Natural Products Diversity from Genus Aspergillus: A Bioinformatics Study for Insecticidal Activity against the Aedes aegypti Vector Aspergillus

Computational predictive approaches present themselves as an attractive alternative in the search for new compounds that may present toxic or growth regulatory activity in Aedes aegypti. Aspergillus species is a producer of secondary metabolites with diverse biological activities. In this study, it was possible to evaluate the pharmacokinetic and toxicological properties in silico, as well as the prediction of the insecticidal biological activities of the juvenile hormone of natural substances produced by the genus Aspergillus. Initially, 187 molecules were cataloged from various species of Aspergillus spp., and only seven molecules of indole alkaloids were found to exhibit a toxic dose 50% (TD50) after conducting toxicity predictions. The molecular dynamic simulation trajectories were utilized to study parameters such as root mean square deviation, root mean square fluctuation, radius of gyration and intermolecular hydrogen bond to understand the behavior and stability of proposed compounds in the binding pocket of the target protein. Resumo Ensaios pré-clínicos demonstraram que uma biomembrana composta de álcool polivinílico a 1%, contendo proteínas de látex (10%) obtidas da planta medicinal Calotropis procera (Aiton) era biocompatível e estimulava a cicatrização de feridas incisionais e excisionais em modelos murinos, e aspectos mecanísticos foram estabelecidos. No estudo atual, foi investigada a eficácia da biomembrana (BioMemCpLP) em promover a cicatrização de úlceras crônicas em pacientes acometidos de hanseníase. O estudo foi iniciado com 28 voluntários. Cinco foram excluídos, posteriormente, por diferentes desconformidades. Úlceras de 15 pacientes foram tratadas continuamente com BioMemCpLP durante 56 dias. Outros cinco pacientes foram tratados apenas com sulfadiazina de prata e mais três pacientes receberam curativos simples de hidrocoloide com alta capacidade de absorção. Em todos os casos, os curativos foram renovados três vezes por semana e as úlceras foram avaliadas semanalmente quanto à contração e ao progresso da cicatrização, durante 56 dias. A extensão da área cicatrizada, nas úlceras tratadas com BioMemCpLP, foi maior do que nos dois grupos controles. Aproximadamente 88% das úlceras tratadas com BioMemCpLP foram totalmente curadas, antes do dia 56 de tratamento, contra 6% em ambos os grupos de controle. Esse resultado não foi correlacionado com idade/sexo, tempo de lesão ou localização das úlceras, deformidade ou se o paciente alcançou ou não a cura da hanseníase. Abstract Pre-clinical assays demonstrated that a 1% polyvinyl alcohol biomembrane containing latex proteins (10%) from the medicinal plant Calotropis procera was biocompatible and stimulated healing of incisional and excisional wounds in murine models, and the mechanistic aspects were established. The efficacy of the biomembrane (BioMemCpLP) to promote healing of chronic ulcers in leprosy patients was investigated. The study started with 28 volunteers. Five were excluded later due to different disconformities. Ulcers from 15 patients were continuously treated with BioMemCpLP for 56 days. Five patients were treated only with silver sulfadiazine and three patients received plain hydrocolloid wound dressings with high absorption capacity. In all cases, wound dressings were renewed three times a week for 56 days and ulcers were evaluated weekly for contraction and healing progress. The extent of the healed area in the ulcers treated with BioMemCpLP was greater than in the control groups. Approximately 88% of ulcers treated with BioMemCpLP were fully healed before day 56, against 6% in both control groups. This result was not correlated with age/gender, duration or location of ulcers, deformity or whether or not the patient was cured of leprosy. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Abstract Arapaima gigas, an emblematic species of the Amazon region and a longstanding primary fishing resource, currently holds a “Data Deficient” status on the International Union for Conservation of Nature Red List, and is listed as an endangered species in Brazil. The Tocantins River is the most extensively modified large tributary of the Amazon Basin, and thus can affect the dynamics of ichthyofaunal populations. Over a period of 1 year, representatives of the fishing communities and fishermen from 25 fishing communities from four municipalities in the lower Tocantins River region were interviewed, and the obtained information was evaluated based on the literature to survey the population abundance status of A. gigas in the region and its impact on local communities. Among the fishermen interviewed, only one reported still encountering and fishing A. gigas on Jaracuera Island. The disappearance of A. gigas in the region are viewed as having economically disastrous consequences for the residents. Additionally, other endemic fish species are no longer observed in this locality either. Abstract This study examined the taxonomic composition and ecological aspects of planktonic green algae (Chlorophyta) in four urban ponds (Parque da Lagoa, Lagoa Grande, Laguneville, and Pindoba) in Feira de Santana, Bahia State, Brazil. We analyzed 96 samples collected bimonthly in 2022 and identified 54 taxa, with the majority (42) classified as uncommon or sporadic. The most common species were Monoraphidium circinale and Lemmermannia komarekii, found in 100% and 95.8% of samples, respectively. Parque da Lagoa had the highest number of taxa (43), followed by Lagoa Grande (40), Laguneville (31), and Pindoba (30). Most taxa were found in water with high levels of oxygenation (5.8-12.3 mg L−1) and conductivity (400-1000 μS cm−1), neutral to slightly alkaline conditions (pH 7-8), and moderate water temperature (26-27 °C). Significant differences in the composition of planktonic green algae and limnological variables were observed among ponds. These studies underscore the importance of implementing actions aimed at the restoration and conservation of urban ponds in Feira de Santana to avoid biodiversity loss and eutrophication while ensuring the provision of critical ecosystem services, such as local climate regulation. Essential oils (EO) have diverse applications, such as antibacterial and antifungal activity. However, they are susceptible to oxidation in the presence of air, light, and moisture. In addition, they are thermally unstable. In this sense, it is necessary to develop techniques to increase the lifespan of EO. In this work, EO was obtained from cloves and characterized by different techniques. The major compounds found were eugenol (83%), eugenol acetate (9%), and β-caryophyllene (8%). The EO was encapsulated by extrusion using a microfluidic device. The sodium alginate was used as wall material, and the CaCl2 solution was used as a crosslinking agent. The microcapsule presented sizes of 164.7 ± 0.3 μm, with an encapsulation yield of 64 ± 14%. Functional characteristic groups of EO were observed in the microcapsule by infrared and Raman spectroscopies. The microcapsule increased the thermal decomposition of the EO from 162 to 230 °C. Release kinetics of the capsule was performed, with an equilibrium time of 72 h and release of 54% of the EO. Finally, the pure EO and encapsulated EO-microcapsules were applied in the Aedes aegypti larvae control, showing mean lethal concentration necessary to eliminate 50% (LC50) values of 74.4 and 96.9 μg mL-1 and lethal concentration necessary to eliminate 90% (LC90) of 106.2 and 133.3 μg mL-1 for pure EO and encapsulated EO, respectively. Estudando a relação entre a fluidez e a quantidade de água/MC, concluindo que o tipo de agregado e as proporções modificam a fluidez e a demanda de água no estado fresco com impacto no UCS e no WER.
Abstract The influence of the sand proportion in mortars Type II was studied, according to the standard N-CMT-2-01-004/02, using three fine aggregates from the region of Morelia, in Michoacan, and determining their influence in the physico-mechanical properties, while considering the use of different cementitious materials (CM): Portland cement plus lime (B1), and Portland cement plus construction cement (B2). Mortar mixtures were designed for B1 and B2 with volume ratios of aggregates/CM, from 2.25 to 4, determining the fluency, the uniaxial compressive strength (UCS) and the wet electrical resistivity (WER). The relation between the fluency and the ratio water/CM were studied, concluding that the aggregate type and the proportions employed modify the fluency and the water demand in the fresh state of the mortar, and consequently impact the UCS and WER.
Resumen Se analizó la influencia de la proporción de arena en morteros Tipo II según norma N-CMT-2-01-004/02 empleando tres tipos de áridos de la región de Morelia, Michoacán determinando su influencia en sus propiedades físico-mecánicas, contemplando el uso de diferentes materiales cementantes (MC): el cemento Portland más cal (B1), cemento Portland más cemento de albañilería (B2). Se elaboraron mezclas para B1 y B2 con relaciones en volumen arena/MC, desde 2.25 hasta 4, determinando la fluidez, resistencia a compresión uniaxial (UCS) y resistividad eléctrica húmeda (WER). The worldwide emergence of viral diseases such as Zika, Dengue, Chikungunya, West Nile and Yellow Fever urge the search for solutions to eliminate their common vector, the Aedes aegypti mosquito. This paper describes the quantitative structure-activity relationship (QSAR) and docking studies of a series of nine 3-(3-aryl-1,2,4-oxadiazol-5-yl)propionic acids (AOPA), 1-9, previously published by our group. Additionally, three new 1,2,4-oxadiazoles, 10-12, have also been synthesized, characterized and studied. The QSAR and docking studies of all compounds, 1-12, clearly indicate that larger hydrophobic substituents such as biphenyl groups attached on position 3 in 1,2,4-oxadiazoles improve the larvicidal activity. It is worthwhile to mention that nanocapsulation of compounds 10-12 were necessary to help their dissolution in water and these three new 1,2,4-oxadiazoles also exhibited approximately equal or higher larvicidal activities compared to the former prototypes at stage L4. Abstract In the present study, a taxonomic review was conducted on representatives of the genus Tetmemorus (Desmidiaceae, Zygnematophyceae) documented within Brazilian territory. This review involved compiling data from the literature and analyzing samples collected throughout the Bahia State, updating our knowledge about this genus in Brazil. For each identified taxon, we provided information such as description, distribution across biomes and states, watersheds, ecological aspects (including habitat and community types), a list of examined (and excluded) materials, and taxonomic comments. Additionally, a taxonomic key for all species reported in Brazil was provided. Resumo O objetivo deste estudo foi relatar o primeiro registro de Trichodina heterodentata em tuviras Gymnotus carapo. Um cultivo de tuviras no município de Jataí, estado de Goiás, na região Centro-Oeste do Brasil, apresentou taxas de mortalidade de peixes de 3-4% por tanque. A análise macroscópica constatou que os peixes tinham lesões na pele e brânquias. Raspados do tegumento e brânquias foram confeccionadas e secas ao ar à temperatura ambiente e impregnadas com nitrato de prata para identificar as espécies de Trichodinidae causadoras das lesões. Os tricodinídeos foram identificados como T. heterodentata, e suas características foram comparadas com as de T. heterodentata registradas para outras espécies de peixes hospedeiros. Observou-se que a presença de T. heterodentata estava associada ao manejo inadequado dos peixes, baixa qualidade da água, altas taxas de densidade de estocagem e manejo nutricional inadequado.
Abstract The aim of this study was to report the first record of Trichodina heterodentata in banded knifefish Gymnotus carapo. Banded knifefish cultivated in the municipality of Jataí, state of Goiás, in the central western region of Brazil, experienced fish mortality rates of 3-4% per tank. Macroscopic analysis found that fish had lesions on their skin and gills. Smears of the integument and gills were confectioned and air-dried at room temperature and impregnated with silver nitrate to identify the Trichodinidae species causing the lesions. The trichodinid were identified as T. heterodentata, and their characteristics were compared to those from T. heterodentata recorded from other species of host fish. It was observed that the presence of T. heterodentata was associated with inadequate fish management, low water quality, high rates of stocking density, and inadequate nutritional management.

Background: Information regarding diagnosis, treatment, and follow-up of patients with type 1 diabetes (PWT1D) in Mexico is limited. We developed an on-line platform Registro Nacional de Pacientes con Diabetes Tipo 1 (RENACED-DT1). Objective: The objective of the study was to describe the characteristics and healthcare of PWT1D registered in RENACED-DT1. Methods: Analyses of 965 PWT1D from July 2014 to January 2018 in different endocrinology clinics around Mexico. Results: Sixty-one percent were female with median age of 21 years, age at diagnosis 11 years, and disease duration at inclusion 8.2 years. Treatment regimen was basal-bolus in 61% and insulin-pumps in 21% (mainly in the private sector); 33.3% with self-monitoring of blood-glucose (SMBG) ≥4 times/day. Mean HbA1c at last follow-up was 8.7 ± 2.1% (72±23 mmol/mol), 18% had HbA1c < 7% (53 mmol/mol), and 35% > 9% (75 mmol/mol). SMBG ≥ 4 times/day was associated with HbA1c < 7%. Time since diagnosis > 10 years, female sex, BMI ≥ 30 kg/m2, SMBG < 4 times/day, and any hypoglycemia were associated with microvascular complications (p < 0.05). Conclusions: Percentage of patients achieving HbA1c < 7% is low; increased blood glucose monitoring is associated with better glycemic control. The achievement of optimal glycemic control must be increased to reduce the incidence of chronic complications and improve quality of life in PWT1D.

THE USE OF FREE SOFTWARES IN PRACTICAL CLASS ABOUT THE MOLECULAR FILTERS OF ORAL BIOAVAILABILITY OF DRUGS. The study of the physical chemical properties to predict the oral bioavailability of drugs is a widely tool used by researches on the Pharmaceutical and Medicinal Chemistry field. Even though it is a deeply studied subject, it is a complex task to teach to pharmacy students. Lipinski stablished that molecular weight, lipophilicity, hydrogen bond donors and acceptors are of great importance for oral bioavailability. In addition, Veber and Lovering added other properties, such as number of rotatable bonds, fraction of sp3 hybridized carbon and polar surface area that amplified the Lipinski Rule of 5. In this sense, the access to chemical databases and the comprehension of this information is an important task to undergraduate and graduate students. Thus, this work aims a new teaching methodology that values the critical interpretation of data and that favors the students comprehension beyond stablished rules.

Sensitivity of hyperparasitic fungi to alternative products for use in the control of papaya black spot

Resumo O uso de mais de uma técnica de controle pode maximizar a redução dos danos causados pelo fungo Asperisporium caricae agente causal da pinta preta na cultura do mamoeiro. O objetivo deste trabalho foi avaliar a sensibilidade dos fungos Hansfordia pulvinata e Acremonium spp. a produtos alternativos com potencial para uso no controle da pinta-preta. Três isolados de Acremonium spp. (A-598, A-602 e A-617) e um isolado de H. pulvinata (H-611) foram cultivados em meio BDA contendo Agro-Mos®, Bion®, Ecolife®, Hortifospk®, Matriz G®, Vitaphol® separadamente. Fungicida Amistar 500WG® foi utilizado como controle positivo e BDA puro, como controle negativo. A toxicidade dos produtos testados foi determinada com base nos valores do índice biológico, provenientes das médias do crescimento micelial, esporulação e germinação de conídios, em cada unidade experimental. Dessa forma foi possível selecionar os produtos classificados como compatíveis para todos os isolados, e testá-los in vivo. Em casa de vegetação, somente os isolados e os isolados com produtos selecionados, foram aplicados em plantas de mamoeiro com sintomas foliares de pinta-preta. Avaliaram-se a incidência de folhas com hiperparasitas e a porcentagem de lesões de pinta-preta colonizadas pelos isolados testados. Assim o isolado H-611 mostrou-se compatível com a maioria dos produtos alternativos testados, exceto com Hortifos®. Os produtos Bion® e Matriz® apresentaram compatibilidade com todos os isolados testados, podendo ser utilizados em conjunto com o Acremonium spp. e H. pulvinata para controlar a pinta-preta do mamoeiro, uma vez que estes produtos não apresentaram toxidade sobre os fungos hiperparasitas.
Abstract The use of more than one control technique can maximize the reduction of the damages caused by the fungus Asperisporium caricae causal agent of the black spot in the papaya crop. The objective of this work was to evaluate the sensitivity of the fungi Hansfordia pulvinata and Acremonium spp. to alternative products with potential for use in the control of the black-spotted ptarmigan. Three isolates of Acremonium spp. (A-598, A-602 and A-617) and an isolate of H. pulvinata (H-611) were grown in BDA medium containing Agro-Mos®, Bion®, Ecolife®, Hortifospk®, Matriz G®, Vitaphol® separately. The Amistar 500WG ® fungicide was used as a positive control and pure BDA as a negative control. The toxicity of the tested products was determined based on the values of the biological index, derived from the means of mycelial growth, sporulation and germination of conidia, in each experimental unit. In this way it was possible to select the products classified as compatible for all isolates, and to test them in vivo. In the greenhouse, only the isolates and isolates with selected products, were applied in papaya plants with foliar symptoms of black-spotted. The incidence of leaves with hyperparasites and the percentage of black-painted lesions colonized by the tested isolates were evaluated. Thus, the H-611 isolate proved to be compatible with most of the alternative products tested, except with Hortifos®. Bion® and Matrix® products were compatible with all tested isolates and could be used in conjunction with Acremonium spp. and H. pulvinata to control the papaya black spot, since these products did not present toxicity on the hyperparasitic fungi.

ABSTRACT Background: Regional information regarding the characteristics of patients with coronavirus disease (COVID)-19 is needed for a better understanding of the pandemic. Objective: The objective of the study to describe the clinical features of COVID-19 patients diagnosed in a tertiary-care center in Mexico City and to assess differences according to the treatment setting (ambulatory vs. hospital) and to the need of intensive care (IC). Methods: We conducted a prospective cohort, including consecutive patients with COVID-19 from February 26, 2020 to April 11, 2020. Results: We identified 309 patients (140 inpatients and 169 outpatients). The median age was 43 years (interquartile range, 33-54), 59.2% men, and 18.6% healthcare workers (12.3% from our center). The median body mass index (BMI) was 29.00 kg/m2 and 39.6% had obesity. Compared to outpatients, inpatients were older, had comorbidities, cough, and dyspnea more frequently. Twenty-nine (20.7%) inpatients required treatment in the IC unit (ICU). History of diabetes (type 1 or 2) and abdominal pain were more common in ICU patients compared to non-ICU patients. ICU patients had higher BMIs, higher respiratory rates, and lower room-air capillary oxygen saturations. ICU patients showed a more severe inflammatory response as assessed by white blood cell count, neutrophil and platelet count, C-reactive protein, ferritin, procalcitonin, and albumin levels. By the end of the study period, 65 inpatients had been discharged because of improvement, 70 continued hospitalized, and five had died. Conclusions: Patients with comorbidities, either middle-age obese or elderly complaining of fever, cough, or dyspnea, were more likely to be admitted. At admission, patients with diabetes, high BMI, and clinical or laboratory findings consistent with a severe inflammatory state were more likely to require IC.
