RESUMO: Existem poucos estudos sobre a extração de compostos bioativos do bagaço de azeitona pelo método de micro-ondas de hidrodifusão e gravidade (MHG). Portanto, este estudo teve como objetivo extrair compostos bioativos do bagaço por MHG utilizando um composto rotacional central (DCCR). O modelo estatístico apresentou diferença significativa para a variável potência (W) nas extrações de compostos fenólicos, apresentando comportamento linear. A análise de regressão linear simples sugere a utilização de 100W de potência para a extração de compostos fenólicos em bagaço de azeitona. Não houve diferenças significativas nas extrações de flavonóides e na capacidade antioxidante dos extratos, em diferentes potências (W) e tempos (min), nos extratos de bagaço de azeitona. ABSTRACT: There are few studies on the extraction of bioactive compounds from olive pomace by the microwave method of hydro diffusion and gravity (MHG). Abstract To date, hydroxyurea is the only effective and safe drug that significantly reduces morbidity and mortality of individuals with Sickle cell disease. Twenty years of real-life experience has demonstrated that hydroxyurea reduces pain attacks, vaso-occlusive events, including acute chest syndrome, the number and duration of hospitalizations and the need for transfusion. The therapeutic success of hydroxyurea is directly linked to access to the drug, the dose used and adherence to treatment which, in part, is correlated to the availability of hydroxyurea. Resumen Objetivo Validar el contenido del guion de un recurso audiovisual para personas que viven con el VIH Métodos Estudio metodológico con enfoque cuantitativo, norteado por el marco referencial teórico metodológico de la psicometría. La validación de contenido fue realizada por jueces especialistas en el área temática mediante el uso del método Delphi. La muestra estuvo compuesta por 22 jueces en el primer análisis de validación y siete jueces en el segundo análisis. Resultados Todos los dominios analizados presentaron coeficiente de validez de contenido (CVC) >0,80, con un CVC total (CVCt) de 0,96, consistencia interna casi perfecta, Alfa de Cronbach de 0,988, ICC de 0,982 [IC95 % 0,969-0,991] y p<0,005 significativo en la primera ronda de evaluación de los especialistas. En el segundo análisis, los criterios usados para la validación de contenido presentaron un CVCt de 0,97, con los criterios de objetividad, simplicidad, claridad, relevancia, precisión, variedad, credibilidad y equilibrio, con un 100 % de aprobación. Conclusión Se validó el guion en cuanto al contenido, lo que demuestra que es un instrumento representativo y pertinente para la elaboración del recurso audiovisual. Abstract Objective The study aimed to validate the script content of an audiovisual resource for people living with HIV. Methods This methodological study had a quantitative approach and was guided by the methodological theoretical framework of psychometrics. Content validation was performed by expert judges in the thematic area using the Delphi technique. The sample consisted of 22 judges in the first validation analysis and seven judges in the second analysis. Results All domains analyzed had a content validity coefficient (CVC) >0.80, with a total CVC (CVCt) of 0.96, almost perfect internal consistency, Cronbach’s alpha of 0.988, ICC of 0.982 [95%CI 0.969-0.991], and p<0.005 which was significant in the first round of expert evaluation. In the second analysis, the criteria used for content validation showed a CVCt of 0.97, with the criteria of objectivity, simplicity, clarity, relevance, accuracy, variety, credibility, and balance achieving 100% approval. Conclusion The script was validated in terms of content, showing to be a representative and relevant instrument for building the audiovisual resource. It contributes to the advancement of scientific knowledge as it presents results with methodological accuracy and innovations in the field of health education for people living with HIV. Resumo Objetivo: Descrever a prevalência da laceração perineal segundo a percepção autorrelatada da puérpera, e analisar os fatores associados à sua ocorrência no Brasil. Métodos: Estudo transversal conduzido em 23.894 puérperas, excluindo-se gestações gemelares, cesarianas e partos com episiotomias entre 2011 e 2012. Razões de prevalência (RP) e intervalos de confiança de 95% (IC95%) da associação entre o evento e as características maternas, feto/recém-nato, obstétricas e manejo clínico foram estimadas em modelos de regressão de Poisson hierarquizados. Resultados: Entre 4.606 puérperas, 49,5% (IC95% 46,1;42,9) autorrelataram laceração perineal. Ser adolescente (RP = 1,12; IC95% 1,02;1,25), primípara (RP = 1,47; IC95% 1,33;1,63), ter tido ganho de peso gestacional excessivo (RP = 1,17; IC95% 1,07;1,29) e ter sido submetida à manobra de Kristeller (RP = 1,18; IC95% 1,08;1,29) elevaram a proporção do desfecho. Conclusão: Os resultados encontrados demandam atenção pré-natal e adequações na assistência ao parto conforme recomendações vigentes.

Resumen Objetivo: Describir la prevalencia de laceración perineal, a partir de la percepción autoinformada de puérperas, y analizar los factores asociados a su aparición en Brasil. Métodos: Estudio transversal entre 2011 y 2012, con 23.894 puérperas, excluyendo embarazos gemelares, cesáreas y partos con episiotomías. Se estimaron razones de prevalencia (RP) e intervalos de confianza del 95% (IC95%) de la asociación entre el evento y las características maternas, feto/recién nacido, obstétricas y de manejo clínico en modelos de regresión jerárquica de Poisson. Resultados: Entre 4.606 mujeres en posparto, el 49,5%(IC95%:46,1;42,9) informó laceración perineal. Ser adolescente (RP = 1,12; IC95% 1,02;1,25), primipara (RP = 1,47; IC95% 1,33;1,63), haber tenido aumento excesivo de peso gestacional (RP = 1,17; IC95% 1,07;1,29) y haber sido sometido a la maniobra de Kristeller (RP = 1,18; IC95% 1,08;1,29) aumentó la proporción de resultados. Conclusión: Los resultados encontrados requieren atención prenatal y ajustes en la atención del parto de acuerdo con las recomendaciones actuales.

Abstract Objective: To describe the prevalence of perineal laceration, based on the self-reported perception of postpartum women, and to analyze factors associated with its occurrence in Brazil. Methods: This was a cross-sectional study conducted with 23,894 postpartum women, excluding twin pregnancies, cesarean sections, and births with episiotomies, between 2011 and 2012. Prevalence ratios (PR) and 95% confidence intervals (95%CI) of association between the event and maternal, fetus/newborn, obstetric and clinical management characteristics were estimated in hierarchical Poisson regression models. Results: Out of 4,606 postpartum women, 49.5% (95%CI 46.1;42.9) self-reported perineal laceration. Being an adolescent (PR = 1.12; 95%CI 1.02;1.25), primipara (PR = 1.47; 95%CI 1.33;1.63), having had excessive gestational weight gain (PR = 1.17; 95%CI 1.07;1.29) and having undergone the Kristeller maneuver (PR = 1.18; 95%CI 1.08;1.29) increased the proportion of the outcome. Conclusion: The results found call for prenatal care and adjustments to childbirth care so as to be in accordance with current recommendations.

Abstract This article analyzes the access to health care that produces vulnerability, as well as the resistance strategies of users. The study, resulting from qualitative research with a cartographic approach, uses the concept-tool “guide-user” to analyze the trajectory of a 48-year-old woman, with visual impairment, accompanied in research at a specialized center in rehabilitation in the municipality of João Pessoa, Paraíba, Brazil. Between October 2020 and April 2021, 12 virtual meetings were held with the user and two interviews with service professionals, recorded in field diaries. The analysis generated the following interpretation axes: Several Deaths in Life, Severina; The fabrication of a Severina body: marks of a prescriptive health of living; and Severina insists on life, in her production of living networks of care. It was evidenced that the organization of health care directed at certain groups of people leads to the production of vulnerabilities and promotes death (or even many deaths) during life. At the same time, possibilities for the construction of other flows were identified, in which it would be possible to constitute, for themselves, means of breaking with what is structured for our routes, allowing to produce holes in the walls.

Resumo Neste artigo, analisa-se o acesso ao cuidado da saúde que produz vulnerabilidades, bem como as estratégias de resistência de usuários(as). No estudo, fruto de pesquisa qualitativa com abordagem cartográfica, recorre-se ao conceito-ferramenta “usuária-guia”, ao analisar a trajetória de uma mulher de 48 anos de idade com deficiência visual, acompanhada em pesquisa num centro especializado em reabilitação da cidade de João Pessoa, Paraíba, Brasil. Entre outubro de 2020 e abril de 2021, foram realizados 12 encontros virtuais com a usuária e duas entrevistas com profissionais do serviço, registrados em diários de campo. A análise gerou os seguintes eixos de interpretação: Várias mortes em vida, Severina; A fabricação de um corpo Severina: marcas de uma saúde prescritora do viver; e Teima a vida Severina, em sua produção de redes vivas de cuidado. Evidenciou-se que a organização da atenção à saúde direcionada a certos grupos leva à produção de vulnerabilidades e promove a morte (ou mesmo muitas mortes) em vida. Ao mesmo tempo, identificaram-se possibilidades de construção de outros fluxos, em que seria possível constituir, para si, meios de romper com o que se estrutura para nossas rotas, permitindo produzir furos nos muros.

ABSTRACT Background: The objective of this study is to characterize the diversity and genetic structure of A. colubrina in three Atlantic Forest fragments in the south of Espírito Santo state, using Inter Simple Sequence Repeats (ISSR) molecular markers. Genomic DNA from 85 trees was analyzed using 12 ISSR primers, generating 147 fragments, of which 109 were polymorphic (74%). Markers were characterized as moderately informative, with a mean polymorphic informational content of 0.34. Results: Low genetic diversity was found for the three fragments and in the total sample, based on Nei’s diversity parameters (H* = 0.26) and Shannon’s index (I* = 0.38). In the dissimilarity analysis, four groups were observed in the dendrogram with an exclusive clustering trend by location. The analysis of molecular variance confirmed that most of the genetic variation is found within populations (73.50%), however, with high genetic differentiation between them (Φ ST = 0.26). This result was supported by the Bayesian approach that indicated genetically structured populations. Conclusion: The data obtained reveal that forest fragmentation affected the diversity and genetic structure of A. colubrina and allow expanding knowledge for the development of effective strategies for the conservation and management of the species. RESUMO: O objetivo deste experimento foi avaliar o desempenho produtivo, infecção por nematódeos gastrointestinais (GIN) e variáveis sanguíneas selecionadas, em cordeiras Corriedale durante a fase de terminação em sistema integrado lavoura-pecuária (ICLS) com duas taxas de lotação. O experimento utilizou 52 cordeiras Corriedale divididas em quatro grupos: sistema de semiconfinamento com baixa taxa de lotação por área (T1) por 70 dias; sistema de semiconfinamento com alta lotação por área (T2) por 70 dias; confinamento por 28 dias, seguido de semiconfinamento com baixa lotação por área (T3) por 42 dias; e confinamento por 28 dias, seguido de semiconfinamento com alta lotação por área (T4) por 42 dias. Peso corporal, ingestão de matéria seca, coleta de sangue e análise fecal para ovos de nematódeos foram avaliados para cada animal. As taxas de lotação não influenciaram no ganho de peso dos cordeiros semiconfinados (T1 e T2) ou nos confinados/semiconfinados (T3 e T4). Todos os tratamentos apresentaram contagens menores de EPG no final do período experimental. Os gêneros de nematódeos identificados durante o experimento foram Haemonchus e Trichostrongylus. Não houve diferença significativa (P > 0.05) para hematócrito, proteína plasmática total e média de eosinófilos entre os tratamentos. Esses resultados sugerem que o confinamento seguido de semiconfinamento em ICLS pode ser uma opção para a produção de cordeiras na fase de terminação e pode ser útil para a redução da GIN sem afetar o desempenho da produção. ABSTRACT: This trial evaluated the production performance, gastrointestinal nematode infection (GIN), and selected blood analytes, in Corriedale lambs during the finishing phase in an integrated crop-livestock system (ICLS) with 2 stocking rates. Fifty-two Corriedale lambs were divided into 4 groups: a semi confinement system with a low stocking rate (T1) for 70 days; a semi confinement system with a high stocking rate (T2) for 70 days; confinement for 28 days, followed by semi confinement with a low stocking rate (T3) for 42 days; and confinement for 28 days, followed by semi confinement with a high stocking rate (T4) for 42 days. Body weight, dry matter intake, blood collection, and faecal analysis of nematode eggs were performed for each animal. The stocking rates did not influence body weight gain for the semiconfined lambs (T1 and T2) or for the confined/semiconfined lambs (T3 and T4). All treatments had lower EPG counts at the end of the experimental period. The nematode genera identified during the experiment trial were Haemonchus and Trichostrongylus. There were no significant differences (P > 0.05) for packed cell volume, total plasma protein, and eosinophil means between treatments. These results suggested that the confinement followed by semi confinement in an ICLS, was an option for lamb production during the finishing phase which could be useful in reducing GIN without affecting the production performance. Hop (Humulus lupulus L.) is a plant widely used as raw material in global beer production, valued for its aroma, flavor and bittering components. Despite Brazil’s third-place position in global beer production, the country still relies on imported hops. Thus, this study describes the chemical composition and volatile oil content of Cascade and Chinook hop cultivars grown in a tropical area in São Paulo state during 2020 and 2021, as well as imported commercial pellets. Hydrodistillation was employed to extract the oils, which were analyzed using GC-FID and GC-MS techniques. Approximately 40% of the cultivated hop plants exhibited volatile oil content within the expected range. Among the 55 identified compounds, β-myrcene, β-caryophyllene, and α-humulene were the major constituents, and several key compounds contributing to hop aroma were detected. Principal component analysis revealed distinct clustering patterns based on cultivar and harvest year. This study reports the first comparative analysis of hops harvested in different years and the volatile oil composition and content of Chinook hops cultivated in Brazil, making a significant contribution to the fields of agronomy and national beer market. These findings highlight Brazil’s potential to become a major hop producer by implementing proper management practices and suitable technologies. Humulus L. Resumo Este estudo teve como objetivo detectar Toxoplasma gondii em produtos cárneos salgados artesanais, comercializados em feiras livres da microrregião Ilhéus-Itabuna, e avaliar a concentração do sal utilizado no preparo e sua influência na viabilidade do parasito. Foram coletadas 125 amostras de diversos produtos cárneos artesanais, vendidos em feiras livres localizadas na microrregião Ilhéus-Itabuna, durante o ano de 2021. Nessas amostras, a fim de detectar a presença do parasito, foram realizadas análises sorológicas de hemaglutinação indireta (HAI) e molecular (PCR). Para a determinação da concentração de cloreto de sódio presente nas amostras, foi realizado o Método de Möhr. Do total, 21 amostras foram destinadas ao bioensaio em camundongos albinos, na intenção de verificar a viabilidade de possíveis cistos teciduais presentes. Dos 125 produtos cárneos, 10 (8%) foram positivos na análise sorológica, sendo quatro linguiças curadas suína, cinco carnes de sol bovina e uma linguiça frescal mista (suíno e frango). Nenhuma amostra foi positiva na análise molecular. No bioensaio, todos os camundongos testaram negativo para a presença do parasito. Das amostras positivas na sorologia, as concentrações de NaCl variaram de 2,9% a 8%. Abstract This study aimed to detect Toxoplasma gondii in artisanal salted meat products sold in street markets in the Ilhéus-Itabuna microregion and to assess the salt concentration used in their preparation and its influence on the parasite’s viability. A total of 125 samples of various artisanal meat products sold in street markets located in the Ilhéus-Itabuna microregion were collected during 2021. Serological analysis using indirect hemagglutination (HAI) and molecular analysis (PCR) were performed on these samples to detect the presence of the parasite. Möhr’s method was utilized to determine the sodium chloride concentration in the samples. Of all samples, 21 were subjected to a bioassay in albino mice to verify the viability of possible tissue cysts. Among the 125 meat products, 10 (8%) tested positive in the serological analysis including four cured pork sausages, five beef sun-dried meats, and one mixed fresh sausage (pork and chicken). None of 125 samples tested positive in the molecular analysis. On bioassay, all mice tested negative for the presence of the parasite. The NaCl concentration in the positive samples ranged from 2.9% to 8%. RESUMO Objetivo: Apreender as estratégias de cuidadores para oferta do alimento à pessoa idosa com disfagia orofaríngea após desospitalização. Método: Pesquisa qualitativa, realizada com cuidadores de pessoas idosas com disfagia orofaríngea, que tiveram alta após internamento em um hospital universitário na Bahia. A coleta de dados foi realizada entre janeiro e fevereiro de 2023, por meio de uma entrevista semiestruturada, cujo dados foram organizados com base na análise de conteúdo e analisados com auxílio do software IRaMuTeQ. Resumo Toxoplasma gondii é um protozoário coccídio de importância zoonótica, causador da toxoplasmose. Embora os tratamentos atuais para a toxoplasmose possam estar associados a efeitos adversos e à eficácia limitada para as diferentes formas biológicas do parasita, evidências sugerem que moléculas alcaloides, como a harmalina e a piperina exibem, efeitos antiparasitários contra protozoários parasitas. Essa investigação teve como objetivo avaliar o efeito in vitro da harmalina e da piperina contra taquizoítos de T. gondii em culturas de células Vero infectadas. Após 24 horas da infecção das células hospedeiras, as culturas foram tratadas com harmalina ou piperina (0,49 a 15,63 µg/mL). Os controles negativo e positivo foram RPMI/DMSO (0,1%) e sulfadiazina (200 µg/mL), respectivamente. Harmalina reduziu significativamente a multiplicação parasitária em 20% em comparação ao controle negativo, enquanto a piperina diminuiu entre 55,56% e 88,89%, de forma dose-dependente. De acordo com uma escala de proporção intracelular de parasitas, observou-se que células Vero com baixa ou moderada proliferação parasitária foram mais prevalentes após tratamento com alcaloides. O estudo demonstrou que os alcaloides tinham efeitos antiparasitários sobre o T. gondii, sendo a piperina mais eficaz. Abstract Toxoplasma gondii is a coccidian protozoan of zoonotic importance that causes toxoplasmosis. Although the current treatments for toxoplasmosis may be associated with adverse effects and limited efficacy for different biological forms of the parasite, evidence suggests that alkaloid molecules such as harmaline and piperine exhibit antiparasitic effects against protozoa parasites. This investigation aimed to evaluate the in vitro effect of harmaline and piperine against T. gondii tachyzoites in infected Vero cell cultures. After 24 hours of host cell infection, the cultures were treated with harmaline or piperine (0.49 to 15.63 µg/mL). Negative and positive controls were RPMI/DMSO (0.1%) and sulfadiazine (200 µg/mL). Harmaline significantly reduced parasite multiplication by 20% compared to the negative control, while piperine decreased between 55.56% and 88.89% in a dose-dependent manner. According to an intracellular parasite proportion scale, it was observed that the Vero cells with low or moderate parasitic proliferation were more prevalent after the alkaloid treatment. Abstract We estimated genetic parameters of two Pinus caribaea var. caribaea progeny tests based on growth traits aiming at the identification of the best selection strategy. This study was conducted in two progeny tests located in Selvíria (MS), Brazil. Tests were set up in block design, one with 76 progenies (area 1) and the other with 99 progenies (area 2). Genetic parameters were estimated using the SELEGEN software through REML/BLUP method. To estimate the genetic gain through selection, four selection methods and ten selection intensities were tested. The intensity of selection that maximizes genetic gain and genetic diversity was calculated by the optimization point. Significant variation was observed between and with progeny for all traits in area 2 and only within plots for height in area 1. Higher values of heritability were observed in area 1. BACKGROUND The impact of nutrient availability on the survival of Mycobacterium leprae and the development of leprosy remains largely unknown. Iron is essential for the survival and replication of pathogens, while vitamin D has been involved with pathogen elimination and immunoregulation. OBJECTIVES We evaluated the influence of dietary iron and vitamin D supplementation and restriction on the inflammatory response of mouse immune cells in vitro. METHODS After 30 days of standard or modified diets, peritoneal cells and splenocytes were stimulated with the alive microorganisms and sonicated antigens of M. leprae, respectively. The production of inflammatory cytokines, reactive oxygen species, and cell proliferation were evaluated. FINDINGS In peritoneal cells, vitamin D supplementation and iron restriction reduced the production of IL-6 and TNF in response to M. leprae, while splenocytes presented a reduction in TNF production under the same conditions. Lower levels of IFN-γ and TNF were observed in both iron-supplemented and iron-deficient splenocytes. Besides, iron supplementation also reduced the production of IL-6 and IL-10. No changes in the production of reactive oxygen species or in cell proliferation were observed related to different diets. MAIN CONCLUSIONS Taken together, these data point to an interference of the status of these nutrients on the interaction between the host and M. leprae, with the potential to interfere with the progression of leprosy. Abstract Objectives To make recommendations on the diagnosis and treatment of post-extubation laryngitis (PEL) in children with or without other comorbidities. Methods A three-iterative modified Delphi method was applied. Specialists were recruited representing pediatric otolaryngologists, pediatric and neonatal intensivists. Questions and statements approached topics encompassing definition, diagnosis, endoscopic airway evaluation, risk factors, comorbidities, management, and follow-up. A consensus was defined as a supermajority >70%. Results Stridor was considered the most frequent symptom and airway endoscopy was recommended for definitive diagnosis. Gastroesophageal reflux and previous history of intubation were considered risk factors. Specific length of intubation did not achieve a consensus as a risk factor. Systemic corticosteroids should be part of the medical treatment and dexamethasone was the drug of choice. No consensus was achieved regarding dosage of corticosteroids, although endoscopic findings help defining dosage and length of treatment. Non-invasive ventilation, laryngeal rest, and use of comfort sedation scales were recommended. Indications for microlaryngoscopy and bronchoscopy under anesthesia were symptoms progression or failure to improve after the first 72-h of medical treatment post-extubation, after two failed extubations, and/or suspicion of severe lesions on flexible fiberoptic laryngoscopy. Conclusions Management of post-extubation laryngitis is challenging and can be facilitated by a multidisciplinary approach.
