Resumo Este estudo teve por objetivo estimar parâmetros genéticos para características de Produção in vitro de Embriões - PIVE das raças Nelore e Senepol. Foram utilizados dados de 1.247 rodadas de fertilização in vitro (1.029 Nelore, 218 Senepol), no total de 148.311 oócitos (116.972 Nelore, 31.339 Senepol), 47.301 embriões (38.722 Nelore, 8.579 Senepol) e 6.323 prenhezes (5.534 Nelore e 789 Senepol). Foram analisadas as variáveis: porcentagem de oócitos viáveis (Pooc), porcentagem de embriões clivados (Pcliv); porcentagem de embriões produzidos (Pemb); porcentagem de prenhezes (Ppren) por rodada/touro; média de oócitos viáveis por doadora (MOD), média de embriões produzidos por doadora (MED) e média de prenhezes por doadora (MPD) de dados fornecidos por empresa parceira entre os anos de 2019 a 2022. Foi utilizado o programa SAS para análise dos efeitos fixos e Correlação Linear de Pearson. Os componentes de variância para cálculo das herdabilidades foram calculados por meio do programa MTDFREML Foram obtidos valores de MOD, MED e MPD para as raças Nelore de 29,94; 10,01; 2,53 e Senepol de 30,12; 8,17; 2,34. De modo geral, a raça Nelore proporcionou melhor produção de embriões em relação à raça Senepol. As estimativas de herdabilidades foram de baixa a média magnitude, sendo para Pcliv (0,16 e 0,04), Pemb (0,14 e 0,08), Ppren (0,02 e 0,15), MED (0,07 e 0,02) e MPD (0,05 e 0,00) para as raças Nelore e Senepol. Porém, indicando a presença de variabilidade genética e possibilidade de seleção. Abstract This study aimed to estimate genetic parameters for traits of in vitro embryo production (IVEP) of Nellore and Senepol cattle. Data from 1,247 rounds of in vitro fertilization (1,029 Nellore, 218 Senepol) were used, totaling 148,311 oocytes (116,972 Nellore, 31,339 Senepol), 47,301 embryos (38,722 Nellore, 8,579 Senepol), and 6,323 pregnancies (5,534 Nellore, 789 Senepol). The variables percentage of viable oocytes (Pooc), percentage of cleaved embryos (Pcleav), percentage of produced embryos (Pemb), percentage of pregnancy (Ppreg) per round per bull, mean number of viable oocytes per donor (MOD), mean number of embryos produced per donor (MED), and mean number of pregnancies per donor (MPD) were analyzed from data provided by a partner company between the years 2019 and 2022. The SAS program was used to analyze fixed effects and Pearson linear correlation. The components of variance for heritabilities were calculated using the MTDFREML program. MOD, MED, and MPD values of 29.94, 10.01, and 2.53 were obtained for Nellore and 30.12, 8.17, and 2.34 for Senepol, respectively. In general, Nellore provided better embryo production compared to Senepol. Heritability estimates showed low to medium magnitude, with values Nellore and Senepol of 0.16 and 0.04 (Pcleav), 0.14 and 0.08 (Pemb), 0.02 and 0.15 (Ppreg), 0.07 and 0.02 (MED), and 0.05 and 0.00 (MPD), respectively. However, it indicates the presence of genetic variability and the possibility of selection. Therefore, there is genetic variability for IVEP traits in both breeds, indicating that they can be used as selection criteria because they are heritable and that Nellore presents better performance for IVEP traits compared to Senepol. Abstract Chromosomal Microarray Analysis (CMA) has increased the comprehension of the mechanisms of copy number variation (CNV) formation, classification of these rearrangements, type of recurrence, and its origin, and has also been a powerful approach to identifying CNVs in individuals with intellectual disability. The aim of this study was to establish the parental origin of de novo pathogenic CNV in a cohort of patients with intellectual disability from the public health system of Goiás-Brazil. CMA was done in 76 trios and we identified 15 de novo pathogenic CNVs in 12 patients with intellectual disability. In a total of 15 de novo pathogenic CNV, 60% were derived from the maternal germline and 40% from the paternal germline. CNV flanked by low copy repeats (LCR) were identified in 46.7% and most of them were of maternal origin. No significant association was observed between paternal age and the mutation rate of de novo CNVs. The presence of high-identity LCRs increases the occurrence of CNV formation mediated by non-allelic homologous recombination and the majority of paternal CNVs are non-recurrent. Cagaiteira, the fruit popularly known as cagaite, belongs to the Myrtaceae family. Seed oil cagaite (Eugenia dysenterica DC) was obtained by three methods of extraction, extraction using an ultrasound (OCU), extraction by mechanical pressing (OCP) and extraction with Soxhlet extractor (OCS) in 3 different times (3, 6 and 9 h) and the content of fatty acids and physicalchemical properties were compared. The rate of saturated fatty acids ranged from 19.46% (OCP03) to 31.18% (OCS09), while the amount of unsaturated fatty acids ranged from 54.72% (OCP03) to 67.64% (OCS09). Linoleic and linolenic acids, important in nutrition food, varied between 32.22-38.11 and 5.55-8.78%, respectively. For oxidative stability, OCUAq (heated ultrasound extraction) samples showed the highest induction periods, showing a positive correlation with antioxidant activity and phenol content, demonstrating the efficiency of the use of ultrasonic extraction to obtain quality oils from cagaite seeds. Resumo Objetivou-se determinar a ocorrência de anticorpos anti-Toxoplasma gondii, Neospora caninum e Leptospira spp., em ovinos e caprinos criados em aldeias da comunidade indígena Xukuru do Ororubá, Pernambuco, Brasil. Foram analisadas 180 amostras de soro de ovinos e 108 amostras de soro de caprinos de ambos os sexos e diferentes idades. Para a pesquisa de anticorpos foi utilizada a técnica de Reação de Imunofluorescência indireta (RIFI), para os protozoários T. gondii e N. caninum e Aglutinação Microscópica (MAT) para Leptospira spp., com ponto de corte de 1:64, 1:50 e 1:100, respectivamente. A frequência de anticorpos anti-T gondii foi de 16,6% (30/180) em ovinos e 11,1% (12/108) em caprinos. A frequência de anticorpos anti-N. caninum foi de 10,55% (19/180) para ovinos e 20,37% (22/108) para caprinos, enquanto para Leptospira spp., 2,2% (4/180) dos ovinos e 1,85% (2/108) dos caprinos reagiram positivamente. Os resultados obtidos neste estudo são inéditos em comunidades indígenas do país e alertam para o monitoramento de caprinos e ovinos da aldeia indígena Xukuru do Ororubá, quanto à ocorrência e impacto produtivo de infecções por T. gondii, N. caninum e Leptospira spp., além da ocorrência de zoonoses como a toxoplasmose e leptospirose na comunidade indígena. Abstract This study aimed to determine the occurrence of anti-Toxoplasma gondii, Neospora caninum, and Leptospira spp. antibodies in sheep and goats raised in villages of the Xukuru do Ororubá indigenous community, Pernambuco, Brazil. A total of 180 serum samples from sheep and 108 serum samples from goats of both sexes and different ages were analyzed. For antibody research, indirect immunofluorescence antibody test (IFAT) were used for the protozoa T. gondii and N. caninum, and microscopic agglutination test (MAT) for Leptospira spp., with a cutoff titer of 1:64, 1:50 and 1:100, respectively. The frequency of anti-T. gondii antibodies was 16.6% (30/180) for sheep and 11.1% (12/108) for goats. The frequency of anti-N. caninum antibodies was 10.55% (19/180) for sheep, and 20.37% (22/108) for goats, while for Leptospira spp., 2.2% (4/180) of sheep and 1.85% (2/108) of goats reacted positively. Abstract Staphylococcus aureus is one of the main pathogens associated with foodborne outbreaks in Brazil and food handlers can carry toxigenic and resistant S. aureus strains. The aims of this study were to verify the frequency of virulence genes, to identify the agr groups and to determine the antimicrobial resistance profile of S. aureus strains isolated from food handlers of pilot kitchens located in São Paulo, Brazil. A total of 74 strains of the Staphylococcus genus were detected and 50% were identified as of the species S. aureus. The enterotoxin genes detection, tst and luk-PV detection, agr typing, mecA detection, ccr complex detection and SCCmec typing were performed using PCR. The antimicrobial resistance testing was performed by the disk diffusion method. The enterotoxin genes were identified in 36 S. aureus, including sea (83.8%). The tst gene was detected in 18.92% of the strains and the luk-PV was detected in only one isolate. Agr typing classified 58.3% of the strains as type I. Seven (18.92%) strains were classified as MRSA and the ccr2 complex was detected in six of these isolates. The SCCmec typing characterized strains as type II, III, IV and V. Moreover, there were also a greater number of resistant strains to penicillin (83.78%) and clarithromycin (67.57%). In conclusion, the study revealed a significant prevalence of S. aureus, and the presence of different virulence genes and a worrying resistance profile in S. aureus strains isolated from food handlers in this country.

RESUMO: A resistência à meticilina no grupo Staphylococcus intermedius (GSI) tem aumentado na clínica de pequenos animais. Membros GSI resistentes à meticilina (GSIRM) têm sido causas de infecções tanto em animais de companhia e humanos. Cassetes cromossômicos estafilocócicos mec (SCCmec) carregam os genes mecA/C, que codificam a transpeptidase PBP2a (PBP2’) responsável pela resistência aos antibióticos β-lactâmicos em estafilococos. Nosso objetivo foi investigar os elementos SCCmec de GSIRM isolados de diferentes amostras clínicas de cães que exibiram CIM de meticilina ≥ 0,5 μg/mL por meio de um sistema automatizado em um Centro Veterinário de Diagnósticos em São Paulo, Brasil. A sensibilidade à meticilina foi determinada por meio do teste de microdiluição em caldo e fitas Oxoid® M.I.C.Evaluator®. A produção de PBP2a foi detectada usando um ensaio de aglutinação de látex. A tipagem dos elementos SCCmec foi realizada de acordo com as diretrizes do International Working Group on the Classification of Staphylococcal Cassette Chromosome Elements (IWG-SCC). SCCmec tipo II (2A), SCCmec tipo III (3A), SCC compostos de um complexo mec de classe A com múltiplos complexos ccr, e elementos SCCmec não tipáveis foram encontrados nesses isolados GSIRM. Variantes que diferem dos elementos SCCmec reconhecidos até o momento pelo IWG-SCC foram encontradas, indicando novos rearranjos no contexto genético de mecA nesses isolados GSIRM caninos.

ABSTRACT: Methicillin resistance in the Staphylococcus intermedius group (SIG) has emerged in small animal practice. Methicillin-resistant SIG (MRSIG) members have been implicated as causes of infections in both companion animals and humans. Staphylococcal cassette chromosome mec (SCCmec) elements carry the mecA/C genes, which encode for the transpeptidase PBP2a (PBP2’) responsible for β-lactam antibiotic resistance in staphylococci. This study examined the SCCmec types of MRSIG isolates from different clinical specimens of dogs that exhibited methicillin MIC ≥ 0.5 μg/mL by an automated identification and susceptibility system in a Center for Veterinary Diagnostics in São Paulo, Brazil. Susceptibility to methicillin was determined by broth microdilution testing, and Oxoid® M.I.C.Evaluator® strips. PBP2a production was detected using a latex agglutination assay. SCCmec typing was performed according to the International Working Group on the Classification of Staphylococcal Cassette Chromosome Elements (IWG-SCC) guidelines. SCCmec type II (2A), SCCmec type III (3A), composite SCC structures consisting of a class A mec gene complex in addition to multiple ccr gene complexes, and non-typable SCCmec elements were reported in these MRSIG isolates. SCCmec type variants differing from those so far acknowledged by IWG-SCC were found, indicating new rearrangements in the genetic context of mecA in these canine MRSIG isolates.

Abstract INTRODUCTION: The present study aimed to assess the seroprevalence and spatial distribution of Neospora caninum and Toxoplasma gondii in dogs. METHODS: Blood samples (n = 241) were collected and analyzed for the presence of anti-N. caninum and anti-T. gondii antibodies. The spatial distribution was evaluated using kernel density estimation (KDE). RESULTS Anti-N. caninum and anti-T. gondii antibodies were detected in 24.06% (58/241) and 9.54% (23/241) of samples, respectively. A heterogeneous spatial distribution of positive dogs was observed across the city. CONCLUSIONS These data are pivotal for better understanding the dynamics of infection caused by these protozoa in the canine population.

Resumo: O objetivo deste trabalho foi selecionar clones de cana-de-açúcar (Saccharum officinarum) superiores, com boa estabilidade e adaptabilidade, ao se considerar a interação genótipo x ambiente em dois ciclos produtivos. Vinte e cinco clones precoces mais cinco clones testemunhas foram avaliados durante dois cortes (cana-soca e cana-planta), em 24 ambientes. Utilizou-se o delineamento experimental de blocos ao acaso, com três repetições. Foram avaliadas toneladas de colmos por hectare e toneladas de pol por hectare. Para a verificação da adaptabilidade e da estabilidade, foram utilizados os métodos de regressão bissegmentada e multivariados (AMMI e GGE biplot). De acordo com os três métodos, que são complementares nas informações desejadas, os clones mais promissores em termos de estabilidade e adaptabilidade geral são G5, G12 e G13; estes dois últimos são os mais próximos do genótipo ideal. O clone G13 é altamente produtivo nos ambientes favoráveis e desfavoráveis, tendo apresentado as maiores médias para tonelada de colmos e de pol por hectare. Os clones G3, G4, G10, G15, G17, G18, G22, G23, G25, G26 e G30 não são recomendados para os 24 ambientes avaliados.

Abstract: The objective of this work was to select superior sugarcane (Saccharum officinarum) clones with good stability and adaptability, considering the genotype x environment interaction in two productive cycles. Twenty-five early clones plus five control clones were evaluated during two cuts (ratoon cane and plant cane) in 24 environments. A randomized complete block design was used, with three replicates. Tons of stems per hectare and tons of pol per hectare were evaluated. To verify adaptability and stability, the bisegmented regression and the multivariate (AMMI and GGE biplot) methods were used. According to the three methods, which are complementary regarding the desired information, the most promising clones in terms of stability and general adaptability are G5, G12, and G13; the last two are closest to the ideal genotype. The G13 clone is highly productive in favorable and unfavorable environments, presenting the highest averages for ton of stems and pol per hectare. The G3, G4, G10, G15, G17, G18, G22, G23, G25, G26, and G30 clones are not recommended for the 24 evaluated environments.

Resumo As doenças cardiovasculares (DCV) são a principal causa de morte e representam uma percentagem significativa das internações. No cenário de minimização dos custos ao sistema de saúde, métodos que identifiquem DCV subclínica seriam importantes. Algumas diretrizes incluem a medida da rigidez aórtica e da espessura íntima-média da artéria carótida como métodos para identificação de DCV subclínica em hipertensos. A velocidade de onda de pulso (VOP) é considerada padrão-ouro para avaliar a rigidez arterial. Nesta revisão, abordamos a fisiopatologia e os determinantes da rigidez arterial e justificamos sua inclusão na avaliação do paciente hipertenso dada a associação direta com o risco cardiovascular, como estabelecido na I Diretriz Brasileira de Prevenção Cardiovascular. Apresentamos ainda os principais estudos genéticos deste fenótipo que, dada sua complexidade, pode ser modulado por dezenas de genes. No entanto, um melhor entendimento da relação genética-rigidez arterial, e até mesmo uma intervenção baseada em genótipos, devem ser investigadas em estudos futuros.

Abstract Cardiovascular diseases (CVD) are the main cause of mortality and it represents a significant percentage of hospitalizations. In the scenario of minimization of costs of the health system, methods that identify subclinical CVD would be important. Some guidelines include the measures of aortic stiffness and intima-media thickness of the carotid artery as methods to identify subclinical CVD in hypertensive patients. The pulse wave velocity (PWV) is the gold standard for the evaluation of arterial stiffness. In this review, we report the pathophysiology, the determinants of arterial stiffness, and justify its inclusion in the assessment of hypertensive patient due its direct association with cardiovascular risk, as show in the I Diretriz Brasileira de Prevenção Cardiovascular. In addition, we raised the main genetic studies of this phenotype, due to its complexity, can be modulated by dozens of genes. However, a better understanding of the relationship genetic-arterial stiffness and, even an intervention based on genotypes, should be achieved in future studies.

The quality of mineral water commercialized in Brazil regarding the microbial content was analyzed and the results were compared with the standards established by the current legislation. Results demonstrated there was no bacterial contamination, but several types of fungi were found. Therefore, bottled mineral water could be considered a possible route for the transmission of filamentous fungi and yeasts.

La artritis reumatoide (AR) es una enfermedad inflamatoria crónica que afecta principalmente las articulaciones, produciendo destrucción articular, deformidad, discapacidad y deterioro de la calidad de vida. Como nuevas opciones de tratamiento, producto del avance tecnológico y del conocimiento de la fisiopatología de la enfermedad, han aparecido recientemente en el mercado los denominados agentes biológicos. Teniendo en cuenta que no hay reportes colombianos sobre la experiencia con infliximab en pacientes con AR, el objetivo de este estudio fue cuantificar en un grupo de pacientes colombianos con AR de larga evolución (13±10 años) y refractarios al tratamiento convencional, el cambio en la calidad de vida, la capacidad funcional, en los síntomas y signos relacionados con la actividad inflamatoria y en la seguridad terapéutica producidos por infliximab. Un total de 31 pacientes, 22 mujeres y 9 hombres, con edades de 51±12 años y un promedio de evolución de enfermedad de 13,2±10,4 años, que permanecieron activos a pesar de recibir metotrexate o leflunomida en combinación con otros agentes modificadores de la enfermedad, bajas dosis de glucocorticoides, analgésicos y antiinflamatorios, recibieron infusiones de infliximab de tres mg/kg dosis durante 22 semanas, siguiendo las recomendaciones de la Asociación Colombiana de Reumatología, en las semanas 0, 2, 6, 14 y 22. Se encontró que la capacidad funcional medida por HAQ-DI pasó de 1,2±0,54 a 0,29±0.28, que la calidad de vida medida por Spanish-AIMS mejoró en su calificación global de 48,4±14.4 a 29,4±8.2 y en cada uno de sus ocho componentes. Hubo también mejoría en los componentes del SF-12, pues la salud física pasó de 39,9±13,7 a 55,5±24,1 y la salud mental de 57,0±24,2 a 74,6±17,0. Aunque el 71% refirió algún evento adverso durante el seguimiento, estos fueron menores y no obligaron a la suspensión del medicamento. Sólo tres pacientes fueron retirados, uno por hipotensión repetitiva al inicio de la infusión, otro por reacción anafiláctica que requirió reanimación y el otro por tuberculosis pleural luego de dos meses de iniciado el medicamento. Se encontró entonces que de acuerdo con los reportes internacionales, la administración de infliximab conlleva a una mejoría significativa en los enfermos con AR refractaria a los tratamientos comunes, evidenciada en los signos y síntomas relacionados con la actividad de la AR, así como en la calidad de vida y en la capacidad funcional. Teniendo en cuenta la posibilidad las reacciones adversas durante la aplicación y que en nuestro país la tuberculosis es un riesgo real, la administración del infliximab debe hacerse en unidades especializadas con personal entrenado y bajo la supervisión estricta del reumatólogo.

Rheumatoid arthritis is a chronic inflammatory process that effects mostly the joints leading to destruction of the joint, deformity, incapacity and deterioration of the quality of life. As a result of technologic advances and the better knowledge of the pathophisiology of the disease, there are new therapeutic options with biologic agents. There are no reports from Colombia about the experience with Infliximab in patients with rheumatoid arthritis. The objective of this study was to quantify the changes in quality of life, functional capacity, signs and symptoms related to the inflammatory activity and the therapeutic efficacy, in a group of Colombian patients with long term rheumatoid arthritis (13±10 years) refractory to conventional treatments. A total of 31 patients, 22 women and 9 men, aged 51±12 years and an average evolution of disease of 13.2±10.4 years, who had active disease in spite of having received Methrotexate or Leflunomide in combination with other agents, such as low doses of glucocorticoids, analgesics and antiinflammatories, received infusions of Infliximab at doses of 3mg/kg during weeks 0, 2, 6, 14, and 22, following the guidelines of the Colombian Rheumatology Association. It was determined that the median functional capacity measured by HAQ-DI went from 1,2±0,54 to 0,29±0.28, that the quality of life measured by Spanish-AIMS improved globally from 48,4±14.4 to 29,4±8.2 in each of its eight components. There was also improvement in the components of SF-12 since the physical health went from 39,9±13,7 to 55,5±24,1 and mental health from 57,0±24,2 to 74,6±17,0. Although 71% of the patients had some type of side effect, they were minor and the medication was not stopped. These findings agree with the international reports that the administration on Infliximab induces a significative improvement in those patients with refractory rheumatoid arthritis. Taking into account the potential side effects and the real risk of tuberculosis in our country, the administration of Infliximab must be carried out in specialized units with well trained personnel and under the strict supervision of a rheumatologist.

Teve também melhoria nos componentes do SF-12, pois a saúde física passou de 39,9±13,7 a 55,5±24,1 e a saúde mental de 57,0±24,2 a 74,6±17,0. Ainda que o 71% referiu algum evento adverso durante o seguimento, estes foram menores e não obrigaram à suspensão do medicamento. Só três pacientes foram retirados, um por hipotensão repetitiva ao início da infusão, outro por reação anafilática que requereu reanimação e o outro por tuberculose pleural depois de dois meses de iniciado o medicamento. Encontrou-se então que de acordo com os reportes internacionais, a administração de infliximab implica uma melhoria significativa nos enfermos com AR refratária aos tratamentos comuns, evidenciada nos signos e sintomas relacionados com a atividade da AR, bem como na qualidade de vida e na capacidade funcional. Tendo em conta a possibilidade às reações adversas durante a aplicação e que em nosso país a tuberculose é um risco real, a administração do infliximab deve fazer-se em unidades especializadas com pessoal treinado e sob a supervisão estrita do reumatólogista.
