Resumo Antecedentes Pacientes com doença grave por coronavírus-19 (COVID-19) podem necessitar do uso de ventilação mecânica (VM) invasiva por um período prolongado. Parâmetros agressivos de VM têm sido associados a alterações na pressão intracraniana (PIC) em pacientes com doenças intracranianas agudas. Elevações significativas da PIC podem comprometer a complacência intracraniana (CIC) e a hemodinâmica cerebrovascular (HVC). No entanto, os efeitos desses parâmetros em indivíduos sem doenças neurológicas ainda não foram sistematicamente avaliados. Objetivo Avaliar a CIC em pacientes em VM com COVID-19 comparados com outros diagnósticos, para melhor caracterizar os efeitos da VM e COVID-19 sobre a CIC. Também foi feita a comparação entre a CIC em pacientes com COVID-19 sem VM e voluntários saudáveis, para avaliar o efeito isolado da COVID-19 sobre a ICC. Métodos Trata-se de um estudo exploratório, observacional com amostra por conveniência. A CIC foi avaliada com um dispositivo não invasivo de monitoramento da PIC. A relação P2/P1 foi calculada dividindo-se a amplitude desses dois pontos, sendo definida como "anormal" quando P2 > P1. A análise estatística foi realizada usando um modelo linear misto com efeitos aleatórios para comparar a relação P2/P1 nos quatro grupos no primeiro dia de monitoramento. Resultados Uma amostra de conveniência com 78 voluntários (15 COVID-19 em VM, 15 sem COVID-19 em VM, 24 com COVID em respiração espontânea e 24 saudáveis) foram prospectivamente incluídos. Não houve diferença nas razões P2/P1 entre pacientes em VM com e sem COVID-19, nem entre pacientes sem VM com COVID-19 ou saudáveis. Abstract Background Patients with severe coronavirus disease-19 (COVID-19) may require the use of invasive mechanical ventilation (MV) for prolonged periods. Aggressive MV parameters have been associated with changes in intracranial pressure (ICP) in patients with acute intracranial disorders. Significant ICP elevation could compromise intracranial compliance (ICC) and cerebrovascular hemodynamics (CVH). However, the effects of these parameters in individuals without neurological disorders have not yet been evaluated. Objective To evaluate ICC in patients on MV with COVID-19 infection compared to other diagnoses, to better characterize the effects of MV and COVID-19 upon ICC. We also compared between the ICC in patients with COVID-19 who did not require MV and healthy volunteers, to assess the isolated effect of COVID-19 upon ICC. Methods This was an exploratory, observational study with a convenience sample. The ICC was evaluated with a noninvasive ICP monitoring device. The P2/P1 ratio was calculated by dividing the amplitude of these two points, being defined as "abnormal" when P2 > P1. The statistical analysis was performed using a mixed linear model with random effects to compare the P2/P1 ratio in all four groups on the first monitoring day. Results A convenience sample of 78 subjects (15 MV-COVID-19, 15 MV non-COVID-19, 24 non-MV-COVID-19, and 24 healthy participants) was prospectively enrolled. RESUMO OBJETIVO Apresentar a metodologia de estratificação das arboviroses urbanas (dengue, zika e chikungunya) pelo índice de receptividade territorial, instrumento de vigilância e controle dessas doenças que considera a heterogeneidade territorial intramunicipal. MÉTODOS Estudo ecológico que utiliza como unidade de análise as áreas de abrangência dos centros de saúde de Belo Horizonte (MG). Para a construção do índice de receptividade territorial foram selecionados indicadores de determinação socioambiental das arboviroses urbanas a fim de integrar à análise de componentes principais. As componentes resultantes foram ponderadas por análise de processos hierárquicos e agregadas por meio de álgebra de mapas. RESULTADOS O índice de receptividade territorial evidenciou grande heterogeneidade das condições de infraestrutura urbana. As áreas classificadas como alta e muito alta receptividade correspondem a aproximadamente 33% da área ocupada e se concentram sobretudo nas regiões de planejamento administrativo Leste, Nordeste, Norte, Oeste e Barreiro, principalmente em áreas limítrofes do município. Quando sobrepostas à densidade de casos de dengue e de ovos de Aedes em 2016, a estratificação pelo índice de receptividade territorial às arboviroses urbanas demonstra que áreas de muito alta receptividade apresentam uma densidade de casos, bem como de ovos de Aedes superior àquela observada nas demais áreas da cidade, o que corresponde a um percentual bastante reduzido do território municipal (13,5%). CONCLUSÕES As análises indicam a necessidade do desenvolvimento de ações de vigilância e controle adequadas para cada contexto, superando, assim, a lógica de alocação homogênea em todo o território.

CONCLUSION The analyses indicate the need for the development of adequate surveillance and control actions for each context, overcoming the logic of homogeneous allocation throughout the territory.

Métodos: Estudo transversal realizado em amostra representativa de residentes (≥12 anos) em Brumadinho, Minas Gerais, incluindo: não expostos (grupo referência); diretamente atingidos pela lama de rejeitos; e residentes em área de mineração. Foram estimadas as prevalências de resultados alterados de hemograma, colesterol total, colesterol lipoproteína de alta densidade (HDL), colesterol lipoproteína de baixa densidade (LDL), triglicérides, aspartato aminotransferase, alanina aminotransferase, creatinina, ureia, estimativa da taxa de filtração glomerular (TFGe) e proteína C-reativa ultrassensível (PCRus). As razões de prevalência (RP) e os intervalos de confiança de 95% (IC95%) de ter o exame alterado foram estimados por meio de modelos lineares generalizados com distribuição de probabilidade Poisson. Estimaram-se modelos brutos e ajustados por faixa etária, sexo, diabetes, índice de massa corporal, tabagismo, hipertensão. Methods: Cross-sectional study carried out on representative sample of residents (≥12 years) in Brumadinho, Minas Gerais, including: 1) non-exposed; 2) directly affected by tailings sludge; 3) residents in mining area. The prevalence of abnormal results of blood count, total, HDL and LDL cholesterol, triglycerides, aspartate aminotransferase, alanine aminotransferase, creatinine, urea, estimate of glomerular filtration rate (eGFR) and high-sensitivity C-reactive protein (hs-CRP) were estimated. The Prevalence Ratios (PR) and 95% Confidence Intervals (95%CI) of having an abnormal laboratory finding were estimated using Generalized Linear Models with Poisson probability distribution. Crude and adjusted models were estimated for age range, gender, diabetes, body mass index, smoking, hypertension. Results: After adjusting, there was no difference in PR between the three populations for most tests, with the exception of the population residing in an area with mining activity and not directly affected by the mud, with a lower chance of having altered total cholesterol (PR: 0.84; 95%CI 0.74–0.95) and a higher chance of having altered HDL cholesterol (PR: 1.26; 95%CI 1.07–1.50), hs-CRP (PR: 1.19; 95%CI 1.04–1.37), and eGFR <60mL/min/1,73 m2 (PR: 1.51; 95%CI 1.05–2.19). Conclusion: No significant differences were found in the prevalence of biochemical and hematological alterations between the populations directly exposed and not exposed to tailings. Abstract RNA interference (RNAi) is a powerful gene silencing technology, widely used in analyses of reverse genetics, development of therapeutic strategies and generation of biotechnological products. Here we present a free software tool for the rational design of RNAi effectors, named siRNA and shRNA designer (SSD). SSD incorporates our previously developed software Strand Analysis to construct template DNAs amenable for the large scale production of mono-, bi- and trivalent multimeric shRNAs, via in vitro rolling circle transcription. We tested SSD by creating a trivalent multimeric shRNA against the vitellogenin gene of Apis mellifera. RT-qPCR analysis revealed that our molecule promoted a decrease in more than 50% of the target mRNA, in a dose-dependent manner, when compared to the control group. Thus, SSD software allows the easy design of multimeric shRNAs, for single or multiple simultaneous knockdowns, which is especially interesting for studies involving large amounts of double-stranded molecules.

Abstract INTRODUCTION: Biomphalaria glabrata is considered to be responsible for the incidence of schistosomiasis in Brazil. Therefore, surveillance of areas where schistosomiasis is prevalent is fundamental for public health planning. This study was aimed to evaluate B. glabrata populations in water bodies of the city of Salvador, determine their distribution, estimate the prevalence of Schistosoma mansoni infections, characterize shed cercariae, and identify transmission foci. METHODS: Malacological surveys were carried out in 17 water collections from Salvador. Snail species were identified based on shell and mantle characteristics. Snails were evaluated for S. mansoni infection by exposure to light and via real time polymerase chain reaction (qPCR) using S. mansoni-18S rRNA subunit specific primers. RESULTS: 1,403 B. glabrata were collected. Classical cercarial shedding indicated that 5 snails (0.4%) were positive for S. mansoni. A higher prevalence of infections was found in Horta de Saramandaia (5.5%) and Lagoa do IAT (1.9%). Non-Schistosoma larvae, such as Xiphidiocercaria, Strigeidae, Spirorchiidae and Clinostomidae, were observed in 3.2% of the snails. S. mansoni DNA was detected in 6.2% snails via qPCR. CONCLUSIONS: B. glabrata is widely distributed in Salvador, as indicated by 7 water collections associated with a risk of schistosomiasis transmission. To our knowledge, this is the first study to identify B. glabrata eliminating cercariae of Clinostomidae, Strigeidae, and Spirorchiidae in Salvador. We propose that qPCR may be employed in combination with classical cercarial shedding. Estimating S. mansoni prevalence in snails by only considering the results of light exposure method classical into account may underestimate the problem.

ABSTRACT Plant-pollinator interactions and their reproductive implications are of central importance to the organization of plant populations and communities in restoration areas. We studied the breeding system and flower visitors of Pleroma trichopodum, a pioneer species of the Atlantic Forest. We attempted to answer three questions: (1) Is P. trichopodum dependent on pollinators and mates for reproduction? (2) What are the pollinators of P. trichopodum? (3) Do tree flower-density and flowering-tree density of P. trichopodum enhance the visitation rate of focal trees and their flowers? We tested the breeding system through pollination treatments. We performed focal observations on 10 trees and analyzed the relationship between tree and flower visitation rates, and the tree flower-density and flowering-tree density with Generalized Linear Mixed Models (GLMM). Pleroma trichopodum sets fruits by selfing and outcrossing, both of which occur only through pollinator visits. Bees visited P. trichopodum, and their visitation increased with tree flower-density. Thus, individuals with greater floral displays may function as magnet species, enhancing the pollination of nearby plant species. This characteristic, allied with the ability to reproduce without a mate (selfing) and a tolerance of soggy soils, make P. trichopodum a candidate for Atlantic Forest restoration.

O proprietário relatava histórico de abortos, nascimentos de crias fracas e mortalidade de animais adultos com perda progressiva de peso. Foram realizadas visitas à propriedade para obtenção do histórico, exame dos animais e coleta de sangue para realização do teste de Woo, hemogramas, testes sorológicos para pesquisa de anticorpos contra tripanossomose, leptospirose, e neosporose e PCR para diagnóstico molecular de Trypanosoma vivax. A identificação de animais com baixos valores no hematócrito foi a principal alteração hematológica identificada no rebanho. Dois animais foram positivos no teste de Woo, sendo visualizados tripanossomas em esfregaços sanguíneos, confirmados por meio de diagnóstico molecular como sendo T. vivax. Identificou-se que 95,23% (40/42) dos animais com hematócrito baixo foram sorologicamente positivos para T. vivax. As condições identificadas na propriedade, como ambiente propício aos vetores mecânicos, a presença de animais silvestres e a introdução de animais de estados onde já haviam sido registrados surtos de tripanossomose provavelmente estiveram associadas à introdução e disseminação do agente no rebanho. O elevado número de animais sorologicamente positivos para tripanossomose 82,51% (151/183) demonstra que praticamente todo o rebanho teve contato com o agente. O rápido estabelecimento das medidas de controle, entre elas a utilização das drogas tripanocidas, contribuiu para o controle do surto. O estudo permitiu comprovar a ocorrência de mais um surto de tripanossomíase tripanossomose no Brasil. O diagnóstico clínico da enfermidade foi dificultado pela semelhança dos sinais clínicos com outras enfermidades e pela possibilidade da associação de duas ou mais doenças no mesmo paciente, o que ressalta a importância do estabelecimento de medidas diagnósticas adequadas como forma de evitar a disseminação da enfermidade e minimizar as perdas econômicas dos produtores.

ABSTRACT: The objective of this study was to investigate the occurrence of trypanosomiasis in a dairy farm in municipality of Timon, State of Maranhão, Brazil. The owner reported abortus, births of weak calves, and mortality of adult animals with progressive weight loss. Visits to the property were carried out to obtain the history, realize animal examination and blood collection for the Woo test, hemograms, serological tests for trypanosomiasis, leptospirosis, and neosporosis and PCR for molecular diagnosis of Trypanosoma vivax. The identification of animals with low values in the hematocrit was the main hematological alteration identified in the herd. Two animals were positive in the Woo test and trypanosomes were visualized in blood smears, confirmed by molecular diagnosis as T. vivax. It was identified that 95.23% (40/42) of the animals with low hematocrit were serologically positive for T. vivax. The conditions identified in the property as an environment propitious to mechanical vectors, the presence of wild animals and the introduction of animals from states where trypanosomiasis outbreaks had already been reported were probably associated with the introduction and dissemination of the agent in the herd. The high number of serologically positive animals for trypanosomiasis 82.51% (151/183) shows that almost all the herd had contact with the agent. The rapid establishment of control measures, including the use of trypanocidal drugs, contributed to the control of the outbreak. The study allowed confirming the occurrence of another outbreak of trypanosomiasis in Brazil. The clinical diagnosis of the disease was difficult by the similarity of the clinical signs of trypanosomiasis with other diseases and the possibility of association of two or more diseases in the same patient, which emphasizes the importance of establishing adequate diagnostic measures as a way to avoid the dissemination of the disease and to minimize the economic losses of the producers.

The guarantee of long-term digital preservation will only be possible with the effective implementation of metadata standards or schemas, because these will determine the description, representation, consistency and persistence of the digital resource/object in information environments, as well as defining interoperability between systems. The adoption of metadata for preservation supports the management of processes for the archiving and maintenance of access to digital information in the long term, guaranteeing authenticity, integrity and reliability. In this scenario, the aim of this study was to identify and describe metadata standards or schemas that could be considered by institutions that are developing their institutional repositories, so that digital preservation might be addressed. To that end, a literature review, and its systematization and content analysis were carried out. The following metadata standards and schemas were selected and analyzed: DUBLIN CORE (DC), MODS, EAD, ANSI/NISO Z39.87, METS and PREMIS. The results indicate that the descriptive and technical metadata DC, MODS, EAD and ANSI/NISO Z39.87 or MIX have a role that is more directed to supporting METS and PREMIS, both in discovering and recording the technical aspects of digital objects, and in the proof of its provenance, authenticity, context and integrity. Encompassing descriptive, structural, administrative and preservation metadata of PREMIS, METS can manage digital objects acting as a Submission Information Package (SIP), Archival Information Package (AIP) or Dissemination Information Package (DIP) in an Open Archival Information System (OAIS).

A garantia de preservação digital a longo prazo só será possível com a adoção efetiva de padrões ou esquemas de metadados, pois são eles que determinarão a descrição, a representação, a consistência e a persistência do recurso/objeto digital no ambiente informacional, além de definir a interoperabilidade entre sistemas. A adoção de metadados para preservação apoia a administração dos processos relativos ao arquivamento e à manutenção do acesso à informação digital a longo prazo, com garantias de autenticidade, de integridade e de confiabilidade. Neste cenário, foi objetivo do trabalho identificar e descrever padrões ou esquemas de metadados que poderiam ser considerados por instituições que estão desenvolvendo seus repositórios institucionais, para que possam contemplar a preservação digital. Para tanto, foi desenvolvida pesquisa bibliográfica e sistematização e análise de conteúdo. Foram selecionados e analisados os seguintes esquemas e padrões de metadados: DUBLIN CORE, MODS, EAD, ANSI/NISO Z39.87, METS e o PREMIS. A análise dos resultados indica que os metadados descritivos e técnicos do DC, MODS, EAD e ANSI/NISO Z39.87 ou MIX, possuem um papel mais direcionado a amparar o METS e o PREMIS, tanto na descoberta e no registro dos aspectos técnicos do objeto digital, como na comprovação de sua procedência, autenticidade, contexto e integridade. Englobando metadados descritivos, estruturais, administrativos e de preservação do PREMIS, o METS pode gerir os objetos digitais atuando como um Pacote de Submissão de Informação (PSI), Pacote de Arquivamento de Informação (PAI) ou Pacote de Disseminação de Informação (PDI) num Sistema Aberto de Arquivamento de Informação (SAAI).

RESUMO: O objetivo deste trabalho foi avaliar a soroconversão em cães imunizados com as vacinas Leishmune® e Leish tec®, através do teste imunocromatográfico rápido DPP® (Dual Path Platform) (DPP LVC) e do ensaio imunoenzimático (EIE) durante um ano após o protocolo vacinal. Trata-se de um estudo onde 28 cães divididos em dois grupos foram imunizados cada um com uma vacina anti - LVC e acompanhados durante um ano através de avaliação clínica e exames laboratoriais. Foi possível acompanhar 22 (78.5%) cães. Nos exames dos tempos 1, 2 e 3 (respectivamente 30 dias, 6 messes e 1 ano após a vacinação) os resultados de todos os cães também foram negativos para LVC, exceto de um cão que recebeu a vacina Leish tec® e soroconverteu no DPP LVC no T2, após 6 meses a vacina. Os exames posteriores deste cão foram negativos. Os resultados do presente estudo demostraram que, em área não endêmica e mesmo em diferentes tempos de avaliação, cães vacinados contra LVC, independente da vacina utilizada, não foram capazes de soroconverter no protocolo utilizado pelo Ministério da Saúde brasileiro (DPP/EIE).

ABSTRACT: The objective of the current research was to assess seroconversion in dogs immunized with Leishmune® and Leish Tec® vaccines using rapid chromatographic immunoassay DPP® (Dual Path Platform) (DPP CVL) and enzyme immunoassay (EIE) up to one year after the vaccination protocol. The study sample comprised 28 dogs divided into two groups, each group immunized with an anti-CVL vaccine and clinically monitored for one year through clinical evaluation and laboratory tests. 22 (78.5%) dog were monitored. During the evaluation time (T1-30 days, T2-6 months, and T3-1 year after vaccination) the results for all dogs were negative for CVL, except for one animal vaccinated with Leish tec® that seroconverted in the DPP CVL test at T2. Subsequent examinations of this dog were negative. Our results showed that in a non-endemic area, even at different evaluation times, dogs vaccinated against CVL with Leishmune® or Leish tec® did not seroconvert in the serological protocol used by the Brazilian Ministry of Health (DPP/EIE).

RESUMO As doenças falciformes são as hemoglobinopatias mais frequentes mundialmente. Nas últimas décadas vivenciamos melhora na sobrevida de portadores destas patologias com a introdução de medidas preventivas e o uso precoce da hidroxiurea. O transplante de medula óssea alogênico (alo TMO) é a única opção terapêutica curativa para as hemoglobinopatias. O mesmo tem sido indicado para pacientes com complicações neurológicas, crises vasoclusivas repetidas, alguma lesão orgânica e alosensibilizados. O uso de doadores relacionados de medula óssea ou cordão umbilical mostrou em 1000 procedimentos realizados sobrevida global de 95% e sobrevida livre de ventos de 90%. O uso de doadores não aparentados não mostrou resultados tão expressivos, mas no futuro métodos melhores de tipagem de HLA e de medidas de suporte podem melhorar estes resultados. No Brasil apenas recentemente o alo TMO foi incluído no âmbito do sistema único de saúde (SUS) como opção terapêutica para portadores de doenças falciformes. O uso de doadores aparentados de MO ou de SCU com regime mieloablativo é considerado hoje tratamento estabelecido, sendo que o uso de doadores alternativos não aparentados ou haploidenticos e o uso de transplante com regime não mieloablativo deve ser considerado apenas em estudos clínicos.

SUMMARY Sickle-cell diseases are the most common inherited hemoglobinopathies worldwide. Improvement in survival has been seen in the last decades with the introduction of careful screening and prevention of complications and the introduction of hydroxyurea. Stem-cell transplantation is currently the only curative option for these patients and has been indicated for patients with neurological events, repeated vaso-occlusive crisis, any organ damage or presence of red blood cell antibodies. Related bone-marrow or cord-blood transplant has shown an overall survival of more than 90% with a disease-free survival of 90% in 1,000 patients transplanted in the last decades. The use of unrelated donors unfortunately has not shown the same good results, but better typing methods and improved support may improve the outcome with this source of stem cells in the future. In Brazil, only recently stem cell transplant from related donors has been included in the procedures performed in the public health system. The use of related bone marrow or cord blood and a myeloablative conditioning regimen are considered standard of care for patients with sickle-cell diseases. Transplants with non-myeloablative regimens, unrelated donors or haploidentical donors should be performed only in controlled clinical trials.

A cultura da pereira (Pyrus communisL.) possui grande potencial de expansão no sul do Brasil devido às condições climáticas e de solo. A carência de informações a respeito da(s) melhor(es) combinação(ões) de cultivares 'copa de pereiras europeias' e porta-enxertos de marmeleiro em relação à compatibilidade de enxertia é um dos fatores que tem limitado o cultivo de pera no país. A utilização de porta-enxertos de marmeleiros apresentam as vantagens de redução do vigor e rápida entrada em produção, porém, quando enxertados com algumas cultivares de 'pereira europeia', pode acarretar incompatibilidade de enxertia. O trabalho foi realizado com o objetivo de avaliar fenotipicamente a compatibilidade morfológica entre cultivares 'copa de pereiras europeias' e porta-enxertos de marmeleiro através das variáveis de crescimento das plantas, como o incremento de diâmetro (mm) do tronco da cultivar e do porta-enxerto; diferença de diâmetro entre a cultivar e o porta-enxerto; incompatibilidade 'translocada'; incompatibilidade 'localizada'; e análise da conexão vascular no ponto de enxertia pela imersão em solução corante de Ácido Fuccínico 0,08%. Os experimentos foram conduzidos em pomares de pereiras europeias na região de Urupema, estado de Santa Catarina, durante os ciclos de cultivo 2011/12, 2012/13 e 2013/14. As combinações de cultivares de pereiras europeias e porta-enxertos avaliados foram a 'Abbè Fetel'/'Adams', 'Decana du Comice'/'Adams', 'Clapp's Favourite'/'EMA', 'Rocha'/'Adams', 'Packham´s Triumph'/'EMA' e 'Santa Maria'/'Adams'. A combinação 'Clapp´s Favourite'/'EMA' apresentou incompatibilidade do tipo 'localizada' pela descontinuidade vascular na região de união do enxerto, impedindo a passagem do corante. As combinações 'Rocha'/'Adams' e 'Abbè Fetel'/'Adams' não apresentaram incompatibilidade de enxertia. Nessas combinações, foram observadas união vascular contínua entre as cultivares e os porta-enxertos. As combinações 'Santa Maria'/'Adams', 'Decana du Comice'/'Adams' e 'Packham´s Triumph'/'EMA' apresentaram boa união na zona de enxertia pela continuidade da linha de união na casca e no lenho. A combinação 'Clapp´s Favourite'/'EMA', devido às regiões de incompatibilidade encontradas, não é uma combinação recomendada nas condições experimentais avaliadas. As combinações 'Abbè Fetel'/'Adams' e 'Rocha'/'Adams' são as mais indicadas para plantio, devido à total compatibilidade encontrada entre a cultivar e o porta-enxerto. As combinações 'Santa Maria'/'Adams', 'Decana du Comice'/'Adams' e 'Packham´s Triumph'/'EMA' podem ser recomendadas, devido à compatibilidade parcial de enxertia, sendo indicadas para alta e super densidades de plantio.

The pear (Pyrus communis L.) crop offers an expansive market opportunity in southern Brazil, because of amicable climatic and soil conditions. However, the lack of studies on compatibility of european pear cultivars and rootstocks is one of the limiting factors on the development of the pear crop in southern Brazil. The use of quinces as rootstocks has the advantages of vigor reduction and earlier bearing trees. However, some of these combinations can present some incompatibility graft. The objective of this study was to evaluate phenotypicaly the morphological compatibility among european pear cultivars and quince rootstocks. The evaluated variable were the section increment of cultivars and rootstocks trunk diameter at the graft union; the diameter difference between scion and cultivars and rootstocks; 'translocated' incompatibility; 'located' incompatibility; and the vascular connection of the graft union by immersion of the base of the plants (under the graft union) in a 0.08% Fuccinic acid solution. The experiments were conducted in an european pear orchards in the region of Urupema municipality, state of Santa Catarina, during the 2011/12, 2012/13 and 2013/14 growing seasons. The european pear cultivars and rootstocks evaluated were: 'Abbé Fetel'/'Adams', 'Decana du Comice'/'Adams', 'Clapp's Favourite'/'EMA', 'Rock'/'Adams', 'Packham's Triumph'/'EMA' and 'Santa Maria'/'Adams'. 'The Clapp´s Favourite'/'EMA' combination showed a `located incompatibility by descontinued vascular graft region, preventing the dye translocation. 'The Rocha'/'Adams' and 'Abbè Fetel'/'Adams' combinations did not present graft incompatibility, showing continued vascular union and no vascular disruption between cultivars and rootstocks. The 'Santa Maria'/'Adams', 'Decana du Comice'/'Adams' and 'Packham´s Triumph'/'EMA' combinations showed a good graft union region with clear and continued bark and wood bond lines. The 'Clapp´s Favourite'/'EMA' combination showed high incompatibility and is not recomented for the experimental conditions evaluated. The 'Abbè Fetel'/'Adams' and 'Rocha'/'Adams' combinations showed the highest graft compatibility among cultivars and rootstocks and are the best recomended combinations. Additionly, the 'Santa Maria'/'Adams', 'Decana du Comice'/'Adams' and 'Packham´s Triumph'/'EMA' combinations showed partial compatibility and can eventually be indicated for high and super plant density when 'Abbè Fetel'/'Adams' and 'Rocha'/'Adams' combinations are not available.

Background β-Glucosidases catalyze the hydrolysis of cellobiose and cellodextrins, releasing glucose as the main product. This enzyme is used in the food, pharmaceutical, and biofuel industries. The aim of this work is to improve the β-glucosidase production by the fungus Lichtheimia ramosa by solid-state fermentation (SSF) using various agroindustrial residues and to evaluate the catalytic properties of this enzyme. Results A high production of β-glucosidase, about 274 U/g of dry substrate (or 27.4 U/mL), was obtained by cultivating the fungus on wheat bran with 65% of initial substrate moisture, at 96 h of incubation at 35°C. The enzymatic extract also exhibited carboxymethylcellulase (CMCase), xylanase, and β-xylosidase activities. The optimal activity of β-glucosidase was observed at pH 5.5 and 65°C and was stable over a pH range of 3.5-10.5. The enzyme maintained its activity (about 98% residual activity) after 1 h at 55°C. The enzyme was subject to reversible competitive inhibition with glucose and showed high catalytic activity in solutions containing up to 10% of ethanol. Conclusions β-Glucosidase characteristics associated with its ability to hydrolyze cellobiose, underscore the utility of this enzyme in diverse industrial processes.

O sucesso do tratamento endodôntico depende da correta determinação do comprimento de trabalho. O término do canal deve ser detectado com precisão e o controle do comprimento de trabalho durante o tratamento deve ser mantido. A proposta dessa investigação foi avaliar a capacidade do localizador foraminal Quill® determinar o comprimento de trabalho, a partir da localização do forame apical, estabelecido no presente estudo em 1 mm aquém do forame apical. Pacientes com indicação para extração dental por motivos ortodônticos ou periodontais foram selecionados, resultando em uma amostra de 24 canais. O termo de consentimento livre e esclarecido foi obtido de cada paciente antes dos tratamentos. Após a administração de anestesia local, a cavidade pulpar foi acessada. Em sequência, as leituras correspondentes ao forame apical mostradas no display do aparelho foram registradas e a lima removida. Com o uso de paquímetro digital, 1 mm foi subtraído da primeira medida. A lima com a nova medida foi introduzida no canal radicular novamente, fixada com resina flow e então o dente foi extraído. Após, uma das paredes do terço apical foi desgastada para vizualização da ponta do instrumento e forame apical. A distância da ponta da lima ao forame apical foi mensurada no MEV. A média das medidas foi de 1,089mm (±0,437mm). O teste t Bicaudal mostrou não haver diferenças significantes (p=0,338) entre os valores experimentais e um valor hipotético testado de 1 mm. O localizador foraminal Quill® foi capaz de determinar um satisfatório comprimento de trabalho para o tratamento endodôntico, estabelecido em 1 mm aquém do forame apical.

The success of an endodontic treatment relies on a correct working length determination. The canal terminus must be detected accurately and a precise control of the working length during the endodontic treatment must be maintained. The aim of this study was to investigate the working length determination in vivo with the Quill Apex Locator® for apical limit established at 1 mm short from the apical foramen (AF). Patients scheduled to dental extraction due to orthodontic or periodontal reasons were selected for this study, resulting in a sample of 24 root canals. Written informed consent was obtained from each patient before the treatment. After the administration of local anesthesia each tooth had its pulp cavity accessed. Next, the reading corresponding to the AF showed on the device's display was recorded and the file was removed. With the use of a digital caliper, 1 mm was subtracted from that first measurement. The file with the new measurement was introduced into to the root canal again then fixed with light curing flow composite and the tooth was extracted. Next, one of the apical third walls was worn out to visualize the instrument's point and the AF. The distance from the file tip to the AF was measured by scanning electron microscopy. The average of the measurements was 1.089±0.437 mm. The Bicaudal t-test showed no significant difference (p=0.338) between the experimental values and the hypothetical value tested at 1 mm. The present investigation confirmed that the Quill Apex Locator® was able to determine the working length with good reliability for the endodontic treatment, established at 1 mm short from the AF.
