RESUMO: Objetivou-se, na construção deste trabalho, detectar Anaplasma marginale por meio do esfregaço sanguíneo e Nested PCR (nPCR) e comparar os resultados com os sinais clínicos apresentados pelos bezerros em uma propriedade leiteira localizada no município de Castanhal, região nordeste do estado do Pará (1°07’19,1”S e 47°53’53,0”W), Amazônia Oriental. Para isso, foram coletadas 192 amostras sanguíneas de 24 animais, divididos em três períodos: 1-20, 21-41 e 42-60 dias de idade. Foram realizados esfregaços sanguíneos e nPCR com iniciadores para o gene msp5. A prevalência de A. marginale foi de 61,46% (118/192) tanto para a técnica de esfregaço sanguíneo quanto para nPCR (msp5). A manifestação de sinais clínicos da anaplasmose também foi significativamente crescente ao longo do estudo (P < 0,0001), sendo menor em animais de 1 a 20 dias, mas expandindo-se entre os de 21 a 41 dias e 42 a 60 dias, esses sinais foram caracterizados por apatia, febre, perda de peso, diarreia, desidratação e mucosas hipocoradas. Quanto a avaliação das técnicas diagnósticas, não houve diferença significativa entre a detecção de A. marginale no esfregaço sanguíneo e na nPCR (P = 0,995), porém se observou aumento riquetsêmico do agente no 47º dia (P < 0,01) em ambos os testes, demonstrando, assim, um padrão de aumento da riquetsemia próximo ao linear ao longo dos 60 dias, com consequente diminuição do volume globular. Assim, demonstra-se que dos 24 animais estudados, 21 se infectaram em algum momento do período estudado, e não houve diferença significativa entre esfregaço sanguíneo e nPCR, em virtude, provavelmente, das parasitemias médias e altas, as quais estiveram diretamente relacionadas com os sinais clínicos e a diminuição do volume globular. ABSTRACT: This study detected Anaplasma marginale in calvesusing blood smears and nested PCR (nPCR) and to compare the results with the clinical signs presented by calves on a dairy farm in the municipality of Castanhal, located northeast of the state Pará (1°07’19.1”S and 47°53’53.0”W), eastern Amazon. To this end, 192 blood samples were collected from 24 animals at 1-20, 21-41 and 42-60 days of age. Blood smears and nPCR with primers for the msp5 gene were performed. The prevalence of A. marginale was 61.5% (118/192) for the blood smear technique and nPCR (msp5). The manifestation of clinical signs of anaplasmosis also increased significantly over the course of the study (P < 0.0001), being lower in animals aged 1-20 days, but increasing among those aged 21-41 and 42-60 days. These signs were characterized by apathy, fever, weight loss, diarrhea, dehydration, and hypochromic mucous membranes. Regarding the evaluation of the diagnostic techniques, no significant difference was observed in the detection of A. marginale between the blood smear and nPCR (P = 0.995), but the agent’s rickets increased on Day 47 (P < 0.01) in both tests, thereby demonstrating a near-linear pattern of increase in rickets over the 60 days, with a consequent decrease in globular volume. This shows that of the 24 animals studied, 21 were infected at some point during the study period. Additionally, there was no significant difference between blood smears and nPCR, probably due to medium and high parasitemia, which were directly related to the clinical signs and decrease in globular volume. Abstract Background Brazil faces notable Breast Cancer (BC) mortality despite lower incidence rates versus developed countries. Despite guidelines from medical societies, Brazilian public policy recommends biennial mammographic screening for women aged 50 to 69. This study investigates sociodemographic and clinical factors related to BC detection methods and clinical stage at diagnosis. Methods The authors conducted a cross-sectional study at a São Paulo tertiary hospital. Patients were divided into ‘symptomatic’ and ‘mammographic’ detection groups. Bivariate analyses by detection method and clinical stage compared groups' profiles in terms of sociodemographic and clinical characteristics. Poisson regression analyses assessed sociodemographic and molecular subtypes´ influence on “mammographic detection” prevalence and “advanced-stage BC”, reporting prevalence ratios and 95 % Confidence Intervals. Results The authors studied 1,536 BC patients admitted from January 2016 to December 2017. The “mammographic detection” group had a higher proportion of patients aged 50‒69 years (62.9 % vs. 44.1 %), white race (63.3 % vs. 51.6 %), Catholic religion (58.2 % vs. 51.1 %), and Luminal A subtype (25.2 % vs. 13.2 %) compared to the “symptomatic detection” group. Patients with early-stage disease were more likely to have higher education levels (8.1 % vs. 5.5 %) and be married (39.8 % vs. 46.6 %) compared to those with advanced-stage. Molecular subtypes were significantly associated with the detection method and stage. The prevalence of advanced-stage disease in "mammographic" (n=313) and "symptomatic" (n=1191) groups was 18.5 % and 55 %, respectively . Mammographic detection significantly reduced advanced-stage BC prevalence (PR = 0.40, 95 % CI 0.31‒0.51). Conclusion Mammographic detection reduces advanced-stage breast cancer prevalence in Brazil, emphasizing the importance of regular screenings, especially among at-risk sociodemographic groups. Enhancing mammographic screening accessibility, lowering the starting age to 40, and extending coverage to include annual mammograms can significantly lower breast cancer mortality in Brazil, benefiting public health and patient outcomes. Abstract Introduction and hypothesis Internal vaginal pessary is among the leading treatments for pelvic organ prolapse (POP); however, it has a high adverse event rate. An external pessary was recently developed as an alternative. The study's objective was to compare the efficacy of external and internal pessaries in treating POP in postmenopausal women. Methods This parallel randomized (1:1 ratio) open-blind study included 40 symptomatic women with stage 2 or 3 POP. They were randomized into two groups: group 1 (internal pessary) and group 2 (external pessary) (n = 20 in each); and evaluated at the start of and 3 months after the treatment. Statistical analysis was performed to compare the results within and between the groups before and after the 3-month treatment. Results The groups were homogeneous, except for the variables previous pregnancies (p = 0.030) and POP-Q score of apical prolapse (p = 0.023) whose values were higher in group 2. A significant improvement in quality of life was observed in both groups after 3 months of follow-up; however, internal pessaries were found to be more effective (p < 0.001). In group 1 there were differences between the initial and final POP-Q scores of anterior (0.004) and apical prolapse (p = 0.005). The complication rate associated with internal pessary use was high (p = 0.044). Conclusions The present data suggested that external pessaries have a similar effect to internal ones for the treatment of POP and improvement of the quality of life of postmenopausal women. Abstract Objective The present study aimed to investigate FOXO3a deregulation in Uterine Smooth Muscle Tumors (USMT) and its potential association with cancer development and prognosis. Methods The authors analyzed gene and protein expression profiles of FOXO3a in 56 uterine Leiomyosarcomas (LMS), 119 leiomyomas (comprising conventional and unusual leiomyomas), and 20 Myometrium (MM) samples. The authors used techniques such as Immunohistochemistry (IHC), FISH/CISH, and qRT-PCR for the present analyses. Additionally, the authors conducted an in-silico analysis to understand the interaction network involving FOXO3a and its correlated genes. Results This investigation revealed distinct expression patterns of the FOXO3a gene and protein, including both normal and phosphorylated forms. Expression levels were notably elevated in LMS, and Unusual Leiomyomas (ULM) compared to conventional Leiomyomas (LM) and Myometrium (MM) samples. This upregulation was significantly associated with metastasis and Overall Survival (OS) in LMS patients. Intriguingly, FOXO3a deregulation did not seem to be influenced by EGF/HER-2 signaling, as there were minimal levels of EGF and VEGF expression detected, and HER-2 and EGFR were negative in the analyzed samples. In the examination of miRNAs, the authors observed upregulation of miR-96-5p and miR-155-5p, which are known negative regulators of FOXO3a, in LMS samples. Conversely, the tumor suppressor miR-let7c-5p was downregulated. Conclusions In summary, the outcomes of the present study suggest that the imbalance in FOXO3a within Uterine Smooth Muscle Tumors might arise from both protein phosphorylation and miRNA activity. FOXO3a could emerge as a promising therapeutic target for individuals with Unusual Leiomyomas and Leiomyosarcomas (ULM and LMS), offering novel directions for treatment strategies. Abstract Background: Melatonin is a hormone produced by the pineal gland and it has antioxidant properties. Aim: This study aimed to evaluate the effects of melatonin on assisted reproductive technologies through a systematic review and a meta-analysis. Materials and methods: Search strategies were used in PubMed and in other databases covering the last 15 years. After screening for eligibility, 17 articles were selected for the systematic review. For the meta-analysis statistics, two groups were formed, the treatment group (with melatonin) and the control group (without melatonin) for various assisted reproduction outcomes. Results: The main results were that no statistical differences were found concerning the clinical pregnancy outcome (p = 0.64), but there was a statistical difference with respect to Mature Oocytes (MII) (p = 0.001), antral follicle count (p = 0.0002), and the fertilization rate (p ≤ 0.0001). Conclusions: Melatonin had beneficial effects such as the improvement in the fertilization rate, although the authors did not obtain significance in the clinical pregnancy rate. SUMMARY OBJECTIVE: Stroke is a chronic health problem that affects all areas of life. The presence of thyroid autoantibodies can augment the severity of stroke. The aim of this work is to investigate whether there is a relationship between the site of stroke involvement and the anti-thyroid peroxidase antibody (anti-TPO) or not. This is the first study in the English-language literature. METHODS: A total of 39 patients with a diagnosis of acute ischemic stroke were included, and the cases under 18 years of age with an infection and the ones with autoimmune diseases other than Hashimoto's thyroiditis were excluded from the study design. The patients’ age, gender, smoking status, comorbid conditions, and stroke localization in brain imaging were recorded. The region involving the anterior circulation area originating from the internal carotid artery was evaluated as anterior, and the region possessing the vertebrobasilar circulation area from the vertebral arteries was considered posterior involvement. Thyroid-stimulating hormone (TSH), triiodothyronine (T3), thyroxine (T4), triglyceride, high-density lipoprotein (HDL), low-density lipoprotein (LDL), C-reactive protein (CRP), sedimentation, and anti-TPO were retrospectively analyzed. RESULTS: As a consequence, gender distribution, smoking, comorbid conditions, TSH, T3, T4, triglyceride, HDL, LDL, CRP, and sedimentation did not differ significantly, while the age of the posterior-located stroke was lower than that of the cases with the anterior. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals.
