Abstract Symbioses between plants and arbuscular mycorrhizal fungi (AMF) provide many advantages, including a reduced need for fertilization. In this study, the effects of AMF inoculation and phosphorus application on the growth and nutritional status of Araucaria angustifolia seedlings were evaluated. The treatments included inoculation with AMF (Gigaspora margarita, Rhizophagus intraradices, Acaulospora colombiana, Acaulospora scrobiculata, or Dentiscutata heterogama), inoculation with a mixture of AMF in 100 g of 50-year-old araucaria forest soil, application of two doses of P2O5 (25 or 100 mg kg-1), and the control. A completely randomized design was used, with seven replicates per treatment. The growth parameters, macronutrient and micro-nutrient contents, root colonization, and mycorrhizal effects on the extracted nutrients were evaluated at 75 and 150 d post germination. Analysis of variance, correlations, and principal component analysis (PCA) were used to evaluate that data. Inoculation with AMF had favorable effects on growth parameters, root colonization, and the extraction of macronutrients and micro-nutrients. In particular, the mixture of AMF from the 50-year-old araucaria forest soil, R. intraradices, and A. colombiana were beneficial. Similarly, the application of 25 mg of P2O5 significantly affected the growth of A. angustifolia seedlings compared to other treatments and increased the efficiency of root colonization.

Resumen La simbiosis entre plantas y hongos micorrízicos arbusculares (FMA) tiene muchas ventajas, entre ellas la reducción de la fertilización. El presente estudio tuvo como objetivo evaluar el efecto de la inoculación de especies de FMA y la aplicación de fósforo sobre el crecimiento y estado nutricional de plántulas de Araucaria angustifolia. Los tratamientos que incluyeron la inoculación de especies de FMA (Gigaspora margarita, Rhizophagus intraradices, Acaulospora colombiana, Acaulospora scrobiculata y Dentiscutata heterogama), aplicación de 100 g de un suelo de bosque de araucaria de 50 años, dos dosis de P2O5 (25 y 100 mg kg-1) y un testigo, se distribuyeron en un diseño completamente al azar, con siete repeticiones por cada tratamiento. Se evaluaron a los 75 y 150 días después de la germinación los parámetros de crecimiento, el contenido de macro y micronutrientes, la colonización radicular y el efecto micorrízico de los nutrientes extraídos. Los datos obtenidos fueron sometidos al análisis de varianza, correlación y componentes principales (PCA). La inoculación de especies de FMA brindaron respuestas favorables sobre los parámetros de crecimiento, colonización radicular y sobre la extracción de macro y micronutrientes, destacando la mezcla con especies de FMA del suelo proveniente del bosque de araucaria de 50 años y las especies Rhizophagus intraradices y Acaulospora colombiana. Así mismo, la aplicación de 25 mg de P2O5 influyo significativamente sobre los parámetros de crecimiento de las plántulas de Araucaria angustifolia en relación con los otros tratamientos, además de incrementar la eficiencia en la colonización radicular.

ABSTRACT The objective of this study was to evaluate the effective temperature for overcoming the dormancy of ‘Fuyu’ persimmon tree buds. Stem samples were collected three times between 2013 and 2014. Stems were maintained in a climate incubator chamber at 3, 6, 9, and 12 °C. For each temperature, five numbers of additional chilling hours (CH) (0, 240, 384, 528, and 672 CH) were studied. The experimental design was completely randomized in a 5 × 4 factorial design (chilling hours × temperatures) with four replications with 10 cuttings. The maintenance of branches at cold temperatures from 3 to 12 °C intensified endodormancy of the buds when the plants were at the beginning of endodormancy. The most effective temperatures for overcome dormancy when the buds were in transition from paradormancy to endodormancy were from 3 to 6 °C. When the buds were already in endodormancy, temperatures of 3, 6, 9, and 12 °C were effective for the accumulation of cold and overcoming dormancy. The increase in the number of chilling hours from 3 to 12 °C induced budburst and the temperature of 12 °C was able to slowly induce and overcome bud dormancy.

RESUMO: O objetivo deste trabalho foi identificar alterações fisiológicas e bioquímicas em gemas de caquizeiro ‘Fuyu’ durante a dormência. Ramos foram coletados entre março e agosto de 2015. A dormência foi avaliada pelo teste biológico de estacas de nós isolados, a 25 °C e fotoperíodo de 16 h. As variáveis analisadas foram umidade ponderal, teor de açúcares solúveis, atividade respiratória, atividade das enzimas superóxido dismutase (SOD), catalase (CAT), ascorbato peroxidase (APX), guaiacol peroxidase (POD) e polifenoloxidase (PPO), teor de peróxido de hidrogênio (H2O2) e peroxidação lipídica. No final de março de 2015 as plantas já estavam dormentes, as folhas e frutos presentes indicam efeito de paradormência. A indução da endodormência pode ter ocorrido em junho de 2015, quando iniciou acúmulo de horas de frio (HF) abaixo de 7,2 °C e maiores HF abaixo de 12 °C, que coincidiu com o período que houve diminuição da umidade e da atividade respiratória, aumento dos açúcares redutores, diminuição da atividade da SOD, CAT, APX e PPO e aumento de H2O2. Após acúmulo de 553 HF abaixo de 12 °C, a capacidade de brotação aumentou e as gemas apresentaram aumento da umidade, diminuição do teor de açúcares redutores, aumento da atividade respiratória, baixa atividade da SOD, CAT, APX e POD e elevados teores de H2O2.

ABSTRACT: This study identified physiological and biochemical changes in ‘Fuyu’ persimmon buds during dormancy. Branches were collected between March and August 2015. Dormancy was evaluated by biological testing of isolated node cuttings at 25 °C and a photoperiod of 16 h. The variables analyzed were water content; reducing sugar content; respiratory activity; activity of superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), guaiacol peroxidase (POD) and polyphenol oxidase (PPO) enzymes; hydrogen peroxide content (H2O2) and lipid peroxidation. At the end of March 2015, the plants were already dormant, and the leaves and fruits present indicated a paradormancy effect. Induction of endodormancy may have occurred in June 2015, when chilling hours (CH) below 7.2 °C and higher CH below 12 °C began to accumulate, which coincided with the period in which there was a decrease in water content and respiratory activity, an increase in reducing sugars, a decrease in SOD, CAT, APX and PPO and an increase in H2O2. After an accumulation of 553 CH below 12 °C, the budburst capacity increased, and the buds presented increased water content, decreased reducing sugars content, increased respiratory activity, low activity in SOD, CAT, APX and POD and high levels of H2O2.

Abstract Fertilization can be a sustainable management practice for the maintenance of the threatened species such as Araucaria angustifolia. Our objective was to evaluate the elemental composition of A. angustifolia fertilized with N, P, and K, and how the fertilization altered the soil fertility. Seven-month-old seedlings were transplanted to 16 dm3 soil pots, and three experiments were carried separately (one for each cited element) in a completely randomized design. Overall, the seedlings tended to accumulate nutrients differently among the compartments in the following order: needles > branches > stem. Application of N reduced the soil pH, boosted Mn concentration in more 10 times, decreased the P uptake, and changed the C:N and C:P ratios. Increased doses of P led to higher uptake rates but promoted a reduction in the absorption of Mg. High rates of K diminished of P, Ca, Mg, Mn, and B, confirming antagonism interaction with Ca and Mg.

ABSTRACT Grafting technique has been presented as an alternative for Araucaria angustifolia propagation. However, vegetative rescuing mature trees and obtaining large numbers of propagules is difficult, because the use of branch grafts has not been indicated due to its plagiotropic and morphogenic characteristic, generally resulting in undesired growth of the graft. So we aimed to describe the unpublished fact occurred in four plants of A. angustifolia grafted with grafts from primary branches that resulted in conversion to trunk. Plant growth and morphology were evaluated ten years after grafting. We verified vertical growth and organization of trunk and persistent branches with defined whorls, same to plants grafted with trunk grafts. Field tests with grafted plants using grafts obtained from the four plants resulted in same development pattern, proving the permanent morphogenic change and opening new perspectives for araucaria vegetative rescue and grafting. We concluded that trunciform shoots can be obtained by A. angustifolia grafting with plagiotropic primary branches. Despite the morphological clarity of trunciform shoots on grafted plagiotropic primary branches, it has not yet been possible to identify the causes of this reversion.

Resumo Araucaria angustifolia (Bert.) O. Kuntze (Araucariaceae), popularmente conhecida como araucária e pinheiro brasileiro, pode ser cultivada em função de suas sementes comestíveis (pinhões), propriedades medicinais e atributos ornamentais. O objetivo foi avaliar o crescimento de parte aérea e radicular e a qualidade geral de mudas de A. angustifolia cultivadas em recipientes com diferentes tamanhos, formas e composição. As sementes foram plantas em cinco recipientes: I - tubetes de polipropileno de 126cm3 (tubetes pequenos); II - tubetes de polipropileno de 290cm3 (tubetes grandes); III - sacos pretos de polietileno de 879cm3; IV- recipientes de tecido não-tecido (TNT) de 275 cm3 (recipiente pequeno de TNT) e; Recipientes de tecido não-tecido (TNT)de 493 cm3 (recipiente grande de TNT). As mudas foram avaliadas aos 210 dias após a semeadura para a determinação das seguintes variáveis: altura (cm), diâmetro do coleto (mm), massa seca da parte aérea (g), massa seca das raízes (g), massa seca total (g), área foliar (cm2), área de raízes (cm2), volume de raízes (cm3), coeficiente de robustez, razão raiz-parte aérea e índice de qualidade de Dickson. A altura foi superior nas mudas cultivadas nos sacos plásticos (27,6 cm) e nos recipientes grandes de TNT (27,02 cm) quando comparados aos tubetes pequenos e grandes (21,75 e 21,78 cm, respectivamente). Os sacos plásticos também apresentaram maiores valores de diâmetro de coleto, biomassa de parte aérea, raízes, biomassa total e índice de qualidade de Dickson. Recipientes grandes e pequenos de TNT promoveram menores área e volume de raizes quando comparados aos tubetes grandes, mas comportaram o mesmo ou melhor crescimento da parte aérea das mudas. Considerando o conjunto das variáveis, o saco de polietileno promoveu melhor crescimento e qualidade das mudas de pinheiro brasileiro em comparação aos demais recipientes.

Abstract Araucaria angustifolia (Bert.) O. Kuntze (Araucariaceae), commonly known as araucaria and Brazilian pine tree, can be grown for their edible pine nuts (pinhões), medicinal properties, and ornamental purposes. The aim was to assess the shoot and root growth and the overall quality of A. angustifolia seedlings grown in containers with different sizes, shapes and composition. Seeds were sowed in five containers: I – 126cm3 polypropylene cone-tainers (small cone-tainers); II – 290cm3 cone-tainers (large cone-tainers); III – 879cm3 polyethylene black bags; IV- 275 cm3 nonwoven fabric (TNT) containers (small TNT container) and; 493 cm3 nonwoven fabric (TNT) containers (large TNT container). Seedlings were evaluated 210 days after sowing, regarding the following variables: aboveground height (cm), root collar diameter (mm), shoot dry mass (g), roots dry mass (g), total dry mass (g), leaf area (cm2), roots area (cm2), roots volume (cm3), sturdiness quotient, root-shoot ratio and Dickson quality index. Height was superior for seedlings grown in the plastic bags (27.6 cm) and the large TNT containers (27.02 cm) when compared to the small and large cone-tainers (21.75 and 21.78 cm, respectively). Plastic bags also presented greater values of root-collar diameter, shoots, roots and total biomass and Dickson Quality index. Small and large TNT containers promoted lower root area and volume when compared to large polyethylene cone-tainers, but allowed for the same or better aboveground growth. Taking all results analyzed together, the polyethylene black bag promoted better growth and quality of Brazilian pine tree seedlings in comparison to the other containers.

Resumo Com a finalidade de analisar a patogenicidade e o antagonismo entre porta-enxertos de videiras e microrganismos de solo, procedeu-se à seleção de bactérias promotoras de crescimento de plantas com efeito antibiótico ao fitopatógeno Ilyonectria liriodendri, isolado monospórico 2T-VSC-101, em testes de antibiose in vitro. O acesso bacteriano que produziu a maior zona de inibição foi utilizado na biotização de mudas dos porta-enxertos de videira P1103, IBBT481 e VR039-16. Da interação entre isolados de bactérias promotoras de crescimento de plantas e o fungo causador da doença pé-preto-da-videira, resultaram três isolados bacterianos com controle sobre o crescimento micelial do fitopatógeno, todos do gênero sp. BurkholderiaEntretanto, a bactéria de maior controle, Burkholderia catarinensis sp. nov. isolado POA 89, mostrou-se prejudicial ao desenvolvimento in vitro dos porta-enxertos, causando-lhes a morte. O cultivo com meio de cultura modificado com pedrisco mostrou-se adequado ao crescimento in vitro dos porta-enxertos e apropriado à observação dos sintomas característicos da doença pé-preto. Das culturas ex vitro com o porta-enxerto IBBT481 e os microrganismos, resultaram efeitos favoráveis às plantas inoculadas com a bactéria em combinação com o fitopatógeno. Também demonstrou a dificuldade de aclimatização e de crescimento desse acesso em casa de vegetação.

Abstract In order to analyze the pathogenicity and antagonism between grapevine rootstocks and soil microorganisms, plant growth-promoting bacteria with antibiotic effect were selected from the phytopathogen Ilyonectria liriodendri, 2T-VSC-101 isolated monosporic, in in vitro antibiosis tests. The bacterial access that produced the largest zone of inhibition was used in the seedling biotization of the grapevine rootstocks P1103, IBBT481 and VR039-16. From the interaction between isolates of plant growth-promoting bacteria and Ilyonectria liriodendri isolate resulted in three bacterial isolates with control of the mycelial growth of the phytopathogen, all of the Burkholderia gender.However, the highest control bacterium, Burkholderia catarinensis sp.nov. isolated POA 89, was found to be detrimental to the in vitro development of rootstocks, causing them to die. The cultivation through culture medium modified with sand was adequate for the in vitro growth of the rootstocks and appropriate for the observation of the black foot disease’s characteristic symptoms. From the ex vitro cultures with the IBBT481 rootstock and the microorganisms, there were results in favorable effects to the plants inoculated with the bacterium in combination with the phytopathogen. It also demonstrated the difficulty of acclimatization and growth of this access in a greenhouse.

RESUMO A enxertia em Araucaria angustifolia é uma alternativa em relação à produção de mudas por sementes. No entanto, a disponibilidade natural de propágulos é muito limitada, uma vez que somente material obtido do tronco (ortotrópico) pode ser utilizado. O objetivo deste trabalho foi avaliar a técnica de enxertia sobre ramos da araucária e o tropismo dos enxertos visando à multiplicação de propágulos. A enxertia foi realizada no início de julho de 2014 por meio da técnica de placagem lenhosa em 20 plantas de Araucaria angustifolia com três anos de idade. Foram utilizadas borbulhas ortotrópicas obtidas de troncos e enxertadas em 43 ramos localizados nos três últimos verticilos superiores, em uma distância de 10 cm do tronco da planta porta-enxerto. Foi avaliada a sobrevivência dos enxertos, a qualidade da cicatrização, o tropismo (ângulo de crescimento), o comprimento e o diâmetro das brotações dos enxertos, bem como o diâmetro dos ramos porta-enxerto e sua relação com o diâmetro da brotação dos enxertos. Os dados foram analisados por meio de ferramentas de estatística descritiva. A enxertia nos ramos da araucária é viável. Apesar das diferenças morfológicas e funcionais, a interação entre enxertos de origem ortotrópica e ramos porta-enxertos plagiotrópicos não interfere no pegamento, na cicatrização e no crescimento inicial dos enxertos, pelo menos até 180 dias após a enxertia. Portanto, a possibilidade de enxertia em vários ramos de uma mesma planta configura uma alternativa para multiplicação de propágulos ortotrópicos. O tropismo das brotações depende da origem do enxerto. Propágulos trunciformes (obtidos de tronco) não são afetados pela interação quando enxertados em ramos porta-enxertos plagiotrópicos, dando origem a brotações com crescimento ortotrópico e morfologia de tronco juvenil. Os ramos do verticilo superior são os mais indicados para enxertia visando à multiplicação de propágulos ortotrópicos, pois estão menos sujeitos à dominância apical.

ABSTRACT The grafting in Araucaria angustifolia is an alternative choice related to the production of seedlings using seeds. However, the natural availability of propagules is very limited, since only material obtained from the trunk (orthotropic) could be used. The aim of this study was to evaluate the grafting technique on branches of Araucaria and the graft tropism in order to have the propagule multiplication. The grafting was held in the beginning of July 2014 using the patch grafting technique in 20 plants of 3-year old Araucaria angustifolia. It was used orthotropic buds obtained from trunks and grafted in 43 branches located in the last three upper whorls, within a distance of 10 cm from the rootstock plant trunk. It was evaluated the graft survivals, the healing quality, the tropism (growth angle), the height and diameter of the graft shoots, as well as the diameter of rootstock branches and its relation to the diameter of the graft. Data were analyzed by descriptive statistical tools. The grafting in Araucaria branches is viable. Despite the morphological and functional differences, the interaction among grafts with orthotropic origin and plagiotropic rootstock branches does not interfere in the setting, healing and the initial growth of the grafts, at least up to 180 days after grafting. Therefore, the possibility of grafting in several branches of a same plant configures an alternative option for the multiplication of orthotropic propagules. The tropism of shoots depends on the graft origin. Trunciform propagules (obtained from the trunk) are not affected by the interaction when grafted on plagiotropic rootstock branches, showing shoots with orthotropic growth and youth trunk morphology. The upper whorl branches are the most suitable for grafting, aiming the multiplication of orthotropic propagules, because they are less subject to the apical dominance.

RESUMO Espécie da floresta subtropical da América do Sul e restando apenas menos de 3 % de vegetação natural, a Araucaria angustifolia é ainda pouco conhecida sobre a distribuição de elementos em suas acículas. Nesse estudo, utilizamos microscopia eletrônica de varredura (MEV) com espectroscopia por dispersão de energia (EDS) para avaliar a distribuição elementar em acículas de A. angustifolia. Foram selecionadas acículas de plantas (28 meses de idade) cultivadas em experimento de adubação: com suprimento de N, P e K; com N e P; com N e K. Na microanálise quatro tipos de tecidos especializados das acículas (epiderme superior, mesófilo paliçádico, mesófilo esponjoso e epiderme inferior) foram avaliados quanto a composição (C, O, P, K, Ca, S e Al). Quando foram detectados cristais determinaram-se as concentrações de 12 elementos (C, O, P, K, Ca, S, Al, Fe, Mg, Na, Si e Cl). Na baixa disponibilidade de P e K, esses elementos foram encontrados em baixas concentrações na epiderme, no mesófilo e em cristais. Na baixa disponibilidade de P ocorreu acúmulo de Ca e K no mesófilo esponjoso, enquanto que na baixa disponibilidade de K ocorreu acúmulo de Ca no mesófilo esponjoso. Em adição, a baixa disponibilidade de K e P favoreceu a formação de cristais, sendo que em baixa disponibilidade de K, os cristais apresentaram mais Ca e Mg. A disponibilidade de P e K no solo afetou a distribuição de elementos em acículas de A. angustifolia, sendo que o tipo de tecido e a formação de cristais foi chave para a dinâmica de nutrientes nas acículas.

ABSTRACT Araucaria angustifolia is a conifer species found in South American subtropical forests that comprises less than 3% of the native vegetation and little is known concerning the accumulation of nutritional elements in its needles. In this study, scanning electron microscopy (SEM) coupled with energy dispersive spectroscopy (EDS) was used to assess the elemental distribution in needles. Needles were selected from 28 month-old plants grown in a fertilization experiment supplied with: 1) N, P, and K; 2) N and P; and 3) N and K. In microanalysis, four types of specialized needle tissues (adaxial epidermis, palisade mesophyll, spongy mesophyll and abaxial epidermis) were evaluated for elemental composition (C, O, P, K, Ca, S and Al). When crystals were detected, the concentrations of 12 elements were determined (C, O, P, K, Ca, S, Al, Fe, Mg, Na, Si, and Cl). Under low soil P and K, these elements were found in low concentrations in the epidermis, mesophyll, and crystals. Under low soil P, Ca and K accumulated in the spongy mesophyll, while under low soil K only Ca accumulated in this tissue. In addition, low soil P or K availability favored the formation of crystals; crystals under low soil K availability had more Ca and Mg. Soil P and K availability affected the distribution of elements in needles of A. angustifolia, in that type of tissue and formation of crystals were key to the nutrient dynamics in needles.

RESUMO Foi desenvolvido um método eficiente de regeneração de plantas para Aspidosperma polyneuron a partir de culturas nodais de plântulas. Sementes maduras foram desinfestadas e os embriões germinaram em meio de cultura Woody Plant Medium (WPM). Segmentos nodais do epicótilo e hipocótilo retirados de plântulas de três semanas crescendo in vitro foram cultivados em meio WPM, suplementado com 6-benziladenina (BA) (2,5; 5,0 e 10 µM), isolado ou combinado com ácido indol-3-butírico (AIB) ou ácido α-naftalenoacético (ANA) (0,5 µM) durante o cultivo inicial e três subcultivos. Para indução de raízes, inicialmente, foram testados tratamentos-pulso de AIB (2,5; 5,0 ou 10 mM) por 15 minutos, seguidos de transferência para meio WPM, sem regulador de crescimento por cinco semanas. Plantas regeneradas foram transplantadas para bandejas contendo solo e substrato Plantmax® (3:1) e depois transferidas para sacos plásticos para aclimatização em casa de vegetação. Os explantes do hipocótilo apresentaram resultados superiores de regeneração de brotações e enraizamento, quando comparados com os do epicótilo. Os maiores números de brotações por explante (7-8) foram obtidos em meios de cultura, suplementados com 10 µM de BA, no terceiro subcultivo. Aadição de 0,5 µM de AIB ou ANA combinada com BAnão influenciou a regeneração de brotações. O tratamento-pulso de AIB (5-10 mM) por 15 minutos induziu 60% de enraizamento. As plantas regeneradas foram aclimatizadas e bem estabelecidas em casa de vegetação com 90% de sobrevivência, após três meses. Baseado nos resultados desse estudo, a micropropagação de Aspidosperma polyneuron a partir de plântulas cultivadas in vitro é viável e pode ser uma boa opção para conservação e propagação dessa espécie importante que está em extinção.

ABSTRACT In this study, an efficient method for regenerating plants from nodal cultures of seedlings was developed for Aspidosperma polyneuron. Mature seeds were surface-sterilized and embryos were germinated in Woody Plant Medium (WPM). Epicotyl and hypocotyl nodal segments, excised from 3-week-old in vitro-grown seedlings, were cultured in WPM medium supplemented with 6-benzyladenine (BA) (2.5, 5.0, and 10µM), alone or combined with indole-3-butyric acid (IBA) or α-naphthaleneacetic acid (NAA) (0.5 µM) for culture initiation and three subcultures. For root induction, IBA (2.5, 5.0, and 10 mM) pulse treatments (15 minutes) were initially applied, followed by transfer to growth regulator-free WPM for five weeks. Regenerated plants were first transplanted to trays containing soil and Plantmax® substrate (3:1) and later to polyethylene bags for acclimatization in a greenhouse. Hypocotyl explants exhibited superior shoot regeneration rate and rooting percentages compared with those of epicotyl explants. The highest numbers of shoots per explant (7−8) were obtained at the third subculture when using the culture medium supplemented with 10 µM BA. When combined with BA, the addition of 0.5 µM IBA or NAA did not affect the regeneration of shoots. An IBA pulse treatment of 5−10 mM for 15 minutes induced 60% of rooting. The regenerated plants were acclimatized and successfully established in a greenhouse, with a 90% survival rate observed after three months. Based on the results of this study, the micropropagation of Aspidosperma polyneuron from in vitro seedlings is feasible and could be a useful tool for the conservation and propagation of this important endangered species.

Araucaria angustifolia apresenta redução da taxa de crescimento durante o ano, podendo ser resposta às condições ambientais desfavoráveis, como ocorre nas fruteiras de clima temperado, que apresentam dormência de gemas como forma de sobrevivência. O objetivo deste trabalho foi avaliar a dinâmica da atividade respiratória de meristemas apicais de ramos plagiotrópicos de Araucaria angustifolia. Foram coletadas amostras de 0,4 g de brotações terminais de ramos plagiotrópicos de plantas jovens e adultas, as quais foram mantidas em 5 mL de solução de cloreto de 2,3,5 trifeniltetrazólio (1,2%) em sala de crescimento a 25°C. Em seguida, foram mantidas em 4 mL de álcool etílico absoluto para leitura por espectrofotometria da absorbância a 560 nm. A atividade respiratória de meristemas apicais de ramos plagiotrópicos de Araucaria angustifolia é variável durante o ano. A maior atividade respiratória ocorre na metade da primavera e a menor atividade respiratória ocorre no inverno, em plantas jovens e adultas. Plantas adultas permanecem em alta atividade respiratória por um período maior, até o início do verão.

Araucaria angustifolia decreases its growth ratio during the fall and winter in response to adverse environmental conditions such as in temperate fruits where bud dormancy as a survival startegy. The aim of this study was to evaluate the dynamics of the respiratory activity of apical meristems of plagiotropic branches of Araucaria angustifolia. To determine the respiratory activity, samples with 0.4 g of apical buds of plagiotropic branches of young and adult plants were collected. These buds were maintained during two hours in 5 mL of 2,3,5-triphenyl tetrazolium chloride (1.2%) in a growth room at 25 °C. Then, it was maintained during one hour in 4 mL of absolute ethyl alcohol and then the absorbance was determined by reading in spectrophotometer at 560 nm. The respiratory activity of apical meristems of plagiotropic branches of Araucaria angustifolia changed during the year, reducing in mid-spring and become lower in the winter in both young and adult plants. Adult plants remained with lower respiratory activity for a longer period, until the beginning of summer.

O objetivo da pesquisa foi avaliar a dinâmica da dormência de gemas de macieira 'Imperial Gala' e de pereira 'Hosui' em região de baixa ocorrência de frio. Os experimentos foram realizados no período de abril a agosto dos anos de 2007 e 2008 com coletas de ramos a cada duas semanas no município de Porto Amazonas, Paraná, Brasil. Na última coleta, um grupo adicional de ramos foi coletado e mantido em refrigerador a temperatura de 4 a 7°C por 1.440h. A avaliação da dormência foi realizada pelo teste biológico de estacas de nós isolados em câmaras de crescimento (BOD) a 25°C e fotoperíodo de 16h. Foram quantificadas horas de frio (HF) e unidades de frio (UF) ocorridas na região. As duas espécies foram avaliadas como experimentos distintos. O delineamento experimental adotado foi o completamente casualizado com 11 tratamentos e quatro repetições para cada espécie. A endodormência mais intensa de gemas de macieira 'Imperial Gala' ocorreu no início de junho em 2007 e da metade de junho ao início de julho em 2008. A endodormência de gemas de pereira 'Hosui' apresentou oscilações entre abril e agosto de 2007 e foi mais intensa de junho ao início de julho em 2008.

The objective of this work was to evaluate the dormancy dynamic of Imperial Gala apple tree buds and Hosui pear tree buds in a region of low chill occurrence. Experiments were conducted between April and August in 2007 and 2008. Branches were collected every two weeks from an orchard at Porto Amazonas (Paraná State, Brazil). On the last sampling day, an additional set of branches was collected and refrigerated between 4°C and 7°C for 1,440 hours. Dormancy was evaluated using a biological test of single node cuttings isolated in growth chambers (GC) at 25°C with 16 hours of light exposure. The number of chill hours (CH) and chill units (CU) for the region were recorded. The two species were evaluated in separate experiments. We used 11 completely randomized treatments with four replicas for each species. The peak of endodormancy for the Imperial Gala apple tree buds occurred in early June 2007 and from middle June to early July in 2008. The endodormancy of the Hosui pear tree buds oscillated between April and August in 2007 and peaked between June and early July in 2008.

A imbuia apresenta sementes recalcitrantes, com forte dormência tegumentar, irregularidade e baixa germinação, dificultando sua propagação natural. O objetivo deste trabalho foi avaliar a germinação in vitro de eixos embrionários de imbuia, testando o efeito de diferentes concentrações de sacarose, de carvão ativado e de formulações salinas para acelerar a produção de mudas. Eixos embrionários zigóticos foram inoculados em meio de cultura MS/2 contendo diferentes concentrações de sacarose (15, 30, 60 ou 90 g L-1), de carvão ativado (0, 1, 2 ou 3 g L-1) e diferentes formulações salinas (MS, WPM, MS/2 ou WPM/2). Os resultados indicaram que a adição de 15 g L-1 de sacarose foi insuficiente para promover a germinação dos eixos embrionários e as demais concentrações promoveram entre 45,5 e 59,4% de germinação. Com relação às formulações salinas, os melhores resultados foram obtidos com os meios de cultura MS e MS/2 obtendo-se 67,7 e 74,2% de germinação, respectivamente. A adição de carvão ativado favoreceu a germinação in vitro dos eixos embrionários, enquanto que na ausência de carvão ativado não ocorreu germinação e houve elevada porcentagem de oxidação. A germinação in vitro de imbuia é viável e pode ser obtida com eixos embrionários zigóticos maduros com a formulação salina MS/2, suplementada com 30 g L-1 de sacarose e 1g L-1 de carvão ativado.

Imbuia has recalcitrant seeds with strong tegumental dormancy, irregular and low germination, which impair natural propagation. The objective of this study was to evaluate the in vitro germination of imbuia embryonic axes by testing the effect of different concentrations of sucrose, activated charcoal and salt formulations in order to accelerate the production of plants. Zygotic embryonic axes were inoculated in MS/2 culture medium with different concentrations of sucrose (15, 30, 60 or 90 g L-1), of activated charcoal (0, 1, 2 or 3 gL-1) and in different salt formulations (MS, WPM, MS/2 or WPM/2). The results indicated that the addition of 15 g L-1 sucrose was not enough to promote germination of embryonic axes and the other concentrations promoted between 45.5 and 59.4% germination. The best results were observed with MS and MS/2 salt formulations obtained 67.7 and 74.2% of germination respectively. The addition of activated charcoal to the media favored the in vitro germination of zygotic embryonic axes, while oxidation of the embryo tissues occurred in media without activated charcoal and germination did not happen. The in vitro germination of imbuia can be achieved with the MS/2 salt formulation supplemented with 30 g L-1 sucrose and 1 g L-1 of activated charcoal.

Lavandula angustifolia Mill. é uma importante planta produtora de óleos essenciais, que apresenta dificuldade de propagação pelo enraizamento de estacas, sendo a produção de mudas realizada nesse trabalho pela técnica de micropropagação, em que foram avaliadas algumas condições do cultivo in vitro. As mudas produzidas pela micropropagação foram cultivadas em São Joaquim (SC). O óleo essencial das inflorescências foi extraído por hidrodestilação, e analisado quanto a sua composição por GC/MS, em dois anos de colheita. O meio de cultura LS apresentou melhor resposta no desenvolvimento das brotações, não sendo observadas brotações com necrose apical. Meios de cultura mais diluídos como o ½ MS e o WPM, apresentaram altas taxas de hiperidricidade e necrose apical das brotações. A citocinina BAP influenciou a multiplicação das brotações, sendo a qualidade mantida com 1,0µM ao longo de quatro subcultivos. A sobrevivência das plantas na aclimatização foi de 80%. Os teores de óleo essencial foram de 4,0% e 5,25%, na primeira e segunda colheita, respectivamente. Os compostos majoritários foram o linalol (46,88% e 37,25%) e acetato de linalila (10,09% e 12,24%).

Lavandula angustifolia Mill. is an important producer of essential oil, which presents the difficulty of propagation stem cuttings. In this work the propagation of plant was carried out by micropropagation technique, in which certain conditions were evaluated in vitro. The micropropagated plants were grown in São Joaquim (SC). The essential oil of the inflorescences was extracted by hydrodistillation and analyzed for their composition by GC/MS in two years of harvest. The culture medium LS showed a better response in the development of shoots. Culture media more diluted as the 1/2 MS and WPM showed high rates of hyperhrydricity and apical necrosis. The BAP influenced the shoot multiplication and the quality being maintained with 1.0 mM over four subcultures. The survival in the acclimatization of plants was 80%. The content of essential oil were 4.0% and 5,25% in the first and second harvest, respectively. The major compounds were linalool (46.88% and 37.25%) and linalyl acetate (10.09% and 12.24%).

A espécie Salix x rubens é cultivada no Planalto Sul Catarinense desde a década de 1940, porém ainda não estão estabelecidas suas exigências quanto da fertilidade de solo. O presente trabalho é o primeiro a avaliar o perfil produtivo dos vimais dessa região, com a aplicação do método adaptado de Summer & Farina (1986), para a recomendação de fertilidade para o cultivo do vimeiro. Esse método, baseado na Lei do Mínimo e na Lei do Máximo, permitiu estabelecer as seguintes faixas recomendadas para a produção de vime nas condições do Planalto Sul Catarinense: pH - entre 5,0 e 6,5; P - 12 a 89 mg dm-3; Mg - entre 3,2 e 7,5; Zn - 5,0 a 8,3 mg dm-3; Cu - 0,8 a 4,6 mg dm-3; e Mn - 20 a 164 mg dm-3. A relação Ca/Mg deve estar entre 1,2 e 2,9. Para o K e o Ca foi possível estabelecer apenas o nível de mínimo, mas não de excesso, respectivamente de 114 mg dm-3 e 5,3 cmol c dm-3. Foi possível ainda definir o nível de excesso para Al e para a relação Al/Ca: 1,7 cmol c dm-3 e 0,28, respectivamente. O solo deve apresentar ainda teores de argila na camada superficial inferiores a 320 g dm-3 para que a expectativa de produção seja máxima.

The species Salix x rubens is being grown on the Southern Plateau of Santa Catarina since the 1940s, but so far the soil fertility requirements of the crop have not been assessed. This study is the first to evaluate the production profile of willow plantations in this region, based on the modified method of Summer & Farina (1986), for the recommendation of fertility levels for willow. By this method, based on the law of Minimum and of Maximum for willow production for the conditions on the Southern Plateau of Santa Catarina, the following ranges could be recommended: pH: 5.0-6.5; P: 12-89 mg dm-3; Mg: 3.2-7.5 mg; Zn: 5.0-8.3 mg dm-3; Cu: 0.8-4.6 mg dm-3; and Mn; 20-164 mg dm-3. The Ca/Mg ratio should be between 1.2 and 2.9. For K and Ca only the lower (sufficiency level), but not the upper threshold (excess) was established, with respectively 114 mg dm-3 and 5.3 cmol c dm-3. It was also possible to determine the upper threshold for Al and the Al/Ca ratio, i.e., 1.7 cmol c dm-3 and 0.28, respectively. For maximum yields, the clay in the soil surface layer should be below 320 g dm-3.
