Apesar de sua importância, o bioma vem sendo cada vez mais suprimido por atividades antrópicas. Neste sentido, este trabalho tem por objetivo analisar e compreender as características físico-hídricas do solo de uma fitofisionomia de Cerrado Ralo presente no Chapadão do diamante (Serra da Canastra-MG), gerando embasamento necessário para valoração e surgimento de futuras formas de manejo a áreas semelhantes. Para tal utilizou-se para prospecção de dados de campo um simulador de chuvas e um infiltrômetro de anéis concêntricos. Os resultados demostraram para área de estudos alta capacidade de infiltração, não exibindo volumes escoados superficialmente mesmo sob precipitações de alta intensidade (57,35mm), expondo altos valores de velocidade de infiltração básica (VIB) 626,56 mm/h. Abstract The Cerrado is characterized as an important Brazilian biome, forming strategic hydrographic basins within its territory that are significant both nationally and internationally. Despite its importance, the biome has been increasingly impacted by anthropogenic activities. In this context, this study aims to analyze and understand the physical-hydraulic characteristics of the soil in a sparse Cerrado physiognomy located in the Chapadão do Diamante (Serra da Canastra-MG), providing the necessary foundation for the assessment and development of future management strategies for similar areas. For field data prospecting, a rain simulator and a concentric ring infiltrometer were used. The results demonstrated a high infiltration capacity in the study area, with no significant surface runoff even under high-intensity precipitation (57.35 mm), revealing high values of basic infiltration velocity (BIV) at 626.56 mm/h. Abstract The cognitive deficit, which is like Alzheimer’s disease and is associated with oxidative damage, may be induced by exposure to streptozotocin. This study aimed to evaluate if the tellurium-containing organocompound, 3j, 5’-arylchalcogeno-3-aminothymidine derivative, interferes with the effects of streptozotocin, as well as to investigate its toxicity in adult mice. Cognitive deficit was induced by two doses of streptozotocin (2.25 mg/kg/day, 48 h interval) intracerebroventricularly. After, the mice were subcutaneously treated with 3j (8.62 mg/kg/day) for 25 days. The effects were assessed by evaluating hippocampal and cortical acetylcholinesterase and behavioral tasks. 3j toxicity was investigated for 10 (0, 21.55, or 43.10 mg/kg/day) and 37 (0, 4.31, or 8.62 mg/kg/day) days by assessing biometric parameters and glucose and urea levels, and alanine aminotransferase activity in blood plasma. 3j exposure did not alter the behavioral alterations induced by streptozotocin exposure. On the other hand, 3j exposure normalized hippocampus acetylcholinesterase activity, which is enhanced by streptozotocin exposure. Toxicity evaluation showed that the administration of 3j for either 10 or 37 days did not cause harmful effects on the biometric and biochemical parameters analyzed. The Ti-19Nb-2.5Fe-6Sn alloy subjected to proper heat treatments is a promising material to be applied in orthopedic implants. This type of Ti alloy presents relatively low cost, good biocompatibility, and reasonable mechanical strength combined with low elastic modulus. In such an alloy, Fe improves mechanical strength while ω phase precipitation can be controlled by Sn addition. In this work, samples of the Ti-19Nb-2.5Fe-6Sn (wt.%) alloy were prepared by arc melting, hot swaging, and solution heat treatment. Results from the literature combined with thermodynamic simulations, differential scanning calorimetry (DSC), and heating/cooling experiments coupled with microstructural analyses were applied to determine the β transus temperature of this alloy. Following, the samples were solution heat treated in the β field and continuously cooled to room temperature at different rates. The effects of cooling rates on intergranular and intragranular α phase precipitation were evaluated. It was found that low cooling rates lead to intergranular precipitation while moderate cooling rates provide more expressive intragranular precipitation. RESUMO: O estudo avaliou os efeitos da suplementação pela via intramuscular de alfa-tocoferol (vitamina E) na qualidade da carne de cordeiros mestiços Santa Inês e Dorper. Os animais foram mantidos em confinamento, sendo alimentados com dieta contendo alto teor de concentrado energético. Oito dias antes do abate os cordeiros foram distribuídos em quatro blocos, de acordo com seu ganho de peso. No sétimo e quarto dias antes do abate eles foram tratados por via intramuscular com 0, 10 ou 20 UI de DL-alfa-tocoferol por kg de peso metabólico. No momento do abate os animais tinham, em média, 138 dias de idade e 43,6 kg de peso vivo. As carcaças foram armazenadas a 2°C por 24 horas. Foram determinados pH (pH24h) e cor (luminosidade, teor de vermelho e teor de amarelo) do músculo Longissimus thoracis, bem como força de cisalhamento, perda por cocção, composição dos ácidos graxos e quantidade de substâncias reativas ao ácido tiobarbitúrico após um e cinco meses de congelamento. Observou-se efeito de desvio da linearidade para luminosidade (L*; P=0.0042) e para o croma amarelo (b*; P=0.0082). A administração intramuscular de 10 e 20 UI de alfa-tocoferol/kg de peso metabólico não influenciou a conservação de ácidos graxos das carcaças, mas melhorou os valores de L* e b*.

ABSTRACT: This study evaluated the effects of intramuscular alpha-tocopherol (vitamin E) supplementation on meat quality characteristics of Santa Inês and Dorper crossbreed lambs. All animals were feed with a high concentrated diet in feedlot. Eight days before slaughter, the animals were distributed into four blocks according to weight gain. At the seventh and fourth days before slaughter, they were intramuscularly treated with 0, 10 or 20 IU of DL-alpha-tocopherol per kg of metabolic body weight. At slaughter they had 138 days of age and 43.6 kg of live weight, in average. Carcasses were stored for 24 hours under refrigeration at 2°C. Longissimus thoracis muscle pH (pH24h) and color (lightness, yellowness and redness) were analyzed and its samples were collected for evaluation of shear force (SF), cooking loss (WLC), fatty acid composition (FA) and thiobarbituric acid reactive substances after one (TBARS1m) and after five months (TBARS5m) of freezing. Linearity deviation effect was observed for lightness (L*; P=0.0042) and yellowness (b*; P=0.0082). Intramuscular administration of 10 or 20 IU of alpha-tocopherol/kg of metabolic weight did not influence the conservation of fatty acid in the carcasses, but benefit L* and b* values.

Objetivou-se determinar as exigências nutricionais de energia e proteína para mantença e ganho de peso de bovinos cruzados, bem como suas eficiências. Foram utilizados 50 bovinos cruzados 3/4 Zebu × 1/4 Holandês, não castrados, com peso inicial de 214±4kg e idade média de 11±0,2 meses. Quatro animais foram usados para referência, dez para o grupo mantença e os 36 animais alimentados à vontade foram distribuídos em delineamento inteiramente ao acaso em esquema fatorial 3×3, sendo três períodos de confinamento (56, 112 ou 168 dias) e três níveis de cálcio e fósforo (baixo, médio e normal) na dieta. Quatro dos animais mantença foram encaminhados ao laboratório de metabolismo e calorimetria da UFMG para medição da produção de calor através da respirometria. Após os abates, foram obtidas duas amostras compostas para cada animal, denominadas carcaça e não carcaça. As exigências de energia líquida para mantença (ELm) e energia metabolizável para mantença (EMm) foram, respectivamente, de 68,9 e 90,1kcal/PCVZ0,75/dia. A eficiência (km) foi de 76,41%. A exigência de ELm obtida na câmara respirométrica foi de 85,5kcal/kg0,75. As equações obtidas para energia líquida de ganho (ELg) e proteína líquida de ganho (PLg) foram: ELg(Mcal/dia) = 0,0505±0,000986 × PCVZ 0,75 × GPCVZ 1,095e PLg (g/dia) = 162,79±18,2546 × GPCVZ - 1,30±5,3010 × ER. As eficiências para deposição de gordura e de proteína foram de 70,04 e 15,12%, respectivamente. Conclui-se que as exigências de ELm para bovinos cruzados 3/4 Zebu × 1/4 Holandês não castrados em crescimento e terminação são de 68,9kcal/PCVZ0,75/dia; e que as exigências de ELg e PLg podem ser obtidas pelas respectivas equações: ELg(Mcal/dia) = 0,0505±0,000986 × PCVZ 0,75 × GPCVZ 1,095e PLg (g/dia) = 162,79±18,2546 × GPCVZ - 1,30±5,3010 × ER.

The aim of this study was to determine the nutritional requirements of energy and protein for maintenance and weight gain of crossbred cattle, as well as their efficiencies. Fifty 3/4 Zebu × 1/4 Holstein crossbred bulls with initial weights of 214±4kg and aged 11±0.2 months on average were used in this experiment. Four animals were used in the reference group; ten bulls were fed at the maintenance level; and the remaining 36 bulls were fed ad libitum and distributed in a completely randomized design in a 3×3 factorial arrangement, which had three feedlot periods (56, 112 or 168 days) and three calcium and phosphorus levels (low, medium and normal) in the diet. Four of the maintenance animals had their heat production measured by respirometry at the Laboratory of Metabolism and Calorimetry of UFMG. After slaughter, composite samples, referred to as carcass and noncarcass samples were obtained from each animal. The net energy requirements for maintenance (NEm) and metabolizable energy for maintenance (MEm) were 68.9 and 90.1 kcal/EBW0.75/day, respectively. The efficiency (km) was 76.41%. The NEm requirement determined in the respirometry chamber was 85.5 kcal/kg0.75. The following equations were obtained for net energy for gain (NEg) and net protein for gain (NPg): NEg (Mcal/day) = 0.0505±0.000986 × EBW0.75× EBWG1.095 and NPg (g/day) = 162.79±18.2546 × EBWG - 1.30±5.3010 × RE. Os efeitos da inclusão de levedura seca de cana-de-açúcar (Saccharomyces cerevisiae) na dieta de vacas lactantes foram avaliados com base no fornecimento de quatro dietas constituídas de diferentes níveis de substituição do farelo de soja pela levedura de cana-de-açúcar (0,00; 0,33; 0,67 e 1,00kg/kg de MS). O delineamento utilizado foi o quadrado latino 4 × 4. Não houve efeito dos tratamentos sobre o consumo de MS, MO, EE, FDNi e NDT, porém ocorreu redução linear no consumo de PB e FDNcp e aumento do consumo de CNF. Diferenças estatísticas não foram verificadas para os coeficientes de digestibilidade aparente da MS, MO, PB, CNF e NDT; no entanto os coeficientes do FDNcp e do EE decresceram linearmente. Não foi observado efeito sobre a produção e eficiência de síntese microbiana, utilização dos componentes nitrogenados e produção e composição do leite. Conclui-se que a levedura seca pode substituir 100% o farelo de soja na alimentação de vacas lactantes.

The effects of the inclusion of sugar cane dry yeast (Saccharomyces cerevisiae) in dairy cows feeding were evaluated based on the provision of four different diets with increasing proportions of sugar cane dry yeast (0.00; 0.33; 0.67 and 1.00kg/kg of DM). The design adopted for this research was the latin square, 4 × 4. It was observed that the levels of yeast inclusion did not affect the intake of DM, OM, EE, NDFi and TDN. However, the CP and NDFcp intake decreased linearly, whereas the NFC intake increased linearly with the increasing levels of yeast. No effect on the digestibility coefficient of DM, OM, CP, NDFcp and NCF was observed. Nevertheless, the EE digestibility coefficient linearly decreased with the yeast inclusion in the experimental diets. There was no treatment effect on the production and efficiency of microbial synthesis, usage of nitrogenous components, as well as milk production and composition. It was concluded that the dried yeast can fully replace the soybean meal in diets of dairy cows.

O objetivo do estudo foi proceder a uma revisão sistemática da literatura sobre a resposta da pressão arterial após o treino de força em hipertensos, procurando identificar o efeito da fadiga nessa resposta. Foi realizada uma pesquisa bibliográfica nas bases de dados SciELO, LILACS e PubMed/MedLine. Foram encontrados 9377 estudos e apenas seis atenderam aos critérios de inclusão estabelecidos e foram incluídos na revisão. Cinco estudos (83,3%) verificaram uma redução na pressão arterial sistólica ou diastólica. Relativamente à fadiga, 66,7% (n = 4) dos estudos relataram que realizaram séries até a fadiga, ao passo que 33,3% (n = 2) relataram que as séries eram interrompidas antes da fadiga. Dos quatro estudos que utilizaram protocolos até a fadiga, 75% (n = 3) observaram redução da pressão arterial, e os dois estudos que interromperam as séries antes da fadiga também evidenciaram redução. A magnitude da redução da pressão arterial foi similar entre os estudos que utilizaram os dois protocolos. Sendo assim, os resultados indicaram que a redução da pressão arterial após o treino de força em sujeitos hipertensos é independente da ocorrência de fadiga.

The objective of this study was to systematically review the literature on blood pressure response after strength training in hypertensive subjects, trying to identify the effect of failure in this response. We performed a literature search in SciELO, LILACS, PubMed / MedLine databases. Out of 9377 studies, only six met the inclusion criteria and were included in the review. Five studies (83.3%) reported a reduction in systolic or diastolic blood pressure. Regarding failure, 66.7% (n = 4) of the studies performed the sets until failure while 33.3% (n = 2) interrupted exercise before failure. Out of the four studies that used protocols until failure, 75% (n=3) observed a reduction in blood pressure, and the two studies who discontinued before the series also showed a reduction of failure. The magnitude of blood pressure reduction was similar among studies that employed both protocols. Thus, the results indicated that the reduction in blood pressure after strength training in subjects with hypertension is independent of the occurrence of failure.

A quina (Strychnos pseudoquina A. St.-Hil) é espécie medicinal do Cerrado Brasileiro. As sementes apresentam dormência e, por isso, foram submetidas aos tratamentos pré-resfriamento a 5ºC durante 7 dias, pré-aquecimento a 40ºC durante 7 dias, pré-embebição em ácido sulfúrico P.A. durante 5 e 15 minutos, pré-embebição em água fervente durante 5 e 15 minutos, pré-embebição em ácido giberélico a 100 e 200 ppm, durante 48 horas, pré-embebição em água destilada durante 24 e 48 horas e escarificação mecânica, visando à superação da dormência. Foram realizadas contagens diárias entre o 2º dia após a implantação do experimento até a estabilização da germinação, que ocorreu no 65º dia. Foram avaliados o índice de velocidade de germinação (IVG) e a porcentagem de germinação. Taxas de germinação acima de 96% foram atingidas em sementes pré-embebidas em água durante 48 horas e substrato umedecido com água ou KNO3.

"Quina" (Strychnos pseudoquina A. St. Hil) is a medicinal plant species from the Brazilian Cerrado. As its seeds show dormancy, they were subjected to the treatments pre-cooling at 5ºC during 7 days, pre-heating at 40ºC during 7 days, pre-soaking in sulfuric acid PA during 5 and 15 min, pre-soaking in boiling water during 5 and 15 min, pre-soaking in 100 and 200 ppm gibberellic acid during 48 h, pre-soaking in distilled water during 24 and 48 h, and mechanical scarification to break dormancy. Counts were daily conducted from the 2nd day after the experiment implementation until the germination stabilization at the 65th day. The germination speed index (GSI) and the germination percentage were evaluated. Germination rates above 96% were reached in seeds pre-soaked in water during 48 h and substrate moistened with water or KNO3.

OBJETIVOS: Avaliar o efeito do antimoniato de meglumina na transferência materno-fetal na geração F1 (prole de matrizes expostas ao composto), e conseqüências em progênies F2. MÉTODOS: Camundongos fêmeas Swiss foram tratados com antimoniato de meglumina, via subcutânea, com administração diária, do sétimo ao 12º dia de gestação (ddg), na dose equivalente a 100mgSb v/kg peso/dia. O grupo controle recebeu apenas o veículo (água destilada). Após o nascimento da prole (geração F1), 59 fêmeas foram examinadas diariamente para determinação do ciclo estral. Quando determinado o ciclo estro, acasalou-se 18 fêmeas com machos da mesma linhagem. No 18º ddg, as fêmeas foram eutanasiadas por câmara de CO2, o abdômen incisado e o útero exposto, quando avaliou-se os sítios de desenvolvimento embrionário e fetal quanto ao número de reabsorções, fetos vivos e mortos. Todos os fetos e placentas foram pesados para calcular o índice placentário. Três placentas de cada ninhada foram separadas para análise microscópica. RESULTADOS: A exposição ao antimoniato de meglumina não interferiu no ciclo estral dos animais tratados, pelo fato de não alterar o intervalo precoital e o índice de fertilidade. Não foram observadas alterações placentárias em progênies F2. CONCLUSÃO: O antimoniato de meglumina não altera a performance reprodutiva das mães expostas cronicamente. Estes dados sugerem que ocorre uma gradual eliminação do antimoniato de meglumina no organismo materno, sem acarretar danos a proles futuras.

OBJECTIVES: Evaluate the effect of Meglumine Antimoniate on maternal-fetal transference in F1 generations (offspring of dams exposed to the drug), and embryotoxicity in F2 generations. METHODS: Female Swiss mice were treated with daily s.c. injection of Meglumine Antimoniate (100mgSb v/kg bw/day) from day 7 until day 12 of pregnancy. The control group received only the vehicle (distilled water). After birth of offspring (F1 generation), 59 females were examined daily for determination of the estral cycle. When the cycle estrus was determined, males were mated with 18 females of the same lineage. On day 18 of pregnancy, females were euthanasied in a chamber of CO2 and after incision of the abdomen, the uterus was exposed. Then, resorptions as well as living and dead fetuses were evaluated, also the number of embryo/fetal implantation sites. Fetuses and their placenta were weighted to calculate the placental index. Three placentas of each litter were separated for microscopic analysis. RESULTS: Administration of the Meglumine Antimoniate did not interfere in the estral cycle of the treated group, since it did not alter the precoital interval and fertility index. Placenta alterations were not observed in the F2 generations. CONCLUSION: Meglumine Antimoniate did not interfere in the reproductive performance, after chronic exposition of dams. Data suggest that there is a gradual elimination of Meglumine Antimoniate by the maternal organism without damaging the future offspring.

Lactococcus lactis, the model lactic acid bacterium, is a good candidate for heterologous protein production in both foodstuffs and the digestive tract. We attempted to produce Streptomyces tendae antifungal protein 1 (Afp1) in L. lactis with the objective of constructing a strain able to limit fungal growth. Since Afp1 activity requires disulfide bond (DSB) formation and since intracellular redox conditions are reportedly unfavorable for DSB formation in prokaryotes, Afp1 was produced as a secreted form. An inducible expression-secretion system was used to drive Afp1 secretion by L. lactis; Afp1 was fused or not with LEISSTCDA, a synthetic propeptide (LEISS) that has been described to be a secretion enhancer. Production of Afp1 alone was not achieved, but production of LEISS-Afp1 was confirmed by Western blot and immunodetection with anti-Afp1 antibodies. This protein (molecular mass: 9.8 kDa) is the smallest non-bacteriocin heterologous protein ever reported to be secreted in L. lactis via the Sec-dependent pathway. However, no anti-fungal activity was detected, even in concentrated samples of induced supernatant. This could be due to a too low secretion yield of Afp1 in L. lactis, to the absence of DSB formation, or to an improper DSB formation involving the additional cysteine residue included in LEISS propeptide. This raises questions about size limits, conformation problems, and protein secretion yields in L. lactis.

Este trabalho foi realizado com os objetivos de gerar uma coleção de genes relacionados ao metabolismo secundário da cana de açúcar e investigar o padrão de expressão gênica de enzimas chaves reguladoras das principais vias biossintéticas ativas nos diferentes tipos de tecidos e situações de estresse físico-químico e biológico a que estão submetidas plantas cultivadas em casas de vegetação, campo ou in vitro. A estratégia de mineração dos dados da database de sequências expressas de cana de açúcar, SUCEST, usando ferramentas de bioinformática, focalizou classes de compostos como isoprenóides e fenilpropanóides que comprovadamente desempenham um papel na resposta de plantas a variações ambientais. Foram identificados e caracterizados genes que codificam enzimas chaves para a síntese de terpenóides, como a sesquiterpeno ciclase (SC); (CHS) para síntese de flavonóides; isoflavona sintase (IFS) envolvida na biossíntese de isoflavonóides que desempenharm importante papel na defesa de plantas e nodulação de raízes; isoflavona redutases (IFR) enzimas chaves para a síntese de fenilpropanóide fitoalexinas, bem como enzimas relacionadas à síntese de precursores de lignina, como a enzima ácido caféico- O- metiltransferase. O efeito do estresse causado por bactérias como Herbaspirillum rubri e Gluconacetobacter diazotroficans também foi avaliado tendo sido constatada a indução da expressão de chalcona sintase (CHS) em plântulas infectadas com esses agentes, sugerindo a ativação da via de flavonóides em resposta a este estresse biológico. Esses resultados apontam para o fato de que as vias do metabolismo de isopropanóides e de fenilpropanóides em cana de açúcar são ativadas de acordo com o estágio de desenvolvimento, especificidade de tecidos e em resposta a situações de estresse. Essas observações deverão ser confirmadas por meio de experimentação genética e bioquímica.

A set of genes related to secondary metabolism was extracted from the sugarcane expressed sequence tag (SUCEST) database and was used to investigate both the gene expression pattern of key enzymes regulating the main biosynthetic secondary metabolism pathways and the major classes of metabolites involved in the response of sugarcane to environmental and developmental cues. The SUCEST database was constructed with tissues in different physiological conditions which had been collected under varied situation of environmental stress. This database allows researchers to identify and characterize the expressed genes of a wide range of putative enzymes able to catalyze steps in the phenylpropanoid, isoprenoid and other pathways of the special metabolic mechanisms involved in the response of sugarcane to environmental changes. Our results show that sugarcane cDNAs encoded putative ultra-violet induced sesquiterpene cyclases (SC); chalcone synthase (CHS), the first enzyme in the pathway branch for flavonoid biosynthesis; isoflavone synthase (IFS), involved in plant defense and root nodulation; isoflavone reductase (IFR), a key enzyme in phenylpropanoid phytoalexin biosynthesis; and caffeic acid-O-methyltransferase, a key enzyme in the biosynthesis of lignin cell wall precursors. High levels of CHS transcripts from plantlets infected with Herbaspirillum rubri or Gluconacetobacter diazotroficans suggests that agents of biotic stress can elicit flavonoid biosynthesis in sugarcane. From this data we have predicted the profile of isoprenoid and phenylpropanoid metabolism in sugarcane and pointed the branches of secondary metabolism activated during tissue-specific stages of development and the adaptive response of sugarcane to agents of biotic and abiotic stress, although our assignment of enzyme function should be confirmed by careful biochemical and genetic supporting evidence.
