Rivaroxaban is a direct factor Xa inhibitor. Its interindividual variability is large and may be connected to the occurrence of adverse drug reactions or drug inefficacy. Pharmacogenetics studies concentrating on the reasons underlying rivaroxaban's inadequate response could help explain the differences in treatment results and medication safety profiles. Against this background, this study evaluated whether polymorphisms in the gene encoding the ABCG2 transporter modify the pharmacokinetic characteristics of rivaroxaban. A total of 117 healthy volunteers participated in two bioequivalence experiments with a single oral dose of 20 mg rivaroxaban, with one group fasting and the other being fed. Ultra-high-performance liquid chromatography coupled with mass spectrometry was employed to determine the plasma concentrations of rivaroxaban, and the WinNonlin program was used to calculate the pharmacokinetics parameters. In the fasting group, the rivaroxaban pharmacokinetic parameters of Vd (508.27 vs 334.45 vs 275.59 L) and t1/2 (41.04 vs 16.43 vs 15.47 h) were significantly higher in ABCG2 421 A/A genotype carriers than in ABCG2 421 C/C and 421 C/A genotype carriers (P<0.05). The mean values of Cmax (145.81 vs 176.27 vs 190.19 ng/mL), AUC0-t (1193.81 vs 1374.69 vs 1570.77 ng/mL·h), and Cl (11.82 vs 14.50 vs 13.01 mL/h) for these groups were lower, but this difference was not statistically significant (P>0.05). These findings suggested that the ABCG2 421 A/A genotype may impact rivaroxaban parameters after a single dose in healthy subjects. This finding must be validated before it is applied in clinical practice. inhibitor inefficacy rivaroxabans s profiles background ABCG 11 2 fed Ultrahighperformance Ultra high performance 508.27 50827 508 27 (508.2 33445 334 45 334.4 27559 275 59 275.5 L t12 t t1 t1/ 41.04 4104 41 04 (41.0 1643 16 43 16.4 1547 15 47 15.4 h 42 AA CC C CA P<0.05. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. The importance of the CTFB is evidenced by the elevated number of citations in the scientific literature in diverse areas of biology, law, anthropology, education, forensic science, and veterinary science, among others. publications problem uptodate up date classifications context exception (CTFB http//fauna.jbrj.gov.br/, httpfaunajbrjgovbr http //fauna.jbrj.gov.br/ , jbrj gov br (http://fauna.jbrj.gov.br/) 2015 Brazil 80 specialists 1 2024 133691 133 691 133,69 125138 125 138 125,13 82.3%, 823 82 3 (82.3% 102000 102 000 102,00 7.69%, 769 7 69 (7.69% 11000 11 11,00 . 3,567 3567 567 (3,56 2,292 2292 2 292 (2,29 1,833 1833 833 (1,83 1,447 1447 447 (1,44 1000 1,00 831 (83 628 (62 606 (60 520 (52 50 users science health biology law anthropology education others http//fauna.jbrj.gov.br/ faunajbrjgovbr //fauna.jbrj.gov.br (http://fauna.jbrj.gov.br/ 201 8 202 13369 13 133,6 12513 12 125,1 82.3% (82.3 10200 10 00 102,0 7.69% 76 6 (7.69 1100 11,0 3,56 356 56 (3,5 2,29 229 29 (2,2 1,83 183 83 (1,8 1,44 144 44 (1,4 100 1,0 (8 62 (6 60 52 (5 5 http//fauna.jbrj.gov.br (http://fauna.jbrj.gov.br 20 1336 133, 1251 125, 82.3 (82. 1020 0 102, 7.69 (7.6 110 11, 3,5 35 (3, 2,2 22 (2, 1,8 18 (1, 1,4 14 4 ( 82.
Abstract In alignment with historical organizational studies (Decker, Hassard, & Rowlinson, 2021; Maclean, Harvey, & Clegg, 2016) and seeking a geographical (re)orientation, more to the Global South, to historical research in organizational and management studies (A. S. M. Costa & Wanderley, 2021; Wanderley & A. Barros, 2019), the article aims to understand the ideological positioning and the political-discursive performance of the newspaper Folha de S. Paulo in the early period of the Brazilian civil-military dictatorship (1964-1985). A theoretical framework was built, supported by two historiographical currents (Business and Dictatorship; History and Press) and by the concepts of discourse, ideology, and power articulated by critical discourse analysis. In terms of documental sources, 112 daily editorials were collected, published from January to June 1964, and available in the historical archive of the researched company. The theoretical and methodological contribution of Norman Fairclough’s (2016) three-dimensional model and its proposed use by J.B. Thompson’s (2011) model of analysis of strategies of ideological constructions were used. As a result, it was possible to identify three different moments in the discursive trajectory of the positioning of the Folha de S. Paulo newspaper concerning the military coup of 1964 in Brazil: (1) significant contribution to the destabilization of the democratic government of President João Goulart, (2) strong alignment with the military coup and its immediate unfoldings; and (3) support for the subsequent Castelo Branco government, but already with the enunciation of some reservations. Finally, it is possible to affirm that this journalistic organization acted strongly not only as an instrument of information but mainly as a producer and reproducer of specific discourses in favor of a particular social group and its subsequent dictatorial acts. Against this background, historical research into the political and socio-economic relations between business, government, and society over time and their unfolding today is becoming increasingly necessary.

Glioblastoma is the most prevalent and malignant brain tumor identified in adults. Surgical resection followed by radiotherapy and chemotherapy, mainly with temozolomide (TMZ), is the chosen treatment for this type of tumor. However, the average survival of patients is around 15 months. Novel approaches to glioblastoma treatment are greatly needed. Here, we aimed to investigate the anti-glioblastoma effect of the combination of matteucinol (Mat) (dihydroxyflavanone derived from Miconia chamissois Naudin) with the chemotherapeutic TMZ in vitro using tumor (U-251MG) and normal astrocyte (NHA) cell lines and in vivo using the chick embryo chorioallantoic membrane (CAM) assay. The combination was cytotoxic and selective for tumor cells (28 μg/mL Mat and 9.71 μg/mL TMZ). Additionally, the combination did not alter cell adhesion but caused morphological changes characteristic of apoptosis in vitro. Notably, the combination was also able to reduce tumor growth in the chick embryo model (CAM assay). The docking results showed that Mat was the best ligand to the cell death membrane receptor TNFR1 and to TNFR1/TMZ complex, suggesting that these two molecules may be working together increasing their potential. In conclusion, Mat-TMZ can be a good candidate for pharmacokinetic studies in view of clinical use for the treatment of glioblastoma.

The aim of the present study was to verify the role of lactate as a signaling molecule in cardiac tissue under physiological conditions. C57BL6/J male mice were submitted to acute running bouts on a treadmill at different exercise intensities (30, 60, and 90% of maximal speed - Smax) under the effect of two doses (0.5 and 5 mM) of α-cyano-4-hydroxycynnamate (CINN), a blocker of lactate transporters. Cardiac lactate levels, activity of the enzymes of glycolytic [hexokinase (HK) and lactate dehydrogenase (LDH)] and oxidative metabolism [citrate synthase (CS)], and expression of genes also related to metabolism [LDH, nuclear factor erythroid 2-related factor 2 (NRF-2), cytochrome oxidase IV (COX-IV), and peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1α)] were evaluated. Elevated cardiac lactate levels were observed after high intensity running at 90% of Smax, which were parallel to increased activity of the HK and CS enzymes and mRNA levels of PGC-1α and COX-IV. No changes were observed in cardiac lactate levels in mice running at lower exercise intensities. Interestingly, prior intraperitoneal administration (15 min) of CINN (0.5 mM) significantly reduced cardiac lactate concentration, activities of HK and CS, and mRNA levels of PGC-1α and COX-IV in mice that ran at 90% of Smax. In addition, cardiac lactate levels were significantly correlated to both PGC-1α and COX-IV cardiac gene expression. The present study provides evidence that cardiac lactate levels are associated to gene transcription during an acute bout of high intensity running exercise.

ABSTRACT This study aimed to assess different prebiotic concentrations and principles, in addition to calcium butyrate, aiming to replace colistin as a growth promoter. The sample consisted of 120 piglets weaned at 22 days old with mean initial weight of 5.475 ± 0.719kg. The animals were assigned to random blocks in six treatments corresponding to the use of the following dietary additives: T1) colistin (40 ppm); T2) β-glucan/mannan-oligosaccharides (0.2%); T3) calcium butyrate (0.1%); T4) β-glucan/mannan-oligosaccharides (0.1%) + fructooligosaccharides (0.01%) + galactooligosaccharides (0.09%); T5) β-glucan/mannan-oligosaccharides (0.1%) + fructooligosaccharides (0.03%) + galactooligosaccharides (0.07%); and T6) β-glucan/mannan-oligosaccharides (0.1%) + fructooligosaccharides (0.05%) + galactooligosaccharides (0.05%). The results showed no difference among treatments for the performance parameters in any of the phases evaluated. For diarrhea incidence and intensity, the results indicated that the treatments with alternative additives had similar effects as the group treated with colistin. A significant difference was found for the profile of propionic acid (0.23% colistin and 0.32%, 0.36%, 0.37% additives) and total fatty acids (0.67% colistin and 0.97% additives) values in the caecum. The supplementation with different compositions and concentrations of prebiotics and butyric acid may viably replace colistin in controlling diarrhea and modulating volatile fatty acid production in the caecum.

ABSTRACT The aim of this study was to evaluate in vitro the probiotic potential and absorption of Saccharomyces cerevisiae for the aflatoxin B1 in simulated fish intestinal tract conditions. Three yeast strains were used, two from brewery: S. cerevisiae RC1 and S. cerevisiae RC3 and one from a fish farming environment: S. cerevisiae A8L2. The selected yeasts were subjected to the following in vitro tests: homologous inhibition, self-aggregation, co-aggregation, antibacterial activity, gastrointestinal conditions tolerance and adsorption of AFB1. All S. cerevisiae strains showed good capability of self-aggregation and co-aggregation with pathogenic bacteria. All yeast strains were able to survive the gastrointestinal conditions. In acidic conditions, the factors (strain vs. time) had interaction (P=0.0317), resulting in significant variation among the strains tested in the time periods analyzed. It was observed that there was also interaction (P=0.0062) in intestinal conditions, with an increased number of cells in the 12-hour period for all strains tested. In the adsorption test, the A8L2 strain was statistically more effective (P<0.005) for both AFB1 concentrations evaluated in this study (10 and 25ng/mL). Thus, it was observed that the strains of S. cerevisiae have potential probiotic and adsorbent of AFB1.

Abstract This study was undertaken to investigate the performance, carcass characteristics, and economic viability of diets including pasta waste in meal form (PWM) for quail. A total of 450 unsexed meat quail (Coturnix coturnix coturnix) were distributed into five treatments (0, 10, 20, 30, and 40% inclusion of PWM) in a completely randomized design with six replicates, each with 15 birds. The following rearing periods were evaluated: Phase 1 - 1 to 21 days; and period of 1 to 42 days. In Phase 1, the PWM levels did not influence feed intake (FI), and the PWM inclusion levels for optimal weight gain (WG) and feed conversion (FC) were estimated at 20.70% and 22.8%, respectively. In the cumulative period of 1 to 42 days, the PWM levels did not lead to differences in FI, WG, or FC. The yields of carcass and cuts were not influenced by the treatments; however, abdominal fat yield decreased as the PWM levels in the diets were increased. Economic analysis showed a reduction of the feeding cost in the studied phases. Pasta waste meal can be included up to the level of 40% without incurring losses in the performance or carcass yield of meat quail.

ABSTRACT The objective of this work was to determine the digestibility of palm kernel meal (Elaeis guineensis) in tambaqui (Colossoma macropomum), in two weight classes: 1 (210 fingerlings of 4.45±1.18g) and 2 (54 juveniles of 115.91±4.01g). The apparent digestibility coefficients (ADC) of dry matter, crude protein and crude energy of the palm kernel meal were evaluated by the substitution of the reference diet methodology, using 0.1% chromic oxide as an external indicator. Data were analyzed by Student's t-test at 5% probability. The dry matter, crude protein and crude energy ADCs of the ingredient were the same (P> 0.05) in the weight classes evaluated. The ADCs observed in classes 1 and 2, respectively, were: dry matter (17.52% and 20.75%), crude protein (62.83% and 63.75%) and crude energy (14.16% and 22.43%). The ability of tambaqui to digest the nutrients of palm kernel meal was not influenced by body weight, and satisfactory protein utilization (63.29%) makes this ingredient a potential alternative source of protein in diets for the species.

ABSTRACT The objective of this study was to evaluate the effect of the partial replacement of elephant grass silage with babassu (Orbignya speciosa) on the performance, intake, digestibility, and carcass weight gain of sheep. Fifty-four castrated male lambs (19.08±2.76kg) were distributed in a completely randomized design and administered one of the following treatments: 0.0, 12.5, 25.0, 37.5 or 50% dry matter (%DM) replacement of elephant grass silage with babassu cake. No difference (P> 0.05) was observed in the dry matter, crude protein, and neutral detergent fiber intake, but the ether extract intake increased (P< 0.05). The digestibility of the dry matter, neutral detergent fiber, and total digestible nutrients were unaffected. The crude protein digestibility decreased (P< 0.05), and the ether extract digestibility increased with the replacement of the elephant grass. There were no changes in mean daily weight gain and carcass weight gain. In the carcasses, a linear increase was observed in the proportion of the ether extract, and the crude protein decreased. The replacement of the silage with the babassu cake by up to 50% did not change the performance of sheep, however it led to an increase in fat deposit in the carcass.

ABSTRACT: Interference between root systems occurs below the ground surface long before the competition between plants is expressed in the shoot. Nutrients stand out for being limited in the soil, requiring supplementation, especially phosphorus. Thus, this study aimed at estimating, from sowing, visual characteristics in the root systems of bean plants from the Carioca variety (Phaseolus vulgaris) when in competition with brachiaria (Urochloa brizantha), considering also the application of phosphorus P2O5. A factorial design with six treatments was used resulting from the combination of three cultivation schemes (beans in isolated crops or in competition with one or two brachiaria plants), and the application or not of phosphate fertilizer (90 kg ha-1 of P2O5), in a completely randomized design with four replications. After the sowing of the species, the visualization results were taken at every 12 hours, by means of the photographic record of the development of the root system in rhizotrons. Greater growth and greater total area of the root system were observed in bean monocultures and when competing with one brachiaria plant in the presence of phosphate fertilization. In contrast, the bean plant in the container with two brachiaria plants invested its resources in greater length and total area of the root system when in the absence of the fertilizer. It can be stated that there is evidence of interference starting three days after the sowing of the species.

ABSTRACT: After years of pesticide application, often indiscriminately, damage has been caused to the environment, as well as to agronomic crops grown in contaminated areas. In water treatment, techniques based on physical and/or physicochemical processes are used, being formed secondary oxidizing agents responsible for the mineralization of contaminants present in the herbicides, thus causing the degradation process. However, few studies have demonstrated the effectiveness of these techniques in soils. The aim of this study was to review the existing studies, presenting the main techniques used for remediation of soils contaminated with pesticides. Emphasis was placed on electrokinetics, advanced oxidative processes, soil washing, chemical or solvent extraction, and combinations of these methods. According to the results of several studies, the combination of techniques, such as electrokinetics and soil washing processes, allows achieving a high efficiency when it is applied for the degradation of agricultural contaminants. The combination of electrolysis with other methods, such as the Fenton’s reagent, ultrasound irradiation, and UV light, also present satisfactory results in removing pesticides in soil treatment. However, Fenton technology used alone is the most promising of the assessed techniques since it can be used for the remediation of several contaminants, especially pesticides, not harming the environment and allowing soil recovery.

The objective of this study was to investigate the effect of whole body vibration (WBV) exercise on oxidative stress markers in a group of women with fibromyalgia (FM) compared to a group of healthy women (CT). Twenty-one women diagnosed with FM and 21 age- and weight-matched healthy women were enrolled the study. Plasma oxidative stress markers (primary outcomes) were evaluated at rest and after WBV, and included thiobarbituric acid reactive substances (TBARS), iron reduction capacity (FRAP), superoxide dismutase antioxidant enzymes activity (SOD), and catalase (CAT). At rest, the FM group had higher TBARS (P<0.001) and FRAP (P<0.001), and lower CAT (P=0.005) compared to the CT. In the CT group, the WBV had no effect on TBARS (P=0.559) and FRAP (P=0.926), whereas it increased both SOD (P<0.001) and CAT (P<0.001). In the FM group, the WBV reduced TBARS (p <0.001), FRAP (P<0.001), and CAT (P=0.005), while it increased SOD (P=0.019). There was an interaction effect (moments vs groups) in the TBARS (effect size=1.34), FRAP (effect size=0.93), CAT (effect size=1.45), and SOD (effect size=1.44) (P<0.001). A single trial of WBV exercise improved all oxidant and antioxidant parameters towards a greater adaptation to the stress response in FM women.

ABSTRACT The objective of this study was to evaluate the bioeconomic performance of calves in the first 60 days of life submitted to three feeding systems. Twenty-four calves (Dutch x Guzerá) were used, 12 males and 12 females, with initial weight of 32.25±4.8kg for females and 36.92±6.8kg for males. The animals were distributed in a completely randomized design, in a factorial scheme (3 x 2). The calves received water at will and six liters of milk replacer a day for 60 days in three different strategies, called the suckling system (SA-30: 3 liters of milk replacer, twice a day until 30 days of age; SA-45: 3 liters of milk replacer, twice a day until 45 days of age; SA-49: 3 liters of milk replacer, twice daily up to 49 days old). The lactation systems studied did not present statistical difference (P>0.05) for the consumption and digestibility of nutrients, except for organic matter (OM) and ethereal extract (EE). There was an interaction (P<0.05) between the suckling system and sexual class for the OM and EE intakes, as well as for the average daily gain, in which HS 49 males presented higher averages in relation to SA 30. The bioeconomic performance of male calves from the lactation system 49 was superior and presented the best cost-benefit ratio among the systems studied.

ABSTRACT This study presents an approach for fault detection and classification in a DC drive system. The fault is detected by a classical Luenberger observer. After the fault detection, the fault classification is started. The fault classification, the main contribution of this paper, is based on a representation which combines the Subctrative Clustering algorithm with an adaptation of Particle Swarm Clustering.
