This study aimed to isolate, purify and characterize secondary metabolites present in Amburana acreana leaves, native to the state of Rondônia, using classical chromatographic methods and 1D, 2D nuclear magnetic resonance spectroscopy. From the hydroethanolic leaves extract of A. acreana, p-hydroxybenzoic acid, vanillic acid, coumarin, campesterol 3-β-D-glucoside and amburoside B, all known compounds, were identified. To the best of our knowledge, this is the first report of isolation and chemical characterization of secondary metabolites in A. acreana. In addition, in vitro assays indicated that amburoside B does not have inhibitory potential on the growth of gram-negative bacteria. Fruits of Schinus terebinthifolia Raddi were investigated. A fast and efficient method for the simultaneous determination of 11 polyphenolic substances by high performance liquid chromatography-diode array detector (HPLC-DAD) was applied. Limits of quantification ranged between 0.78 and 3.14 mg L-1. High concentrations of kaempferol were found. An exploratory evaluation was performed using principal component analysis (PCA) for fruit origin differentiation. Total phenolic content (TPC) ranged from 274 to 517 mg GAE 100 g-1; total flavonoid content (TFC) ranged from 101 to 283 mg QE 100 g-1. In vitro analyses showed that antioxidant capacity with 2,2-diphenyl-1-picrylhydrazyl (DPPH) ranged from 1.7 to 7 µM DPPH 100 g-1. Poly(amidoamine)-type dendrimers (PAMAM) were synthesized by divergent routes, and generations (G0.5, G1.0, G1.5, G2.0 and G2.5) along with hydroxylated half-generation polymers (G0.5-OH, G1.5-OH and G2.5-OH) were characterized by Fourier-transform infrared spectroscopy, thermogravimetric analysis and differential scanning calorimetry. Carbon dioxide (CO2) capture tests were performed using thermogravimetric analysis. Among the absorption temperatures tested, 60 °C was the most promising: 0.77, 1.08, and 1.14 mol CO2 L 1 for G0.5-OH, G1.5-OH and G2.5-OH, respectively, of CO2 and partial pressure of 45 kPa. This showed that dendrimers with larger molecular structures have more hydroxyl groups and consequently capture more CO2. Species from Pavonia genus (Malvaceae) are traditionally used as antioxidant, n anti inflammatory and antimicrobial remedies. Previous studies have revealed their diversity of specialized metabolites widely used in the development of new medicines. This is the first study on Pavonia glazioviana Gurke, a Brazilian species known as “Malva da Chapada”. RESUMO Regiões com climas semiáridos apresentam condições ambientais que resultam em estresses abióticos às plantas. A maior área de produção de melão (Cucumis melo L.) do Brasil, o Estado do Rio Grande do Norte, possui essas características climáticas. O uso de bioestimulantes nesses sistemas de produção pode mitigar os efeitos dos estresses abióticos e aumentar a produtividade e a qualidade dos frutos. O objetivo neste estudo foi avaliar as características fisiológicas e de produção do meloeiro, sob aplicação de bioestimulantes. Os bioestimulantes Acadian®, Folicist® e Nov@® foram aplicados e comparados ao tratamento controle (sem bioestimulantes), em melões ‘Goldex’ e ‘McLaren’. Nov@® proporcionou maior assimilação de CO2, transpiração e condutância estomática, assim como ascorbato peroxidase em ‘Goldex’ e ‘McLaren’. Maior catalase foi obtida pela aplicação de Nov@®, enquanto que o uso de Folicist® resultou em maior teor de malondialdeído. A cultivar McLaren apresentou o melhor desempenho fisiológico e bioquímico e a ‘Goldex’ o melhor rendimento. ABSTRACT Regions with semi-arid climates have environmental conditions that result in abiotic stress to plants. The largest melon (Cucumis melo L.) production area in Brazil, the state of Rio Grande do Norte, has these climatic characteristics. Use of biostimulants in these production systems can mitigate effects of abiotic stress and increase yield and fruit quality. The objective in this study was to evaluate the physiological and production characteristics of melon, under the application of biostimulants. The biostimulants Acadian®, Folicist® and Nov@® were applied and compared to the control treatment (no biostimulants), in ‘Goldex’ and ‘McLaren’ melons. Nov@® led to higher CO2 assimilation, transpiration and stomatal conductance, as well as ascorbate peroxidase, in ‘Goldex’ and ‘McLaren’. Greater catalase was obtained with the application of Nov@®, while the use of Folicist® resulted in higher malondialdehyde content. The cultivar ‘McLaren’ exhibited the best physiological and biochemical performance, and ‘Goldex’ the best yield. Compared to other species, components of Combretum lanceolatum are still with few data reported in the literature. This work describes the compounds identification, antioxidant and antimicrobial activities of C. lanceolatum twigs. Ethanolic extract (EECLT) from C. lanceolatum twigs was submitted to partition obtaining: hexane (HF), diethyl ether (EEF), ethyl acetate (EAF) and hydromethanolic (HMF) fractions. They were analyzed by gas chromatography-mass spectrometry, high performance liquid chromatography coupled to mass spectrometry and direct insertion mass spectrometry and evaluated for antioxidant assay (2,2-diphenyl-1-picrylhydrazyl (DPPH) method) and antimicrobial assay. Fifty-seven compounds were identified in the fractions. In antioxidant assay, EECLT and EAF demonstrated good antioxidant potential (half maximal effective concentration (EC50): 57.9 and 45.4 µg mL–1, respectively), better than the positive control butylated hydroxytoluene (BHT) (EC50: 69.34 µg mL–1). For antimicrobial assay, all fractions presented promising minimal inhibitory concentration (MIC). EECLT presented the best values against Staphylococcus aureus (125 µg mL–1) and Escherichia coli (62.5 µg mL–1) and better than the positive control fluconazole (8 and 16 µg mL–1) against Candida albicans (3.9 µg mL–1) and Candida krusei (15.6 µg mL–1) strains. This paper presents an analytical multiresidue method for the quantification of 7 nitroimidazoles, 19 anthelmintics, 3 amphenicols, and 4 dyes in poultry, pork, beef, horse, shrimp, and fish samples by isotope dilution ultra-high performance liquid chromatography coupled with tandem mass spectrometry (UHPLC-MS/MS) in a single 6 min run. Samples were prepared using a modified quick, easy, cheap, effective, rugged and safe (QuEChERS)-type extraction with ethyl acetate and Na2SO4-NaCl (2:1), concentration into dimethyl sulfoxide (DMSO), and defatting by liquid-liquid extraction (LLE) with n-hexane. No clean-up using solid-phase extraction (SPE) cartridges or dispersive solid-phase extraction (DSPE) was necessary. Abstract Objective To evaluate the association between inflammatory markers and abdominal fat assessed by ultrasound in prepubertal children with and without excess weight. Methods A cross-sectional study involving 241 prepubertal children, 156 with obesity, 37 with overweight, and 48 with normal weight, aged five to ten years, who were followed at a research unit on Childhood Obesity from a teaching hospital belonging to a public health system. The concentration of interleukin-6, tumor necrosis factor-α and C-reactive protein were assessed and regression analyses, considering outcome variables such as abdominal wall and intra-abdominal fat thickness measured by ultrasound, were performed. Results The findings highlighted an association between abdominal fat and inflammatory markers, even in children at this young age group. Subcutaneous fat showed a stronger association with inflammatory biomarkers compared to intra-abdominal fat when performing logistic regression, with a positive association between tumor necrosis factor-α and abdominal wall thickness equal to or greater than the 75th percentile in adjusted logistic regression (OR: 18.12; CI 95 %: 1.57: 209.55). RESUMO O potássio é um dos nutrientes mais importantes para o algodoeiro, sendo um elemento que participa de muitas funções fisiológicas na planta. Além disso, sua adequada disponibilidade favorece melhor desempenho produtivo à cultura. Logo, objetivou-se avaliar nesta pesquisa os componentes agronômicos em cultivares de algodão naturalmente colorido adubadas com doses de potássio no Semiárido brasileiro. Para isto, dois experimentos foram conduzidos na Fazenda Experimental Rafael Fernandes, localizada no município de Mossoró, RN, nos períodos de julho a novembro de 2019 e julho a novembro de 2021. O delineamento experimental foi o de blocos casualizados, arranjados em parcelas subdivididas, com quatro repetições. Nas parcelas, foram alocadas as doses de potássio (0, 60, 120, 180 e 240 kg ha-1 de K2O); e, nas subparcelas, as quatro cultivares de algodão naturalmente colorido (BRS Rubi, BRS Safira, BRS Verde e BRS Topázio). As cultivares de algodão colorido respondem à adubação potássica em função das condições edafoclimáticas as quais estão expostas. ABSTRACT Potassium is one of the most important nutrients for the cotton plant, being an element that participates in many physiological functions. In addition, its adequate availability favors better production performance of the crop. Therefore, the objective of this study was to evaluate the agronomic components in naturally colored cotton cultivars fertilized with potassium doses in the Brazilian semiarid region. For this, two experiments were conducted at Rafael Fernandes Experimental Farm, located in the municipality of Mossoró, RN, from July to November 2019 and July to November 2021. The experimental design was randomized blocks, arranged in split plots, with four repetitions. Potassium doses (0, 60, 120, 180, and 240 kg ha-1 of K2O) were allocated in the plants; and four naturally colored cotton cultivars (BRS Rubi, BRS Safira, BRS Verde, and BRS Topázio) were allocated in the subplots. Colored cotton cultivars respond to potassium fertilization depending on the edaphoclimatic conditions to which they are exposed. BRS Topázio cultivar had higher lint cotton yield and fiber percentage in both growing seasons, highlighting its potential for the region. The dose of 240 kg ha-1 of K2O increased the yield of colored cotton, in second crop cultivation. Resumo Este estudo objetivou avaliar os efeitos da temperatura e motilidade embrionária sobre o desenvolvimento esquelético de jacaré-do-pantanal (Caiman yacare). Os ovos foram incubados com 90% de umidade e empregou-se a temperatura de 29°C por 45 dias. Após, para a incubação do Grupo I a temperatura continuou em 29°C, mas associou-se à injeção de 4-aminopiridina (29°C-4AP, n = 15) aplicada nos dias 46, 47, 48 e 49, do Grupo II permaneceu em 29°C (n = 14) e do Grupo III elevou-se para 33°C (n = 14). A movimentação foi mensurada através do monitor digital Egg Buddy® nos dias 30, 35, 42, 49, 56 e 60 dias. Aos 60 dias, os embriões foram eutanasiados e coletadas amostras embrionárias. Na análise estatística não foram observadas diferenças entre os grupos para o fator temperatura sobre a motilidade embrionária no desenvolvimento esquelético. Em contraste, a motilidade evidenciou diferença estatística no dia 49 para o Grupo I (P < 0,001) e apresentou maiores proporções de nariz e mão. Esses dados demonstraram pela primeira vez que o aumento na motilidade, induzidos farmacologicamente resultam em divergências fenotípicas na proporção de segmentos anatômicos durante a ontogenia pré-natal, podendo alterar efetivamente a adaptação dos animais em ambientes específicos.

Abstract This study aimed to evaluate whether skeletal development of the Pantanal Caiman (Caiman yacare) is similarly influenced by temperature variation and controlled increases in embryo motility. All eggs were incubated at 90% humidity and 29 °C for the first 45 days. Thereafter, the incubation temperature was either maintained at 29 °C and embryos were treated with 4-aminopyridine (4-AP) on days 46, 47, 48, and 49 (Group I, 29 °C 4-AP, n = 15); maintained at 29 °C (n = 14; Group II); or at 33 °C (n = 14, Group III). Embryonic movement was measured using an Egg Buddy® digital monitor on days 30, 35, 42, 49, 56, and 60, at which point embryos were euthanized and samples were collected for analysis. No differences were observed between groups with varying incubation temperatures. In contrast, embryonic motility was greater in embryos treated with 4-AP (P < 0.001) on day 49, and this was associated with higher proportions of snout-vent and hand lengths. This study demonstrates for the first time that pharmacologically induced increases in embryo motility result in phenotypic changes to the proportion of elements during prenatal ontogeny, thereby effectively altering the adaptation of the species to specific environments.

Até o momento, os NETs têm sido implicadas em danos aos tecidos durante processos inflamatórios intensos, principalmente quando sua liberação depende da geração de radicais de oxigênio. Os flavonóides são agentes antioxidantes e anti-inflamatórios, e destes, a quercetina é comumente encontrada em nossa dieta diária. Portanto, a quercetina pode exercer alguma atividade protetora contra o dano tecidual induzido por NETs. Em nossos ensaios in vitro, a quercetina reduziu a liberação das NETs, mieloperoxidase (MPO) e elastase a partir de neutrófilos estimulados com forbol 12-miristato 13-acetato (PMA). A atividade dessas enzimas também foi diminuída na presença de quercetina. A quercetina também reduziu o efeito citotóxico dos NETs sobre células alveolares (linha celular A549). Além disso, os ensaios in silico indicaram interações favoráveis entre a quercetina e as proteínas da NET (MPO e elastase). No geral, nossos resultados demonstram que a quercetina diminui os efeitos deletérios das NETs, reduzindo sua liberação à partir de neutrófilos ativados e diminuindo a atividade enzimática de MPO e elastase, possivelmente por meio de interação direta.

Abstract Neutrophil extracellular traps (NETs) were first reported as a microbicidal strategy for activated neutrophils. Through an immunologic response against several stimuli, neutrophils release their DNA together with proteins from granules, nucleus, and cytoplasm (e.g., elastase and myeloperoxidase). To date, NETs have been implicated in tissue damage during intense inflammatory processes, mainly when their release is dependent on oxygen radical generation. Flavonoids are antioxidant and anti-inflammatory agents; of these, quercetin is commonly found in our daily diet. Therefore, quercetin could exert some protective activity against tissue damage induced by NETs. In our in vitro assays, quercetin reduced NETs, myeloperoxidase (MPO), and elastase release from neutrophils stimulated with phorbol 12-myristate 13-acetate (PMA). The activity of these enzymes also decreased in the presence of quercetin. Quercetin also reduced the cytotoxic effect of NETs on alveolar cells (A549 cell line). Further, in silico assays indicated favorable interactions between quercetin and NET proteins (MPO and elastase). Overall, our results demonstrate that quercetin decreases deleterious cellular effects of NETs by reducing their release from activated neutrophils, and diminishing the enzymatic activity of MPO and elastase, possibly through direct interaction.

Resumo A leishmaniose visceral é uma doença tropical negligenciada. Foram avaliadas a distribuição espacial dos casos de leishmaniose visceral no estado de Alagoas. Todos os casos de LV, registrados pela secretaria de saúde, foram analisados e georreferenciados. Entre 2008 e 2017, 97,1% dos municípios apresentaram classificação esporádica de transmissão. Com a evolução temporal, a incidência de casos de leishmaniose visceral se concentrou na maioria dos municípios da microrregião de Santana do Ipanema-AL. A análise espaço-tempo, se considerada, pode promover o aprimoramento das ações de vigilância e controle da leishmaniose visceral.

Abstract Visceral leishmaniosis is a neglected tropical disease. We evaluated the spatial distribution of cases of visceral leishmaniosis in the state of Alagoas, Brazil. All cases of VL, registered by the health department, were analyzed and georeferenced. Results: Between 2008 and 2017, 97.1% of the municipalities presented sporadic classification of transmission. With temporal evolution, the incidence of cases of visceral leishmaniosis was concentrated in most municipalities in the microregion of Santana do Ipanema-AL. Space-time analysis, if considered, may promote the improvement of surveillance and control actions of visceral leishmaniosis.

Resumo Em uma escala global o Sorghum bicolor (L.) é uma das culturas mais importantes, ocupando o quinto lugar entre todos os cereais. É cultivada na África, Ásia, Oceania e Américas, onde serve como fonte de alimento e ração para humanos e animais, respectivamente. No entanto, a produção é limitada por vários fatores, incluindo pragas e doenças. Entre as pragas que atacam o sorgo, Schizaphis graminum (Rondani), comumente chamado de pulgão verde dos cereais, é a praga mais destrutiva do sorgo. O dano de S. graminum ao sorgo é agravado pelo déficit hídrico que favorece sua ocorrência. Informações limitadas sobre a distribuição espacial da praga e seus inimigos naturais impedem o desenvolvimento de estratégias de manejo ecologicamente corretas. Portanto, o objetivo desta pesquisa foi determinar a distribuição espaço-temporal de S. graminum e seu inimigo natural Coccinella septempunctata (L.) durante a fase vegetativa e reprodutiva da cultura por meio de análise geoestatística. A distribuição espaço-temporal de S. graminum foi influenciada pelo estágio de desenvolvimento do sorgo, paisagem circundante e presença de seu principal predador C. septempunctata. Além disso, a abundância de C. septempunctata foi influenciada pela densidade de S. graminum. Os resultados deste estudo são necessários para o desenvolvimento de estratégias sustentáveis de manejo de pragas contra S. graminum.

Abstract At the global level, Sorghum bicolor (L.), is one of the most important crops, which ranks fifth among all cereals. It is cultivated in Africa, Asia, Oceania and the Americas, where it serves as a source of food and feed for humans and animals, respectively. However, production is constrained by several factors including pests and diseases. Among the pests that are known to attack Sorghum, Schizaphis graminum (Rondani), commonly called the green cereal aphid, is the most destructive pest of sorghum. S. graminum damage to sorghum is worsen by water deficit which favors its occurrence. Limited information on the spatial distribution of the pest and its natural enemies impedes the development of ecologically friendly management strategies. Therefore, the objective of this research was to determine the spatiotemporal distribution of S. graminum and its natural enemy Coccinella septempunctata (L.) during the vegetative and reproductive stage of the crop using geostatistical analysis. The spatiotemporal distribution of S. graminum was influenced by Sorghum development stage, surrounding landscape, and presence of its main predator C. septempunctata. Moreover, the abundance of C. septempunctata was influenced by the density of S. graminum. The findings from this study are required for developing sustainable pest management strategies against S. graminum.

Resumo A busca por herbicidas mais amigáveis ao meio ambiente, visando o controle de pragas agrícolas, aliado a uma menor nocividade à saúde humana e ao meio ambiente tem crescido. Uma alternativa utilizada pelos pesquisadores é a aplicação de produtos do metabolismo secundário de plantas, que são investigados em virtude do seu potencial bioativo. Assim, espécies pertencentes à família Myrtaceae são potenciais nestes estudos, uma vez que esta família é reconhecida por possuir alta atividade biológica. Uma espécie pertencente a este gênero é Psidium cattleyanum, que possui efeito medicinal e seus frutos são utilizados na alimentação humana. Assim, o objetivo desta pesquisa foi avaliar e comparar a fitocitogenotoxicidade de extratos foliares aquosos e etanólicos da espécie P. cattleyanum, a partir de bioensaios vegetais, bem como identificar as principais classes de compostos presentes nos extratos. Para isso, os extratos foram preparados e caracterizados e foram realizados testes biológicos avaliando, em sementes e plântulas de alface e sorgo, as variáveis: porcentagem de germinação, índice de velocidade de germinação, crescimento radicular e crescimento aéreo; em células meristemáticas de alface foram avaliadas as variáveis: fases mitóticas, índice mitótico, alterações nucleares e alterações cromossômicas. Flavonas, flavononas, flavonóis, flavononóis, flavonóides, alcalóides, resinas, xantonas e glicosídeo de antraquinona foram caracterizados no extrato etanólico. Ambos os extratos avaliados, na maior concentração, inibiram o desenvolvimento inicial da planta. Todos os tratamentos causaram alterações nas fases mitóticas e inibiram o índice mitótico. Além disso, os tratamentos promoveram aumento de alterações nucleares e cromossômicas. O mecanismo de ação apresentado foi aneugênico, clastogênico e epigenético. O extrato etanólico foi mais citotóxico, pois teve efeito mais expressivo em menor concentração. Apesar da citotoxicidade dos extratos em estudo, eles promoveram alterações em níveis inferiores ao controle positivo glifosato.

Abstract The search for more environmental friendly herbicides, aiming at the control of agricultural pests, combinated with less harmfulness to human health and the environment has grown. An alternative used by researchers is the application of products of secondary plant metabolism, which are investigated due to their potential bioactivities. Thus, species belonging to the Myrtaceae family are potential in these studies, since this family is recognized for having high biological activity. A species belonging to this genus is Psidium cattleyanum, which has a medicinal effect and its fruits are used in human food. Thus, the objective of this research was to evaluate and compare the phyto-cyto-genotoxicity of aqueous and ethanolic leaf extracts of the specie P. cattleyanum, from plant bioassays, as well as to identify the main classes of compounds present in the extracts. For this, the extracts were prepared, characterized and biological tests were carried out by evaluating, in seeds and seedlings of lettuce and sorghum, the variables: percentage of germination, germination speed index, root growth and aerial growth; and in meristematic lettuce cells the variables: mitotic phases, mitotic index, nuclear alterations and chromosomal alterations. Flavones, flavonones, flavonols, flavononols, flavonoids, alkaloids, resins, xanthones and anthraquinone glycoside were characterized in the ethanolic extract. Both evaluated extracts, in the highest concentration, inhibited the initial plant development. All treatments caused alterations in the mitotic phases and inhibited mitotic index. In addition, the treatments promoted an increase in nuclear and chromosomal alterations. The mechanism of action presented was aneugenic, clastogenic and determined in epigenetic alterations. The ethanolic extract was more cytotoxic, since it had a more expressive effect at a lower concentration. Despite the cytotoxicity of the extracts under study, they promoted alterations at lower levels than the glyphosate positive control.
