Therefore, this study extracted bioactive compounds from pomace by MHG using a central rotational compound (DCCR). The statistical model showed a significant difference for the variable potency (W) in the extractions of phenolic compounds, showing linear behavior. Simple linear regression analysis suggested the use of 100W of power for the extraction of phenolic compounds in olive pomace. Resumen Objetivo Validar el contenido del guion de un recurso audiovisual para personas que viven con el VIH Métodos Estudio metodológico con enfoque cuantitativo, norteado por el marco referencial teórico metodológico de la psicometría. La validación de contenido fue realizada por jueces especialistas en el área temática mediante el uso del método Delphi. La muestra estuvo compuesta por 22 jueces en el primer análisis de validación y siete jueces en el segundo análisis. Abstract Objective The study aimed to validate the script content of an audiovisual resource for people living with HIV. Methods This methodological study had a quantitative approach and was guided by the methodological theoretical framework of psychometrics. Content validation was performed by expert judges in the thematic area using the Delphi technique. The sample consisted of 22 judges in the first validation analysis and seven judges in the second analysis. Results All domains analyzed had a content validity coefficient (CVC) >0.80, with a total CVC (CVCt) of 0.96, almost perfect internal consistency, Cronbach’s alpha of 0.988, ICC of 0.982 [95%CI 0.969-0.991], and p<0.005 which was significant in the first round of expert evaluation. In the second analysis, the criteria used for content validation showed a CVCt of 0.97, with the criteria of objectivity, simplicity, clarity, relevance, accuracy, variety, credibility, and balance achieving 100% approval. Conclusion The script was validated in terms of content, showing to be a representative and relevant instrument for building the audiovisual resource. Abstract Introduction Chronic obstructive pulmonary disease (COPD) not only restricts airflow but also induces sys-temic manifestations in individuals with the disease. Objective To evaluate the effect of a water-based aero-bic exercise program on respiratory muscle strength, thoracic mobility, dyspnea, and functional capacity in patients with COPD. Methods We conducted a blind randomized controlled trial with 22 patients with COPD, dividing them into a control group (CG) and a training group (TG). The TG participated in 24 sessions of a water aerobic exercise program, while the CG only partici-pated in the evaluations. Maximal respiratory pressure (MRP), dyspnea, and functional capacity were measured. Results When comparing the MRP values (cmH2O) in the pre- and post-training conditions, the results revealed a significant improvement in the TG [maximal inspiratory pressure (MIP): 74.8 ± 15.3 vs. 83.9 ± 17.2; maximal expi-ratory pressure (MEP): 141.5 ± 30.7 vs. 157.6 ± 32.9], whereas no difference was observed for the CG (MIP: 55.5 ± 21.8 vs. 54.4 ± 18.4; MEP: 116.2 ± 40.3 vs. 109.3 ± 38.9). Regarding thoracic mobility in the pre- and post-training conditions, no significant difference was found for the CG, whilst for the TG there was a significant in-crease at the axillary level (cm) (5.9 ± 1.8 vs. 7.7 ± 1.1). With respect to functional capacity, there was a signifi-cant increase in walking distance during the six-minute walking test only in the TG when comparing pre- and post-training conditions (462.1 ± 62.9 vs. 538.5 ± 63.7). Lastly, the dyspnea results demonstrated that after the training period there was a major reduction in the scores of Medical Research Council (3.1 ± 0.8 vs. 1.9 ± 0.7) and Borg CR-10 scales (5.2 ± 0.8 vs. 3.7 ± 0.3) only for the TG. Resumo Introdução A doença pulmonar obstrutiva crônica (DPOC) não apenas restringe o fluxo aéreo, mas também induz mani-festações sistêmicas em indivíduos com a doença. Objetivo Avaliar o efeito de um programa de exercícios aeróbicos aquáticos na força muscular respiratória (FMR), mobilidade torácica, dispneia e capacidade funcional em pacientes com DPOC. Métodos Realizou-se um ensaio clínico randomizado cego com 22 pacientes com DPOC, divi-dindo-os em grupo controle (GC) e grupo treinamento (GT). O GT participou de 24 sessões de um programa de exercícios aeróbicos aquáticos, enquanto o GC participou somente das avaliações. Foram me-didas a pressão respiratória máxima, (PRM) dispneia e capa-cidade funcional. Resultados Ao comparar os valores da PRM (cmH2O) nas condições pré e pós-treinamento, os resultados revelaram melhora significativa no GT [pressão inspiratória má-xima (PImáx): 74,8 ± 15,3 vs. 83,9 ± 17,2; pressão expiratória máxima (PEmáx): 141,5 ± 30,7 vs. 157,6 ± 32,9], enquanto não observou-se diferença para o GC (PImáx: 55,5 ± 21,8; vs. 54,4 ± 18,4; PEmáx: 116,2 ± 40,3 vs. 109,3 ± 38,9). Em relação à mobilidade torácica nas condições pré e pós-treinamento, não foi encontrada diferença significativa para o GC, enquanto para o GT houve um aumento significante no nível axilar (cm) (5,9 ± 1,8 vs. 7,7 ± 1,1). Com relação à capacidade funcional, houve aumento significativo da distância percorrida durante o teste de caminhada de 6 minutos apenas no GT quando comparadas as condições pré e pós-treinamento (462,1 ± 62,9 vs. 538,5 ± 63,7). Por fim, os resultados da dispneia demonstraram que após o período de treinamento houve uma redução importante nas pontuações do Medical Research Council (3,1 ± 0,8 vs. 1,9 ± 0,7) e nas escalas Borg CR-10 (5,2 ± 0,8 vs. 3,7 ± 0,3) apenas para o GT. Abstract Toxoplasma gondii is a coccidian protozoan of zoonotic importance that causes toxoplasmosis. Although the current treatments for toxoplasmosis may be associated with adverse effects and limited efficacy for different biological forms of the parasite, evidence suggests that alkaloid molecules such as harmaline and piperine exhibit antiparasitic effects against protozoa parasites. This investigation aimed to evaluate the in vitro effect of harmaline and piperine against T. gondii tachyzoites in infected Vero cell cultures. After 24 hours of host cell infection, the cultures were treated with harmaline or piperine (0.49 to 15.63 µg/mL). Negative and positive controls were RPMI/DMSO (0.1%) and sulfadiazine (200 µg/mL). Harmaline significantly reduced parasite multiplication by 20% compared to the negative control, while piperine decreased between 55.56% and 88.89% in a dose-dependent manner. According to an intracellular parasite proportion scale, it was observed that the Vero cells with low or moderate parasitic proliferation were more prevalent after the alkaloid treatment. The study demonstrated that the alkaloids had antiparasitic effects on T. gondii, with piperine being the most effective. Resumo Toxoplasma gondii é um protozoário coccídio de importância zoonótica, causador da toxoplasmose. Embora os tratamentos atuais para a toxoplasmose possam estar associados a efeitos adversos e à eficácia limitada para as diferentes formas biológicas do parasita, evidências sugerem que moléculas alcaloides, como a harmalina e a piperina exibem, efeitos antiparasitários contra protozoários parasitas. Essa investigação teve como objetivo avaliar o efeito in vitro da harmalina e da piperina contra taquizoítos de T. gondii em culturas de células Vero infectadas. Após 24 horas da infecção das células hospedeiras, as culturas foram tratadas com harmalina ou piperina (0,49 a 15,63 µg/mL). Os controles negativo e positivo foram RPMI/DMSO (0,1%) e sulfadiazina (200 µg/mL), respectivamente. Abstract We estimated genetic parameters of two Pinus caribaea var. caribaea progeny tests based on growth traits aiming at the identification of the best selection strategy. This study was conducted in two progeny tests located in Selvíria (MS), Brazil. Tests were set up in block design, one with 76 progenies (area 1) and the other with 99 progenies (area 2). Genetic parameters were estimated using the SELEGEN software through REML/BLUP method. To estimate the genetic gain through selection, four selection methods and ten selection intensities were tested. The intensity of selection that maximizes genetic gain and genetic diversity was calculated by the optimization point. Significant variation was observed between and with progeny for all traits in area 2 and only within plots for height in area 1. Higher values of heritability were observed in area 1. Abstract Objectives To make recommendations on the diagnosis and treatment of post-extubation laryngitis (PEL) in children with or without other comorbidities. Methods A three-iterative modified Delphi method was applied. Specialists were recruited representing pediatric otolaryngologists, pediatric and neonatal intensivists. Questions and statements approached topics encompassing definition, diagnosis, endoscopic airway evaluation, risk factors, comorbidities, management, and follow-up. A consensus was defined as a supermajority >70%. Results Stridor was considered the most frequent symptom and airway endoscopy was recommended for definitive diagnosis. Gastroesophageal reflux and previous history of intubation were considered risk factors. Specific length of intubation did not achieve a consensus as a risk factor. Systemic corticosteroids should be part of the medical treatment and dexamethasone was the drug of choice. No consensus was achieved regarding dosage of corticosteroids, although endoscopic findings help defining dosage and length of treatment. Non-invasive ventilation, laryngeal rest, and use of comfort sedation scales were recommended. Indications for microlaryngoscopy and bronchoscopy under anesthesia were symptoms progression or failure to improve after the first 72-h of medical treatment post-extubation, after two failed extubations, and/or suspicion of severe lesions on flexible fiberoptic laryngoscopy. Abstract This article analyzes the access to health care that produces vulnerability, as well as the resistance strategies of users. The study, resulting from qualitative research with a cartographic approach, uses the concept-tool “guide-user” to analyze the trajectory of a 48-year-old woman, with visual impairment, accompanied in research at a specialized center in rehabilitation in the municipality of João Pessoa, Paraíba, Brazil. Between October 2020 and April 2021, 12 virtual meetings were held with the user and two interviews with service professionals, recorded in field diaries. Resumo Neste artigo, analisa-se o acesso ao cuidado da saúde que produz vulnerabilidades, bem como as estratégias de resistência de usuários(as). No estudo, fruto de pesquisa qualitativa com abordagem cartográfica, recorre-se ao conceito-ferramenta “usuária-guia”, ao analisar a trajetória de uma mulher de 48 anos de idade com deficiência visual, acompanhada em pesquisa num centro especializado em reabilitação da cidade de João Pessoa, Paraíba, Brasil. Entre outubro de 2020 e abril de 2021, foram realizados 12 encontros virtuais com a usuária e duas entrevistas com profissionais do serviço, registrados em diários de campo. BACKGROUND The impact of nutrient availability on the survival of Mycobacterium leprae and the development of leprosy remains largely unknown. Iron is essential for the survival and replication of pathogens, while vitamin D has been involved with pathogen elimination and immunoregulation. OBJECTIVES We evaluated the influence of dietary iron and vitamin D supplementation and restriction on the inflammatory response of mouse immune cells in vitro. METHODS After 30 days of standard or modified diets, peritoneal cells and splenocytes were stimulated with the alive microorganisms and sonicated antigens of M. leprae, respectively. The production of inflammatory cytokines, reactive oxygen species, and cell proliferation were evaluated. FINDINGS In peritoneal cells, vitamin D supplementation and iron restriction reduced the production of IL-6 and TNF in response to M. leprae, while splenocytes presented a reduction in TNF production under the same conditions. Lower levels of IFN-γ and TNF were observed in both iron-supplemented and iron-deficient splenocytes. Besides, iron supplementation also reduced the production of IL-6 and IL-10. No changes in the production of reactive oxygen species or in cell proliferation were observed related to different diets. MAIN CONCLUSIONS Taken together, these data point to an interference of the status of these nutrients on the interaction between the host and M. leprae, with the potential to interfere with the progression of leprosy. Abstract This study aimed to detect Toxoplasma gondii in artisanal salted meat products sold in street markets in the Ilhéus-Itabuna microregion and to assess the salt concentration used in their preparation and its influence on the parasite’s viability. A total of 125 samples of various artisanal meat products sold in street markets located in the Ilhéus-Itabuna microregion were collected during 2021. Serological analysis using indirect hemagglutination (HAI) and molecular analysis (PCR) were performed on these samples to detect the presence of the parasite. Möhr’s method was utilized to determine the sodium chloride concentration in the samples. Of all samples, 21 were subjected to a bioassay in albino mice to verify the viability of possible tissue cysts. Among the 125 meat products, 10 (8%) tested positive in the serological analysis including four cured pork sausages, five beef sun-dried meats, and one mixed fresh sausage (pork and chicken). None of 125 samples tested positive in the molecular analysis. On bioassay, all mice tested negative for the presence of the parasite. The NaCl concentration in the positive samples ranged from 2.9% to 8%. The results demonstrated that the salt concentration in the collected samples was sufficient to inactivate the parasite T. gondii. Resumo Este estudo teve como objetivo detectar Toxoplasma gondii em produtos cárneos salgados artesanais, comercializados em feiras livres da microrregião Ilhéus-Itabuna, e avaliar a concentração do sal utilizado no preparo e sua influência na viabilidade do parasito. Foram coletadas 125 amostras de diversos produtos cárneos artesanais, vendidos em feiras livres localizadas na microrregião Ilhéus-Itabuna, durante o ano de 2021. Nessas amostras, a fim de detectar a presença do parasito, foram realizadas análises sorológicas de hemaglutinação indireta (HAI) e molecular (PCR). Para a determinação da concentração de cloreto de sódio presente nas amostras, foi realizado o Método de Möhr. Do total, 21 amostras foram destinadas ao bioensaio em camundongos albinos, na intenção de verificar a viabilidade de possíveis cistos teciduais presentes. Dos 125 produtos cárneos, 10 (8%) foram positivos na análise sorológica, sendo quatro linguiças curadas suína, cinco carnes de sol bovina e uma linguiça frescal mista (suíno e frango). ABSTRACT Background: The objective of this study is to characterize the diversity and genetic structure of A. colubrina in three Atlantic Forest fragments in the south of Espírito Santo state, using Inter Simple Sequence Repeats (ISSR) molecular markers. Genomic DNA from 85 trees was analyzed using 12 ISSR primers, generating 147 fragments, of which 109 were polymorphic (74%). Markers were characterized as moderately informative, with a mean polymorphic informational content of 0.34. Results: Low genetic diversity was found for the three fragments and in the total sample, based on Nei’s diversity parameters (H* = 0.26) and Shannon’s index (I* = 0.38). In the dissimilarity analysis, four groups were observed in the dendrogram with an exclusive clustering trend by location. The analysis of molecular variance confirmed that most of the genetic variation is found within populations (73.50%), however, with high genetic differentiation between them (Φ ST = 0.26). This result was supported by the Bayesian approach that indicated genetically structured populations. Conclusion: The data obtained reveal that forest fragmentation affected the diversity and genetic structure of A. colubrina and allow expanding knowledge for the development of effective strategies for the conservation and management of the species. ABSTRACT Objective: To understand caregivers’ strategies for offering food to older adults with oropharyngeal dysphagia after dehospitalization. Method: Qualitative research carried out with caregivers of older adults with oropharyngeal dysphagia, who were discharged after hospitalization at a university hospital in Bahia. Data collection was carried out between January and February 2023 through a semi-structured interview, whose data were organized based on content analysis and analyzed with the help of IRaMuTeQ software. Results: Three categories emerged: Caregivers’ strategies for safely offering food to older adults with dysphagia; Caregiver strategies for oral hygiene for older adults; Recognition of continuity of speech therapy after dehospitalization. Abstract This article aims to present reflective approaches between the concepts of “algorithmic governmentality”, “uncivil society” and “surveillance capitalism”, discussing the points of notional convergence and divergence and proposing possibilities of resistance through the institutional production of the commons. ABSTRACT: This trial evaluated the production performance, gastrointestinal nematode infection (GIN), and selected blood analytes, in Corriedale lambs during the finishing phase in an integrated crop-livestock system (ICLS) with 2 stocking rates. Fifty-two Corriedale lambs were divided into 4 groups: a semi confinement system with a low stocking rate (T1) for 70 days; a semi confinement system with a high stocking rate (T2) for 70 days; confinement for 28 days, followed by semi confinement with a low stocking rate (T3) for 42 days; and confinement for 28 days, followed by semi confinement with a high stocking rate (T4) for 42 days. Body weight, dry matter intake, blood collection, and faecal analysis of nematode eggs were performed for each animal. The stocking rates did not influence body weight gain for the semiconfined lambs (T1 and T2) or for the confined/semiconfined lambs (T3 and T4). All treatments had lower EPG counts at the end of the experimental period. The nematode genera identified during the experiment trial were Haemonchus and Trichostrongylus. There were no significant differences (P > 0.05) for packed cell volume, total plasma protein, and eosinophil means between treatments. These results suggested that the confinement followed by semi confinement in an ICLS, was an option for lamb production during the finishing phase which could be useful in reducing GIN without affecting the production performance. RESUMO: O objetivo deste experimento foi avaliar o desempenho produtivo, infecção por nematódeos gastrointestinais (GIN) e variáveis sanguíneas selecionadas, em cordeiras Corriedale durante a fase de terminação em sistema integrado lavoura-pecuária (ICLS) com duas taxas de lotação. O experimento utilizou 52 cordeiras Corriedale divididas em quatro grupos: sistema de semiconfinamento com baixa taxa de lotação por área (T1) por 70 dias; sistema de semiconfinamento com alta lotação por área (T2) por 70 dias; confinamento por 28 dias, seguido de semiconfinamento com baixa lotação por área (T3) por 42 dias; e confinamento por 28 dias, seguido de semiconfinamento com alta lotação por área (T4) por 42 dias. Peso corporal, ingestão de matéria seca, coleta de sangue e análise fecal para ovos de nematódeos foram avaliados para cada animal. As taxas de lotação não influenciaram no ganho de peso dos cordeiros semiconfinados (T1 e T2) ou nos confinados/semiconfinados (T3 e T4). Abstract Objective: To describe the prevalence of perineal laceration, based on the self-reported perception of postpartum women, and to analyze factors associated with its occurrence in Brazil. Methods: This was a cross-sectional study conducted with 23,894 postpartum women, excluding twin pregnancies, cesarean sections, and births with episiotomies, between 2011 and 2012. Prevalence ratios (PR) and 95% confidence intervals (95%CI) of association between the event and maternal, fetus/newborn, obstetric and clinical management characteristics were estimated in hierarchical Poisson regression models. Results: Out of 4,606 postpartum women, 49.5% (95%CI 46.1;42.9) self-reported perineal laceration. Being an adolescent (PR = 1.12; 95%CI 1.02;1.25), primipara (PR = 1.47; 95%CI 1.33;1.63), having had excessive gestational weight gain (PR = 1.17; 95%CI 1.07;1.29) and having undergone the Kristeller maneuver (PR = 1.18; 95%CI 1.08;1.29) increased the proportion of the outcome. Resumo Objetivo: Descrever a prevalência da laceração perineal segundo a percepção autorrelatada da puérpera, e analisar os fatores associados à sua ocorrência no Brasil. Métodos: Estudo transversal conduzido em 23.894 puérperas, excluindo-se gestações gemelares, cesarianas e partos com episiotomias entre 2011 e 2012. Razões de prevalência (RP) e intervalos de confiança de 95% (IC95%) da associação entre o evento e as características maternas, feto/recém-nato, obstétricas e manejo clínico foram estimadas em modelos de regressão de Poisson hierarquizados. Resultados: Entre 4.606 puérperas, 49,5% (IC95% 46,1;42,9) autorrelataram laceração perineal. Ser adolescente (RP = 1,12; IC95% 1,02;1,25), primípara (RP = 1,47; IC95% 1,33;1,63), ter tido ganho de peso gestacional excessivo (RP = 1,17; IC95% 1,07;1,29) e ter sido submetida à manobra de Kristeller (RP = 1,18; IC95% 1,08;1,29) elevaram a proporção do desfecho. Resumen Objetivo: Describir la prevalencia de laceración perineal, a partir de la percepción autoinformada de puérperas, y analizar los factores asociados a su aparición en Brasil.
