Abstract Mangroves are ecosystems located in the transition zone between land and sea, characterized by periodic flooding that confer to its unique characteristics. Little is known about the transformation of nutrients that occur during the organic matter degradation in this system. In this study, we monitor the nitrogen transformations in soils from three mangroves with distinct levels of contamination using labeled 15NO3-. We also screened the mangroves metagenomes for the presence of genes that encode enzymes involved in denitrification (nirS, nirK, nosZ, norB and narG), anaerobic oxidation of ammonia (anammox) (hh, hao and hzo) and dissimilatory nitrate reduction to ammonium (DNRA) (nrfA). The transformations of 15NO3- indicated the balance of denitrification over anammox and DNRA in all three mangroves, with lower rates of processes in the mangrove affected by oil contamination. The metagenomic analysis detected 56 sequences related to denitrification, 19 with anammox and 6 with DNRA. Genes related with denitrification were phylogenetically distributed among several groups of bacteria (mainly Gammaproteobacteria). Anammox and DNRA related sequences were affiliated with Planctomycetes and Gammaproteobacteria, respectively. Thus, metagenomic and functional approaches supported the description of denitrification, anammox and DNRA rates in mangrove soils, and identified the major bacterial groups involved in these processes.

RESUMO A seleção de bactérias tolerantes a salinidade poderá propiciar maior promoção de crescimento vegetal em solos com estresse salino. Objetivou-se isolar e selecionar isolados bacterianos de cana-de-açúcar, tolerantes à salinidade e promotores de crescimento vegetal, e avaliar a diversidade genética. Para tanto, bactérias endofíticas de raiz e rizosfera, de cana-de-açúcar, foram isoladas, em meio de cultura com e sem NaCl. Avaliou-se a densidade populacional bacteriana, o potencial para a fixação biológica de nitrogênio (FBN), produção de ácido indol acético (AIA), solubilização de fosfato inorgânico e produção da molécula quorum sensing. Além disso, a diversidade genética foi avaliada pela técnica da Eletroforese em Gradiente de Gel Desnaturante (DGGE) do NifH. Testes de tolerância à salinidade também foram realizados. Não houve diferença estatística na densidade populacional. Quanto a FBN, produção de AIA, solubilização de fosfato e produção da molécula quorum sensing, houve alto percentual de isolados positivos. Na técnica do DGGE do gene nifH, o dendrograma indicou alta variabilidade genética. Portanto, houve a seleção de bactérias que possam contribuir para o melhor desenvolvimento da cana-de-açúcar em solos salinos.

ABSTRACT The selection of salinity tolerant bacteria may provide greater promotion of plant growth in soils with salt stress. This study aimed to isolate and select bacterial isolates of sugarcane, tolerant to salinity and plant growth promoters, and assess genetic diversity. To do so, endophytic root and rhizosphere of sugarcane, were isolated in culture medium with and without NaCl. The bacterial population density, the potential for biological nitrogen fixation (BNF), production of indole acetic acid (IAA), inorganic phosphate solubilization and quorum sensing molecule production were evaluated. Furthermore, the genetic diversity was assessed by Denaturing Gradient Gel Electrophoresis (DGGE) of nifH. Salinity tolerance tests were also conducted. There was no statistical difference in population density. As for the potential for BNF, IAA production, phosphate solubilization and quorum sensing molecule production, there was a high percentage of positive isolates. In the technique of DGGE nifH, the dendrogram showed high genetic variability. So there was the selection of bacteria that can contribute to the better development of sugarcane in saline soils.

Considered as one of the most biodiverse biomes, the Amazon has a featured role in the discovery of new species of plants, animals and microorganisms, which may be important for the functionality of different ecosystems. However, studies on the impacts resulted from changes in the Amazon land use on microbial communities and their functions are still limited. In this context, the soil fungal diversity can act as an important indicator of environmental stress caused by land use of the Amazon. This study describes changes in soil fungal communities caused by different systems of land use (primary forest, secondary forest, agroforestry, agriculture and pasture). Communities were observed in each of the areas using denaturing gradient gel electrophoresis (DGGE) of 18S rRNA gene combined with the non-metric multidimensional scaling (NMDS). Unique bands indicated the dominance of particular fungal groups in each of the specific treatments, mainly in areas converted to pasture, which differed greatly from samples of other systems of land use (SLU). The analysis of partial sequence of the 18S rRNA gene of fungi in soils under primary forest, agriculture and pasture showed differences (p = 0.001), evidencing the fungal community response to such changes. Most abundant phyla were the Zygomycota in the soil under primary forest and agricultural land, and Basidiomycota in the soil under pasture. The results show that the Amazon soil is an ecosystem susceptible to environmental changes in regarding the fungi community inhabiting this niche.

Two endophytic strains of Methylobacterium spp. were used to evaluate biofilm formation on sugarcane roots and on inert wooden sticks. Results show that biofilm formation is variable and that plant surface and possibly root exudates have a role in Methylobacterium spp. host recognition, biofilm formation and successful colonization as endophytes.
