ABSTRACT Objective The establishment of reference values for a subset of leukocytes is common in clinical practice, and ethnic variations are strongly associated with disease development. In Brazil, indigenous people are vulnerable to infections, and few studies have described the health and disease conditions of this population. This study aimed to provide reference values for immunological cell subsets in indigenous Brazilians living in the state of Mato Grosso do Sul. Methods Flow cytometry and 4-color combinations of monoclonal antibodies were used to characterize cells. A total of 115 healthy adults, mostly females (72%), were included in the study. The results are presented as mean and median (2.5%-97.5% percentiles) for T and B lymphocytes, CD4+ T cells, CD8+ T cells, Natural Killer cells, monocytes, and dendritic cells, providing an average immunological profile for the population in question. Results The relative medians of CD3+, CD4+, and CD8+ T cells were significantly higher in women than in men in a healthy indigenous population. Conclusion To our knowledge, cell reference data from indigenous Brazilians are unknown in the literature. Abstract Introduction The deficiency of ADA2 (DADA2) is a rare autoinflammatory disease provoked by mutations in the ADA2 gene inherited in a recessive fashion. Up to this moment there is no consensus for the treatment of DADA2 and anti-TNF is the therapy of choice for chronic management whereas bone marrow transplantation is considered for refractory or severe phenotypes. Data from Brazil is scarce and this multicentric study reports 18 patients with DADA2 from Brazil. Patients and methods This is a multicentric study proposed by the Center for Rare and Immunological Disorders of the Hospital 9 de Julho - DASA, São Paulo - Brazil. Patients of any age with a confirmed diagnosis of DADA2 were eligible for this project and data on clinical, laboratory, genetics and treatment were collected. Results Eighteen patients from 10 different centers are reported here. All patients had disease onset at the pediatric age (median of 5 years) and most of them from the state of São Paulo. Vasculopathy with recurrent stroke was the most common phenotype but atypical phenotypes compatible with ALPS-like and Common Variable Immunodeficiency (CVID) was also found. All patients carried pathogenic mutations in the ADA2 gene. Acute management of vasculitis was not satisfactory with steroids in many patients and all those who used anti-TNF had favorable responses. ABSTRACT The objective of this study was to evaluate the effects of different levels of microencapsulated carvacrol and cinnamaldehyde on metabolizable energy, feed digestibility, intestinal morphometric analysis, and antioxidant parameters in broilers. A completely randomized design with five treatments and eight replications of eight broilers (Cobb male) was used, and collections were carried out at 20 and 42 days of age. Carvacrol and cinnamaldehyde concentrations (mg/kg) were: 0 (control), 50, 100, 150, and 200. Carvacrol and cinnamaldehyde improved apparent metabolizable energy but did not change protein and ether extract digestibility. Supplementation increased intestinal villus height and villi:crypt ratio; in which 100 mg/kg produced the greatest villus height. Serum uric acid levels were lower in birds receiving supplementation. Improvement in the activity of glutathione peroxidase and glutathione-S-transferase was observed, while lower uric acid, thiobarbituric acid reactive substances, and reactive oxygen species levels were observed. RESUMO Objetivo Esta revisão de escopo teve como objetivo mapear evidências sobre a suplementação com açaí combinada com exercícios físicos em estudos experimentais em animais e / ou humanos. Métodos A busca considerou seis bases de dados eletrônicas além da triagem de referências relevantes. O processo de seleção e extração de dados foi realizado por dois autores independentes. As características do estudo, estratégias de suplementação de açaí (forma, tempo de intervenção, e quantidade ingerida) e exercícios (tipos, intensidade e duração), seus resultados foram resumidos. Resultados Um total de 342 estudos foram inicialmente alcançados e somente 11 foram elegíveis (5 com animais e 6 com humanos). Em animais, a suplementação de açaí e os exercícios indicaram benefícios na tolerância ao exercício e melhorias em vários parâmetros hemodinâmicos, bem como melhorias significativas nos marcadores hepáticos e no metabolismo da glicose. Em humanos, a suplementação de açaí indicou resultados positivos no aumento do tempo de exaustão para 90% do VO2máx e no aumento da intensidade correspondente ao limiar anaeróbio. Conclusão Concluiu-se que pesquisas futuras envolvendo animais e humanos devem examinar a suplementação de açaí e exercícios com (a) modelos de obesidade para testar o efeito da adiponectina na composição corporal por meio de parâmetros histológicos e histoquímicos (b) protocolos de dano muscular excêntrico com incorporação de variáveis de qualidade muscular para avaliação da recuperação; (c) suplementação crônica de açaí e treinamento de força; (d) comparação das diferentes formas de suplementação de açaí em protocolos de exercícios.

ABSTRACT Objective This scoping review aimed to map evidence on açai supplementation combined with exercise in animal and/or human experimental studies. Methods The search considered six electronic databases and screening of relevant references. The selection process and data extraction were performed by two independent authors. The study characteristics, and AS (e.g., form, intervention time, amount ingested) and exercise (e.g., types, intensity, and duration) strategies were summarized, as well as their reported results. Results From an initial total of 342 studies identified; 11 (5 with animal and 6 with human models) were eligible. In animals, açai supplementation and exercise led to benefits in exercise tolerance and improvements in several hemodynamic parameters, as well as significant improvements in liver markers and glucose metabolism. In humans, açai supplementation indicated positive results in increasing exhaustion time to 90% of VO2max and increasing intensity at the anaerobic threshold. Conclusion We conclude that future research involving animals and humans should examine açai supplementation and exercise with (a) obesity models to test the effect of adiponectin on body composition with analysis of histological and histochemical parameters; (b) eccentric injury protocols with the incorporation of muscle quality variables to assess recovery; (c) chronic açai supplementation and strength training; (d) comparison of different forms of açai supplementation in exercise protocols.

ABSTRACT Pseudomonas aeruginosa is one of the most common bacterium with a broad spectrum of human-associated infections. It is intrinsically resistant to many antimicrobial drugs, making carbapenems crucial in clinical management. The emergence and dissemination of carbapenemases among P. aeruginosa clinical isolates is a serious public health concern as it limits the options for the treatment of bacterial infections. Here, we described the molecular and epidemiological characteristics of 28 carbapenem-resistant P. aeruginosa strains isolated from patients hospitalized in an intensive care unit (ICU). The antimicrobial susceptibility of carbapenem-resistant P. aeruginosa strains was determined by broth microdilution. The presence of resistance genes was evaluated by PCR and DNA sequencing. Additionally, alterations in genes encoding P. aeruginosa outer membrane proteins were analyzed by PCR as well as SDS-PAGE. Clinical characteristics of the patients and the economic impact of hospitalization on the public health system were evaluated. PCR amplification showed that the blaKPC-2 and blaTEM genes were identified in three isolates (11%) and blaSHV gene in two isolates (7%). Outer membrane profiles obtained by SDS–PAGE indicated that the OprD porin was either absent or was produced at very low levels. A PCR assay using oprD-specific primers failed to show the presence of mutations in this gene. P. aeruginosa strains were isolated from 28 patients, among whom 43% (12/28) had sepsis, 31% (9/28) had respiratory failure, and 31% (9/28) had systemic arterial hypertension. A high mortality rate (39%) was observed in these patients, with an average duration of hospitalization of 34.6 days and a median cost of 3.275 dollars per patient. The production of carbapenemase was not the main mechanism of resistance in these strains. All carbapenem-resistant P. aeruginosa were isolated from patients hospitalized in the ICU. Besides the high mortality rate, many patients remained hospitalized for several days, resulting in a high cost of hospitalization for the public health system. Therefore, the evolution of this resistance and its dissemination should be actively monitored among critically ill patients to improve their health conditions.

O objetivo deste estudo foi caracterizar o efeito do alumínio (Al) na concentração de zinco (Zn), manganês (Mn), ferro (Fe) e cobre (Cu) em quatro clones de batata (Macaca e Dakota Rose: sensíveis ao Al; e SMIC148-A e Solanum microdontum: tolerantes ao Al) crescendo em solução nutritiva (pH 4,0) com 0, 50, 100, 150 e 200mg Al L-1. A concentração de Zn e Fe em raízes diminuiu linearmente com o aumento dos níveis de Al nos clones Macaca, SMIC148-A e Dakota Rose e aumentou linearmente em S. microdontum. Na parte aérea, a concentração de Zn mostrou resposta quadrática ao Al em S. microdontum e SMIC148-A, enquanto no clone Dakota Rose houve uma resposta cúbica. Nos clones S. microdontum, SMIC148-A e Dakota Rose, a concentração de Fe mostrou resposta quadrática ao Al. A concentração de Mn em raízes diminuiu linearmente em relação ao Al nos clones Macaca e SMIC148-A e aumentou linearmente em S. microdontum. Para Dakota Rose e SMIC148-A, a concentração de Mn mostrou uma resposta quadrática em relação ao Al em raízes e parte aérea. A concentração de Mn na parte aérea aumentou de forma cúbica com os níveis de Al no clone Dakota Rose. Na parte aérea, não houve alteração na concentração de Zn e Fe na Macaca e de Mn nos clones Macaca e S. microdontum. Em raízes e na parte aérea, a concentração de Cu aumentou linearmente no clone Dakota Rose e mostrou resposta quadrática no clone Macaca. A concentração de Cu mostrou resposta quadrática com os níveis de Al em raízes dos clones S. microdontum e SMIC148-A. Na parte aérea, a concentração de Cu aumentou linearmente no clone S. microdontum e diminuiu linearmente no clone SMIC148-A com o aumento nos níveis de Al. Portanto, a acumulação excessiva de Al afetou negativamente a absorção e a distribuição de Zn, Fe, Mn e Cu nas raízes e na parte aérea dos clones de batata. A resposta da concentração de Cu na parte aérea ao Al foi menos alterada nos clones Al-tolerantes que naqueles Al-sensíveis. A tolerância ao Al em S. microdontum pode estar conectada com os maiores níveis de Zn, Fe e Mn nas raízes.

The objective of this study was to evaluate the effects of aluminum (Al) on the zinc (Zn), manganese (Mn), iron (Fe) and copper (Cu) concentrations in four potato clones (Macaca and Dakota Rose: both Al-sensitive clones; and SMIC148-A and Solanum microdontum: both Al-tolerant-clones), grown in a nutrient solution (pH 4.00) with 0, 50, 100, 150 and 200mg Al L-1. Root Zn and Fe concentrations decreased linearly with the increase of Al levels in Macaca, SMIC148-A and Dakota Rose and increased linearly in S. microdontum. Shoot Zn concentration showed a quadratic relationship with Al in S. microdontum and SMIC148-A, but a curvilinear response in Dakota Rose. Shoot Fe concentration showed a quadratic relationship with Al in S. microdontum, SMIC148-A and Dakota Rose. Root Mn concentration decreased linearly in Macaca and SMIC148-A, and increased linearly in S. microdontum with Al levels. Mn concentration showed a quadratic relationship with Al in roots of Dakota Rose and in shoot of SMIC148-A, and increased curvilinearly with Al levels in shoot of Dakota Rose. In shoot, there was no alteration in Zn, Fe and Mn in Macaca and Mn concentration in S. microdontum. Roots and shoot Cu concentration increased linearly in Dakota Rose, and showed quadratic relationship with Al in Macaca. Roots Cu concentration showed a quadratic relationship with Al levels in S. microdontum and SMIC148-A. Shoot Cu concentration increased linearly in S. microdontum, and decreased linearly in SMIC148-A. Therefore, the excessive Al accumulation affected the uptake and distribution of Zn, Fe, Mn and Cu in roots and shoot of potato clones.The response of shoot Cu concentration to Al was less altered in the Al-tolerant clones than was in Al-sensitive clones. Aluminum tolerance in S. microdontum may be connected with greater levels of Zn, Fe and Mn in the roots.

Dezenove amostras de Rosmarinus officinalis foram extraídas por destilação por arraste a vapor em uma planta piloto e os óleos essenciais foram avaliados quanto a composição química e características físico-químicas. Os rendimentos de óleo essencial variaram de 0,37% (média de 1999) a 0,49% (média de 1998). Vinte componentes foram identificados nos óleos essenciais. Os componentes majoritários foram a-pineno (40,55 a 45,10%), 1,8-cineol (17,40 a 19,35%), canfeno (4,73 a 6,06%) e verbenona (2,32 a 3,86%). A média por safra dos parâmetros físico-químicos avaliados resultaram em 0,8887g/cm³ para densidade específica, 1,4689 para índice de refração e +11,82º para rotação óptica, sendo que as variações encontradas para os parâmetros químicos e físico-químicos dos óleos essenciais não variaram significativamente em função do ano de produção.

Nineteen samples of Rosmarinus officinalis were extracted by steam distillation in a pilot plant and evaluated in terms of chemical compositions and physico-chemical characteristics. The volatile oil yields ranged from 0.37% (1999 harvest) to 0.49% (1998 harvest). Twenty components were identified in the oils. The major components were alpha-pinene (40.55 to 45.10%), 1,8-cineole (17.40 to 19.35%), camphene (4.73 to 6.06%) and verbenone (2.32 to 3.86%). The physico-chemical parameters averaged 0.8887 g/cm³ for specific gravity, 1.4689 for refractive index, and +11.82° for optical rotation, and there were no significant variations in either the chemical or physico-chemical data in the different years.
