This study aimed to isolate, purify and characterize secondary metabolites present in Amburana acreana leaves, native to the state of Rondônia, using classical chromatographic methods and 1D, 2D nuclear magnetic resonance spectroscopy. From the hydroethanolic leaves extract of A. acreana, p-hydroxybenzoic acid, vanillic acid, coumarin, campesterol 3-β-D-glucoside and amburoside B, all known compounds, were identified. To the best of our knowledge, this is the first report of isolation and chemical characterization of secondary metabolites in A. acreana. In addition, in vitro assays indicated that amburoside B does not have inhibitory potential on the growth of gram-negative bacteria. ABSTRACT Purpose: Smoking is a recognized risk factor for bladder BC and lung cancer LC. We investigated the enduring risk of BC after smoking cessation using U.S. national survey data. Our analysis focused on comparing characteristics of LC and BC patients, emphasizing smoking status and the latency period from smoking cessation to cancer diagnosis in former smokers. Materials and Methods: We analyzed data from the National Health and Examination Survey (2003-2016), identifying adults with LC or BC history. Smoking status (never, active, former) and the interval between quitting smoking and cancer diagnosis for former smokers were assessed. We reported descriptive statistics using frequencies and percentages for categorical variables and median with interquartile ranges (IQR) for continuous variables. Results: Among LC patients, 8.9% never smoked, 18.9% active smokers, and 72.2% former smokers. Former smokers had a median interval of 8 years (IQR 2-12) between quitting and LC diagnosis, with 88.3% quitting within 0-19 years before diagnosis. For BC patients, 26.8% never smoked, 22.4% were active smokers, and 50.8% former smokers. Former smokers had a median interval of 21 years (IQR 14-33) between quitting and BC diagnosis, with 49.3% quitting within 0-19 years before diagnosis. Conclusions: BC patients exhibit a prolonged latency period between smoking cessation and cancer diagnosis compared to LC patients. Despite smoking status evaluation in microhematuria, current risk stratification models for urothelial cancer do not incorporate it. Our findings emphasize the significance of long-term post-smoking cessation surveillance and advocate for integrating smoking history into future risk stratification guidelines. Abstract Objective: To examine if the substitution of different screen time intervals with light physical activity (LPA), moderate to vigorous physical activity (MVPA) and sleep is associated with cardiovascular indicators and inflammatory markers in children. Methods: This is a cross-sectional study developed with 186 children aged between six and 11 years old from public schools in southern Brazil. CRF was measured with the 6-minute running and walking test, following the Brazil Sports Project procedures. The percentage of fat was evaluated through DXA. LPA and MVPA were measured using accelerometers. Sleep and screen time were assessed by questionnaires answered by parents. Leptin and C-reactive protein were measured by fasting blood collection. Systolic and diastolic blood pressure were determined through a digital sphygmomanometer. Isotemporal substitution models were used for statistical analysis. Results: Replacing 1 h of screen time with MVPA was associated with lower BMI, systolic and diastolic blood pressure, fat percentage, leptin, and C-reactive protein. When screen time was substituted for sleep time, lower waist circumference was observed. Regarding the substitution of 1 h of screen time with LPA, significant values were found only for leptin. Conclusion: The replacement of screen time with physical activities of different intensities and sleep time was associated with benefits in cardiovascular indicators and inflammatory markers in childhood. Abstract Robotic milking systems are successful innovations in the development of dairy cattle. The objective of this study was to analyse the milking characteristics and behavior of dairy cows of different calving orders in “milk first” robotic milking systems. The data were collected from a commercial herd located in the Midwest region of Minas Gerais (Brazil), which uses an automatic milking system (AMS TM, DeLaval). Were analysed 26,574 observations of 235 Holstein cows were available. Data were evaluated by multivariate analysis of variance and the Tukey test. - Tthe characteristics milk flow and milking efficiency were more favourable for multiparous cows (p <0.01), while the time in the stall was more favourable for primiparous females (p <0.01). The values of handling time were better in the primiparous cows (p <0.01). Primiparous cows had higher amounts of kick-off (p <0.001), and multiparous cows had higher incomplete milkings (p <0.001). The number of incomplete milkings showed a higher ratio in terms of reduction in milk production in 26.6% in primiparous cows and 26.7% in multiparous cows (p <0.01). Resumo Uma nova espécie do recentemente descoberto tridentíneo miniatura, Rhinotridens, é descrita da bacia Amazônica no Brasil, Colômbia e Peru. A nova forma difere de Rhinotridenschromocaudatus, espécie-tipo do gênero, pela nadadeira caudal hialina, o barbilhão rictal vestigial ou externamente ausente, a nadadeira anal longa, com 22–29 raios segmentados, além de várias outras características do esqueleto. Uma descrição osteológica da nova espécie é fornecida com base em exemplares diafanizados e corados, e características de morfologia externa são revelados através de microscopia eletrônica de varredura (MEV), que pela primeira vez mostram detalhes da estrutura labial de Tridentinae. Os sete caracteres morfológicos previamente listados como sinapomorfias de Rhinotridens são confirmados na espécie aqui descrita, corroborando sua alocação genérica. Abstract A new species of the recently discovered miniature tridentine catfish, Rhinotridens, is described from the Amazon basin in Brazil, Colombia, and Peru. It differs from Rhinotridens chromocaudatus, type-species of the genus, in having the caudal fin mostly hyaline, a rictal barbel vestigial or externally absent, a longer anal fin with 22–29 segmented rays, and numerous other features from the skeleton. An osteological account of the new species is provided based on cleared and stained specimens, and traits of its external morphology are revealed through scanning electron microscopy (SEM), which for the first time reveal details of the lip structure of tridentines. The seven morphological characters previously listed as synapomorphies for Rhinotridens are confirmed in the species described herein, corroborating its generic allocation. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. ABSTRACT Objective Polycystic ovary syndrome (PCOS) is an endocrine disease characterized by metabolic, reproductive, and psychological manifestations. Growth and differentiation factor 15 (GDF-15) is a cytokine associated with metabolic and inflammatory disorders. Metformin is commonly used for the treatment of PCOS. We investigated the relationship between GDF-15 levels and PCOS, the effect of metformin on GDF-15 levels, and potential biologic pathways related to GDF-15. Subjects and methods The study included 35 women with PCOS and 32 women without PCOS (controls). Both groups were compared in terms of GDF-15 levels. Additional analysis was conducted on samples from 22 women with PCOS who were treated with either metformin (n = 7) or placebo (n = 15), retrieved from a previous randomized, controlled trial. Levels of GDF-15 were measured using MILLIPLEX. The biologic pathways related to GDF-15 were evaluated using the databases STRING, SIGNOR, and Pathway Commons. The statistical analysis was conducted using the software SPSS. Results Levels of GDF-15 were higher in the PCOS group compared with the non-PCOS group (p = 0.039). Among women with PCOS, GDF-15 levels were higher in those treated with metformin compared with placebo (p = 0.007). The proteins related to GDF-15 overlapped between the databases, and a significant interaction was found between GDF-15 and proteins related to PCOS and its complications, including those related to estrogen response, oxidative stress, ovarian infertility, interleukin (IL)-18, IL-4, the ratio of advanced glycation end products to their receptor (AGE/RAGE), leptin, transforming growth factor beta (TGF-β), adipogenesis, and insulin. Conclusion The findings of the present study suggest a relationship between GDF-15 and PCOS and a potential increase in GDF-15 levels with metformin treatment. An additional finding was that GDF-15 could be involved in biologic pathways related to PCOS complications. RESUMO As esponjas estão entre as piores dispersoras do reino animal, no entanto, algumas esponjas de água doce produzem corpos de resistência chamados gêmulas, que têm sido usados como a principal explicação para a ampla distribuição de alguns desses organismos. Esse é o caso de Tubella pennsylvanica Potts 1882, que foi descrita originalmente para Pennsylvania, EUA e foi relatada com uma ampla distribuição, mas com informações escassas e confusas que validem seus registros e seus mecanismos de dispersão. Ela foi registrada para a Amazônia e foi a primeira espécie deTubella reportada fora da Amazônia. No intuito de confirmar o status taxonômico destas ocorrências no Brasil, coletamos espécimes de três locais na bacia amazônica e comparamos suas medidas e características com o material tipo de T. pennsylvanica. Com base na análise dos espécimes, consideramos todos os registros para o Brasil de T. pennsylvanica como sendo uma espécie diferente, a qual descrevemos como Tubella manauara n. sp. Além disso, fornecemos as primeiras imagens das espículas do holótipo de T. pennsylvanica. A nova espécie difere do material tipo de T. pennsylvanica por apresentar espinhos maiores nas megascleras acanthoxeas, e maior tamanho de suas espículas. Os espécimes de Tubella manauara n. sp apresentam variações morfológicas de acordo com o tipo de substrato no qual a esponja se desenvolveu. ABSTRACT Sponges may be among the worst dispersers in the animal kingdom, however, some freshwater sponges produce resting bodies called gemmules, which have been used as the main explanation for the widespread distribution of some of those organisms. This is the case of Tubella pennsylvanica Potts 1882, which was originally described from Pennsylvania, USA, and is reported with a worldwide distribution but with scarce and confusing information to validate its taxonomy and dispersal mechanism. It has been registered in the Amazon and is the first species of Tubella reported outside the Amazon. In order to confirm the taxonomic status of the occurrences in Brazil, we collected specimens from three sites in the Amazon basin and compared their measurements and characteristics with the type material of T. pennsylvanica. Based on this specimen analysis, we consider all records in Brazil of Tubella pennsylvanica to be a different species, which is here described as Tubella manauara n. sp. In addition, we provide the first images of the spicules of the holotype of Tubella pennsylvanica. The new species differs from the type material of Tubella pennsylvanica by presenting larger spines in the megasclere acanthoxeas, and larger size of the spicules. The specimens of Tubella manauara n. sp show morphological variations according to the type of substrate on which the sponge has developed. Progenesis QI (PQI) is a multiplatform bioinformatics tool that facilitates the identification workflow for metabolomics experiments. PQI uses fragmentation data provided by MassBank of North America (MoNA) libraries, among others, for metabolite annotation. However, PQI does not officially support MoNA libraries and other libraries based on structure-data files (.sdf). This paper describes the development and application of a software named MoNA to Progenesis QI Library Converter, allowing PQI and MoNA by correcting the fragmentation data of the library for Progenesis readability. We evaluated several public experimental datasets, including human plasma, plant extracts, cultured cells, bacteria, rat serum, and rat hippocampus. The results showed that it is mandatory to proceed with file conversion of each library to allow PQI to access fragmentation information from .msp (main spectra profile) files. ABSTRACT Inoculating corn with diazotrophic bacteria as growth promoters has been demonstrated to be an efficient agricultural practice in Brazil, mainly due to the root stimulation they provide to plants. This study investigates the corn (Zea mays L.) root architecture in a greenhouse assay where A. baldaniorum Sp245 and H. seropedicae ZAE94 strains were inoculated and evaluated for 22 days under two N levels: 0.6 and 6 mmol L-1 of N. Short-term bioassays were conducted to assess the plant’s response to the addition of indole-acetic acid, two bacterial populations, and two corn genotypes, utilizing image capture software WinRhizo Pro. The growth and distribution of tips, crossing, and length of fine roots were determined to be the most sensitive aspects to inoculation and indole-acetic acid induction. These responses were influenced by the genotype and the number of bacterial cells present. Biomass accumulation analyses quantified these modifications after a 22-day period. Additionally, the growth response was found to be more significant when applying the Hs-ZAE94 strain to plants fertilized with a higher dose of nitrogen (6.0 mmol L-1), and this response was positively correlated with bioassay data. Resumo A soja Bt (transgênica) foi desenvolvida para controlar insetos-praga como Anticarsia gemmatalis e Chrysodeixis includens. Este objetivo foi alcançado com sucesso; no entanto, recentemente, alguns autores afirmaram que as plantas de soja Bt têm sido mais suscetíveis do que a soja não-Bt a Bemisia tabaci MEAM1. Além disso, não se sabe se plantas de soja Bt infestadas por B. tabaci tornam-se menos resistentes às pragas-alvo. Portanto, este estudo teve como objetivo avaliar: (i) se a infestação prévia de B. tabaci pode comprometer a resistência da soja Bt e não Bt a C. includens; (ii) os efeitos de infestações de B. tabaci no crescimento de plantas de soja Bt e não Bt; e (iii) se a alimentação de B. tabaci reduz os teores de clorofila e carotenoides das plantas de soja. Plantas de soja Bt e não Bt pré-infestadas com B. tabaci não apresentaram alterações na resistência a C. includens. Plantas de soja Bt infestadas com B. tabaci apresentaram menor altura de planta do que plantas não infestadas. Diferentemente, as plantas de soja não-Bt não apresentaram redução no crescimento das plantas devido à alimentação de B. tabaci. Plantas de soja Bt sofreram redução de matéria seca apenas sob infestação dupla (B. tabaci e C. includens), enquanto plantas de soja não Bt apresentaram redução de matéria seca quando infestadas por B. tabaci e C. includens ou apenas por C. includens. Abstract Transgenic Bt soybean plants have been developed to control insect pests, such as Anticarsia gemmatalis and Chrysodeixis includens. This objective has been achieved successfully; however, recently, some authors claimed that Bt soybean plants have been more susceptible than non-Bt soybean to Bemisia tabaci MEAM1. In addition, it is unknown whether Bt soybean plants infested by B. tabaci become less resistant to target pests. Therefore, this study aimed to evaluate: (i) whether the previous infestation with B. tabaci can compromise Bt and non-Bt soybean resistance to C. includens; (ii) the effects of B. tabaci infestations on Bt and non-Bt soybean plant growth; and (iii) whether B. tabaci feeding reduces contents of chlorophyll and carotenoids of soybean plants. RESUMO O arroz apresenta altos custos de produção no Brasil devido ao elevado uso de agroquímicos. Algumas rizobactérias promotoras de crescimento de plantas (PGPR) podem ser utilizadas como alternativa aos fertilizantes e produtos fitossanitários. Este trabalho teve como objetivo caracterizar bactérias endofíticas obtidas de raízes de arroz. A caracterização dos isolados foi baseada na morfologia da colônia, resistência a antibióticos, uso de fontes de carbono, atividade enzimática (catalase, amilase, protease, celulase e lipase), solubilização de fosfato inorgânico e região intergênica do 16S-23S rDNA. Com base na morfologia, 68% dos isolados apresentaram crescimento rápido, 46% apresentaram produção abundante de muco e 77% apresentaram colônia viscosa. Todos os isolados foram resistentes ao ácido nalidíxico e 16% deles à estreptomicina. A maioria dos isolados (90%) utilizou monossacarídeos e dissacarídeos no ensaio da fonte de carbono. A maioria dos isolados (95%) foi positiva para catalase e 63,6% foram positivas para amilase, protease, lipase e celulase. Além disso, 59% foram capazes de solubilizar o fosfato. A média do índice enzimático para amilase, celulase e protease foi de 2,8, 3,5 e 1,7, respectivamente. A análise de similaridade revelou alta diversidade entre os isolados, com índice de similaridade de 70% (baseado nas características morfológicas) e 60% (baseado na análise da região intergênica 16S-23S rDNA). ABSTRACT Rice production in Brazil incurs high costs due to the significant use of agrochemicals. Some plant growth-promoting rhizobacteria (PGPR) can be used as alternative to fertilizers and phytosanitary products. Thus, the objective of this study was to characterize endophytic bacteria isolated from roots of rice plants. The isolates were characterized based on colony morphology, antibiotic resistance, carbon sources utilization, enzyme activity (catalase, amylase, protease, cellulase, and lipase), inorganic phosphate solubilization, and the 16S-23S rDNA intergenic region. Morphologically, 68% of the isolates presented a rapid growth rate, 46% presented abundant mucus production, and 77% formed viscous colonies. All isolates were resistant to nalidixic acid and 16% presented resistance to streptomycin. The majority (90%) used monosaccharides and disaccharides in carbon source assays. Most of the isolates (95%) were positive for catalase and 63.6% were positive for amylase, protease, lipase, and cellulase activities. Additionally, 59% of them were able to solubilize phosphate. The mean enzymatic index for amylase, cellulase, and protease was 2.8, 3.5, and 1.7 respectively. The similarity analysis revealed high diversity among the isolates, with similarity indices of 70% (based on morphological characteristics) and 60% (based on the intergenic region 16S-23S rDNA). Resumo A cana-de-açúcar é uma das maiores commodities agrícolas, quando se considera o volume de exportação e o número de empregos gerados. A produção no Nordeste brasileiro, em geral, é baixa, decorrente de diversos fatores, entre eles, a irregularidade de distribuição das chuvas, o que realça a importância de estudos com vistas a minorar os efeitos deletérios do estresse hídrico. Nesse cenário, objetivou-se avaliar o piruvato de cálcio como atenuante do déficit hídrico sobre a qualidade agroindustrial da cana-de-açúcar no segundo ciclo de cultivo. O experimento foi conduzido em casa de vegetação da Universidade Federal de Campina Grande, onde foram testados cinco genótipos de cana-de-açúcar (G1- RB863129, G2- RB92579, G3- RB962962, G4- RB021754 e G5- RB041443) submetidos a três estratégias de manejo da irrigação (E1- Irrigação plena, E2- déficit hídrico com aplicação de 30 mM de piruvato de cálcio e E3- déficit hídrico sem aplicação de piruvato de cálcio), distribuídos em blocos casualizados, em esquema fatorial 5 × 3, com três repetições. O genótipo RB021754 sob déficit hídrico e sem aplicação foliar do piruvato de cálcio aumentou o teor de fibra (13,2%) e o peso do bolo úmido da cana (143,5 g). Abstract Sugarcane is one of the largest agricultural commodities when considering the export volume and the number of jobs generated. Sugarcane production in the Brazilian Northeast region is generally low due to several factors, including the irregular rainfall distribution, which highlights the importance of studies aimed at mitigating the deleterious effects of water stress. In this scenario, this study aimed to evaluate calcium pyruvate as a water deficit attenuator on the agro-industrial quality of sugarcane in the second cycle of cultivation. The experiment was conducted out under greenhouse conditions of the Federal University of Campina Grande, where five sugarcane commercial genotypes tested (G1- RB863129, G2- RB92579, G3- RB962962, G4- RB021754, and G5- RB041443) and three irrigation management strategies (E1- full irrigation, E2- water deficit with application of 30 mM of calcium pyruvate, and E3- water deficit without calcium pyruvate application), distributed in randomized blocks in 5 × 3 factorial arrangement with three replications. The RB021754 genotype under water deficit and without foliar application of calcium pyruvate increased the fiber content (13.2%) and the sugarcane moist cake weight (143.5 g). RESUMO A epidemiologia baseada em esgotos é uma importante ferramenta de saúde pública com grande aplicabilidade no enfrentamento à pandemia de COVID-19, visto que indivíduos infectados são capazes de excretar partículas de SARS-CoV-2. Diante desse contexto, este estudo objetivou correlacionar o número de casos de COVID-19 com variáveis físicas e químicas e presença do vírus em amostras de esgoto bruto provenientes de uma estação de tratamento de esgoto (ETE) do município de Lavras (MG) coletadas entre as 19ª e 25ª semanas epidemiológicas. As amostras foram concentradas pelo método de adsorção em membrana eletronegativa, e o RNA de SARS-CoV-2 foi extraído, detectado e quantificado por reação da transcriptase reversa seguida pela reação em cadeia da polimerase (RT-qPCR) utilizando-se oligonucleotídeos e sondas para o gene N. Ademais, as variáveis pH, sólidos suspensos totais e demanda química de oxigênio foram avaliadas. Os casos de COVID-19 foram obtidos de boletins epidemiológicos disponibilizados pela prefeitura. Na 24ª semana epidemiológica, observou-se redução na concentração viral (cópias L-1) concomitantemente às medidas de enfrentamento à pandemia iniciadas pelo município. Correlação positiva forte e significativa (p < 0,05) entre carga viral e número de casos confirmados foi obtida por meio do teste de Spearman, entretanto não houve correlação entre a presença do vírus e as características físicas e químicas do esgoto. Sugere-se, portanto, que novas pesquisas sejam realizadas com a finalidade de rastrear a concentração de SARS-CoV-2 em águas residuárias, corroborando, desse modo, com o enfrentamento à pandemia e a obtenção de sistemas de vigilância epidemiológica por meio do monitoramento de esgotos.
ABSTRACT Wastewater-based epidemiology is an important public health tool with great applicability when facing the COVID-19 pandemic, since infected individuals are able to excrete SARS-CoV-2 particles. In this context, this study aimed to correlate the number of cases of COVID-19 with physical and chemical variables and the presence of the virus in raw sewage samples from a sewage treatment plant (STP) in the municipality of Lavras, Minas Gerais, Brazil, collected between the 19th and 25th epidemiological weeks. The samples were concentrated by electronegative membrane adsorption method and SARS-CoV-2 RNA was extracted, detected, and quantified by RT-qPCR using oligonucleotides and probes for the N gene. Moreover, the variables pH, total suspended solids, and chemical oxygen demand were evaluated. COVID-19 cases were obtained from epidemiological bulletins made available by the municipal government. From the 24th epidemiological week on, a reduction in viral concentration (copies L-1) was observed concomitantly with the pandemic response countermeasures initiated by the municipality. A strong and significant positive correlation (p<0.05) between viral load and number of confirmed cases was obtained through Spearman’s test. However, there was no correlation between the presence of the virus and the physical and chemical characteristics of the sewage. Therefore, it is suggested that further research be carried out in order to track SARS-CoV-2 concentrations in wastewater, thus corroborating with the fight against the pandemic and the achievement of epidemiological surveillance systems based on sewage monitoring.
