This study aimed to isolate, purify and characterize secondary metabolites present in Amburana acreana leaves, native to the state of Rondônia, using classical chromatographic methods and 1D, 2D nuclear magnetic resonance spectroscopy. From the hydroethanolic leaves extract of A. acreana, p-hydroxybenzoic acid, vanillic acid, coumarin, campesterol 3-β-D-glucoside and amburoside B, all known compounds, were identified. To the best of our knowledge, this is the first report of isolation and chemical characterization of secondary metabolites in A. acreana. The in silico study aimed to suggest phytochemicals from Commiphora leptophloeos leaves essential oil as therapies against coronavirus disease (COVID-19) (or severe acute respiratory syndrome coronavirus 2, SARS-CoV-2). Fruits of Schinus terebinthifolia Raddi were investigated. A fast and efficient method for the simultaneous determination of 11 polyphenolic substances by high performance liquid chromatography-diode array detector (HPLC-DAD) was applied. Species from Pavonia genus (Malvaceae) are traditionally used as antioxidant, n anti inflammatory and antimicrobial remedies. RESUMO Regiões com climas semiáridos apresentam condições ambientais que resultam em estresses abióticos às plantas. A maior área de produção de melão (Cucumis melo L.) do Brasil, o Estado do Rio Grande do Norte, possui essas características climáticas. O uso de bioestimulantes nesses sistemas de produção pode mitigar os efeitos dos estresses abióticos e aumentar a produtividade e a qualidade dos frutos. O objetivo neste estudo foi avaliar as características fisiológicas e de produção do meloeiro, sob aplicação de bioestimulantes. Os bioestimulantes Acadian®, Folicist® e Nov@® foram aplicados e comparados ao tratamento controle (sem bioestimulantes), em melões ‘Goldex’ e ‘McLaren’. Nov@® proporcionou maior assimilação de CO2, transpiração e condutância estomática, assim como ascorbato peroxidase em ‘Goldex’ e ‘McLaren’. ABSTRACT Regions with semi-arid climates have environmental conditions that result in abiotic stress to plants. The largest melon (Cucumis melo L.) production area in Brazil, the state of Rio Grande do Norte, has these climatic characteristics. Use of biostimulants in these production systems can mitigate effects of abiotic stress and increase yield and fruit quality. The objective in this study was to evaluate the physiological and production characteristics of melon, under the application of biostimulants. The biostimulants Acadian®, Folicist® and Nov@® were applied and compared to the control treatment (no biostimulants), in ‘Goldex’ and ‘McLaren’ melons. Nov@® led to higher CO2 assimilation, transpiration and stomatal conductance, as well as ascorbate peroxidase, in ‘Goldex’ and ‘McLaren’. Greater catalase was obtained with the application of Nov@®, while the use of Folicist® resulted in higher malondialdehyde content. The cultivar ‘McLaren’ exhibited the best physiological and biochemical performance, and ‘Goldex’ the best yield. Folicist® promotes physiological adaptations to stress without impairing photosynthetic activity. RESUMO O balanço hídrico é uma ferramenta com diversas aplicações na agricultura, incluindo a avaliação da adequação de um ambiente para o cultivo de uma determinada cultura, através da compreensão de como os recursos hídricos operam no sistema. Esta informação é vital para determinar se a água disponível satisfaz a necessidade da cultura. Assim, objetivou-se calcular o balanço hídrico em um Argissolo Vermelho-Amarelo para o cultivo de Coffea canephora no município de Cruzeiro do Sul, Acre, utilizando uma série histórica de 36 anos para avaliar o potencial e as restrições da cultura de acordo com a água disponível. Os resultados mostraram que, de forma geral, o cultivo de Coffea canephora é adequado para a região de estudo. Os períodos de déficit hídrico e de excesso de água são bem definidos, ocorrendo de abril a agosto e de setembro a março, respectivamente. Verificou-se que apenas a fase de floração do cafeeiro coincide com o período de déficit hídrico na região, exigindo assim irrigação para evitar comprometer essa etapa do desenvolvimento do fruto. Em relação ao Indice de Satisfação da Necessidade de Água (ISNA), observou-se que, quando calculado em uma escala mensal, indica baixo risco climático para o cultivo de Coffea canephora na região. ABSTRACT Water balance is a tool that has various applications in agriculture, including assessing whether an environment is suitable for growing a specific crop by providing information on how water resources function in the system. This information is essential for determining whether the available water satisfies the crop’s demand. Thus, the objective of this work was to calculate the water balance of Typic Hapludults for growing coffee (Coffea canephora) crops in Cruzeiro do Sul, Acre, Brazil, using a 36-year historical series to assess the potential and limitation of the crop according to the water availability. The results showed that coffee crops are, in general, suitable for growing in the study region. Water deficit and water surplus periods are well-defined, from April to August and September to March, respectively. Flowering was the only stage of coffee crops that coincides with the water deficit period in the region; thus, irrigation is necessary to prevent compromising this stage and fruit development. The Water Requirement Satisfaction Index (WRSI), calculated on a monthly basis, indicates a low climate risk for coffee crops in the region. However, when calculated on a daily basis, the WRSI shows a medium to high climate risk for coffee crops in several periods. The present work aims to study the synthesis and application of waste-made magnetic nanoparticles in the recovery of adulterated markings. A set of analytical-grade materials and metallic waste magnetic nanoparticles were synthesized for comparison purposes. Chemical characterization tests by X-ray fluorescence, morphological tests by scanning electron microscopy (SEM), and magnetization tests by vibrating sample magnetometer (VSM) were performed. The efficacy of the synthesized nanoparticles was tested on 1020 steel plates previously engraved and adulterated by sanding. Asphaltene aggregation mechanism is still a nebulous and controversial issue. Many authors argue that the aggregation of its components is due to π-stacking interactions, nonetheless, alkyl chains, heteroatoms, and even acidic groups are also constituents of asphaltenes and can play an important role in the chemistry of asphaltene. Excimers from pyrene are easily detected by fluorescence and were used in the current study to probe the effect of the side chain on the π-stacking capacity of pyrene. Pyrene was acylated with acyl chains varying from two to twelve carbon atoms, and the effect of the variation of the acyl chain length on excimer formation was followed by fluorescence emission spectroscopy. The results revealed that as the side chain grows from two to twelve carbon atoms the excimer/monomer ratio decreases and the excimer heat of formation drops from ΔH = -6.00 to -1.29 kcal mol-1. Resumo Introdução: O papel da tomografia computorizada torácica de ultrabaixa dose (ULDCT) em contexto de urgência é desconhecido. Objetivo: Este estudo avaliou a viabilidade da ULDCT em adultos na urgência com suspeita de pneumonia. Foram medidos a dose de radiação, o tempo entre a solicitação e os relatórios do exame, prescrições de antimicrobianos também foram correlacionadas. Métodos: Este estudo avaliou a viabilidade da ULDCT em adultos na urgência com suspeita de pneumonia. Foram medidas a dose de radiação, o tempo entre a solicitação e a disponibilização dos relatórios do exame, e também foram correlacionadas as prescrições de antimicrobianos. Entre outubro de 2017 e dezembro de 2018, incluímos prospectivamente adultos com suspeita de pneumonia adquirida na comunidade, sem diagnóstico definitivo por avaliação clínica. Os critérios de exclusão abrangeram condições clínicas que pudessem afetar a interpretação da imagem. Resultados: Foram incluídos 131 pacientes elegíveis (idade média de 43,5 anos; 57% homens) encaminhados para ULDCT. Dezoito (13,7%) foram excluídos. A dose média de radiação da ULDCT foi de 0,14 mSv. O intervalo de tempo entre a solicitação médica e o resultado da ULDCT foi de 43 minutos. Conclusão: A ULDCT mostrou-se viável para pacientes adultos com suspeita de pneumonia em contexto de urgência. Os relatórios foram disponibilizados num intervalo razoável e os resultados negativos foram associados a uma baixa taxa de prescrição de antibióticos.

Abstract Background: The role of ultra-low-dose chest CT (ULDCT) in the emergency department is unknown. Purpose: This study sought to examine feasibility of ULDCT in adults presenting to the emergency department for whom pneumonia was suspected. Radiation dose and time interval between medical request and ULDCT results were measured, antibiotic prescriptions were correlated. Methods: Between October 2017 and December 2018, we prospectively enrolled consecutive healthy adult patients in a single-center for whom community-acquired pneumonia was suspected and for whom no definitive diagnosis was possible by clinical judgment. Exclusion criteria comprised all chronic conditions that could impact severity and interpretation of image. Results: 131 eligible patients (mean age 43.5 years; 57% men) were enrolled to ULDCT. Eighteen (13.7%) patients were excluded. Average CT radiation dose was 0.14 mSv. Time interval between medical request and ultra-low-dose chest CT result was 43 minutes. Only 15.9% of patients with negative CT received antibiotics. Conclusion: Ultra-low-dose chest CT was feasible for almost all patients in a group of healthy adults presenting to the emergency department with suspected community-acquired pneumonia. Results were made available in a reasonable interval, and negative ones were associated with a low antibiotic prescription rate.

Foi conduzido um experimento de campo, delineado em blocos casualizados completos, onde foram testadas cinco doses de Se (0, 15, 30, 45 e 60 g ha-1), por aplicação foliar, em dois híbridos de cebola (Andrômeda e Rio das Antas). Foram analisadas variáveis de crescimento, nutricional e produtividade. O Se aumentou a altura e o acúmulo de massa seca em plantas de cebola, com máxima na dose 60 g ha-1 de Se, mesma dose em que foi obtida a maior produtividade comercial (78,15 t ha-1) e total (86,03 t ha-1) de bulbos em Rio das Antas, todavia, para Andrômeda, não ocorreu efeito do Se sobre a produtividade. As doses de Se também aumentaram o teor desse elemento nos bulbos. ABSTRACT The common climatic characteristics in semi-arid regions can cause abiotic stress and be a limiting factor for crops. Under these conditions, the use of beneficial elements such as selenium (Se) is an alternative to mitigate the harmful effects of abiotic stresses and increase yield. Therefore, this research aimed to evaluate the agronomic performance of onion grown in a semi-arid region under the application of Se. A field experiment was performed in a randomized block design, where five doses of Se (0, 15, 30, 45, and 60 g ha-1), by foliar application, were tested on two onion hybrids (Andrômeda and Rio das Antas). Growth, nutritional and yield variables were analyzed. Se increased the height and accumulation of dry mass in onion plants, with a maximum at the 60 g ha-1 dose of Se, the same dose at which the highest commercial (78.15 t ha-1) and total (86.03 t ha-1) bulb yields were obtained in Rio das Antas; however, for Andrômeda, there was no effect of Se on yield. RESUMO O objetivo deste trabalho foi avaliar a eficiência de aplicação de diferentes pontas de bicos associada a taxas crescentes de aplicação. Foi conduzido um experimento de campo no delineamento em blocos ao acaso, em esquema fatorial 3 x 4, com quatro repetições. O primeiro fator consistiu em três tipos de bicos: leque plano simples, cone oco e cone oco com eletrificação das gotas; e o segundo fator por quatro taxas de aplicação (50, 100, 200 e 250 L ha-1). A tecnologia eletrostática e o cone oco aumentaram o volume depositado nas folhas dos estratos superior e médio do feijoeiro, independentemente da dose de aplicação utilizada. ABSTRACT The objective of this work was to evaluate the application efficiency of different nozzle tips associated with increasing application rates. A field experiment was conducted in a factorial 3 × 4 randomized block design with four replicates. The first factor consisted of three types of nozzle tips (simple flat fan, hollow cone, and hollow cone with the electrification of the drops), and the second factor constituted the four application rates (50, 100, 200, and 250 L ha-1). Resumo Este estudo objetivou avaliar os efeitos da temperatura e motilidade embrionária sobre o desenvolvimento esquelético de jacaré-do-pantanal (Caiman yacare). Os ovos foram incubados com 90% de umidade e empregou-se a temperatura de 29°C por 45 dias. Após, para a incubação do Grupo I a temperatura continuou em 29°C, mas associou-se à injeção de 4-aminopiridina (29°C-4AP, n = 15) aplicada nos dias 46, 47, 48 e 49, do Grupo II permaneceu em 29°C (n = 14) e do Grupo III elevou-se para 33°C (n = 14). A movimentação foi mensurada através do monitor digital Egg Buddy® nos dias 30, 35, 42, 49, 56 e 60 dias. Aos 60 dias, os embriões foram eutanasiados e coletadas amostras embrionárias. Na análise estatística não foram observadas diferenças entre os grupos para o fator temperatura sobre a motilidade embrionária no desenvolvimento esquelético. Em contraste, a motilidade evidenciou diferença estatística no dia 49 para o Grupo I (P < 0,001) e apresentou maiores proporções de nariz e mão. Esses dados demonstraram pela primeira vez que o aumento na motilidade, induzidos farmacologicamente resultam em divergências fenotípicas na proporção de segmentos anatômicos durante a ontogenia pré-natal, podendo alterar efetivamente a adaptação dos animais em ambientes específicos.

Abstract This study aimed to evaluate whether skeletal development of the Pantanal Caiman (Caiman yacare) is similarly influenced by temperature variation and controlled increases in embryo motility. All eggs were incubated at 90% humidity and 29 °C for the first 45 days. Thereafter, the incubation temperature was either maintained at 29 °C and embryos were treated with 4-aminopyridine (4-AP) on days 46, 47, 48, and 49 (Group I, 29 °C 4-AP, n = 15); maintained at 29 °C (n = 14; Group II); or at 33 °C (n = 14, Group III). Embryonic movement was measured using an Egg Buddy® digital monitor on days 30, 35, 42, 49, 56, and 60, at which point embryos were euthanized and samples were collected for analysis. No differences were observed between groups with varying incubation temperatures. In contrast, embryonic motility was greater in embryos treated with 4-AP (P < 0.001) on day 49, and this was associated with higher proportions of snout-vent and hand lengths. This study demonstrates for the first time that pharmacologically induced increases in embryo motility result in phenotypic changes to the proportion of elements during prenatal ontogeny, thereby effectively altering the adaptation of the species to specific environments.

Resumo As armadilhas extracelulares de neutrófilos (NETs) foram relatadas pela primeira vez como uma estratégia microbicida para neutrófilos ativados. Por meio de uma resposta imunológica contra vários estímulos, os neutrófilos liberam seu DNA ligado a proteínas de grânulos, núcleo e citoplasma (por exemplo, elastase e mieloperoxidase). Até o momento, os NETs têm sido implicadas em danos aos tecidos durante processos inflamatórios intensos, principalmente quando sua liberação depende da geração de radicais de oxigênio. Os flavonóides são agentes antioxidantes e anti-inflamatórios, e destes, a quercetina é comumente encontrada em nossa dieta diária. Portanto, a quercetina pode exercer alguma atividade protetora contra o dano tecidual induzido por NETs. Em nossos ensaios in vitro, a quercetina reduziu a liberação das NETs, mieloperoxidase (MPO) e elastase a partir de neutrófilos estimulados com forbol 12-miristato 13-acetato (PMA). A atividade dessas enzimas também foi diminuída na presença de quercetina. A quercetina também reduziu o efeito citotóxico dos NETs sobre células alveolares (linha celular A549). Além disso, os ensaios in silico indicaram interações favoráveis entre a quercetina e as proteínas da NET (MPO e elastase). No geral, nossos resultados demonstram que a quercetina diminui os efeitos deletérios das NETs, reduzindo sua liberação à partir de neutrófilos ativados e diminuindo a atividade enzimática de MPO e elastase, possivelmente por meio de interação direta.

Abstract Neutrophil extracellular traps (NETs) were first reported as a microbicidal strategy for activated neutrophils. Through an immunologic response against several stimuli, neutrophils release their DNA together with proteins from granules, nucleus, and cytoplasm (e.g., elastase and myeloperoxidase). To date, NETs have been implicated in tissue damage during intense inflammatory processes, mainly when their release is dependent on oxygen radical generation. Flavonoids are antioxidant and anti-inflammatory agents; of these, quercetin is commonly found in our daily diet. Therefore, quercetin could exert some protective activity against tissue damage induced by NETs. In our in vitro assays, quercetin reduced NETs, myeloperoxidase (MPO), and elastase release from neutrophils stimulated with phorbol 12-myristate 13-acetate (PMA). The activity of these enzymes also decreased in the presence of quercetin. Quercetin also reduced the cytotoxic effect of NETs on alveolar cells (A549 cell line). Further, in silico assays indicated favorable interactions between quercetin and NET proteins (MPO and elastase). Overall, our results demonstrate that quercetin decreases deleterious cellular effects of NETs by reducing their release from activated neutrophils, and diminishing the enzymatic activity of MPO and elastase, possibly through direct interaction.
