ABSTRACT: The tetrazolium test quickly assesses seed viability, the efficiency of which depends on methodological adjustments for each species. Therefore, the objective was to evaluate the viability of Anadenanthera colubrina seeds using this test. The seeds were aged for 12 h and 24 h to obtain differences in physiological quality. Initial quality was assessed by the following tests: germination, emergence, GSI and ESI, and MGT and MET. For the tetrazolium test, a completely randomized experimental design was used in a 3 × 9 factorial arrangement consisting of three accelerated aging (AA) conditions (0, 12, and 24 h of AA) and nine combinations between concentrations of tetrazolium salt and immersion time in this salt (0.075% / 4 h, 0.075% / 6h, 0.075% / 8h, 0.1% / 4 h, 0.1% / 6 h, 0.1% / 8 h, 0.5% / 4 h, 0.5% / 6 h, and 0.5% / 8 h), with four replications of 25 seeds, evaluated at temperatures of 35 °C and 40 °C. The seeds were classified as viable and vigorous, viable and not vigorous, and inviable. The tetrazolium test was effective for estimating the viability of A. colubrina seeds. To make this estimation, the seeds should be chipped in the region opposite the micropyle, then pre-moistened in water for 10 h, followed by removal of the seed coat and immersion in a 0.1% tetrazolium solution for 4 h at 35 °C. RESUMO: O teste de tetrazólio avalia de forma rápida a viabilidade das sementes, cuja eficiência depende de ajustes metodológicos para cada espécie. ABSTRACT Among the main problems that compromise the development of forest species are water and salt stresses, especially in the early stages of development, when seedlings are more sensitive. Thus, the objective of this study was to evaluate the morphophysiological and biochemical changes in Enterolobium contortisiliquum seedlings subjected to abiotic stresses. The experimental design was randomized complete blocks (RCB), composed of five treatments, with four replicates, and the experimental plot consisted of twenty plants. From the 31st day after sowing, when the seedlings reached approximately 15 cm in height, they began to receive the treatments, which consisted of different levels of electrical conductivity of irrigation water (0.3; 2; 4; 6; 8 dS m-1) for salt stress and periods of water restriction (0; 4; 8; 12 and 16 days without irrigation) for water stress. Abstract: Melon is one of the most consumed cucurbits and commercially significant cucurbits in the world. It is planted predominantly through seedlings, and seed quality is essential in this process. Among the rapid tests used in seed analysis, the pH testing of exudate has shown promise for several cultivated species. Thus, this study aimed to verify the viability of melon (Cucumis melo L.) seeds through the pH testing of exudate. For this, seeds of the hybrids Premier and Supreme (three lots each) were subjected to germination and vigor tests (first germination count, tetrazolium, accelerated aging, emergence speed index, and emergence). The pH testing of exudate was evaluated using phenolphthalein and sodium carbonate solutions. The results indicated the possibility of classifying the lots into three quality levels for both hybrids, similar to most tests used to assess initial quality. Resumo: O meloeiro é uma das cucurbitáceas mais consumidas e de maior interesse comercial do mundo. Seu plantio é realizado predominantemente por meio de mudas, sendo a qualidade das sementes fundamental neste processo. Dentre os testes rápidos utilizados na análise de sementes, o pH do exsudato tem se mostrado promissor para várias espécies cultivadas. Com isso, objetivou-se verificar a viabilidade de sementes de melão (Cucumis melo L.) por meio do teste de pH do exsudato. Para isso, sementes dos híbridos Premier e Supreme (três lotes de cada) foram submetidas aos testes de germinação e vigor (primeira contagem de germinação, tetrazólio, envelhecimento acelerado, índice de velocidade de emergência e emergência). O teste de pH de exsudato foi avaliado com a utilização de soluções de fenolftaleína e carbonato de sódio. Os resultados indicaram que para ambos os híbridos foi possível classificar os lotes em três níveis de qualidade, semelhantes a maioria dos testes utilizados na avaliação da qualidade inicial. RESUMO Pitaya (Hylocereus undatus e H. costataricensis) encontra-se no grupo de frutas exóticas e seu cultivo no Brasil está em expansão, sobretudo na região semiárida do Nordeste. Nessa região os problemas de quantidade e qualidade hídrica normalmente ocasionam estresses ambientais limitando à sobrevivência das plantas, principalmente durante a germinação e estabelecimento de plântulas. Sendo assim, objetivou-se verifi car a ação mitigadora de tratamentos pré-germinativos em sementes de pitaya sob estresse salino. O experimento foi conduzido em delineamento inteiramente casualizado, em esquema fatorial 2 × 6, correspondente a duas espécies de pitayas H. undatus e H. costatariciensis e seis tratamentos pré-germinativos (T1 = 0,0 MPa (controle); T2 = estresse salino (-0,4 MPa); T3 = hidrocondicionamento + estresse salino; T4 = ácido giberélico + estresse salino; T5 = ácido salicílico + estresse salino; T6 = tiametoxan + estresse salino). Após 20 dias de semeadura, analisou-se a germinação, índice de velocidade de germinação, comprimentos de partes aérea e raiz primária, massa seca total, açúcares solúveis totais e aminoácidos livres totais. H. costaricensis mostrou-se mais tolerante à salinidade que H. undatus. O ácido salicílico estimulou a germinação e o crescimento de H. undatus, além de aumentar o teor de açúcares solúveis em H. costaricensis. O hidrocondicionamento das sementes atenuou o estresse salino durante a germinação de H. undatus e favoreceu o ganho de massa seca. O ácido giberélico estimulou a germinação de H. undatus e o crescimento de plântulas desta e de H. costaricensis, além disso aumentou os níveis de açúcares solúveis em H. undatus.

ABSTRACT Pitaya (Hylocereus undatus and H. costaricensis) is found in the group of exotic fruits and its cultivation in Brazil is expanding, especially in the semi-arid region of the Northeast. In this region, the problems of water quantity and quality usually cause environmental stresses which limit the survival of plants, especially during germination and seedling establishment. Thus, the objective was to evaluate the mitigating action of pre-germination treatments in pitaya seeds under salt stress. The experiment was conducted in a completely randomized design, in a 2 × 6 factorial scheme, corresponding to two species of pitaya (H. undatus and H. costaricensis) and six pre-germination treatments (T1 = 0.0 MPa (control); T2 = salt stress (-0.4 MPa); T3 = hydropriming + salt stress; T4 = gibberellic acid + salt stress; T5 = salicylic acid + salt stress; T6 = thiamethoxam + salt stress). After 20 days of sowing, germination, germination speed index, shoot length, primary root length, total dry mass, total soluble sugars and total free amino acids were analyzed. H. costaricensis was more tolerant to salinity than H. undatus. Salicylic acid stimulated the germination and growth of H. undatus, besides increasing the soluble sugar content in H. costaricensis. Seed hydropriming attenuated salt stress during germination of H. undatus and favored dry mass gain. Gibberellic acid stimulated the germination of H. undatus and the growth of H. undatus and H. costaricensis seedlings, in addition to increasing the levels of soluble sugars in H. undatus.

RESUMO Sementes de sabiá (Mimosa caesalpiniifolia Benth.) apresentam dormência tegumentar, dificultando a absorção de água e oxigênio, consequentemente retardando a germinação. Dessa forma, objetivou-se avaliar métodos para superação de dormência associados ao condicionamento fisiológico sobre germinação e vigor de sementes de sabiá, considerando a eficiência, praticidade e custo do tratamento. O delineamento experimental foi inteiramente casualizado, em esquema fatorial 4 x 5 (quatro métodos de superação de dormência x cinco tipos de condicionamento fisiológico), constituindo vinte tratamentos, com quatro repetições de vinte e cinco sementes. Os métodos de superação de dormência utilizados foram desponte, imersão das sementes em água quente a 100 °C por três minutos; imersão em ácido sulfúrico por 10 minutos e sementes intactas. Para determinação do tempo de condicionamento fisiológico das sementes, realizou-se a curva de embebição com os diferentes agentes condicionantes. O condicionamento fisiológico foi feito em papel filtro umedecido com soluções de manitol nos potenciais de -0,2 MPa (16 horas), -0,4 MPa (24 horas) e -0,6 MPa (36 horas) e apenas com água destilada para o hidrocondicionamento (12 horas) e sementes sem condicionamento. As variáveis analisadas foram primeira contagem de germinação, germinação, comprimento radicular, comprimento da parte aérea e massa seca de plántulas. Os dados foram submetidos à análise de variância pelo teste F e teste de Tukey a 5% de probabilidade. A superação de dormência de sementes de M. caesalpiniifolia deve ser realizada com o uso de água quente (100 °C) durante três minutos, sem necessidade do condicionamento fisiológico.

ABSTRACT Seeds of ‘sabiá’ (Mimosa caesalpiniifolia Benth.) have coat dormancy, which hampers the absorption of water and oxygen, thereby delaying germination. Thus, this study aimed to evaluate methods for overcoming dormancy associated with priming on germination and vigor of seeds of ‘sabiá’, considering the efficiency, practicality and cost of treatment. The experimental design was completely randomized, in a 4 x 5 factorial scheme (four methods for overcoming dormancy x five types of priming), constituting 20 treatments with four replicates of 25 seeds. The methods for overcoming dormancy used were tip removal, seed immersion in hot water at 100 °C for three minutes, immersion in sulfuric acid for 10 minutes and intact seeds. For the determination of physiological conditioning of seeds, the imbibition curve with the different priming agents was constructed. Priming was done on filter paper moistened with solutions of mannitol at the potentials of -0.2 MPa (16 hours), -0.4 MPa (24 hours) and -0.6 MPa (36 hours) and only with distilled water for the hydropriming (12 hours) and seeds without priming. The variables analyzed were first count of germination, germination, root length, shoot length and dry weight of seedlings. The data were subjected to analysis of variance by F test and Tukey test at 5% probability. The dormancy of M. caesalpiniifolia seeds should be overcome with the use of hot water (100 °C) for three minutes, without the need for priming.

Resumo A salinidade é um dos principais estresses abióticos que afeta a germinação e crescimento de plantas em regiões áridas e semiáridas. Sendo assim, algumas técnicas alternativas podem ser utilizadas para amenizar os efeitos negativos da exposição ao sal, dentre elas, o hidrocondicionamento, que consiste na embebição controlada das sementes, suficiente para promover a ativação das fases iniciais da germinação (fases I e II), sem que ocorra protrusão da raiz primária. Dessa forma, objetivou-se avaliar o efeito do hidrocondicionamento na germinação e vigor de sementes de Piptadenia moniliformis submetidas ao estresse salino. Para determinação do tempo de hidrocondicionamento, realizou-se a curva de embebição, cujo tempo escolhido foi de 38 horas. Para simulação do estresse salino durante a germinação de sementes, prepararam-se soluções com o cloreto de sódio (NaCl) nos potenciais -0,3, -0,6, -0,9 e -1,2 MPa. O teste de germinação foi realizado com sementes hidrocondicionadas e não-hidrocondicionadas, semeadas em folhas de papel germitest® umedecidos com as soluções salinas e mantidos em câmara de germinação tipo B.O.D. (Biochemical Oxygen Demand) a 25°C, durante 21 dias. O delineamento experimental foi o inteiramente casualizado em esquema fatorial 2 x 4, com quatro repetições de 25 sementes, sendo o primeiro fator formado por sementes hidrocondicionadas e não-hidrocondicionadas e o segundo aos potenciais do estresse salino (-0,3, -0,6, -0,9 e -1,2 MPa). As variáveis analisadas foram: germinação, índice de velocidade de germinação, altura de plântulas, comprimento de raiz e massa seca de plântulas. O hidrocondicionamento reduz os efeitos negativos do estresse salino sobre o vigor das sementes de P. moniliformis, até o limite de até -0,9 MPa.

Abstract Salinity is one of the main abiotic stresses affecting the germination and growth of plants in arid and semiarid regions. In this context, some alternative techniques can be used to mitigate the negative effects of the exposure to salts, including seed hydropriming, which consists in the controlled imbibition of the seeds, sufficient to promote the activation of the initial stages of the germination (stages I and II) without leading to primary root protrusion. This study aimed to evaluate the effect of hydropriming on the germination and vigor of Piptadenia moniliformis seeds subjected to salt stress. Hydropriming time was determined based on the construction of an imbibition curve, and the time chosen was 38 hours. To simulate the salt stress during the seed germination, solutions were prepared with sodium chloride (NaCl) at potentials of -0.3, -0.6, -0.9 and -1.2 MPa. The germination test was performed with hydroprimed and non-hydroprimed seeds, sown on Germitest® paper sheets moistened with saline solutions and kept in a B.O.D. (Biochemical Oxygen Demand) germination chamber at 25°C for 21 days. The experimental design was completely randomized in a 2 x 4 factorial scheme, with four replicates of 25 seeds, in which the first factor was formed by hydroprimed and non-hydroprimed seeds, whereas the second factor corresponded to the salt stress potentials (-0.3, -0.6, -0.9 and -1.2 MPa). The variables analyzed were: germination, germination speed index, seedling height, root length and seedling dry mass. Hydropriming reduces the negative effects of salt stress on the vigor of Piptadenia moniliformis seeds, up to the limit of -0.9 MPa.

RESUMO O teste de tetrazólio constitui uma das alternativas promissoras para determinar a viabilidade de sementes dormentes, recalcitrantes e de germinação lenta. Objetivou-se adequar a metodologia do teste de tetrazólio para estimar a viabilidade e vigor em sementes de Piptadenia stipulacea (Benth.) Ducke. Para isso, uma amostra de sementes foi classificada em três sublotes utilizando peneiras de crivo redondo, correspondendo a seis, cinco e quatro milímetros de diâmetro, respectivamente. Inicialmente, avaliou-se o teor de água, curva de embebição, germinação, emergência, índice de velocidade de germinação e emergência, tempo médio de germinação e emergência de plântulas. Na segunda etapa, utilizou-se o delineamento experimental inteiramente casualizado, em esquema fatorial 3 x 9, sendo três sublotes e nove combinações entre concentrações e tempo de imersão no sal de tetrazólio (0,05%/2 h; 0,05%/4 h; 0,05%/6 h; 0,075%/2 h; 0,075%/4 h; 0,075%/6 h; 0,1%/2 h; 0,1%/4 h; 0,1%/6 h), totalizando vinte e sete tratamentos com quatro repetições de 25 sementes, avaliados separadamente a 35 e 40 °C. Para cada tratamento, as sementes foram divididas em quatro classes: sementes viáveis e vigorosas (classe I), viáveis e não vigorosas (classe II), inviáveis (classe III) e mortas (classe IV). O período de 8 h de pré-embebição e 4 h de coloração na solução de tetrazólio a 0,075%, sob 35 ou 40 °C, é adequado para estimar a viabilidade e vigor de sementes de P. stipulacea.

ABSTRACT The tetrazolium test is one of the promising alternatives to determine the viability of dormant, recalcitrant and slow-germinating seeds. The objective of this study was to adapt the tetrazolium test methodology to estimate the viability and vigor of Piptadenia stipulacea (Benth.) Ducke seeds. For this, a sample of seeds was classified into three sublots using round sieves, corresponding to six, five and four millimeters in diameter, respectively. Moisture content, imbibition curve, germination, emergence, germination and emergence speed indices, mean germination time and seedling emergence were initially evaluated. The second stage was conducted using a completely randomized experimental design, in 3 x 9 factorial scheme, corresponding to three sublots and nine combinations between concentrations and time of immersion in tetrazolium salt (0.05%/2 h; 0.05%/4 h; 0.05%/6 h; 0.075%/2 h; 0.075%/4 h; 0.075%/6 h; 0.1%/2 h; 0.1%/4 h; 0.1%/6 h), totaling twenty-seven treatments with four replicates of 25 seeds, evaluated separately at 35 and 40 °C. For each treatment, the seeds were divided into four classes: viable and vigorous seeds (class I), viable and non-vigorous (class II), unviable (class III) and dead (class IV). The period of 8 h of pre-imbibition and 4 h of staining in tetrazolium solution at 0.075%, under 35 or 40 °C, is adequate to estimate the viability and vigor of P. stipulacea seeds.

Resumo: Objetivou-se verificar a eficiência de índices físicos e bioquímicos na determinação da maturidade fisiológica de sementes de Tabebuia aurea, bem como avaliar o repouso pós-colheita dos frutos com diferentes estádios de maturação sobre a germinação e vigor. Para isto, classificou-se 200 frutos com a coloração do epicarpo em verde-escuro (estádio I), verde-claro (estádio II) e castanho-claro (estádio III). Inicialmente, nos frutos recém-colhidos, avaliou-se o comprimento, largura e peso e, para as sementes, o comprimento, largura e espessura, peso de mil sementes, teor de água, condutividade elétrica e composição química. Em seguida, utilizou-se o fatorial de três estádios de maturação e quatro períodos de repouso dos frutos (0, 5, 10 e 15 dias) e avaliou-se o teor de água, germinação, índice de velocidade de germinação, comprimento de raiz, comprimento da parte aérea e massa seca de plântulas. Os índices físicos e bioquímicos podem ser utilizados como indicadores da maturação em sementes de T. aurea, com exceção do comprimento dos frutos, espessura das sementes, condutividade elétrica e teor de proteínas. O máximo de germinação e vigor de sementes de T. aurea foi obtido em frutos nos estádios I (verde-escuro) ou II (verde-claro), associados ao repouso pós-colheita por quinze dias.

Abstract: The purpose of this research was to verify the efficiency of physical and biochemical indices in determining the physiological maturity of Tabebuia aurea seeds, as well as to evaluate the post-harvest storage of fruits with different maturation stages on germination and vigor. For this, 200 fruits were classified as dark green (stage I), light green (stage II), and light brown (stage III) epicarp. Freshly harvested fruits were evaluated for length, width, and weight and their seeds for length, width, thickness, thousand-seed weight, water content, electrical conductivity, and chemical composition. Subsequently, a factorial consisting of three maturation stages and four storage periods of fruits (0, 5, 10 and 15 days) were used, and water content, germination, germination speed index, root length, shoot length, and seedling dry matter were evaluated. Physical and biochemical indices can be used as maturation indicators in T. aurea seeds, except fruit length, seed thickness, electrical conductivity, and protein content. The maximum germination and vigor of T. aurea seeds were obtained in fruits at stages I (dark green) or II (light green) associated with post-harvest storage of fifteen days.

Ducke apresenta germinação lenta e desuniforme, devido à dormência tegumentar, além disto, não há indicações padronizadas quanto à temperatura e substrato para condução do teste de germinação em laboratório. Objetivou-se avaliar métodos de superação de dormência, temperaturas e substratos na germinação e vigor de sementes de P. stipulacea. Para tanto, instalaram-se dois experimentos, sendo o primeiro em delineamento inteiramente casualizado, com 4 repetições de 25 sementes e os tratamentos para superação de dormência foram: imersão em água a 100 °C por 1; 2; 3; 4; 5 e 6 min; imersão em ácido sulfúrico concentrado (H2SO4 - 98% P.A) durante 1; 4; 7; 10 e 13 minutos; escarificação em lixa n° 80; desponte na região oposta à micrópila e sementes intactas. Aos 21 dias após a semeadura, avaliou-se emergência, índice de velocidade de emergência e tempo médio de emergência. No experimento II, o delineamento experimental foi o inteiramente casualizado em esquema fatorial 4 x 6, com quatro repetições de 25 sementes, utilizando os substratos entre areia, sobre papel, rolo de papel e entre vermiculita nas temperaturas de 20; 25; 30; 35; 40 °C e alternada 20-30 °C, as variáveis analisadas foram primeira contagem, germinação, tempo médio de germinação e índice de velocidade de germinação. Para superação de dormência de sementes de P. stipulacea, recomenda-se o tratamento pré-germinativo desponte na região oposta à micrópila. Pode-se utilizar a temperatura de 30 °C e 20-30 °C com o substrato rolo de papel, na avaliação da germinação e vigor de sementes de P. stipulacea.

ABSTRACT Piptadenia stipulacea (Benth.) Ducke presents slow and uneven germination due to integumentary dormancy. In addition, there are no standardized indications as to the temperature and substrate for conducting the germination test in the laboratory. Thus, the objective was to evaluate methods of dormancy overcoming, temperatures and substrates in the germination and vigor of P. stipulacea seeds. For this, two experiments were installed, the first one in a completely randomized design, with 4 replicates of 25 seeds and treatments to overcome dormancy were: immersion in water at 100 °C for 1; 2; 3; 4; 5 and 6 min; immersion in concentrated sulfuric acid (H2SO4 - 98% P.A) for 1; 4; 7; 10 and 13 min; scarification on sandpaper no 80; in the region opposite the micropyle and intact seeds. At 21 days after sowing, emergency, emergency speed index and mean emergency time were evaluated. In experiment II, the experimental design was completely randomized in a 4 x 6 factorial scheme, with four replicates of 25 seeds, using the substrates between sand, paper, paper roll and vermiculite at temperatures of 20; 25; 30; 35; 40 °C and alternated 20-30 °C, the variables analyzed were first count, germination, mean germination time and germination speed index. In order to overcome seed dormancy of P. stipulacea, it is recommended that the pre-germinating treatment be set in the region opposite to the micropyle, and to evaluate the germination and vigor of these seeds, the temperature of 30 °C and 20-30 °C can be used with the roller substrate of paper.

RESUMO Objetivou-se estabelecer o procedimento adequado do teste de tetrazólio para estimar a viabilidade em sementes de quiabo. Inicialmente, as sementes de quatro lotes de quiabo foram imersas em álcool (98%) por 60 minutos e seguida de 18 horas de embebição em água destilada a 25 ºC. Após este período, as sementes foram seccionadas longitudinalmente, imersas em soluções de tetrazólio de 0,025; 0,050 e 0,075% por 60, 90 e 120 minutos e mantidas em estufa a 35 e 40 °C. Posteriormente, as sementes foram avaliadas quanto à viabilidade, tendo como indicação a coloração dos tecidos (vermelho brilhante rosa ou vermelho carmim forte). Paralelamente, foi instalado o teste de germinação, cujos resultados serviram como referência para o teste de tetrazólio. O experimento foi conduzido em delineamento inteiramente casualisado, em esquema fatorial 3 x 3 + 1, (três concentrações do sal de tetrazólio x três períodos de imersão e a testemunha) em quatro repetições de 50 sementes para cada tratamento. O teste de tetrazólio para estimar a viabilidade em sementes de quiabo deve ser realizado em solução de tetrazólio de 0,075%, por 90 minutos, a 35 °C.

ABSTRACT This study aimed to establish an adequate tetrazolium test procedure to estimate okra seed viability. Initially, seeds from four okra lots were immersed in alcohol (98%) for 60 minutes, followed by an 18-h soak in distilled water at 25 °C. Then, the seeds were longitudinally sectioned, immersed in 0.025, 0.050 and 0.075% tetrazolium solutions for 60, 90 and 120 minutes and maintained in an oven at 35 and 40 °C. Subsequently, the seeds were evaluated for viability, based on tissue color (bright red, pink or dark Carmine red). Simultaneously, a germination test was conducted, and its results were used as a reference for the tetrazolium test. The experiment was carried out in a completely randomized design, in a 3 x 3 + 1 factorial scheme (three concentrations of tetrazolium salt x three immersion periods, plus the control) in four replicates of 50 seeds for each treatment. The tetrazolium test to estimate okra seed viability should be conducted in 0.075% tetrazolium solution, for 90 minutes, at 35 °C.

RESUMO O teste de envelhecimento acelerado consiste em avaliar o vigor das sementes em condições de elevada temperatura e umidade, com a finalidade de identificar diferenças na qualidade fisiológica de lotes com germinação semelhante. Dessa forma, objetivou-se avaliar a eficiência do teste de envelhecimento acelerado para classificar os lotes de sementes de P. moniliformis em diferentes níveis de vigor. Inicialmente os lotes de sementes foram avaliados por meio da emergência de plântulas, índice de velocidade de emergência, comprimento da parte aérea e da raiz, além da massa seca total de plântulas, além da determinação do grau de umidade, antes e após cada período de envelhecimento das sementes. O delineamento experimental foi o inteiramente casualizado, em esquema fatorial 3 x 4 (três lotes de sementes e quatro períodos de 24, 48, 72 e 96 horas), e avaliado separadamente nas temperaturas de 38 e 41 °C. O teste de envelhecimento acelerado conduzido sob a temperatura de 41 °C durante 24 horas é a combinação mais adequada para separar os lotes de P. moniliformis em diferentes níveis de vigor, pois possibilitou a obtenção de resultados semelhantes à classificação dos lotes em relação à qualidade inicial.

ABSTRACT The accelerated aging test consists of evaluating the vigor of the seeds under conditions of high temperature and humidity, with the purpose of identifying the physiological quality of lots with similar germination. The objective of this study was to evaluate the efficiency of the accelerated aging test for classifying seed lots of P. moniliformis at different levels of vigor. Initially, the seed lots were assessed by means of seedling emergence, life velocity index, shoot length and root length, total dry mass of seedlings and determination of water content, before and after each period of aging. The experimental design for the whole world was based on a 3 × 4 factorial scheme (three seed lots and four periods of 24, 48, 72 and 96 h duration), with separate evaluations at temperatures of 38 °C and 41 °C. The accelerated aging test conducted at a temperature of 41 °C for 24 h was a more suitable combination for separating batches of P. moniliformis according to different levels of vigour, and made it possible to obtain results similar to the classification of lots in relation to the initial quality.

Resumo: Os testes rápidos têm sido imprescindíveis na avaliação do potencial fisiológico de sementes, sendo o de tetrazólio um dos que vêm sendo empregados pelas empresas produtoras de sementes. Assim, objetivou-se estabelecer o procedimento adequado para o teste de tetrazólio em sementes de Libidibia ferrea (Mart. ex Tul.) L.P. Queiroz var. ferrea. Para isso, testaram-se três concentrações da solução de tetrazólio (0,05; 0,075 e 0,1%) e três períodos de coloração (1, 3 e 6 horas) sob as temperaturas de 35 e 40 ºC. As sementes também foram avaliadas pelo teste de germinação visando a comparação com a viabilidade obtida pelo teste de tetrazólio. O delineamento experimental foi o inteiramente casualizado em esquema fatorial 3 x 3 + 1 (três concentrações x três períodos + uma testemunha = teste de germinação), para cada temperatura. As médias de sementes viáveis no teste de tetrazólio foram comparadas pelo teste de Tukey (p ≤ 0,05), enquanto a comparação entre as médias de sementes viáveis com o teste de germinação foi realizada pelo teste de Dunnett (p ≤ 0,05). Recomenda-se que o teste de tetrazólio para L. ferrea seja realizado na concentração de 0,05% por três horas de coloração, sob 35 °C ou 40 °C.

Abstract: Rapid tests have been essential to evaluate the physiological potential of seeds and the tetrazolium test is one of those which have been used by seed companies. The objective of the study was to establish the procedure for the tetrazolium test in Libidibia ferrea (Mart. Ex Tul.) L.P. Queiroz var. ferrea seeds. For this, three tetrazolium solution concentrations (0.05, 0.075 and 0.1%) and three staining periods (1, 3 and 6 hours) at temperatures of 35 to 40 °C were tested. The seeds were also evaluated by the germination test, whose result was compared with viability by the tetrazolium test. The experimental design was completely randomized in a factorial design 3 x 3 + 1 (three concentrations x three periods + a control = germination test) for each temperature. Viable seeds in the tetrazolium test were compared by Tukey’s test (p ≤ 0.05), while the comparison between the viable seeds with germination test was carried out by Dunnett’s test (p ≤ 0.05). It is recommended that the tetrazolium test for L. ferrea is carried out at a concentration of 0.05% for three-hour staining under 35 °C or 40 °C.

Resumo: A qualidade fisiológica de sementes utilizada na produção agrícola é um dos principais fatores que deve ser considerado no momento da implantação da cultura. Objetivou-se avaliar a qualidade de sementes de maxixeiro, cv. Liso de Calcutá, provenientes de plantas oriundas de sementes previamente tratadas com bioestimulante (0, 5, 10, 15 e 20 mL. kg-1 de sementes) e cultivadas sob dois níveis de salinidade da água de irrigação (0,5 e 3,5 dS. m-1). Inicialmente, determinou-se o teor de água das sementes e realizou-se os testes de germinação, envelhecimento acelerado, emergência em casa de vegetação, índice de velocidade de emergência, altura e massa seca da parte aérea das plântulas. Constatou-se que a qualidade fisiológica de sementes de maxixeiro oriundas de plantas submetidas ao estresse salino é afetada de maneira negativa, e que o uso de bioestimulante, aplicado via semente, proporciona efeito benéfico na qualidade fisiológica das sementes produzidas na geração seguinte.

Abstract: The physiological quality of seeds used in agricultural production is one of the main factors that should be considered when implanting the culture. This study aimed at evaluating the quality of bur gherkin seeds, cv. Liso de Calcutá, coming from plants grown from seeds that were previously treated with biostimulant (0, 5, 10, 15 and 20 mL. kg-1 seeds) and cultivated in two irrigation water salinities (0.5 and 3.5 dS. m-1). Initially, the water content of seeds was determined and the following tests were performed: germination, accelerated aging, greenhouse emergence, emergence speed index, height and dry mass of seedling aerial part. It was found that the physiological quality of bur gherkin seeds coming from plants that were subjected to salt stress is negatively affected, and that the use of biostimulant, applied via seeds, provides beneficial effects on the physiological quality of seeds produced in the next generation.

Resumo O teste de condutividade elétrica é considerado rápido, prático e objetivo na diferenciação de lotes de sementes, no entanto, alguns fatores podem influenciar nos resultados. O objetivo deste trabalho foi avaliar a qualidade fisiológica de lotes de sementes de P. moniliformis por meio do teste de condutividade elétrica. Para isto, foram testados dois lotes em três quantidades de sementes (25, 50 e 75 sementes) e nove tempos de embebição (2, 4, 6, 12, 24, 36, 48, 60 e 72 horas) em três volumes de água (50, 75 e 100 mL) a 25 °C. Para avaliação da qualidade inicial dos lotes determinou-se a porcentagem de germinação, índice de velocidade de germinação, tempo médio de germinação, comprimento e massa seca de plântulas. Adotou-se o delineamento inteiramente casualizado, em esquema fatorial 2x3x9 (2 lotes, 3 quantidade de sementes, 9 períodos de embebição) avaliados para cada volume separadamente, realizando-se a comparação entre médias pelo teste Tukey a 5% de probabilidade. De acordo com os resultados, recomenda-se o uso do teste de condutividade elétrica em sementes deP. moniliformis utilizando 50 mL de água destilada com 50 sementes por 6 horas ou com 75 sementes em 75 mL de água durante 2 horas, a 25 °C.

The electrical conductivity test is a quick, practical and objective test force; however, its effectiveness varies according to the species, quantity of seed, water volumes, soaking temperature, among other factors. The objective of this study was to evaluate the physiological quality of P.moniliformis seeds lots by means of the electrical conductivity test, testing different methodologies. For this, two lots were tested in three quantities of time (25, 50 and 75 seeds) and nine soaking times (2, 4, 6 , 12, 24 , 36, 48, 60 and 72 hours) in three volumes of water (50, 75 and 100 mL) at 25 °C. To evaluate the initial quality of the lots, germination percentage, germination speed index, average time of germination, length and dry mass of seedlings were determined. The use of the electrical conductivity test of P. moniliformis seeds is recommended using 75 seeds in 75 mL of water for 2 hours at 25 °C or seeds in 50 mL distilled water with 50 seeds per 6 hours at 25 °C.

RESUMOOs testes de germinação e de vigor são componentes essenciais do processo de controle de qualidade das empresas produtoras de sementes. Diante disso, nesta pesquisa o objetivo foi estudar a metodologia do teste de condutividade elétrica, verificando sua eficiência na identificação de diferentes níveis de vigor de lotes de sementes de coentro. Para tanto, utilizaram-se quatro cultivares (Português, Super Verdão, Tabocas e Verdão), cada uma representada por quatro lotes de sementes, que inicialmente foram submetidas às avaliações do grau de umidade, germinação, primeira contagem de germinação e emergência de plântulas. O teste de condutividade elétrica foi realizado com 25 e 50 sementes embebidas em 50 e 75 mL de água destilada, a 25 ºC, por 2, 8 e 24 horas. O teste de condutividade elétrica é eficiente para a avaliação do potencial fisiológico de sementes de coentro quando conduzido com 50 sementes imersas em 50 mL de água destilada, a 25 ºC, após duas horas de embebição.

ABSTRACTThe tests of germination and vigour are essential components of the quality control process of seed producers. Therefore, the aim of this research was to study the methodology of the test of electrical conductivity, verifying its efficiency in identifying different levels of vigour in batches of coriander seed. To do this, four cultivars were used (Português, Super Verdão, Tabocas and Verdão), each represented by four batches of seed, which were initially subjected to evaluations of moisture content, germination, germination first count and seedling emergence. The test of electrical conductivity was carried out on 25 to 50 seeds soaked in 50 and 75 mL of distilled water at 25 °C for 2, 8 and 24 hours. The test of electrical conductivity is efficient to evaluate the physiological potential of coriander seeds when carried out with 50 seeds immersed in 50 mL of distilled water at 25 °C, after soaking for two hours.
