Effects of a supplemented diet containing 7 probiotic strains (Honeybeeotic) on honeybee physiology and immune response: analysis of hemolymph cytology, phenoloxidase activity, and gut microbiome

Abstract Background In this study, a probiotic mixture (Honeybeeotic) consisting of seven bacterial strains isolated from a unique population of honeybees (Apis mellifera ligustica) was used. That honeybee population was located in the Roti Abbey locality of the Marche Region in Italy, an area isolated from human activities, and genetic contamination from other honeybee populations. The aim was to investigate the effects of this probiotic mixture on the innate immunity and intestinal microbiome of healthy common honeybees in two hives of the same apiary. Hive A received a diet of 50% glucose syrup, while hive B received the same syrup supplemented with the probiotics, both administered daily for 1 month. To determine whether the probiotic altered the immune response, phenoloxidase activity and hemolymph cellular subtype count were investigated. Additionally, metagenomic approaches were used to analyze the effects on gut microbiota composition and function, considering the critical role the gut microbiota plays in modulating host physiology. Results The results revealed differences in hemocyte populations between the two hives, as hive A exhibited higher counts of oenocytoids and granulocytes. These findings indicated that the dietary supplementation with the probiotic mixture was safe and well-tolerated. Furthermore, phenoloxidase activity significantly decreased in hive B (1.75 ± 0.19 U/mg) compared to hive A (3.62 ± 0.44 U/mg, p < 0.005), suggesting an improved state of well-being in the honeybees, as they did not require activation of immune defense mechanisms. Regarding the microbiome composition, the probiotic modulated the gut microbiota in hive B compared to the control, retaining core microbiota components while causing both positive and negative variations. Notably, several genes, particularly KEGG genes involved in amino acid metabolism, carbohydrate metabolism, and branched-chain amino acid (BCAA) transport, were more abundant in the probiotic-fed group, suggesting an effective nutritional supplement for the host. Conclusions This study advocated that feeding with this probiotic mixture induces beneficial immunological effects and promoted a balanced gut microbiota with enhanced metabolic activities related to digestion. The use of highly selected probiotics was shown to contribute to the overall well-being of the honeybees, improving their immune response and gut health.

Resumen Spondias mombin es un árbol nativo de la región amazónica; su explotación se produce de manera extractiva, siendo aún incipiente el cultivo de la especie. El objetivo de este estudio fue evaluar la divergencia genética entre genotipos de S. mombin y cuantificar la contribución relativa de 12 características morfológicas de frutos y semillas, para obtener datos que permitan investigaciones futuras relativas a la conservación y domesticación de la especie. Se evaluaron 60 genotipos; de cada uno se analizaron diez frutos. Los descriptores utilizados para la caracterización de los frutos fueron ocho: masa del fruto, peso de la pulpa, volumen, longitud, anchura, espesor, contenido total de sólidos solubles y pH; los descriptores de semillas fueron cuatro: masa, longitud, anchura y espesor. Los datos se evaluaron por medio de componentes principales y agrupaciones obtenidas por métodos UPGMA y Tocher, de la matriz de disimilitud (distancia euclideana media) con ayuda del programa GENES. A través del análisis de los componentes principales, se verificó que los tres primeros componentes explican 83.63 % de la variación acumulada. Las características que más contribuyen a la discriminación de los genotipos fueron: anchura del fruto, peso de la pulpa, pH, longitud y espesor de semillas, y son las más significativas para la selección de genotipos de S. mombin. Las contribuciones más pequeñas a la diversidad se deben a: la masa del fruto, anchura de semilla, espesor, volumen y longitud del fruto, así como a la masa de las semillas y el contenido de sólidos solubles totales. Ambos métodos de agrupación, UPGMA y Tocher, revelaron que existe divergencia genética entre los genotipos analizados. Los genotipos 37 y 41 son los más divergentes respecto a los otros; por eso son ideales para programas de mejoramiento genético con miras a la domesticación de la especie.
Abstract The species Spondias mombin, is native to the Amazonian region. Since these trees’ cultivation is incipient, their exploitation is done through extraction techniques. The aim of the present study was to assess the genetic divergences between S. mombin genotypesand to quantify the relative contribution from 12 morphological traits of the species’ fruits and seeds, as well as to collect data able to subsidize future research on the species conservation and domestication. 60 genotypes were assessed in total, and ten fruits of each genotype were analyzed. Eight descriptors were used for fruit characterization, namely: fruit mass, pulp weight, volume, length, width, thickness, total soluble solids content and hydrogenionic potential. The seed descriptors were mass, length, width and thickness. The data were assessed through the principal components and groupings by applying the UPGMA and Tocher methods. They were analyzed in the GENES software, based on the dissimilarity matrix (Euclidean distance average). The analysis applied to the principal components showed that the first three components explained 83 % accumulated variation. The main traits contributing to the genotype discrimination were fruit width, fruit pulp weight, pH, seed length and thickness, and the most responsive traits to S. mombin genotype selection. The features fruit mass, seed width, fruit thickness, fruit volume, fruit length, seed mass and total soluble solid content presented the smallest contribution to diversity. The grouping methods UPGMA and Tocher evidenced genetic divergence between the analyzed genotypes. Genotypes 37 and 41 were more divergent than the others, what makes them promising for crossings in future genetic enhancement programs focused on the species’ domestication.

Different roles of resilience in depressive patients with history of suicide attempt and no history of suicide attempt

Objective: Suicidal ideation is modulated by several risk and protective factors. The aim of this study was to evaluate differences between patients with a history of suicide attempt and those with no such history, with special attention to depression, interpersonal sensitivity, humiliation, and resilience. Methods: One hundred consecutively admitted patients with an index depressive episode were recruited. The Brief Symptom Inventory, Humiliation Inventory, and Resilience Scale for Adult were administered. Results: Scores for humiliation, interpersonal sensitivity, and depression were higher in subjects with history of suicide attempt, while higher scores for resilience were observed in the group with no such history. Different patterns of relationships among the variables of interest were found in the two groups. Resilience dimensions such as social resources and familial cohesion were strongly and negatively correlated with humiliation, interpersonal sensitivity, and depression in subjects with a past suicide attempt. Conclusions: Resilience factors can modulate and reduce the impact of suicide risk. Assessing risk and protective factors could enhance the ability to intervene appropriately.

RESUMO Theobroma subincanum (cupuí) é espécie frutífera nativa da Amazônia, cujos frutos são apreciados pela população local, para consumo tanto in natura, como na forma de suco, néctar ou refresco. Estudos sobre a biologia reprodutiva podem fornecer informações e estratégias para a conservação dos frutos e melhoramento vegetal da espécie. O presente estudo objetivou analisar a receptividade de estigmas e a viabilidade polínica de T. subincanum em diferentes horários durante a antese. O estudo foi desenvolvido em um fragmento florestal localizado no perímetro urbano do município de Alta Floresta, MT. No período de florescimento da espécie foram coletados botões florais ou flores em sete horários diferentes: 22h; 02h; 06h; 10h; 14h; 18h e às 22h. A viabilidade polínica foi estimada com o reativo de Alexander (1969) e a receptividade estigmática com o uso do peróxido de hidrogênio a 3%. As médias de viabilidade polínica foram submetidas à análise de variância, enquanto que a receptividade estigmática foi analisada pelo percentual médio em cada intervalo. Foram caracterizados quatro estádios florais com base na abertura floral, sendo que a partir das 6h da manhã já foram encontradas flores totalmente abertas (estádio IV). A percentagem de viabilidade polínica não foi afetada pelos horários de coleta. Na análise da receptividade estigmática foi observado que, em todos os estádios florais, o estigma encontrava-se receptivo, entretanto, os maiores percentuais de receptividade foram encontrados entre as 2h e às 10h do mesmo dia, sendo estes os horários mais propícios para a fertilização. A coleta do pólen em T. subincanum pode ser realizada em qualquer um dos horários avaliados neste estudo, visto que o mesmo se mantem com um alto percentual de viabilidade.
ABSTRACT Theobroma subincanum (cupuí) is a fruit species native to the Amazon region. Fruits are enjoyed by local people and consumed both as fresh fruit such as juice, nectar or soft drinks. Reproductive biology studies provide contributions to conservation strategies and plant improvement. The present study aimed to analyze the stigmatic receptivity and pollen viability of T. subincanum. This study was developed in a forest fragment located in the urban perimeter of the municipality of Alta Floresta, MT. In the flowering period, flower buds or flowers were collected in seven different times, as follows: 10 pm, 02 am, 06 am, 10 am, 2 pm, 6 pm, and 10 pm. Pollen viability was estimated by reactive Alexander (1969) and stigmatic receptivity using 3% hydrogen peroxide. Pollen viability averages were submitted to analysis of variance, while stigmatic receptivity was analyzed by average percentage in each interval. Four floral stages were characterized based on flower opening, and from 6 am, fully opened flowers have already been found (stage IV). The percentage of pollen viability was not affected by collection times. In the stigmatic receptivity analysis, it was observed that in all floral stages, stigma was receptive; however, the highest percentages of stigmatic receptivity were found from 2 am to 10 am of the same day, which is the most propitious time for fertilization. Pollen collection of T. subincanum may be performed in any of schedules evaluated in this study, since it is held with high viability percentage.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

INTRODUCTION: In the present work we describe the mental health condition of L'Aquila population in the aftermath of the earthquake in terms of structural, process and outcome perspectives. METHOD: Literature revision of the published reports on the L'Aquila earthquake has been performed. RESULTS: Although important psychological distress has been reported by the population, capacity of resilience can be observed. However if resilient mechanisms intervened in immediate aftermath of the earthquake, important dangers are conceivable in the current medium-long-term perspective due to the long-lasting alterations of day-to-day life and the disruption of social networks that can be well associated with mental health problems. CONCLUSIONS: In a condition such as an earthquake, the immediate physical, medical, and emergency rescue needs must be addressed initially. However training first responders to identify psychological distress symptoms would be important for mental health triage in the field.
INTRODUZIONE: Il presente lavoro intende descrivere la condizione della salute mentale della popolazione dell'Aquila a seguito del terremoto in termini di prospettive di struttura, di processo ed esito. METODO: È stata eseguita una revisione di letteratura relativa gli articoli pubblicati sul terremoto dell'Aquila. RISULTATI: Anche se la popolazione ha manifestato un disagio psicologico importante, viene rilevata capacità di resilienza. Tuttavia, se sono intervenuti meccanismi resilienti nel periodo immediatamente successivo al terremoto, è verosimile considerare pericoli importanti a medio-lungo termine a causa della lunga durata delle quotidiane difficoltà di vita e della disgregazione delle reti sociali che possono essere ben associate a problemi di salute mentale. CONCLUSIONI: In una situazione quale quella di un terremoto le esigenze immediate di soccorso per problematiche di ordine fisico, medico e di emergenza devono essere affrontate per prime. Tuttavia sarebbe importante la formazione dei soccorritori di primo intervento per identificare i sintomi di stress psicologico e permettere un triage per la salute mentale sul campo.
