Salivary cytokine profile in patients with oral lichen planus

Resumen Este estudio tuvo como objetivo determinar las concentraciones salivales de interleucina (IL)-1β, IL-2, IL-10, IL-23, factor de crecimiento transformante (TGF)-β y factor de necrosis tumoral (TNF)-α, nitrato (subproducto de la oxidación del óxido nítrico) y cortisol en pacientes con liquen plano oral (OLP). En este estudio transversal se incluyeron veinte pacientes diagnosticados con OLP y 20 voluntarios sanos (HV) del mismo sexo. Saliva entera no estimulada Se recolectó por la mañana, se determinaron las concentraciones de citocinas y cortisol en saliva mediante ensayo inmunoabsorbente ligado a enzimas (ELISA), se determinó nitrato mediante un analizador de óxido nítrico, se encontraron concentraciones salivales mayores de IL-2 (p<0,003), IL- 23 (p<0,04) y TGF-β (p=0,05) en pacientes con OLP en comparación con HV. No se encontraron diferencias significativas en los niveles salivales de TNF-α, IL-1β e IL-10. Las concentraciones de nitrato fueron marcadamente aumentó en pacientes con OLP (1227,0 ± 738,8 µM/mg de proteína total), en comparación con HV (261,6 ± 166,8 µM/mg; p<0,0001). Los niveles de cortisol salival también fueron más altos en los pacientes con OLP que en los controles (2,79 ± 1,39 vs. 1,94 ±1,21 ng/mg; p<0,048). Los niveles de óxido nítrico en saliva aumentaron notablemente en pacientes con OLP, lo que sugiere una relación de esta molécula con la muerte celular y el daño tisular observado en las lesiones de OLP.
Abstract This study aimed to determine salivary concentrations of interleukin (IL)-1β, IL-2, IL-10, IL- 23, transforming growth factor (TGF)-β, tumor necrosis factor (TNF)-α, nitrate (a by-product of nitric oxide oxidation), and cortisol in patients with oral lichen planus (OLP). Twenty patients diagnosed with OLP and 20 sex-matched healthy volunteers (HV) were included in this cross-sectional study. Unstimulated whole saliva was collected in the morning. Salivary cytokine and cortisol concentrations were determined by enzyme-linked immunosorbent assays (ELISA). Nitrate was measured in a nitric oxide analyzer. We found higher salivary concentrations of IL-2 (p<0.003), IL-23 ( p<0.04), and TGF-β (p=0.05) in patients with OLP compared to HV. No significant differences were found in salivary levels of TNF-α, IL-1β, or IL-10. Nitrate concentrations were markedly increased in OLP patients (1,227.0 ± 738.8 µM/mg total protein) when compared to HV (261.6 ± 166.8 µM/mg; p<0.0001). Salivary cortisol levels were also higher in OLP patients (2.79 ± 1.39 vs. 1.94 ±1.21 ng/mg; p<0.048). The markedly increased salivary levels of nitric oxide in patients with OLP suggest a relationship of this molecule with the cell death and tissue damage observed in these lesions.

RESUMO Objetivo Verificar o repertório lexical de crianças falantes do português brasileiro aos 24 e 30 meses e a associação entre a quantidade de palavras faladas e as variáveis: nível socioeconômico, escolaridade dos pais, presença de irmãos no convívio familiar, frequentar ou não escola e uso exacerbado de tablets e celulares pelas crianças. Método 30 pais de crianças com 24 meses, residentes no estado de São Paulo participaram do estudo. Por meio de plataformas de videoconferência eles foram submetidos à anamnese fonoaudiológica, entrevista com o serviço social e preencheram o “Inventário MacArthur de Desenvolvimento Comunicativo - Primeiras Palavras e Gestos”, quando seus filhos tinham 24 e 30 meses. Foi aplicada estatística indutiva inferencial, quantitativa e qualitativa. Resultados A mediana das palavras emitidas foi de 283 aos 24 meses e 401 aos 30 meses, indicando aumento em torno de 118 palavras após seis meses. A criança estar frequentando ambiente escolar apresentou relação significativa com o aumento do vocabulário. Conclusão O estudo reforça o crescimento do vocabulário conforme o avanço da idade e corrobora o fato de as crianças com 24 meses já possuírem um repertório maior que 50 palavras. ABSTRACT Purpose To check the lexical repertoire of Brazilian Portuguese-speaking children at 24 and 30 months of age and the association between the number of words spoken and the following variables: socioeconomic status, parents’ education, presence of siblings in the family, whether or not they attend school, and excessive use of tablets and cell phones. Methods 30 parents of children aged 24 months living in the state of São Paulo participated in the study. Using videoconferencing platforms, they underwent a speech-language pathology anamnesis, an interview with social services, and then they completed the “MacArthur Communicative Development Inventory - First Words and Gestures” as soon as their children were 24 and 30 months old. Quantitative and qualitative inferential inductive statistics were applied. Results the median number of words produced was 283 at 24 months and 401 at 30 months, indicating an increase of around 118 words after six months. The child attending a school environment had a significant relationship with increased vocabulary. Conclusion The study reinforces the fact that vocabulary grows with age and corroborates the fact that children aged 24 months already have a repertoire greater than 50 words. Those who attend school every day produce at least 70 more words than those who do not. Abstract Sjogren’s disease (SjD) is an autoimmune disease that is characterized not only by the sicca symptoms it causes but also by its systemic nature, which is capable of several and not yet fully understood extraglandular manifestations. To gain a clearer understanding of these manifestations as well as a better practical approach, a panel of experts from the Brazilian Society of Rheumatology conducted a systematic review and meta-analysis on the identification of epidemiologic and clinical features of the extraglandular manifestations present in ESSDAI (EULAR Sjogren´s syndrome disease activity index), followed by a voting panel with recommendations for clinical practice. This publication is complementary to others already published and covers cutaneous and hematological manifestations, with prevalence data generated by a meta-analysis of 13 clinical or laboratory manifestations and 6 clinical management recommendations. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. RESUMO Objetivos: verificar a validação de constructo de um instrumento de avaliação da assistência a pessoas vivendo com HIV na Atenção Primária à Saúde. Métodos: estudo metodológico, realizado em 2021 com 260 profissionais de saúde, em Recife, PE. Realizou-se a validação baseada na estrutura interna, nesta etapa, utilizou-se a análise fatorial exploratória e confirmatória, e a validade baseada na teoria de resposta ao item. Resultados: a validação determinou a retenção de cinco fatores e 63 itens. A consistência interna do instrumento e qualidade do ajustamento foi de 0,90, o índice de Tukey-Lewis foi de 0,915 e o índice de ajuste comparativo foi de 0,918 na análise fatorial confirmatória. A indicação para a maioria absoluta dos itens é de ajuste adequado. Abstract Objectives: To identify somatic mutations in tumors from young women with triple-negative or luminal breast cancer, through targeted sequencing and to explore the cancer driver potential of these gene variants. Methods: A customized gene panel was assembled based on data from previous sequencing studies of breast cancer from young women. Triple-negative and luminal tumors and paired blood samples from young breast cancer patients were sequenced, and identified gene variants were searched for their driver potential, in databases and literature. Additionally, the authors performed an exploratory analysis using large, curated databases to evaluate the frequency of somatic mutations in this gene panel in tumors stratified by age groups (every 10 years). Results: A total of 28 young women had their tumoral tissue and blood samples sequenced. Using a customized panel of 64 genes, the authors could detect cancer drivers in 11/12 (91.7 %) TNBC samples and 11/16 (68.7 %) luminal samples. Among TNBC patients, the most frequent cancer driver was TP53, followed by NF1, NOTCH1 and PTPN13. In luminal samples, PIK3CA and GATA3 were the main cancer drivers, and other drivers were GRHL2 and SMURF2. CACNA1E was involved in both TN and luminal BC. The exploratory analysis also indicated a role for SMURF2 in luminal BC development in young patients. Conclusions: The data further indicates that some cancer drivers are more common in a specific breast cancer subtype from young patients, such as TP53 in TNBC and PIK3CA and GATA3 in luminal samples. These results also provide additional evidence that some genes not considered classical cancer-causing genes, such as CACNA1E, GRHL2 and SMURF2 might be cancer drivers in this age group. Resumo O objetivo deste estudo foi detectar formas tripomastigotas de Trypanosoma vivax em esfregaço sanguíneo, DNA de T. vivax e anticorpos anti-T. vivax, em amostras de búfalos, criados na Baixada Maranhense, Brasil. Foram coletadas amostras de sangue de 116 búfalos e 25 espécimes de ectoparasitas. Para o diagnóstico, foi realizada a confecção de esfregaços sanguíneos a fim de observar formas compatíveis com Trypanosoma spp., a sorologia por Ensaio Imunoenzimático Indireto ((iELISA), a Imunocromatografia de fluxo lateral (Imunotest®) e o diagnóstico molecular por meio da Reação em Cadeia da Polimerase (PCR). Não foram observadas formas compatíveis com Trypanosoma spp. nos esfregaços sanguíneos. Dentre as 116 amostras de soro analisadas, 79,31% e 76,72% mostraram-se positivas nos ensaios de ELISA e teste rápido, respectivamente. Uma amostra foi positiva no teste molecular. Foram coletados 25 piolhos da espécie Haematopinus tuberculatus. Quando submetidos a PCR para a detecção do DNA de T. vivax, todos mostraram-se negativos. Os espécimes de piolhos estavam negativos para T. vivax. Não houve diferença estatística significante (p<0,05) em relação à idade e sexo dos animais e a presença de T. vivax na região. Abstract The aim of this study was to detect trypomastigote forms of Trypanosoma vivax, in blood smears, DNA of T. vivax and anti-T. vivax antibodies in samples from buffalos reared in the lowlands of Maranhão, Brazil. Blood samples were collected from 116 buffalos and 25 ectoparasite specimens. Blood smears were produced to diagnose forms compatible with Trypanosoma spp.; the indirect enzyme-linked immunosorbent assay (iELISA) and lateral-flow immunochromatography (Imunotest®) serological tests were used; and the polymerase chain reaction (PCR) was used to make molecular diagnoses. No forms compatible with Trypanosoma spp. were observed in blood smears. Among the 116 serum samples analyzed, 79.31% and 76.72% were positive in the ELISA and rapid tests, respectively. One sample was positive in the molecular test. Twenty-five lice of the species Haematopinus tuberculatus were collected. When subjected to PCR for detection of DNA of T. vivax, all of them were negative. The louse specimens were negative for T. vivax. There were no statistically significant differences (p < 0.05) in the presence of T. vivax in this region, in relation to the animals’ age and sex. It can be concluded that these protozoa are circulating in the buffalo herd of the lowlands of Maranhão displaying crypitc parasitemias. Resumo A alteração no marco legal do saneamento (Lei 14.026, de 15 de julho de 2020) passou a exigir licitação no Brasil, mesmo nos casos de prestação do serviço de saneamento antes delegada pelos municípios às companhias estaduais, mediante contrato de programa. O objetivo foi identificar os atores que se beneficiaram com a mudança na lei e observar a efetividade social do processo de desestatização, que ocorreu em abril de 2021 com o leilão dos serviços de abastecimento de água e esgotamento sanitário prestados pela Companhia Estadual de Águas e Esgotos do Rio de Janeiro (CEDAE) em 35 municípios do estado. Adotou-se a análise exploratória de dados secundários e a metodologia da integração sistêmica, que possibilita identificar o papel e a atuação de atores sociais na concessão dos serviços operados pela CEDAE no Rio de Janeiro. Concluiu-se que os governos federal, estadual e municipais, o Banco Nacional de Desenvolvimento Econômico e Social e grupos privados ganharam com as concessões, mas quanto à efetividade social dos serviços, observou-se que é menor a proporção de população de baixa renda com redes de afastamento do esgoto sanitário nos municípios do estado operados pela iniciativa privada e que as tarifas praticadas são em média maiores ao comparar com os municípios que participaram do leilão. Abstract Changes in the regulatory and legal framework for Brazil’s water and sanitation sector (Law 14,026, July 15, 2020) require competitive bidding for service contracts, even in cases where the provision of services was previously delegated to state-owned utilities under program contracts. The aim of this study was to identify the actors who benefited from these changes to the legislation and assess the social effectiveness of the privatization of water supply and sewerage services in the state of Rio de Janeiro by investigating the auction of services provided by the public utility CEDAE in four blocks comprising 35 municipalities in April 2021. We conducted an exploratory analysis of secondary data and used the systemic integration method to identify the main actors involved in the concession process and the role they played. It is concluded that federal, state, and municipal governments, the Brazilian Development Bank, and the concessionaires have gained from the concessions, while the percentage of low-income populations connected to the sewage collection or drainage network is lower and tariffs are higher in municipalities served by the private sector when compared to the municipalities encompassed by the auction. ABSTRACT Background: Malaria is a parasitosis conditioned by several factors. This study sought to analyze the spatial distribution of malaria considering environmental, socioeconomic, and political variables in São Félix do Xingu, Pará, Brazil, from 2014 to 2020. Methods: Epidemiological, cartographic, and environmental data were obtained from the Ministry of Health, Brazilian Geographical and Statistical Institute, and National Space Research Institute. Statistical and spatial distribution analyses were performed using chi-squared tests of expected equal proportions and the kernel and bivariate global Moran’s techniques with Bioestat 5.0 and ArcGIS 10.5.1. Results: The highest percentage of cases occurred in adult males with brown skin color, mainly placer miners, with a primary education level, living in rural areas, who were infected with Plasmodium vivax and with parasitemia of two or three crosses as diagnosed by the thick drop/smear test. The disease had a non-homogeneous distribution, with distinct annual parasite indices associated with administrative districts and clusters of cases in locations with deforestation, mining, and pastures close to Conservation Units and Indigenous Lands. Thus, a direct relationship between areas with cases and environmental degradation associated with land use was demonstrated, along with the precarious availability of health services. Pressure on protected areas and epidemiological silence in Indigenous Lands were also noted. Conclusions: Environmental and socioeconomic circuits were identified for development of diseases associated with precarious health services in the municipality. These findings highlight the need to intensify malaria surveillance and contribute to the systematic knowledge of malaria’s epidemiology by considering the complexity of its conditioning factors. RESUMO Objetivo: descrever a percepção de pais sobre o uso de telas de mão por crianças e elaborar cartilha para o uso saudável desses dispositivos. Métodos: 102 pais de crianças entre 18 e 71 meses responderam um questionário sobre uso de telas. A análise foi descritiva e não paramétrica. Após a apreciação das respostas dos cuidadores e revisão da literatura, a cartilha foi construída. O material foi disponibilizado a três juízes para análise e sugestões. Resultados: o uso de telas portáteis pelas crianças foi confirmado pela maioria dos respondentes, e uma parcela das crianças permanece por duas horas ou mais na frente delas. Os pais apontaram as telas como fonte de estímulos para aprender a falar, mas se preocupam com a reprodução de comportamentos inapropriados dos personagens e consumismo. Os juízes responderam ao questionário, atribuindo pontuação máxima à maioria das perguntas. O texto da cartilha reuniu informações sobre como crianças aprendem a falar, esclareceu sobre tempo, conteúdo do uso de telas e apresentou estratégias de estimulação de linguagem. Conclusão: as respostas dos pais ao questionário, a revisão de estudos sobre o tema, somadas às análises dos juízes, forneceram elementos para a elaboração de cartilha sobre o uso adequado desses dispositivos. ABSTRACT Purpose: to describe the perception of parents about the use of handheld screens by children and to prepare a booklet for the healthy use of these devices. Methods: 102 parents of children aged between 18 and 71 months answered a questionnaire on the use of screen. The analysis was a descriptive and non-parametric one. After assessing parents’ responses and reviewing the literature, the booklet was prepared. The material was made available to three judges for analysis and suggestions. Results: the use of portable screens by children was confirmed by most respondents, and some children spend two hours or more in front of them. Parents believe that screens are a source of stimuli for learning how to speak, but they are concerned about the reproduction of some characters’ inappropriate behavior and consumerism. The judges answered the questionnaire assigning maximum scores to most questions. The text of the booklet gathered information about how children learn to speak, clarified about time and content of screen use, and presented language stimulation strategies. Conclusion: parents' responses to the questionnaire, together with a review of studies on the subject and the judges' analyses, provided elements for the preparation of a booklet on the proper use of these devices. The Amazon suffers from the occurrence of mining along its hydrographic routes, which releases toxic substances into the aquatic environment. The objective of this study was to evaluate the risk of contamination by As and Hg that indigenous peoples are exposed to through the consumption of fish in an area impacted by mining activity in the basin of the Curuá and Baú Rivers, Altamira-PA. In total, 55 fish specimens and 30 surface water samples were collected. The decomposition of the fish samples was done by wet method with the use of nitric acid, hydrogen peroxide, and microwaves. The quantification of total mercury (THg) and total arsenic (TAs) was carried out by inductively coupled plasma optical emission spectrometry with vapor generation (VG-ICP OES). The methodology proposed by the U.S. Environmental Protection Agency was used. The results showed that TAs presents safe levels; however, THg presents a sample with levels up to (4.6 µg g 1), exceeding the permitted levels (1 µg g 1). The risk assessment shows that only the consumption of fish, linked to Hg, represents the existence of deleterious risks to indigenous peoples (hazard quotient (HQ)) > 1). Bioaccumulation factor (BAF) in fish represents a risk to indigenous people, because of the Hg’s presence. The study identified that illegal mining activities are responsible for Hg pollution in the Baú and Curuá Rivers. Abstract Changes in the regulatory and legal framework for Brazil’s water and sanitation sector (Law 14,026, July 15, 2020) require competitive bidding for service contracts, even in cases where the provision of services was previously delegated to state-owned utilities under program contracts. The aim of this study was to identify the actors who benefited from these changes to the legislation and assess the social effectiveness of the privatization of water supply and sewerage services in the state of Rio de Janeiro by investigating the auction of services provided by the public utility CEDAE in four blocks comprising 35 municipalities in April 2021. We conducted an exploratory analysis of secondary data and used the systemic integration method to identify the main actors involved in the concession process and the role they played. It is concluded that federal, state, and municipal governments, the Brazilian Development Bank, and the concessionaires have gained from the concessions, while the percentage of low-income populations connected to the sewage collection or drainage network is lower and tariffs are higher in municipalities served by the private sector when compared to the municipalities encompassed by the auction.
Resumo A alteração no marco legal do saneamento (Lei 14.026, de 15 de julho de 2020) passou a exigir licitação no Brasil, mesmo nos casos de prestação do serviço de saneamento antes delegada pelos municípios às companhias estaduais, mediante contrato de programa. O objetivo foi identificar os atores que se beneficiaram com a mudança na lei e observar a efetividade social do processo de desestatização, que ocorreu em abril de 2021 com o leilão dos serviços de abastecimento de água e esgotamento sanitário prestados pela Companhia Estadual de Águas e Esgotos do Rio de Janeiro (CEDAE) em 35 municípios do estado. Adotou-se a análise exploratória de dados secundários e a metodologia da integração sistêmica, que possibilita identificar o papel e a atuação de atores sociais na concessão dos serviços operados pela CEDAE no Rio de Janeiro. Concluiu-se que os governos federal, estadual e municipais, o Banco Nacional de Desenvolvimento Econômico e Social e grupos privados ganharam com as concessões, mas quanto à efetividade social dos serviços, observou-se que é menor a proporção de população de baixa renda com redes de afastamento do esgoto sanitário nos municípios do estado operados pela iniciativa privada e que as tarifas praticadas são em média maiores ao comparar com os municípios que participaram do leilão.

Résumé Cet article résulte d’une recherche sur la place des cas d’enseignement comme outils de médiation dans la formation enseignante. L’étude a été menée auprès de quatre professeurs débutants au cours de leur première année de travail à l’école primaire, dans une commune de l’intérieur du Pernambuco, Brésil. La possibilité d’utiliser des cas d’enseignement comme stratégies de médiation formative pour le développement professionnel des enseignants débutants y a été étudiée. Les données ont été collectées dans le cadre de cas d’enseignement et par le biais de groupes de discussion, au sein d’un réseau collectif de réflexion sur les défis et les besoins en matière de formation. Abstract This article results from research into teaching cases as mediation in teacher training. The study was conducted with four novice teachers in their first year of work in the early years of elementary school, in a municipality in the countryside of the state of Pernambuco, Brazil. We investigated the possibility of using teaching cases as training strategies to mediate the professional development of novice teachers. Data was collected using teaching cases and discussion groups, in a collective network of reflections on training challenges and needs. Resumen Este artículo es el resultado de una investigación sobre casos de enseñanza como mediación en la formación de profesores. The use of hand screens by children is increasingly precocious and routine. Objective: to analyze the perception of parents about the use of hand screens by children and to elaborate a booklet about the beneficial use of screens. Method: parents/guardians of children aged between 18 and 71 months were invited to answer a questionnaire on the use of hand screens. The collected data were analyzed by descriptive and percentage statistics. The parents' answers, the literature review on language development and the use of hand screens, the illustration by a designer and the analysis of three reviewers served as the basis for the construction of the booklet. The results analyzed on 102 responses showed that the use of handheld screens by children was 85.3%, with a time of use of up to 1 hour per day (64.3%) before the pandemic, with an increase of 1 more hour, after its start. The understanding of communication development is restricted to the use of speech (62.7%) for most parents. The booklet addressed language acquisition milestones and clarified about the time, content of the use of hand screens, in addition to presenting stimulation strategies. Conclusion: the use of screens by preschool children, regardless of the pandemic, was already frequent. Parents do not relate language delay and hand screens. The booklet brought clarifications and strategies on how to make them allies in language development.
O uso de telas de mão por crianças está cada vez mais precoce e rotineiro. Objetivo: analisar a percepção de pais sobre o uso das telas das crianças e elaborar a cartilha a respeito do uso do perfil das telas. Método: pais/responsáveisde crianças entre 18 e 71 meses foram convidados a responder sobre uso de telas de mão. Os dados informados foram analisados e analisados por análise de dados. Como a revisão da literatura sobre os pais e a ilustração e a análise de telas de mão, a ilustração e a análise de criação fizeram o desenvolvimento da linguagem de base para a construção de cartilha.Os resultados antes da tela sobre as crianças 102 apontaram que o uso de respostas à mão 85, por 3%, com tempo de uso 1 hora da pandemia (64,3%), com aumento de 1 hora, após o início. Abstract Sjogren's syndrome (SS) is an autoimmune disease characterized by lymphocytic infiltration of the exocrine glands and other organs, associated with sicca syndrome but also with systemic involvement with varying degrees of severity. Despite their importance, some systemic manifestations, mainly liver, gastrointestinal, and pancreatic are not routinely evaluated. To address these manifestations, the Sjögren's Syndrome Committee of the Brazilian Society of Rheumatology conducted a broad systematic review of the literature on studies investigating prevalence and diagnosis of these symptoms in Sjogren´s patients and made recommendations based on the findings. Agreement between the experts was achieved using the Delphi method. This is the second part of this guideline, providing 6 recommendations for liver, gastrointestinal, and pancreatic care of SS patients.
