Resumo Esse artigo objetiva analisar estudos sobre lazer entre universitários da área da saúde, a partir de revisão da literatura. Utilizou-se os descritores: lazer e estudantes, e seus respectivos termos em espanhol e inglês na base de dados Bireme, resultando no final da revisão em 32 artigos. Observou-se um predomínio dos seguintes interesses do lazer: desportivos, sociais, intelectuais e artísticos, com maior número de artigos apresentando vivências nos esportes. Notou-se também que a ingestão de álcool no âmbito do lazer é bastante mencionada por esta população. Há uma predileção por práticas corporais no tempo livre, em relação aos demais interesses no contexto do lazer, havendo pouco conhecimento sobre os diferentes conteúdos do lazer. Discute-se a necessidade de criar estratégias que incentivem o lazer entre os estudantes para melhorar a qualidade de vida. Ressalta-se que são profissionais da saúde em formação e é possível que suas orientações futuras sofram influência de suas práticas pessoais.

Abstract This article set out to analyze studies on leisure activities among university students in the healthcare area. It conducted a search using the following key words: leisure and students, and their corresponding terms in Spanish and English in the Bireme database, which resulted in 32 articles at the end of the review. In the articles analyzed there was a predominance of the following leisure interests: sports, social, intellectual and artistic activities, with a greater number of articles presenting experiences in sports. It was also noted that the intake of alcohol in the leisure context is often mentioned by this population group. There was a predilection for physical exercised/sport during leisure time, compared to other leisure interests and little knowledge about any other possibilities for leisure. This article also discusses the need to develop strategies that promote leisure among these students, seeking to improve their quality of life. It is noteworthy that they are health professionals in the training phase and it is quite possible that their future orientations might be influenced by their personal practices.

A otimização das condições de cultivo para a produção de α-amilase por um termofílico Bacillus sp. cepa SMIA-2 foi realizada. Além disso, a hidrólise enzimática do amido, proveniente de várias fontes tais como batata, mandioca e milho, foi também investigada. A produção de α-amilase por Bacillus sp. SMIA-2, cultivado em meio líquido contendo amido (5 g.L-1) como fonte de carbono e suplementado com 0,5 g.L-1 de proteínas do soro de leite e 2 g.L-1 de peptona, alcançou o máximo em 32 horas com níveis de 37 U.mL-1. O microrganismo foi capaz de utilizar diversas fontes de carbono, porém a atividade da amilase variou com cada fonte. O amido foi a melhor fonte de carbono para a secreção da amilase, enquanto a sacarose, lactose, maltose, galactose e glicose não foram muito efetivas. Uma redução na concentração de amido de até 2,5 g.L-1 no meio de cultura melhorou o crescimento do organismo e a atividade enzimática. Em altas concentrações de amido, a produção da enzima foi comparativamente menor. Em relação às fontes de nitrogênio orgânico e inorgânico, a peptona (2 g.L-1) foi considerada a melhor. Considerando a quantidade de proteínas do soro de leite no meio de cultivo, a concentração de 0,25 g.L-1 foi considerada a mais efetiva para a secreção da α-amilase pelo microrganismo. A produção máxima da atividade enzimática foi observada a 50 °C e pH 8,5. A enzima foi capaz de degradar todos os amidos testados. A hidrólise do amido de batata resultou num alto rendimento de açúcares redutores em comparação às outras fontes de amido. Amido solúvel e amido de mandioca ocuparam, respectivamente, a segunda e terceira posição em relação à liberação dos açúcares redutores, enquanto que a amilase estudada mostrou apenas uma ligeira afinidade pelo amido de milho. Com o aumento da temperatura da reação para 70 °C, a hidrólise dos substratos, com exceção do amido solúvel, resultou em maiores quantidades de açúcares redutores.

The optimization of culture conditions for the production of α-amylase by the thermophilic Bacillus sp strain SMIA-2 was carried out. In addition, the enzymatic hydrolysis of starch from several sources, such as potato, cassava and corn was investigated. Alpha-amylase production by Bacillus sp SMIA-2 cultivated in liquid cultures containing starch as carbon source and supplemented with 0.5 g.L-1 whey protein and 2.0 g.L-1 peptone reached a maximum of 37 U.mL-1 at 32 hours. The microorganism was capable of utilizing several carbon sources, but amylase activity varied with each source. Starch was the best carbon source for amylase secretion, while lactose, maltose, sucrose, galactose, and glucose were not very effective. Decreasing starch concentration in the medium to 2.5 g.L-1 improved organism growth and enzyme activity. At higher starch concentrations, enzyme production was comparatively lower. Among the various organic and inorganic nitrogen sources, peptone (2.0 g.L-1) was found to be the best. Regarding the amount of whey protein in the medium, the concentration of 0.25 g.L-1 was considered the most effective for amylase secretion by the organism. Maximum amylase activity was observed at 50 °C and pH 8.5. The enzyme was able to degrade all the starches tested. Potato starch hydrolysis resulted in a higher yield of reducing sugars in comparison to the other starches. Soluble and cassava starch were, respectively, in second and third positions regarding the liberation of reducing sugars, while the amylase studied showed a slightly lower affinity for corn starch. Increasing the reaction temperature to 70 °C resulted in higher levels of reducing sugars after the hydrolysis of the substrates, except for soluble starch.

A produção de alfa-amilase por um termofilico, Bacillus sp SMIA-2, cultivado em meio líquido contendo amido solúvel como fonte de carbono, alcançou uma atividade máxima de 37 U/mL em 32 horas. Estudos sobre a caracterização da amilase revelaram que a temperatura ótima desta enzima foi 90ºC. A enzima foi estável por 1 hora a temperaturas de 40 e 50ºC enquanto a 90ºC, 66% da atividade máxima foi perdida. Entretanto, na presença de 5 mM de CaCl2, a enzima foi estável a 90ºC por 30 minutos e manteve cerca de 58% de sua atividade residual por 1 hora. O pH ótimo da enzima encontrado foi de 8.5. Após a incubação da enzima por 2 horas a pH 9.5 e 11.0 foi observado um decréscimo de aproximadamente 6.3% e 16.5% da atividade original. Em pH 6.0 a enzima perdeu cerca de 36% de sua atividade original. A enzima foi fortemente inibida por Co2+, Cu2+, e Ba2+, porém pouco afetada por Mg2+, Na+ e K+. Na presença de 2.0 M de NaCl, 63% da atividade da amilase foi mantida após 2 horas de incubação a temperatura de 45ºC. A amilase exibiu atividade acima de 70% quando incubada por 1 hora a 50ºC em presença de sódio dodecil sufato (SDS). Entretanto, uma baixa atividade residual foi obtida quando na presença do hipoclorito de sódio e uma completa inibição quando a enzima foi incubada em peróxido de hidrogênio. A compatibilidade da amilase produzida pelo Bacillus sp SMIA-2, em relação a alguns detergentes comerciais mostrou que a enzima manteve 86%, 85%, e 75% da atividade após 20 minutos de incubação a 50ºC na presença dos detergentes Omo®, Campeiro® e Tide®, respectivamente.

alpha-Amylase production by thermophilic Bacillus sp strain SMIA-2 cultivated in liquid cultures containing soluble starch as a carbon source and supplemented with 0.05% whey protein and 0.2% peptone reached a maximum activity at 32 h, with levels of 37 U/mL. Studies on the amylase characterization revealed that the optimum temperature of this enzyme was 90ºC. The enzyme was stable for 1 h at temperatures ranging from 40-50ºC while at 90ºC, 66% of its maximum activity was lost. However, in the presence of 5 mM CaCl2, the enzyme was stable at 90ºC for 30 min and retained about 58% residual activity after 1 h. The optimum pH of the enzyme was found to be 8.5. After incubation of enzyme for 2 h at pH 9.5 and 11.0 was observed a decrease of about 6.3% and 16.5% of its original activity. At pH 6.0 the enzyme lost about 36% of its original activity. The enzyme was strongly inhibited by Co2+, Cu2+ and Ba2+, but less affected by Mg2+, Na+ and K+. In the presence of 2.0 M NaCl, 63% of amylase activity was retained after 2 h incubation at 45ºC. The amylase exhibited more than 70% activity when incubated for 1 h at 50ºC with sodium dodecyl sulphate. However, very little residual activity was obtained with sodium hypochlorite and with hydrogen peroxide the enzyme was completely inhibited. The compatibility of Bacillus sp SMIA-2 amylase with certain commercial detergents was shown to be good as the enzyme retained 86%, 85% and 75% of its activity after 20 min incubation at 50ºC in the presence of the detergent brands Omo®, Campeiro® and Tide®, respectively.
