08), were polymorphic between bulks. Linkage distances were estimated based on co-segregation data of the 32 susceptible plants and indicated distances of 28.7 centimorgans (cM) between HtP and bnlg198 and 23.5 cM between HtP and dupssr25. The same set of susceptible plants was also genotyped with markers polymorphic between L30HtPHtPRtRt and L40htphtprtrt in order to find markers linked to the rt gene. Marker bnlg197, from chromosome 3L (bin 3.06), was found linked to rt at a distance of 9.7 cM. This is the first report on the chromosomal locations of these newly described genes.

Durante muitos anos, a cercosporiose, causada pelo fungo Cercospora zeae-maydis Tehon & Daniels, não foi considerada importante para a cultura do milho (Zea mays, L.) no Brasil. Entretanto, a recente utilização de práticas culturais como o plantio direto e o cultivo sob pivôs centrais favoreceram o aumento de sua severidade e incidência, de forma que a doença é hoje considerada uma das mais importantes da cultura. Isolados de C. zeae-maydis podem ser distinguidos em dois grupos genéticos (I e II) baseados em marcadores AFLP e polimorfismos das regiões ITS e rDNA 5.8S. Até o momento, no entanto, a implicação biológica de tal distinção não é conhecida. Este trabalho objetivou determinar se isolados dos dois grupos genéticos diferem em agressividade em milho. Para tal, sintomas de um híbrido suscetível foram avaliados sob condições de casa de vegetação após inoculação com 9 e 11 isolados de C. zeae-maydis dos grupos I e II, respectivamente. Plantas no estádio V3 foram inoculadas através do depósito de sementes de sorgo colonizadas pelo patógeno no cartucho. Os sintomas foram avaliados 30 após com uma escala visual. Em média, isolados do grupo genético II foram mais agressivos que os do grupo I, com índices médios de doença de 3.1 e 2.3, respectivamente. Também observamos diferenças entre experimentos que sugerem diferenças em adaptabilidade dos grupos I e II a ambientes diferentes. Este é o primeiro relato de diferenças em agressividade entre isolados dos dois grupos genéticos de C. zeae-maydis.

For many years, the gray leaf spot disease (GLS) caused by the fungus Cercospora zeae-maydis Tehon & Daniels, was not considered an important pathogen of maize (Zea mays, L.) in Brazil. However, the recent adoption of agronomical practices such as no-tillage and cultivation under central pivot irrigation systems increased the incidence and severity to the extent that GLS is now one of the most important diseases of maize. Isolates of C. zeae-maydis can be distinguished by two genetic groups (I and II) based on AFLP markers and on polymorphisms of the ITS and 5.8S rDNA regions. Until now, however, the biological implications of this distinction remain unclear. This study investigated whether isolates from the two genetic groups differ in aggressiveness towards maize. For this, symptoms of a susceptible hybrid were evaluated under greenhouse conditions with 9 and 11 isolates of C. zeae-maydis from groups I and II, respectively. Plants in the V3 growth stage were inoculated by placing sorghum seeds colonized with the pathogen in the leaf whorl and symptoms were evaluated with a visual rating scale 30 days later. On average, isolates of genetic group II were more aggressive than those of group I, with mean disease scores of 3.1 and 2.3, respectively. Differences were also observed between experiments, which suggested that group I and II might also differ in their fitness under different environments. This is the first report on differences in aggressiveness between the two genetic groups of C. zeae-maydis.
