Abstract This study highlights the importance of fungi, specifically Botrytis cinerea Pers., in the search for bioactive compounds with therapeutic potential. Extraction approaches using Soxhlet, and maceration methods were applied of the fungus to explore secondary metabolites production. The compound mannitol was separated from the crude extract through nuclear magnetic resonance. The results indicated a positive effect on the inhibitory action of the acetylcholinesterase enzyme for ethyl acetate fractions obtained from the broth. Additionally, significant cytotoxic effects were observed in neoplastic cell lines, with IC 50 values of 3.5 μg/mL, 5.6 μg/mL, and 8.5 μg/ mL for colon cancer cells, monocytes, and human glioma, respectively. Antimicrobial activity was also evident in B. cinerea extracts and fractions, particularly in the ethyl acetate fractions from the broth. Larvicidal activity was observed in the chloroform fractions of the broth, with CL50 values of 20,824 μg/mL and 83,401 μg/mL. Furthermore, morphological changes in larvae were observed when exposed to the fungus’s extracts and fractions. The results suggest that B. cinerea extracts and fractions have the potential to identify substances with applications in biological activities, such as cytotoxic, antimicrobial, and larvicidal actions. RESUMO Objetivos: correlacionar as variáveis sociodemográficas, econômicas, de trabalho, padrão do sono, nível de ansiedade e qualidade de vida ao desenvolvimento da Síndrome de Burnout em docentes do ensino superior. Métodos: estudo observacional, analítico, transversal e com abordagem quantitativa. A coleta dos dados ocorreu de agosto a novembro de 2022, com 140 docentes de uma instituição de ensino superior privada, da Amazônia Ocidental Brasileira. ABSTRACT Objectives: to correlate the development of Burnout Syndrome in higher education professors with the following variables: sociodemographic, economic, work, sleep pattern, level of anxiety and quality of life. Methods: observational, analytical, cross-sectional study with a quantitative approach. Data collection took place from August to November 2022, with 140 professors from a private higher education institution in the Western Brazilian Amazon. Results: professors who carried out activities outside institutional hours and who had sleep duration < 5 hours, presented lower scores in the personal fulfillment dimension of burnout, with (p=0.002) and (p=0.001), respectively. RESUMO Objetivo Demonstrar a predição do ar exalado dos pacientes submetidos à ventilação mecânica no diagnóstico à beira leito da pneumonia associada à ventilação mecânica. Métodos Foram coletadas amostras de ar através do ramo expiratório do circuito de ventilação mecânica ao longo da internação de pacientes na unidade de terapia intensiva do Hospital São José, em Criciúma (SC). O trabalho analisou 83 participantes subdivididos em dois grupos, com e sem pneumonia associada à ventilação mecânica. Resultados Por meio da análise de três padrões de ar identificados, conseguiu-se um valor preditivo para diagnóstico de pneumonia associada à ventilação mecânica. As análises das amostras das primeiras 12 horas de ventilação mecânica invasiva foram capazes de predizer a pneumonia associada à ventilação mecânica (p = 0,018). Entretanto, nenhuma das outras amostras de ar coletadas ao longo da internação dos pacientes foi capaz de caracterizar gravidade ou predizer, em precoce ou tardia, a pneumonia associada à ventilação mecânica. ABSTRACT Objective To predict exhaled air in patients undergoing mechanical ventilation during bedside diagnosis of ventilator-associated pneumonia. Methods Air samples were collected through the expiratory branch of the mechanical ventilation circuit during the hospitalization of patients at the intensive care unit of Hospital São José in Criciúma (SC), Brazil. In this study, 83 participants were divided into two groups, namely, the group with and the group without ventilator-associated pneumonia. Results The analysis of three air patterns revealed a predictive value for the diagnosis of ventilator-associated pneumonia. The analyses of samples from the first 12 hours of invasive mechanical ventilation were able to predict ventilator-associated pneumonia (p = 0.018). However, none of the other air samples collected during hospitalization were useful in identifying the severity or predicting early or late ventilator-associated pneumonia. Método: Se trata de un ensayo clínico aleatorizado, doble ciego, controlado y con un diseño prospectivo. Durante la realización de este estudio se incluyeron 43 mujeres divididas entre los grupos A (pomada sin nanopartículas de plata n = 23) y B (pomada con nanopartículas de plata n = 20) clínicamente sanas y usuarias del sistema unificado de salud. Resultados: No hubo diferencias significativas al comparar antes y después del uso de la pomada para la prueba de IVA (p = 0,15), la prueba de Schiller (p = 0,15), los cambios celulares (p = 0,47) y el análisis microbiológico (p = 0,89) mediante citología. ABSTRACT Objective: To assess the feasibility of incorporating technology as a new alternative for treating topics on cervical lesions. Method: This is a randomized, double-blind, controlled clinical trial with a prospective design. During the realization of this study, 43 women were included and divided between groups A (ointment without silver nanoparticles n = 23) and B (ointment with silver nanoparticles n = 20) clinically healthy and who used the unified health system. Results: There were no significant differences when comparing before and after the use of ointment for IVA test (p = 0.15), Schiller test (p = 0.15), cellular changes (p = 0.47) and microbiological analysis (p = 0.89) through cytology. After use, no adverse reaction was observed in the sample studied. Conclusion: Based on the results identified in this study, identified that the product is safe and does not promote adverse events. Abstract The Brazilian native species Cestrum intermedium, known as mata-boi, induces hepatotoxicity and death when ingested by cattle. While most studies on this species focus on toxicological features, our study is the first to describe the anatomy and in vitro biological activities of Cestrum intermedium. We investigated adult leaves and stems by histochemistry, described their anatomy, performed physical-chemical analysis, determined in vitro antioxidant and antimicrobial activities, and identified secondary metabolites. A few noteworthy anatomical features were the anomocytic stomata on the abaxial surface and the absence of trichomes, in addition to the circular shaped petiole with two projections on the adaxial surface. Histochemical analysis showed chemical markers such as alkaloids, usually reported as toxic, and terpenoids. Potassium nitrate (ATR-FTIR) and lupeol palmitate (NMR) were detected on the crude stem extract. Thermogravimetric and physical-chemical analysis provided fingerprint parameters for the species. Minimal Inhibitory Concentration (MIC) assay revealed that Staphylococcus aureus, Staphylococcus epidermidis, and Candida albicans were weakly inhibited by extract samples. Chloroform and ethyl acetate fractions presented high phenolic content, which resulted in in vitro antioxidant activity. These novel features expand the knowledge about this species, considering that previous studies mainly focused on its toxicity. Our study also provided characteristics that may help in avoiding misidentification between Cestrum members, especially when taxonomic keys cannot be employed, as in the absence of flowers and fruits.

Abstract The aim of this study was to evaluate the association of different concentrations of Trolox® and the addition of a fixed concentration of DHA in the freezing of semen of Mangalarga Marchador stallions. To that end, 16 ejaculates were frozen in the following extenders: E1) BotuCrio® (BC; Control); E2) BC + 50 ngml-1 DHA + 30 µM Trolox® (BCDHA30T); E3) BC + 50 ngml-1 DHA + 40 µM Trolox® (BCDHA40T); E4) BC + 50 ngml-1 DHA + 50 µM Trolox® (BCDHA50T). All the tested extenders were similar in preserving different kinematic parameters, cell functional integrity, compacted DNA, and high and intermediate mitochondrial activity (P>0.05). However, sperm cryopreserved in BCDHA40T showed higher velocities than sperm frozen in the control extender (P<0.05). The 30 µM concentration of Trolox® was worse for sperm motility and the 50 µM concentration of Trolox® did not adequately preserve the structural integrity of the membranes in an extender containing DHA when compared to the BotuCrio® (P<0.05) extender. The use of Trolox® in freezing extenders containing DHA did not maximize the effect of BotuCrio®, except for in the case of sperm velocity parameters when at a concentration of 40 µM.

Abstract Studies have shown that Caesalpinia pulcherrima extracts promote antioxidant, healing, immunomodulating and antiparasitic activities and its polysaccharides can be used as functional food. In this sense, this work had as objective the isolation and characterization of a polysaccharide-like pectin, extracted from the C. pulcherrima leaves and its possible applications as an antioxidant and immunomodulator agent. The molecule was characterized by high performance liquid chromatography, fourier transform infrared spectroscopy and nuclear magnetic resonance spectroscopy. Its antioxidant potential was evaluated through the methods of phosphomolybdenum, ABTS radical scavenging [2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid], DPPH (1,1-diphenyl-2-picrylhydrazyl) and nitric oxide radical. The immunostimulating effects of pectin were tested in splenocytes to evaluate its toxic, proliferative and cell activator and immunomodulatory potential. The polysaccharide obtained has structural characteristics similar to pectins. Pectin showed high in vitro antioxidant activity for ABTS radical scavenging, moderate activity for phosphomolybdenum and low activity for DPPH and nitric oxide. In vitro immunomodulation assays showed that pectin obtained did not promote a cytotoxic effect (viability > 90%). The increase in cytosolic ROS levels indicates a possible mechanism of cell activation without causing damage. Immunophenotyping showed that pectin increased a subpopulation of CD8+ T lymphocytes and monocytes. In addition, it promoted a mostly pro-inflammatory response confirmed by the production of cytokines IL-2, -4, -6, IFN-γ and TNF-α. These results reinforce the ethnopharmacological use of C. pulcherrima leaves and expand the use of this plant for future applications as herbal medicines.

Abstract Effects were assessed of the dilutants TRIS and ACP - 101c® with the addition of different guinea fowl (Numida meleagris) egg yolk concentrations. Fifteen ejaculates were collected from five goats of the Anglo Nubian breed. The ejaculates were pooled and then divided into 12 groups, two control groups (GC1 TRIS, with 2.5% Gallus gallus domesticus hen egg yolk GOGD), (GC2 Control Group ACP - 101c®, with the addition of 2.5% Gallus gallus domesticus hen egg yolk GOGD) and ten experimental groups (EG), containing TRIS and ACP added with different concentrations of egg yolk from guinea hen (Numida meleagris) (TRIS 2,5% GONM; TRIS 5% GONM; TRIS 10% GONM; TRIS 15% GONM; TRIS 20% GONM; ACP® 2,5% GONM; ACP® 5% GONM; ACP® 10% GONM; ACP® 15% GONM; ACP® 20% GONM). Then cryopreservation was carried out and the samples stored in liquid nitrogen (-196 °C). After seven days, the samples were thawed and assessed for spermatic kinetics, immunofluorescence and sperm morphology. Analysis of GOMN by the CASA system showed that the various parameters were similar to those of GOGD (P>0.05). The membrane integrity, mitochondrial potential and the acrosome were not influenced by the treatment (P>0.05) nor by the dilutant used for cryopreservation (P>0.05). The spermatic morphology was also preserved by the different GOGD and GONM concentrations in the ACP® and TRIS dilutants, with no statistically significant differences (P<0.05). It was concluded that Numida meleagris egg yolk, as external membrane cryoproctant added to the dilutants ACP-101c® and TRIS, improved goat semen quality.

Resumen El objetivo fue evaluar la correlación entre los grados-día de crecimiento (GDC), altura del dosel (AD) y índice de área foliar (IAF) para el pasto Guinea irrigado considerando la diferente combinación de las tasas de nitrógeno y boro y los ciclos de rebrote. El experimento se llevó a cabo de agosto/2017 a agosto/2018, en un experimento de campo y la región climática se clasifica como subtropical. Realizó un experimento factorial (4x3x11), con cuatro repeticiones y corrido en un diseño de bloques completamente aleatorio. Tratos: fertilización con boro: 0, 1 y 2 kg ha-1; fertilización con nitrógeno: 250, 500, 750 y 1,000 kg ha-1 distribuidos en 11 ciclos de crecimiento/corta. Hemos observado la alta correlación entre GDC y los parámetros evaluados para AD y IAF. Los resultados de nitrógeno mostraron una baja influencia en la altura del dosel y el índice de área foliar, y los resultados de boro no presentaron influencia en estos parámetros.

Abstract This study aims to evaluate the correlation among growing degree-days (GDD), canopy height (CH), and leaf area index (LAI) for Guinea grass irrigated, considering the different combinations of the nitrogen and boron rates and cycles of regrowth. The experiment was carried out from August/2017 to August/2018 in a field experiment, and the climate of the region is classified as subtropical. A randomized block design arranged in a factorial scheme (4x3x11) with four replications was used. The treatment was (i) boron fertilization: 0, 1, and 2 kg ha-1; and (ii) nitrogen fertilization: 250, 500, 750, and 1,000 kg ha-1 distributing to 11 growth/cutting cycles. We observed the high correlation between GDD and the parameters evaluated of CH and LAI. The nitrogen results showed a low influence on canopy height and the leaf area index, and boron results did not influence these parameters.

Abstract We evaluated the efficacy of xylitol varnishes on the remineralization of newly erupted permanent and deciduous teeth in vitro and in situ. Human enamel specimens were randomly allocated to 8 groups (n = 15/group). Artificial caries lesions were produced and enamel alterations were quantified by surface/subsurface hardness and transverse microradiography. The blocks were then treated with the following varnishes: DuraphatTM; 20 wt% xylitol (146 μm) varnish; 20 wt% milled xylitol (80 μm) varnish, and placebo varnish, and removed after 6 h of immersion in artificial saliva. In vitro the blocks were subjected to pH-cycles for 8 days. In situ fifteen subjects wore palatal appliances containing four pre-demineralized and treated enamel specimens, for 5 days. Data were statistically analyzed by ANOVA/Tukey and Kruskal-Wallis/Tukey’s test (p < 0.05). The %SHR in both studies was significantly increased by xylitol and Duraphat™ varnishes when compared to placebo. Considering subsurface remineralization, only the xylitol varnishes were able to significantly reduce the enamel lesion. Xylitol varnishes can be promising alternatives to promote enamel remineralization of newly erupted permanent and deciduous teeth.

RESUMO O objetivo deste estudo foi identificar o uso de dispositivos metodológicos para mudanças de comportamento e a prática de educação em saúde aos indivíduos com Diabetes Mellitus (DM). Estudo descritivo, exploratório, de abordagem transversal, desenvolvido no município de Aracaju, Sergipe, com 138 profissionais, médicos e enfermeiros da Estratégia Saúde da Família, utilizando entrevista estruturada, tendo como referencial teórico-metodológico o Modelo de Atenção à Condição Crônica. Os resultados mostraram que a maioria dos profissionais referiu desenvolver atividades de educação em saúde, sendo os temas mais referidos: alimentação saudável e exercício físico. Com relação ao uso de abordagens para mudança de comportamento dos usuários, a mais utilizada foi a entrevista motivacional, seguida do Modelo Transteórico de Mudança (MTT). Todos os profissionais que utilizam o MTT concordam com sua aplicabilidade. Conclui-se que a prática de educação em saúde para pessoas com DM é desenvolvida, enquanto o uso de dispositivos metodológicos para mudanças de comportamento ainda é subutilizado, sobretudo o Grupo Operativo.

ABSTRACT The objective of this study was to identify the use of methodological devices for behavioral changes and practice of health education for individuals with Diabetes Mellitus (DM). A descriptive, exploratory, cross-sectional study developed in the city of Aracaju, Sergipe, Brazil, with 138 professionals, doctors and nurses of the Family Health Strategy, using a structured interview, having as a theoretical-methodological reference the Model of Care to Chronic Conditions. The results showed that most of the professionals referred to developing health education activities, with the most mentioned themes being: healthy eating and physical exercise. Regarding the use of approaches to change the behavior of users, the most used was the motivational interview, followed by the Transtheoretical Model of Change (MTT). All professionals who use MTT agree with its applicability. It is concluded that the practice of health education for people with DM is developed, while the use of methodological devices for behavior changes is still underutilized, especially the Operative Group.

Estudo descritivo, exploratório, de abordagem transversal, desenvolvido no município de Aracaju, Sergipe, com 138 profissionais, médicos e enfermeiros da Estratégia Saúde da Família, utilizando entrevista estruturada, tendo como referencial teórico-metodológico o Modelo de Atenção à Condição Crônica. Os resultados mostraram que a maioria dos profissionais referiu desenvolver atividades de educação em saúde, sendo os temas mais referidos: alimentação saudável e exercício físico. Com relação ao uso de abordagens para mudança de comportamento dos usuários, a mais utilizada foi a entrevista motivacional, seguida do Modelo Transteórico de Mudança (MTT). Todos os profissionais que utilizam o MTT concordam com sua aplicabilidade. Conclui-se que a prática de educação em saúde para pessoas com DM é desenvolvida, enquanto o uso de dispositivos metodológicos para mudanças de comportamento ainda é subutilizado, sobretudo o Grupo Operativo.

ABSTRACT The objective of this study was to identify the use of methodological devices for behavioral changes and practice of health education for individuals with Diabetes Mellitus (DM). A descriptive, exploratory, cross-sectional study developed in the city of Aracaju, Sergipe, Brazil, with 138 professionals, doctors and nurses of the Family Health Strategy, using a structured interview, having as a theoretical-methodological reference the Model of Care to Chronic Conditions. The results showed that most of the professionals referred to developing health education activities, with the most mentioned themes being: healthy eating and physical exercise. Regarding the use of approaches to change the behavior of users, the most used was the motivational interview, followed by the Transtheoretical Model of Change (MTT). All professionals who use MTT agree with its applicability. It is concluded that the practice of health education for people with DM is developed, while the use of methodological devices for behavior changes is still underutilized, especially the Operative Group.

RESUMO Objetivo: identificar estímulos que interferem na adaptação de longevos no domicílio, após internação na unidade de terapia intensiva e alta hospitalar. Método: estudo descritivo, com abordagem qualitativa, realizado com longevos que estiveram internados na unidade de terapia intensiva e já se encontravam em domicílio, após alta hospitalar. A coleta de dados ocorreu através de entrevista aberta e foi realizada na unidade de terapia intensiva do hospital e no domicílio dos longevos. As visitas domiciliares aconteceram entre dezembro de 2017 e fevereiro de 2018, e foram realizadas em sete cidades. Utilizou-se a análise de conteúdo de Bardin, e a discussão pautou-se na Teoria de Adaptação de Callista Roy. Os cuidados éticos e legais foram rigidamente respeitados. Resultados: participaram do estudo 11 longevos, com idade entre 80 e 94 anos. A partir dos depoimentos dos participantes emergiram duas categorias: Estímulos que contribuem para respostas comportamentais adaptáveis em longevos e Estímulos que afetam negativamente a adaptação dos longevos. Conclusão: os principais estímulos que contribuíram para um comportamento adaptativo dos longevos foi o retorno para o seu domicílio, o apoio familiar e a rede de apoio social. Em contrapartida, os estímulos que afetaram negativamente a adaptação foi o medo, a falta de informação e dificuldades na continuidade do cuidado. Ressalta-se o papel fundamental dos profissionais de saúde da unidade de terapia intensiva e da unidade hospitalar no preparo do longevo e sua família para a alta hospitalar e retorno domiciliar.

ABSTRACT Objective: to identify stimuli that interfere with the adaptation of long-lived elders at home after hospitalization in the intensive care unit and hospital discharge. Method: a descriptive study with a qualitative approach, conducted with elderly individuals who were hospitalized in the intensive care unit and were already at home after hospital discharge. Data collection took place through open interviews and was performed at the intensive care unit of the hospital and at the home of the elderly. Home visits took place between December 2017 and February 2018, and were conducted in seven cities. Bardin's content analysis was used, and the discussion was based on Callista Roy's Adaptation Theory. Ethical and legal cares were strictly respected. Results: eleven long-lived elders, aged between 80 and 94 years old, took part in the study. Two categories emerged from the participants' testimonies: Stimuli that contribute to adaptive behavioral responses in the long-lived elders and Stimuli that negatively affect the adaptation of the long-lived elders. Conclusion: the main stimuli that contributed to the adaptive behavior of the long-lived elders were the return to their homes, family support and the social support network. In contrast, the stimuli that negatively affected adaptation were fear, lack of information and difficulties in continuing the care. The critical role of the health professionals in the intensive care unit and the hospital unit in preparing longevity the long-lived elders their families for hospital discharge and home return is highlighted.

RESUMEN Objetivo: identificar los estímulos que interfieren en la adaptación de los ancianos a sus domicilios, después de la internación en una unidad de cuidados intensivos y del alta hospitalaria. Método: estudio descriptivo de abordaje cuantitativo realizado con ancianos que estuvieron internados en una unidad de cuidados intensivos y ya se encontraban de regreso en sus domicilios después del alta hospitalaria. Los datos se recolectaron por medio de entrevistas abiertas que se realizaron en una unidad de cuidados intensivos de un hospital y en los domicilios de los ancianos. Las visitas domiciliarias tuvieron lugar entre diciembre de 2017 y enero de 2018 en siete ciudades. Se utilizó el análisis de contenido de Bardin, y la discusión se pautó sobre la base de la Teoría de Adaptación de Callista Roy. Se respetaron estrictamente los reparos éticos y legales. Resultados: del estudio participaron 11 ancianos, de 80 a 94 años de edad. A partir de las declaraciones de los participantes surgieron dos categorías: Estímulos que ayudan a lograr respuestas comportamentales adaptables en los ancianos y Estímulos que afectan negativamente la adaptación de los ancianos. Conclusión: los principales estímulos que favorecen un comportamiento de adaptación en los ancianos fueron el regreso a sus domicilios, el apoyo familiar y la red de apoyo social. Como contrapartida, los estímulos que ejercieron un efecto negativo sobre la adaptación fueron el miedo, la falta de información y las dificultades para proseguir con los cuidados. Se resalta el papel fundamental de los profesionales de la salud que trabajan en unidades de cuidados intensivos y en unidadades hospitalarias en la preparación de los ancianos y de sus familias para el alta hospitalaria y el regreso a sus domicilios.

Resumo Objetivos: adaptar culturalmente e validar o Safe Childbirth Checklist (SCC) da OMS para os hospitais brasileiros. Métodos: realizou-se uma pesquisa metodológica com técnicas de consenso e etapas de adaptação transcultural. O SCC original passou por três etapas de adaptação e validação: 1- grupo nominal com painel de especialistas; 2- conferência de consenso em duas maternidades escolas, em reuniões com profissionais que utilizariam a lista; 3- pré-teste com questionário estruturado aos profissionais de saúde das duas maternidades (n=40) após 30 dias de sua utilização. Critérios de validação contemplaram as validades de face e conteúdo, adequação aos protocolos nacionais, terminologia e viabilidade no contexto local. Resultados: o SCC adaptado para o Brasil foi denominado Lista de Verificação para o Parto Seguro - Brasil (LVPS-BR), contendo 49 itens. Na primeira etapa, os 29 itens do SCC original foram aprovados com algumas adaptações (ex.: CD4 substituído por Teste Rápido para HIV). Na segunda etapa, ocorreram ajustes em alguns dos 29 itens e acrescentaram-se 24 itens. Na terceira etapa, excluíram-se três itens, agruparam-se dois e acrescentou-se um. Conclusões: o processo de validação disponibilizou uma LVPS potencialmente útil para o contexto brasileiro, apresentando indícios de validade e viabilidade para o contexto nacional.

Abstract Objectives: to culturally adapt and validate the WHO Safe Childbirth Checklist (SCC) in Brazilian hospitals. Methods: a methodological study was carried out with consensus techniques and cross-cultural adaptation stages. The original SCC underwent three adaptation and validation stages: 1- nominal group with a panel of experts; 2- consensus conference at two maternity schools, in meetings with professionals who would use the list; 3- pre-test with a structured questionnaire for health professionals from both maternities (n=40) after 30 days of using the checklist. Validation criteria contemplated the content validity, adequated to Brazilian protocols, terminology and feasibility for local context. Results: the adapted SCC in Brazil was called the Lista de Verificação para o Parto Seguro - Brasil (LVPS-BR) (Checklist for Safe Childbirth -Brazil) and included 49 items. In the first stage, the 29 items of the original SCC were approved with some adaptations (e.g. CD4 was replaced by the Rapid HIV Test). In the second stage, some of the 29 items were adjusted and added 24 items more. In the third stage, three items were excluded, two were grouped and one more was added. Conclusions: the validation process provided a potentially useful LVPS for the Brazilian context, presenting validity and feasibility evidences for the Brazilian context.
