O objetivo deste estudo foi verificar a eficiência e aplicabilidade de sensores de umidade relativa do ar intergranular atrelados a sensores digitais de temperatura no sistema de termometria de silos armazenando grãos de soja. ABSTRACT Grains are hygroscopic materials and, during storage, they are subject to exchanging moisture with the environment according to temperature and relative air humidity, making it important to monitor these factors. Resumo Fundamento: A Cardiomiopatia Chagásica Crônica (CCC) é causada por um processo inflamatório induzido pelo Trypanosoma cruzi, que leva à miocardite com fibrose reativa e reparativa. A CCC progride com alterações de perfusão miocárdica e eventos histopatológicos que afetam a Aptidão Cardiorrespiratória (ACR). Objetivos: Avaliamos os efeitos do Treinamento Físico Aeróbico (TFA) na perfusão miocárdica e nos comprometimentos morfológicos e funcionais relacionados à inflamação e fibrose em hamsters sírios com CCC. Como objetivo secundário, analisamos as áreas de secção transversa do músculo esquelético. Métodos: Hamsters com CCC e seus respectivos controles foram divididos em quatro grupos: CCC sedentário, CCC-TFA, controle sedentário e controle TFA. Sete meses após a infecção, os animais foram submetidos à ecocardiografia, à cintilografia de perfusão miocárdica e ao teste de esforço cardiopulmonar. TFA de intensidade moderada foi realizado durante cinquenta minutos, cinco vezes por semana, por oito semanas. Posteriormente, os animais foram reavaliados. A análise histopatológica foi realizada após os procedimentos acima mencionados. O nível de significância foi estabelecido em 5% em todas as análises (p<0,05). Resultados: Animais com CCC sedentários apresentaram piores Defeitos de Perfusão Miocárdica (DPM) ao longo do tempo, Fração de Ejeção do Ventrículo Esquerdo (FEVE) reduzida, e apresentaram mais inflamação e fibrose quando comparados aos demais grupos (análise ANOVA mista). Por outro lado, o TFA foi capaz de mitigar a progressão do DPM, atenuar a inflamação e a fibrose e melhorar a eficiência da ACR em animais CCC-TFA. Abstract Background: Chronic Chagas cardiomyopathy (CCC) is caused by an inflammatory process induced by Trypanosoma cruzi, which leads to myocarditis with reactive and reparative fibrosis. CCC progresses with myocardial perfusion abnormalities and histopathological events that affect cardiorespiratory fitness (CRF). Objectives: We evaluated the effects of aerobic physical training (APT) on myocardial perfusion and on morphological and functional impairments related with inflammation and fibrosis in Syrian hamsters with CCC. As a secondary objective, we analyzed the cross-sectional areas of the skeletal muscle. Methods: Hamsters with CCC and their respective controls were divided into four groups: CCC sedentary, CCC-APT, sedentary control and APT control. Seven months after infection, the animals underwent echocardiography, myocardial perfusion scintigraphy and cardiopulmonary exercise testing. Moderate-intensity APT was performed for fifty minutes, five times a week, for eight weeks. Subsequently, the animals were reassessed. Histopathological analysis was conducted after the above-mentioned procedures. The level of significance was set at 5% in all analyses (p<0.05). Results: CCC sedentary animals presented worse myocardial perfusion defects (MPD) over time, reduced left ventricle ejection fraction (LVEF) and showed more inflammation and fibrosis when compared to other groups (mixed ANOVA analysis). Conversely, APT was able to mitigate the progression of MPD, ameliorate inflammation and fibrosis and improve CRF efficiency in CCC-APT animals. Conclusions: Our study demonstrated that APT ameliorated cardiac dysfunction, MPD, and reduced inflammation and fibrosis in CCC hamster models. Additionally, CCC-SED animals presented skeletal muscle atrophy while CCC-APT animals showed preserved skeletal muscle CSA. RESUMO Neste estudo objetivou-se descrever o estágio de fluxo de secagem constante, bem como obter o teor de água crítico durante a secagem convectiva da pasta de folhas de jambu trituradas em diferentes condições e caracterizar as propriedades físicas do material após a secagem. A secagem foi realizada em estufa de circulação de ar com velocidade de 1,0 m s-1 nas temperaturas de 60, 70 e 80 ºC e espessuras da camada de 0,005 e 0,01 m. Para a determinação do estágio de taxa constante, foram calculados os fluxos de massa e a taxa de secagem. Os polinômios segmentados foram utilizados para estimar o teor de água crítico. Nas condições analisadas houve um comportamento típico de secagem, demonstrando a existência de um expressivo período de taxa constante de secagem para a pasta de folhas de jambu trituradas. ABSTRACT The objective of this study was to describe the constant drying flux stage, as well as obtaining the critical moisture content during the convective drying of jambu leaf paste crushed under different conditions and characterizing the physical properties of the material after drying. Drying was carried out in an air circulation oven at a speed of 1.0 m s-1 and temperatures of 60, 70 and 80 ºC and layer thicknesses of 0.005 and 0.01 m. To determine the constant rate stage, the mass fluxes and the drying rate were calculated. Segmented polynomials were used to estimate the critical moisture content. The growth of the oil and gas industry implies an understanding of corrosion and ways to mitigate pipeline failures. The objective of this research is to evaluate the propagation of localized corrosion induced by flow in different concentrations of sodium chloride and in the presence of corrosion inhibitor, associated with the wall shear stress. ABSTRACT This study aimed to obtain the sorption isotherms of grain sorghum flour, fit mathematical models to experimental data, and recommend safe water levels for preserving the material at different temperatures. Sorghum grains with an initial moisture content of 34% dry basis (db) were subjected to drying at a temperature of 60 °C, reaching moisture contents ranging from 32.20 to 9.8% db. Water activity was obtained using Hygropalm Aw1 equipment placed inside a BOD chamber at temperatures of 10, 20, 30, and 40 °C. The experimental data were fitted to mathematical models frequently used to predict the isotherms of plant products. The modified Oswin model presented the best fit to estimate the sorption isotherms of sorghum flour. ABSTRACT The objective of this study was to evaluate the L-arginine supplemental effects for broilers in the final rearing phase (29 and 42 days), during which they were exposed to heat stress and fed diets with different metabolizable energy levels. Performance, carcass parameters, and physiological and biochemical parameters related to lipid metabolism were evaluated. A completely randomized design was used, in a 3 × 2 factorial arrangement, corresponding to three metabolizable energy levels (3000, 3150, and 3300 kcal/kg) in diets supplemented or not with L-arginine (0.66%). Eight replicates composed by eight broilers each (experimental units; boxes) per treatment were evaluated, totaling 48 experimental units. Performance was evaluated in the period from 29 to 42 days old, and carcass and blood parameters were measured at 42 days old. L-arginine supplementation improved feed conversion and blood profile, mainly due to the reduction of total and LDL-cholesterol levels. Broilers that received 3300 kcal/kg of metabolizable energy showed the greatest weight gain, regardless of supplementation or not with arginine. ABSTRACT Phosphites can be considered an additional strategy to be included in disease management programs. Therefore, this study aims to evaluate the application of potassium phosphite (KPhi) in common bean seeds, in the reduction of intensity of root rot (Rhizoctonia solani) and in the compatibility with Rhizobium tropici, a nitrogen-fixing bacterium. The emergence speed index (ESI) and the percentage of total emergence were evaluated in seeds inoculated with R. solani and treated with KPhi. The number of nodules per root system, dry nodule mass, quantified the compatibility Phi x R. tropici, dry mass, nitrogen content, and nitrogen accumulation in the shoots. KPhi promoted a higher ESI and increase in plant stand compared to the control treatment. Abstract Background Controlling blood pressure and glycemic levels is a challenge that requires innovative solutions. Objective To assess the feasibility of implementing a text message intervention among low-income primary care patients, as well as to assess self-reported behavioral change. Methods A set of 200 text messages was developed on healthy eating, physical activity, adherence, and motivation. Participants from Vale do Mucuri, MG, Brazil diagnosed with diabetes or hypertension or undergoing screening for those diseases, received 5 to 8 messages per week for 6 months. They answered a questionnaire to report their satisfaction and behavioral changes. Results Of the 136 patients, 117 (86.0%) answered the questionnaire. Most reported that the messages were very useful (86.3%), easy to understand (90.6%), and were very helpful for behavioral change (65.0%); 84.6% reported that they had started eating healthier. The most frequent reported lifestyle changes were: improved diet quality (85.5%), reduced portions (65.8%), and weight loss (56.4%). The majority of patients shared the messages (60.7%) with family or other acquaintances, considered the number of messages to be adequate (89.7%) and would recommend the program to others (95.7%). Conclusion An intervention based on text messages to promote behavioral change in patients with hypertension or diabetes in primary care is feasible in low-resource settings. ABSTRACT Sorghum ( Sorghum bicolor L. Moench) is a crop of paramount importance for Brazil, used in human and animal food. Considering the reduced number of studies on the influence of using a densimetric table in seed processing and its respective reprocessing to the equipment, this study aimed to evaluate the physiological quality of processed sorghum seeds reprocessed in a densimetric table and stored for three months. The treatments consisted of four collection points in the terminal part of the densimetric table, representing the processing material: upper part (P1), upper middle part (P2), middle part (P3), and lower part (D). Subsequently, the samples that were reprocessed in the table came from P3 and were divided into the upper part, which was followed to bagging (P3R1), and the lower part (P3R2), which followed for disposal, totaling six treatments. Storage was performed in a refrigerated environment, using a completely randomized design, with the means being compared by Tukey’s test at 5%. We concluded that the reprocessing of sorghum seeds in the densimetric table can improve the physical quality of a lot due to its standardization at the end of the stages, contributing to the improvement of the physiological quality and use of the material that would be discarded. Abstract Background Management delays imply worse outcomes in rheumatoid arthritis (RA) and, therefore, should be minimized. We evaluated changes in diagnostic and treatment delays regarding RA in the last decades in Brazil. Methods Adults fulfilling the ACR/EULAR (2010) criteria for RA were assessed. Delays in diagnosis and treatment, and the frequencies of early management initiation within thresholds (windows of opportunity) of 3, 6, and 12 months from symptoms onset were evaluated. The Mann–Kendall trend test, chi-squared tests with Cramer’s V effect sizes and analysis of variance were conducted. Results We included 1116 patients: 89.4% female, 56.8% white, mean (SD) age 57.1 (11.5) years. A downward trend was found in diagnostic (tau = - 0.677, p < 0.001) and treatment (tau = - 0.695, p < 0.001) delays from 1990 to 2015. The frequency of early management increased throughout the period, with ascending effect sizes across the 3-, 6-, and 12-month windows (V = 0.120, 0.200 and 0.261, respectively). Despite all improvements, even in recent years (2011–2015) the diagnostic and treatment delays still remained unacceptably high [median (IQR): 8 (4–12) and 11 (5–17) months, respectively], with only 17.2% of the patients treated within the shortest, 3-month window. Conclusion The delays in diagnosis and treatment of RA decreased during the last decades in Brazil. Improvements (effect sizes) were greater at eliminating extreme delays (≥ 12 months) than in attaining really short management windows (≤ 3 months). RESUMO A modelagem da distribuição diamétrica em florestas naturais é uma ferramenta importante para compreender a dinâmica de espécies nativas lenhosas, auxiliando na tomada de decisão para o manejo e a recuperação de áreas degradadas. Dessa forma, o objetivo deste trabalho foi ajustar funções de densidade probabilística para avaliar a estrutura diamétrica de três espécies arbóreas de interesse econômico, como Campomansesia xanthocarpa Marl. Ex. O. Berg, Piptadenia gonoacantha (Mart.) J.F. Macbr. e Zeyheria tuberculosa (Vell.) Bureau ex Verl., em um fragmento de Floresta Estacional Semidecidual no estado de São Paulo, Brasil. Os dados foram provenientes de 83 parcelas temporárias de 10 m x 20 m distribuídas sistematicamente ao longo de 164 ha. As funções Log-normal, Gamma e Weibull foram ajustadas para as três espécies por meio do pacote fitdistrplus no programa R. O teste de aderência de Kolmogorov-Smirnov foi aplicado a 5% de probabilidade para avaliar os ajustes. As funções foram .selecionadas por meio do Critério de Informação Akaike (AIC) e do Critério de Informação Bayesiano de Schwaarz (BIC), além da análise gráfica das funções ajustadas. Log-normal, Gamma, and Weibull functions were fitted to the three species using the fitdistrplus-package in the R program. Kolmogorov-Smirnov's adherence test was used to evaluate the fits at a 5% probability level. The functions were selected employing Akaike's Information Criterion (AIC) and Schwarz's Bayesian Information Criterion (BIC), in addition to a graphical analysis of the fitted functions. The results indicated that the three species diameter structure is positively asymmetric, representing the exponential pattern, representing continuous natural regeneration. AIC and BIC statistics indicated the Log-normal function to describe the diameter distribution of C. xanthocarpa and Z. tuberculosa, while the Gamma function was the most appropriate for P. gonoacantha. Resumo O reparo por excisão de nucleotídeos (NER) atua reparando danos no DNA, como lesões causadas por cisplatina. A proteína Xeroderma Pigmentosum complementation group C (XPC) está envolvida no reconhecimento de danos pela via de reparação global do genoma pelo NER (GG-NER) e tem sido estudada em diferentes organismos devido à sua importância em outros processos celulares. Neste trabalho, estudamos proteínas do NER em Trypanosoma cruzi e Trypanosoma evansi, parasitos de humanos e animais, respectivamente. Modelos tridimensionais das proteínas XPC de T. cruzi e T. evansi foram feitos e observou-se poucas diferenças estruturais entre estas proteínas. Durante testes, a inserção do gene XPC de T. evansi (TevXPC) em T. cruzi resultou em crescimento celular mais lento em condições normais. Após o tratamento com cisplatina, T. cruzi superexpressando seu próprio gene XPC (TcXPC) foi capaz de recuperar as taxas de divisão celular mais rapidamente do que T. cruzi expressando o gene TevXPC. Com base nesses testes, sugere-se que TevXPC (sendo uma proteína exógena em T. cruzi) interfere negativamente nos processos celulares em que TcXPC (a proteína endógena) está envolvida. Isso provavelmente ocorreu pois TevXPC é capaz de interagir com algumas moléculas ou proteínas endógenas de T.cruzi, mas é incapaz de interagir com outras. Isso reforça a importância do correto funcionamento de XPC dentro da célula.

Abstract Nucleotide excision repair (NER) acts repairing damages in DNA, such as lesions caused by cisplatin. Xeroderma Pigmentosum complementation group C (XPC) protein is involved in recognition of global genome DNA damages during NER (GG-NER) and it has been studied in different organisms due to its importance in other cellular processes. In this work, we studied NER proteins in Trypanosoma cruzi and Trypanosoma evansi, parasites of humans and animals respectively. We performed three-dimensional models of XPC proteins from T. cruzi and T. evansi and observed few structural differences between these proteins. In our tests, insertion of XPC gene from T. evansi (TevXPC) in T. cruzi resulted in slower cell growth under normal conditions. After cisplatin treatment, T. cruzi overexpressing its own XPC gene (TcXPC) was able to recover cell division rates faster than T. cruzi expressing TevXPC gene. Based on these tests, it is suggested that TevXPC (being an exogenous protein in T. cruzi) interferes negatively in cellular processes where TcXPC (the endogenous protein) is involved. This probably occurred due interaction of TevXPC with some endogenous molecules or proteins from T.cruzi but incapacity of interaction with others. This reinforces the importance of correctly XPC functioning within the cell.

Chikungunya virus (CHIKV) is a re-emerging disease caused by an alphavirus of the Togaviridae family. Since its first description in 1952, the disease has spread worldwide, affecting populations in both tropical and temperate countries. To date, there is no licensed vaccine or specific pharmacological treatment. Therefore, there is an increasing urgency in developing new antiviral drugs capable of specifically inhibiting viral replication. In the present work, we report the synthesis and antiviral activity evaluation of nineteen naphthoquinone derivatives, containing a sulfonamide or sulfonate group. Cell viability assays indicated a low toxic potential for all tested compounds and inhibitory assays against CHIKV identified five compounds with potent activity. The compounds were also evaluated for their virucidal potential, and the results demonstrated that compound 11a exhibited a virucidal effect higher than 70% in the treatment with 20 µM. Furthermore, in silico studies were performed to predict the antiviral drug targets.

RESUMO Neste trabalho propomos e analisamos medidas de relaxamento do distanciamento social, e seus impactos no sentido epidemiológico, a partir das simulações de um modelo matemático compartimental do tipo SEIRD (Suscetível-Exposto-Infectado-Recuperado-Morto) generalizado, que implicitamente leva em conta medidas de quarentena, considerando a estimativa da dinâmica de espalhamento da doença do coronavírus (COVID-19) sob perturbações/incertezas. O objetivo principal é avaliar os efeitos nas projeções da epidemia da COVID-19 no Brasil e, em particular, no estado do Rio de Janeiro. Diferentes estratégias de relaxamento das medidas de distanciamento social são investigadas para determinar quais delas são viáveis e menos perigosas para a população. Os resultados destacam a necessidade de manter políticas de distanciamento social para controlar a propagação da doença. Especificamente, o cenário de relaxamento abrupto do distanciamento social, implementado após a ocorrência do pico de casos diagnosticados positivamente, pode prolongar a epidemia, com um aumento significativo do número previsto de casos confirmados e de mortes. Um cenário ainda pior pode ocorrer se a política de relaxamento do distanciamento social é implementada antes da evidência do controle epidêmico, indicando a importância da escolha adequada de quando parar e começar a relaxar as medidas de distanciamento social. A abordagem empregada aqui, e as análises subsequentes aplicadas no Brasil e Rio de Janeiro, pode ser usada para outros locais.

ABSTRACT In this work, we propose and analyze social distancing relaxation measures and their epidemiological impacts through simulations of a compartmental mathematical model. Our generalized SEIRD model implicitly takes into account social distancing measures, considering the estimate of the spreading dynamics of COVID-19 under disturbances/uncertainties. The main objective is to assess the effects on the projections of the COVID-19 epidemic in Brazil and, in particular, in the state of Rio de Janeiro. Different relaxation strategies of social distancing measures are investigated to determine which ones are viable and less dangerous for the population. The results highlight the need to maintain social distancing policies to control the spread of the disease. Specifically, the scenario considered as abrupt relaxation measures, implemented after the peak of positively diagnosed cases has occurred, can prolong the epidemic, with a significant increase in the expected number of confirmed and death cases. An even worse scenario can occur if the relaxation policy is implemented prior to the evidence of epidemiological control, indicating the importance of the proper choice of when to stop and start to relax social distancing measures. The approach employed here, and the subsequent analyzes applied to Brazil and Rio de Janeiro, can be used for other locations.

Abstract Background Species of Amaranthus genus are common in agricultural areas of Brazil. Such weeds are problematic, and they bring complexity to the management mainly due to herbicide resistance. Thus, the monitoring and mapping of chlorimuron-resistant Amaranthus spp. is necessary to detect resistance in different locations of Brazil. Objective Elaborate discriminating dose of distinction between Amaranthus spp. populations that are susceptible and resistant to chlorimuron and monitor the resistance dispersal throughout five crops. Methods 33 pigweed populations from the main grain producing properties in Brazil by means of dose-response curves were evaluated. For the D dose, chlorimuron dose of 20 g ha-1 ai was considered. Once the discriminating dose was identified, a monitoring screening of the dispersal of resistance of Amaranthus spp. to chlorimuron was conducted with 226 samples between the 2016 and 2020 crops. Results The discriminating dose (“base line”) considered ideal to control susceptible plants was 20 g ha-1 of chlorimuron. Among 226 pigweed samples evaluated in the five years of monitoring, 74% of populations were considered susceptible (S), while those classified as resistant (R) and segregating (r) did not exceed 26.0%. Conclusions By comparing susceptible biotypes of Amaranthus spp. with international scientific literature standards and leaflet averages, it could be safely concluded that the discriminating dose of chlorimuron is 20 g ha-1. Resistance of Amaranthus spp. to chlorimuron in Brazil is present in the main soybean producing regions evaluated, with a frequency of 26% of the total samples evaluated.
